Abstract:Thrombolysis is conventionally regarded as dissolution of the fibrin matrix of thrombi by plasmin, a protease generated by plasminogen activators from its inactive precursor, plasminogen. Typically plasminogen activation occurs on the surface of the clot, where fibrin behaves as a cofactor in this process, and plasmin also initiates its proteolytic action at the fluid-solid interface. Although the basic reactions of the plasminogen/plasmin system in fluid phase are well characterized in terms of classical enzymology, they cannot explain completely the interfacial fibrinolytic events. Recently new methods have been introduced for quantitative evaluation of plasminogen activation on gel-phase fibrin and heterogenous-phase proteolysis, an overview of the new methodology is presented. Following formation of an interfacial lytic zone, fibrin dissolution proceeds through propagation of this zone to the core of the clot, which depends on diffusion and permeation phenomena affected by the composition of thrombi. Phospholipids (originating from platelets) form a diffusion barrier to the thrombolytic agents and also bind some of them; structural cellular proteins (namely myosin) interact with the fibrin fibers masking their cofactor and plasmin-cleavage sites. The contribution of these recent findings to our understanding of the limitations of current thrombolytic therapy is discussed. Finally, attention is focused on the termination of thrombus-associated proteolytic action in an environment abundant in proteinase inhibitors. Thus, combining together the interfacial events in the initiation, progress and termination of thrombolysis, a concept for modeling the thrombus as a temporary fibrinolytic compartment is presented.
Evaluation of Risk Factors for Peste des Petits Ruminants Virus in Sheep and Goats at the Wildlife-Livestock Interface in Punjab Province, Pakistan

Abstract:Peste des petits ruminants virus (PPRV) is causing infectious disease with high morbidity and mortality rate in domestic and wild small ruminants of Pakistan with valuable economical losses. The present study was carried out to investigate risk factors of PPRV in domestic small ruminants which were present in the vicinity of wildlife parks. A total of 265 sera samples (27 wild ruminants and 238 domesticated small ruminants) from apparently healthy animals from two different wildlife parks were collected and analysed for PPRV antibodies. Also, 20 nasal swabs from domestic small ruminants showing respiratory signs were collected to check for presence of PPRV antigen. Competitive ELISA revealed highest proportions of anti-PPRV antibodies in domestic small ruminants around the Wildlife Park at Lahore (35%) as compared to Faisalabad (13%), with no existence of PPRV antibodies in tested serum of wild ruminants at these parks. Higher seropositivity was observed in females (25.6%) than in males (5.1%) and in goats (34.5%) compared to sheep (11.2%). The results of N-gene based RT-PCR highlight the absence of PPRV due to lack of current PPR outbreak in the region during study period. Even though grazing was not a significant risk factor, there is still a possibility of wildlife-livestock interactions for feed and water reservoirs, resulting in spillover of PPR to wildlife. Keeping in view the high seropositivity and risk of PPR, vaccination should be adopted to avoid circulation of PPRV among wild and domestic small ruminants (sheep and goats).

Comparative Immunology Microbiology and Infectious Diseases

Abstract:Background Toxoplasma gondii is an apicomplexan protozoan parasite that infects one-third of the population of the world, including humans, animals, birds, and other vertebrates. The present investigation is the first molecular attempt in the Malakand Division of Pakistan to determine the epidemiology and phylogenetic study of Toxoplasma gondii infecting small ruminants. Methodology A total of (N = 450) blood samples of sheep were randomly collected during the study period (December 2020 to November 2021), and DNA detection was done using PCR by amplifying ITS-1 genes. SPSS.20 and MEGA-11 software were used for statistical significance and phylogenetic analysis. Results The overall prevalence of T. gondii infection among sheep was 14.44 % (65/450). A high infection rate was found in more than five-year-olds at 18.33 % (11/60). Sequencing and BLAST analysis of PCR-positive samples confirmed the presence of T. gondii . Randomly, three isolates were sequenced and submitted to GenBank under accession numbers (PP028089–PP028091), respectively. The BLAST analysis of the obtained sequences based on the ITS-1 gene showed 99 % similarities with reported genotypes found in goats of Malakand, Pakistan (PP028089) and dogs of Brazil (MF766454). The study concludes that T. gondii is notably prevalent among the sheep population in the region, emphasizing the significant role of risk factors in disease transmission across animals and potentially to humans. Further research, zoonotic potential analysis, and targeted control measures are warranted to address and manage this parasitic infection effectively.

Abstract:Background and aim: Trypanosomiasis, a parasitic infection caused by various Trypanosoma species, poses a significant threat to global livestock, affecting both human health and economic sectors. This study aimed to estimate the prevalence of Trypanosoma evansi in Southern Punjab, Pakistan, focusing on key ruminant species, including camels, cattle, buffaloes, goats, and sheep. Materials and methods: A total of 240 blood samples, comprising 48 samples from each animal species (camel, cattle, buffaloes, goat, and sheep) were collected from three districts in Southern Punjab. The collected samples were subjected to thin smear microscopy, DNA extraction, and polymerase chain reaction (PCR) amplification. The molecular characterization was conducted using the TBR primer set, which targeted repeated satellite DNA regions and the cytochrome oxidase II gene of T. evansi. Results: About 22.08% (53/240) of overall samples were positive for trypanosomiasis, with prevalence rates being 23.75% (19/80), 21.25% (17/80), and 21.75% (17/80) for districts Muzaffargarh, Lodhran, and Bahawalpur, respectively. 5.83% (14/240) of samples tested for T. evansi using PCR were positive in the districts of Muzaffargarh 7.50% (6/80), Lodhran 5.00% (4/80), and Bahawalpur 5.00% (4/80). Among the animals tested, camels had the highest positivity rate. The microscopic examination confirmed infection rates of 45.83% (22/48) for camels, 18.75% (9/48) for cattle, 8.33% (4/48) for buffaloes, 18.75% (9/48) for goats, and 18.75% (9/48) for sheep (p < 0.001). PCR results did not reveal substantial differences (p < 0.05) in prevalence: camels 12.50% (6/48), cattle 6.25% (3/48), buffaloes 0% (0/48), goats 8.33% (4/48), sheep 2.08% (1/48); while distinct disparities were detected district-wise: Muzaffargarh 23.75% (19/80), Lodhran 21.25% (17/80), and Bahawalpur 21.25% (17/80). The PCR results for these districts were insignificantly different: 7.50% (6/80), 5% (4/80), and 5% (4/80). The microscopic infection rate in camels from Bahawalpur was 56.30% (9/16). The microscopic analysis in Buffaloes reported a 6.30% (1/16) infection rate, but PCR results indicated no infections (0%) in any district. A significant difference (p < 0.001) in identifying Trypanosoma species was found between positively and negatively tested animals in both microscopic and PCR methods. Conclusion: This study emphasizes the necessity of regularly using PCR-based screening for its superior sensitivity and specificity over traditional microscopy. The varying occurrence of trypanosomiasis among districts reflects the intricate nature of this diseases epidemiology in the region. Reducing economic losses from trypanosomiasis in Southern Punjab, Pakistan, requires targeted interventions, such as vector control measures and farmer education.

Abstract:This study was designed to determine the prevalence of Cryptosporidium infections in sheep herds around tributaries in Pakistan and to evaluate the influence of potential risk factors. Four hundred forty-eight samples from 26 sheep herds were screened microscopically and then confirmed through PCR amplification of the 18S SSU rRNA gene. Our result revealed that out of 448 samples, 107 were found positive by microscopy and 77 positive by PCR, with prevalence rates of 23.88% and 17.18% in different breeds of sheep. A high statistical difference exists between the P-value (P <0.05) when comparing diarrheic and non-diarrheic sheep, breeds, ages, genders, and seasons. The highest infection rate was found in the Australian breed (8.25%); season-wise, the highest prevalence was recorded in summer. Furthermore, sheep-secreting diarrhoeal faces were 41% young sheep and lambs. This reveals that young animals are more susceptible to infection than adults, and cryptosporidial infection decreases with increasing age of the animal. This is the first report on the prevalence of Cryptosporidium in sheep sampled near the tributaries in Pakistan. The implications of this study's findings are clear; immediate action is necessary to prevent further spread and potentially devasting harm.

Abstract:A comparison has been made of the sequence of the tyrosine-containing tryptic peptide derived from the purified, unfractionated, lysine-rich histone complement of the calf thymus, rat thymus and calf liver. Two tyrosine-containing peptides, one of which is a fragment of the other, have been obtained from each tissue. The major tyrosine-containing peptide was isolated by a single electrophoretic step in about a 50%, yield. The results indicate that the sequence of amino acids around the single tyrosine present in the lysine-rich molecule is constant from tissue to tissue. However, this sequence varies among the molecular species comprising the lysine-rich histone complement of a single tissue. Within a tissue, a conservative amino acid replacement in the residue penultimate to tyrosine (from the amino terminus) has been detected. The sequence of the tyrosine-containing region was found to be Lys-Ala-Leu-Ala-AlaGly-Gly-Tyr-Asp-Val-Glu-Lys.
Gastrointestinal Parasitic Infections in Markhor Wild Goat (Capra Falconeri) at Chitral National Parks, Khyber Pakhtunkhwa, Pakistan

Abstract:Markhor (Capra Falconeri), is notably endangered ungulate native to the northern mountains of Chitral, Pakistan, specifically within the Chitral Goal National Park and Tooshe Shaha Conservancy Area (TSC), is facing a serious threat from gastrointestinal (GI) parasites. These parasites adversely affect the health of Markhor, resulting in pathological and immunological degeneration. The conservation plan for Markhor is further complicated by climate change, introducing challenges, particularly in terms of hunting trophies. This study, conducted from October 2022 to May 2023. The primary goal was to evaluate the prevalence and distribution of gastrointestinal parasites in Markhor within the Chitral national parks. A total of n=200 fecal samples were processed and analyzed using flotation, sedimentation techniques, and the McMaster slide method to identify the prevailing parasites in these samples, 64% tested positive for various helminthes eggs and larvae. The prevalence rates for specific helminth species were identified as follows: strongyles 20%, trichostrongyles 14.5%, ostertagia 11%, haemonchus 8.5%, and trichuris 10%. Notably, 44% of positive animals exhibited co-infections. The collected fecal samples detected no eggs or larvae of trematodes (flukes) or cestodes (tapeworms). An overall protozoan infection proportion of 47.5% indicated that nearly half of the fecal samples were positive for protozoan parasites. Eimeria was present in 31.5% of the samples, while Entamoebiosis caused by Entamoeba was found in 15.5%. Gastrointestinal parasites were observed to be widely distributed among Markhor, posing a significant burden of various parasites and increasing the risks of morbidity and mortality. This research highlights the health challenges resulting from parasitic infections in the Markhor (Capra Falconeri) population as substantial contributors to poor health outcomes. It calls for collaborative efforts among stakeholders to address these challenges and ensure the long-term survival of Markhor conservation. Furthermore, it highlights the importance of preserving the natural habitat of Markhor populations in the northern Chitral region to prevent infections and habitat degradation.

Molecular detection of Anaplasma, Babesia, Theileria, and Trypanosoma infection in cattle and buffaloes in India

Abstract:Tick-borne pathogens pose a significant global threat, causing substantial economic losses to the dairy industry. In India, tropical theileriosis, anaplasmosis, babesiosis, and trypanosomiasis are major hemo-parasitic diseases affecting bovines. A cross-sectional study was conducted to determine the prevalence of hemo-parasites in different farms in India. PCR assays were employed to detect carrier status, using gene targets msp1b, tams1, rap-1, ama1, and ITS1 for A. marginale, T. annulata, B. bovis, B. bigemina, and Trypanosoma species, respectively. Out of the 578 apparently healthy animals screened, 30.45% (95% CI: 26.84-34.32%) were infected with at least one hemo-parasite. Cattle showed an overall positivity of 32.87%, while buffaloes had a prevalence of 15.19%, which was statistically significant (p < 0.001). Interestingly, prevalence was higher in indigenous cattle (47.81%) compared to cross-breeds (25.53%) and exotics (14.62%), with a statistically significant difference (p < 0.001). The prevalence of hemo-parasites varied widely among the farms, ranging from 5.77 to 100%. A. marginale was the most prevalent parasite (23.70% of animals), followed by T. annulata (13.67%), Babesia species (1.90%), and Trypanosoma species (1.56%). Enzootic instability was observed in six of the eight farms, indicating a potential for future outbreaks. Co-infection was detected in 60 out of 176 animals positive for hemo-parasites, with 59 animals co-infected with A. marginale and T. annulata, and only one cross-breed cattle infected with both Anaplasma marginale and Babesia bigemina. The findings highlight the prevalence of hemo-parasites in farms, underscoring the need for whole-herd screening, treatment of infected animals, and improvement in farm management practices to prevent production losses caused by these pathogens. Abstract:Introduction: Gastrointestinal (GI) parasites are major health concerns in both humans and domestic animals. Livestock farming is one of the common livelihood practices in rural Nepal. The proximity at human and domestic animal interface increases the chances of dissemination of enteric parasites, especially those of zoonotic importance. This study was aimed at finding the parasite prevalence and risk factors in both humans and their domestic animals in Jirel community. Materials and methods: A field survey was conducted on the Jirel ethnic people and their domestic animals in Dolakha district, where a total of 152 fresh fecal samples from humans and domestic animals (cow, pigs, goats, chickens, ducks, and pigeons) were collected. The feces were examined by wet mounts and concentration techniques. A structured questionnaire survey was carried out among the local people and owners of the domestic animals to gather sociodemographic information, awareness, and hygienic practices in relation to parasite transmission. Results: The enteric parasite prevalence was found to be highest in goats (80.0%;12/15), followed by pigs (55.55%;5/9), cows (45.45%;6/11), chickens (11.7%;4/34), and humans (1.41%;1/71), while the fecal samples of ducks and pigeons did not contain any parasites. The only parasite identified in humans was Ascaris lumbricoides. Similarly, three genera of GI parasites (Eimeria sp., Strongyloides sp, and Trichuris sp.) from goats, two genera each from cow (Eimeria sp. and Strongyloides sp.), pigs (Entamoeba sp. and A. suum), and chickens (Eimeria sp. and Ascaridia galli), were detected. Conclusions: Based on the direct field observation, questionnaire survey and laboratory analysis, it is concluded that the Jirel community people are aware of health and hygiene; however, intervention measures are necessary to prevent parasitic infection in their domestic animals.

Prevalence of gastrointestinal parasites in domestic pigs from family farms in northeastern Argentina

Abstract:Domestic pigs serve as significant hosts and reservoirs for multiple parasite species, some specific to pigs and many others of zoonotic importance. This study aimed to assess the prevalence of intestinal parasites in pigs within a rural area in northeastern Argentina. We also examined demographic information, breeding conditions, and exposure factors associated with parasite presence. Pig feces were subjected to coprological examination through flotation and sedimentation techniques. Modified Ziehl-Neelsen technique was employed to examine oocysts of Cryptosporidium spp. In total, 29 family farms with pig pens were analyzed, and 42 stool samples were collected from pigs on these farms. At the farm level, the presence of at least one parasite species was recorded in 27 rural houses (93.1%). We found that 90.4% of pigs were parasitized, with a specific parasitoses of 10 species, with a maximum of six species in a single host. The most prevalent protozoa were Entamoeba spp. (57.1%) and Blastocystis sp. (45.2%), followed by Iodamoeba butschlii (33.3%), Neobalantidium coli (21.4%), coccidia (14.2%), Cryptosporidium spp. (9.5%) and Giardia spp. (2.3%). The most prevalent helminths were Strongylidae eggs (52.3%), Ascaris spp. (14.2%) and Trichuris spp. (2.3%). We advocate for an urgent need to implement a comprehensive prophylaxis program prioritizing general hygiene practices such as regular cleaning, removal of fecal material and renewal of drinking water. Additionally, vaccination and deworming protocols should be implemented. Furthermore, this study highlights the necessity for molecular-level evaluations to detect potential zoonotic genotypes of the identified protozoa.

Abstract:Pilocarpine isosteres have been synthesized and characterized with regard to their in vitro muscarinic properties. The results indicate that the carbonyl oxygen of the lactone function of pilocarpine is of primary importance for agonist activity with the ether oxygen being of lesser or secondary importance. An X-ray structure determination for the hydrogen O,O'-ditoluoyltartrate salt of thiolactone pilocarpine isostere 2a has been performed. This compound has an unusual pharmacological profile exhibiting M1-agonist selectivity as well ass presynaptic antagonism. As a result this compound is also viewed as having therapeutic potential for Alzheimer's disease. A model for the binding of pilocarpine and other muscarinic agonists to the third transmembrane helix of the human m1 muscarinic receptor has been developed.

Abstract:Background and Aim: Eimeria spp. are gastrointestinal protozoans that affect animal productivity, thereby causing symptoms that range from bloody diarrhea to death. These symptoms cause economic losses to farmers. The distribution of Eimeria spp. in cattle has, therefore, been reported to have spread widely, especially in the tropics and subtropics. Indonesia is a tropical country at high risk of Eimeria infections. This study aimed to identify the prevalence and risk factors related to the levels of eimeriosis in beef cattle originating from different geographic areas in Indonesia. Materials and Methods: Here, 817 fecal samples were collected from beef cattle in Indonesia, including 282 calves, 535 adults, 530 males, and 287 females. In addition, 156 semi-intensively and 661 intensively managed cattle were randomly collected. Then, fecal samples were analyzed by parasitology examinations. Results: Screening examination using the sugar flotation modification method showed that Eimeria spp. were prevalent in Indonesia, as 65.4% of the bacterial strain was detected. The prevalence of identified Eimeria spp. in Indonesia was highest in North Maluku (Maluku Island) (94.1%), whereas the lowest levels were observed in West Java (24.0%) (Java Island). The prevalence was also found to be higher in males (79.3%) than females (51.9%). Similarly, levels in semi-intensively managed cattle (66.7%) were higher than those subjected to intensive management (65.9%). However, its prevalence in calf and adult cattle was similar. Conclusion: Bovine eimeriosis spp. were detected at high prevalence in Indonesia, and high-level risks were observed in infected males, including those under the semi-intensive management. In addition, although the results from oocyst examinations were based on qualitative analysis, the endemicity levels of Eimeria spp. among farms in Indonesia should be considered because Eimeria spp. were distributed in most parts of Indonesia. Based on the results of this study, we provide the first information about the prevalence of bovine eimeriosis from different geographical locations in Indonesia, which have differing climates associated with the level of the existing risk factors. Hence, farmers are advised to pay more attention to strict biosecurity techniques on their farms, thereby favoring the early control of bovine eimeriosis.

Abstract:Global human population growth requires the consumption of more meat such as beef to meet human needs for protein intake. Cattle parasites are a constant and serious threat to the development of the beef cattle industry. Studies have shown that parasites not only reduce the performance of beef cattle, but also negatively affect the profitability of beef agriculture and have many other impacts, including contributing to the production of greenhouse gases. In addition, some zoonotic parasitic diseases may also threaten human health. Therefore, ongoing cattle parasite research is crucial for continual parasite control and the development of the beef cattle industry. Parasitism challenges profitable beef production by reducing feed efficiency, immune function, reproductive efficiency, liveweight, milk yield, calf yield and carcass weight, and leads to liver condemnations and disease transmission. Globally, beef cattle producers incur billions (US$) in losses due to parasitism annually, with gastrointestinal nematodes (GIN) and cattle ticks causing the greatest economic impact. The enormity of losses justifies parasitic control measures to protect profits and improve animal welfare. Geographical differences in production environment, management practices, climate, cattle age and genotype, parasite epidemiology and susceptibility to chemotherapies necessitate control methods customized for each farm. Appropriate use of anthelmintics, endectocides and acaricides have widely been shown to result in net positive return on investment. Implementing strategic parasite control measures, with thorough knowledge of parasite risk, prevalence, parasiticide resistance profiles and prices can result in positive economic returns for beef cattle farmers in all sectors.

Abstract:A stain combining periodic acid with the fluorochrome acriflavine (the fluorescent periodic acid-acriflavine [FPAA] reaction) was applied to specimens of normal adult human skin for study of the subepidermal basement membrane (BM). Advantages over conventional PAS technique were enhanced delineation of structural details and color contrast. In specimens from all body regions studied the BM showed variations in thickness, reticulation or stratification, and defects ranging from fragmentation to absence. These variations were least pronounced in plantar skin. Correlations of BM changes with basal layer morphology were noted.
The Prevalence of Infection with Gastrointestinal Helminths in Free-Range Poultry of Zabol, Iran

Abstract:Background: As an ecological and ethical method employed for poultry farming, free-range chicken keeping is growing and is important in fostering rural farmers' livelihoods and community prosperity. This study aimed to determine the nature and prevalence of helminth infections in native poultry in Zabol, Iran. Methods: Between 2022 and 2023, 160 chickens were acquired and slaughtered ethically, and then their gastrointestinal tracts were taken and formalin preserved in Zabol, Iran. The parasites were isolated from gastrointestinal mucus in the laboratory and characterized using a microscope and specific methods such as Acetocarmine staining. Results: Among 160 dissected birds, 92 (57.5%) were presented with gastrointestinal parasites, with 64 (40%) containing cestodes, 16 (10%) containing nematodes, and 12 (7.5%) infected by both. Observations were made of seven different species. Ascaridia galli (A. galli) (10%), Subulura brumpti (7.5%), and Heterakis gallinarum (3.12%) represent the three main nematode species. While Raillietina tetragona (R. tetragona) (33.12%) was the most prevalent cestode, R. echinobothrida (6.25%), R. magninumida (5%), and Cotugnia digonopora (3.12%) were less common. Conclusion: The study revealed that free-range chickens in the arid Zabol region had a high prevalence of gastrointestinal parasites. There is a strong correlation between chicken foraging behavior and exposure to contaminated soil, with certain nematodes and cestodes constituting the most prevalent, negatively affecting bird growth, productivity, and health. Therefore, improving poultry welfare by applying parasite control measures and implementing appropriate management strategies is essential.

Abstract:Ectoparasite infestations are one of the major problems affecting goat and sheep farming. Disease resulting from these infestations can cause changes in physical appearance, such as severe lesions on the skin, and economic consequences in the form of significantly reduced selling prices. This study aimed to determine the prevalence of ectoparasites in the Boyolali district, Central Java, Indonesia. A total of 651 sheep and goats were surveyed in this study. The parasites were collected via skin scraping, twister, or manually from clinically infected goats and sheep in traditional farms. All of the ectoparasites collected were successfully identified. The prevalence of ectoparasites in ruminants in Boyolali was 97.8% (637/651). The species make-up was as follows: Bovicola caprae 97.8% (637/651), Linognathus africanus 39% (254/651), Haemaphysalis bispinosa 3.5% (23/651), Ctenocephalides spp. 0.2% (1/651), and Sarcoptes scabiei 5.2% (34/651). The predilection sites were in the face, ear, and leg areas, and in the axillary, dorsal, abdomen, and scrotum regions of the surveyed animals. An evaluation of farmers' attitudes to ectoparasites was performed using a questionnaire. The findings of this study imply that animals in the investigated area are highly exposed to ectoparasite infestations. Given the importance of ectoparasites in both livestock and human communities, specifically in the health domain, more research into appropriate control strategies is necessary.

Abstract:Sheep production is a major source of income for resource poor farmers of the world. The main purposeof sheep raising is to fulfill the needs for mutton, milk, and wool production. Parasitic diseases, one ofthe principal hindrances in the development of commercial livestock business, are facilitated throughfavorable climatic conditions and lack of awareness. A variable prevalence ranging from 18.63 to 100% ofdifferent gastrointestinal parasite (GI) species in sheep were reported from different parts of the world.Incidence, prevalence, and intensity of parasitic infections is a multifactorial phenomenon and depends onenvironment, geographic location, pasture condition, host characteristics, grazing habits, and nutritionalstatus. Prevalence of parasitic infections not only adversely affects animal health but also causes hugeeconomic losses up to billions of rupees by decrease in milk, meat and wool production, retarded growth,parasite control measures and death of animals. For the control of GI parasites in small ruminants especiallykept by resource-poor farmers, it is better to identify the burden and types of helminths along withspecific risk factors associated with helminthosis of a specific area. Factors responsible for developmentof resistance are lavish use of anti-parasitic drugs, poor efficacy of anti-parasitic agents, inadequate doselevel, low protein diet and environmental toxicity. Due to limitations of chemotherapy during the pastdecade, use of plants with anthelmintic properties (ethnoveterinary medicine) are under considerationaround the world. A number of plant species have been identified with anti-parasitic properties that maydisturb the regular physiological functions of parasites. Use of plants with anthelmintic properties isconsidered the best choice for control of parasites in the present scenario. In this review, sheep production,GI parasitic infection of sheep, medicinal plants, and their uses for control have been described.
