The high selection pressure from glyphosate use has led to the evolution of resistance to glyphosate in weeds. We investigated the molecular mechanisms of three recently discovered glyphosate-resistant Eleusine indica populations (R1, R2 and R3). The results showed that R1 and R2 had double Thr102Ile and Pro106Ser mutation and a single mutation of Pro106Leu in the 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) gene, respectively. Escherichia coli containing the mutated EPSPS genes was tolerant to glyphosate. EPSPS activity in R1 and R2 plants was higher than in the sensitive plants. There was no amino acid substitution in EPSPS gene in R3. However, expression of EPSPS in R3 plants was higher than in glyphosate-susceptible (S) population (13.8-fold) after glyphosate treatment. EPSPS enzyme activity in both R3 and S plants was inhibited by glyphosate, while shikimate accumulation in R3 was significantly lower than for the S population. Further analysis revealed that the genome of R3 contained 28.3-fold more copies of the EPSPS gene than that of susceptible population. EPSPS expression was positively correlated with copy number of EPSPS. In conclusion, mutation of the EPSPS gene and increased EPSPS expression are part of the molecular mechanisms of resistance to glyphosate in Eleusine indica.

Abstract:Background: The herbicide glyphosate inhibits the enzyme 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS). Overexpression of the EPSPS gene is one of the molecular mechanisms conferring glyphosate resistance in weeds, but the transcriptional regulation of this gene is poorly understood. The EPSPS gene was found to be significantly up-regulated following glyphosate treatment in a glyphosate-resistant Eleusine indica population from southern China. To further investigate the regulation of EPSPS overexpression, the promoter of the EPSPS gene from this E. indica population was cloned and analyzed. Results: Two upstream regulatory sequences, Epro-S (862 bp) and Epro-R (877 bp), of EPSPS were obtained from glyphosate-susceptible (S) and -resistant (R) E. indica plants, respectively, by high-efficiency thermal asymmetric interlaced polymerase chain reaction (HiTAIL-PCR). The Epro-S and Epro-R sequences were 99% homologous, except for two insertions (3 and12 bp) in the R sequence. The 12-base insertion in the Epro-R sequence was located in the 5' untranslated region (UTR) pyrimidine nucleotide-rich (Py-rich) stretch element. Promoter activity tests showed that the 12-base insertion resulted in significant enhancement of Epro-R promoter activity, whereas the 3-base insertion had little effect on Epro-R promoter activity. Conclusion: Alterations in the 5' UTR Py-rich stretch element of EPSPS are responsible for glyphosate-induced EPSPS overexpression. Thus, EPSPS transcriptional regulation confers glyphosate resistance in this E. indica population. © 2018 Society of Chemical Industry.

Abstract:Glyphosate-resistant crops have been a huge economic success for genetic engineering. The creating of new glyphosate-resistant plants would increase the available choices for planting and lower the price of genetically modified crop seeds. A novel G6 gene from Pseudomonas putida that encoded 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) was previously isolated. The G6 gene was transfected into rice via Agrobacterium-mediated transformation. The transgenic rice obtained was confirmed by PCR, Southern, and Western blots. The lab experiment and field trials further confirmed that the transgenic rice can survive glyphosate spraying at a dose of 8 g L−1. In contrast, conventional rice was killed at a weed control glyphosate spray dose of 1 g L−1. Altogether, the present study showed that the G6 gene works well in rice in vivo for glyphosate-resistance.
Role of physiological mechanisms and EPSPS gene expression in glyphosate resistance in wild soybeans (Glycine soja)

Abstract:The physiological mechanisms underlying glyphosate resistance in wild soybean germplasm and relevant EPSPS gene expression were evaluated. These germplasms were selected by gradually increasing glyphosate selection pressure started from 2010. As indicated by a whole-plant dose response bioassay, ZYD-254 plants were resistant to glyphosate at concentrations of 1230gaeha(-1), but the susceptible plants (ZYD-16) were unable to survive in the presence of 300gaeha(-1) glyphosate. The ED50 values of resistant germplasm were approximately 8.8 times of the susceptible germplasm. Chlorophyll content was significantly decreased in ZYD-16 plants in comparison with ZYD-254 plants. ZYD-16 plants accumulated 10.1 times more shikimate in leaves at 5days after glyphosate treatment at 1230gaeha(-1) than ZYD-254 did. GST activity differed between ZYD-254 and ZYD-16 in three tissues. It was highest in leaves. There were no significant differences in EPSPS1 or EPSPS3 expression between two germplasms before exposure to glyphosate treatment. After glyphosate treatment, there was a 2- to 4-fold increase in EPSPS1 mRNA levels in ZYD-254, but there was no change in EPSPS3 mRNA levels in ZYD-254 or ZYD-16.

Abstract:This study was aimed to clone a glyphosate-resistant gene and analyze its glyphosate-resistance in Escherichia coli.A glyphosate-resistant strain G6PP was isolated from extremely glyphosate-polluted soil,which could grow on medium with 200 μmol/L glyphosate.It was identified as Pseudomonas putida by scanning electron microscopy and 16S rDNA.Total DNA was isolated from G6PP.The EPSPS was cloned by PCR,which encoded 440 amino acid residues.EPSPS was then subcloned into the expression vector pET-28a and the recombinant prokaryotic expression vector(pET-G6) was transformated into E.coli BL21(DE3).A fusion protein of about 46 ku was obtained with IPTG induction.E.coli BL21(DE3) harboring pET-G6 plasmid could tolerate 150 μmol/L glyphosate in M63 liquid medium.The EPSPS G6 had glyphosate-resistant activities and might be used to develop glyphosant-resistant transgenic crops.

Abstract:Gene mutation is a basic evolutionary mechanism in plants under selection pressure of herbicides. Such mutation has pleiotropic effects on plant growth. We systemically investigated the effects of Pro106Leu (P106L), Pro106Ser (P106S), and Thr102Ile + Pro106Ser (TIPS) mutations on EPSPS functionality and fitness traits in Eleusine indica at the biochemical and physiological levels. The affinity of natural EPSPS for glyphosate was 53.8 times higher than that for phosphoenolpyruvate (PEP), as revealed by the dissociation constant; the constant decreased in both the P106L (39.9-fold) and P106S (46.9-fold) mutants but increased in the TIPS (87.5-fold) mutant. The Km (PEP) values of the P106L, P106S, and TIPS mutants were 2.4-, 0.7-, and 4.1-fold higher than that of natural EPSPS, corresponding to resistance levels of 2.5, 1.9, and 11.4, respectively. The catalytic efficiency values (maximum reaction rates) were 0.89-, 0.94-, and 0.26-fold higher than that of natural EPSPS. The levels of metabolites related to amino acids and nucleotides were significantly reduced in the mutated plants. The fitness costs were substantial for the biomass, total leaf area, seed number, and seedling emergence throughout the growth period in the plants with P106L and TIPS mutations. These results provide insights into EPSPS kinetics and their effect on plant growth.

Abstract:We have selected an upland cotton (Gossypium hirsutum L.) cell line (R1098) that is highly tolerant to glyphosate. This cell line was developed by in vitro selection with gradually increasing glyphosate concentrations, and its mechanisms conferring glyphosate tolerance were studied. Based on a whole-plant dose–response bioassay, R1098 plants were tolerant to glyphosate at a concentration of 1500gaeha−1 glyphosate (1.5× the recommended field rate) whereas the control plants (Coker 312) were unable to survive at 150gaeha−1 glyphosate. Coker 312 accumulated 13.1 times more shikimate in leaves at 5days after glyphosate treatment (1500gaeha−1) than that of R1098. Two distinct cDNAs for 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS), EPSPS-1 and EPSPS-2, were isolated from R1098. Both cDNAs were 97.7% identical within the common protein-coding region and the predicted sequences of the mature proteins were greater than 83% identical with EPSPS proteins from other known higher plants. In comparison to the glyphosate-susceptible cotton Coker 312, sequence analysis of the EPSPS-1 gene indicated that R1098 has an alanine insertion at nucleotide position 1216 resulting in frameshift. It leads to two copy functional EPSPS genes in R1098. There was no difference between R1098 and Coker 312 in EPSPS mRNA levels before glyphosate treatment. However, its treatment caused a 2–4 times increase in the basal EPSPS mRNA level in R1098.

Abstract:In order to introduce the glyphosate resistant EPSPS-G6 gene into cotton genome, this study used the cultivar CCRI-49 as receptor parent and the glyphosate herbicide as screening agent. 26 genetic transformation events were obtained through the method of pollen tube pathway (PTP). At the same time, the results of PCR identification of EPSPS-G6 gene also indicated that the target gene was integrated into cotton genome.Furthermore, the non-transgenic cotton TM-1 and CCRI-49 both died after the treatment of 15 mmol/L glyphosate, which showed they were not resistant to glyphosate. However, all of 26 transformation events expressed a high resistance to 20 mmol/L glyphosate; 11 of them still expressed an excellent resistance to 40 mmol/L glyphosate.

Abstract:Alzheimer's disease (AD) is the most frequent form of dementia in elderly individuals and its incidence and prevalence increases with age. This risk of AD is increased in the presence of genetic and demographic factors including apolipoprotein E 4 allele, lower education, and family history of AD. There are medical risk modifiers including systemic hypertension, diabetes mellitus, cardiovascular disease, and cerebrovascular disease that increase the vulnerability for AD. By contrast, there are lifestyle risk modifiers that reduce the effects of AD risk factors include diet and physical and cognitive activity. Our research has consistently shown that it is the interactions among these risk factors with the pathobiological cascade of AD that determine the likelihood of a clinical expression of AD-either as dementia or mild cognitive impairment. However, the association between "vulnerability" and "protective" factors varies with age, since the effects of these factors on the risk for AD may differ in younger (age < 80) versus older (age > 80) individuals. The understanding of the dynamic of these factors at different age periods will be essential for the implementation of primary prevention treatments for AD.

Abstract:The purpose of this study was to analysis the genetic characters of transgenic glyphosate -resistance cotton with EPSPS gene,using the glyphosate-resistant cotton transformation events G 6-1-G6-26 as the materials, and their non-transgenic genetic background cultivar CCRI-49 as control .Field resistance test in T 1 plants showed that the 20 of 26 transformation events were consistent with the ratio of 3∶1 and the other transformation events were inconsistent with single-gene Mendelian inheritance according to χ2 test.Field resistance test in T 2 plants showed 152 homozygous resistance lines were obtained derived from 25 transformation events ,respectively .Fifty-seven lines derived from 15 transformation events were consistent with the ratio 3∶1 through the analysis of heterozygous resist-ance lines.Furthermore,every transformation event had lines which were inconsistent with the ratio of 3∶1,and 10 transformation events did not have lines which were consistent with the ratio of 3∶1 .These suggested the integration and genetic mode of exogenous gene transformed by the pollen tube pathway method were complex .
Expression of Cytochrome P450 CYP81A6 in Rice: Tissue Specificity, Protein Subcellular Localization, and Response to Herbicide Application.

Abstract:The cytochrome P450 gene CYP81A6 confers tolerance to bentazon and metsulfuron-methyl, two selective herbicides widely used for weed control in rice and wheat fields. Knockout mutants of CYP81A6 are highly susceptible to both herbicides. The present study aimed to characterize the CYP81A6 expression in rice. Quantitative real-time polymerase chain reaction (PCR) analyses demonstrated that foliar treatment of bentazon (500 mg/L) greatly induced expression of CYP81A6 in both wild-type (Jiazhe B) and its knockout mutant (Jiazhe mB): a 10-fold increase at 9 h before returning to basal levels at 24 h in Jiazhe B, while in the mutant the expression level rose to >20-fold at 12 h and maintained at such high level up to 24 h post exposure. In contrast, metsulfuron-methyl (500 mg/L) treatment did not affect the expression of CYP81A6 in Jiazhe B within 80 h; thereafter the expression peaked at 120 h and returned gradually to basal levels by Day 6. We suggest that a metabolite of metsulfuron-methyl, 1H-2,3-benzothiazin-4-(3H)-one-2,2-dioxide, is likely to be responsible for inducing CYP81A6 expression, rather than the metsulfuron-methyl itself. Use of a promoter-GUS reporter construct (CYP81A6Pro::GUS) demonstrated that CYP81A6 was constitutively expressed throughout the plant, with the highest expression in the upper surfaces of leaves. Subcellular localization studies in rice protoplasts showed that CYP81A6 was localized in the endoplasmic reticulum. These observations advance our understanding of CYP81A6 expression in rice, particularly its response to the two herbicides.
Pro-106-Ser mutation and EPSPS overexpression acting together simultaneously in glyphosate-resistant goosegrass (Eleusine indica)

Abstract:Glyphosate has been used for more than 15 years for weed management in citrus groves in the Gulf of Mexico, at up to 3–4 applications per year. Goosegrass (Eleusine indica (L.) Gaertn.) control has sometimes failed. In this research, the mechanisms governing three goosegrass biotypes (Ein-Or from an orange grove, and Ein-Pl1 and Ein-Pl2 from Persian lime groves) with suspected resistance to glyphosate were characterized and compared to a susceptible biotype (Ein-S). Dose-response and shikimate accumulation assays confirmed resistance of the resistant (R) biotypes. There were no differences in glyphosate absorption, but the R biotypes retained up to 62–78% of the herbicide in the treated leaf at 96 h after treatment (HAT), in comparison to the Ein-S biotype (36%). The 5-enolpyruvylshikimate-3-phosphate synthase (EPSPS) activity in the Ein-Or and Ein-S biotypes was over 100-fold lower than the Ein-Pl1 and Ein-Pl2 ones. The latter showed a high EPSPS-basal activity, a mutation at Pro-106-Ser position in the EPSPS gene, and EPSPS overexpression. The EPSPS basal and EPSPS overexpression were positively correlated. The R goosegrass biotypes displayed poor glyphosate translocation. Furthermore, this grassweed showed, for the first time, two mechanisms at the target-site level (Pro-106-Ser mutation + EPSPS overexpression) acting together simultaneously against glyphosate.

An unusual presentation of Lyme arthritis.

Abstract:A patient eventually found to have Lyme arthritis presented with a symmetrical polyarthritis clinically resembling both rheumatoid arthritis and an enthesopathy but with a negative RA latex test. A history of tick bite was obtained only on followup questioning. Her Lyme serology was positive and her response to antibiotic treatment was complete.
Multiple resistance of Echinochloa phyllopogon to synthetic auxin, ALS-, and ACCase-inhibiting herbicides in Northeast China

Abstract:Echinochloa phyllopogon is a self-pollinating allotetraploid weed and a serious threat to global rice production. One sensitive and three multiple-resistant populations collected from two provinces of Northeast China were used to analyze the mechanism of multiple resistance of E. phyllopogon to penoxsulam, metamifop, and quinclorac. Compared with the sensitive population LN12, LN1 showed higher resistance to these three herbicides; LN24 showed medium resistance to penoxsulam and metamifop and higher resistance to quinclorac (274-fold); HLJ4 showed low resistance to penoxsulam and high resistance to metamifop and quinclorac. Target sequence analysis showed no mutations in acetolactate synthase or acetyl-CoA carboxylase genes. In-vitro enzyme activity analysis showed that the activity of the target enzyme of multiple herbicide-resistant populations was similar to that of the sensitive population. The P450 inhibitor, malathion, noticeably increased the sensitivity of LN1, LN24, and HLJ4 to penoxsulam, LN1 to metamifop, and HLJ4 to quinclorac. Under all four treatments, the GSTs activities of resistant and sensitive populations showed an increasing trend from day 1 to day 5, but the sensitivity and activity of GSTs were higher in the multiple-resistant population than that in the sensitive population LN12. This study identified the development of multiple-resistant E. phyllopogon populations that pose a serious threat to rice production in rice fields in Northeast China, preliminarily confirming that multiple-resistance was likely due to non-target-site resistance mechanisms. These populations of E. phyllopogon are likely to be more difficult to control.

Abstract:Genetically engineered (GE) rice endogenous epsps (5-enolpyruvoylshikimate-3-phosphate synthase) gene overexpressing EPSPS can increase glyphosate herbicide-resistance of cultivated rice. This type of epsps transgene can enhance the fecundity of rice crop-weed hybrid offspring in the absence of glyphosate, stimulating great concerns over undesired environmental impacts of transgene flow to populations of wild relatives. Here, we report the substantial alteration of phenology and fitness traits in F1-F3 crop-wild hybrid descendants derived from crosses between an epsps GE rice line and two endangered wild rice (Oryza rufipogon) populations, based on the common-garden field experiments. Under the glyphosate-free condition, transgenic hybrid lineages showed significantly earlier tillering and flowering, as well as increased fecundity and overwintering survival/regeneration abilities. In addition, a negative correlation was observed between the contents of endogenous EPSPS of wild, weedy, and cultivated rice parents and fitness differences caused by the incorporation of the epsps transgene. Namely, a lower level of endogenous EPSPS in the transgene-recipient populations displayed a more pronounced enhancement in fitness. The altered phenology and enhanced fitness of crop-wild hybrid offspring by the epsps transgene may cause unwanted environmental consequences when this type of glyphosate-resistance transgene introgressed into wild rice populations through gene flow.

Abstract:Background: Echinochloa crus-galli is the most troublesome and widespread weed of most rice-growing regions of the world. Cyhalofop-butyl, a herbicide within the acetyl-CoA carboxylase (ACCase) chemical group, has been extensively used to control barnyardgrass in rice. The repeated exposure to cyhalofop-butyl has led to resistance evolution in E. crus-galli populations. Results: In this study, we identified a population of E. crus-galli (R-HN) in a rice field in Hunan, China, that developed resistance to cyhalofop-butyl at 4.49-fold the recommended field dose. No known target mutation was detected in the ACCase gene of the R-HN population by ACCase sequencing compared to sensitive populations. Both cytochrome P450 (CYP450) and glutathione S-transferase (GST) inhibitors could not significantly reverse the resistance to cyhalofop-butyl. The nontarget-site resistance (NTSR) mechanism was investigated by transcriptome sequencing. Validation of the screened candidate genes by quantitative real-time (qRT)-PCR revealed that six glycosyltransferases (GTs) and four ATP-binding cassette (ABC) transporter genes were consistently upregulated in the R-HN population. Five GTs and one ABC transporter genes were constitutively upregulated after cyhalofop-butyl treatment in the R-HN population. Molecular docking results showed that the significant binding energy of GT79, GT75L6 and GT74E among all candidate genes. Conclusion: Thus, the GT genes appear to be directly implicated in NTSR to cyhalofop-butyl in the R-HN populations through metabolic enhancement, but their functional characterization needs to be studied. © 2024 Society of Chemical Industry.
A Novel 5-Enolpyruvoylshikimate-3-phosphate (epsp) Synthase Transgene for Glyphosate Resistance Stimulates Growth and Fecundity in Weedy Rice (oryza Sativa) Without Herbicide

Abstract:Understanding evolutionary interactions among crops and weeds can facilitate effective weed management. For example, gene flow from crops to their wild or weedy relatives can lead to rapid evolution in recipient populations. In rice (Oryza sativa), transgenic herbicide resistance is expected to spread to conspecific weedy rice (Oryza sativa f. spontanea) via hybridization. Here, we studied fitness effects of transgenic over-expression of a native 5-enolpyruvoylshikimate-3-phosphate synthase (epsps) gene developed to confer glyphosate resistance in rice. Controlling for genetic background, we examined physiological traits and field performance of crop-weed hybrid lineages that segregated for the presence or absence of this novel epsps transgene. Surprisingly, we found that transgenic F-2 crop-weed hybrids produced 48-125% more seeds per plant than nontransgenic controls in monoculture- and mixed-planting designs without glyphosate application. Transgenic plants also had greater EPSPS protein levels, tryptophan concentrations, photosynthetic rates, and per cent seed germination compared with nontransgenic controls. Our findings suggest that over-expression of a native rice epsps gene can lead to fitness advantages, even without exposure to glyphosate. We hypothesize that over-expressed epsps may be useful to breeders and, if deployed, could result in fitness benefits in weedy relatives following transgene introgression.
Target-Site and Metabolic Resistance Mechanisms to Penoxsulam in Late Watergrass (Echinochloa phyllopogon) in China

Abstract:Echinochloa phyllopogon, a malignant weed in Northeast China's paddy fields, is currently presenting escalating resistance concerns. Our study centered on the HJHL-715 E. phyllopogon population, which showed heightened resistance to penoxsulam, through a whole-plant bioassay. Pretreatment with a P450 inhibitor malathion significantly increased penoxsulam sensitivity in resistant plants. In order to determine the resistance mechanism of the resistant population, we purified the resistant population from individual plants and isolated target-site resistance (TSR) and nontarget-site resistance (NTSR) materials. Pro-197-Thr and Trp-574-Leu mutations in acetolactate synthase (ALS) 1 and ALS2 of the resistant population drove reduced sensitivity of penoxsulam to the target-site ALS, the primary resistance mechanisms. To fully understand the NTSR mechanism, NTSR materials were investigated by using RNA-sequencing (RNA-seq) combined with a reference genome. High-performance liquid chromatography-tandem mass spectrometry (HPLC-MS/MS) analysis further supported the enhanced penoxsulam metabolism in NTSR materials. Gene expression data and quantitative reverse transcription polymerase chain reaction (qRT-PCR) validation confirmed 29 overexpressed genes under penoxsulam treatment, with 16 genes concurrently upregulated with quinclorac and metamifop treatment. Overall, our study confirmed coexisting TSR and NTSR mechanisms in E. phyllopogon's resistance to ALS inhibitors.
