Abstract:Intensive use of organophosphate and carbamate insecticides has led to the development (of resistance) in many target species that are important in agriculture. (Acetylcholinesterase(AChE))(insensitive) to organophosphate and carbamate insecticides has been identified as a major resistance mechanism in numerous insect species. Acetylcholinesterase gene mutations conferring insecticide (resistance) in insect pests are summarized briefly. Furthermore, the effects of acetylcholinesterase gene mutations on (acetylcholinesterase) protein structures and functions, the characterization of (modified) (acetylcholinesterase) protein, the effects of acetylcholinesterase gene mutations on fitness, and the effects of the combination of various mutations on insecticide resistance were discussed. These gene mutations responsible for conferring insensitivity may open up the way for designing new (anti-)resistant compounds.

Abstract:Acetylcholinesterase (AChE; EC 3.1.1.7) is a primary target of many insecticides including organophosphates (OP) and carbamates (CB). Because AChE is expressed in all invertebrate and vertebrate animals as a key enzyme of the cholinergic system, the toxicity of anticholinesterase insecticides to mammals and non-target species such as beneficial insects has been a great concern. In addition, the intensive use of OP and CB insecticides has resulted in the development of resistance in many insect pests, which has limited the use of anticholinesterase insecticides. Many aces encoding AChEs have been sequenced from a variety of vertebrates, insects and other invertebrates, and crystal structures of four AChEs have been determined in the past 20 years. Although the primary motifs and the three dimensional (3D) structures of different AChEs are similar, differences among AChEs are obvious. The catalytic properties and inhibition kinetics of AChEs from different groups of insects and mammals may be quite different, and two AChEs from a single insect may also show distinct differences. These differences may provide new opportunities for designing more selective insecticides for pest management.

Abstract:Acetylcholinesterase (AChE) is the primary target of organophosphate (OP) and carbamate (CB) insecticides. Many insect species have been shown to have two different AChE genes. The amino acid identity between the two lepidopteran AChEs is lower than 40%, and potential differences in enzymatic function have not been characterized. In this study, the cDNAs encoding two AChEs (Boma-AChE1 and Boma-AChE2) from Bombyx mandarina were sequenced, and the corresponding proteins were heterologously expressed to compare their enzymatic properties and interactions with insecticides in vitro. Both of these enzymes had high specific activities for acetylthiocholine iodide. Studies on substrate and inhibitor specificities confirmed that both enzymes belong to AChE. Insecticide inhibition assays indicated that Boma-AChE1 was more sensitive than Boma-AChE2 to eight of the nine insecticides tested. However, Boma-AChE2 was more sensitive than Boma-AChE1 to one of the OP insecticides, heptenophos. The results suggested that two AChEs from a lepidopteran insect have distinct catalytic properties and responses to different inhibitors.

Abstract:Acetylcholinesterase (AChE), which is encoded by the ace gene, catalyzes the hydrolysis of the neurotransmitter acetylcholine to terminate nerve impulses at the postsynaptic membrane. AChE is a primary target of many insecticides including organophosphates (OP) and carbamates (CB). In this study, full-length cDNA sequences of two ace genes (Nlace1 and Nlace2) were sequenced from the brown planthopper (BPH) Nilaparvata lugens, the most destructive insect pest of rice crops. Nlace1 cDNA is 2842 nucleotides long and contains an ORF potentially encoding a 790 amino acid peptide. Nlace2 cDNA is 2852bp in length and contains an ORF that potentially encodes a 672 amino acid peptide. NlAChE1 has an identity of 40% with NlAChE2 at the amino acid sequence level. Phylogenetic analysis of 59 AChEs from 32 animal species showed that NlAChE1 is most closely related to AChE1s from Blattella germanica and Nephotettix cincticeps, while NlAChE2 is most closely related to AChE2 from N. cincticeps. Quantitative RT-PCR analysis showed that Nlace1 is expressed at a much higher level than Nlace2 in all developmental stages and tissues, demonstrating that NlAChE1 may be the dominant AChE form of the two enzymes. This result will help reveal the resistance mechanism of N. lugens to organophosphorous and carbamate insecticides and promote development of more selective insecticides targeting the main NlAChE1.

Abstract:Recent advances in the studies of acetylcholinesterase (AChE, EC 3.1.1.7) genes from insects were reviewed. The gene structure, including ten extrons of insect AChE gene (ace), was discussed. All the ace genes reported from insects were summed up, and the phylogenetic tree was constructed based on the reported complete sequences of ace genes from insects. The ace structure characteristics, the functional sites of ace genes, the effect of ace mutation(s) on insecticide resistance and their relationships were also discussed. Finally, the problems and prospects in the studies of insect ace were analyzed.
Identification and characterization of ace1-type acetylcholinesterase in insecticide-resistant and -susceptible Propylaea japonica (Thunberg)

Abstract:Characterization and gene cloning of acetylecholinesterase (AChE) in the insecticide-resistant (R) and -susceptible (S) insects have been reported in the past. However, the studies focused mostly on herbivorous pests, rather than predacious species, such as ladybird beetles. Using R and S Propylaea japonica (thunberg), a full-length cDNA sequence (2928 bp) of the ace1-type AChE gene was determined for the first time. The ace1 encoding a protein of 645 amino acids contained typical conserved motifs, such as FGESAG domains, catalytic triad, acyl pocket, oxyanino hole, choline binding site, peripheral anionic site, omega loop and conserved aromatic residues. R P. japonica displayed 50-times greater resistance to chlorpyrifos or mathamidophos with a significantly lower AChE sensitivity to paraoxon, malaoxon, chlorpyrifos or methamidophos than its S counterpart. Five amino acids in the ace1 of R P. japonica differed from those found in S P. japonica. One of them, F358S, located in the acyl-binding pocket, might play a crucial role in the resistance of the insect to organophosphates (OPs). Whereas, K493E and I538V, which were close to some of the conserved aromatic amino acids (i.e., H509, Y511, and W499) in the gorge, and G571R and T576A near C593 that formed the disulfide bonds with C471, might also involve in the change of insecticide resistance in P. japonica. AChE insensitivity and amino acid replacements, particularly F358S, might be the determining factors in the alteration of OPs-resistance in P. japonica.

Abstract:Athetis lepigone is one of the most severe polyphagous pests, and it has developed resistance to different chemical insecticides. Insects primarily rely on the olfactory system to recognize various environmental chemicals, including xenobiotics such as insecticides. Here, we expressed two A. lepigone pheromone-binding proteins (AlepPBP2 and AlepPBP3), and observed they had higher binding affinities to phoxim than other insecticides, with Ki was 3.30 ± 0.38 μM and 3.27 ± 0.10 μM, respectively. Molecular dynamics simulation, binding mode analysis, and computational alanine scanning showed that six residues (Phe15, Phe39, Ile55, Leu65, Ile97, and Phe122) of AlepPBP2 and three residues (Phe12, Ile52, and Ile134) of AlepPBP3 maybe as potential residues that can change protein ability to bind an organophosphorus insecticide phoxim. Then, we used site-directed mutagenesis assay to mutate these residues into alanine, respectively. Subsequently, the binding assays displayed that Phe15, Phe39, and Ile97 of AlepPBP2, Phe12 and Ile134 of AlepPBP3 caused a significant decrease of AlepPBPs binding ability to phoxim, suggesting they should play crucial roles in the AlepPBPs/phoxim interactions. Our findings could further advance in using PBPs as unique targets to design and develop precise and environmentally-friendly pest control agents with high insecticidal potential using a computer-aided drug design (CADD) approach.

Abstract:We develop an information-theoretic analysis of dependencies between image wavelet coefficients. The dependencies are measured using mutual information, which has a direct link with data compression and estimation performance. Mutual information can be computed analytically for the special case where the image is a stationary autoregressive (AR)-1 Gaussian process. We have also developed methods to compute mutual information experimentally from wavelet domain image data. Our mutual-information analysis provides a mechanism for model evaluation and comparison.

Abstract:Two acetylcholinesterase (ace) genes have been reported in many insect species. In pests such as Helicoverpa assulta and Plutella xylostellas, ace1 gene encodes the predominant synaptic enzyme that is the main target of organophosphorus (OP) and carbamate pesticides. It has been reported that pesticide selection has an impact on the ace gene evolution. The domesticated silkworm, Bombyx mori, also has two ace genes. We studied ace gene expression and enzyme activities in silkworm as this has not faced pesticide selection over the past decades. The expression levels of two ace genes, Bm-ace1 and Bm-ace2, were estimated by quantitative real-time polymerase chain reaction. Bm-ace2 was expressed more highly than Bm-ace1 in all tested samples of different developmental stages or tissues, suggesting ace2, rather than ace1, is the major type of acetylcholinesterase (AChE) in Bombyx mori. This is inconsistent with the aforementioned lepidopterons agricultural pests, partly be due to the widespread use of pesticides that may induce high expression of the ace1 gene in these pests. Besides high expression in the head, Bm-ace1 also expresses highly in the silk glands and Bm-ace2 is abundant in the germline, implying both ace genes may have potential non-hydrolytic roles in development. Furthermore, we found that the mRNA levels of two ace genes and their ratios (ace2/ace1) change day to day in the first and third instars. This challenges the conventional method of estimating enzymatic activity using crude extract as an enzyme solution, as it is a mixture of AChE1 and AChE2. An efficient and simple method for separating different AChEs is necessary for reliable toxicological analyses.

Abstract:Acetylcholinesterase (AChE, EC 3.1.1.7) is an important enzyme with a typical function of degrading the neurotransmitter acetylcholine. Although two ace genes were reported in Plutella xylostella, their function differences remain largely unknown. The chemically synthesized siRNAs (si‐Pxace1 and si‐Pxace2) were injected into the second instar larvae to knock down Pxace1 and Pxace2, either respectively or simultaneously. The mRNA abundance of Pxace1 and Pxace2 was significantly reduced to 7–33.5% of the control levels at 72 h after siRNA injection. The AChE activities were significantly decreased at 96 h after treatment. Silencing of Pxace1 or Pxace2 resulted in mortality of 33.9 and 22.9%, respectively. The survivors in siRNA‐treated groups had apparent growth inhibition such as reduction in larvae weights and lengths, malformation and motor retardation. Knockdown of Pxace1 apparently affected more on larvae growth than that of Pxace2, suggesting that Pxace1 had more important roles than Pxace2. Both Pxace1 and Pxace2 genes might have atypical functions in regulating larvae growth and motor ability.
Transcriptomics and proteomic-based study of the acaricidal mechanism of paeonol against Aleuroglyphus ovatus

Abstract:Aleuroglyphus ovatus, a common mite harming stored grain, poses a threat to human health. Paeonol is the main component isolated from peony roots and exhibits various medicinal values. In our previous study, we found that paeonol shows strong acaricidal activity. In the present study, the acaricidal mechanism of paeonol against A. ovatus was uncovered through RNA-seq transcriptomics and TMT proteomics. According to the transcriptomics results, paeonol treatment noticeably up-/downregulated genes associated with “oxidoreductase activity” (peptidase activity, ATPase activity, and hydrolase activity). As shown by proteomic analysis, concordant changes were identified in proteins linked with hydrolase activity, serine-type peptidase activity, antioxidant activity, and oxidoreductase activity. Paeonol target proteins were FMO, ACAD8, GPX, CTSL, and PRSS1, as determined by transcriptomics-proteomics correlation analysis. We propose that paeonol may play an acaricidal role by interfering with enzyme activities in A. ovatus, especially serine-type peptidase, thereby causing an imbalance in amino acid ratios and eventually leading to the death of A. ovatus. However, the specific mechanism of action needs to be further studied. In addition, we verified the key enzymes at the transcriptional level.

Abstract:Glyphodes pyloalis Walker (Lepidoptera: Pyralididae) is a common pest in sericulture and has developed resistance to different insecticides. However, the mechanisms involved in insecticide resistance of G. pyloalis are poorly understood. Here, we present the first whole-transcriptome analysis of differential expression genes in insecticide-resistant and susceptible G. pyloalis. Clustering and enrichment analysis of DEGs revealed several biological pathways and enriched Gene Ontology terms were related to detoxification or insecticide resistance. Genes involved in insecticide metabolic processes, including cytochrome P450, glutathione S-transferases and carboxylesterase, were identified in the larval midgut of G. pyloalis. Among them, CYP324A19, CYP304F17, CYP6AW1, CYP6AB10, GSTs5, and AChE-like were significantly increased after propoxur treatment, while CYP324A19, CCE001c, and AChE-like were significantly induced by phoxim, suggesting that these genes were involved in insecticide metabolism. Furthermore, the sequence variation analysis identified 21 single nucleotide polymorphisms within CYP9A20, CYP6AB47, and CYP6AW1. Our findings reveal many candidate genes related to insecticide resistance of G. pyloalis. These results provide novel insights into insecticide resistance and facilitate the development of insecticides with greater specificity to G. pyloalis.

Abstract:Acetylcholinesterase (AChE, EC 3.1.1.7), which is encoded by the ace gene, is a key enzyme in both cholinergic system and other non-neuronal tissues. AChE is the target of organophosphates (OP) and carbamates (CA) insecticides. Although AChEs have been extensively studied in lepidopteran pests, no research has been carried out on butterflies, a major subgroup in Lepidoptera. In this study, we cloned two ace genes (Mcace1 and Mcace2) from the Glanville fritillary butterfly (Melitaea cinxia, Lepidoptera: Nymphalidae). The full-length cDNA of Mcacel was 2433bp, containing a 2073bp ORF encoding 690 amino acids, a 227bp 5'-untranslated region (5'-UTR), and a 133bp 3'-UTR with a poly(A) tail. The cDNA of Mcace2 was 2400bp in length, containing a 1911bp ORF encoding 636 amino acids, a 368bp 5'-UTR, and a 121bp 3'-UTR with a poly(A) tail. Both the deduced proteins, McAChE1 and McAChE2, possess typical features of AChE family, such as catalytic triad, acyl pocket and choline-binding site. Phylogenetic analysis indicated that McAChE1 and McAChE2 exhibited the highest identity to the AChE1 and AChE2 of moths, respectively. We compared the amino acids at the sites where substitution may result in insecticide resistance between McACHEs and the earlier reported AChEs, and found that no amino acid substitution related to insecticide resistance occurs in either McAChE1 or McAChE2. These implied that both McAChE1 and McAChE2 may be sensitive to OP and CA insecticides. The result is compatible with our expectation as no insecticides were used in the habitat of the Glanville fritillary.

Abstract:Two acetylcholinesterase genes, Ace1 and Ace2, have been fully cloned and sequenced from both organophosphate-resistant and susceptible clones of cotton aphid. Comparison of both nucleic acid and deduced amino acid sequences revealed considerable nucleotide polymorphisms. Further study found that two mutations occurred consistently in all resistant aphids. The mutation F139L in Ace2 corresponding to F115S in Drosophila acetylcholinesterase might reduce the enzyme sensitivity and result in insecticide resistance. The other mutation A302S in Ace1 abutting the conserved catalytic triad might affect the activity and insecticide sensitivity of the enzyme. Phylogenetic analysis showed that insect acetylcholinesterases fall into two subgroups, of which Ace1 is the paralogous gene whereas Ace2 is the orthologous gene of Drosophila AChE. Both subgroups contain resistance-associated AChE genes. To avoid confusion in the future work, a nomenclature of insect AChE is also suggested in the paper. (C) 2004 Elsevier Ltd. Abstract:This report presents a case of nodular colloid degeneration in a 76-year-old man with a solitary facial lesion. The skin lesion was a yellowish indurated plaque on the left mandibular area. Histopathologic examination revealed the amorphous colloid material in the upper dermis. The Congo red stain was negative, no birefringence was observed under polarized light, and elastic stain showed positivity in both the colloid and the surrounding fibers. To the best of our knowledge, nodular colloid degeneration is a rare entity and has only been reported in published literature a few times in the last 8 decades.
Female rats exhibit greater susceptibility to early alcohol-induced liver injury than males.

Abstract:It is known that women develop hepatic injury more rapidly and with exposure to less ethanol than men; however, mechanisms remain unclear. The purpose of this study was to determine if an enteral alcohol delivery model could be used to study susceptibility of females to alcohol-induced liver injury. Male and female Wistar rats (age- or weight-matched) were given ethanol (11-12 g.kg-1.day-1) continuously for up to 4 wk via intragastric feeding, and control rats received a high-fat diet without ethanol. There were no significant differences in body weight among the groups studied. Furthermore, mean ethanol concentrations, their cyclic pattern in urine, and rates of ethanol elimination were also not different between the genders under these conditions. Ethanol treatment elevated serum aspartate aminotransferase levels in male rats to 126 +/- 10 IU/l after 4 wk. In females, however, values increased more rapidly and reached significantly higher values at 4 wk (168 +/- 18 IU/l). Steatosis, inflammation, and necrosis assessed histologically also developed more rapidly and were more severe in females than males. Steatosis due to ethanol exposure, which was localized in centrilobular areas in males, was panlobular in the female. Moreover, endotoxin in plasma, intercellular adhesion molecule 1 expression in hepatic sinusoidal-lining cells, and the number of infiltrating inflammatory cells in the liver were 2-2.5-fold greater in females than males. These changes possibly account for increased hepatic injury due to ethanol in the female.
Two Bombyx mori acetylcholinesterase genes influence motor control and development in different ways

Abstract:Among its other biological roles, acetylcholinesterase (AChE, EC 3.1.1.7), encoded by two ace in most insects, catalyses the breakdown of acetylcholine, thereby terminating synaptic transmission. ace1 encodes the synaptic enzyme and ace2 has other essential actions in many insect species, such as Chilo suppressalis and Plutella xylostella. The silkworm, Bombyx mori, has been domesticated for more than two thousand years and its aces have no history of pesticide exposure. Here, we investigated the functional differences between two ace genes, BmAce1 and BmAce2, in the silkworm. qPCR analysis indicated that BmAce1 is highly expressed in muscle and BmAce2 is more ubiquitously expressed among tissues and enriched in the head. Both genes were separately suppressed using chemically synthesized siRNAs. The mRNA abundance of the two ace genes was significantly reduced to about 13% - 75% of the control levels after siRNA injection. The AChE activities were decreased to 32% to 85% of control levels. Silencing BmAce2 resulted in about 26% mortality, faster and higher than the 20% in the siBmAce1-treated group. Silencing BmAce1 impacted motor control and development to a greater extent than silencing BmAce2, although both treatment groups suffered motor disability, slowed development and reduced cocoons. Both genes have essential, differing biological significance.

Abstract:The molecular mechanism was reviewed in terms of AChE insensitivity to carbamate and organophosphate insecticides in cotton aphid(Aphis gossypii Glover).Cotton aphid is the first insect to be found with two different AChE genes by RT-PCR approach,providing the direct molecular evidence for the hypothesis that some insects may have another AChE gene besides the one orthologous to the AChE gene found in Drosophila melanogaster.The two genes,sharing only 48% similarity at the amino acid level in cotton aphid,were distant by phylogenetic analysis,and were speculated to be diverged long before the differentiation of insects.Some point mutations were found to correlate with the insecticide resistance in both AChE-1 and AChE-2.Further study with the expressed recombined AChE-1 enzymes with S431F and A302S mutations verified that these two mutations conferred the enzyme insensitivity to pirimicarb and most of other insecticides tested,but with exceptions of increased enzyme sensitivity to some insecticides.Also,the biochemical analysis with the in vitro expressed AChE-1 and AChE-2 indicated that AChE-1,rather than AChE-2,seemed to take the main role in the cholinergic synapses and subsequently to be the target for carbamate and organophosphate insecticides.Some related questions that need to be further clarified were discussed.
Extra-Axial Ependymoma Presenting as a Cerebellopontine Angle Mass.

Abstract:Ependymomas are glial tumors most commonly located within the posterior fossa (60%Y70% in both adults and children), classically originating from the ependymal cells of the fourth ventricle (1,2). As ependymomas may exist anywhere along the nueroaxis, a minority of ependymomas have been documented in supratentorial and spinal cord sites (1,2). Isolated extra-axial ependymomas at the cerebellopontine angle (CPA) are rare, particularly when no fourth ventricle involvement is identified. It has been suggested that deposition of heterotopic ependymal rests in surrounding tissue during fetal development could give rise to extra-axial ependymomas (1). Ependymomas may be cystic or demonstrate areas of necrosis and hemorrhage (3). On histologic examination, pleomorphic cells with rare mitoses form perivascular pseudorosettes and occasional ependymal canals (3). Vestibular schwannomas and meningiomas combine for 80% to 95% of CPA lesions, with epidermoid cysts being the next most common (4). Only rarely do ependymomas present as primary CPA lesions and, for this reason, these tumors are often initially misdiagnosed. A 48-year-old man presented with a 2-month history of worsening headaches, lightheadedness, progressive ataxia, nausea, and vomiting. The patient was found to have mild ipsilateral sensorineural hearing loss on the audiogram despite subjective perception of normal hearing. A noncontrast head computed tomography showed a subtle mass effect on the right lateral margin of the fourth ventricle by a noncalcified extra-axial mass centered near the CPA (Fig. 1). Axial T2-weighted magnetic resonance imaging at the level of the foramen of Luschka showed a cerebrospinal fluid cleft separating the extra-axial mass from the cerebellum and the brainstem. The lesion did not extend through the foramen of Luschka or the porus acusticus and was homogeneously hypointense to brain on T1 and hyperintense on T2. After the administration of contrast, the mass showed mild to moderate heterogeneous enhancement (Fig. 2). The imaging appearance of this lesion is inconsistent with that of the more common CPA lesions, meningioma and schwannoma, as well with the less frequently encountered choroid plexus papilloma. Schwannomas tend to be isointense to the brain on precontrast T1, have variable intensity on T2 based on cystic degeneration of the tumor, and enhance intensely with contrast (4). They also usually have a concentric growth around the component extending into the internal auditory canal (IAC) (4). Abstract:Helicoverpa armigera is a major devastating insect pest on a wide range of vegetables and cash crops. Insecticides are presently indispensable for its control in nearly all crops. H. armigera has acquired resistance against almost all insecticides because of the activity of detoxification enzymes used for the defensive mechanism. The current research was carried out to evaluate the activity of detoxification enzymes, i.e., acetylcholinesterase and alkaline and acid phosphatases in chlorpyrifos-, bifenthrin-, lufenuron-, lambda cyhalothrin-, and emamectin benzoate-treated larvae of H. armigera. The maximum AChE activity was recorded in emamectin benzoate-treated larval samples followed by chlorpyrifos, lufenuron, lambda cyhalothrin and bifenthrin, respectively, while the highest alkaline phosphatases’ activity was recorded in emamectin and the lowest in bifenthrin-treated H. armigera. As far as acid phosphatases’ activity is concerned, the highest activity was noted in lufenuron samples while the lowest in lambda cyhalothrin samples, respectively. Comparatively, activities of alkaline and acid phosphatases were higher than AChE. The elevated activities of detoxification enzymes can possibly lead to increase in resistance development against synthetic chemical insecticides.
