Abstract:Targeting mitochondria has emerged as an effective approach in recent tumor immunotherapy. While nanosized carriers have been utilized for therapeutic drug delivery, their retention in lysosomes diminishes their efficacy in targeting mitochondria. Despite the use of polycationic materials and peptides to facilitate lysosomal escape through the proton sponge effect and destabilization of lysosomal membranes, concerns persist regarding their stability and biocompatibility. Our objective is to exploit Oroxylin A (OA), a polyphenolic compound, to synthesize metal-polyphenolic complexes, capitalizing on OA's inherent lysosomal self-modifying properties to achieve efficient lysosomal escape. Herein, we report the fabrication of polyphenol-based OA@Mn@3-TYP nanoenhancers, assembled with hyaluronic acid (HA) and denoted as OMT NEs, designed for effective lysosomal escape and targeted drug delivery to mitochondria. These nanoenhancers exhibit a positive targeting ability towards tumor cells and facilitate lysosomal escape through spontaneous glucuronidation and sialic acid transporter mechanisms. The incorporation of OA and 3-TYP in the nanoenhancers leads to a cascading effect resulting in mitochondrial dysfunction and subsequent release of mitochondrial DNA (mtDNA). Upon activation of the Mn2+-enhanced cGAS-STING pathway by the released mtDNA, a synergistic anti-PD-1 immunotherapy effect is augmented, ultimately enhancing anti-tumor treatment.
Unraveling Ros Conversion Through Enhanced Enzyme‐Like Activity with Copper‐Doped Cerium Oxide for Tumor Nanocatalytic Therapy

Abstract:A high degree of surface oxygen mobility and a low level of oxygen vacancy formation energy is exposed over the CeO2/Cu‐W surface, which is employed as a mimetic enzyme of superoxide dismutase (SOD) to convert O2∙‐ →H2O2 and peroxidase (POD) to realize H2O2→∙OH for oncotherapy. Nanozyme catalytic therapy for cancer treatments has become one of the heated topics, and the therapeutic efficacy is highly correlated with their catalytic efficiency. In this work, three copper‐doped CeO2 supports with various structures as well as crystal facets are developed to realize dual enzyme‐mimic catalytic activities, that is superoxide dismutase (SOD) to reduce superoxide radicals to H2O2 and peroxidase (POD) to transform H2O2 to ∙OH. The wire‐shaped CeO2/Cu‐W has the richest surface oxygen vacancies, and a low level of oxygen vacancy (Vo) formation energy, which allows for the elimination of intracellular reactive oxygen spieces (ROS) and continuous transformation to ∙OH with cascade reaction. Moreover, the wire‐shaped CeO2/Cu‐W displays the highest toxic ∙OH production capacity in an acidic intracellular environment, inducing breast cancer cell death and pro‐apoptotic autophagy. Therefore, wire‐shaped CeO2/Cu nanoparticles as an artificial enzyme system can have great potential in the intervention of intracellular ROS in cancer cells, achieving efficacious nanocatalytic therapy.

Abstract:Nanozymes have shown great clinical application potential in the treatment of tumors with reactive oxygen species (ROS). However, conventional nanozyme technologies face challenges such as low catalytic activity and unknown therapeutic mechanisms for tumor therapy. Herein, we designed platinum (Pt)/nanoceria (CeO2) hybrid nanozymes using Pt single-atom and cluster engineering on the CeO2 surface, named Pt/CeO2-NR and Pt/CeO2-R respectively. The results showed that Pt/CeO2-R had higher enzyme-like activity than Pt/CeO2-NR because the Pt clusters (Pt0 and Pt2+) stabilized the oxygen vacancies better than the single Pt atoms (Pt2+). Further studies showed that Pt/CeO2-R exhibited selective toxicity to tumor cells, induced tumor cell apoptosis, and inhibited cell migration. In addition, we studied the antitumor mechanism of Pt/CeO2-R with stable isotopic labeling by amino acids in cell culture (SILAC)-based quantitative proteomics. Mechanistically, large amounts of the ROS produced by Pt/CeO2-R affected both the mitochondria and the endoplasmic reticulum and ultimately induced tumor cell apoptosis. Additionally, Pt/CeO2-R inhibited tumor cell migration by downregulating the proteins on the cell membrane surface. This study provided an approach for optimizing the enzyme-like activities of CeO2 through the efficient use of platinum engineering, and explored the mechanisms by which Pt/CeO2-R killed tumor cells and inhibited cell migration during the treatment of tumors.

Abstract:Twenty non-diabetic controls, 50 diabetics without retinopathy and 29 diabetics with retinopathy, all well-matched age groups, were studied in males and females, respectively. Moreover, diabetic patients with retinopathy were divided into the following two subgroups: mild retinopathy and severe retinopathy. Platelet aggregation was elevated in the male diabetics with mild or severe retinopathy. There was no difference in platelet volume between the groups. Intraplatelet ADP contents were significantly decreased in the female diabetics with severe retinopathy showing no high platelet aggregability. There was no significant difference in intraplatelet ATP contents between groups. These results indicated not only that platelet aggregation might be generally high in the diabetics with retinopathy, but also that dense granule release might occur readily in vivo in the female diabetics with severe retinopathy and therefore platelet aggregation was not elevated in the group.
pH‐Activatable Copper‐Biomineralized Proenzyme for Synergistic Chemodynamic/Chemo‐Immunotherapy against Aggressive Cancers

Abstract:Artificial enzymes have demonstrated therapeutic benefit against diverse malignant tumors, yet their antitumor potencies are still severely compromised by non‐selective catalysis, low atomic utilization efficiency, and undesired off‐target toxicity. Herein, we report that the peroxidase‐like biomineralized copper (II) carbonate hydroxide nanocrystals inside single albumin nanocages (CuCH‐NCs) act as a pH‐activatable proenzyme to achieve tumor‐selective and synergistic chemodynamic/chemo‐immunotherapy against aggressive triple‐negative breast cancers (TNBCs). These CuCH‐NCs show pH‐sensitive Cu2+ release, which spontaneously undergoes glutathione (GSH)‐mediated reduction into Cu+ species for catalyzing the evolution of H2O2 into hydroxyl radicals in a single‐atom‐like manner to cause chemodynamic cell injury, and simultaneously activates non‐toxic disulfiram to cytotoxic complex for yielding selective chemotherapeutic damage via blocking cell proliferation and amplifying cell apoptosis. CuCH‐NCs exhibit considerable tumor‐targeting capacity with deep penetration depth, thus affording preferable efficacy against orthotopic breast tumors through synergistic chemodynamic/chemotherapy, together with good in vivo safety. Moreover, CuCH‐NCs arouse distinct immunogenic cell death effect and upregulate PD‐L1 expression upon disulfiram combination, and thus synergize with anti‐PD‐L1 antibody to activate adaptive and innate immunities, together with relieved immunosuppression, finally yielding potent antitumor efficacy against both primary and metastatic TNBCs. Thermoresponsive and Substrate Self-Cycling Nanoenzyme System for Efficient Tumor Therapy

Abstract:Cerium oxide (CeO2-x) performs well in photothermal and catalytic properties due to its abundance of oxygen vacancies. Based on this, we designed a thermosensitive therapeutic nanoplatform to achieve continuous circular drug release in tumor. It can solve the limitation caused by insufficient substrate in the process of tumor treatment. Briefly, CeO2-x and camptothecin (CPT) were wrapped in an agarose hydrogel, which could be melted by the photothermal effect of CeO2-x. At the same time, the local temperature increase provided photothermal treatment, which could induce the apoptosis of tumor cell. After that, CPT was released to damage the DNA in tumor cells to realize chemical treatment. In addition, CPT could active nicotinamide adenine dinucleotide oxidase to react with O2 to increase the intracellular H2O2. After that, the exposed CeO2-x could catalyze H2O2 to generate cytotoxic reactive oxygen species for chemodynamic therapy. More importantly, CeO2-x could catalyze H2O2 to produce O2, which could combine with the catalytic action of CPT to construct a substrate self-cycling nanoenzyme system. Overall, this self-cycling nanoplatform released hypoxia in the tumor microenvironment and built a multimode tumor treatment, which achieved an ideal antitumor affect.
A MXene-Based Bionic Cascaded-Enzyme Nanoreactor for Tumor Phototherapy/Enzyme Dynamic Therapy and Hypoxia-Activated Chemotherapy

Abstract:Abstract The enzyme-mediated elevation of reactive oxygen species (ROS) at the tumor sites has become an emerging strategy for regulating intracellular redox status for anticancer treatment. Herein, we proposed a camouflaged bionic cascaded-enzyme nanoreactor based on Ti 3 C 2 nanosheets for combined tumor enzyme dynamic therapy (EDT), phototherapy and deoxygenation-activated chemotherapy. Briefly, glucose oxidase (GOX) and chloroperoxidase (CPO) were chemically conjugated onto Ti 3 C 2 nanosheets, where the deoxygenation-activated drug tirapazamine (TPZ) was also loaded, and the Ti 3 C 2 -GOX-CPO/TPZ (TGCT) was embedded into nanosized cancer cell-derived membrane vesicles with high-expressed CD47 (m e TGCT). Due to biomimetic membrane camouflage and CD47 overexpression, m e TGCT exhibited superior immune escape and homologous targeting capacities, which could effectively enhance the tumor preferential targeting and internalization. Once internalized into tumor cells, the cascade reaction of GOX and CPO could generate HClO for efficient EDT. Simultaneously, additional laser irradiation could accelerate the enzymic-catalytic reaction rate and increase the generation of singlet oxygen ( 1 O 2 ). Furthermore, local hypoxia environment with the oxygen depletion by EDT would activate deoxygenation-sensitive prodrug for additional chemotherapy. Consequently, m e TGCT exhibits amplified synergistic therapeutic effects of tumor phototherapy, EDT and chemotherapy for efficient tumor inhibition. This intelligent cascaded-enzyme nanoreactor provides a promising approach to achieve concurrent and significant antitumor therapy.

Abstract:The ternary PdMoP composite nanoenzymes enable an irreversible cascade of immunogenic tumor cell ferroptosis by promoting reactive oxygen species (ROS) production and GSH/GPX4 depletion and reversing the immunosuppressive tumor microenvironment (TME) for the enzyodynamic effect‐enhance ferroptosis and sonocatalysis‐enabled tumor immunotherapy of breast cancer. The engineered multicomponent nanoenzymes feature remarkable potential for the clinical treatment of cancer. Immunotherapy has become one of the most effective therapeutic modalities for achieving long‐term cancer remission, but the available immunotherapeutic strategies suffer from modest response rates owing to the insufficient immunogenicity of tumor cells. In this work, a nanomedicine strategy for maintaining highly immunogenic tumor cells by inducing cascade‐mediated immunogenic tumor‐cell ferroptosis is proposed and developed. A PdMoP nanoplatform is engineered that not only induces initial immunogenic tumor cell ferroptosis through its multienzyme‐mimicking activities but also accelerates Mo(IV)‐to‐Mo(VI) transition, which aggravates glutathione (GSH) depletion for deactivating glutathione peroxidase 4 (GPX4) enzyme and lead to excessive radical production for promoting p53 expression and reducing SLC7A11, thereby resulting in efficient ferroptosis and apoptosis. Additionally, PdMoP nanoparticles induce the breakdown of hydrogen peroxide into oxygen to alleviate tumor hypoxia, working synergistically with GSH depletion to reverse the immunosuppressive tumor microenvironment. Significant ferroptosis (through the classical p53‐SLC7A11‐GPX4 pathway) is monitored in both in vitro cellular level and in vivo tumor models, achieving effective tumor suppression and elimination. The distinct ultrasound‐enhanced enzyodynamic therapy strategy represents a simple and effective paradigm for treating cancer by nanocatalytic medicine and catalytic biomaterials.

Ultrathin 2D Pd/Cu Single-Atom MOF Nanozyme to Synergistically Overcome Chemoresistance for Multienzyme Catalytic Cancer Therapy

Abstract:Single-atom nanozymes (SAzymes) have obtained increasing interest to mimic natural enzymes for efficient cancer therapy, while challenged by chemoresistance from cellular redox homeostasis and the interface of reductive species in tumor microenvironment (TME). Herein, a dual single-atomic ultrathin 2D metal organic framework (MOF) nanosheet of multienzyme (Pd/Cu SAzyme@Dzy) is prepared to synergistically overcome chemoresistance for multienzyme enhanced cancer catalytic therapy. The Pd SAzyme exhibits peroxidase (POD)-like catalytic activity for overcoming chemoresistance via disturbing cellular redox balance. This is further enhanced by cascade generation of more ∙OH via Cu+ -catalyzed POD-like reactions, initiated by in situ-reduction of Cu2+ into Cu+ upon GSH depletion. This process can also avoid the consumption of ∙OH by endogenous reductive GSH in TME, ensuring the adequate amount of ∙OH for highly efficient therapy. Besides, the DNAzyme is also delivered for gene therapy of silencing cancer-cell-targeting VEGFR2 protein to further enhance the therapy. Based on both experiments and theoretical calculations, the synergetic multienzyme-based cancer therapy is examined and the enhancement by the cascade tumor antichemoresistance is revealed.

Abstract:Cerium oxide nanozymes have emerged as a new type of bio-antioxidants in recent years. CeO2 nanozymes possess enzyme mimetic activities with outstanding free radical scavenging activity, facile synthesis conditions, and excellent biocompatibility. Based on these extraordinary properties, use of CeO2 nanozymes has been demonstrated to be a highly versatile therapeutic method for many diseases, such as for inflammation, rheumatoid arthritis, hepatic ischemia-reperfusion injury and Alzheimer's disease. In addition to that, CeO2 nanozymes have been widely used in the diagnosis and treatment of cancer. Many examples can be found in the literature, such as magnetic resonance detection, tumour marker detection, chemotherapy, radiotherapy, photodynamic therapy (PDT), and photothermal therapy (PTT). This review systematically summarises the latest applications of CeO2-based nanozymes in cancer research and treatment. We believe that this paper will help develop value-added CeO2 nanozymes, offering great potential in the biotechnology industry and with great significance for the diagnosis and treatment of a wide range of malignancies.
Synergizing Pyroelectric Catalysis and Enzyme Catalysis: Establishing a Reciprocal and Synergistic Model to Enhance Anti-Tumor Activity

Abstract:Nanozyme activity is greatly weakened by the microenvironment and multidrug resistance of tumor cells. Hence, a bi-catalytic nanoplatform, which promotes the anti-tumor activity through "charging empowerment" and "mutual complementation" processes involved in enzymatic and pyroelectric catalysis, by loading ultra-small nanoparticles (USNPs) of pyroelectric ZnSnO3 onto MXene nanozyme (V2CTx nanosheets), is developed. Here, the V2CTx nanosheets exhibit enhanced peroxidase activity by reacting V3+ with H2O2 to generate toxic ·OH, accelerated by the near-infrared (NIR) light mediated heat effect. The resulting V4+ is then converted to V3+ by oxidizing endogenous glutathione (GSH), realizing an enzyme-catalyzed cycle. However, the cycle will lose its persistence once GSH is insufficient; nevertheless, the pyroelectric charges generated by ZnSnO3 USNPs continuously support the V4+/V3+ conversion and ensure nanoenzyme durability. Moreover, the hyperthermia arising from the V2CTx nanosheets by NIR irradiation results in an ideal local temperature gradient for the ZnSnO3 USNPs, giving rise to an excellent pyroelectric catalytic effect by promoting band bending. Furthermore, polarized charges increase the tumor cell membrane permeability and facilitate nanodrug accumulation, thereby resolving the multidrug resistance issue. Thus, the combination of pyroelectric and enzyme catalysis together with the photothermal effect solves the dilemma of nanozymes and improves the antitumor efficiency.

Abstract:Nanozymes have attracted extensive interest owing to their high stability, low cost and easy preparation, especially in the field of cancer therapy. However, the relatively low catalytic activity of nanozymes in the tumor microenvironment (TME) has limited their applications. Herein, we report a novel nanozyme (PtFe@Fe3O4) with dual enzyme-like activities for highly efficient tumor catalytic therapy. PtFe@Fe3O4 shows the intrinsic photothermal effect as well as photo-enhanced peroxidase-like and catalase-like activities in the acidic TME, thereby effectively killing tumor cells and overcoming the tumor hypoxia. Importantly, a possible photo-enhanced synergistic catalytic mechanism of PtFe@Fe3O4 was first disclosed. We believe that this work will advance the development of nanozymes in tumor catalytic therapy.
A thermo-sensitive hydrogel with prominent hemostatic effect prevents tumor recurrence via anti-anoikis-resistance

Abstract:Tumor cells can survive when detached from the extracellular matrix (ECM) or lose cell–cell connections, a phenomenon known as anoikis-resistance (AR). AR is closely associated with tumor cell metastasis and recurrence, enabling tumor cells to disseminate, migrate, and invade after detachment. To address this issue, a novel intervention method combining intraoperative hemostasis with multifunctional nanozyme driven-enhanced chemodynamic therapy (ECDT) has been proposed, which holds the potential to weaken the AR capability of tumor cells and suppress tumor recurrence. Here, a nanocomposite containing a dendritic mesoporous nanoframework with Cu 2+ was developed using an anion-assisted approach after surface PEG grafting and glucose oxidase (GOx) anchoring (DMSN-Cu@GOx/PEG). DMSN-Cu@GOx/PEG was further encapsulated in a thermal-sensitive hydrogel (H@DMSN-Cu@GOx/PEG). DMSN-Cu@GOx/PEG utilizes its high peroxidase (POD) activity to elevate intracellular ROS levels, thereby weakening the AR capability of bladder cancer cells. Additionally, through its excellent catalase (CAT) activity, DMSN-Cu@GOx/PEG converts the high level of hydrogen peroxide (H 2 O 2 ) catalyzed by intracellular GOx into oxygen (O 2 ), effectively alleviating tumor hypoxia, downregulating hypoxia-inducible factor-1α (HIF-1α) expression, inhibiting epithelial-mesenchymal transition (EMT) processes, and ultimately suppressing the migration and invasion of bladder cancer cells. Interestingly, in vivo results showed that the thermosensitive hydrogel H@DMSN-Cu@GOx/PEG could rapidly gel at body temperature, forming a gel film on wounds to eliminate residual tumor tissue after tumor resection surgery. Importantly, H@DMSN-Cu@GOx/PEG exhibited excellent hemostatic capabilities, effectively enhancing tissue coagulation during post-tumor resection surgery and mitigating the risk of cancer cell dissemination and recurrence due to surgical bleeding. Such hydrogels undoubtedly possess strong surgical application. Our developed novel nanosystem and hydrogel can inhibit the AR capability of tumor cells and prevent recurrence post-surgery. This study represents the first report of using dendritic mesoporous silica-based nanoreactors for inhibiting the AR capability of bladder cancer cells and suppressing tumor recurrence post-surgery, providing a new avenue for developing strategies to impede tumor recurrence after surgery.

Abstract:Diagram of in vivo catalytic immunotherapy mechanism of SS‐MSN@Au‐BOM NPs SS‐MSN@Au‐BOM NPs exhibit GOx‐POD‐CAT‐cascade catalysis and significantly disrupts tumor growth by catalyzing endogenous hydrogen peroxide to generate cytotoxic ROS and essential oxygen. In addition, this catalytic action facilitates immunogenic cell death, enhances dendritic cell maturation, and T‐cell activation, which synergistically enhances anti‐tumor effects. The treatment of solid tumors remains a significant challenge due to the complex, immunosuppressive tumor microenvironment (TME). This manuscript introduces the self‐oxygenated biomimetic nanozyme system, SS‐MSN@Au‐BOM, which innovatively combines catalytic therapy with immunotherapy to modulate the TME for enhanced therapeutic efficacy. Employing mesoporous silicon nanoparticles doped with disulfide bonds, the system integrates gold nanozymes that exhibit glucose oxidase (GOx)‐like and peroxidase (POD)‐like activities, along with a catalase (CAT) function derived from bacterial outer membrane (BOM) coatings. SS‐MSN@Au‐BOM significantly disrupted tumor growth by catalyzing endogenous hydrogen peroxide to generate cytotoxic ROS and essential oxygen, enhancing intratumoral ROS levels while alleviating hypoxia. This catalytic action facilitates immunogenic cell death, enhances dendritic cell maturation, and T‐cell activation. Notably, the system shows excellent biocompatibility, effective tumor targeting, and prolonged systemic circulation. SS‐MSN@Au‐BOM offers a dual‐functional approach that effectively disrupts the TME and promotes robust antitumor immunity. This study paves the way for further clinical investigation and the development of enzyme‐based therapeutics, potentially transforming the landscape of cancer treatment.

Abstract:Nanocatalytic therapy, based on the nanozyme-triggered production of reactive oxygen species (ROS) in tumor microenvironment (TME), has demonstrated potential in tumor therapy, but nanozymes still face challenges of activity and specificity that compromise the therapeutic efficacy. Herein, we develop an innovative strategy using a single-atom nanozyme to initiate cascade enzymatic reactions in TME for tumor-specific treatment. The cobalt (Co)-single-atom nanozyme, with Co- N coordination on N -doped porous carbon ( Co-SAs@NC ), displays catalase-like activity that decompose cellular endogenous H 2 O 2 to produce O 2 , and subsequent oxidase-like activity that converts O 2 into cytotoxic superoxide radicals to efficiently kill tumor cells. By incorporation with doxorubicin, the therapy achieves a significantly enhanced antitumor effect in vivo . Our findings show that cascade TME-specific catalytic therapy combining with chemotherapy is a promising strategy for efficient tumor therapy.

High-Performance Self-Cascade Pyrite Nanozymes for Apoptosis–Ferroptosis Synergistic Tumor Therapy

Abstract:As next-generation artificial enzymes, nanozymes have shown great promise for tumor catalytic therapy. In particular, their peroxidase-like activity has been employed to catalyze hydrogen peroxide (H2O2) to produce highly toxic hydroxyl radicals (•OH) to kill tumor cells. However, limited by the low affinity between nanozymes with H2O2 and the low level of H2O2 in the tumor microenvironment, peroxidase nanozymes usually produced insufficient •OH to kill tumor cells for therapeutic purposes. Herein, we present a pyrite peroxidase nanozyme with ultrahigh H2O2 affinity, resulting in a 4144- and 3086-fold increase of catalytic activity compared with that of classical Fe3O4 nanozyme and natural horseradish peroxidase, respectively. We found that the pyrite nanozyme also possesses intrinsic glutathione oxidase-like activity, which catalyzes the oxidation of reduced glutathione accompanied by H2O2 generation. Thus, the dual-activity pyrite nanozyme constitutes a self-cascade platform to generate abundant •OH and deplete reduced glutathione, which induces apoptosis as well as ferroptosis of tumor cells. Consequently, it killed apoptosis-resistant tumor cells harboring KRAS mutation by inducing ferroptosis. The pyrite nanozyme also exhibited favorable tumor-specific cytotoxicity and biodegradability to ensure its biosafety. Enzyme-Empowered "Two Birds with One Stone" Strategy for Amplifying Tumor Apoptosis and Metabolic Clearance

Abstract:Nanomedicine has reshaped the landscape of cancer treatment. However, its efficacy is still hampered by innate tumor defense systems that rely on adenosine triphosphate (ATP) for fuel, including damage repair, apoptosis resistance, and immune evasion. Inspired by the naturally enzymatic reaction of glucose oxidase (GOx) with glucose, here a novel "two birds with one stone" technique for amplifying enzyme-mediated tumor apoptosis and enzyme-promoted metabolic clearance is proposed and achieved using GOx-functionalized rhenium nanoclusters-doped polypyrrole (Re@ReP-G). Re@ReP-G reduces ATP production while increasing H2O2 concentrations in the tumor microenvironment through GOx-induced enzymatic oxidation, which in turn results in the downregulation of defense (HSP70 and HSP90) and anti-apoptotic Bcl-2 proteins, the upregulation of pro-apoptotic Bax, and the release of cytochrome c. These processes are further facilitated by laser-induced hyperthermia effect, ultimately leading to severe tumor apoptosis. As an enzymatic byproduct, H2O2 catalyzes the conversion of rhenium nanoclusters in Re@ReP-G nanostructures into rhenate from the outside in, which accelerates their metabolic clearance in vivo. This Re@ReP-G-based "two birds with one stone" therapeutic strategy provides an effective tool for amplifying tumor apoptosis and safe metabolic mechanisms.

Abstract:Overcoming apoptosis resistance to achieve efficient breast cancer treatment remains a challenge. The precise induction of another form of programmed cell death, pyroptosis, is an excellent alternative for treating cancer. We developed ultrasound (US)‐enhanced enzyme dynamic (enzyodynamic) therapy (EDT) by employing LaFeO3 perovskite nanocrystals as a substrate to increase the rate of deleterious reactive oxygen species (ROS) generation for intensive cell pyroptosis. LaFeO3 nanocrystals possess quadruple enzyme‐mimicking activities, including oxidase‐, peroxidase‐, glutathione peroxidase‐, and catalase‐mimicking activities, which undertake the dominant therapeutic task through cascade catalytic reactions, including the reversal of hypoxic microenvironment, depletion of endogenous glutathione, and continuous output of ROS. US exogenous stimulation increases the transition rate of the intermediate complex to Fe (II) and favors incremental ROS production, by which the ROS burst‐induced pyroptosis process is accomplished through the ROS‐TXNIP‐NLRP3‐GSDMD pathway. Both in vitro and in vivo antineoplastic outcomes affirmed the ascendancy of LaFeO3 nanozyme‐induced pyroptosis. This work highlights the critical role of US coupled with nanocatalytic reactors in pyroptosis‐dominant breast cancer treatment with the apoptosis resistance circumvention feature.
