Abstract:An amperometric immunosensor based on a composite electrode of single-walled carbon nanotubes and ionic liquid n-octylpyridinum afluorophosphate (SWCNT-ILE) was developed for the determination of tetrodotoxin (TTX). Compared with the glassy carbon electrode (GCE), the electrode combined advantages of carbon nanotubes and ionic liquid, which exhibited the excellent antifouling ability of p-nitrophenol (PNP) so that it remarkably improved the stability of the p-nitrophenyl phosphate-based sensor. Combining the enzyme-linked immune sorbent assay (ELISA) by alkaline phosphatase (AP) and magnetic particles immobilized with antigens, a real-time assay of tetrodotoxin was developed by amperometric immunosensors. Under the optimium condition, the developed sensor demonstrated a linear range of tetrodotoxin from 2 to 45 ng/mL with a low detection limit of 5 ng/mL. Furthermore, the amperometric immunosensor was applied to determine TTX in real samples and could be used as an effective and sensitive sensor for direct detection of tetrodotoxin within 20 min.

Abstract:Saxitoxin (STX) and brevetoxin (PbTX-2), which are produced by marine dinoflagellates, are highly-toxic marine toxins targeting separate sites of the α subunit of voltage-dependent sodium channels (VDSCs). In this work, a portable cardiomyocyte-based potential biosensor is designed for rapid detection of STX and PbTX-2. This potential biosensor is constructed by cardiomyocyte and microelectrode array (MEA) with a label-free and real-time wireless 8-channel recording system which can dynamically monitor the multisite electrical activity of cardiomyocyte network. The recording signal parameters, spike amplitude, firing rate and 50% of spike potential duration (SPD50) extracted from extracelluar field potential (EFP) signals of the potential biosensor is analyzed to quantitatively evaluate toxicological risk of STX and PbTX-2. Firing rate of biosensor signals presents high sensitivity to STX with the detection limit of 0.35ng/ml within 5min. SPD50 shows high sensitivity to PbTX-2 with the detection limit of 1.55ng/ml within 5min. Based on the multi-parameter analysis, cardiomyocyte-based potential biosensor will be a promising tool for rapid detection of these two toxins.

Abstract:Tetrodotoxin (TTX), a potent neurotoxin, has recently been detected in marine fauna and waters in multiple locations outside its historical range and is projected to increase in both stability and bioavailability in acidifying oceans. We report the first biosensor designed to detect sub-ng mL-1 concentrations of TTX directly in natural seawater. The compact sensor chip employs a competitive enzyme strategy with a glucose oxidase-TTX conjugate that generates an amperometric response in the presence of a ferricyanide redox mediator. The biorecognition layer synthesis is confirmed by attenuated total reflection Fourier transform infrared spectroscopy. Cyclic voltammetry and electrochemical impedance spectroscopy are used to probe the stability and hydrophilicity of the biorecognition layer. We investigate the effects of nonspecific adsorption on sensor results and find that bovine serum albumen is an excellent blocking agent for preventing NSA when incubating with filtered seawater samples. The biosensor has a limit of detection of 0.053 ng mL-1 and a sensitivity of 87 nA ng-1 mL. Requiring no extraction or preconcentration steps with a time-to-results of under an hour, our biosensor provides a new tool for detecting TTX in natural waters, which can lead to more proactive monitoring and higher throughput testing.

Abstract:PbTx-2 and okadaic acid (OA) are two typical marine toxins that are highly toxic and harmful to human health. The approach based on citrate-capped gold nanoparticles (Cit-AuNPs) and specific aptamers to construct label-free colorimetric sensors is a widely used method for marine toxin detection. However, the potential interactions between Cit-AuNPs and target molecules have always been ignored, which may result in wrong analytical results due to shortcomings in the Cit-AuNPs. To overcome these shortfalls, in this work, AuNPs were synthesized using tyrosine as a reducing and capping agent, and a robust colorimetric aptasensor based on tyrosine-capped AuNPs (Tyr-AuNPs) was constructed for the label-free detection of marine toxins. Tyr-AuNPs presented better stability compared to Cit-AuNPs due to the stronger binding of amine groups on tyrosine to AuNPs through the Au-N bond. Interactions between Tyr-AuNPs and PbTx-2 were analyzed through UV-vis and isothermal titration calorimetry methods and the results validated the robustness of the Tyr-AuNPs. Colorimetric aptasensors were established for PbTx-2 and OA detection with a linear range of 0.05-4 ppm and limits of detection of 2.25 ppb and 5.19 ppb, respectively. These results demonstrate that the developed colorimetric aptasensor can be a robust and promising method for marine toxin detection.
Non-toxic immunosensor for highly efficient detection of tetrodotoxin by electrochemical and colorimetric dual-mode sensing platform

Abstract:Antibody-based immunoassay is an important tool for detecting tetrodotoxin (TTX), a strong neurotoxic with high mortality rate. However, the traditional immunoassays always suffer from low sensitivity, high batch errors of the TTX conjugate and potential harmful to operators. In this work, we fabricated a monoclonal antibody (mAb) with good affinity and high selectivity against TTX, and selected a phage-displayed mimotope with specific binding to the anti-TTX mAb. Based on the mAb and mimotope, a non-toxic and dual-mode (electrochemical and colorimetric signal) immunosensor was developed for the highly efficient and sensitive detection of TTX. A biomimetic mineralized material encapsulated with anti-TTX mAb and HRP (e.g. HRP/anti-TTX ) was used as the recognition element and signal probe. The developed electrochemical/colorimetric dual-mode sensing platform realized the simultaneous sensing of TTX: a wide detection range from 1.0 × 10 -4 to 1.0 × 10 3 μg/mL, with a detection limit as low as 1.0 × 10 -5 μg/mL by electrochemical analysis; a linear range from 1.05 × 10 -3 - 3.8×10 -1 μg/mL, accompanied by a detection limit of 1.83 × 10 -4 μg/mL by colorimetric analysis based on a smart phone. To the best of our knowledge, we report for the first time on the development of electrochemical/colorimetric dual-mode sensing platform with non-toxic recognizing strategy for detecting TTX, which is highly desirable for detecting and monitoring toxins residue in food samples.

Abstract:Tetrodotoxin (TTX) is a marine toxin responsible for many intoxications around the world. Its presence in some pufferfish species and, as recently reported, in shellfish, poses a serious health concern. Although TTX is not routinely monitored, there is a need for fast, sensitive, reliable, and simple methods for its detection and quantification. In this work, we describe the use of an automated patch clamp (APC) system with Neuro-2a cells for the determination of TTX contents in pufferfish samples. The cells showed an IC50 of 6.4 nM for TTX and were not affected by the presence of muscle, skin, liver, and gonad tissues of a Sphoeroides pachygaster specimen (TTX-free) when analysed at 10 mg/mL. The LOD achieved with this technique was 0.05 mg TTX equiv./kg, which is far below the Japanese regulatory limit of 2 mg TTX equiv./kg. The APC system was applied to the analysis of extracts of a Lagocephalus sceleratus specimen, showing TTX contents that followed the trend of gonads > liver > skin > muscle. The APC system, providing an in vitro toxicological approach, offers the advantages of being sensitive, rapid, and reliable for the detection of TTX-like compounds in seafood.

Abstract:Saxitoxin (STX) belongs to the family of marine biological toxins, which are major contaminants in seafood. The reference methods for STX detection are mouse bioassay and chromatographic analysis, which are time-consuming, high costs, and requirement of sophisticated operation. Therefore, the development of alternative methods for STX analysis is urgent. Electrochemical analysis is a fast, low-cost, and sensitive method for biomolecules analysis. Thus, in this study, an electrolyte-insulator-semiconductor (EIS) sensor based on aptamer-modified two-dimensional layered Ti3C2Tx nanosheets was developed for STX detection. The high surface area and rich functional groups of MXene benefited the modification of aptamer, which had specific interactions with STX. Capacitance-voltage (C-V) and constant-capacitance (ConCap) measurement results indicated that the aptasensor was able to detect STX with high sensitivity and good specificity. The detection range was 1.0 nM to 200 nM and detection limit was as low as 0.03 nM. Moreover, the aptasensor was found to have a good selectivity and two-week stability. The mussel tissue extraction test suggested the potential application of this biosensor in detecting STX in real samples. This method provides a convenient approach for low-cost, rapid, and label-free detection of marine biological toxins.

Abstract:With the rapid development of modern industry,the threat from marine toxins to human health has be?come increasingly serious,especially the paralytic biological poison. This paper presents a new method of cell-based potential sensor to detect shellfish toxicity. The cardiomyocyte combined with microelectrode array(MEA)by microelectronic processing technique which construct a novel cardiomyocyte potential sensor. The sensor can not on?ly real-time monitoring,but also could output good extracellular field potential signal with stability and consistency, which cardiomyocytes seeded on the MEA high-closely. By testing saxitoxin(STX)and analyzing the characteristics of signal parameters,the experimental results illustrate the STX have obvious inhibitory effect for electrical activity of cardiomyocyte. And the peak potential amplitude and frequency have obvious inhibition of concentration depen?dence,and presented the different concentration sensitivity. The LOD(limit of detection)of conventional standard laboratory toxicity detection on mouse bioassay method is 40μg/100 g,while the LOD and detection range of cardio?myocyte potential sensor is 1.004 ng/mL and 25 nmol/L~1 600 nmol/L,respectively. By comparative experiments, this method has great LOD and the detection is relatively simple and easy to fit standardization. With further im?provement,the method has good prospects for development on detection rapidly toxicity of marine toxins.
A smartphone-based fluorescent biosensor with metal-organic framework biocomposites and cotton swabs for the rapid determination of tetrodotoxin in seafood

Abstract:Background: Tetrodotoxin (TTX) is a potent neurovirulent marine biotoxin that is present in puffer fish and certain marine animals. It is capable of causing severe neurotoxic symptoms and even death when consumed through contaminated seafood. Due to its high toxicity, developing an effective assay for TTX determination in seafood has significant benefits for food safety and human health. Currently, it remains challenging to achieve on-site determination of TTX in seafood. To facilitate mass on-site assays, more affordable technologies utilizing accessible equipment that require no skilled personnel are needed. Results: A smartphone-based portable fluorescent biosensor is proposed for TTX determination by using metal-organic framework (MOF) biocomposites and cotton swabs. Oriented antibody (Ab)-decorated and fluorescent quantum dot (QD)-loaded MOF biocomposites (QD@MOF*Ab) are rapidly synthesized for binding targets and fluorescent responses by utilizing the tunability of zinc-based MOF. Moreover, facile Ab-immobilized household cotton swabs are utilized as TTX capture tools. TTX forms sandwich immune complexes with QD@MOF*Ab probes, achieving signal amplification. These probes are excited by a portable device to generate bright fluorescent signals, which can be detected by the naked eye, and TTX quantitative results are obtained using a smartphone. When observed with the naked eye, the limit of detection (LOD) is 0.4 ng/mL, while intelligent quantitation presents an LOD of 0.13 ng/mL at logarithmic concentrations of 0.2-400 ng/mL. Significance: This biosensor is convenient to use, and an easy-to-operate analysis is completed within 15 min, thus demonstrating excellent performance in terms of detection speed and portability. Furthermore, it successfully determines TTX contents in puffer fish and clam samples, demonstrating its potential for monitoring seafood. Herein, this work provides a favorable rapid sensing platform that is easily portable.

Abstract:The goal of this work was to obtain rapid sampling technique to measure transient metabolites in vivo. First, a pulse of glucose was added to a culture of the yeast Saccharomyces cerevisiae growing aerobically under glucose limitation. Next, samples were removed at 2 to 5 s intervals and quenched using methods that depend on the metabolite measured. Extracellular glucose, excreted products, as well as glycolytic intermediates (G6P, F6P, FBP, GAP, 3-PG, PEP, Pyr) and cometabolites (ATP, ADP, AMP, NAD(+), NADH) were measured using enzymatic or HPLC methods. Significant differences between the adenine nucleotide concentrations in the cytoplasm and mitochondria indicated the importance of compartmentation for the regulation of the glycolysis. An improved efficient biochemical detection method to marine toxins with a smartphone-based portable system—Bionic e-Eye

Abstract:It is an undeniable fact that the effect of marine toxins becomes increasingly serious to people. Paralytic shellfish poisons (PSP) are well-known sodium channel-blocking marine toxins, which block the conduction of nerve impulses and lead to a series of neurological disorders symptoms. Similarly, as inhibitors of serine/threonine protein phosphatases type 1 (PP1) and 2A (PP2A), diarrheic shellfish poisons (DSP) can destroy cytoskeleton and affect intracellular signal transduction, which causes some symptoms of gastrointestinal food allergy. Since the conventional methods to detect marine toxins had some disadvantages (e.g., complicated pretreatment, time-consuming detection, huge instruments and off-site test), bionic electronic eye (Bionic e-Eye) was designed and studied for the in-field fast measurement and real-time on-line analysis to marine toxins. Bionic e-Eye installed the homemade software-iPlate and completed the integration of image acquisition and further data processing. With the cooperation of ELISA, Bionic e-Eye used two color models-HSV and RGB to evaluate its bioanalytical performance in saxitoxin (STX) assay and okadaic acid (OA), which were the standard representations of PSP and DSP, respectively. Moreover, the microtiter plate reader (MTPR) was compared with Bionic e-Eye in the detection of marine toxins. The results demonstrated that Bionic e-Eye using saturation had the comparable precision, dynamic range, detection limit and sensitivity to MTPR in the two assays. Therefore, Bionic e-Eye has great potential to be a field-test platform and becomes the supplement of conventional off-site detection methods in measurement domain of marine toxins. Abstract:Tetrodotoxin (TTX), which is found in various marine organisms, including pufferfish, shellfish, shrimp, crab, marine gastropods, and gobies, is an effective marine toxin and the cause of many seafood poisoning incidents. Owing to its toxicity and threat to public health, the development of simple, rapid, and efficient analytical methods to detect TTX in various food matrices has garnered increasing interest worldwide. Herein, we reviewed the structure and properties, origin and sources, toxicity and poisoning, and relevant legislative measures of TTX. Additionally, we have mainly reviewed the state-of-the-art progress of analytical methods for TTX detection in the past five years, such as bioassays, immunoassays, instrumental analysis, and biosensors, and summarized their advantages and limitations. Furthermore, this review provides an in-depth discussion of the most advanced biosensors, including cell-based biosensors, immunosensors, and aptasensors. Overall, this study provides useful insights into the future development and wide application of biosensors for TTX detection.

Abstract:Saxitoxin (STX), is one of the most dangerous and widespread paralytic shellfish toxins, causing a severe threat to the ecosystem and human health. So, it is important and highly essential to develop novel techniques for STX detection in a convenient, desirable, and low-cost manner. Herein, this study developed an electrolyte-insulator-semiconductor (EIS) sensor covered with a layer-by-layer prepared, positively-charged weak polyelectrolyte layer of poly (allylamine hydrochloride) (PAH) for the label-free detection of STX. The specific aptamer (Apt) sensitive to STX was electrostatically adsorbed onto the PAH layer. This leads to a preferentially flat orientation of the Apt within the Debye length, thus yielding a reduced charge-screening influence and a higher sensor signal. Each step of sensor surface modification, i.e. PAH adsorption, immobilization of Apt, and attachment of STX, was monitored by capacitance-voltage (C-V) and constant-capacitance (ConCap) measurements. Furthermore, atomic force microscopy (AFM) was employed to characterize the surface morphology and roughness of the PAH layer. Fluorescence microscopy was used to confirm the effective immobilization of Apt onto the PAH-modified EIS sensor. The results showed that the detection range of this aptasensor for STX detection was 0.5-100 nM and the detection limit was as low as 0.05 nM. Furthermore, this aptasensor showed good selectivity and 9 days' stability. The mussel tissue extraction test suggested that this aptasensor can be used to detect STX in real samples. This aptasensor provides a convenient approach for moderate, rapid, and label-free detection of marine biological toxins.

Abstract:A method was established for determining tetrodotoxin (TTX) in bivalve mollusks by hydrophilic interaction liquid chromatography-triple quadrupole mass spectrometry (HILIC-MS/MS) using kasugamycin as the internal standard for quantification. Samples were subjected to ultrasonic extraction with methanol-water (1:1, v/v) containing 0.5% acetic acid, protein precipitation with acetonitrile, clean-up using a cation exchange solid phase extraction cartridge, elution with acetonitrile:water (1:1, v/v) containing 0.3% hydrochloric acid, neutralization with ammonia before HILIC-MS/MS analysis. The average recovery of the samples spiked at 3 levels ranged in 84.6%-98.1% with the relative standard deviation less than 7.2%. Using this method, the contamination of TTX in 429 bivalve mollusk samples collected in the local markets during 2018 and 2020 was investigated. The detection rates were 12.0-18.8%, following the order of oyster > mussel > clam > scallop. High contaminated oysters and mussels with TTX were found in July to August. Moreover, TTX analogs found in bivalve mollusks included 4-epiTTX, 5,6,11-trideoxyTTX, 4,9-anhydroTTX, and 5-deoxyTTX/11-deoxyTTX.

Abstract:Tetrodotoxins (TTXs) are potent marine neurotoxins involved in poisoning cases in humans after consumption of some marine organisms. A cell-based assay (CBA) providing toxicological information is a commonly used method for their detection because of its high sensitivity. However, matrix effects may interfere in the CBA. In this work, five insoluble cyclodextrin polymers, with different chemicals structures (βCDP-14-DS2, βCDP-13-DS2, βCDP-13-DS4, βCDP-14-DS4 and βCDP-N-DS5) have been investigated as novel clean-up materials for oyster extracts containing TTXs. The best recoveries have been achieved with βCDP-14-DS2 (∼100 %), which allowed exposing cells to 27-fold higher oyster matrix concentrations and decreased the LOQ down to 46 μg equiv. TTX/kg. Then, the applicability of this strategy has been demonstrated with oyster extracts from The Netherlands and comparing the results with immunoassay and liquid chromatography-tandem mass spectrometry (LC-MS/MS). This clean-up strategy combined with CBA could be implemented for TTXs detection in monitoring programs to ensure seafood safety.

We presented a facile dual-mode aptamer-based biosensor (aptasensor) for ultrasensitive fluores-cence and surface-enhanced Raman spectroscopy (SERS) detection of TTX by using gold nanoparticles (AuNPs)-embedded metal-organic framework (MOF) nanohybrids (AuNPs@MIL-101) because of their superior properties. A TTX-specific aptamer labelled with fluorescence and Raman reporter cyanine-3 (Cy3-aptamer) was selected as the recognition element and signal probe. Without immobilisation processing steps, Cy3-aptamers were effec-tively adsorbed onto the surface of AuNPs@MIL-101, thereby generating both fluorescence quenching and SERS enhancement. The preferential binding of TTX towards the Cy3-aptamer triggered the release of rigid Cy3-aptamer-TTX complexes from the AuNPs@MIL-101 surface, which resulted in recovered fluorescence signals and weakened SERS signals. Switched fluorescence and SERS intensities exhibited excellent linear relationships with logarithms of TTX concentrations of 0.01-300 ng/mL, and ultrahigh detection sensitivities of 6 and 8 pg/mL, respectively, were obtained. Furthermore, two quantitative detection approaches for TTX-spiked puffer fish and clam samples obtained satisfactory spiked recoveries and coefficient of variation (CV) values. Notably, the dual-mode aptasensor also successfully determined natural TTX-contaminated samples, showing excellent practical applications. The results indicated that this dual-mode measurement not only was ultrasensitive and simple but also markedly boosted analysis reliability and precision. This study is the first to propose a dual-mechanism AuNPs@MIL-101-based aptasensor for detection of trace TTX and provides a favourable pathway for devel-oping multimode sensing platforms for various applications.

Abstract:Saxitoxin (STX) is a highly toxic and widely distributed paralytic shellfish toxin (PSP), posing a serious hazard to the environment and human health. Thus, it is highly required to develop new STX detection approaches that are convenient, desirable, and affordable. This study presented a label-free electrolyte-insulator-semiconductor (EIS) sensor covered with a layer-by-layer developed positively charged Poly (amidoamine) (PAMAM) dendrimer. An aptamer (Apt), which is sensitive to STX was electrostatically immobilized onto the PAMAM dendrimer layer. This results in an Apt that is preferably flat inside a Debye length, resulting in less charge-screening effect and a higher sensor signal. Capacitance-voltage and constant-capacitance measurements were utilized to monitor each step of a sensor surface variation, namely, the immobilization of PAMAM dendrimers, Apt, and STX. Additionally, the surface morphology of PAMAM dendrimer layers was studied by using atomic force microscopy and scanning electron microscopy. Fluorescence microscopy was utilized to confirm that Apt was successfully immobilized on a PAMAM dendrimer-modified EIS sensor. The results presented an aptasensor with a detection range of 0.5 - 100 nM for STX detection and a limit of detection was 0.09 nM. Additionally, the aptasensor demonstrated high selectivity and 9-day stability. The extraction of mussel tissue indicated that an aptasensor may be applied to the detection of STX in real samples. An aptasensor enables marine toxin detection in a rapid and label-free manner.

Abstract:We have established an assay that relies on aptamer and isothermal amplification for the tetrodotoxin (TTX)detection. The method uses triple cycle amplification (strand displacement amplification combined with catalytic hairpin assembly) and fluorescent reporter as an output signal. Free TTX and cDNA compete for binding to aptamer-modified magnetic beads. The cDNA collected by magnetic separation then used as a primer to trigger triple cycle amplification to obtain more ssDNA. The ssDNA combined with the reporter probe, and the original quenched fluorescence can be recovered. In addition, a linear relationship between fluorescence spectrum and different target concentrations is revealed. This method allows TTX to be detected by fluorometry with a detection limit as low as 0.265 pg mL(-1). It was applied to clams and shellfish, achieving recoveries ranging from 100% to 107.33% and 99.67%-116.67%, respectively. The results were consistent with the commercial TTX ELISA kit. This assay is highly sensitive, reliable and has a good specificity. Therefore, it provides a better alternative to the standard method for quantitative detection of TTX. (C) 2020 Elsevier B.V.
