Abstract:Nanoliter high-performance liquid chromatography shows low consumption of solvents and samples, offering one of the best choices for arsenic speciation in precious samples in combination with inuctively coupled plasma mass spectrometry. A systematic investigation on coupling nanoliter high-performance liquid chromatography to inductively coupled plasma mass spectrometry from instrument design to injected sample volume and mobile phase was performed in this study. Nanoflow mobile phase was delivered by flow splitting using a conventional high-pressure pump with reuse of mobile phase waste. Dead volume was minimized to 60 nL for the sheathless interface based on the previously developed nanonebulizer. Capillary columns for nanoliter high-performance liquid chromatography were found to be sensitive to sample loading volume. An apparent difference was also found between the mobile phases for nanoliter and conventional high-performance liquid chromatography. Baseline separation of arsenite, arsenate, monomethylarsenic, and dimethylarsenic was achieved within 11 min on a 15 cm C-18 capillary column and within 12 min on a 25 cm strong anion exchange column. Detection limits of 0.9-1.8 mu g/L were obtained with precisions variable in the range of 1.6-4.2%. A good agreement between determined and certified values of a certified reference material of human urine (GBW 09115) validated its accuracy along with good recoveries (87-102%).

Abstract:A novel method for speciation analysis of inorganic arsenic was developed by on-line hyphenating microchip capillary electrophoresis (chip-CE) with hydride generation atomic fluorescence spectrometry (HG-AFS). Baseline separation of As(III) and As(V) was achieved within 54s by the chip-CE in a 90mm long channel at 2500V using a mixture of 25mmoll−1 H3BO3 and 0.4mmoll−1 CTAB (pH 8.9) as electrolyte buffer. The precisions (RSD, n=5) ranged from 1.9 to 1.4% for migration time, 2.1 to 2.7% for peak area, and 1.8 to 2.3% for peak height for the two arsenic species at 3.0mgl−1 (as As) level. The detection limits (3σ) for As(III) and As(V) based on peak height measurement were 76 and 112μgl−1 (as As), respectively. The recoveries of the spikes (1mgl−1 (as As) of As(III) and As(V)) in four locally collected water samples ranged from 93.7 to 106%.
Speciation of inorganic and organic arsenic forms using HPLC-ICP-MS technique in aqueous matrices

Abstract:The aim of the study was to develop, optimize and validate an analytical method for the simultaneous detection and quantification of As species (As (III), dimethylarsinate (DMA), monomethylarsonate (MMA) and As (V)) from various water matrices (drinking water, surface water and groundwater) using the technique of high performance liquid chromatography coupled with inductively coupled plasma mass spectrometry (HPLC-ICP-MS). The separation of the species was achieved using an Agilent G3154-65002 precolumn, 4.6 mm x 10 mm, and an Agilent G3154-65001 column, 4.6 mm x 150 mm, at ambient temperature. The mobile phase used was 1M NaH2PO4 and 0.1 mM EDTA-2Na solution at a flow rate of 1 mL/min. Linear regression lines were drawn in the range 10-50 µg/L, the coefficients of determination (R2) obtained for each species were higher than 0.99. The quantification limits (LOQ) varied in the range 1.8 µg/L As(V) to 2.0 µg/L As(III), and in the range 3.9 µg/L DMA to 4.1 µg /L MMA for all studied matrices. These limits allow the quantification of As species in drinking water samples, surface water and groundwater at the trace level.

Abstract:Abstract An on-line method capable of the separation of arsenic species was developed for the speciation of arsenite As(III), arsenate As(V), monomethylarsenic (MMA) and dimethylarsenic acid (DMA) in biological samples. The method is based on the combination of high-performance liquid chromatograph (HPLC) for separation, UV photo oxidation for sample digestion and hydride generation atomic fluorescence spectrometry (HGAFS) for sensitive detection. The best separation results were obtained with an anion-exchange AS11 column protected by an AG11 guard column, and gradient elution with NaH2PO4 and water as mobile phase. The on-line UV photo oxidation with 1.5% K2S2O8 in 0.2 mol L–1 NaOH in an 8 m PTFE coil for 40 s ensures the digestion of organoarsenic compounds. Detection limits for the four species were in the range of 0.11–0.15 ng (20 μL injected). Procedures were validated by analysis of the certified reference materials GBW09103 freeze-dried human urine and the results were in good agreement with the certified values of total arsenic concentration. The method has been successfully applied to speciation studies of blood arsenic species with no need of sample pretreatment. Speciation of arsenic in blood samples collected from two patients after the ingestion of realgar-containing drug reveals slight increase of arsenite and DMA, resulting from the digestion of realgar.

Abstract:It has been challenging to analyze inorganic arsenic (iAs) with anion exchange HPLC-Electrospray Ionization-Mass spectrometry (HPLC-ESI-MS), because arsenite (As(III)) is difficult to retain on column and the salts in mobile phase causes ionization suppression of iAs. To address these issues, a method has been developed involving the determination of arsenate (As(V)) with mixed mode HPLC-ESI-MS and the conversion of As(III) to As(V) for total iAs. As(V) was separated from other chemicals on Newcrom B, a bi-modal HPLC column involving anion exchange and reverse phase interaction. The elution employed a two-dimensional gradient, including a formic acid gradient to elute As(V) and a concurrent alcohol gradient to elute organic anions used in sample preparations. As(V) was detected by Selected Ion Recording (SIR) in negative mode at m/z = 141 with a QDa (single quad) detector. As(III) was quantitatively converted to As(V) by mCPBA oxidation and measured for total iAs. By replacing salt with formic acid in elution, the ionization efficiency for As(V) was greatly enhanced in ESI interface. The limit of detection (LOD) for As(V) and As(III) were 0.0263 μM (1.97 ppb) and 0.0398 μM (2.99 ppb), respectively. The linear range was 0.05-1 μM. The method has been used to characterize iAs speciation change in the solution and precipitation in a simulated iron-rich groundwater caused by air exposure.

Abstract:Multi-elemental speciation analysis in a single run reduces costs, waste and time, and also sample consumption, which is favorable to a large set of instable samples. This work demonstrates mercury and arsenic speciation in a single run using high performance liquid chromatography with inductively coupled plasma mass spectrometric detection (HPLC/ICP-MS). Thiol compounds in mobile phases interfered with the elution of arsenite because of coordination action, which was overcome by gradient elution (elution of As(III) by thiol-free mobile phases at first). Four arsenic forms (As(III), DMA, MMA and As(V)) and three mercury species (Hg(II), MeHg and EtHg) were separated and detected by HPLC/ICP-MS using 4 mM tetrabutylammonium hydroxide (TBAH) (pH 6.0) in the first 1.0 min and 4 mM TBAH + 20 mM L-cysteine (pH 6.0) in the next 9.0 min. The detection limits were obtained at 0.078 for As(III), 0.081 for DMA, 0.078 for MMA and 0.15 mu g L-1 for As(V), and 0.016 for Hg(II), 0.027 for MeHg and 0.032 mu g L-1 for EtHg, and the reproducibilities (50 mu g L-1 for each As-species and 10 mu g L-1 for each Hg-species) were all lower than 3.7%. The proposed method was successfully applied for speciation analyses of arsenic and mercury in five lotus roots, which indicated DMA (3.5-7.9 mu g kg(-1)) as the only arsenic species and MeHg (0.6-1.0 mu g kg(-1)) as the only mercury species. Analyses of two certified reference materials demonstrated good agreement between determined values and certified values. All results prove its advantages including high sensitivity and high analytical efficiency, which are beneficial for routine speciation analyses of arsenic and mercury in a single run in environmental and food fields.

Abstract:The current paper describes a method for determining As(III) and As(V) concentrations in wastewater samples. The approach is based on the extraction of As(III) from water samples using a non-polar organic solvent in a 10-12 M HCl medium. The extract contains As(III), while the raffinate contains As(V). Distilled water can be used to re-extract As(III) from the organic solvent. ICP-MS was used to analyze As concentrations in the reextract and the raffinate. The average recovery of arsenic by the proposed method is 99.6 %. It was calculated as the sum of As(III) and As(V) concentrations. The collected data demonstrated high repeatability. The standard addition procedure demonstrated the selectivity and accuracy of the employed method. The relative error of recovery of the standard As(III) addition is about 2 %, which is similar to the RSD found under repeatability conditions.

Abstract:This work establishes a hyphenated methodology coupling HPLC with ICP-MS for simultaneous speciation analysis of arsenic, mercury and lead for the first time. Four arsenicals (As(III), DMA, MMA and As(V)), four mercurials (Hg(II), MeHg, EtHg and PhHg) and three lead compounds (Pb(II), TML and TEL) were simultaneously analyzed within only 8 min with acceptable resolution (2.0-8.2 for As, 1.6-6.1 for Hg and 2.7-4.0 for Pb). The detection limits were 0.036-0.20 for As-species, 0.023-0.041 for Hg-species, and 0.0076-0.14 mu g L-1 for Pb-species. The developed method was applied for the measurement of five lotus seed samples, indicating the presence of DMA (19.6-28.2 mu g kg(-1)), TML (1.4-2.9 mu g kg(-1)), MeHg (1.2-4.8 mu g kg(-1)) and EtHg (0.8-2.2 mu g kg(-1)). This method provides a promising tool for studying the toxic, metabolic and bioavailable behaviors of arsenic, mercury and lead.

Abstract:The optimum chromatographic separation conditions, polyatomic ions interference correction, and extraction parameters were investigated for simultaneous extraction and quantification of As(III), dimethylarsinic acid (DMA), monomethylarsonic acid (MMA), As(V) and Cr(VI) with excellent resolution. The ammonium salts (NH4H2PO4 and NH4NO3) were examined as mobile phases at different pH values by altering the composition during the chromatographic run in gradient elution mode using high-performance liquid chromatography – inductively coupled plasma – mass spectrometry (HPLC-ICP-MS). The 10 mM and 70 mM NH4NO3 composition at pH 8.7 enabled sufficient baseline separation of the target As and Cr species at a total run time of 18 minutes. The identified polyatomic ion interferences were corrected by an inter-element method using mathematical equations. The optimum extraction conditions containing a mixture of 0.375 M (NH4)2HPO4 and 50 mM EDTA at neutral pH as an extraction buffer, extraction temperature of 150 °C and volume of 15 mL were attained by a factorial design technique. The lowest concentrations of the selected elemental species that can be reliably detected by the procedure were in the range from 0.064 to 0.682 ng g−1. The accuracy of the developed procedure was demonstrated by achieving percentage recoveries of 96.8% for As(V) and 97.7% for Cr(VI) using San Joaquin Soil standard reference material 2709a. Proposed method was validated by achieving percentage recoveries from 84.9 to 104% for As species and Cr(VI) using spiking and recovery studies at 3x and 25x the limit of quantification values of the target species. The selected As and Cr species in real sediment were monitored using the optimized analytical method. The differences in concentrations of Cr(VI), As(III), MMA and As(V) between the seasonal state of the river were statistically significant (p ˂0.05) at a 95% confidence level as determined by analysis of variance (ANOVA).

Abstract:A method is presented for arsenic speciation analysis of an oyster sample using ion chromatography coupled with an inductively coupled plasma mass spectrometry (ICP-MS) instrument. A strong anion exchange resin was employed with a step gradient elution of 0.1 mM/0.1 M K 2 SO 4 at pH 10.2. Arsenobetaine and dimethylarsinic acid were determined following extraction based on trypsin enzymolysis with 95-100% extraction efficiency. Limits of detection in the range 0.1-0.3 mg kg m 1 of arsenic were obtained for organic arsenic species. No inorganic arsenic was detected. Validation was performed using TORT-2 as a certified reference material. Although high performance liquid chromatography (HPLC) coupled to ICP-MS is an effective method for speciation analysis it is not always necessary to obtain such a detailed picture. A simple liquid chromatographic separation technique based upon mini-column technology is presented. It was developed to obtain a fast, efficient and reliable separation of inorganic from organic, i.e. assumed toxic from non-toxic, arsenic and selenium species suitable for use as an initial screening method for environmental analysis. Two types of strong anion exchange resin were tested. Excellent separation was obtained for both min-column resins and analysis times were within 7 min. Limits of detection obtained for inorganic arsenic, organic arsenic, selenomethionine, Se IV and Se VI were 1.6, 1.8, 66, 32 and 22 µg kg m 1 , respectively.

Abstract:Six arsenic species, namely arsenite (AsIII), arsenate (AsV), monomethylarsonic acid (MMA), dimethylarsinic acid (DMA), arsenobetaine (AsB) and arsenocholine (AsC) were speciated using a combination of high-performance liquid chromatography and inductively coupled plasma mass spectrometry (HPLC-ICP-MS). Under optimum chromatographic conditions, six arsenic species were well separated, and the performance of the combined system (HPLC-ICP-MS) for the species was determined. The limits of detection were calculated in the range of 0.14-0.29 ng/mL, and the corresponding quantification limits ranged between 0.45 and 0.97 ng mL-1 for the species. Spike recovery experiments performed on rice samples were used to validate the method's applicability to complex matrices. The recovery results calculated ranged between 93 and 109%, validating the method's applicability. Triplicate measurements for all spiked samples recorded percent relative standard deviation values below 10%.

Abstract:A simple capillary zone electrophoresis procedure was developed for the separation of arsenic species (AsO2 2−, AsO4 2−, and dimethylarsinic acid, DMA). Both counter-electroosmotic and co-electroosmotic (EOF) modes were investigated for the separation of arsenic species with direct UV detection at 185 nm using 20 mmol L−1 sodium phosphate as the electrolyte. The separation selectivity mainly depends on the separation modes and electrolyte pH. Inorganic anions (Cl−, NO2 −, NO3 − and SO4 2−) presented in real samples did not interfere with arsenic speciation in either separation mode. To improve the detection limits, sample-stacking techniques, including large-volume sample stacking (LVSS) and field-amplified sample injection (FASI), were investigated for the preconcentration of As species in co-CZE mode. Less than 1 μmol L−1 of detection limits for As species were achieved using FASI. The proposed method was demonstrated for the separation and detection of As species in water.

Abstract:The accurate determination of arsenic species in waters is still an environmental challenge because of the rapid redox transformation of As. So, a procedure for on-site separation of As(III), As(V), methylarsonic acid, and dimethylarsinic acid in natural waters has been developed. The procedure involves passing a water sample through tandem cation- and anion-exchange resin columns in the field. Selective retention of arsenic species occurs in resin cartridges followed by selective elution. The eluate is collected and analyzed by inductively coupled plasma mass spectrometry. The standard reference material CASS-5 was diluted 10 times and analyzed directly to validate accuracy of measurements, and SLRS-2 was used to validate on-site separation procedure, respectively. The results agreed well with the certified values of standard reference materials. The proposed procedure was applied to the on-site separation of arsenic species from natural waters.

Abstract:It is known that arsenic has different toxicological properties dependent upon both its oxidation state for inorganic compounds, as well as the different toxicity levels exhibited for organic arsenic compounds. The field of arsenic speciation analysis has grown rapidly in the recent years, especially with the utilization of chromatographic analysis. Extraction method as sequential extraction combined to microwave assisted digestion were applied to water and sediments samples. This paper describes the essential background and toxicity of arsenic in environment, and more importantly, some currently used chromatographic applications and sample handling procedures necessary to accurately detect arsenic in its various forms. Experiments and work using ultra high-performance liquid chromatography and inductively coupled plasma optical emission spectrometry for arsenic speciation of environmental samples are presented in this research.

Abstract:This work describes the method of a selective hydride generation-cryotrapping (HG-CT) coupled to an extremely sensitive but simple in-house assembled and designed atomic fluorescence spectrometry (AFS) instrument for determination of toxicologically important As species. Here, an advanced flame-in-gas-shield atomizer (FIGS) was interfaced to HG-CT and its performance was compared to a standard miniature diffusion flame (MDF) atomizer. A significant improvement both in sensitivity and baseline noise was found that was reflected in improved (4 times) limits of detection (LODs). The yielded LODs with the FIGS atomizer were 0.44, 0.74, 0.15, 0.17 and 0.67 ng L(-1) for arsenite, total inorganic, mono-, dimethylated As and trimethylarsine oxide, respectively. Moreover, the sensitivities with FIGS and MDF were equal for all As species, allowing for the possibility of single species standardization with arsenate standard for accurate quantification of all other As species. The accuracy of HG-CT-AFS with FIGS was verified by speciation analysis in two samples of bottled drinking water and certified reference materials, NRC CASS-5 (nearshore seawater) and SLRS-5 (river water) that contain traces of methylated As species. As speciation was in agreement with results previously reported and sums of all quantified species corresponded with the certified total As. The feasibility of HG-CT-AFS with FIGS was also demonstrated by the speciation analysis in microsamples of exfoliated bladder epithelial cells isolated from human urine. The results for the sums of trivalent and pentavalent As species corresponded well with the reference results obtained by HG-CT-ICPMS (inductively coupled plasma mass spectrometry).

Abstract:As a redox-sensitive and multi-species trace element, the chemical behavior of arsenic in various biological sources was closely related to human health. Direct and sensitive speciation analysis of arsenic from complex matrices is ideally required since it enables obtaining of multidimensional information concerning arsenic concentration, existing forms, redox states and molecular structures without sample pretreatment. Herein, an online speciation analysis method allowing molecular characterization of both inorganic (As(III), As(V)) and organic arsenic species (MMA, DMA, AsB and AsC) at trace level from original bio-matrices (e.g. crayfish, laver and kelp) was developed based on ambient ionization mass spectrometry, namely extraction electrospray ionization-hydride generation-microwave plasma torch-mass spectrometry (EESI-HG-MPT-MS). The novel ambient ionization source of MPT was combined with a hydride generator, resulting in a unique mass spectrum behavior for As. By using a home-made sample introduction device and incorporating HG-MPT, the sensitivity of analytes and matrix tolerance was significantly improved. The detection limits of As(III), As(V), MMA, DMA, AsB and AsC were 0.01, 0.02, 0.08, 0.08, 0.05 and 0.02 μg/kg respectively. Online spike experiment was also conducted, showing the recovery of 94-111%, with an RSD less than 10%. The developed method may play a role in studying the chemical or biological behaviors of arsenic in various environment.
