Abstract:Prostaglandins (PGs) of the E series, are detectable at inflamed sites and exert pro-inflammatory actions, such as vasodilatation and potentiation of plasma exudation induced by other mediators (see Flower 1977). However, with animal models of chronic inflammation, high doses of PGEs exert anti-inflammatory actions in vivo (see Bonta & Parnham 1978). Recently, we observed that local administration of PGE,, at 1000-fold lower doses than those used by other authors, during the early phase of spongeinduced granulomatous inflammation, enhances subsequent granuloma formation, whereas administration during the later phase inhibits granuloma formation (Bonta & Parnham 1979). In fact, in rats which were deficient in the fatty acid precursors of endogenous PGs, exogenous PGEl inhibited granuloma formation when given at the low daily dose of 50ng. We now report similar proand anti-granuloma effects with low doses of PGE, (the most commonly detected PG at inflamed sites) and additionally show that the later inhibitory effects of PGE, on granuloma formation are capable of counteracting the early stimulatory effects. We have also compared the inhibitory action of PGE, with other PGs including prostacyclin (PGIJ, a novel PG (Moncada et al 1976) which has been found to exert similar actions to PGE,, including vasodilatation and increased vascular permeability (Peck & Williams 1978; Higgs et al 1978b; Murota et al 1978). Granulomatous inflammation was induced in rats, using subcutaneously implanted, carrageenan-soaked, polyether sponges (2 per rat) with indwelling cannulae (Bonta et a1 1979), and the PGs were injected either with the 1 ml 2% Na carrageenan on implantation (day 1) and/or through the cannulae (in 0.51111) at different times, as described earlier (Bonta & Parnham (Moncada et a1 1976). All rats were killed on day 8, sponges removed and granuloma formation expressed as the dry weight of tissue surrounding the sponges (Bonta et a1 1979). In some experiments, both adrenals from each rat were removed and weighed together, their weights being expressed in terms of 1OOg body weight. Table 1 shows that, while injection of PGE, (2 pg) into sponges on implantation (day 1) had no effect on granuloma formation, measured after 8 days, the same daily dose administered on days 1-3 enhanced granuloma formation after 8 days. This action is similar to the granuloma stimulation produced by PGE, (1 pg), which was thought to be secondary to an initial vasodilating action, following injection on day 1 (Bonta & Parnham 1979). Furthermore, the inhibition of 8 day granuloma formation caused by administration of PGEa on days 4-7 (Table 1) was identical to the effect of PGE, administered over the same period (Bonta & Parnham 1979). It should be noted that the increase in control (saline-treated) granuloma weights with progressively later treatment periods, is coinci dental, since, in a large number of control experiments, granuloma weight was unrelated to timing of saline treatment, despite considerable variation in granuloma weights (Bonta et al 1979). Furthermore, it was previously found that this variation in granuloma weights did not alter the effects of added PGEl (Bonta & Parnham 1979). Thus, PGE, injected on day 1 enhanced granuloma weight when control values ranged from 0.32 to 0.78 g and when injected on days 4-7, PGE, inhibited granuloma weight in experiments in which control values ranged from 0.35 to 1.03 g. Abstract:Abstract Background Titanium and its alloys have been widely employed for bone tissue repair and implant manufacturing. The rapid development of three-dimensional (3D) printing technology has allowed fabrication of porous titanium scaffolds with controllable microstructures, which is considered to be an effective method for promoting rapid bone formation and decreasing bone absorption. The purpose of this systematic review was to evaluate the osteogenic potential of 3D-printed porous Ti6Al4V (Ti64) scaffold for repairing long bone defects in animal models and to investigate the influential factors that might affect its osteogenic capacity. Methods Electronic literature search was conducted in the following databases: PubMed, Web of Science, and Embase up to September 2021. The SYRCLE's tool and the modified CAMARADES list were used to assess the risk of bias and methodological quality, respectively. Due to heterogeneity of the selected studies in relation to protocol and outcomes evaluated, a meta-analysis could not be performed. Results The initial search revealed 5858 studies. Only 46 animal studies were found to be eligible based on the inclusion criteria. Rabbit was the most commonly utilized animal model. A pore size of around 500–600 μm and porosity of 60–70% were found to be the most ideal parameters for designing the Ti64 scaffold, where both dodecahedron and diamond pores optimally promoted osteogenesis. Histological analysis of the scaffold in a rabbit model revealed that the maximum bone area fraction reached 59.3 ± 8.1% at weeks 8–10. Based on micro-CT assessment, the maximum bone volume fraction was found to be 34.0 ± 6.0% at weeks 12. Conclusions Ti64 scaffold might act as a promising medium for providing sufficient mechanical support and a stable environment for new bone formation in long bone defects. Abstract:Critical-sized bone defect repair remains a substantial challenge in clinical settings and requires bone grafts or bone substitute materials. However, existing biomaterials often do not meet the clinical requirements of structural support, osteoinductive property, and controllable biodegradability. To treat large-scale bone defects, the development of three-dimensional (3D) porous scaffolds has received considerable focus within bone engineering. A variety of biomaterials and manufacturing methods, including 3D printing, have emerged to fabricate patient-specific bioactive scaffolds that possess controlled micro-architectures for bridging bone defects in complex configurations. During the last decade, with the development of the 3D printing industry, a large number of tissue-engineered scaffolds have been created for preclinical and clinical applications using novel materials and innovative technologies. Thus, this review provides a brief overview of current progress in existing biomaterials and tissue engineering scaffolds prepared by 3D printing technologies, with an emphasis on the material selection, scaffold design optimization, and their preclinical and clinical applications in the repair of critical-sized bone defects. Furthermore, it will elaborate on the current limitations and potential future prospects of 3D printing technology. STATEMENT OF SIGNIFICANCE: 3D printing has emerged as a critical fabrication process for bone engineering due to its ability to control bulk geometry and internal structure of tissue scaffolds. The advancement of bioprinting methods and compatible ink materials for bone engineering have been a major focus to develop optimal 3D scaffolds for bone defect repair. Achieving a successful balance of cellular function, cellular viability, and mechanical integrity under load-bearing conditions is critical. Hybridization of natural and synthetic polymer-based materials is a promising approach to create novel tissue engineered scaffolds that combines the advantages of both materials and meets various requirements, including biological activity, mechanical strength, easy fabrication and controllable degradation. 3D printing is linked to the future of bone grafts to create on-demand patient-specific scaffolds.

Abstract:Every year, millions of people around the world undergo bone graft and artificial prosthesis transplants which engenders the repair and/or replacement of native bone for treating bone defects and injury. As a result, bone has become the second most transplanted tissue globally and there is a growing need for advances in bone tissue engineering (BTE) to ensure improved quality of life. The advent of 3D bone scaffolds has led to a paradigm shift in this field. These scaffolds act as an extra cellular matrix (ECM) providing a 3D environment for cell adhesion, proliferation, and differentiation. The scaffolds so fabricated, must meet the mechanical and biological criteria for successful clinical translation. Initial iterations of these scaffolds were not constructed with precise geometries. This review looks at the various advances in scaffold designs, materials and 3D printing techniques employed, which enable us to fabricate scaffolds with tailored architecture and control its functionality. These advances allow us to optimize the parameters in the fabrication of these scaffolds, such that we can create structures with desired characteristics like controllable biodegradability using the least amount of material possible along with a precise pore size and high permeability for purposes of osteo-induction and osseointegration. This is followed by the computational analysis carried out on the samples to ensure they are suitable for a given application. Finally, this paper talks about regulatory hurdles (ethical, legal and social) faced by researchers, in addition to the various technical challenges, when it comes to the real-life implementation of this technology.

Abstract:The need for bone grafts is tremendous, and that leads to the use of autograft, allograft, and bone graft substitutes. The biology of the bone is quite complex regarding cellular composition and architecture, hence developing a mineralized connective tissue graft is challenging. Traditionally used bone graft substitutes including metals, biomaterial coated metals and biodegradable scaffolds, suffer from persistent limitations. With the advent and rise of additive manufacturing technologies, the future of repairing bone trauma and defects seems to be optimistic. 3D printing has significant advantages, the foremost of all being faster manipulation of various biocompatible materials and live cells or tissues into the complex natural geometries necessary to mimic and stimulate cellular bone growth. The advent of new-generation bioprinters working with high-precision, micro-dispensing and direct digital manufacturing is aiding in ground-breaking organ and tissue printing, including the bone. The future bone replacement for patients holds excellent promise as scientists are moving closer to the generation of better 3D printed bio-bone grafts that will be safer and more effective. This review aims to summarize the advances in scaffold fabrication techniques, emphasizing 3D printing of biomimetic bone grafts.

Abstract:One thousand pregnant women belonging to upper and upper-middle socio-economic class admitted at term to Aga Khan Maternity Home, Karimabad weie screened for HBs Ag by reversed passive haemagglutination (Green Cross Corporation, Japan).HBs Ag positive sera were tested for HBe Ag by passive baemagglutination technique. Sera obtained from venous blood samples of babies born to all HBs Ag carrier mothers were tested for HBs Ag. Follow up of babies could not be done due to non cooperation of mothers and inadequate addresses. Of 1000 pregnant women tested 39 (3.9%) were HBsAg positive and 6(15.4%) of these were HBeAg positive. None of the babies born to HBs Ag carrier mothers had HBs antigenaemia at birth.

Abstract:Three-dimensional (3D) printing technology is developing as a dominant tool for biomedical engineering by supporting 3D cell culture within compound 3D biomimetic buildings. Biomaterial and Tissue engineering has developed as a favorable alternative method in the treatment of bones, teeth, and organs. This paper summarizes the current research status and attention of the 3D biomaterials scaffolds in bone tissue engineering applications. Several 3D scaffolds fabricated from several types of biodegradable materials have been established. The crucial topics of 3D printing techniques are recognized and deliberated with the future improvement of innovative biomaterials. There has been a prompt development in the applications of 3D printing in engineering customized implants, drug delivery devices, prostheses, and 3D scaffolds for regenerative medicine and tissue engineering. Medical 3D printing technologies are classified into the following categories: Fused Deposition Modeling (FDM), Extrusion-based 3D bioprinting, Selective Laser Sintering (SLS)/Selective Laser melting (SLM), Electron Beam Manufacturing (EBM), Stereolithography (SLA) and Digital Light Processing (DLP) printing techniques, and their clinical applications, different types of biomaterials currently used by researchers, and key limitations are discussed in detail. Iris localization using colour segmentation and circular Hough transform

Abstract:This paper presents the enhancement technique of iris localization using circular Hough transform and colour segmentation. Circular Hough transform localized the inner and outer boundaries of the iris using images from each colour channels namely; R, G and B and H, S and V. Combination of the colour channels was also experimented. The experiments were conducted on the iris images obtained from healthy women free from Human Papilloma Virus (HPV). The result from the enhancement technique shows that when images of gray scale and combination of S and V channels are used, both outer and inner boundaries of the irises able to achieve 100% correct localization.

Abstract:Proceedings of the Institution of Mechanical Engineers, Part L: Journal of Materials: Design and Applications, Ahead of Print. When treating orthopaedic damage or illness and accidental fracture, bone grafting remains the gold standard of treatment. In cases where this approach doesn't seem achievable, bone tissue engineering can offer scaffolding as a substitute. Defective and fractured bone tissue is extracted and substituted with porous scaffold structures to aid in the process of tissue regeneration. Three-dimensional bioprinting has demonstrated enormous promise in recent years for producing scaffold structures with the necessary capabilities. In order to create composite biomaterial inks for three-dimensional bioprinting, four different materials were combined such as silk fibroin, bone particles (B), synthetic biopolymer poly (ε-caprolactone), and Fenugreek (F). These biomaterials were mixed together in certain proportion to develop a silk fibroin + bovine bone + polycaprolactone + fenugreek powder composites biomaterial which was later three-dimensional bioprinted to fabricated composite bio-scaffold. The biomechanical, structural, and biological elements of the manufactured composite scaffolds were characterized in order to determine their suitability as a possible biomaterial for the production of bone tissue. The in vitro bioactivity of the composite scaffolds was assessed in the simulated body fluids, and the swelling and degradation characteristics of the two developed scaffolds were analyzed separately over time. The results showed that the mechanical durability of the composite scaffolds was enhanced by the bovine bone particles, up to a specific concentration in the silk fibroin matrix. Furthermore, the incorporation of bone particles improved the bioactive composite scaffolds' capacity to generate hydroxyapatite in vitro. The combined findings show that the three-dimensional printed bio-composites scaffolds have the required mechanical strength and may be applied to regeneration of bone tissue and restoration, since they resemble the characteristics of native bone.

Abstract:Computer-aided design/computer-aided manufacturing and 3-dimensional (3D) printing techniques have revolutionized the approach to bone tissue engineering for the repair of craniomaxillofacial skeletal defects. Ample research has been performed to gain a fundamental understanding of the optimal 3D-printed scaffold design and composition to facilitate appropriate bone formation and healing. Benchtop and preclinical, small animal model testing of 3D-printed bioactive ceramic scaffolds augmented with pharmacological/biological agents have yielded promising results given their potential combined osteogenic and osteoinductive capacity. However, other factors must be evaluated before newly developed constructs may be considered analogous alternatives to the "gold standard" autologous graft for defect repair. More specifically, the 3D-printed bioactive ceramic scaffold's long-term safety profile, biocompatibility, and resorption kinetics must be studied. The ultimate goal is to successfully regenerate bone that is comparable in volume, density, histologic composition, and mechanical strength to that of native bone. In vivo studies of these newly developed bone tissue engineering in translational animal models continue to make strides toward addressing regulatory and clinically relevant topics. These include the use of skeletally immature animal models to address the challenges posed by craniomaxillofacial defect repair in pediatric patients. This manuscript reviews the most recent preclinical animal studies seeking to assess 3D-printed ceramic scaffolds for improved repair of critical-sized craniofacial bony defects.

Abstract:The emergence of bone tissue engineering as a trend in regenerative medicine is forcing scientists to create highly functional materials and scaffold construction techniques. Bone tissue engineering uses 3D bio-printed scaffolds that allow and stimulate the attachment and proliferation of osteoinductive cells on their surfaces. Bone grafting is necessary to expedite the patient’s condition because the natural healing process of bones is slow. Fused deposition modeling (FDM) is therefore suggested as a technique for the production process due to its simplicity, ability to create intricate components and movable forms, and low running costs. 3D-printed scaffolds can repair bone defects in vivo and in vitro. For 3D printing, various materials including metals, polymers, and ceramics are often employed but polymeric biofilaments are promising candidates for replacing non-biodegradable materials due to their adaptability and environment friendliness. This review paper majorly focuses on the fused deposition modeling approach for the fabrication of 3D scaffolds. In addition, it also provides information on biofilaments used in FDM 3D printing, applications, and commercial aspects of scaffolds in bone tissue engineering.

Abstract:Orthopedic tumor resection, trauma, or degenerative disease surgeries can result in large bone defects and often require bone grafting. However, standard autologous bone grafting has been associated with donor site morbidity and/or limited quantity. As an alternate, allografts with or without metallic or polyether-etherketone have been used as grafting substitutes. However, these may have drawbacks as well, including stress shielding, pseudarthrosis, disease-transmission, and infection. There is therefore a need for alternative bone substitutes, such as the use of mechanically compliant three-dimensional (3D)-printed scaffolds. Several off-the-shelf materials are available for low-cost fused deposition 3D printing such as polylactic acid (PLA) and polycaprolactone (PCL). We have previously described the feasibility of 3D-printed PLA scaffolds to support cell activity and extracellular matrix deposition. In this study, we investigate two medical-grade filaments consistent with specifications found in American Society for Testing and Materials (ASTM) standard for semi-crystalline polylactide polymers for surgical implants, a pure polymer (100M) and a copolymeric material (7415) for their cytocompatibility and suitability in bone tissue engineering. Moreover, we assessed the impact on osteo-inductive properties with the addition of beta-tricalcium phosphate (β-TCP) minerals and assessed their mechanical properties. 100M and 7415 scaffolds with the additive β-TCP demonstrated superior mesenchymal stem cells (MSCs) differentiation detected via increased alkaline phosphatase activity (6-fold and 1.5-fold, respectively) and mineralized matrix deposition (14-fold and 5-fold, respectively) in vitro . Furthermore, we evaluated in vivo compatibility, biosafety and bone repair potential in a rat femur window defect model. 100M +β - TCP implants displayed a positive biosafety profile and showed significantly enhanced new bone formation compared to 100M implants evidenced by μCT (39 versus 25% bone volume/tissue volume ratio) and histological analysis 6 weeks post-implantation. These scaffolds are encouraging composite biomaterials for repairing bone applications with a great potential for clinical translation. Further analyses are required with appropriate evaluation in a larger critical-sized defect animal model with long-term follow-up.

Abstract:The treatment of bone defects has always posed challenges in the field of orthopedics. Scaffolds, as a vital component of bone tissue engineering, offer significant advantages in the research and treatment of clinical bone defects. This study aims to provide an overview of how 3D printing technology is applied in the production of bone repair scaffolds. Depending on the materials used, the 3D-printed scaffolds can be classified into two types: single-component scaffolds and composite scaffolds. We have conducted a comprehensive analysis of material composition, the characteristics of 3D printing, performance, advantages, disadvantages, and applications for each scaffold type. Furthermore, based on the current research status and progress, we offer suggestions for future research in this area. In conclusion, this review acts as a valuable reference for advancing the research in the field of bone repair scaffolds.

Abstract:Due to the increase in traffic accidents, industrial contingencies, and natural disasters, there is a high demand for bone regeneration biomaterials. Although bone can regenerate and self-repair, it is difficult to do so when the defect area exceeds the critical repair area due to trauma, tumour resection, or congenital diseases. The traditional methods for bone repair are autograft, allograft, and xenograft. However, these methods have flaws and limitations, such as complications in the donor bone area, a limited number of donors, rejection, and infectious diseases. However, in recent years, the use of additive manufacturing technologies in bone tissue engineering has increased. Three-dimensional printing (3DP) is gaining popularity among the various technology options due to its ability to directly print porous composite with designed shape, controlled chemistry, and interconnected porosity. Some of these inorganic composites are biodegradable and have proven ideal for bone tissue engineering, with the ability to deliver growth factors or drugs to specific sites. The purpose of this study is to discuss recent advancements in 3D printed bone tissue engineering composite, as well as current challenges and future directions.

Abstract:Treatment of large-size bone defects is difficult, and acquiring autografts may be challenging due to limited availability. A synthetic patient-specific bone substitute can be developed by using 3D printing technologies in such cases. In the present study, we have developed photocurable composite resins with poly(trimethylene carbonate) (PTMC) containing a high percentage of biodegradable bioactive strontium-substituted nanohydroxyapatite (SrHA, size 30-70 nm). These photocurable resins have then been employed to develop high-surface-area 3D-printed bone substitutes using the digital light processing (DLP) technique. To enhance the surface area of the 3D-printed substitute, cryogels alone and functionalized with bioactive components of bone morphogenetic protein (BMP) and zoledronic acid (ZA) were filled within the 3D-printed scaffold/substitute. The scaffolds were tested in vitro for biocompatibility and functionality in vivo in two therapeutically relevant rat models with large bone defects (4 mm). The porosities of 3D printed scaffolds were found to be 60.1 ± 0.9%, 72.9 ± 0.5%, and 74.3 ± 1.6% for PTMC, PTMC-HA, and PTMC-SrHA, respectively, which is in the range of cancellous bone (50-95%). The thermogravimetric analysis demonstrated the fabrication of 3D printed composites with HA and SrHA concentrations of 51.5 and 57.4 wt %, respectively, in the PTMC matrix. The tensile Young's modulus (E), compressive moduli, and wettability increased post incorporation of SrHA and HA in the PTMC matrix. In vitro and in vivo results revealed that SrHA integrated into the PTMC matrix exhibited good physicochemical and biological properties. Furthermore, the osteoactive molecule-functionalized 3D printed composite scaffolds were found to have an adequate osteoconductive and osteoinductive surface that has shown increased bone regeneration and defect repair in both tibial and cranial bone defects. Our findings thus support the use of PTMC-SrHA composites as next-generation patient-specific synthetic bioactive biodegradable bone substitutes.

Abstract:Cell Transplantation, Volume 33, Issue , January-December 2024. Damage to bones resulting from trauma and tumors poses a significant challenge to human health. Consequently, current research in bone damage healing centers on developing three-dimensional (3D) scaffolding materials that facilitate and enhance the regeneration of fractured bone tissues. In this context, the careful selection of materials and preparation processes is essential for creating demanding scaffolds for bone tissue engineering. This is done to optimize the regeneration of fractured bones. This study comprehensively analyses the latest scientific advancements and difficulties in developing scaffolds for bone tissue creation. Initially, we clarified the composition and process by which bone tissue repairs itself. The review summarizes the primary uses of materials, both inorganic and organic, in scaffolds for bone tissue engineering. In addition, we present a comprehensive study of the most recent advancements in the mainstream techniques used to prepare scaffolds for bone tissue engineering. We also examine the distinct advantages of each method in great detail. This article thoroughly examines potential paths and obstacles in bone tissue engineering scaffolds for clinical applications.
