Abstract:The fall armyworm (FAW), Spodoptera frugiperda (J. E. Smith, 1797), known as an important agricultural pest around the world, is indigenous to the tropical–subtropical regions in the Western Hemisphere, although its distribution has expanded over large parts of America, Africa, Asia and Oceania in the last few years. The pest causes considerable costs annually coupled with its strong invasion propensity. Temperature is identified as the dominant abiotic factor affecting herbivorous insects. Several efforts have reported that temperature directly or indirectly influences the geographic distribution, phenology and natural enemies of the poikilothermal FAW, and thus may affect the damage to crops, e.g., the increased developmental rate accelerates the intake of crops at higher temperatures. Under some extreme temperatures, the FAW is likely to regulate various genes expression in response to environmental changes, which causes a wider viability and possibility of invasion threat. Therefore, this paper seeks to review and critically consider the variations of developmental indicators, the relationships between the FAW and its natural enemies and the temperature tolerance throughout its developmental stage at varying levels of heat/cold stress. Based on this, we discuss more environmentally friendly and economical control measures, we put forward future challenges facing climate change, we further offer statistical basics and instrumental guidance significance for informing FAW pest forecasting, risk analyses and a comprehensive management program for effective control globally.

Abstract:The fall armyworm (FAW), Spodoptera frugiperda, is regarded as one of the world's most harmful plant pests. This research examines the molecular response processes of FAW to low temperature (4°C) and high temperature (40°C) when gene expression is compared to controls (25°C). A total of 211,967 unigenes were collected, at least 14,338 of which were annotated with gene descriptions, gene ontology terms, and metabolic pathways. There were 50 Gene Ontology (GO) functional sub-groups and 21 EggNOG words as a result. Differentially expresses genes (DEGs) with log2FC ≥ 2 were identified and compared at various temperatures. In comparison to the 25°C treated group, we discovered 199 and 1,248 individual DEGs co-regulated at 4 and 40°C, respectively. Comparing transcriptome profiles for differential gene expression revealed a number of DEGs, including cytochrome P450, odorant binding proteins (OBPs), and immune system genes previously implicated in cold and high temperature stresses. The enrichment pathways were identified using Kyoto Encyclopedia of Genes and Genomics (KEGG) analysis, and heatmaps of similar unigenes from both treatment groups (T4 and T40) were plotted. We used quantitative reverse transcription PCR (RT-qPCR) to confirm the RNA-seq data on 10 up- and down-regulated DEGs. These findings provide a foundation for future understanding of FAW adaptation mechanisms and the underlying basis underlying the response to low and high temperatures.

Abstract:Spodoptera frugiperda (J.E. Smith) is a significant economic pest that has recently invaded Africa and Asia. However, much of the information regarding its ecological capabilities in these newly invaded environments remains largely unknown. In this study, the life history traits of the fall armyworm under conditions of increased temperature, different photoperiods, and varying humidity levels were systematically evaluated. Among 43 studies, a total of 20 studies were included in the analysis by passing the screening criteria, and random-effects meta-analysis, fixed-effects meta-analysis, and meta-regression were conducted. It has been found that with the increase in temperature above 20 °C, various physiological indicators of the fall armyworm are significantly enhanced. When the temperature reaches 32 °C, the physiological activities of S. frugiperda are at their highest point. As the temperature increases, the duration of each developmental stage of the fall armyworm decreases significantly, accompanied by an increase in oviposition quantity and period in females. Additionally, the pupal development time is shortened, which leads to an increase in the lifespan of the adult moth. Using temperature and relative humidity as environmental variables, the optimal survival conditions for each insect state of the fall armyworm were calculated. These findings can assist in predicting the population dynamics of the fall armyworm and in formulating appropriate and practical management strategies.

Abstract:How herbivorous insects adapt to host plants is a key question in ecological and evolutionary biology. The fall armyworm, (FAW) Spodoptera frugiperda (J.E. Smith), although polyphagous and a major pest on various crops, has been reported to have a rice and corn (maize) feeding strain in its native range in the Americas. The species is highly invasive and has recently established in China. We compared behavioral changes in larvae and adults of a corn population (Corn) when selected on rice (Rice) and the molecular basis of these adaptational changes in midgut and antennae based on a comparative transcriptome analysis. Larvae of S. frugiperda reared on rice plants continuously for 20 generations exhibited strong feeding preference for with higher larval performance and pupal weight on rice than on maize plants. Similarly, females from the rice selected population laid significantly more eggs on rice as compared to females from maize population. The most highly expressed DEGs were shown in the midgut of Rice vs. Corn. A total of 6430 DEGs were identified between the populations mostly in genes related to digestion and detoxification. These results suggest that potential adaptations for feeding on rice crops, may contribute to the current rapid spread of fall armyworm on rice crops in China and potentially elsewhere. Consistently, highly expressed DEGs were also shown in antennae; a total of 5125 differentially expressed genes (DEGs) s were identified related to the expansions of major chemosensory genes family in Rice compared to the Corn feeding population. These results not only provide valuable insight into the molecular mechanisms in host plants adaptation of S. frugiperda but may provide new gene targets for the management of this pest.

Abstract:The brown planthopper (Nilaparvata lugens, BPH), white-backed planthopper (Sogatella furcifera, WBPH) and small brown planthopper (Laodelphax striatellus, SBPH) are important rice pests in Asia. These three species differ in thermal tolerance and exhibit quite different migration and overwintering strategies. To understand the underlying mechanisms, we sequenced and compared the transcriptome of the three species under different temperature treatments. We found that metabolism-, exoskeleton- and chemosensory-related genes were modulated. In high temperature (37 degrees C), heat shock protein (HSP) genes were the most co-regulated; other genes related with fatty acid metabolism, amino acid metabolism and transportation were also differentially expressed. In low temperature (5 degrees C), the differences in gene expression of the genes for fatty acid synthesis, transport proteins and cytochrome P450 might explain why SBPH can overwinter in high latitudes, while BPH and WBPH cannot. In addition, other genes related with moulting, and membrane lipid composition might also play roles in resistance to low and high temperatures. Our study illustrates the common responses and different tolerance mechanisms of three rice planthoppers in coping with temperature change, and provides a potential strategy for pest management.

Abstract:Temperature is an important environmental factor in agriculture, affecting individual organisms and the entire farmland ecosystem. Global warming has become more tangible, which may negatively affect pest biological control due to the generally weak thermal tolerance of natural enemies. The mirids Cyrtorhinus lividipennis and Tytthus chinensis (Insecta: Hemiptera: Miridae) are important natural predators of planthoppers and leafhoppers in Asian paddy fields. However, the effects of thermal stress on these predators remain poorly understood. We investigated the thermal tolerance, fitness, predation abilities, and transcriptomic response of T. chinensis and C. lividipennis at elevated temperatures. T. chinensis was more heat tolerant than both Nilaparvata lugens (its prey) and C. lividipennis. T. chinensis not only exhibited better development, survival, reproduction, and predation capacities compared with C. lividipennis but also showed stronger competitiveness when the two mirid predators co-persisted under high-temperature conditions. To understand the underlying mechanisms, we sequenced their transcriptomes at different temperatures. Heat shock protein (HSP) genes were identified and analyzed due to their high co-regulation during heat treatment. Quantitative polymerase chain reaction results showed that T. chinensis induces HSPs expression quickly and strongly over a wider temperature range in response to heat stress compared with C. lividipennis. Taken together, we highlighted the potential of T. chinensis as a biological control agent in future global warming conditions and provided insight into the thermal adaption of mirid species.
Genomic divergences between the two polyphagous Spodoptera relatives provide cues for successful invasion of the fall armyworm

Abstract:The fall armyworm Spodoptera frugiperda recently invaded China, ravaging crops in many provinces. Deciphering the possible genetic basics for its successful invasion is critical for innovative and specific control for this gluttonous pest. Here we generated comparative genomic analyses between S. frugiperda and its native relative, S. litura, which differs in host preference, locomotivity and production behavior. We demonstrated that S. frugiperda genes are enriched in taste sensory perception and nervous system, obviously different from those of S. litura. Potential host adaptation genes showed generally an elevated ratio of non-synonymous substitution rate to synonymous substitution rate, suggesting a faster evolution during the divergence of the two species. Focusing on these sets of genes, we identified 23 genes being under positive selection in S. frugiperda. Among them are two notable genes involved in sensory perception, gustatory receptor (GR) and an acetaldehyde oxidase, which are important for host detection in invasion and expansion processes. Another two genes are mitochondrial adenosine triphosphate synthase β subunit and ferritin heavy chain, which may be associated with the enhanced locomotivity and resistance, which fascinated long-distance migration needed for invasion and rapid expansion. Another interesting gene is chorion protein, in which positive selection sites in S. frugiperda were found and a replacement in one site is predicted to affect the protein function, which might be associated with competent reproductivity in S. frugiperda to ensure genetic resources for expansion.
Composition and Expression of Heat Shock Proteins in an Invasive Pest, the Rice Water Weevil (coleoptera: Curculionidae)

Abstract:Abstract For poikilothermic groups such as insects, the capacity to adapt to different temperature regimes is particularly important for survival. To investigate the possible role of heat shock proteins (Hsps) in the invasive pest, the rice water weevil (Lissorhoptrus oryzophilus Kuschel, Coleoptera: Curculionidae), we first analyzed the composition and expression profile of Hsp families under sub-lethal temperatures of 0 °C and 43 °C, using the quantitative realtime polymerase chain reaction. Eight genes coding Hsp90, Hsp70, and small Hsps were upregulated under heat stress, while only 1 Hsp70 gene and 1 Hsp90 gene were up-regulated under cold stress. Results indicate that Hsps from all families except Hsp60 are responsible for the capacity of L. oryzophilus to tolerate temperature stress, although more genes were up-regulated, and more rapidly, under heat stress than under cold stress. Secondly Hsp expression patterns in diapausing and non-diapausing female adults were investigated. The results showed that rice water weevils in diapause up-regulated no Hsp gene but they downregulated 4 small Hsps, 2 Hsp90, 1 Hsp70 and 1 Hsp60 genes.

Abstract:In mid-May, 2019, the fall armyworm (FAW) Spodoptera frugiperda invaded Jiangxi Province, China, and caused extensive damage to corn crops. However, little attention has been given to the life-history traits of the FAW. In the present study, we systematically investigated the life-history traits of the newly invasive FAW on corn leaves at 19, 22, 25, 28, and 31°C under a photoperiod of LD 15:9 hr. The FAW thrived on the corn leaves with short developmental periods, high survival rates of larvae and pupae, very high mating success rates, and high fecundity. The pupal developmental stage was significantly longer in males than females at all temperatures, thus resulting in a protogyny phenomenon. The pupal weight was heaviest after a relatively shorter larval development stage at a higher temperature (25°C); thus, the FAW did not follow the temperature–size rule. Females were smaller than males, indicating sexual size dimorphism. A small proportion of females delayed their pre-oviposition period and began to lay eggs on the 7th to 9th day after adult emergence. There were positive relationships between pupal weight and larval developmental time and between adult weight and fecundity. There was a negative relationship between fecundity and longevity. These findings can help us to predict the population dynamics of the FAW on corn and to develop a suitable and practical management strategy.

High and Low Temperatures Differentially Affect Survival, Reproduction, and Gene Transcription in Male and Female Moths of Spodoptera frugiperda

Abstract:In this study, we found that both heat and cold stresses significantly affected the survival and reproduction of both sexes in Spodoptera frugiperda adults, with larvae showing relatively higher extreme temperature tolerance. Further transcriptomic analysis in adults found remarkable differences and similarities between sexes in terms of temperature stress responses. Metabolism-related processes were suppressed in heat stressed females, which did not occur to the same extend in males. Moreover, both heat and cold stress reduced immune activities in both sexes. Heat stress induced the upregulation of many heat shock proteins in both sexes, whereas the response to cold stress was insignificant. More cold tolerance-related genes, such as cuticle proteins, UDP-glucuronosyltransferase, and facilitated trehalose transporter Tret1, were found upregulated in males, whereas most of these genes were downregulated in females. Moreover, a large number of fatty acid-related genes, such as fatty acid synthases and desaturases, were differentially expressed under heat and cold stresses in both sexes. Heat stress in females induced the upregulation of a large number of zinc finger proteins and reproduction-related genes; whereas cold stress induced downregulation in genes linked to reproduction. In addition, TRPA1-like encoding genes (which have functions involved in detecting temperature changes) and sex peptide receptor-like genes were found to be differentially expressed in stressed moths. These results indicate sex-specific heat and cold stress responses and adaptive mechanisms and suggest sex-specific trade-offs between stress-resistant progresses and fundamental metabolic processes as well as between survival and reproduction.

Abstract:Wheat, a major worldwide staple food crop, is relatively sensitive to a changing environment, including high temperature. The comprehensive mechanism of heat stress response at the molecular level and exploitation of candidate tolerant genes are far from enough. Using transcriptome data, we analyzed the gene expression profiles of wheat under heat stress. A total of 1705 and 17 commonly differential expressed genes (DEGs) were identified in wheat grain and flag leaf, respectively, through transcriptome analysis. Gene Ontology (GO) and pathway enrichment were also applied to illustrate the functions and metabolic pathways of DEGs involved in thermotolerance of wheat grain and flag leaf. Furthermore, our data suggest that there may be a more complex molecular mechanism or tighter regulatory network in flag leaf than in grain under heat stress over time, as less commonly DEGs, more discrete expression profiles of genes (principle component analysis) and less similar pathway response were observed in flag leaf. In addition, we found that transcriptional regulation of zeatin, brassinosteroid and flavonoid biosynthesis pathways may play an important role in wheat’s heat tolerance. The expression changes of some genes were validated using quantitative real-time polymerase chain reaction and three potential genes involved in the flavonoid biosynthesis process were identified.
Molecular mechanisms of cytochrome P450-mediated detoxification of tetraniliprole, spinetoram, and emamectin benzoate in the fall armyworm, Spodoptera frugiperda (J.E. Smith)

Abstract:The fall armyworm (FAW) Spodoptera frugiperda (J.E. Smith) is a highly damaging invasive omnivorous pest that has developed varying degrees of resistance to commonly used insecticides. To investigate the molecular mechanisms of tolerance to tetraniliprole, spinetoram, and emamectin benzoate, the enzyme activity, synergistic effect, and RNA interference were implemented in S. frugiperda. The functions of cytochrome P450 monooxygenase (P450) in the tolerance to tetraniliprole, spinetoram, and emamectin benzoate in S. frugiperda was determined by analysing changes in detoxification metabolic enzyme activity and the effects of enzyme inhibitors on susceptibility to the three insecticides. 102 P450 genes were screened via transcriptome and genome, of which 67 P450 genes were differentially expressed in response to tetraniliprole, spinetoram, and emamectin benzoate and validated by quantitative real-time PCR. The expression patterns of CYP9A75, CYP340AA4, CYP340AX8v2, CYP340L16, CYP341B15v2, and CYP341B17v2 were analysed in different tissues and at different developmental stages in S. frugiperda. Silencing CYP340L16 significantly increased the susceptibility of S. frugiperda to tetraniliprole, spinetoram, and emamectin benzoate. Furthermore, knockdown of CYP340AX8v2, CYP9A75, and CYP341B17v2 significantly increased the sensitivity of S. frugiperda to tetraniliprole. Knockdown of CYP340AX8v2 and CYP340AA4 significantly increased mortality of S. frugiperda to spinetoram. Knockdown of CYP9A75 and CYP341B15v2 significantly increased the susceptibility of S. frugiperda to emamectin benzoate. These results may help to elucidate the mechanisms of tolerance to tetraniliprole, spinetoram and emamectin benzoate in S. frugiperda.
Comparative transcriptome analysis provides comprehensive insight into the molecular mechanisms of heat adaption in Plutella xylostella

Abstract:Plutella xylostella is one of the most destructive pests for cruciferous vegetables, and is adaptability to different environmental stressors. However, we still know little about the molecular mechanisms of how P. xylostella adapt to thermal stress. Here, the comparative transcriptome analysis was conducted from the samples of control (27 °C, CK) and heat treatment (40 °C, 40 T) P. xylostella. The results showed 1253 genes were differentially expressed, with 624 and 629 genes up- and down-regulated respectively. The annotation analysis demonstrated that "Energy production and conversion", "Protein processing in endoplasmic reticulum", "Peroxisome" and "Tyrosine metabolism" pathways were significantly enriched. Additionally, we found the expression levels of heat shock protein genes (Hsps), cuticle related genes and mitochondrial genes were significantly up-regulated in 40 T insects, suggesting their vital roles in improving adaption to heat stress. Importantly, the SOD activity and MDA content of P. xylostella were both identified to be increased under high temperature stress, indicating the elevated antioxidant reactions might be involved in response to heat stress. In conclusion, the present study offered us an overview of gene expression changes after 40 °C treatments, and found some critical pathways and genes of P. xylostella might play the critical roles in resisting heat stress.
The Effect of Temperatures and Hosts on the Life Cycle of Spodoptera frugiperda (Lepidoptera: Noctuidae)

Abstract:The interactions between ambient temperatures and host plants are central to the population dynamics of invasive animal species. Despite significant research into the effects of temperatures, the performance of invasive species is also influenced by host plants. The effects of different temperatures (20, 25, and 30 °C) and host plants (maize, sorghum, and coix seed) were tested on the mortality, development, reproduction, and population parameters of the fall armyworm (FAW), Spodoptera frugiperda (J E Smith) (Lepidoptera: Noctuidae), using an age-stage, two-sex life table. The results support the hypothesis that temperature and the species of the host plant significantly influences the performance of FAW. Feeding on maize at 30 °C resulted in a lower mortality rate, a shorter developmental time and longevity, a higher fecundity, intrinsic rate of natural increase (r), finite rate of increase (λ), and net reproductive rate (R0). However, at 20 °C, the host plant could eliminate temperature-mediated synergism in FAW performance, which did not reach statistical significance at 20 °C. Similar results induced by a relatively low temperature (20 °C) on different host plants were also found in the age-stage specific survival curves (sxj), fecundity (mx), maternity (lxmx), and reproductive value (vxj) curves of FAW. Consequently, we also need to pay more attention to FAW outbreaks on different host plants mediated by relatively low temperatures.

Abstract:Understanding a population's fitness heterogeneity and genetic basis of thermal adaptation is essential for predicting the responses to global warming. We examined the thermotolerance and genetic adaptation of Plutella xylostella to exposure to hot temperatures. The population fitness parameters of the hot-acclimated DBM strains varied in the thermal environments. Using genome scanning and transcription profiling, we find a number of genes potentially involved in thermal adaptation of DBM. Editing two ABCG transporter genes, PxWhite and PxABCG, confirmed their role in altering cuticle permeability and influencing thermal responses. Our results demonstrate that SNP mutations in genes and changes in gene expression can allow DBM to rapidly adapt to thermal environment. ABCG transporter genes play an important role in thermal adaptation of DBM. This work improves our understanding of genetic adaptation mechanisms of insects to thermal stress and our capacity to predict the effects of rising global temperatures on ectotherms.

Abstract:The fall armyworm, Spodoptera frugiperda Smith (Lepidoptera: Noctuidae), a common agricultural pest known for its extensive migration and wide host ranges, causes considerable harm to maize (Zea mays L.). Additionally, through an interactive study between S. frugiperda larvae and six maize varieties aiming to understand the insect's adaptability and resistance mechanisms, our analysis revealed that both the COI and Tpi genes identified S. frugiperda as the corn strain. Further examination of the larvae showed significant differences in nutritional indices, digestive, and detoxification enzyme activities. Special maize varieties were found to offer higher efficiency in nutrient conversion and assimilation compared with common varieties. This study revealed adaptations in S. frugiperda's digestive and detoxification processes in response to the different maize varieties. For instance, larvae reared on common maize exhibited elevated amylase and lipase activities. Interestingly, detoxification enzyme activities exhibited different patterns of variation in different maize varieties. The Pearson correlation analysis between nutritional indices, enzyme activities, and the nutritional content and secondary metabolites of maize leaves provided deeper insights into the pest's adaptability. The results highlighted significant relationships between specific nutritional components in maize and the physiological responses of S. frugiperda. Overall, our findings contribute substantially to the understanding of S. frugiperda's host plant adaptability, offering critical insights for the development of sustainable pest management strategies.

Abstract:As an invasive species, Bemisia tabaci Mediterranean (MED) has notable potential to adapt to a wide range of environmental temperatures, which enables it to successfully spread after invasion and occupy habitats over a wide latitude range. It has been postulated that chromatin remodeling mechanisms are related to the rapid acquisition of adaptive traits and thermal resistance in invasive species; however, relevant experimental evidence is scarce. To identify the molecular characteristics and assess the role of chromatin remodelers in thermal stress within invasive MED and native Asia II 1 of the B. tabaci species complex, we identified 13 switching defective/sucrose non-fermenting (SWI/SNF) and 10 imitation switch (ISWI) family members in the B. tabaci genome, analyzed their molecular characteristics and structures, and identified key mutation sites between MED and Asia II 1, then cloned the catalytic subunits, and revealed the difference in thermal tolerance function. The results showed that the expression levels of Bt-BRM-1 and Bt-BRM-2 were significantly higher in MED than in Asia II 1 during heat stress, and Bt-BRM-2 expression was significantly higher during cold stress. In addition, RNA interference results indicated that the two target genes had similar temperature tolerance function in the both two cryptic species. This study is the first to identify and analyze the molecular characteristics of SWI/SNF and ISWI family members and reveal their potential key roles in temperature tolerance in poikilothermic ectotherms. The results will assist in understanding the underlying temperature adaptation mechanism of invasive insects and will enrich stress adaptation research systems from an epigenetic perspective.

Abstract:The fall armyworm (FAW), Spodoptera frugiperda (J.E. Smith), is one of the most highly polyphagous invasive pests causing serious damage to maize crops in China. However, little is known about the gut immune responses to the environment, particularly along the migration routes in Jianghuai, China, throughout the autumn and winter. In this study, high-throughput sequencing and real-time quantitative PCR (RT-qPCR) were employed to examine the variations in immune genes and gut microbiome communities between captive and wild fall armyworm populations. Results showed that the diversity and community of the gut's microbes were higher in wild populations, and the average weighted UniFrac distance between bacterial taxa varied. A wide variety of immune genes were more abundant in the wild populations than in others. Results indicated that diets and different survival conditions impacted the gut microbiota and immune system of S. frugiperda, which was crucial for environmental adaptation. These differences in gut microbiota and immune responses between wild and captive Fall armyworms are critical for comprehending the symbiotic relationship between microbes, immune genes, and hosts. They also highlight the need for increased focus on developing more effective and environmentally friendly pest control methods.

Abstract:Temperature is one of the critical abiotic factors influencing the range, richness, activity, life history and viability of insects. The persistence trend of thermal stress on biological and reproductive traits were studied for invasive fall armyworm, Spodoptera frugiperda (J. E. Smith) by exposing early fourth instar larvae to thermal stress treatments (42° and 45 °C) for single and multiple times along with control (27 °C) and reared them till F 2 generation. Survival of larvae was minimum in 45 °C single (88%) and multiple thermal stress (92.5%). The carry over effect of thermal stress was evident up to F 2 generation for developmental attributes with prolonged developmental duration, reduced maturation success and female biased sex ratio. Reproductive parameters such as gonadosomatic index, eupyrene sperm count, fecundity and mating frequency reduced drastically as a result of thermal stress in F 0 generation and the effect persisted up to F 2 generation on fecundity and gonadosomatic index. The biochemical response due to thermal stress was investigated by analyzing metabolic reserves in the adult stage. The reduction in water content and accumulation of total sugar, glycogen, lipid and protein content was observed in adults of all three generations. A trade-off between reproduction and survival strategies was evident under thermal stress with the allocation of resources between them. Thus, our study provides a basic understanding on persistence of thermal stress across generations both at biological and biochemical level and offers insights into further studies on demography of fall armyworm under current and future climate change scenario.

Abstract:The fall armyworm (Spodoptera frugiperda, J.E. Smith) is one of the most important agricultural pests in the world and causes serious damage to many significant crops. Insect gut microbiota plays a vital role in host immunity, digestion, and development, helping the higher organism colonize in a new environment. However, the effects of different diets on midgut microbial composition and host immunity in S. frugiperda remain unclear. So far, no reports have compared the gut microbiota of fall armyworm reared using an artificial diet compared to corn leaf in Guangzhou, China. High-throughput 16S rRNA sequencing technology was applied to gain insight into the composition of the gut microbiota of S. frugiperda feeding on corn leaf (field diet) and on a starch-rich artificial diet (lab diet). The fall armyworm gut microbiota was dominated by the bacterial phyla Firmicutes and Proteobacteria. Despite the difference in diet, the core bacterial community was represented by the genus Enterococcus. However, the bacterial community is dominated by a few phylotypes, namely operational taxonomical units 1 (OTU1) (Enterococcus casseliflavus), OTU3 (Enterobacteriaceae), OTU2 (Weissella), and OTU4 (Clostridium), accounting for 97.43% of the total OTUs in the complete dataset. A significant difference was identified in the bacterial communities between the "lab diet" and the "field diet" groups. OTU1 and OTU2 were significantly higher in the "field diet" group, whereas OTU3 and OTU4 were higher in the "lab diet" group. A phylogenetic investigation of the communities by reconstruction of unobserved states (PICRUSt) predicted functional analysis indicates the presence of several genes associated with plant biomass degradation. Importantly, antibiotic-mediated perturbation of the midgut microbial community significantly impacts the expression profile of the important immune genes of the host. Furthermore, the oral reintroduction of gut bacterial isolates (E. mundtii and E. gallinarum) significantly enhances host resistance to AcMNPV infection. Taken together, our results indicate that diet composition is an important driver in shaping insect gut microbiome and immune gene expression, ultimately playing an important role in the pest defense system.
