Abstract:The spermatozoon ultrastructure of Moerella iridescens was observed by electron microscopy.Images showed that the sperm structure of Moerella iridescens is that of a typical primitive type that includes a head,middle piece,and tail.The head is composed of an acrosome and nucleus.The acrosome exhibits a crooked,upside-down V shape,and the acrosome displays an electron dense outside and lower electron density inside.The sub-acrosomal space has no special structure except homogeneous low electron density materials.The slender and spiral nucleus has a shallow front nuclear pocket but no posterior nuclear pocket.The middle piece of the sperm is composed of mitochondria and two centrioles.Four mitochondria could be seen in cross-sections of the middle piece,while we observed 2~4 mitochondria in longitudinal sections.Overall,there were10 mitochondria in the middle piece of sperm.The flagella of sperm are slender and exhibit typical 9+2 microtubular structure.Images also showed that there were two distinct differences in morphological configuration between this sperm and sperm from other molluscs.First,the sperm morphology is a rare spiral shape,while sperm from bivalve mollusks are typically spherical,cylindrical,or oval.Second,the number of mitochondria found inside sperm is also atypical,where other bivalve mollusks typically possess 4~5,but10 were found in sperm from M.iridescens.The atypical sperm morphology described here is conducive to enhancing sperm motility,and thereby improving the successful rate of fertilization.
Spermatological characters of Stephanostomum bicoronatum (Digenea, Acanthocolpidae), an intestinal parasite of the Brown meagre Sciaena umbra (Teleostei: Sciaenidae) off the Tunisian coast

Abstract:The ultrastructural spermatozoon organisation of the acanthocolpid digenean Stephanostomum bicoronatum have been described by micrographs of transmission electron microscopy (TEM). Live digeneans were isolated from the digestive tract of the Brown meagre Sciaena umbra (Teleostei: Sciaenidae), obtained from artisanal fishing off the coast of La Chebba (Tunisia Mediterranean Sea). The ultrastructural study reveals that the male gamete of S. bicoronatum is a filamentous cell tapered at both extremities, which present typical characteristics of digenean spermatozoa of the type III, namely two axonemes of the 9 + '1' pattern of trepaxonematan Platyhelminthes, nucleus, one mitochondrion, external ornamentation of the plasma membrane associated with cortical microtubules and located in the posterior part of the anterior region of the sperm cell, two parallel bundles of cortical microtubules, and maximum number of cortical microtubules in the anterior part of the spermatozoon. In addition, the male gamete of S. bicoronatum shows spine-like bodies, a large amount of glycogen granules and a posterior extremity containing only the nucleus. The results are compared and discussed according to the existing data in other digeneans, particularly with those of the family Acanthocolpidae.

Abstract:Sperm motility is crucial for successful fertilization. Highly decorated doublet microtubules (DMTs) form the sperm tail skeleton, which propels the movement of spermatozoa. Using cryo-electron microscopy (cryo-EM) and artificial intelligence (AI)-based modeling, we determined the structures of mouse and human sperm DMTs and built an atomic model of the 48-nm repeat of the mouse sperm DMT. Our analysis revealed 47 DMT-associated proteins, including 45 microtubule inner proteins (MIPs). We identified 10 sperm-specific MIPs, including seven classes of Tektin5 in the lumen of the A tubule and FAM166 family members that bind the intra-tubulin interfaces. Interestingly, the human sperm DMT lacks some MIPs compared with the mouse sperm DMT. We also discovered variants in 10 distinct MIPs associated with a subtype of asthenozoospermia characterized by impaired sperm motility without evident morphological abnormalities. Our study highlights the conservation and tissue/species specificity of DMTs and expands the genetic spectrum of male infertility.

Abstract:The present study aimed to investigate the macroscopic and histological structure of the gastrointestinal tract (GIT) of Larimichthyscrocea (Richardson, 1846). It consists of esophagus, stomach regions, pyloric caeca, intestinal regions, and rectum. Sixteen tubular light yellowish pyloric caeca of similar sizes were observed in all individuals. The digestive wall consists of mucosa, submucosa, muscularis, and adventitia. No major differences appeared in the structure of the tunica, epithelial cell types, connective tissues and musculature glands of L.crocea GIT. The mucosal epithelia in the oesophagus has longitudinal branched folds with frontward and hindmost zones. The gastric tunica mucosa has a characteristic folded structure and can be divided into three regions. The intestinal tunica mucosa is characterized by villi structures and numerous mucus-secreting cells. Mucus-secreting goblet cells were strongly positive to AB at pH 2.5 in the oesophagus (excluding gastro-oesophageal junction) and intestine mucosal regions, which indicates an abundance of carboxylate mucins. The surface epithelia of the gastric mucosa is PAS-positive and AB-negative. SEM examination revealed that cells in the epithelium of the esophagus have an unbroken apical layer and goblet cells. The intestinal coefficient (IC) of L.crocea was 0.80 ± 0.21, consistent with a carnivorous or omnivorous habit. Our study adds the knowledge of the digestive system of L.crocea and might be useful in the management of L.crocea stocks.

Abstract:Acanthopagrus schlegelii is an autochthonous teleost species concerning the remarkable economic importance and prevalent fish cultivated in China as well as in different nations of South-East Asia. Little is known about the digestive tract (DT) morphology of A. schlegelii. Therefore, anatomical and histomorphological aspects of A. schlegelii DT were examined by light and scanning electron microscope (SEM). Anatomically, DT of A. schlegelii was mainly formed of oesophagus, well-developed stomach and fingers like pyloric caeca (four in number), intestinal regions and rectum. Histomorphologically, oesophagus occurred as a shorter tube-like organ presenting a longitudinal folded mucosa that connects oropharynx cavity to stomach. Stomach was a muscular thick-wall organ that included three regions, thickly longitudinal folds were observed in the first (cardiac) and last (pyloric) regions, whereas the second (fundic) region showed folds in different directions. Long villi were observed within pyloric caeca and anterior intestine. Tunica muscularis appeared narrow in the anterior intestine, whereas thicker in the posterior part of the intestine. Collectively, anatomical and histomorphological aspects of A. schlegelii DT are consistent with the carnivorous habit of this species. These data could be a potential source to modify better methods of nutrition and identify the DT pathogenic conditions in farming of this fish.

Abstract:The structure of the oviduct of female Macrobrachium rosenbergii during spawning was examined with optical microscope and transmission electron microscope(TEM).In Macrobrachium rosenbergii,a pair of oviducts originates from the anterolateral part of the ovary ends at the female genital pores situated in the basipodite of the 3rd thoracic appendage.The oviduct wall,folded in some areas, consists of a basal lamina and a layer of columnar epithelial cells.There are two types of epithelial cells,one with light cytoplasm and the other with dark cytoplasm,found in the oviduct wall.The free surface of the epithelial cells possesses long dense microvilli.And beneath the microvilli,there is a thick layer of secreted substance.The nucleus is in oval shape with one or several nucleoli.The cytoplasm contains abundant mitochondria and rough endoplasmic reticulums.After shedding the eggs,the cell membrane and the microvilli break and the cytoplasm leaks out.The layer of secreted substance disappears,only a little residue of it remains.

Abstract:The ultrastructure of spermiogenesis and the mature spermatozoon in Catenotaenia pusilla (Cestoda: Catenotaeniidae) is described. Spermiogenesis is characterized by the presence of a single axoneme which grows on the outside of a cytoplasmic extension at an angle of 45°. Flagellar rotation and proximodistal fusion are produced in this process. The centrioles lack striated roots and an intercentriolar body. In the mature spermatozoon four different regions are described. The anterior extremity is capped by an apical cone and presents two helical crest-like bodies of unequal length. The axoneme, of the 9 + '1' pattern of the Trepaxonemata, presents a periaxonemal sheath. The cortical microtubules form a spiral pattern at an angle of about 40° to the hypothetical spermatozoon axis. The nucleus is kidney- to horseshoe-shaped in cross section. Granules and proteinaceus walls are not observed in the spermatozoon of C. pusilla.

Abstract:Acanthocephalans are dioecious parasites that gain sexual maturity in the alimentary canal of their definitive hosts (gnathostome vertebrates). This initial survey by light and transmission electron microscopy was conducted on the functional organization of the ovarian balls and uterine bell in mature females and on Saefftigen's pouch and the copulatory bursa in males. We studied these structures via the example of Centrorhynchus globocaudatus (Palaeacanthocephala) in Falco tinnunculus and Buteo buteo, from the Province of Ferrara (Northern Italy). Our study confirms that the ovarian balls have surface microvilli and consist of a multinucleate supporting syncytium and a cellular region with oogonial syncytium, single germ cells, zygotes, and shelled eggs. Germ cells are embedded in the supporting syncytium. The ultrastructural features of these components and data on fertilization, shell formation, and release from the ovarian ball, alongside insights into the likely egg sorting function of the uterine bell, are provided. We also present light and electron microscopy observations of Saefftigen's pouch and a suggestion regarding its hydrostatic functioning in the eversion of the copulatory bursa.

Abstract:Sperm ultrastructure has been successfully used in gastropod taxonomy and here we attempted to identify potential synapomorphies within the Hygrophila, a group of freshwater lung-bearing gastropods, by examining sperm morphology and ultrastructure in two hygrophilan species of the genus Choanomphalus Gerstfeldt, 1859 (Lymnaeoidea: Planorbidae): Ch. riparius (Westerlund, 1865) and Ch. rossmaessleri (A. Schmidt, 1851). The spermatozoa of both species, like those of other Hygrophila, are divided into a head with an acrosome and nucleus, a midpiece containing a mitochondrial derivative, a glycogen piece, and an endpiece. The acrosome consists of an apical vesicle and a lucent-cored pedestal, plugged basally by compact electron-dense material (acrosomal plug) and separated from the nucleus by a membrane-bound vesicle (basal cap). The midpiece bears apically four surface ridges and contains four glycogen-filled helices inside the mitochondrial derivative. The midpiece is separated from the glycogen piece by an annulus composed of electron-dense material at the tip of the mitochondrial derivative and a hollow cylinder, but the latter appears to consist of two fused structures, making the annulus essentially tripartite. Spermatozoa of the two species show interspecific differences: in Ch. rossmaessleri , the acrosomal plug lacks a stem and the midpiece ridges differ in size, while Ch. riparius has spermatozoa with a stemmed plug and equal-sized ridges. The results of this and earlier studies suggest that sperm ultrastructure can provide synapomorphies for hygrophilan groups at different taxonomic levels. The Lymnaeoidea, one of the two superfamilies of the Hygrophila, appears to be characterised by a lucent-cored acrosomal pedestal, multiple glycogen helices and a tripartite annulus. The presence of a basal plug in the acrosomal pedestal is a possible synapomorphy for the clade uniting Planorbini Rafinesque, 1815 and Segmentinini F.C. Baker, 1945. In Planorbini including the two species of Choanomphalus , the pedestal is separated from the nucleus by a membrane-bound basal cap, a feature readily distinguishing this tribe from Segmentinini and other Lymnaeoidea. The spermatozoa of the two Choanomphalus species differ from those of other Planorbini in having two nuclear keels instead of one, but this character may not be taxonomically reliable.

Abstract:Spermatozoon formation in Caryophyllaeides fennica (Schneider, 1902) is characterised by the following: (1) apical electron-dense material in the zone of differentiation, (2) typical striated roots situated unconventionally in opposite directions in early stages of spermiogenesis, (3) intercentriolar body composed of three electron-dense and two electron-lucent plates, (4) free flagellum and a flagellar bud that correspond to a greatly reduced flagellum and (5) rotation of free flagellum and a flagellar bud to the median cytoplasmic process at 90°. The development of two flagella of significantly unequal length clearly supports a derived form of spermiogenesis in the Caryophyllidea. New for cestodes is a finding of two additional striated roots situated opposite each other, in conjunction with both the flagellar bud and free flagellum. Mutual position of additional striated roots and typical striated roots is parallel in early stages and perpendicular in advanced stages of spermiogenesis. A complete proximodistal fusion gives rise to a mature spermatozoon consisting of one axoneme, parallel cortical microtubules, a nucleus and a moderately electron-dense cytoplasm with glycogen particles, detected by a technique of Thiéry (J Microsc 6:987-1018, 1967), in the principal regions (II, III, IV). Electron tomography analysis of the free flagellum and one axoneme of a mature spermatozoon of C. fennica provides clear evidence, for the first time, that two tubular structures are present in the central axonemal electron-dense core. Phylogenetically important aspects of spermiogenesis of the Caryophyllidea with one axoneme, and other cestodes with one or two axonemes, are briefly reviewed and discussed.

Abstract:The work gave a detailed systematic morphology by optic and scan electron microscopy of two parasites; Raillietina echinobothriida Megnin, 1880 and Spirora meleagaris n. sp. infecting domesticated turkey, with some important description characters. SEM revealed that tegument of R. echinobothriida exhibits, filamentous, microtriches and sensory papillae densely covered the tegument of entire body, rostellum armed with two rows of hummer-shaped hooks and provide by 16-20 rows of small, rose thorn-shaped accessory spines. In addition, a number of taxonomic features in S. meleagaris n. sp. that differ from other species of the genus, mouth circular, bounded by a cuticular three circles plates, five pairs of cephalic papillae, an inner circle of two pairs situated on the wall of the buccal cavity, one pairs of larges sub-median amphids, and an outer circle of two pairs papillae. Buccal cavity supported by four chitinious cusped molar teeth anteriorly directed .Vulva near the end of the first third of the body, vulvular lips prominent. The male has unique rose like shaped pedunculated and unarranged numerous distributed sessile cervical papillae at the second third of the body that are distinguishable from other spirorid.

Abstract:The muscle system and the FMRFamide immunopositive components of the nervous system of flatworms Acrolichanus auriculatus (Digenea, Allocreadiidae), an intestine parasite of acipenserid fishes, were studied for the first time by immunocytochemical, histochemical methods and confocal laser scanning microscopy. Scanning electron microscopy in A. auriculatus revealed a consistent pattern in the number and arrangement of ciliated and unciliated sensory papillae along the digenean body. Morphological data demonstrated the presence of the circular, longitudinal and diagonal muscles in different body regions of A. auriculatus . The musculature of the oral and ventral suckers, the digestive system, as well as the muscles extending radially from the genital pore are described. The study revealed the presence of the FMRFamide immunopositive staining in the central and peripheral nervous systems of A. auriculatus : in the neurons and neurites of the brain, brain commissure, in three pairs of the longitudinal nerve cords and commissures. FMRFamide immunopositive nerve elements innervate the attachment organs and the compartments of the digestive, reproductive and excretory systems.

Molecular and morphological characterization of Dollfustrema bagarii (Digenea: Bucephalidae) metacercariae from aquaculture channel catfish (Ictalurus punctatus) in northern Vietnam

Abstract:White grub metacercariae were found in the livers and kidneys of diseased specimens of an introduced channel catfish, Ictalurus punctatus (Rafinesque), in Vietnam. Based on morphological features and 28S rDNA sequence analysis, the isolated metacercariae were identified as Dollfustrema bagarii (Digenea: Bucephalidae) Moravec & Sey. Histopathological examination shows that encysted metacercariae can change the tissue structure of the infected organs and is often accompanied by haemorrhaging and the presence of eosinophilic granular cell infiltration. Degenerative changes were also observed.

Abstract:Ultrastructure of mature spermatozoon of the Turbo cornutus was observed by using transmission electron microscopy.The mature sperm contains two parts: head and tail.The head comprises a winebottle-shaped acrosome and a pillar-shaped nucleus.The contents in the acrosome is homogeneous,but the electron density is less than nucleus;the subacrosomal space is long-tubule in shape,and it becomes trumpet-shaped at base,which is characterized by filling with granular materials.The nucleus is highly condensed;there exist front nuclear pocket and posterior nuclear pocket.The tail is comprised of mid-piece and end-piece.Five mitochondria surrounding the proximal and distal contrioles make up a mid-piece;The slim and whip-alike end piece consists of an central axoneme,which is a typical '9+2' structure,and a plasmamembrane.A comparison was made between the ultrastrucutre of Turbo cornutus and related gastropods,and an analysis of the phylogenetic signifance was also presented in this paper.

Abstract:Comparing the morphological characteristics of the CNS and PNS of adults and larvae of Renicola parvicaudatus, we identified both continuity in the organization of their neurons and synapses, as well as significant differences in the structure of ganglia, neuron somata and sensory endings. The excretory system of adult worms is characterized by the presence of unique terminal cells with several flame tufts, which are not typical either for the larvae of this species or for other digeneans studied so far. The digenean complex life cycle includes various morphological forms with different locomotory and behavioral activities, and the functional specialization of their nervous system is of importance for the transmission of these parasites. Adult digeneans acquire many adaptive features associated with the final settlement in a vertebrate host. Our study describes the general morphology and ultrastructure of the nervous system of the adult renicolid digenean Renicola parvicaudatus parasitizing the renal tubules of herring gulls. Using immunocytochemical and electron microscopic methods, we identified the distinctive characteristics of ganglia and synapses in the studied species. A comparative analysis of the organization of the nervous system of adult individuals and their continuously‐swimming stylet cercariae revealed a number of stage‐related differences in the composition of ganglia, the distribution of serotonin‐ and FMRFamide‐immunoreactive neurons, the cytomorphology of neuron somata and free sensory endings. Thus, in adults, the presence of FMRFamide‐positive neuron somata, accessory muscle bundles in the ganglionic cortex, and eight types of neuronal vesicles was detected, but no glia‐like elements were identified. Their neurons are characterized by a larger volume of cytoplasm and also show greater ultrastructural diversity. Although the sensory papillae of adults do not vary in their external morphology as much as those of larvae, their sensory bulbs are more diverse in cytomorphology. Following our previous data on the "support" cell processes related to various tissues of the larvae and considered as glia‐like structures, we also briefly present the identified features of the parenchyma, attachment organs and excretory system of adult individuals. The excretory system of adult R. parvicaudatus is characterized by the presence of unique terminal cells with several flame tufts, which are not typical either for the larvae of this species or for other digeneans studied so far. We also used molecular phylogenetic analysis to clarify species identification.

Abstract:Summary Sperm development and structure in the wood-inhabiting fungal and plant-feeding nematode, Bursaphelenchus luxuriosae , were studied using transmission electron microscopy to evaluate interspecific similarities and differences of spermatozoa in nematodes. In general, spermatogenesis in B. luxuriosae fits the ‘rhabditid’ pattern supported by morphological and phylogenetic analysis of the order Rhabditida. Spermatocyte development includes formation of complexes of fibrous bodies (FBs) with membranous organelles (MOs), the complexes dissociate in the spermatids into separate components, and the immature sperm contain MOs but lack FBs, which transform into a dense matrix of sperm cytoplasm. The female spermatheca contains mature spermatozoa as bipolar cells subdivided into a pseudopod devoid of organelles and a main cell body containing a nucleus without a nuclear envelope, numerous mitochondria, and peripheral MOs as pouches opening to the exterior via pores. Data on B. luxuriosae are used for analysis of variable quantitative and morphological characteristics of spermatozoa in Aphelenchoidea. General size of spermatozoa and their MOs have little value for comparative analysis. The MO knobbles look uniform in immature spermatozoa of each aphelenchoidid species studied and may be considered as a taxonomically specific ultrastructural feature. The presence or absence of FBs in immature spermatozoa demonstrates the diagnostic value of the aphelenchoidid sperm structure at the species level. Analysis also shows close similarity of spermatozoa in Bursaphelenchus spp. and Caenorhabditis elegans , the model species with comprehensive data on sperm biology. Abstract:Presenting new molecular and scanning electron microscope (SEM) features, this study gives additional data to the better knowledge of Thaparocleidus vistulensis (Siwak, 1932) (Monopisthocotyla, Ancylodiscoididae), a parasite of the European catfish Silurus glanis Linnaeus, 1758 (Siluriformes, Siluridae) cultured in a commercial fish farm in Hungary. In addition, notes on the early development of sclerotized anchors are also provided. The main morphological difference of T. vistulensis compared to other congeneric species is associated with the male copulatory organ, which exhibits 5–7 loops in the middle of the penis length and a long open V-shaped sclerotized accessory piece, dividing terminally into two parts, securing the terminal part of the penis tube. The present study provides for the first time molecular characterization data based on the 2694 bp long nucleotide sequence of rDNA (ITS1, 5.8S, ITS2, and flanked with partial 18S and partial 28S) submitted in GenBank with the accession number OR916383. A phylogenetic tree based on ITS1 sequences supports a well-defined clade including T. vistulensis , forming a sister group with T. siluri, a species-specific monopisthocotylan parasite to S. glanis . The morphological characterization of T. vistulensis, especially for the male copulatory organ, together with the molecular data in the present study, extends knowledge about this monopisthocotylan species and provides new information for future phylogeny studies.

Abstract:The ultrastructure of mature spermatozoa of the giant clam bivalve Scapharca broughtoni was investigated by transmission electron microscopy for the first time. The mature spermatozoon consists of a head which is composed of a cone-shaped acrosome, a round nucleus, and a tail region. A subacrosomal space contains an axial rod and a basal plate, the latter lying between the acrosome and the nucleus. Although the nucleus lacks an anterior invagination, an inverted shallow V-shaped posterior invagination is present within the nucleus. Within the middle portion of the spermatozoon lie five spherical mitochondria while the long whip-like end portion is composed of an axoneme with the typical 9 + 2 structure. Our conclusion is that the spermatozoon of S. broughtoni is of the type I anacrosomal "aquasperm", and the morphology of acrosome and nucleus are an adaptation to external fertilization. The morphology of the male reproductive system was studied in Cherax quadricarinatus . The testes and vasa deferentia were dissected, fixed, cut and stained. Testes appear as two parallel and opalescent strands; they present many testicular lobes, each lobe containing cells in the same stage of the spermatogenic cycle. A vas deferens arises from the external side of each testis and three parts were clearly distinguished: proximal vas deferens (PVD), middle vas deferens (MVD) and distal vas deferens (DVD). The PVD is opalescent and highly convoluted, the MVD is pale white in colour and convoluted, but wider in diameter than the PVD, while the DVD shows the widest diameter, is straight and is white in colour. A single-layered epithelium is recognized in the vas deferens; with cylindrical cells in the PVD and cuboid cells in the MVD and DVD. The formation of the spermatophore starts at the PVD, while the secondary layer of the spermatophore seems to be added at the MVD. At the DVD, the highly coiled spermatophore is surrounded by the periodic acid Schiff-positive sticky components of the secondary layer. Many aspects of spermatophore formation in C. quadricarinatus differ from those of other Astacida. The applied aspects of this study for aquaculture purposes are discussed.

Rab2 and Rab6 are Implicated in Acrosome Formation during Spermatogenesis in Eriocheir sinensis: Based on Sperm Proteome

Abstract:BACKGROUND: Rab proteins are GTP-dependent small proteins that function as regulators of intracellular vesicle transport, fusion, and localization. However, few studies have investigated their function in Decapoda reproduction. The Eriocheir sinensis sperm has no tail and the nuclei are uncondensed. With the acrosome forming the majority of the sperm mass, it provides an ideal model for studying acrosome formation.METHODS: We firstly analyzed the sperm proteome using LC-MS/MS. To study the functions of Rab2 and Rab6, related to the Golgi apparatus, in the acrosome formation during spermatogenesis, the genes of Rab2 and Rab6 were cloned based on the testis transcriptome of E.sinensis and poly-clonal antibodies were prepared. The presence of 2 Rab proteins was confirmed in the testis and sperm by western blot. We further observed the characteristics of target 2 Rab proteins using immunofluorescence (IF).RESULTS: A total of 1247 proteins including 7 Rab proteins, Rab1, Rab2, Rab5, Rab6, Rab11, Rab14, and Rab18 were identified in the sperm proteome. The IF results showed that Rab2 co-localizes with GM130, a cis-Golgi matrix protein, in the spermatagonia and spermatocytes. In the early spermatids, Rab2 and Rab6 participate in the formation of pre-acrosomal vesicles. In maturing spermatids, both Rab2 and Rab6 settle on the acrosomal membrane but present different characteristics wrapping the pre-acrosome. In the mature sperm, Rab2 localizes in the perinuclear theca surrounding the nuclei cup, while Rab6 remains on the acrosomal membrane.CONCLUSIONS: Our research found 7 Rab proteins based on the analysis of the sperm proteome in E.sinensis, and confirmed the involvement of Rab2 and Rab6 in acrosome formation. These findings provide a foundation for studying the functions of Rab proteins during spermatogenesis in Decapoda animals.
