Abstract:Theranostic sutures are derived from innovative ideas to enhance wound healing results by adding wound diagnostics and therapeutics to typical sutures by functionalizing them with additional materials. Here, we present a new direct electrospinning method for the fast, continuous, inexpensive, and high-throughput production of versatile nanofibrous-coated suture threads, with precise control over various essential microstructural and physical characteristics. The thickness of the coating layer and the alignment of nanofibers with the thread's direction can be adjusted by the user by varying the spooling speed and the displacement between the spinneret needle and thread. To show the flexibility of our method for a range of different materials selected, gelatin, polycaprolactone, silk fibroin, and PEDOT:PSS (poly(3,4-ethylene dioxythiophene):poly(styrene sulfonate)) were the resultant nanofibers characterized by scanning electron microscopy (SEM) imaging and conductivity tests. In a series of in vitro and ex vivo tests (pig skin), sutures were successfully tested for their flexibility and mechanical properties when used as weaving and knotting sutures, and their biocompatibility with a keratinocyte cell line. For temperature-based drug-releasing tests, two fluorescent molecules as drug models with high and low molecular weight, namely fluorescein isothiocyanate-dextran (20 kDa) and rhodamine B (470 Da), were used, and their steady release with incremental increase of temperature to 37 °C over 120 min was seen, which is appropriate for bacterial treatment drugs. Given the advantages of the presented technique, it seems to have promising potential to be used in future medical applications for wound closure and bacterial infection treatment via a temperature-triggered drug release strategy.
Mechanical Stability of Custom and Anatomical Femoral Stems: An Experimental Study in Human Femora

Abstract:In this study we have compared the mechanical stability of custom (n=8) and anatomical (n=8) uncemented femoral components, following insertion into human cadaveric femurs, during simulated single leg stance and stair climbing. In the custom group two specimens were excluded from the study due to detachment of the greater trochanter during cyclical loading. As a consequence of their mechanical behaviour both types of stems could be divided into subgroups of “unstable” and “stable” implants. In the course of one thousand loading cycles three anatomical stems and one custom stem migrated more than 1 mm, which was interpreted as mechanical loosening. This difference in rate of mechanical loosening was not significant. However, the majority of the stems were remarkably stable and showed micromotion of less than 18 μm and migration of less than 35 μm at the proximal implant-bone interface. The corresponding figures for the tip of the stems were 243 μm and 170 μm, respectively. During torsional loading the custom stems showed less rotatory motion than the anatomical stem (p<0.05). There were no significant differences in the magnitude of cyclical micromotion or migration for the two types of femoral stems.

Abstract:Polymeric nanofibers are of great interest in biomedical applications, such as tissue engineering, drug delivery and wound healing, due to their ability to mimic and restore the function of natural extracellular matrix (ECM) found in tissues. Electrospinning has been heavily used to fabricate nanofibers because of its reliability and effectiveness. In our research, we fabricated poly(ε-caprolactone)-(PCL), magnesium oxide-(MgO) and keratin (K)-based composite nanofibers by electrospinning a blend solution of PCL, MgO and/or K. The electrospun nanofibers were analyzed by scanning electron microscopy (SEM), Fourier transform infrared spectroscopy (FTIR), mechanical tensile testing and inductively-coupled plasma optical emission spectroscopy (ICP-OES). Nanofibers with diameters in the range of 0.2-2.2 µm were produced by using different ratios of PCL/MgO and PCL-K/MgO. These fibers showed a uniform morphology with suitable mechanical properties; ultimate tensile strength up to 3 MPa and Young's modulus 10 MPa. The structural integrity of nanofiber mats was retained in aqueous and phosphate buffer saline (PBS) medium. This study provides a new composite material with structural and material properties suitable for potential application in tissue engineering.

Abstract:Strong and tough biofibers based on regenerated silk fibroins (RSFs) are designed by a dual‐crosslinking strategy. Covalent bonds between tyrosines are introduced by photo crosslinking and coordination interactions of Zn2+ ions with serines are built between RSFs, forming a dual‐crosslinked network. Dual‐crosslinked RSF fibers exhibit excellent mechanical performances, temperature adaptability, fatigue resistance, biocompatibility and biodegradability, making them ideal candidates for sutures. High‐performance bio‐based fibers have unique advantages in biocompatibility for biomedical applications, such as sutures, hernia patches, and blood vessels. Herein, strong and tough biofibers based on regenerated silk fibroins (RSFs) are designed by a dual‐crosslinking strategy and prepared continuously in situ via a single‐channel microfluidic device. During the wet spinning process, covalent bonds between tyrosines are introduced by photo crosslinking and coordination interactions of Zn2+ ions with serines are built between RSFs, forming a dual‐crosslinked, and inter‐connected network and effectively improving the mechanical performances of RSF fibers. By optimizing the wet spinning parameters and applying an optimal post‐stretching treatment, dual‐crosslinked RSF fibers exhibit an excellent tensile strength of 891 MPa, a remarkable Young's modulus of 15.0 GPa, a large tensile strain of 20.1%, and an outstanding toughness of 114 MJ m−3, which are superior to other regenerated biofibers. In addition to the outstanding mechanical performances, dual‐crosslinked RSF fibers also possess excellent temperature adaptability, fatigue resistance, biocompatibility, and biodegradability, making them ideal candidates for biomedical applications. In vivo tests of dual‐crosslinked RSF fibers as sutures demonstrate a good wound healing performance with smaller scar and lower inflammation.

Abstract:Abstract In laboratory conditions, composite sutures based on polylactide (PLA) containing chitin nanofibrils modified with polyethylene glycol (CN‐PEG) and poviargol (silver nanoparticles stabilized with poly( N ‐vinylpyrrolidone)) were obtained, studied, and used as a prototype. Surgical sutures threads with the addition of CN‐PEG have stable mechanical properties both in air and in a buffer simulating the environment of a living organism. The yield strength of oriented threads decreased by an average of 15%, whereas for non‐oriented threads the decrease was 3–4 times. The strength values in simple units of unfilled PLA, PLA containing 5 wt % CN‐PEG, and PLA with 1 wt % Poviargol were on average 50% higher than the national standard 31620‐2012. The results of in vivo experiments on albino rats (cross‐linking skin and muscle tissue in the linea alba area) showed that composite sutures are best for suturing muscle tissue, whereas unfilled PLA sutures are more suitable for suturing skin. When suturing muscle tissue, suturing with composite sutures increased the number of collagen fibers of different diameters.

Abstract:As one of the most widely used medical device, sutures face challenges related to surgical site infections (SSIs) and lack of subcutaneous traceability. In the present study, we have developed a facile and effective approach using peptide‐AIE nanofibers (NFs‐K18) to create fluorescent‐traceable antimicrobial sutures, which have been applied to four commercially available sutures. The functionalized sutures of PGAS‐NFs‐K18 and PGLAS‐NFs‐K18 exhibited fluorescence with excellent penetration from 4 mm chicken breasts. They also demonstrated remarkable stability after 24 h of white light illumination and threading through chicken breasts 10 times. These sutures efficiently generated ROS, resulting in a significant suppression of four clinical bacteria, with the highest antimicrobial rate of nearly 100%. Moreover, the sutures exhibited favorable hemocompatibility and biocompatibility. In vivo experiments demonstrated that the optimized PGLAS‐NFs‐K18 suture displayed potent antimicrobial activity against MRSA, effectively inhibiting inflammation and promoting tissue healing in both skin wound and abdominal wall wound models, outperforming the commercially available Coated VICRYL® Plus Antibacterial suture. Importantly, PGLAS‐NFs‐K18 exhibited sensitive subcutaneous traceability, allowing for accurate in situ monitoring of its degradation. Abstract:Two types of fibers were prepared by using bio- based materials: a mono-filament made from poly(3- hydroxybutyrate-co-3-hydroxyhexanoate) (PHBHHx) and a multi-filament made from poly(3-hydroxybutyrate-co-3- hydroxyvalerate) (PHBV) and polylactic acid (PLA) blend. The two fibers were evaluated for mechanical properties, biocompatibility and degradability for the potential appli- cation as medical sutures. The PHBHHx fiber showed remarkable biocompatibility by H.E. Stainning, with very little impact to the surrounding tissues. The degradation of the fiber was observed by SEM after implantation for 36 weeks, and the major degradation product was detected after 96 weeks. Consistently, the PHBHHx fiber main- tained more than half of the mechanical properties after 96 weeks. The other fiber was prepared by twisting PHBV/PLA blend strands to a bunch, and showed high biocom- patibility and relatively high degradability. The bunched structure loosed after 36 weeks of implantation. These low- cost and easily prepared fibers have great potential in medical applications, since they could avoid the formation of fibrous capsule, reduce the size of scar, and degrade into non-toxic and even beneficial products.

Abstract:Multifunctional microfibers with long‐lasting analgesia capabilities are presented for sutures from microfluidic spinning. Their morphology, microstructure, mechanical property, and the analgesia effect can be easily yet precisely mastered in the microfluidic spinning process. The promising mechanical property, suturing ability, and effective long‐term analgesic performance of the resulting multifunctional microfibers open up a way for their clinical surgery application. Sutures are the most commonly used wound repair method after surgery. However, addressing delayed recovery and pain management remains a significant challenge. Here, microfibers are developed from microfluidic spinning with long‐lasting analgesia capabilities for sutures. By using a solvent extraction manner, the polycaprolactone (PCL) microfibers encapsulated with ropivacaine (ROP), a well‐known analgesic, can be continuously obtained from microfluidics. The intrinsic property of PCL and the advantage of microfluidic spinning technique impart the microfiber with highly controlled morphologies, mechanical strengths, as well as drug release. After exploring their biocompatibility both at in vitro and in vivo levels, the microfibers are directly applied to wound suture. The results demonstrate the lasting analgesic effect of the microfiber on mice with incision pain, highlighting its potential as promising suture for post‐surgery treatments. It is anticipated that the multifunctional analgesic sutures produced through microfluidic spinning will pave the way for utilizing fibers as effective sutures in clinical incision wound treatment.

Abstract:Bile acid‐based PU fibers demonstrate tunable mechanical properties and self‐tightening behavior when exposed to heat or an infrared (IR) laser. In vivo experiments confirm that bio‐derived polymer fibers not only possess outstanding tissue compatibility, but also exhibit superior healing efficiency as the smart sutures. Materials made from bile acids can benefit from the excellent biocompatibility and rigid skeleton of these natural compounds. To address concerns of the biocompatibility of shape‐memory polymers (SMPs) in biomedical applications, a series of polyurethanes (PUs) is developed using bile acid as the hard segment and oligo(ethylene glycol) as the soft segment through a one‐pot polymerization. These polymers can be easily processed into skin‐like fibers, tubes, strips, etc., with tunable mechanical properties, e.g., elongation at break (483%) and high tensile strength (40.9 MPa). Their glass transition temperatures spread across a wide range, enabling them to exhibit dual, triple, and quadruple shape memory effects (SMEs). Due to intermolecular interactions, these PUs display a maximum self‐healing efficiency of 96% in tensile strength for 1.5 h. In vitro cytotoxicity tests, hemolysis evaluations and healing efficiency in vivo all indicate their good biocompatibility and biosafety. These bio‐derived PUs offer an option of shape memory polymers easy to make, with good tissue compatibility for applications as surgical sutures, artificial muscles, and soft materials.

Abstract:Keratin is an important protein used in wound healing and tissue recovery. In this study, keratin was modified chemically with iodoacetic acid (IAA) to enhance its solubility in organic solvent. Poly(hydroxybutylate-co-hydroxyvalerate) (PHBV) and modified keratin were dissolved in hexafluoroisopropanol (HFIP) and electrospun to produce nanofibrous mats. The resulting mats were surface-characterized by ATR-FTIR, field-emission scanning electron microscopy (FE-SEM) and electron spectroscopy for chemical analysis (ESCA). The purem-keratin mat was cross-linked with glutaraldehyde vapor to make it insoluble in water. The biodegradation testin vitro showed that the mats could be biodegraded by PHB depolymerase and trypsin aqueous solution. The results of the cell adhesion experiment showed that the NIH 3T3 cells adhered more to the PHBV/m-keratin nanofibrous mats than the PHBV film. The BrdU assay showed that the keratin and PHBV/m-keratin nanofibrous mats could accelerate the proliferation of fibroblast cells compared to the PHBV nanofibrous mats.

Abstract:Recently, the design of polymer nanofibers using the electrospinning process has attracted much interest. Particularly the use of natural polymers has promoted many advantages in their biomedical applications. However, the combination of multiple natural polymers remains a great challenge in terms of electrospun production and applied performance. From this perspective, the current investigation highlights the study of the preparation of electrospun nanomaterial scaffolds based on combined natural polymers for improved wound healing performance. First, we have synthesized a crosslinked polymer by reacting microcrystalline cellulose (MC) and chitosan (CS) biopolymer via the intermediate of citric acid as a crosslinking agent. Then a natural propolis biomolecule was incorporated into the polymer network. Different MC/CS blend ratios of 90/10 and 70/30 were then used and various machine parameters were optimized to obtain nanofiber scaffolds with excellent strength and structures. SEM, IR, physicochemical, mechanical, and morpho-logical characterization were then performed. SEM evaluation revealed homogeneous and bead-free nanofibrous structures, with well-defined morphology and a random deposition that could accurately mimic the extracellular matrix of native skin. The calculated average nanofiber diameters for the MC/CS blend ratios at 90/10 and 70/30 were 431.4 and 441.2 nm, respectively. The results showed that when the chitosan amount increased, larger nanofibers with narrow diameter distribution appeared. The prepared nanomaterials had a significant and close water vapor permeability of about 1735.12 and 1698.52 g per m per day for the two blend ratios of 90/10 and 70/30, respectively. The examination of swelling behavior revealed a noteworthy enhancement in hydrophilicity, a necessary attribute for improved healing efficacy. FT-IR analysis confirmed the success and the stability of the chemical crosslinking reaction between the two biopolymers before nanofiber conception. Excellent mechanical properties were acquired, based on the chitosan content. Both developed nanofiber scaffolds exhibited high tensile strength and Young's modulus values. The incorporation of 30% chitosan versus 10% results in an increase in tensile strength of 11% and 14% in Young's modulus. Therefore, we could adjust the different mechanical properties simply by varying the mixing rate of the electrospun polymers. Using epithelial HepG2 cells, viability and kinetic cell adhesion assays were assessed to obtain biological evaluation. No cytotoxicity was observed and good cytocompatibility was confirmed. Functionalized nanofiber biomaterials with different MC/CS ratios substantiated significant bactericidal effectiveness against Gram-positive and Gram-negative bacterial culture strains. The novel functional electrospun wound dressing scaffold demonstrated effective and promising biomedical performance, healing both acute and chronic wounds.

Abstract:Braided polyamide sutures are frequently used in dermatologic surgery for wound closure. However, braided sutures promote bacteria proliferation. In order to prevent wound complications due to this effect, antibacterial sutures should be used. The main objective of this study is the development of new non-absorbable antibacterial polyamide braided suture. This paper suggests new coating process that leads to obtain suture uniformly covered by antibacterial film enclosing chitosan, which is known for its antibacterial benefit. Mechanical properties and surface morphology of developed sutures were investigated by using mechanical tests. Sutures surfaces were also examined by scanning electron microscope, to perceive spreading of coating product on suture surface. In order to identify potential reactions between chemical compounds present in coating solution and suture material, sutures were analyzed by ATR-IF spectroscopy. It has been demonstrated that many eventual bonds between compounds present in coating solutions and polyamide macromolecular chain may occur. The existence of these bonds implies the fixation of biopolymer coating on suture surface. It has been demonstrated that uniform surface may be obtained by progressively applying coating solution containing little amount of chitosan on suture surface. We have also found that developed coating process has not affected mechanical properties of suture, which still meet United States Pharmacopeia requirement. Finally, antibacterial effects against four colonies, very widespread in hospitals, were studied. Prominent antibacterial effects of braided polyamide suture against two gram-positive ( S Aureus, S epidermidis) and two gram-negative ( E coli and P aeruginosa) colonies are presented. Optimal result of best properties is obtained by applying three layers of biopolymer coating comprising 1% chitosan and 10% citric acid. The new developed suture coating process appears as a promising method for obtaining important antibacterial effect with smooth suture surface.

Abstract:The mechanical mismatch between soft hydrated tissues and sutures has become a common negative impact on wound healing process. A novel method of coating multilayer polymer shells is thus reported to improve the mechanical performance of hydrogel sutures. It is suitable for tissue patching and shows advantages of convenient, efficient, and biosafety. Specifically, a precursor hydrogel (Cu@CMC) consisted of carboxymethyl chitosan and copper modified by carbon dots was used as the inner sheath, and then bonding the precursor hydrogel sheath with toughening polyethylene glycol network by anchoring sites composited from rigid chitosan shell integrated a whole structure. Subsequently, the whole system was soaked with EtOH, and rapid dehydration of EtOH was used to accelerate the entanglement process between the two coatings by constricting the molecular chains. Finally, an ideal suture (Cu-fiber) with both toughness and rigidness was obtained. The data showed that the tensile strength and biosafety of the hydrogel sutures prepared by the new strategy were significantly improved, and the skin, liver and vessel of rodents can be sutured without secondary damage. Moreover, it can inhibit inflammation response and promote the healing process of skin wound, indicating that the Cu-fiber will become a great candidate for tissue patching.

Abstract:Surgical sutures are essential in wound closure, with different wound sites demanding sutures with specific mechanical properties and biodegradation rates. While silk sutures have a rich history of clinical use, they are typically regarded as nonabsorbable due to their slow degradation in the human body. In this study, we present the development of an absorbable surgical silk suture (ASS) that features controllable biodegradability and mechanical properties, along with excellent biocompatibility and antibacterial attributes. The ASS is created through a straightforward enzymatic hydrolysis process using proteinase K on degummed braided silk sutures. Additionally, we enhance the mechanical properties of the ASS by applying a regenerated silk fibroin (RSF) coating technology, which uses vapor treatment under water and methanol atmospheres. This combination of enzymatic hydrolysis and RSF coating technology allows for the customization of ASS's biodegradation rates while ensuring that the suture diameter and tensile strength comply with the United States Pharmacopeia (USP) standards for absorbable sutures. This innovative approach addresses the dynamic needs of medical professionals and patients alike, providing a novel solution for wound closure.

Abstract:Bioinspired membranes as extracellular matrices are desired for skin regeneration. Herein, we fabricate four systems of fibrillar membranes composed of polycaprolactone (PCL), aminolyzed PCL, denatured collagen type I (C), and the blend of collagen hydrolysate types I and III (bC) or elastin (E), fabricated by the electrospinning process. The superficial features by SEM and optical profilometry, mechanical properties by puncture strength assays, and biological evaluation with a human keratinocyte cell line (HaCaT) for one day of aPCL6bCE (aPCL/collagen hydrolysate type I and III/Elastin), aPCL6CE (aPCL/denaturated Col I/elastin), aPCL6bC (aPCL/collagen hydrolysate type I and III), and PCL6bCE (PCL/collagen hydrolysate type I and III/Elastin) membranes are described. The aPCL6bCE and aPCL6CE fibers resulted in better structural properties for culturing cells, while aPCL6bC and PCL6bCE membranes have shown lower viability for keratinocytes, probably due to their different chemical surface and nanotopography. The roughness of these materials varied from 4.53 to 114.4 μm, depending on their diameter values. Puncture strength results were not significantly different within 2.25–2.66 MPa. Our results suggest that aPCL6bCE and aPCL6CE fibers have potential applications in skin tissue engineering, where the synergy among aminolyzed PCL and E seems to be a prerequisite to developing such activity.
Biodegradable Silk Fibroin Matrices for Wound Closure in a Human 3D Ex Vivo Approach

Abstract:In this study, the potential of silk fibroin biomaterials for enhancing wound healing is explored, focusing on their integration into a human 3D ex vivo wound model derived from abdominoplasties. For this purpose, cast silk fibroin membranes and electrospun nonwoven matrices from Bombyx mori silk cocoons were compared to untreated controls over 20 days. Keratinocyte behavior and wound healing were analyzed qualitatively and quantitatively by histomorphometric and immune histochemical methods (HE, Ki67, TUNEL). Findings reveal rapid keratinocyte proliferation on both silk fibroin membrane and nonwoven matrices, along with enhanced infiltration in the matrix, suggesting improved early wound closure. Silk fibroin membranes exhibited a significantly improved early regeneration, followed by nonwoven matrices (p < 0.05) compared to untreated wounds, resulting in the formation of multi-layered epidermal structures with complete regeneration. Overall, the materials demonstrated excellent biocompatibility, supporting cell activity with no signs of increased apoptosis or early degradation. These results underscore silk fibroin's potential in clinical wound care, particularly in tissue integration and re-epithelialization, offering valuable insights for advanced and—as a result of the electrospinning technique—individual wound care development. Furthermore, the use of an ex vivo wound model appears to be a viable option for pre-clinical testing.

Abstract:Recent advancements in the field of biomedical engineering have underscored the pivotal role of biodegradable materials in addressing the challenges associated with tissue regeneration therapies. The spectrum of biodegradable materials presently encompasses ceramics, polymers, metals, and composites, each offering distinct advantages for the replacement or repair of compromised human tissues. Despite their utility, these biomaterials are not devoid of limitations, with issues such as suboptimal tissue integration, potential cytotoxicity, and mechanical mismatch (stress shielding) emerging as significant concerns. To mitigate these drawbacks, our research collective has embarked on the development of protein-based composite materials, showcasing enhanced biodegradability and biocompatibility. This study is dedicated to the elaboration and characterization of an innovative suture fabricated from human serum albumin through an extrusion methodology. Employing a suite of analytical techniques—namely tensile testing, scanning electron microscopy (SEM), and thermal gravimetric analysis (TGA)—we endeavored to elucidate the physicochemical attributes of the engineered suture. Additionally, the investigation extends to assessing the influence of integrating biodegradable organic modifiers on the suture's mechanical performance. Preliminary tensile testing has delineated the mechanical profile of the Filament Suture (FS), delineating tensile strengths spanning 1.3 to 9.616 MPa and elongation at break percentages ranging from 11.5 to 146.64%. These findings illuminate the mechanical versatility of the suture, hinting at its applicability across a broad spectrum of medical interventions. Subsequent analyses via SEM and TGA are anticipated to further delineate the suture's morphological features and thermal resilience, thereby enriching our comprehension of its overall performance characteristics. Moreover, the investigation delves into the ramifications of incorporating biodegradable organic constituents on the suture's mechanical integrity. Collectively, the study not only sheds light on the mechanical and thermal dynamics of a novel suture material derived from human serum albumin but also explores the prospective enhancements afforded by the amalgamation of biodegradable organic compounds, thereby broadening the horizon for future biomedical applications.

Mechanical fabrication and evaluation of bioresorbable barbed sutures with different barb geometries

Abstract:Bioresorbable polymeric sutures are gaining interest from surgeons and patients as they reduce surgical stress and trauma. This study involves two bioresorbable polymers, namely, catgut and poly(4-hyrdorxybutyrate) (P4HB) that are used widely in cosmetic procedures. P4HB barbed sutures are favorably used in rhytidectomy (micro-facelifts) procedures while catgut sutures are widely used for external wound closure after surgical interventions. This study involves the mechanical fabrication of catgut and P4HB barbed sutures and compares their mechanical and anchoring properties. Barbed sutures were fabricated with two different barb geometries namely, straight and curved barbs. The mechanical properties were evaluated via tensile testing, and the anchoring performance was studied by means of a suture-tissue pull-out protocol using porcine dermis tissue which was harvested from the medial dorsal site. The fabricated barbed sutures of both materials showed a similar trend compared to non-barbed sutures of decreases in failure stress, strain at failure, and work to rupture or toughness which was reduced by about 70%. At the same time there was a 15% increase in the initial modulus or stiffness of the barbed sutures. The pull-out force for the barbed sutures with straight barbs was similar for both P4HB (5.04±0.8N) and catgut (4.47±3.8N), and as expected, were higher than that of non-barbed sutures of the same size. It was also observed that barbed sutures with curved barbs also required a higher pull-out force than those sutures with straight barbs. It was concluded that by barbing sutures with different barb shapes and geometries, a range of barbed suture products could be fabricated, each meeting the closure requirements for different types of tissue and therefore being suitable for different surgical procedures.
Fabrication of Chitosan/Silk Fibroin Composite Nanofibers for Wound-dressing Applications

Abstract:Chitosan, a naturally occurring polysaccharide with abundant resources, has been extensively exploited for various biomedical applications, typically as wound dressings owing to its unique biocompatibility, good biodegradability and excellent antibacterial properties. In this work, composite nanofibrous membranes of chitosan (CS) and silk fibroin (SF) were successfully fabricated by electrospinning. The morphology of electrospun blend nanofibers was observed by scanning electron microscopy (SEM) and the fiber diameters decreased with the increasing percentage of chitosan. Further, the mechanical test illustrated that the addition of silk fibroin enhanced the mechanical properties of CS/SF nanofibers. The antibacterial activities against Escherichia coli (Gram negative) and Staphylococcus aureus (Gram positive) were evaluated by the turbidity measurement method; and results suggest that the antibacterial effect of composite nanofibers varied on the type of bacteria. Furthermore, the biocompatibility of murine fibroblast on as-prepared nanofibrous membranes was investigated by hematoxylin and eosin (H&E) staining and MTT assays in vitro, and the membranes were found to promote the cell attachment and proliferation. These results suggest that as-prepared chitosan/silk fibroin (CS/SF) composite nanofibrous membranes could be a promising candidate for wound healing applications.

Abstract:Interpolyelectrolyte complexes (IPECs) of polysaccharides are multifunctional polymer materials that improve the mechanical and physicochemical properties of individual polysaccharides. In this study, highly porous (>90%) materials based on IPECs of versatile natural polysaccharides, chitosan (30 and 1,200 kDa) and pectin, are obtained by freeze‐drying technique. To enhance the interaction between chitosan and pectin macromolecules, the latter are chemically functionalized with dialdehyde groups. The chitosan‐/aldehyde‐functionalized pectin (Chit/AF‐Pect) polyelectrolyte complex sponges obtained are characterized using SEM, FTIR spectroscopy, and TGA. The swelling capacity study reveals a higher swelling ratio of IPEC sponges with an increase in both the molecular weight and content of chitosan: for Chit30/AF‐Pect, the swelling ratio rises from 327% to 480%, while for Chit1200/AF‐Pect, from 681% to 1,066%. Additionally, the in vitro degradation test demonstrates higher stability of Chit1200/AF‐Pect sponges in comparison with those of Chit30/AF‐Pect: after 4 days of incubation, the weight losses are found to be 9%–16% and 18%–41%, respectively. The cytotoxicity study shows that Chit30/AF‐Pect sponges are noncytotoxic, with cell viability values >70%. Furthermore, the Chit30/AF‐Pect sponges, obtained at chitosan:pectin weight ratio of 5:1, exhibit bactericidal activity against Escherichia coli BIM B‐984 G, Pseudomonas aeruginosa BIM B‐807 G, Staphylococcus aureus BIM B‐1841, and slightly inhibit the growth of Enterococcus faecalis BIM B‐1530 G. These findings indicate that the obtained Chit30/AF‐Pect sponges can be used to create wound dressings for wound healing applications.
