Abstract:The microbial electrochemical systems (MESS) demonstrate advantages in wastewater treatment for enhanced organic contaminants removal by electron flow. Comprehensive investigation of anode metabolism for fermentable substrate, represented by glucose, under electrochemical regulation was essential to understand the operation mechanism of MESs. The anodic COD (chemical oxygen demand) degradation reaction w simulated as a superposition of first-order (background COD remove) and zeroorder reaction (anode-respiring COD remove) to COD concentration. Even though the background COD degradation wasn't directly related with anodic respiration to form electron flow, it was accelerated by increasing current rather than the changed electrode potentials. Statistically, the kinetic constant of background COD degradation generally exhibited a positive linear correlation (R-2 = 0.7822) with current. In overall COD removal increment enhanced by electron flow, the improvement of background COD degradation accounted for a relatively stable proportion of 33%. The anode-respiring and background metabolism successively exhibited competition (100-500 mg L-1) and synergy (500-1500 mg L-1) with increasing COD concentration, before being restricted by excessive COD loading (above 1500 mg L-1). The stratification and function differentiation were observed in anode biofilm. The dark-red dense inner layer close to anode surface was directionally enriched anode-respiring bacteria (ARB) like Geobacter (16.23%) and Pseudomonas (13.04%), while the white incompact outer layer separated inner layer from exogenous bacteria interference as a transition layer and assisted for the degradation of fermentable substrate. The anode biofilm helped for the progressive degradation of glucose and the formation of stable electrochemical active inner layer under persistent exogenous bacteria interference. A Novel Microbial Electrolysis Cell (MEC) Reactor for Biological Sulfate-Rich Wastewater Treatment Using Intermittent Supply of Electric Field

Abstract:Microbial electrolysis cells (MEC), coupled with sulfate-reducing bacteria (SRB), was used to degrade sulfate-rich wastewater that is deficient in electron donors. However, because of the impact of electric double layers (EDLs), salt crystals formed on the electrode, thereby potentially retarding the continuity of sulfate removal during the whole operation. Here, an improved MEC reactor using intermittent electric field was established. It works better in sulfate removal for a longer period, which was higher than the conventional MEC reactor by 2.18-fold after 10 days. Observation on the activity of lactic dehydrogenase (LDH) and ATP revealed that the formation of salt crystals on the electrode led to plasmatorrhexis. Conversely, improved reactor contributed to extracellular substances production and prevented the salt crystal formation, which was conducive to biofilm formation as further verified by detection through SEM. Electrochemical impedance spectroscopy tests showed that the cathodic microorganisms accelerated electron transfer whereas the salt crystals increased the charge transfer resistance. High-throughput sequencing analysis illustrated that improved reactor could maintain the competitiveness of SRB in the microbial community for a longer period. Moreover, the improved reactor resulted in high species diversity, thereby showing the significant resistance of the microorganisms to arduous environments. (C) 2017 Elsevier B.V. Electrical Stimulation Accelerated Phenanthrene Biodegradation Coupling with Nitrate Reduction in Groundwater

Abstract:Phenanthrene (PHE) and nitrate co-contamination widely exists in the groundwater. Anaerobic bioremediation of the two suffered from the slow removal rate and limited electron transfer within microbes. Herein electrical stimulation (ES) was proved to stimulate the simultaneous removal of PHE and nitrate anaerobically. High removal of PHE and nitrate (90.94 % and 100 %, respectively) was obtained within 24 h in the ES-bioreactor, two times higher than the control anaerobic bioreactor (32.16 %). ES-bioreactor exhibited a good PHE removal performance within initial PHE ranging from 5 to 25 mg/L. In ES-bioreactor, extracellular polymeric substances increased PHE bioavailability and acted as protective capsules. Nicotinamide adenine dinucleotide (NADH) and cytochrome C (cyt-C) facilitated the extracellular and intracellular electron exchange during PHE biotransformation. ES significantly selected Acetobacterium, PHE-degraders (f_Bacteroidetesvadin HA17, Limno-bacter and Stenotrophomonas) and electrogens (Clostrium_sensu_stricto_13) to participate in PHE decomposition. Acetobacterium decomposed complex organics to acetate, proving accessible substrates for electrogens to main-tain an efficient electron flow. Enzymes regulating bacterial chemotaxis, flagellar assembly and carbohydrate metabolism were up-regulated with ES, suggesting the indispensable role of such enzymatic processes in ES-bioreactor. Overall, this work demonstrates ES can promote anaerobic bioremediation of PHE and nitrate in groundwater and, thus, opens up a new bioelectrochemistry application scenario.
Enhanced Anaerobic Wastewater Treatment by a Binary Electroactive Material: Pseudocapacitance/Conductance-Mediated Microbial Interspecies Electron Transfer.

Abstract:Anaerobic digestion (AD) is a promising method to treat organic matter. However, AD performance was limited by the inefficient electron transfer and metabolism imbalance between acid-producing bacteria and methanogens. In this study, a novel binary electroactive material (Fe3O4@biochar) with pseudocapacitance (1.4 F/g) and conductance (10.2 μS/cm) was exploited to store-release electrons as well as enhance the direct electron transfer between acid-producing bacteria and methanogens during the AD process. The mechanism of pseudocapacitance/conductance on mediating interspecies electron transfer was deeply studied at each stage of AD. In the hydrolysis acidification stage, the pseudocapacitance of Fe3O4@biochar acting as electron acceptors proceeded NADH/NAD+ transformation of bacteria to promote ATP synthesis by 21% which supported energy for organics decomposition. In the methanogenesis stage, the conductance of Fe3O4@biochar helped the microbes establish direct interspecies electron transfer (DIET) to increase the coenzyme F420 content by 66% and then improve methane production by 13%. In the complete AD experiment, electrons generated from acid-producing bacteria were rapidly transported to methanogens via conductors. Excess electrons were buffered by the pseudocapacitor and then gradually released to methanogens which alleviated the drastic drop in pH. These findings provided a strategy to enhance the electron transfer in anaerobic treatment as well as guided the design of electroactive materials.
Microbial approach towards anode biofilm engineering enhances extracellular electron transfer for bioenergy production

Abstract:Applying microbial electrolysis cells (MEC) is a biological approach to enhance the growth of high amounts of electroactive biofilm for extracellular electron transfer. The electroactive biofilm degrades the organics by oxidizing them at the anode and producing electrical energy. Addition of waste-activated sludge (WAS) with fat grease oil (FOG) produces an optimal reactor environment for microbial growth to enhance the exchange of electrons between cells via microbial electrolysis. The present work aimed to investigate the microbial approach to increase the extracellular electron transfer (EET) in microbial electrolysis cells. Results revealed that metabolites in electroactive microbes (EAM) grow viable cells that initiate high EET at anode sites. At optimum WAS with FOG addition, volatile fatty acid and current generation yield production was 2.94 ± 0.19 g/L and 17.91 ± 7.23 mA, accompanied by COD removal efficiency of 89.5 ± 14.4%, respectively. This study introduces a novel approach to anode biofilm engineering that significantly enhances extracellular electron transfer, offering a fresh perspective on bioenergy production. Our approach, which demonstrates that anodic biofilm enhances intercellular electron transfer, increases NADH-NAD ratio, and increases metabolite yield-fluxes, has the potential to revolutionize bio-electricity production. Results indicated that the electrolysis highlights MEC performance in power generation of 788 mV with 200 mL of anode volume of active viable cells by utilizing WAS with 11% FOG. The achievements of this study provide critical parameters for the anode biofilm engineering, demonstrating how growth cell volume, intercellular electron transfer, and increases in NADH-NAD ratio are evidence of an increase in the EET, compelling evidence for the resilience treatment and efficient current production. These findings are significant in advancing our understanding of bioenergy production.
Novel electro-assisted micro-aerobic cathode biological technology induces oxidative demethylation of N, N-dimethylformamide for efficient ammonification of refractory membrane-making wastewater

Abstract:Recalcitrant and toxicological membrane-making wastewater displays negative impacts on environment, and this is difficult to treat efficiently using conventional hydrolytic acidification. In this study, a novel electro-assisted biological reactor with micro-aerobic cathode (EABR-MAC) was developed to improve the biodegradation and ammonification of N, N-dimethylformamide (DMF) in membrane-making wastewater, and the metabolic mechanism using metagenomic sequencing as comprehensively illustrated. The results showed that EABR-MAC significantly improved the ammonification of refractory organonitrogen and promoted DMF oxidative degradation by driving the electron transferred to the cathode. Additionally, the inhibition rates of oxygen uptake rate and nitrification in EABR-MAC were both lower under different cathode aeration frequency conditions. Microbial community analysis indicated that the functional fermentation bacteria and exoelectrogens, which were correlated with COD removal, ammonification, and detoxification, were significantly enriched upon electrostimulation, and the positive biological connections increased to form highly connected communities instead of competition. The functional genes revealed that EABR-MAC forcefully intervened with the metabolic pathway, so that DMF converted to formamide and ammonia by oxidative demethylation and formamide hydrolysis. The results of this study provide a promising strategy for efficient conversion of organonitrogen into ammonia nitrogen, and offer a new insight into the effects of electrostimulation on microbial metabolism.

Abstract:The formation and fouling control of dynamic membrane (DM) are critical to the performance of anaerobic dynamic membrane bioreactors (AnDMBRs), especially in conditions with high solids concentrations. This study introduced an electro-AnDMBR (EC) for sludge anaerobic digestion over a 120-day period, comparing its performance to a control AnDMBR (CK) without external voltage. The EC system applied a voltage of 0.5 V, resulting in improved filtration performance and enhanced digestion efficiency, with sludge concentrations exceeding 50 g/L. The DM layer in the EC system, influenced by the electric field, was observed to be thinner, more porous, and composed of fewer polysaccharides, proteins, and cells. Calculation based on the extended Derjaguin-Landau-Verwey-Overbeek theory revealed that the alleviated DM fouling in the EC system was due to a 27.9% increase in the energy barrier for sludge-membrane adhesion, facilitated by the electric field's interaction with modified sludge properties. In addition, the EC system exhibited stronger Lewis acid-base interactions within the sludge flocs, indicative of enhanced hydrogen-bonding interactions and improved electron transfer efficiency. The sludge in the EC system showed a 1.23-fold increase in conductivity and a 2.25-fold increase in capacitance compared to the CK system, along with increased electroactivity of extracellular polymeric substances (EPS), promoting extracellular electron transfer within the microbial communities. Moreover, the application of an electrical field significantly altered the microbial community structure, fostering a more interconnected microbial network and promoting multiple methanogenic pathways, which contributed to the overall enhanced performance of the EC system.

Abstract:An electrically stimulated anaerobic–aerobic coupled system was developed to improve the biodegradation of refractory phenolics. Expected 4-nitrophenol, 2, 4-dinitrophenol, and COD removals in the system with aerobic cathodic and anaerobic anodic chambers were approximately 53.7%, 45.4%, 22.3% (intermittent mode) and 37.9%, 19.8%, 17.3% (continuous mode) higher than that in the control system (26.0 ± 6.4%, 30.7 ± 7.1%, 49.8 ± 3.0%). 2, 4-dichlorophenol removal in the system with aerobic anodic and anaerobic cathodic chambers was approximately 28.5% higher than that in the control system (71.4 ± 5.7%). The contribution of the aerobic cathodic/anodic chambers to the removal of phenolic compounds was higher than that of the anaerobic cathodic/anodic chambers. The species related to phenolic biodegradation (Rhodococcus, Achromobacter, PSB-M-3, and Sphingobium) were enriched in the cathodic and anodic chambers of the system. These results showed that intermittent electrical stimulation could be a potential alternative for the efficient degradation of refractory phenolics.

Abstract:As an efficient wastewater pretreatment biotechnology, electrostimulated hydrolysis acidification (eHA) has been used to accelerate the removal of refractory pollutants, which is closely related to the effects of electrostimulation on microbial interspecies associations. However, the ecological processes underpinning such linkages remain unresolved, especially for the microbial communities derived from different niches, such as the electrode surface and plankton. Herein, the principles of cross-niche microbial associations and community assembly were investigated using molecular ecological network and phylogenetic bin-based null model analysis (iCAMP) based on 16S rRNA gene sequences. The electrostimulated planktonic sludge and electrode biofilm displayed significantly (P < 0.05) 1.67 and 1.53 times higher organic nitrogen pollutant (azo dye Alizarin Yellow R) degradation efficiency than non-electrostimulation group, and the corresponding microbial community composition and structure were significantly (P < 0.05) changed. Electroactive bacteria and functional degraders were enriched in the electrode biofilm and planktonic sludge, respectively. Notably, electrostimulation strengthened the synergistic microbial associations (1.8 times more links) between sludge and biofilm members. Additionally, both electrostimulation and cross-niche microbial associations induced greater importance of deterministic assembly. Overall, this study highlights the specificity of cross-electrode surface microbial associations and ecological processes with electrostimulation and advances our understanding of the manipulation of sludge microbiomes in engineered wastewater treatment systems.
Anaerobic oxidation of methane coupled with extracellular electron transfer to electrodes

Abstract:Anaerobic oxidation of methane (AOM) is an important process for understanding the global flux of methane and its relation to the global carbon cycle. Although AOM is known to be coupled to reductions of sulfate, nitrite, and nitrate, evidence that AOM is coupled with extracellular electron transfer (EET) to conductive solids is relatively insufficient. Here, we demonstrate EET-dependent AOM in a biofilm anode dominated by Geobacter spp. and Methanobacterium spp. using carbon-fiber electrodes as the terminal electron sink. The steady-state current density was kept at 11.0 ± 1.3 mA/m2 in a microbial electrochemical cell, and isotopic experiments supported AOM-EET to the anode. Fluorescence in situ hybridization images and metagenome results suggest that Methanobacterium spp. may work synergistically with Geobacter spp. to allow AOM, likely by employing intermediate (formate or H2)-dependent inter-species electron transport. Since metal oxides are widely present in sedimentary and terrestrial environments, an AOM-EET niche would have implications for minimizing the net global emissions of methane.
Micro-aeration assisted with electrogenic respiration enhanced the microbial catabolism and ammonification of aromatic amines in industrial wastewater

Abstract:Improvement of refractory nitrogen-containing organics biodegradation is crucial to meet discharged nitrogen standards and guarantee aquatic ecology safety. Although electrostimulation accelerates organic nitrogen pollutants amination, it remains uncertain how to strengthen ammonification of the amination products. This study demonstrated that ammonification was remarkably facilitated under micro-aerobic conditions through the degradation of aniline, an amination product of nitrobenzene, using an electrogenic respiration system. The microbial catabolism and ammonification were significantly enhanced by exposing the bioanode to air. Based on 16S rRNA gene sequencing and GeoChip analysis, our results indicated that aerobic aniline degraders and electroactive bacteria were enriched in suspension and inner electrode biofilm, respectively. The suspension community had a significantly higher relative abundance of catechol dioxygenase genes contributing to aerobic aniline biodegradation and reactive oxygen species (ROS) scavenger genes to protect from oxygen toxicity. The inner biofilm community contained obviously higher cytochrome c genes responsible for extracellular electron transfer. Additionally, network analysis indicated the aniline degraders were positively associated with electroactive bacteria and could be the potential hosts for genes encoding for dioxygenase and cytochrome, respectively. This study provides a feasible strategy to enhance nitrogen-containing organics ammonification and offers new insights into the microbial interaction mechanisms of micro-aeration assisted with electrogenic respiration.
Validation of Effective Roles of Non-Electroactive Microbes on Recalcitrant Contaminant Degradation in Bioelectrochemical Systems.

Abstract:Bioelectrochemical systems (BESs) have been widely investigated for recalcitrant waste treatment mainly because of their waste removal effectiveness. Electroactive microbes (EMs) have long been thought to contribute to the high effectiveness by interacting with electrodes via electron chains. However, this work demonstrated the dispensable role of EMs for enhanced recalcitrant contamination degradation in BESs. We revealed enhanced p-fluoronitrobenzene (p-FNB) degradation in a BES by observing a defluorination efficiency that was three times higher than that in biodegradation or electrochemical processes. Such an improvement was achieved by the collaborative roles of electrode biofilms and planktonic microbes, as their individual contributions to p-FNB degradation were found to be similarly stimulated by electricity. However, no bioelectrochemical activity was found in either the electrode biofilms or the planktonic microbes during stimulated p-FNB degradation; because no biocatalytically reductive or oxidative turnovers were observed on cyclic voltammetry curves. The non-involvement of EMs was further proven by the similar microbial community evolution for biofilms and planktonic microbes. In summary, we proposed a mechanism for indirect electrical stimulation of microbial metabolism by electrochemically generating the active mediator p-fluoroaniline (p-FA) and further degradation by a sequential combination of electrochemical p-FNB reduction and biological p-FA oxidation by non-EMs.
The convergence of lactic acid microbiomes and metabolites in long-term electrofermentation

Abstract:Regulating electron transfer in predominantly fermentative microbiomes has broad implications in environmental, chemical, food, and medical fields. Here we demonstrate electrochemical control in fermenting food waste, digestate, and wastewater to improve lactic acid production. We hypothesize that applying anodic potential will expedite and direct fermentation towards lactic acid. Continued operation that introduced epi/endophytic communities (Lactococcus, Lactobacillus, Weissella) to pure culture Lactiplantibacillus plantarum reactors with static electrodes was associated with the loss of anode-induced process intensification despite 80% L. plantarum retention. Employing fluidized electrodes discouraged biofilm formation and extended electrode influence to planktonic gram-positive fermenters using mediated extracellular electron transfer. While short-term experiments differentially enriched Lactococcus and Klebsiella spp., longer-term operations indicated convergent microbiomes and product spectra. These results highlight a functional resilience of environmental fermentative microbiomes to perturbations in redox potential, underscoring the need to better understand electrode induced polymicrobial interactions and physiological impacts to engineer tunable open-culture or synthetic consortia.
Stressful encounters with social work clients: a descriptive account based on critical incidents.

Abstract:This article presents the findings of an analysis of 130 critical incidents reported by social workers in Israel. Almost all the incidents turned out to be upsetting events that caused the writers a great deal of pain, frustration, and self-doubt. Content analysis yielded four main categories of incidents or events: (1) client hostility and aggression toward the worker, (2) client behaviors that violated the unwritten rules of the profession or the workers' expectations, (3) inherent professional dilemmas, and (4) workers' personal issues. Although the events may not be typical of worker-client interactions, they tend to undermine the workers' professional confidence and color their entire work experience. Recommendations are made for training that better prepares social workers for the difficult situations they may encounter and for supervision that helps them to deal with such situations more effectively.

Abstract:Increasing industrialization and urbanization have contributed to a significant rise in wastewater discharge and exerted extensive pressure on the existing natural energy resources. Microbial fuel cell (MFC) is a sustainable technology that utilizes wastewater for electricity generation. MFC comprises a bioelectrochemical system employing electroactive biofilms of several aerobic and anaerobic bacteria, such as Geobacter sulfurreducens, Shewanella oneidensis, Pseudomonas aeruginosa, and Ochrobacterum pseudiintermedium. Since the electroactive biofilms constitute a vital part of the MFC, it is crucial to understand the biofilm-mediated pollutant metabolism and electron transfer mechanisms. Engineering electroactive biofilm communities for improved biofilm formation and extracellular polymeric substances (EPS) secretion can positively impact the bioelectrochemical system and improve fuel cell performance. This review article summarizes the role of electroactive bacterial communities in MFC for wastewater treatment and bioelectricity generation. A significant focus has been laid on understanding the composition, structure, and function of electroactive biofilms in MFC. Various electron transport mechanisms, including direct electron transfer (DET), indirect electron transfer (IET), and long-distance electron transfer (LDET), have been discussed. A detailed summary of the optimization of process parameters and genetic engineering strategies for improving the performance of MFC has been provided. Lastly, the applications of MFC for wastewater treatment, bioelectricity generation, and biosensor development have been reviewed.
Co-occurrence of Direct and Indirect Extracellular Electron Transfer Mechanisms during Electroactive Respiration in a Dissimilatory Sulfate Reducing Bacterium

Abstract:Extracellular electron transfer (EET) propels microbial fuel cell (MFC) technology and contributes to the mobility of redox active minerals and microbial syntrophy in nature. Sulfate-reducing bacteria (SRB), especially the genus corrode metal electrodes but are of interest for sulfate-containing MFCs providing wastewater treatment. Although extensive studies on SRB-mediated metal electrode corrosion have been done, there remain knowledge gaps on SRB EET to electrodes. We aimed to determine SRB EET mechanisms towards improving SRB performance in MFC wastewater treatment. Our MFCs with Hildenborough ( H), a model SRB, indicated that H can harvest and send electrons to the carbon cloth electrode. Electricity production with a maximum power density of ∼0.074 W/m was observed when the ratio of lactate (electron and carbon donor) to sulfate (electron acceptor) was 60:20 and 0:10 in the anodic and cathodic chamber, respectively. Patterns in current production compared to variations of electron donor/acceptor ratios in the anode and cathode suggested that attachment of H to the electrode and biofilm density were critical for effective electricity generation. Analysis of H biofilms at different conditions (planktonic dissimilatory sulfate reduction respiration vs. electroactive respiration) by electron microscopy indicated H utilized filaments that resemble nano-pili to attach on electrodes and facilitate EET from cell-to-cell and to the electrode. Proteomics profiling of electroactive respiration proteins indicated H adapted to electroactive respiration by presenting more pili-, flagellar-related proteins and histidine kinases on electrodes. To investigate the role of pili and biofilm, we grew two H mutants in MFCs under the same conditions. The mutant with a deletion of the major pilus-producing gene yielded less voltage and far less attachment to the electrode, suggesting the importance of pili in EET. The mutant with a deficiency in biofilm formation, however, did not eliminate current production indicating the existence of indirect EET. Untargeted metabolomics profiling showed flavin-based metabolites, potential electron shuttles, were dysregulated between respiration modes. This work revealed the metabolic flexibility of H to thrive in less than ideal conditions with solid surfaces as both an electron acceptor (growth on anode) and donor (growth on cathode) by using a combination of direct and indirect EET mechanisms. Understanding H EET mechanism could enhance the application of H in MFCs treating wastewater.

Abstract:The performance and stability of a bioelectrochemical anaerobic digester (BeAD), continuously augmented with electroactive microorganisms (EAMs), were investigated. The BeAD showcased superior performance, sustaining the high COD removal efficiency and methane production rate of 76.5 % and 0.67 L/(L.d), respectively, in a stable state. Prominently, it exhibited remarkable resilience under hydraulic and organic shock loads, adeptly recuperating from disturbances up to 1000 % of its stable condition. This resilience of up to 300 % shock load was driven by increased levels of electron transport components such as quinones and riboflavins, which act as electron shuttles. However, after extreme shock exposures from 500 % to 1000 %, despite the spike in inhibitory by-products such as humic acids and ammonia, the upregulation of the mtr complex was pivotal in recovering and sustaining methane production. These insights emphasize the BeAD's capability to bolster both performance and stability, thereby providing a potent strategy for practical application of bioelectrochemical systems.

Abstract:Electric fields have been documented to promote the humification of organic matter, but the mechanism is still unclear. Herein, our proposed a new pathway for hydroxyl radical ( OH) generation through electroactive bacteria enhanced electron transfer, resulting in accelerated humification of organic matter during electric field-assisted aerobic composting (EAC). The results indicated that electric field significantly enhanced the generation of reactive oxygen species (ROS) and humification of organic matter, and the anode and cathode of EAC are the main production regions of singlet oxygen (1O2, maximum yield 83.2–132.8 μmol·kg − 1) and OH (maximum yield 2343.4–2905.2 μmol·kg − 1), respectively. Further investigations suggested that electroactive bacteria accumulated on electrodes increased the generation of OH by promoting the transfer of electrons from electrode to oxygen, whereas quenching the generated OH significantly suppressed the process of organic matter humification. Our findings suggest that the OH production through microbe-induced electron transfer is a primary cause of organic matter humification, representing an electric field-assisted strategy overlooked biochemical process.

Abstract:With applications in bioremediation, biosensing, and bioenergy, microbial electrochemical systems are a rapidly growing, multidisciplinary field within biological, chemical, and materials science. Since these systems use living microorganisms as biocatalysts, it is important to understand how microbial physiology, specifically biofilm formation, affects these electrochemical systems. Specifically, the literature lacks research that assesses the effects of biofilm on metabolic current output in mediated electron transfer systems. In this study, Rhodobacter capsulatus and Pseudomonas putida GPo1 were used as model, nonpathogenic strains that facilitate electron transfer via diffusible redox mediators. Nitric oxide has gained attention in biomedicine as a gaseous signaling molecule, which at sublethal concentrations may either augment or inhibit biofilm formation depending on the bacterial species. In R. capsulatus, nitric oxide treatment was associated with increased current yield and improved biofilm formation. However, in P. putida-GPo1, nitric oxide treatment corresponded to significantly reduced current output, as well as biofilm dispersal. In addition to highlighting the use of electrochemical tools to assess the effects of nitric oxide in biofilm formation, these findings demonstrate that biofilm-based mediated electron transfer systems benefit from the increased electrochemical output and enhanced cell adhesion, which is promising for more robust applications compared to their planktonic counterpart

Abstract:The formation and bioelectric performance of anode biofilms in microbial electrochemical systems (MESs) are sensitive to oxygen. Investigating the temporal-spatial structure of anode biofilms will help elucidate the interfaces between oxygen and bacteria, thereby facilitating the applications of MESs in wastewater treatment and energy recovery. Here, use of optical coherence tomography, frozen sections, and a microsensor revealed that the aerobic biofilms exhibited a multilayered sandwich structure with a sparse gap between the aerobe- and amphimicrobe-enriched outer layer and the dense exoelectrogen-enriched inner layer, whereas the anaerobic biofilm consisted of only a single dense layer. Our results showed that the inner layer of aerobic anode biofilms performed electricity generation, whereas the outer layer only consumed oxygen. In this case, electron donor diffusion through the outer layer became the limiting factor in electricity generation by the bioanode. Consequently, as the anode biofilms matured, current generation decreased.
