Abstract:[Background] Bottleneck problems such as poor yield and low productivity had been hindering the progress in commercial production of polymalic acid from glucose by Aureobasidium pullulans.[Objective] To enhance polymalic acid yield and productivity from Aureobasidium pullulans BK-10 by optimizing the fermentation medium and conditions.[Methods] Single-factor experiment,Plackett-Burman design and orthogonal experiment were adopted to evaluate the importance of various fermentation medium ingredients and optimize kinds of fermentation conditions.A 5 L fermentor experiment was conducted to verify the above optimized conditions.Results] The optimum fermentation medium consisted of 100 g/L glucose,1.5 g/L urea,0.20 g/L KH2PO4,0.20 g/L ZnSO4,0.05 g/L MgSO4,0.75 g/L KCl,30 g/L CaCO3,0.01％ Tween-80,the optimal fermentation temperature was found 26 ℃ and the optimal working volume was 50 mL of fermentation medium in a 250 mL flask.[Conclusion] Yield of polymalic acid from glucose reached 0.71 g/g with an increase of 18.33％,and the productivity was 0.89 g/(L·h) which was increased by 71.15％ as compared with the unoptimized conditions.These results show great economical potential for commercial production of polymalic acid even or L-malic acid by fermentation from glucose.

Abstract:Poly (beta-l-malic acid) (PMLA) is a water-soluble polyester with many attractive properties in chemical industry and medicine development. However, the low titer of PMLA in the available producer strains limits further industrialization efforts and restricts its many potential applications. In order to solve this problem, a new strain with the distinguished high productivity of PMLA was isolated from fresh plants samples. It was characterized as the candidate of Aureobasidium pullulans based on the morphology and phylogenetic analyses of the internal transcribed spacer sequences. After the optimization of culture conditions, the highest PMLA concentration (62.27 g l(-1)) could be achieved in the shake flask scale. In addition, the contribution of the carbon flux to exopolysaccharide (EPS) and PMLA could be regulated by the addition of CaCO3 in the medium. This high-level fermentation process was further scaled up in the 10 l benchtop fermentor with a high PMLA concentration (57.2 g l(-1)) and productivity (0.35 g l(-1) h(-1)), which are the highest level in all the literature. Finally, the suitable acid hydrolysis conditions of PMLA were also investigated with regard to the production of l-malic acid, and the kinetics of PMLA acid hydrolysis was modeled to simulate the whole degradation process. The present work paved the road to produce this multifunctional biomaterial (PMLA) at industrial scale and promised one alternative method to produce l-malic acid in the future.

Abstract:Melanin has good nutritional and medicinal value; however, its extraction rate is extremely low. This study explored the edible and medicinal fungus Inonotus hispidus fruiting body melanin (IHFM) extraction process and solid-state fermentation conditions. The results showed that the best way to extract IHFM is the compound enzymatic method, with complex enzyme 26.63 mg/g, liquid material ratio 5:1, enzymatic hydrolysis 80 min, pH 4.61, and enzymolysis temperature at 36.07 °C. The yield of IHFM was 23.73 ± 0.57%, which was equivalent to 1.27 times before optimization. The best solid medium formula was normal pH, rice 20 g per cultivation bottle, maltose 22 g/L, beef extract 4.4 g/L, carbon-nitrogen ratio 5:1, and liquid-to-material ratio 1.1:1, where the IHFM yield was 31.80 ± 1.34%, which was equivalent to 1.7 times that before optimization. In summary, solid-state fermentation and extraction optimization greatly improved the yield of melanin, provided a reference to produce melanin, and laid a foundation for the development and utilization of melanin.
High Yield And Cost-Effective Production Of Poly(Gamma-Glutamic Acid) With Bacillus Subtilis

Abstract:Poly(gamma-glutamic acid) (gamma-PGA) is a promising biopolymer with many potential industrial and pharmaceutical applications. To reduce the production costs, the effects of yeast extract and L-glutamate in the substrate for gamma-PGA production were investigated systematically at shake flask scale. The results showed that lower concentrations of yeast extract (40 g/L) and L-glutamate (30 g/L) were beneficial for the cost-effective production of gamma-PGA in the formulated medium. By maintaining the glucose concentration in the range of 3-10 g/L via a fed-batch strategy in a 10-L fermentor, the production of gamma-PGA was greatly improved with the highest gamma-PGA concentration of 101.1 g/L, a productivity of 2.19 g/L . h and a yield of 0.57 g/g total substrate, which is about 1.4- to 3.2-fold higher than those in the batch fermentation. Finally, this high-density fermentation process was successfully scaled up in a 100-L fermentor. The present work provides a powerful approach to produce this biopolymer as a bulk chemical in large scale.
Extraction, physicochemical properties, and antioxidant activity of natural melanin from Auricularia heimuer fermentation

Abstract:In this study, single-factor experiments, Box-Behnken design (BBD), and response surface methodology (RSM) were employed to investigate the effects of alkali-soluble pH, acid precipitation pH, and microwave time on the extraction yield of Auricularia heimuer melanin (AHM) from fermentation. Ultraviolet-visible spectrum (UV-Vis), Fourier transform infrared (FT-IR) spectroscopy, scanning electron microscope (SEM), and high-performance liquid chromatography (HPLC) were used to analyze the extracted AHM. The solubility, stability, and antioxidant activities of AHM were also measured. The results showed that alkali-soluble pH, acid precipitation pH, and microwave time significantly affected the AHM yield, with the following optimized microwave-assisted extraction conditions: alkali-soluble pH of 12.3, acid precipitation pH of 3.1, and microwave time of 53 min, resulting in an AHM extraction yield of 0.4042%. AHM exhibited a strong absorption at 210 nm, similar to melanin from other sources. FT-IR spectroscopy also revealed that AHM exhibited the three characteristic absorption peaks of natural melanin. The HPLC chromatogram profile of AHM showed a single symmetrical elution peak with a 2.435 min retention time. AHM was highly soluble in alkali solution, insoluble in distilled water and organic solvents, and demonstrated strong DPPH, ·OH, and ABTS free radical scavenging activities. This study provides technical support to optimize AHM extraction for use in the medical and food industries.

Optimization of Production Process Conditions for Mulberry Juice Fermentation with Lactic Acid Bacteria Using Dual Response Surface Methodology

Abstract:Mulberry fruit juice fermented with lactic acid bacteria is a nutrient-rich healthcare food.In order to improve production level of mulberry fruit juice fermentation and product quality,production process conditions including inoculation amount,fermentation time,and fermentation temperature were optimized using dual response surface methodology based on means and variance by taking lactic acid output as the evaluation index.The results showed that the process conditions optimized by dual response surface methodology had good stability.The optimal process conditions were as follows: fermentation time 6.85 d,fermentation temperature 35 ℃,and inoculation amount of lactic acid bacteria 3.13%(volume fraction).The predicted mean of lactic acid yield was 9.636 g/L,and the variance was 0.212 8.Verification test to the optimized process conditions showed that mean of lactic acid yield was 9.634 g/L,and the variance was 0.215 1 whose absolute error was less than 1%,meaning that the fermentation efficiency reached a high level.Under the optimized process conditions,the content of glucose and fructose in fermented mulberry fruit juice was decreased by 82% and 61% respectively compared with mulberry juice before fermentation,indicating that most sugar in mulberry fruit juice had been converted into lactic acid and thus making it more beneficial to human health.

Abstract:Agaricus bitorquis (Quel.) Sacc. Chaidam (ABSC) is a wild edible fungus uniquely found in the Tibet Plateau. ABSC is rich in polysaccharides that are considered biologically active. This study aimed to determine the feasibility of enhancing exopolysaccharide (EPS) production by ABSC in shake flask culture by supplementing the fermentation medium with anthocyanin extract. Different concentrations of Lycium ruthenicum Murr. (LRM) anthocyanin crude extract were tested on ABSC fermentation. The activity of phosphoglucose isomerase (PGI), phosphoglucose mutase (PGM), and phosphomannose isomerase (PMI), enzymes presumably involved in EPS synthesis by ABSC, was determined. ABSC transcriptomic profile in response to the presence of anthocyanins during fermentation was also investigated. LRM anthocyanin crude extract (0.06 mg/mL) was most effective in increasing EPS content and mycelial biomass (by 208.10% and 105.30%, respectively, P < 0.01). The activity of PGI, PGM, and PMI was increased in a medium where LRM anthocyanin extract and its main components (proanthocyanidins and petunia anthocyanin) were added. RNA-Seq analysis showed that 349 genes of ABSC were differentially expressed during fermentation in the medium containing anthocyanin extract of LRM; 93 genes were up-regulated and 256 genes down-regulated. From gene ontology enrichment analysis, differentially expressed genes were mostly assigned to carbohydrate metabolism and signal transduction categories. Collectively, LRM anthocyanins extract positively affected EPS production and mycelial biomass during ABSC fermentation. Our study provides a novel strategy for improving EPS production and mycelial growth during ABSC liquid submerged fermentation.
High-yield production of β-arbutin by identifying and eliminating byproducts formation

Abstract:β-Arbutin is a plant-derived glycoside and widely used in cosmetic and pharmaceutical industries because of its safe and effective skin-lightening property as well as anti-oxidant, anti-microbial, and anti-inflammatory activities. In recent years, microbial fermentation has become a highly promising method for the production of β-arbutin. However, this method suffers from low titer and low yield, which has become the bottleneck for its widely industrial application. In this study, we used β-arbutin to demonstrate methods for improving yields for industrial-scale production in Escherichia coli. First, the supply of precursors phosphoenolpyruvate and uridine diphosphate glucose was improved, leading to a 4.6-fold increase in β-arbutin production in shaking flasks. The engineered strain produced 36.12 g/L β-arbutin with a yield of 0.11 g/g glucose in a 3-L bioreactor. Next, based on the substrate and product's structural similarity, an endogenous O-acetyltransferase was identified as responsible for 6-O-acetylarbutin formation for the first time. Eliminating the formation of byproducts, including 6-O-acetylarbutin, tyrosine, and acetate, resulted in an engineered strain producing 43.79 g/L β-arbutin with a yield of 0.22 g/g glucose in fed-batch fermentation. Thus, the yield increased twofold by eliminating byproducts formation. To the best of our knowledge, this is the highest titer and yield of β-arbutin ever reported, paving the way for the industrial production of β-arbutin. This study demonstrated a systematic strategy to alleviate undesirable byproduct accumulation and improve the titer and yield of target products. Abstract:The optimization of fermentation process,extraction and separation of chaetoglobosin A were studied with HPLC analytical techniques to establish high yield,low cost,pollution-free scale fermentation process.[Results] The fermentation process of chaetoglobosin A was optimized by Uniform Design Method.The fermentation medium in the optimal combination of carbon and nitrogen source were determined sucrose 17 g/L,starch concentration of 6 g/L respectively,peptone concentration was 0.5 g/L and fermented for 13 d.The results showed that eluent ethyl acetate was the suitable solvent for the preparation of test samples while considering both extract ratio and fungicidal activity.Found eluant ethyl acetate,petroleum ether(1∶3),developing solvent system ethyl acetate,petroleum ether,formic acid(37∶63∶1) as the best separation conditions.[Conclusions] The ultimate production of 0.17 g/L under the optimum conditions could be reached,which was increased by 13.3% compared to 0.15 g/L under the original fermentation conditions.

Abstract:Melanin is a natural pigment ubiquitously present in living organisms, including bacteria, fungi, plants, and animals. Melanin produced by the edible mushroom Auricularia auricula has a remarkable potential for resource development. Several A. auricula strains planted across China were collected and analyzed for mycelial growth rate and colony RGB value for color block. Further, the effects of various nutrients on melanin formation, including different carbon and nitrogen sources were evaluated to optimize medium for submerged fermentation. The pathways involved in the biosynthesis of melanin in A. auricula were investigated using an enzyme inhibitor assay and intermediate determination. In addition, the functional activity of purified A. auricula melanin was assessed. The highest melanin yield (1.797 g/L) was displayed by strain AU‐3 in medium I. A. auricula melanin was composed of eumelanin, pheomelanin and 1,8‐dihydroxynaphthalene melanin, and the biosynthetic pathways involved were Raper‐Mason and 1,8‐dihydroxynaphthalene melanin pathway. High-level production of pullulan and its biosynthesis regulation in Aureobasidium pullulans BL06

Abstract:Pullulan has many potential applications in the food, pharmaceutical, cosmetic and environmental industries. However, the yield and molecular properties of pullulan produced by various strains still need to be promoted to fit the application needs. A novel yeast-like strain Aureobasidium pullulans BL06 producing high molecular weight (Mw) pullulan (3.3 × 10 6 Da) was isolated and identified in this study. The remarkable Mw of pullulan produced by A. pullulans BL06 was the highest level ever reported thus far. To further regulate the biosynthesis of pullulan in A. pullulans BL06, three gene knockout strains A. pullulans BL06 ΔPMAs, A. pullulans BL06 Δmel, and A. pullulans BL06 ΔPMAsΔmel, were constructed. The results showed that A. pullulans BL06 ΔPMAs could produce 140.2 g/L of moderate Mw (1.3 × 10 5 Da) pullulan after 120 h of fermentation. The highest yield level of pullulan to date could vastly reduce its production cost and expand its application scope and potential. The application experiments in food preservation showed that the moderate-Mw pullulan obtained in this work could reduce the weight loss of celery cabbages and mangos by 12.5% and 22%, respectively. Thus, the novel strains A. pullulans BL06 and A. pullulans BL06 ΔPMAs possessed unlimited development prospects in pullulan production at various Mw ranges and pullulan applications in multiple fields.

Abstract:Owing to the growing need for natural materials in different fields, studying melanin production from biological sources is imperative. In the current study, the extracellular melanin pigment was produced by the fungus Curvularia soli AS21 ON076460. The factors that affect the production of melanin were optimized by the Plackett-Burman design (P-BD). The effect of gamma irradiation on melanin productivity was investigated. The maximum melanin yield (3.376 mg/L) was elicited by a stimulus of gamma irradiation at 1.0 kGy. The results evoked that, Curvularia soli AS21 ON076460 melanin exhibited excellent antimicrobial activity against all tested bacteria and fungi. Klebsiella pneumoniae ATCC 13883 and P. digitatum were mostly affected by melanin registering the inhibition zone diameters of 37.51 ± 0.012 and 44.25 ± 0.214 mm, respectively. Moreover, Curvularia soli AS21 ON076460 melanin indicated a significant antiviral efficacy (77% inhibition) of Herpes simplex virus (HSV1). The melanin pigment showed antioxidant activities with IC 50 of 42 ± 0.021 and 17 ± 0.02 μg/mL against DPPH and NO, respectively. Melanin had cytotoxic action against human breast cancer and skin cancer cell lines (Mcf7and A431) as well as exerting a low percentage of cell death against normal skin cell lines (Hfb4). Melanin was effective in wound management of human skin cells by 63.04 ± 1.83% compared with control (68.67 ± 1.10%). The novelty in the study is attributed to the possibility of using gamma rays as a safe method in small economic doses to stimulate melanin production from the fungi that have been isolated. In summary, melanin produced from fungi has significant biological activities that encourage its usage as a supportive medical route.

Abstract:α-Arbutin is an effective skin-whitening cosmetic ingredient and hyperpigmentation therapy agent. It can be synthesized by one-step enzymatic glycosylation of hydroquinone (HQ), but limited by the low yield. Amylosucrase (Amy-1) from Xanthomonas campestris pv. campestris 8004 was recently identified with high HQ glycosylation activity. In this study, whole-cell transformation by Amy-1 was optimized and process scale-up was evaluated in 5000-L reactor. In comparison with purified Amy-1, whole-cell catalyst of recombinant E. coli displays better tolerance against inhibitors (oxidized products of HQ) and requires lower molar ratio of sucrose and HQ to reach high conversion rate (> 99%). Excess accumulation of glucose (0.6–1.0 M) derived from sucrose hydrolysis inhibits HQ glycosylation rate by 46–60%, which suggests the importance of balancing HQ glycosylation rate and sucrose hydrolysis rate by adjusting the activity of whole-cell catalyst and HQ-fed rate. Using optimal conditions, 540 mM of final concentration and 95% of molar conversion rate were obtained within 13–18 h in laboratory scale. For industrial scale-up production, 398 mM and 375 mM of final concentration with high conversion rates (~ 95%) were obtained in 3500-L and 4000-L of reaction volume, respectively. These yields and productivities (4.5–4.9 kg kL−1 h−1) were the highest by comparing to the best we known. Hence, high-yield production of α-arbutin by batch-feeding whole-cell biotransformation was successfully achieved in the 5000-L reaction scale.

Abstract:The secondary metabolite Monacolin K (MK), derived from Monascus, exhibits potential lipid-lowering and anti-tumor effects. In this study, a mutant strain M. purpureus M400–6 was generated through X-ray treatment, resulting in an average MK production of 181.97 mg/L (a 54.20% increase compared to the parent strain M. purpureus M1) over five successive generations. Furthermore, optimization of medium compositions using response surface methodology led to a further enhancement in MK production by the mutant strain to 266.63 mg/L. Moreover, based on the outcomes of single-factor experiments, the addition of L-citrulline demonstrated a significantly higher level of enhancement in MK production. Therefore, a comparative transcriptome analysis was conducted to unveil the impact of L-citrulline on MK biosynthesis in M. purpureus. The data revealed a significant upregulation of glycolysis pathways, while oxidative phosphorylation including the TCA cycle and the electron transport chain, exhibited significant downregulation. Furthermore, the metabolisms of serine, glycine, threonine, alanine, aspartic acid and glutamate were found to be significantly disturbed. Additionally, the expressions of MPs and MK biosynthetic gene cluster showed a significant downregulation and upregulation respectively. Overall, these findings provide insights into enhancing MK production in submerged fermentation and elucidate the impact of L-citrulline on secondary metabolism.

Abstract:Natural pigments are playing important roles in our daily lives. They not only make products colorful but also provide various health benefits for humans. In addition, Pycnoporus genus, listed as food- and cosmetic-grade microorganism, is one of the promising organisms for developing natural pigments. In this study, a new fungal strain with high efficiency in producing intense orange pigments was isolated and identified as Pycnoporus sanguineus SYBC-L7. Different agro-industrial wastes were applied to evaluate the growth and pigment production of strain SYBC-L7. SYBC-L7 can grow rapidly and effectively produce pigments using wood chips as substrate in solid-state fermentation (SSF). Culture conditions were also optimized for value-added pigments production and the optimum production conditions were glucose as carbon source, ammonium tartrate as nitrogen source, initial pH 6.0, and relative humidity of 65%. Pigment components, cinnabarinic acid, tramesanguin, and 2-amino-9-formylphenoxazone-1-carbonic acid were confirmed by liquid chromatography–mass spectrometry. Meanwhile, an agar plate diffusion assay was performed to evaluate the antimicrobial activity of the pigment. These pigments showed more significant inhibition of Gram-positive than Gram-negative bacteria. The results showed that Pycnoporus sanguineus SYBC-L7 was able to cost-effectively produce intense natural orange pigments with antibacterial activity in SSF, which is the basis of their large-scale production and application.

Abstract:The potential economic values and applications of natural yellow and red pigments in the food industry, cosmetic, and textile cannot be underestimated. So the improvement of a bioprocess for better production of Monascus pigments (MPs) using nutrition regulation under submerged fermentation was carried out. An organic acid together with fermentation conditions such as pH has proved to be effective for pigment enhancement as well as cell growth. The combined effect of 10 g/L peptone and 1.5 g/L pyruvic acid yielded yellow (99.81 ± 5.25 AU/ml) and red (97.22 ± 6.12 AU/ml) pigments which are about 5 folds compared to control. Further investigation indicated that the pigment production in the acidic medium is better than the nearly neutral medium. The highest yellow (574.76 ± 6.18 AU/ml) and red (570.98 ± 7.66 AU/ml) pigment production were attained at the initial pH of 5.0. The study again showed that the effect of fermentation time on pigments, pH curve, and biomass production did not reach the decline phase given an indication of adequate nutrient in the broth for probable growth and pigment production.The present study gives the basic information on which the scaled-up production of valuable metabolites by filamentous fungi in submerged fermentation could be carried on.

Metabolic engineering of Aureobasidium melanogenum for the overproduction of putrescine by improved L-ornithine biosynthesis

Abstract:Aureobasidium melanogenum HN6.2 is a high siderophore-producing yeast-like fungal strain. After blocking siderophore biosynthesis and attenuating the expression of the ornithine carbamoyltransferase gene (the OTC gene), the obtained D-LCFAO-cre strain produced 2.1 ± 0.02 mg of intracellular L-ornithine per mg of the protein. The overexpression of the L-ornithine decarboxylase gene (the SPE1-S gene) from Saccharomyces cerevisiae in the mutant D-LCFAO-cre could make the transformant E-SPE1-S synthesize 3.6 ± 0.1 of intracellular ornithine per mg of protein and produce 10.5 g/L of putrescine. The further overexpression of the ArgB/C gene encoding bifunctional acetylglutamate kinase/N-acetyl-gamma-glutamyl-phosphate reductase in the transformant E-SPE1-S caused the transformant E-SPE1-S-ArgB/C to accumulate L-ornithine (4.2 mg/mg protein) and to produce 21.3 g/L of putrescine. During fed-batch fermentation, the transformant E-SPE1-S-ArgB/C could produce 33.4 g/L of putrescine, the yield was 0.96 g/g of glucose, and the productivity was 0.28 g/L/h. The putrescine titer was much higher than that produced by most engineered strains obtained thus far.

Abstract:Monascus pigments (MPs) have been used in the food industry for more than 2,000 years and are known for their safety, bold coloring, and physiological activity. MPs are mainly yellow (YMPs), orange (OMPs), and red (RMPs). In this study, a mutant strain Monascus purpureus H14 with high production of water-soluble YMPs (WSYMPs, λ max at 370 nm) was generated instead of primary YMPs ( λ max at 420 nm), OMPs ( λ max at 470 nm), and RMPs ( λ max at 510 nm) produced by the parent strain M. purpureus LQ-6 through dual mutagenesis of atmospheric and room-temperature plasma and heavy ion beam irradiation (HIBI), producing 22.68 U/ml extracellular YMPs and 10.67 U/ml intracellular YMPs. WSYMP production was increased by 289.51% in optimal conditions after response surface methodology was applied in submerged fermentation. Application of combined immobilized fermentation and extractive fermentation improved productivity to 16.89 U/ml/day, 6.70 times greater than with conservative submerged fermentation. The produced WSYMPs exhibited good tone stability to environmental factors, but their pigment values were unstable to pH, light, and high concentrations of Ca 2+ , Zn 2+ , Fe 2+ , Cu 2+ , and Mg 2+ . Furtherly, the produced exYMPs were identified as two yellow monascus pigment components (monascusone B and C 21 H 27 NO 7 S) by UHPLC-ESI-MS. This strategy may be extended to industrial production of premium WSYMPs using Monascus .
