Abstract:Melatonin (MLT) is a methoxyindole that has potent antioxidant actions, anti-inflammatory, and antiapoptotic capacity. However, its in vitro antibacterial capacity has been the least studied of its properties. Dimethylsulfoxide (DMSO) has been the most used solvent for these tests, but it shows an antimicrobial effect if it is not dissolved. CyreneTM is a new solvent that has emerged as an alternative to DMSO. Therefore, this study aimed to determine the antimicrobial capacity of MLT by MIC assays, using CyreneTM as a solvent. Likewise, the solubility of MLT in this solvent and whether it exerted any effect on bacterial growth at different percentages was also determined. Different dilutions of MLT in CyreneTM with different concentrations, were prepared. No growth inhibition caused by MLT was observed. The growth inhibition observed was because of CyreneTM. The maximum amount of MLT that can be diluted in 100% Cyrene is 10 mg/mL, but this percentage of solvent shows a bactericidal effect. Therefore, it must be dissolved at 5% to avoid this effect, so only 4 mg/mL of MLT can be diluted in it. Therefore, if no other solvents are available, the in vitro antibacterial role of MLT cannot be adequately assessed.

Abstract:A new melatonin sulfonate derivative sodium 4-(3-(2-acetamidoethyl)-5-methoxy-1H-indol-1-yl) butane-1-sulfonate (MLTBS) with higher water solubility (695 times) and lower cytotoxicity than natural melatonin (MLT) was synthesized, yet with the same sleep aid function. The poor solubility of MLT in water has been improved with a simple chemical reaction, which solves the poor solubility of melatonin in water, overcoming the safety problem caused by adding organic reagents such as dimethyl sulfoxide (DMSO) and ethanol to increase the solubility. Moreover, the modified MLT still has the same sleep aid effect as the natural MLT and higher biological safety. As a novel potential drug for sleep aid, the new MLT derivative could also flourish the application and research of this molecule in medicine and biology.

Abstract:Melatonin is one of the most significant hormones that have an impact on biological activities. Melatonin is a strange molecule that humans and animals produce in their pineal glands. A small gland found in the center of the brain is known as the pineal body. This research aims to synthesize a new melatonin derivative, assess its biological properties, and design a new HPLC analysis method. In the first stage, benzyl chloride was used to make a novel derivative of this chemical. The melting point was used to check the purity of the produced derivative. The molecular structure of the substance has been confirmed using: Fourier transform infrared (FT-IR), Carbon-13(C13)nuclear magnetic resonance (13C-NMR), Differential scanning calorimetry (DSC) and Proton nuclear magnetic resonance (1H-NMR) spectral studies, and elemental analysis. The antibacterial, antioxidant, and antitumor capabilities of the produced melatonin derivative were then studied using various microbe and antioxidant methods in the second stage. Although the new derivative gave a high antioxidant effect in APTS-99.03% and OH- 99.29% than BHT control, the produced derivative has a relatively negative impact on microorganisms (Staphylococcus aureus 7 mm) and moderate anticancer effect. The method for analyzing the novel derivative in biological samples using HPLC–UV was devised as the final stage using %64 acetonitrile. A PH 4 shows a higher peak and a shorter retention time (3.52 min and absorbance 250 nm).

Abstract:Abstract Antimicrobial resistance (AMR) is a current major health issue, both for the high rates of resistance observed in bacteria that cause common infections and for the complexity of the consequences of AMR. Pathogens like Staphylococcus aureus , Pseudomonas aeruginosa , Klebsiella pneumoniae , Mycobacterium tuberculosis among others are clear examples of antibiotic-resistant threats. Biosurfactants have recently emerged as a potential new generation of anti-adhesive and anti-biofilm agents; mannosylerythritol lipids (MELs) are biosurfactants produced by a range of fungi. A range of structural variants of MELs can be formed and the proportion of each isomer in the fermentation depends on the yeast used, the carbon substrate used for growth and the duration of the fermentation. In order to allow assessment of the possible functions of MELs as antimicrobial molecules, small quantities of MEL were produced by controlled fermentation. Fermentations of the yeast Pseudozyma aphidis using rapeseed oil as a carbon source yielded up to 165 g MELs /kg Substrate . The MELs formed by this strain was a mixture of MEL-A, MEL-B, MEL-C and MEL-D. The MELs produced were tested against S. aureus ATCC 6538 on pre-formed biofilm and on co-incubation biofilm experiments on silicone discs; showing a disruption of biomass, reduction of the biofilm metabolic activity and a bacteriostatic/bactericidal effect confirmed by a release of oxygen uptake $$p_{{{\text{O}}_{2} }}$$ p O 2 , the reduction of citrate synthase activity and scanning electron microscopy. The results show that MELs are promising antimicrobial molecules for biomedical technological applications that could be studied in detail in large-scale systems and in conjunction with animal tissue models.

Abstract:To our knowledge, this is the first study on the influence of water, ethyl acetate and methanol as the extraction solvent upon the bioactive compound composition, antioxidant capacities and enzyme inhibitory activities of Marrubium lutescens isolates. The contents of the total phenolic compounds varied between 9.94 and 54.80 mg gallic acid equivalent/g extract and the level of total flavonoids were between 4.45 and 27.20 mg quercetin equivalent/g extract. Twenty-four compounds were identified using liquid chromatography – electrospray ionization tandem mass spectrometry (LC-ESI-MS/MS). Rosmarinic acid, luteolin 7-glucoside, chlorogenic acid, caffeic acid and hyperoside were the primary compounds present for all of the used solvents. The most compounds (23) were detected in the methanol extract. Six antioxidant assays revealed that M. lutescens extracts showed interesting antioxidant activities with a significant variation between the extracts. The results revealed that the highest tyrosinase and α-amylase inhibitory activities was detected in the ethyl acetate extract. This work confirmed that the nature of solvent influenced the levels and the types of the extracted biomolecules. Moreover, the obtained results suggest that M. lutescens may be a source of bioantioxidants for industrial additives.

Abstract:In this current research work has a main aim to develop a novel Melatonin (MLT: BCS class II drug)-loaded nanoemulsion for the improvement in solubility. A developed novel-NE was to evaluate and compare the efficacy of Melatonin-loaded mucoadhesive nanoemulsion (CS-MLT-NE) with Melatonin-loaded nanoemulsion (MLT-NE) and MLT-solution (MLT-S) pharmacodynamically and also determine the quantity of MLT in the brain after the nasal administration. MLT-NE was prepared with the help of an advanced ultrasonication method after the screening of excipients and the development of pseudoternary phase diagram study. We got the optimized nanoformulation via three-factor four-level central-composite design (CCD). We optimized %oil, %Smix, and sonication time (second) based on selected independent variables. Independent and dependent variables were selected and optimized on the basis constraints oil (2.0%), 10%Smix, and 120 s sonication time. The dependent variables found the experimental their values 49.37 ± 4.09 nm (Y1: Globule size), 0.257 ± 0.0004 (Y2: PDI), − 16.20 ± 1.10 mV (Y3: Zeta Potential), and 98.03 ± 0.59% (Y4: %Transmittance), respectively. The shape of optimized-MLT-NE was found spherical with the pH, viscosity, and drug content (7.40 ± 0.08, 37.86 ± 7.01 cp, and 98.27 ± 0.17%), respectively. Zeta-Potential of CS-MLT-NE was altered from −ve to +ve after the addition of CS followed small increment of globule-size. CS-MLT-NE showed the great mucoadhesive nature as compared to MLT-NE, and MLT-S with a retention time of 0.641 min and m/z of 233.20/174.20 for MLT. A linear range was established from 0.50 to 1500.0 ng mL−1 with %accuracy (92.41–98.61%) and inter- and intraday %precision (2.49–4.93%). Cmax was enhanced (AUC)0−24 which was greatly significant (p < 0.001) in the rat’s brain when it compared with i.n. and intravenous-treated group. Moreover, FST (forced swimming, climbing, and immobility) and locomotor activity test of CS-MLT-NE (i.n.) showed highly improved behavioral analysis as compared to the MLT-NE and MLT-S after 24.0 h of study. CS-MLT-NE showed highly significant role (p < 0.001) for the improvement in brain bioavailability and treatment of depression.Graphical abstract

Abstract:In this work, the dispersive solid phase extraction (dSPE) of melatonin using graphene (G) mixtures with sepiolite (SEP) and bentonite (BEN) clays as sorbents combined with fluorescence detection has been investigated. The retention was found to be quantitative for both G/SEP and G/BEN 4/96 and 10/90 w/w mixtures. G/clay 4/96 w/w mixtures were selected to study the desorption process since the retention was weaker, thus leading to easier desorption. MeOH and aqueous solutions of the nonionic surfactant Brij L23 were tested as desorbents. For both clays and an initial sample volume of 25 mL, a percentage of melatonin recovery close to 100% was obtained using 10 or 25 mL of MeOH as desorbent. Further, using a G/SEP mixture, 25 mL as the initial sample volume and 5 mL of MeOH or 60 mM Brij L23 solution as the desorbent, recoveries of 98.3% and 90% were attained, respectively. The whole method was applied to herbal tea samples containing melatonin, and the percentage of agreement with the labeled value was 86.5%. It was also applied to herbal samples without melatonin by spiking them with two concentrations of this compound, leading to recoveries of 100 and 102%.

Abstract:Melatonin (MLT) is a natural indolic hormone and its antioxidant activity has been reported through various mechanisms. MLT interacts with the estrogen signaling pathway by inhibiting aromatase enzyme, and it has been suggested that these new compounds may have endocrine‐related adverse effects due to their selective estrogen modulator activity. In this study, we investigated the aromatase inhibitory and estrogen receptor agonist/antagonist effects as well as possible antioxidant activity of MLT analogue 5‐floro indole derivatives for being potential drug candidates in the prevention and treatment of oxidative stress related diseases. Among the 34 compounds tested, three compounds showed antioxidant effect through radical scavenging activity and reducing the formation of intracellular reactive oxygen species. They also showed varying potencies of antiestrogenic and/or aromatase inhibitory activity which should be considered as a potential untargeted effect of MLT and its analogues.

Abstract:Sulfur and nitrogen mustards, bis(2-chloroethyl)sulfide and tertiary bis(2-chloroethyl) amines, respectively, are vesicant warfare agents with alkylating activity. Moreover, oxidative/nitrosative stress, inflammatory response induction, metalloproteinases activation, DNA damage or calcium disruption are some of the toxicological mechanisms of sulfur and nitrogen mustard-induced injury that affects the cell integrity and function. In this review, we not only propose melatonin as a therapeutic option in order to counteract and modulate several pathways involved in physiopathological mechanisms activated after exposure to mustards, but also for the first time, we predict whether metabolites of melatonin, cyclic-3-hydroxymelatonin, N1-acetyl-N2-formyl-5-methoxykynuramine, and N1-acetyl-5-methoxykynuramine could be capable of exerting a scavenger action and neutralize the toxic damage induced by these blister agents. NLRP3 inflammasome is activated in response to a wide variety of infectious stimuli or cellular stressors, however, although the precise mechanisms leading to activation are not known, mustards are postulated as activators. In this regard, melatonin, through its anti-inflammatory action and NLRP3 inflammasome modulation could exert a protective effect in the pathophysiology and management of sulfur and nitrogen mustard-induced injury. The ability of melatonin to attenuate sulfur and nitrogen mustard-induced toxicity and its high safety profile make melatonin a suitable molecule to be a part of medical countermeasures against blister agents poisoning in the near future.
In silico Exploration of a Novel ICMT Inhibitor with More Solubility than Cysmethynil against Membrane Localization of KRAS Mutant in Colorectal Cancer

Abstract:Background: ICMT (isoprenylcysteine carboxyl methyltransferase) is an enzyme that plays a key role in the post-translational modification of the K-Ras protein. The carboxyl methylation of this protein by ICMT is important for its proper localization and function. Cysmethynil (2-[5-(3-methylphenyl)-l-octyl-lH-indolo-3-yl] acetamide) causes K-Ras mislocalization and interrupts pathways that control cancer cell growth and division through inhibition of ICMT, but its poor water solubility makes it difficult and impractical for clinical use. This indicates that relatively high amounts of cysmethynil would be required to achieve an effective dose, which could result in significant adverse effects in patients. Objective: The general objective of this work was to find virtually new compounds that present high solubility in water and are similar to the pharmacological activity of cysmethynil. Methods: Pharmacophore modeling, pharmacophore-based virtual screening, prediction of ADMET properties (absorption, distribution, metabolism, excretion, and toxicity), and water solubility were performed to recover a water-soluble molecule that shares the same chemical characteristics as cysmethynil using Discovery Studio v16.1.0 (DS16.1), SwissADME server, and pkCSM. Results: In this study, ten pharmacophore model hypotheses were generated by exploiting the characteristics of cysmethynil. The pharmacophore model validated by the set test method was used to screen the "Elite Library®" and "Synergy Library" databases of Asinex. Only 1533 compounds corresponding to all the characteristics of the pharmacophore were retained. Then, the aqueous solubility in water at 25°C of these 1533 compounds was predicted by the Cheng and Merz model. Among these 1533 compounds, two had the optimal water solubility. Finally, the ADMET properties and Log S water solubility by three models (ESOL, Ali, and SILICOS-IT) of the two compounds and cysmethynil were compared, resulting in compound 2 as a potential inhibitor of ICMT. Conclusion: According to the results obtained, the identified compound presented a high solubility in water and could be similar to the pharmacological activity of cysmethynil.

Abstract:Dihydrolevoglucosenone (Cyrene) is a bio-based molecule, derived in two simple steps from cellulose, which demonstrates significant promise as a dipolar aprotic solvent. The dipolarity of dihydrolevoglucosenone is similar to NMP, DMF and sulpholane. Dihydrolevoglucosenone demonstrates similar performance to NMP in a fluorination reaction and the Menschutkin reaction.

Abstract:This study explores a binary solvent system composed of biobased Cyrene and its derivative Cygnet 0.0 for application in membrane technology and in biocatalytic synthesis of polyesters. Cygnet-Cyrene blends could represent viable replacements for toxic polar aprotic solvents. The use of a 50 wt % Cygnet-Cyrene mixture makes a practical difference in the production of flat sheet membranes by nonsolvent-induced phase separation. New polymeric membranes from cellulose acetate, polysulfone, and polyimide are manufactured by using Cyrene, Cygnet 0.0, and their blend. The resultant membranes have different morphology when the solvent/mixture and temperature of the casting solution change. Moreover, Cyrene, Cygnet 0.0, and Cygnet-Cyrene are also explored for substituting diphenyl ether for the biocatalytic synthesis of polyesters. The results indicate that Cygnet 0.0 is a very promising candidate for the enzymatic synthesis of high molecular weight polyesters.
Phytochemical analysis, biological activities of methanolic extracts and an isolated flavonoid from Tunisian Limoniastrum monopetalum (L.) Boiss: an in vitro and in silico investigations

Abstract:In recent years, due to the dramatic increase of the bacteria resistance to antibiotics and chemotherapeutic drugs, an increasing importance is given to the discovery of novel bioactive molecules, more potent than those in use. In this contest, methanol extracts of different parts of the medicinal plant Limoniastrum monopetalum (L.) Boiss. (Plumbaginaceae), widely occurring in Tunisia, were prepared to evaluate the antimicrobial and antiproliferative activities. The methanol extract of the roots showed the highest antibacterial activity against E. coli, S. aureus and E. faecalis, whereas the stem extract exhibited the highest antiproliferative effects towards a Hela cell line. Analysis of volatile fractions, using gas chromatography-mass spectrometry (GC-MS) and gas chromatography-flame ionization detector (GC-FID) techniques, led to the identification of camphor as the most abundant constituent, which represented from 84.85 to 99.48% of the methanol extracts. Multiple chromatographic separation of the methanol leaf extract afforded the flavonoid maeopsin-6-O-glucoside (S1) and a few fractions that were subjected to biological activity assays. One fraction exhibited interesting antibacterial activity against E. coli and E. faecalis (MIC values of 62.5 and 78.12 µg/mL, respectively), and antiproliferative effects against Hela and A549 cells (IC50 = 226 and 242.52 μg/mL, respectively). In addition, in silico studies indicated that maesopsin-6-O-glucoside, which was moderately active against Staphylococcus aureus, strongly interacted with the active site of the accessory gene regulator protein A (AgrA) of Staphylococcus aureus.
Dihydrolevoglucosenone (Cyrene), a Biobased Solvent for Liquid–Liquid Extraction Applications

Abstract:Dihydrolevoglycosenone, commercially known as Cyrene, is a versatile biobased solvent reported for various applications including being a medium of chemical reactions and membrane manufacture. In this work, application of Cyrene in liquid–liquid extractions has been investigated. Four ternary systems have been assessed at 298.15 K, 323.15 K, and 348.15 K, keeping Cyrene and methylcyclohexane constant and changing the third compound to toluene, cyclohexanol, cyclohexanone, and cyclopentyl methyl ether. Studying these selected ternary systems yielded an indication of applicability of Cyrene in related industrial separation processes such as aromatic/aliphatic separations and in separations of oxygenates in the cyclohexane oxidation process. Key factors are biphasic system formation, distribution coefficients, and selectivity. All ternary systems showed type I liquid–liquid phase behavior with a selective extraction of ternary components from methylcyclohexane by Cyrene. The highest selectivity at 298.15 K was found for cyclohexanol with 61.4 ± 4.33, followed by cyclohexanone with 44.1 ± 8.63, while toluene and cyclopentyl methyl ether had a selectivity of, respectively, 12.0 ± 0.89 and 6.4 ± 0.08. Although Cyrene is applicable in all four ternary systems, the miscibility gap is narrow for the oxygenated species, indicating a limited operation window for liquid–liquid extraction which will be more severe at higher temperatures. Overall, the studies showed that there are certainly application windows for Cyrene. In the case of oxygenate extraction, a process with Cyrene appears energy-saving because the energy duty appears to be lower than when using water, the best alternative solvent, which is a low boiling solvent for this purpose.Cyrene extracts solutes more polar than methylcyclohexane selectively from the hydrocarbon phase.This article has not yet been cited by other publications.

DMSO is widely used to solubilize drugs of therapeutic applications and studies indicated that 10% v/v concentration did not modify culture viability when used to treat human peripheral blood mononuclear cells (PBMC). However, some DMSO concentrations could influence lymphocyte activation and present anti-inflammatory effects. Therefore, the objective of this study was to evaluate the effect of DMSO on lymphocyte activation parameters. Cell viability analysis, proliferation, and cytokine production were performed on PBMC from six healthy subjects by flow cytometry. The results indicated that 2.5% v/v DMSO concentrations did not modify lymphocytes viability. DMSO at 1% and 2% v/v concentrations reduced the relative proliferation index of lymphocytes and at 5% and 10% v/v concentrations reduced the percentage of total lymphocytes, cluster of differentiation 4⁺ (CD4⁺) T lymphocytes and CD8⁺ T lymphocytes interferon-γ (IFN-γ), tumor necrosis factor-α (TNF-α) and interleukin-2 (IL-2) producers. Thus, it was concluded that DMSO has an in vitro anti-inflammatory effect by reducing lymphocyte activation demonstrated with proliferation reduction and the decrease of cytokine production.

Abstract:Sulfamerazine (SMR) is a drug used as an antibacterial agent in the treatment of some pathologies, such as bronchitis, prostatitis and urinary tract infections. Although this drug was developed in 1945 and, due to its toxicity, was partially displaced by penicillin, due to the current problem of bacterial resistance, compounds such as SMR have regained validity. In this context, the thermodynamic study of SMR in cosolvent mixtures of acetonitrile (MeCN) + ethanol (EtOH) at nine temperatures (278.15–318.15 K) is presented. The solubility of SMR was determined by UV–Vis spectrophotometry, following the guidelines of the shake-flask method. The solubility process was endothermic in all cases; thus, the minimum solubility was reached in pure EtOH at 278.15 K, and the maximum solubility was reached in pure MeCN at 318.15 K. Both the solution process and the mixing process were entropy-driven. On the other hand, the solubility data were modeled by using the van't Hoff–Yalkowsky–Roseman model, obtaining an overall average relative deviation of 3.9 %. In general terms, it can be concluded that the solution process of SMR in {MeCN (1) + EtOH (2)} mixtures is thermodependent, favored by the entropy of the solution and mixture; additionally, the van't Hoff–Yalkowsky–Roseman model allows very good approximations to be obtained and is a simple model that starts from only four experimental data.

Abstract:In this work, the use of a hydrophobic natural deep eutectic solvent (NADES) as supported liquid membrane (SLM) for hollow fiber liquid-phase microextraction (HF-LPME) of sulfonamides is proposed. Different NADES based on l-menthol and naturally occurring organic acids (citric acid, formic acid, acetic acid, decanoic acid, nonanoic acid, lactic acid, valeric acid, isobutyric acid, hexanoic acid, octanoic acid, and 2-methylbutyric acid) were prepared at different molar ratios. Important solvent properties to work as SLM were studied such as stability in the fiber and compatibility with HPLC. Then, the ability of the prepared NADES to extract 8 antibiotics of the family of sulfonamides (SAs) from water by HF-LPME was studied. From them, the combination of formic acid:L-menthol (1:1) was selected as optimum since it provided a better and more balanced extraction of SAs. The optimized HF-LPME procedure was applied to the analysis of an artificial water containing humic acids, tap and river water samples, showing a good clean-up ability for all the tested samples. Calibration curves showed good linearity for all the SAs and samples with linear regression coefficients (r2) greater than 0.992. Relative recoveries ranged from 53 to 121%, with relative standard deviation values between 5 and 21%, depending upon the analyte and type of water sample. The limits of detection of the method varied from 0.63 to 2.85 µg/L for tap water, 0.70 to 3.23 µg/L for the Jarama river and 0.61 to 2.52 µg/L for the Manzanares river. Besides, the proposed method can be considered sustainable, according to AGREEprep tool, and greener than reported methods in the literature based in other microextraction techniques.

Abstract:Dimethyl sulfoxide (DMSO), an organosulfur compound, is widely used as the gold standard solvent in biological research. It is used in cell culture experiments and as a component of formulations in in vivo studies. Unfortunately, parameters related to sulfur metabolism are often not taken into account when using DMSO. Therefore, in this work we aim to show that the addition of DMSO to the culture medium (even in amounts commonly considered acceptable) alters some parameters of sulfur metabolism. For this study, we used three cell lines: a commercially available Caco-2 line (HTB-37, ATCC) and two lines created as part of our early studies (likewise previously described in the literature) to investigate the anomalies of sulfur metabolism in mucopolysaccharidosis. As the negative effects of DMSO on the cell membrane are well known, additional experiments with the partial loading of DMSO into polymerosomes (poly(ethylene glycol) methyl ether-block-poly(lactide-co-glycolide), PEG-PLGA) were performed to eliminate these potentially disruptive effects. The results show that DMSO is a source of interference in studies related to sulfur metabolism and that there are not just simple effects that can be corrected in the final result by subtracting control values, since complex synergisms are also observed.
