Abstract:Extracellular matrix (ECM) derived from decellularized tissues and organs has been used as a biological scaffold in a variety of pre-clinical and clinical applications. However, the lack of mechanical properties and shape controllability is a drawback. In contrast, synthetic polymers can be easily engineered with good mechanical performance, but they have limited biological functionality. In this study, we explored a new method to electrospun cartilage-derived extracellular matrix (cECM) and polycaprolactone (PCL) composite nanofibrous membranes. The cartilage is a compact tissue for which electrospinning is difficult. To overcome this problem, the cartilage was sliced into pieces, milled into powders, and digested into a looser structure. The cECM/PCL (mass ratio 50:50) hybrid nanofibers appeared to be smooth, thinner, and uniform with enhanced mechanical properties and wettability compared to the electrospun PCL. Meanwhile, the presence of cECM in the cECM/PCL nanofibrous membranes significantly promoted chondrocyte proliferation in vitro and facilitated cartilage regeneration in vivo. All these results indicate that the cECM/PCL nanofibrous membranes fabricated with good mechanical properties and biocompatibility may represent a promising scaffold for cartilage regeneration. Moreover, this work may offer a convenient and cost-effective method to synthesize ECM-based hybrid nanofibrous scaffolds for other tissue applications.
Enhanced Cellular Function of Human Vascular Endothelial Cell on Poly (Ε-Caprolactone)/gelatin Coaxial-Electrospun Scaffold

Abstract:An extracellular matrix-like poly (ε-caprolactone) (PCL)/gelatin core-shell nanofibrous scaffold with high hydrophilicity was developed by coaxial-elecrospinning. However, the behavior of vascular endothelial cells (ECs) on the modified scaffold remains limited. In this study, human umbilical vein ECs (HUVECs) were seeded onto PCL scaffolds with or without gelatin. Morphological changes of HUVECs were observed under confocal laser scanning microscopy (LSCM). HUVECs’ adhesion, proliferation and apoptosis were detected by MTT assay and flow cytometry (FCM). Our results showed that HUVECs on PCL/gelatin scaffolds with identical polygonal and cobblestone-like characteristics reached confluence after 7 days. Modification of PCL nanofibers significantly promoted the attachment of HUVECs onto scaffolds within 1 hour. Compared to pristine PCL, a two-fold increase in proliferation of HUVECs was also observed after 7 days, whereas the apoptosis of HUVECs was obviously reduced by 40% on the modified scaffolds. In summary, these results indicated modified PCL/gelatin scaffold developed by coaxial-elecrospinning can increase the adhesion, proliferation, and suppress apoptosis of HUVECs, suggesting it has a great potential and promising vascular graft in tissue engineering applications.
Electrospun Composite Poly(L-Lactic Acid)/Tricalcium Phosphate Scaffolds Induce Proliferation and Osteogenic Differentiation of Human Adipose-Derived Stem Cells

Abstract:Development of tissue-engineered bone constructs has recently focused on the use of electrospun composite scaffolds seeded with stem cells from various source tissues. In this study, we fabricated electrospun composite scaffolds consisting of beta-tricalcium phosphate (TCP) crystals and poly(L-lactic acid) (PLA) at varying loading levels of TCP (0, 5, 10, 20 wt%) and assessed the composite scaffolds' material properties and ability to induce proliferation and osteogenic differentiation of human adipose-derived stem cells (hASCs) in the presence of osteogenic differentiating medium. The electrospun scaffolds all exhibited a nonwoven structure with an interconnected porous network. With the addition of TCP, the fiber diameter increased with each treatment ranging from 503.39 +/- 20.31 nm for 0 wt% TCP to 1267.36 +/- 59.03 nm for 20 wt% TCP. Tensile properties of the composite scaffolds were assessed and the overall tensile strength of the neat scaffold (0 wt% TCP) was 847 +/- 89.43 kPA; the addition of TCP significantly decreased this value to an average of 350.83 +/- 38.57 kPa. As the electrospun composite scaffolds degraded in vitro, TCP was released into the medium with the largest release occurring within the first 6 days. Human ASCs were able to adhere, proliferate and osteogenically differentiate on all scaffold combinations. DNA content increased in a temporal manner for each scaffold over 18 days in culture although for the day 12 timepoint, the 10 wt% TCP scaffold induced the greatest hASC proliferation. Endogenous alkaline phosphatase activity was enhanced on the composite PLA/TCP scaffolds compared to the PLA control particularly by day 18. It was noted that at the highest TCP loading levels of 10 and 20 wt%, there was a dramatic increase in the amount of cell-mediated mineralization compared to the 5 wt% TCP and the neat PLA scaffold. This work suggests that local environment cues provided by the biochemical nature of the scaffold can accelerate the overall osteogenic differentiation of hASCs and encourage rapid ossification.

Abstract:We have detected and partially characterized factors that promote the directed migration of mouse epidermal cells in a modified Boyden chamber assay. Smooth muscle cells grown in culture were found to secrete a potent chemoattractant for epidermal cells. This activity was further characterized and compared to the chemotactic activities found in wound fluid and conditioned medium from 3T3 L1 cells and with interleukin 1. The migration of epidermal cells during wound healing in vivo might be regulated by such factors.
Optimization of Consecutive Signalized Intersections Based on Combined Algorithms: Comparing Results with Microsimulation

Abstract:The primary objective of this research is to optimize signal timing in consecutive signalized intersections. In this paper, the combination of genetic programming (GP) with genetic algorithms (GA) and neural network (NN) with genetic algorithm (GA) were used and compared in order to optimize signal timing in consecutive signalized intersections. First, genetic programming and neural network were constructed from existing signal timing data to predict the delay of intersections. Then genetic algorithm was applied to optimize these predictive networks (GP and NN). The results and comparisons of timing process and error percentage showed that neural network is more efficient than genetic programming. However, the ability of genetic programming in producing formula is a specific characteristic which makes it more applicable than neural network. Finally, for validating the results, Aimsun and Synchro micro simulation software were used, and accuracy of our models was approved.

Abstract:Tissue engineering scaffolds as three-dimensional substrates may serve as ideal templates for tissue regeneration by simulating the structure of the extracellular matrix (ECM). Many biodegradable synthetic polymers, either hydrophobic, like Poly-ε-caprolactone (PCL), or hydrophilic, like Poly(Vinyl Alcohol) (PVA), are widely used as candidate bioactive materials for fabricating tissue engineering scaffolds. However, a combination of good cytocompatibility of hydrophilic polymers with good biomechanical performance of hydrophobic polymers could be beneficial for the in vivo performance of the scaffolds. In this study, we aimed to fabricate biodegradable fibrous scaffolds by combining the properties of hydrophobic PCL with those of hydrophilic PVA and evaluate their properties in comparison with pristine PCL scaffolds. Therefore, single-layered PCL scaffolds, sequential tri-layered (PVA/PCL/PVA), and core-shell (PVA as shell and PCL as core) composite scaffolds were developed utilizing the electrospinning technique. The material structural and biomechanical properties of the electrospun scaffolds, before and after their hydrolytic degradation over a seven-month period following storage in phosphate-buffered saline (PBS) at 37 °C, were comprehensively compared. In addition, human embryonic kidney cells (HEK-293) were cultured on the scaffolds to investigate potential cell attachment, infiltration, and proliferation. The results demonstrated the long-term efficacy of core-shell biodegradable fibrous scaffolds in comparison to single-layers PCL and tri-layers PVA/PCL/PVA, not only due to its superior morphological characteristics and mechanical properties, but also due to its ability to promote homogeneous cell distribution and proliferation, without any external chemical or physical stimuli.

Abstract:Background: Enhancing the efficiency of cell-based skin tissue engineering (TE) approaches is possible via designing electrospun scaffolds possessing natural materials like amniotic membrane (AM) with wound healing characteristics. Concentrating on this aim, we fabricated innovative polycaprolactone (PCL)/AM scaffolds through the electrospinning process. Methods: The manufactured structures were characterized by employing scanning electron microscope (SEM), attenuated total reflection-Fourier transform infrared (ATR-FTIR) spectroscopy, tensile testing, Bradford protein assay, etc. In addition, the mechanical properties of scaffolds were simulated by the multiscale modeling method. Results: As a result of conducting various tests, it was concluded that the uniformity and distribution of fibers decreased with an increase in the amniotic content. Furthermore, PCL-AM scaffolds contained amniotic and PCL characteristic bands. In the case of protein release, greater content of AM led to the release of higher amounts of collagen. Tensile testing revealed that scaffolds' ultimate strength increased when the AM content augmented. The multiscale modeling demonstrated that the scaffold had elastoplastic behavior. In order to assess cellular attachment, viability, and differentiation, human adipose-derived stem cells (ASCs) were seeded on the scaffolds. In this regard, SEM and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assays showed significant cellular proliferation and viability on the proposed scaffolds, and these analyses illustrated that higher cell survival and adhesion could be achieved when scaffolds possessed a larger amount of AM. After 21 days of cultivation, particular keratinocyte markers, such as keratin I and involucrin, were identified through utilizing immunofluorescence and real-time polymerase chain reaction (PCR) tests. The markers' expressions were higher in the PCL-AM scaffold with a ratio of 90:10 v v-1 compared with the PCL-epidermal growth factor (EGF) structure. Moreover, the presence of AM in the scaffolds resulted in the keratinogenic differentiation of ASCs even without employing EGF. Consequently, this state-of-the-art experiment suggests that the PCL-AM scaffold can be a promising candidate in skin bioengineering. Conclusion: This study showed that mixing AM with PCL, a widely used polymer, in different concentrations can overcome PCL disadvantages such as high hydrophobicity and low cellular compatibility.
3D-Printed PCL Scaffolds Coated with Nanobioceramics Enhance Osteogenic Differentiation of Stem Cells

Abstract:With advances in bone tissue engineering, various materials and methods have been explored to find a better scaffold that can help in improving bone growth and regeneration. Three-dimensional (3D) printing by fused deposition modeling can produce customized scaffolds from biodegradable polyesters such as polycaprolactone (PCL). Although the fabricated PCL scaffolds exhibited a lack of bioactivity and poor cell attachment on their surfaces, herein, using a simple postfabrication modification method with hydroxyapatite (HA) and bioglasses (BGs), we obtained better cell proliferation and attachment. Biological behavior and osteosupportive capacity of the 3D-printed scaffolds including PCL, PCL/HA, PCL/BG, and PCL/HA/BG were evaluated in this study, while human adipose tissue-derived mesenchymal stem cells (hADSCs) were cultured on the scaffolds. The cell morphology, attachment, and proliferation were investigated using scanning electron microscopy (SEM), 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay, and 4',6-diamidino-2-phenylindole (DAPI) staining. In the next step, the ability of stem cells to differentiate into osteoblasts was evaluated by measuring alkaline phosphatase (ALP) activity, calcium deposition, and bone-related gene and protein expression. In the end, the expression levels of miR-20a, miR-125a, and their target genes were also investigated as positive and negative regulators in osteogenesis pathways. The results showed that the coated scaffolds with bioceramics present a more appropriate surface for cell adhesion and proliferation, as well as efficient potential in inducing osteoconduction and osteointegration compared to PCL alone and control. The PCL/HA/BG scaffold exhibited higher in vitro cell viability and bone formation compared to the other groups, which can be due to the synergistic effect of HA and BG. On the whole, this tricomponent 3D-printing scaffold has a promising prospect for bone tissue engineering applications.

Abstract:Polycaprolactone (PCL), poly (lactic acid) (PLA) and hydroxyapatite (HA) are frequently used as materials for tissue engineering. In this study, PCL/PLA/HA nanofiber mats with different weight ratio were prepared using electrospinning. Their structure and morphology were studied by FTIR and FESEM. FTIR results demonstrated that the HA particles were successfully incorporated into the PCL/PLA nanofibers. The FESEM images showed that the surface of fibers became coarser with the introduction of HA nanoparticles into PCL/PLA system. Furthermore, the addition of HA led to the decreasing of fiber diameter. The average diameters of PCL/PLA/HA nanofiber were in the range of 300-600 nm, while that of PCL/PLA was 776 +/- 15.4 nm. The effect of nanofiber composition on the osteoblast-like MC3T3-E1 cell adhesion and proliferation were investigated as the preliminary biological evaluation of the scaffold. The MC3T3-E1 cell could be attached actively on all the scaffolds. The MTT assay revealed that PCL/PLA/HA scaffold shows significantly higher cell proliferation than PCL/PLA scaffolds. After 15 days of culture, mineral particles on the surface of the cells was appeared on PCL/PLA/HA nanofibers while normal cell spreading morphology on PCL/PLA nanofibers. These results manifested that electrospun PCL/PLA/HA scaffolds could enhance bone regeneration, showing their marvelous prospect as scaffolds for bone tissue engineering.

Abstract:Background: Many patients lose their organs or tissues due to disease, trauma, or a variety of genetic disorders. Tissue engineering is a multidisciplinary science to regenerate or restore tissue or organ function and an appropriate scaffold is the first and certainly a crucial step in tissue engineering strategies.Objective: The purpose of this study is to fabricate and evaluate the in-vitro response of porous nano Polycaprolactone (PCL)/ chitosan/ multi-wall carbon nanotube (MWCNTs) scaffold for tissue regeneration.Material and Methods: In this experimental research, a novel scaffold containing MWCNTs in polycaprolactone/chitosan nanofibrous scaffold was synthesized by electrospinning technique.Results: According to scanning electron microscopy SEM images, by increasing the number of MWCNT in the scaffold by 2%, the average diameter decreased significantly for fabricated scaffolds with 5% MWCNTs. Based on the results, the scaffolds plunged from submicron to nanoscale fibers at about 80 nm. In addition, by adding more MWCNT to the nanofibrous scaffold, the biodegradation rate was decreased by 32%. However, mechanical characterization demonstrates that the higher level of MWCNT increases young modulus by 96%, and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazolium bromide (MTT) assay illustrated that MWCNTs could enhance bioactivity and cell- scaffold relationship in addition to alkaline phosphatase (ALP).Conclusion: MWCNT significantly improves the physical and mechanical properties of fabricated scaffolds and in-vitro assessment demonstrated that the prepared nanofibrous scaffold containing 4% MWCNT could be a very useful biocompatible material for tissue engineering.
Artificial decellularized extracellular matrix improves the regenerative capacity of adipose tissue derived stem cells on 3D printed polycaprolactone scaffolds

Abstract:Ideal tissue engineering frameworks should be both an optimal biological microenvironment and a shape and stability providing framework. In this study we tried to combine the advantages of cell-derived artificial extracellular matrix (ECM) with those of 3D printed polycaprolactone (PCL) scaffolds. In Part A, both chondrogenic and osteogenic ECMs were produced by human adipose derived stem cells (hASCs) on 3D-printed PCL scaffolds and then decellularized to create cell free functionalized PCL scaffolds, named acPCL and aoPCL respectively. The decellularization resulted in a significant reduction of the DNA content as well as the removal of nuclei while the ECM was largely preserved. In Part B the bioactivation and the effect of the ac/aoPCL scaffolds on the proliferation, differentiation, and gene expression of hASCs was investigated. The ac/aoPCL scaffolds were found to be non-toxic and allow good adhesion, but do not affect proliferation. In the in vitro investigation of cartilage regeneration, biochemical analysis showed that acPCL scaffolds have an additional effect on chondrogenic differentiation as gene expression analysis showed markers of cartilage hypertrophy. The aoPCL showed a large influence on the differentiation of hASCs. In control medium they were able to stimulate hASCs to produce calcium alone and all genes relevant investigated for osteogenesis were significantly higher expressed on aoPCL than on unmodified PCL. Therefore, we believe that ac/aoPCL scaffolds have a high potential to improve regenerative capacity of unmodified PCL scaffolds and should be further investigated.

Dynamics-Based Nonlinear Acceleration Control With Energy Shaping for a Mobile Inverted Pendulum With a Slider Mechanism

Abstract:A nonlinear controller to accelerate a mobile inverted pendulum (MIP) with a slider mechanism is proposed. The concept of this paper is to control translational acceleration and deceleration of the MIP in a dynamically reasonable manner. The body angle and slider displacement are controlled to maintain reference states where the MIP is statically unstable but dynamically stable, which leads to a constant translational acceleration due to instability of the vehicle. The accelerating motion is like a sprinter moving from the crouch start, and it fully exploits the dynamics of the MIP. To achieve it, the total energy of the system is shaped to have the minimum at the given reference states and the system is controlled to converge to them. The controller can achieve various properties through the energy-shaping procedure. In particular, an energy function that will lead to safe operation of the MIP is proposed. The function ensures that motion of the MIP is restricted within predefined regions. The controller also returns the system back to the reference states with state-dependent gains that become large if the system comes close to falling over. The effectiveness of the controller is verified in simulations. Finally, a new physical MIP was constructed, and experiments were carried out with an additional friction compensation method. The results show that the proposed controller works well in the presence of uncertainty, such as noise and modeling errors.
Decellularized extracellular matrix-decorated 3D nanofiber scaffolds enhance cellular responses and tissue regeneration

Abstract:The use of decellularized extracellular matrix products in tissue regeneration is quite alluring yet practically challenging due to the limitations of its availability, harsh processing techniques, and host rejection. Scaffolds obtained by either incorporating extracellular matrix (ECM) material or coating the surface can resolve these challenges to some extent. However, these scaffolds lack the complex 3D network formed by proteins and growth factors observed in natural ECM. This study introduces an approach utilizing 3D nanofiber scaffolds decorated with dECM to enhance cellular responses and promote tissue regeneration. Notably, the dECM can be customized according to specific cellular requirements, offering a tailored environment for enhanced therapeutic outcomes. Two types of 3D expanded scaffolds, namely radially aligned scaffolds (RAS) and laterally expanded scaffolds (LES) fabricated by the gas-foaming expansion were utilized. To demonstrate the proof-of-concept, human dermal fibroblasts (HDFs) seeded on these scaffolds for up to 8 weeks, resulted in uniform and highly aligned cells which deposited ECM on the scaffolds. These cellular components were then removed from the scaffolds through decellularization (e.g., SDS treatment and freeze-thaw cycles). The dECM-decorated 3D expanded nanofiber scaffolds can direct and support cell alignment and proliferation along the underlying fibers upon recellularization. An in vitro inflammation assay indicates that dECM-decorated LES induces a lower immune response than dECM-decorated RAS. Further, subcutaneous implantation of dECM-decorated RAS and LES shows higher cell infiltration and angiogenesis within 7 and 14 days than RAS and LES without dECM decoration. Taken together, dECM-decorated 3D expanded nanofiber scaffolds hold great potential in tissue regeneration and tissue modeling. STATEMENT OF SIGNIFICANCE: Decellularized ECM scaffolds have attained widespread attention in biomedical applications due to their intricate 3D framework of proteins and growth factors. Mimicking such a complicated architecture is a clinical challenge. In this study, we developed natural ECM-decorated 3D electrospun nanofiber scaffolds with controlled alignments to mimic human tissue. Fibroblasts were cultured on these scaffolds for 8 weeks to deposit natural ECM and decellularized by either freeze-thawing or detergent to obtain decellularized ECM scaffolds. These scaffolds were tested in both in-vitro and in-vivo conditions. They displayed higher cellular attributes with lower immune response making them a good grafting tool in tissue regeneration.

Abstract:The interconnected porous structures that mimic the extracellular matrix support cell growth in tissue engineering. Nanofibers generated by electrospinning can act as a vehicle for therapeutic cell delivery to a neural lesion. The incorporation of carbon nanomaterials with excellent electrical conductivity in nanofibers is an attractive aspect for design of a nanodevice for neural tissue regeneration. In this study, nanoscaffolds were created by electrospinning poly(ε-caprolactone) (PCL) and three different types of carbon nanomaterials, which are carbon nanotubes, graphene, and fullerene. The component of carbon nanomaterials in nanofibers was confirmed by Fourier transform infrared spectroscopy. The fiber diameter was determined by scanning electron microscopy, and it was found that the diameter varied depending on the type of nanomaterial in the fibers. The incorporation of carbon nanotubes and graphene in the PCL fibers increased the contact angle significantly, while the incorporation of fullerene reduced the contact angle significantly. Incorporation of CNT, fullerene, and graphene in the PCL fibers increased dielectric constant. Astrocytes isolated from neonatal rats were cultured on PCL-nanomaterial nanofibers. The cell viability assay showed that the PCL-nanomaterial nanofibers were not toxic to the cultured astrocytes. The immunolabeling showed the growth and morphology of astrocytes on nanofiber scaffolds. SEM was performed to determine the cell attachment and interaction with the nanoscaffolds. This study indicates that PCL nanofibers containing nanomaterials are biocompatible and could be used for cell and drug delivery into the nervous system.

Abstract:Polycaprolactone (PCL), poly (lactic acid) (PLA) and hydroxyapatite (HA) are frequently used as materials for tissue engineering. In this study, PCL/PLA/HA nanofiber mats with different weight ratio were prepared using electrospinning. Their structure and morphology were studied by FTIR and FESEM. FTIR results demonstrated that the HA particles were successfully incorporated into the PCL/PLA nanofibers. The FESEM images showed that the surface of fibers became coarser with the introduction of HA nanoparticles into PCL/PLA system. Furthermore, the addition of HA led to the decreasing of fiber diameter. The average diameters of PCL/PLA/HA nanofiber were in the range of 300-600 nm, while that of PCL/PLA was 776 +/- 15.4 nm. The effect of nanofiber composition on the osteoblast-like MC3T3-E1 cell adhesion and proliferation were investigated as the preliminary biological evaluation of the scaffold. The MC3T3-E1 cell could be attached actively on all the scaffolds. The MU assay revealed that PCL/PLA/HA scaffold shows significantly higher cell proliferation than PCL/PLA scaffolds. After 15 days of culture, mineral particles on the surface of the cells was appeared on PCL/PLA/HA nanofibers while normal cell spreading morphology on PCL/PLA nanofibers. These results manifested that electrospun PCL/PLA/HA scaffolds could enhance bone regeneration, showing their marvelous prospect as scaffolds for bone tissue engineering.
Complete amino-acid sequence of a functional unit from a molluscan hemocyanin (Helix pomatia).

Abstract:From the beta c-hemocyanin (beta c-Hc) of the vineyard snail, Helix pomatia, the functional unit d (Mr approximately equal to 50,000-55,000) was isolated by limited proteolysis and gel chromatography. A small quantity of functional unit d was obtained intact, but the major part in the form of two peptides (Mr approximately equal to 43,000 and 10,000, respectively) connected by a disulfide bridge. After reduction and carboxymethylation, these were separated from each other and cleaved by conventional methods. The peptides were isolated by gel chromatography and HPLC, and sequenced manually or automatically. The complete sequence of Helix beta c-Hc d comprises 410 residues plus 3 residues at the N-terminus seemingly resulting from incomplete cleavage. There is apparently only one carbohydrate side-chain. Comparison of this gastropodan hemocyanin sequence to the partial sequence of a cephalopodan Hc C-terminal unit revealed sufficient identities to state that the functional units of molluscan hemocyanins have arisen by a series of gene duplications. On the other hand, there is practically no homology with arthropodan hemocyanins except for one section of 42 residues which is clearly homologous. This section corresponds to the "Copper B" site of Panulirus interruptus hemocyanin. It is also found in tyrosinases from Neurospora crassa, Streptomyces glaucescens, and mouse. In the N-terminal half of Helix beta c-Hc d there are other sections clearly homologous to the tyrosinases, but overall homology is limited. The second copper-binding site was not identified but must be completely distinct from the "Copper A" binding site of arthropodan hemocyanins. It is suggested that molluscan and arthropodan hemocyanins have evolved independently from a common ancestral mononuclear copper protein.

Abstract:The frequent association of affective disorder and multiple sclerosis raises important aetiological and clinical considerations. Reported here are seven patients who presented with symptoms of multiple sclerosis and who had a preceding history of bipolar affective disorder. The hypothesis that bipolar illness may be the initial manifestation of multiple sclerosis is discussed with reference to relevant clinical and epidemiological research on the topic.

Abstract:<p>Nanofibrous scaffolds hold great promise in tissue engineering owing to their extracellular matrix (ECM)-mimicking architectures. Electrospinning, with its ease for producing nanofibrous scaffolds, has therefore been widely employed for various tissue engineering applications. However, electrospun nanofibrous scaffolds have faced the inherent challenge of three-dimensional (3D) cell distribution due to the small sizes of interconnected pores in these scaffolds when conventional approach of scaffold fabrication with subsequent cell seeding is adopted, which severely limits their applications in repairing/regenerating human body tissues with thick and vascularized structures. In this study, we demonstrate a method to directly place living endothelial cells within bioactive nanofibrous scaffolds in 3D through concurrent emulsion electrospinning and coaxial cell electrospraying. Using this concurrent manufacturing method, endothelial cells are encapsulated in hydrogel microspheres and deposited along with vascular endothelial growth factor (VEGF)-containing nanofibers in the scaffold fabrication process, resulting in nanofibrous scaffolds with 3D embedded cell-encapsulated microspheres. After selective disruption of the hydrogel microspheres, the encapsulated endothelial cells are released, yielding bioactive nanofibrous scaffolds with tissue-like 3D cell-incorporated nanofibrous structures. It is shown that cell viability is well preserved (>98%) during the concurrent manufacturing process and that a deep cell distribution (∼100 μm) through the scaffold thickness has been achieved. With combined structural and biochemical cues via the 3D cell-incorporated architectures, endothelial cells can freely stretch, display enhanced intercellular connections, and maintain the phenotype in the bioactive nanofibrous scaffolds. Our investigations offer a promising platform technology for creating bioactive nanofibrous scaffolds with 3D cell incorporation and for overcoming inherent problems of electrospun nanofibrous scaffolds, which should open new avenues for biomanufacturing tissue-mimicking constructs with vascularized structures and complex anatomy.</p>
