Abstract:Microplastics (MPs), the emerging environmental pollutants, have attracted global attention due to the potential public health challenge and ecological security risk. Recent studies suggested liver as a vulnerable organ to MPs exposure, evidenced by abnormal hepatic lipid metabolism upon MPs intake in multiple animal species. How-ever, the specific changes of lipid metabolism in mammalian livers, as well as the underlying mechanisms, remain to be elucidated. In the present study, C57BL/6 mice were randomly assigned to normal drinking water or drinking water containing 100 mu g L-1 or 1000 mu g L-1 polystyrene (PS) MPs for 8 weeks. MPs exposure exerted no significant effect on body weight, serum triglyceride or total cholesteryl esters. However, mice showed impaired glucose tolerance and hepatic lipid deposition in response to high-dose MPs administration. Further lipidomic analysis showed significant alteration in hepatic lipid species particularly with free fatty acids (FFAs) and tri-acylglycerols (TAGs) in mice exposed to MPs. Meanwhile, the liver transcriptional profile indicated MPs exposure-induced differentially expressed genes (DEGs) were enriched in pathways of lipid metabolism and unfolded protein response. Furthermore, most altered lipid species were significantly correlated with DEGs enriched in lipid metabolic signaling. These findings provide lipidomic and transcriptional signatures of liver in response to MPs exposure, which will shed light on further understanding of the metabolic toxicity of MPs.
Microfluidic-based in vitro thrombosis model for studying microplastics toxicity

Abstract:The potential impact of microplastics (MPs) on health has caused great concern, and a toxicology platform that realistically reproduces the system behaviour is urgently needed to further explore and validate MP-related health issues. Herein, we introduce an optically assisted thrombus platform to reveal the interaction of MPs with the vascular system. The risk of accumulation has also been evaluated using a mouse model, and the effect of MPs on the properties of the thrombus are validated via in vitro experiments. The microfluidic system is endothelialized, and the regional tissue injury-induced thrombosis is then realized through optical irradiation. Whole blood is perfused with MPs, and the invasion process visualized and recorded. The mouse model shows a cumulative risk in the blood with continuous exposure to MPs (P-value < 0.0001). The on-chip results show that MP invasion leads to decreased binding of fibrin to platelets (P-value < 0.0001), which is consistent with the results of the in vitro experiments, and shows a high risk of thrombus shedding in real blood flow compared with normal thrombus. This work provides a new method to further reveal MP-related health risks.
Exploring the Trophic Transfer and Effects of Microplastics in Freshwater Ecosystems: A Focus on Bellamya Aeruginosa to Mylopharyngodon Piceus

Abstract:Microplastics (MPs) can enter aquatic food webs through direct ingestion from the environment or indirectly via trophic transfer, but their fate and biological effects within local freshwater food chains remain largely unexplored. In this study, we conducted the first investigation on the trophic transfer and impacts of fluorescently labeled polystyrene microplastics (PS-MPs) (100-nm and 10-μm) in a model freshwater food chain consisting of the snail Bellamya aeruginosa and the commercially important fish Mylopharyngodon piceus, both prevalent in Chinese freshwater ecosystems. Quantitative analysis revealed substantial accumulation of MPs in B. aeruginosa, reaching an equilibrium state within 12 h of exposure. While steady-state was not observed, a pronounced time-dependent bioaccumulation of MPs was evident in M. piceus over a five-week period following dietary exposure through the consumption of contaminated B. aeruginosa. Notably, MPs of both sizes underwent translocation from the gastrointestinal tract to the muscle tissue in M. piceus. High-throughput sequencing of the gut microbiota revealed that exposure to 100-nm MPs significantly altered the microbial community composition in M. piceus, and both particle sizes led to increased relative abundance of potentially pathogenic bacterial genera. Our findings provide novel insights into the trophic transfer, tissue accumulation, and biological impacts of MPs in a model freshwater food chain, highlighting the need for further research to assess the ecological and food safety risks associated with microplastic pollution in freshwater environments.

Abstract:Microplastics (MPs) are pervasive across ecosystems, presenting substantial risks to human health. Developing a comprehensive review of MPs is crucial due to the growing evidence of their widespread presence and potential harmful effects. Despite the growth in research, considerable uncertainties persist regarding their transport dynamics, prevalence, toxicological impacts, and the potential long-term health effects they may cause. This review thoroughly evaluates recent advancements in research on MPs and their implications for human health, including estimations of human exposure through ingestion, inhalation, and skin contact. It also quantifies the distribution and accumulation of MPs in various organs and tissues. The review discusses the mechanisms enabling MPs to cross biological barriers and the role of particle size in their translocation. To ensure methodological rigor, this review adheres to the PRISMA guidelines, explicitly detailing the literature search strategy, inclusion criteria, and the quality assessment of selected studies. The review concludes that MPs pose significant toxicological risks, identifies critical gaps in current knowledge, and recommends future research directions to elucidate the prolonged effects of MPs on human health. This work aims to offer a scientific framework for mitigating MP-related hazards and establishes a foundation for ongoing investigation.

Abstract:Microplastics (MPs) can pose ecological risk to the environment and have the potential to negatively affect human health, raising serious public concerns. It is recognized that MPs could act as a vector for various environmental pollutants including heavy metals and potentially influencing their mobility, fate, and bioavailabilty in the environment. However, knowledge on the mechanisms underpinning the interaction processes between MPs and heavy metals is far from clear. This review discusses the effects of MPs on the adsorption/desorption, speciation and bioavailability, and toxicity of various heavy metals. The present review also systematically identifies the environmental factors (e.g., pH, ionic strength, and organic matters) that could affect their interaction processes. This work aims to establish a meaningful perspective for a comprehensive understanding of the indirect ecological risks of MPs as vectors for contaminants. The work also provides a reference for the development of better regulatory strategies in mitigating the negative effects caused by the co-existence of MPs and heavy metals.

Abstract:Microplastics (MPs) are a significant environmental health issue and increasingly greater source of concern. MPs have been detected in oceans, rivers, sediments, sewages, soil and even table salts. MPs exposure on marine organisms and humans has been documented, but information about the toxicity of MPs in mammal is limited. Here we used fluorescent and pristine polystyrene microplastics (PS-MPs) particles with two diameters (5 μm and 20 μm) to investigate the tissue distribution, accumulation, and tissue-specific health risk of MPs in mice. Results indicated that MPs accumulated in liver, kidney and gut, with a tissue-accumulation kinetics and distribution pattern that was strongly depended on the MPs particle size. In addition, analyses of multiple biochemical biomarkers and metabolomic profiles suggested that MPs exposure induced disturbance of energy and lipid metabolism as well as oxidative stress. Interestingly, blood biomarkers of neurotoxicity were also altered. Our results uncovered the distribution and accumulation of MPs across mice tissues and revealed significant alteration in several biomarkers that indicate potential toxicity from MPs exposure. Collectively, our data provided new evidence for the adverse consequences of MPs.

Abstract:The effect of microplastics on the ecological environment and human health has become a topical issue， and research on the risks and harmful effects of MPs on human health in particular has attracted widespread attention. Due to the characteristics of small size， low degradability， and easy migration， MPs continuously migrate from the environment to the human body， and their main exposure pathways are oral ingestion， inhalation， and dermal contact， with the main exposure media being food， drinking water， dust， personal care products， etc. MPs have been detected in organs， fluids， and excreta of digestive， respiratory， cardiovascular， reproductive systems， etc. The abundance range of MPs in the human body is 0-1 206.94 particles per gram. After entering the human body， MPs can cause cytotoxicity， mitochondrial toxicity， DNA damage， cell membrane damage， and other effects on human cells and organs， leading to serious consequences such as local inflammation， ecological imbalance， metabolic disorders， etc.， in various systems. Owing to their small specific surface area， they can also adsorb pollutants such as heavy metals， organic pollutants， antibiotics， pathogens， and harmful microorganisms， causing combined toxicity and immunotoxicity. In the end， we highlighted general deficiencies in existing studies and provided directions for future research on the influence of MPs on human health.
Effects of Microplastic (MP) Exposure at Environmentally Relevant Doses on the Structure, Function, and Transcriptome of the Kidney in Mice

Abstract:As a common emerging environmental pollutant, microplastics (MPs) have been detected in a variety of environmental media and human bodies. The potential toxic effects and mechanisms of MPs need to be revealed urgently. MPs can be deposited in the kidney, and exposure to high doses of MPs can cause nephrotoxicity in experimental animals. In this study, we investigated the effects of exposure to polystyrene microplastics (PS-MPs) at environmentally relevant doses (0.1 and 1 mg/L) on kidney structure, function, and transcriptome in mice. We found that mice exposed to PS-MPs in drinking water for eight weeks had no change in body weight or kidney coefficient. PS-MPs administration decreased the levels of blood urea nitrogen (BUN) in mice, while serum creatinine (CRE) and uric acid (UA) concentrations were unaffected. Through using periodic acid–Schiff (PAS) and Masson staining, we discovered that the glomerular tuft area increased in the PS-MP-treated mice, while the degree of renal fibrosis remained unchanged. Furthermore, renal cortex transcriptomic analysis identified 388 and 303 differentially expressed genes (DEGs) in the 0.1 and 1 mg/L dose groups, respectively. The DEGs were highly enriched in mitochondrial-related terms and pathways of thermogenesis and oxidative phosphorylation. Moreover, protein–protein interaction (PPI) network analysis revealed that cytochrome b-c1 complex subunit 10 (UQCR11) and cytochrome c oxidase subunit 3 (MT-CO3) were important node proteins. These findings suggest that environmental exposure to MPs can cause abnormalities in renal structure and filtration function and that long-term exposure to MPs may be a risk factor for renal disease.

Abstract:Global plastic use has consistently increased over the past century with several different types of plastics now being produced. Much of these plastics end up in oceans or landfills leading to a substantial accumulation of plastics in the environment. Plastic debris slowly degrades into microplastics (MPs) that can ultimately be inhaled or ingested by both animals and humans. A growing body of evidence indicates that MPs can cross the gut barrier and enter into the lymphatic and systemic circulation leading to accumulation in tissues such as the lungs, liver, kidney, and brain. The impacts of mixed MPs exposure on tissue function through metabolism remains largely unexplored. To investigate the impact of ingested MPs on target metabolomic pathways, mice were subjected to either polystyrene microspheres or a mixed plastics (5 micron) exposure consisting of polystyrene, polyethylene and the biodegradability and biocompatible plastic, poly-(lactic-co-glycolic acid). Exposures were performed twice a week for four weeks at a dose of either 0, 2, or 4 mg/week via oral gastric gavage. Our findings demonstrate that, in mice, ingested MPs can pass through the gut barrier, be translocated through the systemic circulation, and accumulate in distant tissues including the brain, liver, and kidney. Additionally, we report on the metabolomic changes that occur in the colon, liver and brain which show differential responses that are dependent on dose and type of MPs exposure. Lastly, our study provides proof of concept for identifying metabolomic alterations associated with MPs exposure and adds insight into the potential health risks that mixed MPs contamination may pose to humans.

Abstract:As emerging pollutants, direct and indirect adverse impacts of micro(nano)plastics (MPs/NPs) are raising an increasing environmental concern in recent years due to their poor biodegradability and difficulty in recycling. MPs/NPs can act as carriers of bacteria, viruses, or pollutants (such as heavy metals and toxic organic compounds), and may potentially change the toxicity and bioavailability of pollutants. Ingested or attached MPs/NPs can also be transferred from low-trophic level organisms to high-nutrient organisms or even the human body through the food chain transfer process. This article reviews the emerging field of micro- and nanoplastics on organisms, including the separate toxicity and toxicity of compound after the adsorption of organic pollutants or heavy metals, as well as possible mechanism of toxicological effects and evaluate the nano- and microplastics potential adverse effects on human health. The inherent toxic effects MPs/NPs mainly include the following: physical injury, growth performance decrease and behavioral alteration, lipid metabolic disorder, induced gut microbiota dysbiosis and disruption of the gut’s epithelial permeability, neurotoxicity, damage of reproductive system and offspring, oxidative stress, immunotoxicity, etc. Additionally, MPs/NPs may release harmful plastic additives and toxic monomers such as bisphenol A, phthalates, and toluene diisocyanate. The vectors’ effect also points out the potential interaction of MPs/NPs with pollutants such as heavy metals, polycyclic aromatic hydrocarbons, organochlorine pesticides, polychlorinated biphenyls, perfluorinated compounds, pharmaceuticals, and polybrominated diphenyl ethers. Nevertheless, these potential consequences of MPs/NPs being vectors for contaminants are controversial.

Abstract:Microplastics (MPs) acted as environmental emerging pollutants have been widely distributed in natural waters. Due to their small particle size and large specific surface area, MPs can be adsorbed or accumulated in aquatic organisms, resulting in potential toxicity to mammals or humans through dietary routes. In this review, we discussed the distribution of MPs in natural waters, aquatic organisms, and humans. Pretreatment methods for the determination of MPs in biological samples were summarized, which are closely related to tissue composition, especially for human samples. Further elucidation of the potential toxicity and underlined mechanisms of MPs in mammals was carried out to parallel it in humans. This review provides a comprehensive understanding of the ecological and health risks of MPs originating from natural waters, as well as perspectives for more specific studies on the toxicity of MPs to mammals or humans.

Abstract:Particle size is one of the most important factors in determining the biological toxicity of microplastics (MPs). In this study, we attempted to examine the systemic toxicity of polystyrene MPs of different sizes (0.5 μm MP1 and 5 μm MP2) in C57BL/6 J mice. After the mice were given oral gavage of MPs for 8 consecutive weeks, histopathology and molecular biology assays, 16 S rRNA sequencing of the gut microbiota, and untargeted metabolomics were performed. The results showed that MPs were distributed in the organs in a size-dependent manner, with smaller particles demonstrating greater biodistribution. Further analysis indicated that exposure to MPs caused multi-organ damage through distinct toxicity pathways. Specifically, exposure to 0.5 μm MP1 led to excessive accumulation and induced more serious inflammation and mechanical damage in the spleen, kidney, heart, lung, and liver. However, 5 μm MP2 led to more severe intestinal barrier dysfunction, as well as gut dysbiosis and metabolic disorder in association with neuroinflammation. These results are helpful in expanding our knowledge of the toxicity of MPs of different sizes in mammalian models.

A quantitative study of nanoplastics within cells using magnetic resonance imaging

Abstract:Concerns regarding the environmental hazards and health risks of nanoplastics (NPs) are increasing. However, quantifying of NPs in vivo remains challenging. In this study, we propose a strategy for using magnetic resonance imaging (MRI) to quantify NPs internalized by mouse macrophages. Model NPs (Fe3O4@PS) with more homogeneous sizes and morphologies were obtained by encapsulating Fe3O4 in polystyrene. A standard curve was generated by linearly fitting the intensity and concentration charts to the Fe3O4@PS MRI data. The mass of Fe3O4@PS captured by the mouse macrophages was estimated using a standard curve. An explanation of how the standard curves were created and used is provided in the text. The accuracy of the MRI results was demonstrated using, inductively coupled plasma (ICP). Quantitative results from MRI and ICP revealed that the mouse macrophage uptake increased as NPs concentrations decreased. According to the ICP results, when the NPs exposure concentration was 10 μg/mL, the uptake rate by mouse macrophages was 63.0 %. The quantitative MRI results were slightly lower than those for ICP, with an uptake rate of 57.7 % in mouse macrophages at the same concentration. Therefore, MRI provides a new perspective for quantitative NPs analysis.

Abstract:Microplastics (MPs) have received an increasing attention because of their ubiquitous presence and aquatic toxicity associated with MPs and MP-bound contaminants in the natural water. This review is to critically examine the chemical additives leached from MPs, the altered contaminant behaviors and the resulting changes in their aquatic ecotoxicity. Available data suggest that heavy metals Zn, Cr, Pb, and Cd regulated and present in plastics at the sub-mg g-1 to mg g-1 level can leach a significant amount depending on MPs size, aging, pH, and salinity conditions. MP-bound organic contaminants are primarily additive-derived (e.g., brominated diphenyl ethers, nonylphenol, and bisphenol A) at the µg g-1 to mg g-1 level, and secondarily pyrogenic and legacy origins (e.g., PAHs and PCBs) in the range of ng g-1 and mg g-1. MPs tend to have higher but more variable sorption capacities for organic compounds than metals (1.77 ± 2.34 vs. 0.82 ± 0.94 mg g-1). MPs alter the behavior of heavy metals through the electrostatic interactions and surface complexation, while the transport of additive derived organic compounds are altered primarily through hydrophobic effect as supported by a positive correlation (R2 = 0.71) between the logarithmic MPs-adsorbed concentrations and octanol/water partition coefficients (KOW) of organic compounds. MPs constitute less than 0.01% of the total mass of aquatic particulates in typical waters, but play a discernible role in the local partitioning and long-distance movement of contaminants. MPs alone exert higher toxicity to invertebrates than algae; however, when MPs co-occur with pollutants, both synergistic and antagonistic toxicities are observed depending mainly on the ingestibility of MPs, the extent of sorption, MPs as a transport vector or a sink to scavenge pollutants. We finally suggest several key areas of future research directions and needed data concerning the role of MPs in mitigating pollutant leaching, transport and risk under conditions mimicking natural and polluted waters.
Direct identification and visualisation of real-world contaminating microplastics using Raman spectral mapping with multivariate curve resolution-alternating least squares

Abstract:Microplastics (MPs) contamination is ubiquitous in environmental matrices worldwide. Moreover these pollutants can be ingested by organisms and transported to organs via the circulatory system. Although efficient methods for the analysis of MPs derived from environment matrices and organisms' tissue samples have been developed after special sample pre-treatment, there remains a need for an optimised approach allowing direct identification and visualisation these MPs in real environmental matrices and organismal samples. Herein, we firstly used a multivariate curve resolution-alternating least squares (MCR-ALS) analysis of Raman hyperspectral imaging data to direct identification and visualisation of MPs in a complex serum background. Four common MPs types including polyethylene (PE), polystyrene (PS), polypropylene (PP) and polyethylene terephthalate (PET) were identified and visualised either individually or in mixtures within spiked samples at an 8-μm spatial resolution. Moreover, Raman imaging based on MCR-ALS was successfully applied in fish faeces biological samples and environmental sand samples for in situ MPs identification directly without washing or removal of organic matter. The current results demonstrate Raman imaging based on MCR-ALS as a novel imaging approach for direct identification and visualisation of MPs, through extraction of MPs' chemical spectra within a complicated biological or environmental background whilst eliminating overlapping Raman bands and fluorescence interference.

Abstract:Microplastics have become emerging contaminants, causing widespread concern about their potential toxic effects. In this study, the uptake and tissue accumulation of polystyrene microplastics (PS-MPs) in zebrafish were detected, and the toxic effects in liver were investigated. The results showed that after 7 days of exposure, 5 μm diameter MPs accumulated in fish gills, liver, and gut, while 20 μm diameter MPs accumulated only in fish gills and gut. Histopathological analysis showed that both 5 μm and 70 nm PS-MPs caused inflammation and lipid accumulation in fish liver. PS-MPs also induced significantly increased activities of superoxide dismutase and catalase, indicating that oxidative stress was induced after treatment with MPs. In addition, metabolomic analysis suggested that exposure to MPs induced alterations of metabolic profiles in fish liver and disturbed the lipid and energy metabolism. These findings provide new insights into the toxic effects of MPs on fish.

Abstract:Microplastics (MPs) have attracted significant attention due to their global distribution in living environments. Although some studies have reported MP-induced hepatotoxicity in mouse models, a systematic approach to MP-mediated liver toxicity was still lacking. Therefore, we used a mouse model to study the sub-chronic effects of MP exposure on the liver. Female C57BL/6 mice, aged 6 weeks, received an oral administration of 0.3 mg of Nile Red-labeled polystyrene (PS) microplastics, with particle sizes of 0.5 μm (submicron) and 5 μm (micron), via gavage, while control mice received vehicle only. Each mouse was exposed to MPs twice a week for 12 weeks. After sacrifice, the levels of MP accumulation, oxidative stress, inflammation, and pathological changes were measured in the mouse liver, and blood samples were collected for serum biochemistry analysis. Our results demonstrated that 0.5 μm PS-MPs were accumulated in mouse livers post-MP exposure, but not in the 5 μm MP exposure group. Simultaneously, increased levels of glucose, triglyceride, alanine transaminase (ALT), aspartate transaminase (AST), superoxide dismutase, 4-hydroxy-2-nonenal-mercapturic acid (HNE-MA), interleukin-6, and lipid droplets were found in the 0.5 μm MP exposure group, while the fewer responses, including elevated liver weight index, glucose, high-density lipoprotein, AST, and decreased HNE-MA were observed in 5 μm MP exposure group. These results indicate that sub-chronic exposure to submicron MPs causes MP deposition in mouse livers, which further induces oxidative stress, increases inflammatory cytokines and perturbs glucose and lipid homeostasis, which might trigger more severe metabolic dysfunction or non-alcoholic steatohepatitis-like hepatotoxicity.

Abstract:Although irregularly-shaped label-free microplastics (MPs) are predominantly distributed in the environment, non-destructive analysis of environmentally relevant MPs in organisms is still challenging. The purpose of the study is to suggest in vivo visual evidence of the uptake and effect of environmentally relevant MPs in organism. Transparent irregularly-shaped high-density polyethylene was selected as an environmentally relevant model MP and exposed to brine shrimp (Artemia franciscana). As a result, we suggest the application of SEM/EDX and coherent anti-Stokes Raman scattering (CARS) microspectroscopy as complementary tools to secure in vivo visual evidence of irregularly-shaped unlabeled MPs in living organisms without chemical digestion for biodistribution observations. Biological transmission electron microscopy also provides how ingested MPs physically affects the digestive tract in the brine shrimp which is rarely reported. In terms of environmental implications, this study would advance ecotoxicological research on microplastic pollution by providing a cutting-edge tool for investigating the bioavailability and ecotoxicity of environmentally relevant MPs in ecosystems.

Abstract:Microplastic (MP) is ubiquitous in the environment which appeared as an immense intimidation to human and animal health. The plastic fragments significantly polluted the ocean, fresh water, food chain, and other food items. Inadequate maintenance, less knowledge of adverse influence along with inappropriate usage in addition throwing away of plastics items revolves present planet in to plastics planet. The present study aims to focus on the recognition and advance detection technologies for MPs and the adverse effects of micro- and nanoplastics on human health. MPs have rigorous adverse effect on human health that leads to condensed growth rates, lessened reproductive capability, ulcer, scrape, and oxidative nervous anxiety, in addition, also disturb circulatory and respiratory mechanism. The detection of MP particles has also placed emphasis on identification technologies such as scanning electron microscopy, Raman spectroscopy, optical detection, Fourier transform infrared spectroscopy, thermo-analytical techniques, flow cytometry, holography, and hyperspectral imaging. It suggests that further research should be explored to understand the source, distribution, and health impacts and evaluate numerous detection methodologies for the MPs along with purification techniques.

Abstract:Microplastics (MPs) have become increasingly serious global problems due to their wide distribution and complicated impacts on living organisms. To obtain a comprehensive overview of the latest research progress on MPs, we conducted a bibliometric analysis combined with a literature review. The results showed that the number of studies on MPs has grown exponentially since 2010. Recently, the hotspot on MPs has shifted to terrestrial ecosystems and biological health risks, including human health risks. In addition, the toxic effects, identification and quantification of MPs are relatively new research hotspots. We subsequently provide a review of MPs studies related to health risks to terrestrial higher mammals and, in particular, to humans, including detection methods and potential toxicities based on current studies. Currently, MPs have been found existing in human feces, blood, colon, placenta and lung, but it is still unclear whether this is associated with related systemic diseases. In vivo and in vitro studies have demonstrated that MPs cause intestinal toxicity, metabolic disruption, reproductive toxicity, neurotoxicity, immunotoxicity through oxidative stress, apoptosis and specific pathways, etc. Notably, in terms of combined effects with pollutants and neurotoxicity, the effects of MPs are still controversial. Future attention should be paid to the detection and quantification of MPs in human tissues, exploring the combined effects and related mechanisms of MPs with other pollutants and clarifying the association between MPs and the development of pre-existing diseases. Our work enhances further understanding of the potential health risks of MPs to terrestrial higher mammals.
