Abstract:Microwave thermotherapy (MWTT), as a treatment for tumors, lacks specificity and requires sensitizers. Most reported microwave sensitizers are single-metal organic frameworks (MOFs), which must be loaded with ionic liquids to enhance the performance in MWTT. Meanwhile, MWTT is rarely combined with other treatment modalities. Here, we synthesized a novel Fe-Cu bimetallic organic framework FeCuMOF (FCM) by applying a hydrothermal method and further modified it with methyl polyethylene glycol (mPEG). The obtained FeCuMOF@PEG (FCMP) showed remarkable heating performance under low-power microwave irradiation; it also acted as a novel nanoparticle enzyme to catalyze hydrogen peroxide decomposition, producing abundant reactive oxygen species (ROS) to deplete glutathione and prevent ROS clearance from tumor cells during chemodynamic treatment. The FCMP was biodegradable and demonstrated excellent biocompatibility, allowing it to be readily metabolized without causing toxic effects. Finally, it was shown to act as a suitable agent for T2 magnetic resonance imaging (MRI) in vitro and in vivo. This new bimetallic nanostructure could successfully realize two tumor treatment modalities (MWTT and chemodynamic therapy) and dual imaging modes (MRI and microwave thermal imaging). Our findings represent a breakthrough for integrating the diagnosis and treatment of tumors and provides a reference for developing new microwave sensitizers.

A multifunctional nanoplatform for improving microwave hyperthermia by a combination therapy of vessel disruptive agent and immune modulator

Abstract:Microwave (MW) hyperthermia is one of the safest and most efficient minimally invasive tumor treatment methods, it is restricted by the bottlenecks of the heat sink effect and ineffective immune activation. Herein, a multifunctional nano platform with the load of nano immune modulator bimetallic metal-organic framework (BM), tumor vessel destructive agent and prodrug for gas production is developed for improving MW hyperthermia. Specifically, the combretastatin A4 phosphate (CA4P) was a vessel destructive agent to reduce MW heat loss by destructing the tumor blood vessel. Moreover, the as designed BM can scavenge the endogenic reactive oxygen species, which is conducive to hydrogen sulfide gas (H2S) that produced by bismuth sulfide (Bi2S3) to activate immune cells. Our in vivo experimental results demonstrate the destruction of tumor blood vessels coupled with the activated immune system results in the remarkable antitumor effect. This study provides an efficient strategy to improve MW hyperthermia by a combination of vasculature-targeting therapy with systemic immunity.
Microwave-activated Cu-doped zirconium metal-organic framework for a highly effective combination of microwave dynamic and thermal therapy

Abstract:Percutaneous microwave ablation (PMA) is a thermoablative method used as a minimally invasive treatment for liver cancer. However, the application of PMA is limited by its insufficient ROS generation efficiency and thermal effects. Herein, a new microwave-activated Cu-doped zirconium metal-organic framework (MOF) (Cu Zr MOF) used for enhanced PMA has a significantly improved microwave sensitizing effect. Owing to the strong inelastic collisions between ions confined in numerous micropores, CuZr MOF has strong microwave sensitivity and high thermal conversion efficiency, which can significantly improve microwave thermal therapy (MTT). Moreover, because of the existence of Cu 2+ ions, a further benefit of CuZr MOF is their Fenton-like activity, in particular, microwaves used as an excitation source for microwave dynamic therapy (MDT) can improve the Fenton-like reaction to maximize the synergistic effectiveness of cancer therapy. Importantly, Cu Zr MOF can inhibit the production of heat shock proteins (HSPs) by producing abundant ROS to enhance tumor destruction. Mechanistically, we found that Cu Zr MOF + MW treatment modulates ferroptosis-mediated tumor cell death by targeting the HMOX1/GPX4 axis. In summary, this study develops a novel CuZr MOF microwave sensitizer with great potential for synergistic treatment of liver cancer by MTT and MDT.

Synergistic Photothermal Tumor Immunotherapy by 1-MT Based on Zeolitic Imidazolate Framework-8 with pH-High Sensitivity

Abstract:Background: A successful immune response against tumors depends on various cellular processes. Hence, there is an urgent need to construct a proficient nanoplatform for immunotherapy that can concurrently regulate the activities of various cells participating in the immune process. We have developed zeolitic imidazolate framework-8 (ZIF-8) formula, with good pH sensitivity, which is conducive to the release of drugs in the tumor site (acidic environment) and significantly improves immunotherapy. This is achieved through the coordinated action of different therapeutic agents, such as the photothermal agent polydopamine (PDA), the chemodrug camptothecin (CPT), and the immunomodulator 1-methyl-D-tryptophan (1-MT). Materials and methods: In this study, we evaluated the antitumor effect of PDA/(CPT + 1-MT) @ZIF-8 (PCMZ) nanoparticles (NPs) in vitro and in vivo and investigated the molecular mechanism of PCMZ NPs in tumor suppression via photothermal-chemo-immunotherapy. Results: MTT and Annexin V-FITC/PI double staining apoptosis test showed that PCMZ NPs could induce apoptosis of 4T1 cell, and PCMZ NPs could cause 4T1 cell necrosis under 808 nm laser irradiation. The objective is to establish a unilateral breast cancer model in mice and investigate the effect of PCMZ NPs on tumor growth and tumor suppression in tumor bearing mice. The results showed that PCMZ NPs showed good heating effect in vivo and effectively inhibited tumor growth under 808 nm laser irradiation. In addition, PCMZ NPs could induce the immunogenic death of tumor cells, promote the maturation of DCs, inhibit IDO pathway, and finally differentiate T cells into cytotoxic T cells and helper T cells, so as to effectively activate the anti-tumor immune response. Conclusion: The PCMZ NPs, possessing good photothermal conversion capabilities due to join of PDA, effectively overcome two main challenges in immunotherapy: insufficient stimulation of the immune response and evasion of the immune system. This provides a robust platform against invasive cancer and recurrent tumors.

Abstract:Developing high-performance nanomedicines to enhance antitumor efficacy remains a hot point in the field of biomedicine. In this study, we designed a versatile nanocomposite (FeS₂@COF-HA/AIPH) integrating covalent organic frameworks (COF) functionalized with pyrite (FeS₂) for synergistic photothermal (PTT), chemodynamic (CDT), thermodynamic (TDT) therapies, and immunotherapy. The superior photothermal effects and catalytic capabilities of FeS₂@COF enabled a minimally invasive PTT/CDT combination. The nanoplatform, with its mesoporous structure, also served as a drug delivery system, encapsulating the thermos-decomposable initiator AIPH. The hyaluronic acid (HA) coating not only improved tumor-targeting efficiency but also prevented nonspecific AIPH release. Under near-infrared (NIR) irradiation, the localized hyperthermia triggered AIPH decomposition, generating toxic alkyl radicals (•R) for TDT, further enhancing CDT efficiency. The combination of PTT, CDT, TDT, and immunotherapy led to potent antitumor effects with minimal systemic toxicity, both in vitro and in vivo. Notably, the nanoplatform effectively reprogrammed tumor-associated macrophages (TAMs) from an M2 to M1 phenotype, boosting antitumor immunity. This multifunctional platform thus offers a promising strategy for integrated PTT, CDT, TDT, and immune activation in tumor therapy.

Abstract:Efforts have been made to improve the therapeutic efficiency of tumor treatments, and metal–organic frameworks (MOFs) have shown excellent potential in tumor therapy. Monotherapy for the treatment of tumors has limited effects due to the limitation of response conditions and inevitable multidrug resistance, which seriously affect the clinical therapeutic effect. In this study, we chose to construct a multiple cascade synergistic tumor drug delivery system MIL−101(Fe)−DOX−TCPP−MnO2@PDA−Ag (MDTM@P−Ag) using MOFs as drug carriers. Under near−infrared (NIR) laser irradiation, 5,10,15,20−tetrakis(4−carboxyphenyl)porphyrin (TCPP) and Ag NPs loaded on MDTM@P−Ag can be activated to generate cytotoxic reactive oxygen species (ROS) and achieve photothermal conversion, thus effectively inducing the apoptosis of tumor cells and achieving a combined photodynamic/photothermal therapy. Once released at the tumor site, manganese dioxide (MnO2) can catalyze the decomposition of hydrogen peroxide (H2O2) in the acidic microenvironment of the tumor to generate oxygen (O2) and alleviate the hypoxic environment of the tumor. Fe3+/Mn2+ will mediate a Fenton/Fenton−like reaction to generate cytotoxic hydroxyl radicals (·OH), while depleting the high concentration of glutathione (GSH) in the tumor, thus enhancing the chemodynamic therapeutic effect. The successful preparation of the tumor drug delivery system and its good synergistic chemodynamic/photodynamic/photothermal therapeutic effect in tumor treatment can be demonstrated by the experimental results of material characterization, performance testing and in vitro experiments.

Abstract:Imaging-guided therapy has considerable potential in tumor treatment. Different treatments have been integrated to realize combinational tumor therapy with improved therapeutic efficiency. Herein, the conventional metal-organic framework (MOF) MIL-100 is utilized to load curcumin with excellent encapsulation capacity. Polydopamine-modified hyaluronic acid (HA-PDA) is coated on the MIL-100 surface to construct engineering MOF nanoparticles (MCH NPs). The HA-PDA coating not only improves the dispersibility and stability of NPs but also introduces a tumor-targeting ability to this nanosystem. A two-stage augmented photothermal conversion capability is introduced to this nanosystem by encapsulating curcumin in MIL-100 pores and then coating HA-PDA on the surface, which confer the MCH NPs with strong photothermal conversional efficiency. After being intravenously injected into xenograft HeLa tumor-bearing mice, MCH NPs prefer to accumulate at the tumor site and achieve photoacoustic imaging-guided chemo-/photothermal combinational tumor therapy, generating nearly complete tumor ablation. Engineering MOFs is an efficient platform for imaging-guided combinational tumor therapy, as confirmed by in vitro and in vivo evaluations.

Abstract:Abstract Backgrounds Microwave sensitization nanoplatform, integrating multiple functional units for improving tumor selectivity, is of great significance for clinical tumor microwave treatment. Lanthanide europium metal organic framework (EuMOF) is expected to be a theranostic nanoplatform owing to its unique luminescent and microwave sensitization properties. However, it is difficult to be applied to complicated biological systems for EuMOF due to its rapid degradation induced by the solvent molecular and ionic environment. In this work, a luminescent EuMOF nanocomposite (EuMOF@ZIF/AP-PEG, named EZAP) was designed, which brought the multifunctional characteristics of microwave sensitization, fluorescence imaging and drug loading. Results Lamellar EuMOF was synthesized by a hydrothermal method. Through the charge adsorption mechanism, the zeolite imidazole framework (ZIF) structure was intensively assembled on the surface of EuMOF to realize the protection. Then, through in-situ Apatinib drug loading and PEG modification, EZAP nanocomposite was finally obtained. Apatinib (AP) was a kind of chemotherapy drug approved by Food and Drug Administration for targeted therapy of tumors. PEG modification increased long-term circulation of EZAP nanocomposite. The physical and chemical structure and properties of EuMOF@ZIF (EZ) were systematically represented, indicating the successful synthesis of the nanocomposite. The toxic and side effects were negligible at a safe dose. The growth of human liver cancer cells and murine liver cancer cells in vitro was significantly inhibited, and the combined microwave-thermal therapy and chemotherapy in vivo achieved high anti-cancer efficacy. Moreover, EZAP nanocomposite possessed bright red fluorescence, which can be applied for tumor imaging in tumor-bearing mice in vivo. Conclusion Therefore, EZAP nanocomposite showed high microwave sensitization, excellent fluorescence properties and outstanding drug loading capacity, establishing a promising theranostic nanoplatform for tumor therapy and fluorescence imaging. This work proposes a unique strategy to design for the first time a multifunctional nanoplatform with lanthanide metal organic frameworks for biological applications in tumor therapy and diagnosis. Abstract:When microwave hyperthermia (MWH) array antenna technology is used to treat breast cancer, how to effectively target and heat deep tumors and reduce thermal damage to healthy tissues is still a challenge in clinical applications. In this study, the synergistic MWH effect of conformal-array antennas (CAA) and a novel microwave-thermal-sensitive nanomaterial (MTSN) was investigated for the treatment of subcutaneous deep in situ breast cancer. At the beginning of the study, the thermal damage score was used to evaluate the therapeutic efficacy of the CAA. It was found that although array antenna technology can achieve effective heating of deep tumors, its damage to healthy tissues is unacceptable. Consequently, we developed a novel MTSN, ZIF-8@HA, whose unique structure significantly enhanced the absorption of MW energy and MW thermal conversion efficiency in the local tumor region. The MW thermal conversion efficiency of ZIF-8@HA achieved was as high as 46.46% in in vivo MW heating experiments. In the phantom that simulates the electromagnetic environment of the human body, the microwave-thermal sensitization (MTS) effect is also significant, and the reduction in the average thermal damage score of healthy tissues by more than 10% was verified through measurements using the coaxial probe method and COMSOL simulations. Cellular experiments confirmed that the combination of ZIF-8@HA and MW irradiation could significantly reduce the survival rate of tumor cells. In addition, cross-tissue MW heating experiments revealed the advantages of ZIF-8@HA combined with the CAA. Finally, phantom experiments confirmed that the synergistic use of the CAA with ZIF-8@HA significantly accelerated the local heating rate of deep tumors, reduced the time required for the tumor region to achieve 100% thermal damage, and effectively minimized the thermal damage to healthy tissues.

Abstract:Microwave ablation (MWA) holds promise as a tumor treatment modality, yet incomplete ablation with low-frequency MWA (iMWA) remains a significant obstacle to survival, while the high-frequency MWA poses safety concerns. To address these challenges, we introduce a novel bimetallic iMWA nanosensitizer, CFT-NPs, constructed via metal-coordination interactions between tannic acid (TA) and iron/calcium dual metal ions (Fe3+/Ca2+). The Fe3+/Ca2+ in CFT-NPs work in harmony to convert electromagnetic energy into heat, thereby bolstering the effectiveness of iMWA while maintaining good biosafety. Furthermore, the pH-triggered rupture of CFT-NPs release TA, Fe3+, and Ca2+. The liberated Fe3+ undergoes reduction by TA, and when combined with Ca2+, they trigger ferroptosis and Ca2+ overload respectively, significantly amplifying the therapeutic effects of iMWA. Utilization iMWA with CFT-NPs results in lipid oxidation, mitochondrial dysfunction, and disruption of redox balance, both in vitro and in vivo. Notably, this nanosensitizer exerts a robust antitumor effect, effectively inhibiting tumor growth and downregulating the expression of matrix metalloproteinases, which is associated with tumor metastasis. Abstract:In the framework of modified mean-field density-functional theory, effects of a uniform electric field on the interfacial properties of a model dipolar fluid [Teixeira and Telo da Gama, J. Phys.: Condens. Matter 3, 111 (1991)] are studied. Both density and orientational order-parameter profiles of the planar vapor-liquid interface are obtained as a function of the field strength. For the dipolar fluids with reduced dipole moment μ0*⩽1, we find that the field (under the condition μ0*E*/T*≪1) can shift the surface tension by few percent. We also find that the electric field actually reduces the thermodynamical surface tension but enhances the mechanical surface tension at the equimolar dividing surface. To detect these field effects on the surface tension we estimate the field strength which can be as high as 108 V/m.

Abstract:Currently, precise ablation of tumors without damaging the surrounding normal tissue is still an urgent problem for clinical microwave therapy of liver cancer. Herein, we synthesized Mn-doped Ti MOFs (Mn-Ti MOFs) nanosheets by in-situ doping method and applied them for microwave therapy. Infrared thermal imaging results indicate Mn-Ti MOFs can rapidly increase the temperature of normal saline, attributing to the porous structure improving microwave-induced ion collision frequency. Moreover, Mn-Ti MOFs show higher 1O2 output than Ti MOFs under 2 W of low-power microwave irradiation due to the narrower band-gap after Mn doping. At the same time, Mn endows the MOFs with a desirable T1 contrast of magnetic resonance imaging (r2/r1 = 2.315). Further, results on HepG2 tumor-bearing mice prove that microwave-triggered Mn-Ti MOFs nearly eradicate the tumors after 14 days of treatment. Our study offers a promising sensitizer for synergistic microwave thermal and microwave dynamic therapy of liver cancer.

Abstract:The development of smart drug delivery systems (SDDSs) based on engineered nanomaterials is important for clinical applications. Nevertheless, controllable administration of chemotherapeutic drugs for deep tumors and the avoidance of side effects caused by off-targeting during delivery remain a great challenge. Herein, a stimulus-responsive system of mesoporous nanospheres (composed of Cu@Fe2C@mSiO(2)) with good magnetothermal effect is introduced into the tumor microenvironment. This system plays an important role in image-guided controllable targeted drug delivery that is independent of tumor depth. Aggregation-induced emission luminogen-based fluorescence imaging and magnetic resonance imaging were utilized since these techniques visualize the delivery process in real time. In addition, the degraded nanocarriers showed high catalytic activity for Fenton and Fenton-like reactions, upregulating the level of hydroxyl radicals (center dot OH) in cancer cells to realize chemodynamic therapy. The induced center dot OH led to the overexpression of pho-STAT3, activating the STAT3 signaling pathway, eventually inducing cancer cell apoptosis. Through metabolic monitoring, this SDDS is removed from the body after its degradation in vivo. The synergistically enhanced therapeutic effect was obtained in the chemo-chemodynamic therapy of 4T1 tumor-bearing mice, offering a platform for efficient cancer therapy with a personalized theranostic strategy. Abstract:Microwave ablation (MWA) is recognized as the mainstream choice for tumor thermal ablation, but the rapid diffusion of heat and high tumor recurrence rate are still the primary bottlenecks in the clinic application of MWA. Hexagonal tungsten oxide nanomaterials could be potentially exploited as microwave sensitizers to elevate MWA efficacy due to the inherent tunnel structure and ions intercalation characteristics. Importantly, the flexible redox states of W element make it promising candidates for chemodynamic therapy (CDT) to further improve microwave-mediated cancer treatment. Herein, multifunctional hexagonal phase NaxWO3 nanocrystals modified with polyvinylpyrrolidone are prepared. Such NaxWO3 nanocrystals could not only function as microwave sensitizers to produce microwave heating effects for cancer cell killing, but also act as nano-catalyst to decompose hydrogen peroxide into highly oxidative hydroxyl radicals and simultaneously consume intracellular glutathione for an enhanced CDT effect. In addition, the hyperthermia generated by microwave irradiation could further upgrade the efficacy of CDT, achieving a combined effect of MWA and heat-amplified CDT. Notably, NaxWO3 nanocrystals enable the effective destruction of tumor tissues under microwave irradiation without inducing significant side effects to normal tissues. Consequently, our findings propose an innovative and safe nanotechnology strategy to reinforce the therapeutic outcome of MWA and CDT.
Synergistic Microwave Hyperthermia Treatment for Subcutaneous Deep In Situ Breast Cancer Using Conformal Array Antenna and Microwave-Thermal-Sensitive Nanomateria

Abstract:When microwave hyperthermia (MWH) array antenna technology is used to treat breast cancer, how to effectively target and heat deep tumors and reduce thermal damage to healthy tissues is still a challenge in clinical application. In this study, the synergistic MWH effect of Conformal-Array antenna (CAA) and novel Microwave-Thermal-Sensitive nanomaterial (MTSN) was investigated for the treatment of subcutaneous deep in situ breast cancer. At the beginning of the study, the thermal damage score was used to evaluate the therapeutic efficacy of CAA. It was found that although array antenna technology can achieve effective heating of deep tumors, its damage to healthy tissues is unacceptable. Consequently, we developed a novel MTSN, ZIF-8@HA, whose unique structure significantly enhanced the absorption of MW energy and MW thermal conversion efficiency in the local tumor region. The MW thermal conversion efficiency of ZIF-8@HA achieved a high of 46.46% in vivo MW heating experiments. In the phantom that simulates the electromagnetic environment of the human body, the microwave-thermal sensitization (MTS) effect is also significant, and the reduction in the average thermal damage score of healthy tissue by more than 10% was verified through measurements using the coaxial probe method and COMSOL simulations. Cellular experiments confirmed that the combination of ZIF-8@HA and MW irradiation could significantly reduce the survival rate of tumor cells. In addition, cross-tissue MW heating experiments revealed the advantages of ZIF-8 @ HA combined with CAA. Finally, phantom experiments confirmed that the synergistic use of CAA with ZIF-8@HA significantly accelerated the local heating rate of deep tumors, reduced the time required for the tumor region to achieve 100% thermal damage, and effectively minimized thermal damage to healthy tissues.
