Abstract:The pea aphid, Acyrthosiphon pisum, is a major pest of legume crops, exhibiting distinct polymorphism in terms of wings and body color. We found that, under crowded conditions, the red morph A. pisum produced more winged offspring than the green morph. The signaling pathways involved in aphid wing determination, like insulin and ecdysone, also play important roles in regulating growth, development, and metabolism. Thus, here, we examined the association between the wing-producing ability and the growth rate, development time, reproductive capacity, and energy metabolism in these two color morphs. The growth rate of red morphs was significantly higher than that of green morphs, whereas green morphs produced more offspring during the first 6 days of the adult stage. Red morphs accumulated higher levels of glycogen and triglycerides and consumed more triglycerides during starvation; however, green aphids consumed more trehalose during food deprivation. Red aphids exhibited stronger starvation tolerance, possibly due to their higher triglyceride catabolic activity. Furthermore, the expression levels of genes involved in the insulin pathway, glycolysis, and lipolysis in red aphids were higher than those in green aphids. These results suggest that the wing-producing ability of the pea aphid may be associated with its growth and metabolism, which may be due to the shared regulatory signaling pathways.

Abstract:Many insects are capable of developing into either long-winged or short-winged (or wingless) morphs, which enables them to rapidly match heterogeneous environments. Thus, the wing polymorphism is an adaptation at the root of their ecological success. Wing polymorphism is orchestrated at various levels, starting with the insect's perception of environmental cues, then signal transduction and signal execution, and ultimately the transmitting of signals into physiological adaption in accordance with the particular morph produced. Juvenile hormone and ecdysteroid pathways have long been proposed to regulate wing polymorphism in insects, but rigorous experimental evidence is lacking. The breakthrough findings of ecdysone receptor regulation on transgenerational wing dimorphism in the aphid Acyrthosiphon pisum and of insulin signaling in the planthopper Nilaparvata lugens greatly broaden our understanding of wing polymorphism at the molecular level. Recently, the advent of high-throughput sequencing coupled with functional genomics provides powerful genetic tools for future insights into the molecular bases underlying wing polymorphism in insects.

Abstract:Wing polymorphism contributes significantly to the success of a wide variety of insects. However, its underlying molecular mechanism is less well understood. The migratory planthopper (BPH), Nilaparvata lugens, is one of the most extensively studied insects for wing polymorphism, due to its natural features of short- and long-winged morphs. Using the BPH as an example, we first surveyed the environmental cues that possibly influence wing developmental plasticity. Second, we explained the molecular basis by which two insulin receptors (InR1 and InR2) act as switches to determine alternative wing morphs in the BPH. This finding provides an additional layer of regulatory mechanism underlying wing polymorphism in insects in addition to juvenile hormones. Further, based on a discrete domain structure between InR1 and InR2 across insect species, we discussed the potential roles by which they might contribute to insect polymorphism. Last, we concluded with future directions of disentangling the insulin signalling pathway in the BPH, which serves as an ideal model for studying wing developmental plasticity in insects.This article is part of the themed issue 'Evo-devo in the genomics era, and the origins of morphological diversity'.
Transcriptome Analysis of the Regulatory Mechanism of FoxO on Wing Dimorphism in the Brown Planthopper, Nilaparvata Lugens (hemiptera: Delphacidae)

Abstract:The brown planthopper (BPH), Nilaparvata lugens, can develop into either short-winged (SW) or long-winged (LW) adults according to environmental conditions, and has long served as a model organism for exploring the mechanisms of wing polyphenism in insects. The transcription factor NlFoxO acts as a master regulator that directs the development of either SW or LW morphs, but the underlying molecular mechanism is largely unknown. Here, we microinjected SW-destined morphs with double stranded-RNA (dsRNA) targeting NlFoxO (dsNlFoxO) to change them into LW-winged morphs. In parallel, SW-destined morphs microinjected with dsRNA targeting the gene encoding green fluorescence protein (dsGfp) served as a negative control. The forewing and hindwing buds of 5th-instar nymphs collected at 24, 36, and 48 h after eclosion (hAE) were used for RNA sequencing. We obtained a minimum of 43.4 million clean reads from forewing and hindwing buds at a single developmental time. Differentially expressed genes (DEGs) were significantly enriched in various Gene Ontology (GO) terms, including cellular process, binding, and cell part. Kyoto Encyclopedia of Genes and Genomes (KEGG) enrichment pathway analysis showed that up-regulated genes in dsNlFoxO-treated forewing and hindwing buds were largely associated with the cell cycle and DNA replication. Furthermore, most up-regulated genes displayed higher expression at 24-, and 36-hAE relative to 48 hAE, indicating that wing cells in LW-destined wings might actively proliferate during the first 36 h in 5th-instar nymphs. Our findings indicated that LW development in BPH was likely dependent on the duration of cell proliferation in the 5th-instar stage, which sheds light on the molecular basis of wing polymorphism in insects.
Postnatal Wing Morph of Pea Aphids Regulates Hemolymph Trehalose Levels

Abstract:Trehalose, a nonreducing disaccharide composed of two glucose molecules, functions as a critical energy source in various insect tissues and organs and is the predominant sugar component of the hemolymph. The pea aphid, Acyrthosiphon pisum, exhibits higher hemolymph trehalose levels than other insects. However, the dynamics of hemolymph trehalose levels throughout its life stages remain unclear owing to the challenges associated with obtaining hemolymph from these small insects. Therefore, this study was conducted to quantify hemolymph trehalose levels in A. pisum using a fluorescent trehalose sensor (Tre-C04), which enhances green fluorescent protein fluorescence through the binding of trehalose to a ligand-binding protein fused to the fluorophore. Trehalose levels were successfully quantified in minimal hemolymph samples from individual aphids, with measurements spanning from the first nymphal stage to the adult stage in both the winged and wingless forms of A. pisum. Hemolymph trehalose levels remained relatively stable throughout the life cycle but exhibited a gradual increase with each developmental stage. Notably, adult winged aphids exhibited significantly higher hemolymph trehalose levels than wingless aphids. Given that wing morph determination occurs early in the nymphal stage, these findings suggest that hemolymph trehalose levels are regulated post-wing morph development. Further investigation of the expression of genes associated with trehalose metabolism revealed that trehalose phosphate synthase 2 levels were downregulated in early-stage wingless adults, whereas insulin-related peptide 5 levels were upregulated in wingless aphids. These findings indicate that A. pisum synthesizes trehalose during the winged adult stage to serve as an energy source for flight.
The maternal vGluT2 and embryonic mGluR3 signaling relay system controls offspring wing dimorphism in pea aphid

Abstract:Transgenerational phenotypic plasticity (TPP) refers to the phenomenon that environmental conditions experienced by one generation can influence the phenotype of subsequent generations to adapt to the environment without modification of their DNA sequences. Aphid wing dimorphism is a textbook example of TPP by which a maternal aphid perceives the environmental cues to decide the wing morph of her offspring. However, the signaling mechanism from mother to daughter remains unclear. In this study, we showed that the population density and physical contact caused high proportion of winged offspring in the pea aphid Acyrthosiphon pisum. Its vesicular glutamate transporter 2 (ApvGluT2) and metabotropic glutamate receptor 3 (ApmGluR3) were identified by tissue-specific RNA-seq as differentially expressed genes in the head and embryo respectively between solitary and more densely housed maternal aphids. Elevated expression of brain ApvGluT2 and embryonic ApmGluR3 led to increases in the winged proportion. Knockdown of either gene inhibited phosphorylation of ApFoxO in embryos. Furthermore, EMSA showed that dephosphorylated ApFoxO directly bound to the promotor of hedgehog (ApHh), a morphogen gene for wing development, to repress its transcription in stage 20 embryos, causing a lower winged proportion. Our results demonstrated that brain vGluT2 and embryonic mGluR3 coordinately relayed the maternal physical contact signals and control wing development in offspring, showcasing a novel regulatory mechanism underlying physical contact-dependent, transgenerational wing dimorphism in aphids.

Abstract:Author summary The highly conserved insulin/insulin-like growth factor signaling pathway plays a pivotal role in growth, development, and various physiological processes across a wide phylogeny of organisms. Unlike a single InR in the model species such as the fruit fly Drosophila melanogaster and the nematode Caenorhabditis elegans, most insect lineages have two or even three InR copies. However, the function of the alternative InRs remains elusive. Here, we created a homozygous mutation for a second insulin receptor (InR2) in the wing-dimorphic brown planthopper (BPH), Nilaparvata lugens, using the clustered regularly interspaced palindromic repeats/CRISPR-associated (CRISPR/Cas9) system. Our findings revealed that InR2 possesses functions distinct from the BPH InR homologue (NlInR1), indicating that multiple InR paralogues may have evolved independently and may have functionally diversified in ways more complex than previously expected in invertebrates. A single insulin receptor (InR) gene has been identified and extensively studied in model species ranging from nematodes to mice. However, most insects possess additional copies of InR, yet the functional significance, if any, of alternate InRs is unknown. Here, we used the wing-dimorphic brown planthopper (BPH) as a model system to query the role of a second InR copy in insects. NlInR2 resembled the BPH InR homologue (NlInR1) in terms of nymph development and reproduction, but revealed distinct regulatory roles in fuel metabolism, lifespan, and starvation tolerance. Unlike a lethal phenotype derived from NlInR1 null, homozygous NlInR2 null mutants were viable and accelerated DNA replication and cell proliferation in wing cells, thus redirecting short-winged-destined BPHs to develop into long-winged morphs. Additionally, the proper expression of NlInR2 was needed to maintain symmetric vein patterning in wings. Our findings provide the first direct evidence for the regulatory complexity of the two InR paralogues in insects, implying the functionally independent evolution of multiple InRs in invertebrates.
Comparative transcriptional analysis provides insights of possible molecular mechanisms of wing polyphenism induced by postnatal crowding in Aphis gossypii

Abstract:Abstract Background Aphis gossypii is a worldwide sap-sucking pest with a variety of hosts and a vector of more than 50 plant viruses. The strategy of wing polyphenism, mostly resulting from population density increasing, contributes to the evolutionary success of this pest. However, the related molecular basis remains unclear. Here, we identified the effects of postnatal crowding on wing morph determination in cotton aphid, and examined the transcriptomic differences between wingless and wing morphs. Results Effect of postnatal crowding on wing determination in A. gossypii was evaluated firstly. Under the density of 5 nymphs·cm − 2 , no wing aphids appeared. Proportion of wing morphs rised with the increase of density in a certain extent, and peaked to 56.1% at the density of 20 nymphs·cm − 2 , and reduced afterwards. Then, transcriptomes of wingless and wing morphs were assembled and annotated separately to identify potentially exclusively or differentially expressed transcripts between these two morphs, in which 3 126 and 3 392 unigenes annotated in Nr (Non-redundant protein sequence) database were found in wingless or wing morphs exclusively. Moreover, 3 187 up- and 1 880 down-regulated genes were identified in wing versus wingless aphid. Pathways analysis suggested the involvement of differentially expressed genes in multiple cellular signaling pathways involved in wing morphs determination, including lipid catabolic and metabolism, insulin, ecdysone and juvenile hormone biosynthesis. The expression levels of related genes were validated by the reverse transcription quantitative real time polymerase chain reaction (RT-qPCR) soon afterwards. Conclusions The present study identified the effects of postnatal crowding on wing morphs induction and demonstrated that the critical population density for wing morphs formation in A. gossypii was 20 nymphs·cm − 2 . Comparative transcriptome analysis provides transcripts potentially expressed exclusively in wingless or wing morph, respectively. Differentially expressed genes between wingless and wing morphs were identified and several signaling pathways potentially involved in cotton aphid wing differentiation were obtained.

Abstract:Sexual dimorphism and wing polyphenism are important and evolutionarily conserved features of many insect species. In this article, we found a cross-talk linking sexual differentiation with wing polyphenism in the brown planthopper (BPH) Nilaparvata lugens (order: Hemiptera). Knockdown of the sex determination gene Transformer-2 in N. lugens (NlTra-2) in nymph caused females to develop into infertile pseudomales containing undeveloped ovaries. Whereas males treated with dsNlTra-2 exhibited normal morphology, but lost fertility. Knockdown of NlTra-2 in adult females (maternal RNAi) resulted in long-winged female offspring, indicating that maternal RNAi changed the wing morphs in female offspring. In addition, silencing of NlTra-2 down-regulated the expression of the forkhead transcription factor FoxO (NlFoxO), and simultaneously up-regulated the expression of phosphatidylinositol-3-OH kinase (PI(3) K)-protein kinase B (NlAkt), the two critical genes in the insulin signaling pathway. Furthermore, the long-winged effect caused by maternal dsNlTra-2 RNAi could be reversed by silencing of NlInR1 and NlAkt, leading to short-winged morphs. We propose that there is a cross-talk between the sexual differentiation and wing polyphenism pathways mediated by NlTra-2 during embryonic stages.

Abstract:Significance Phenotypic plasticity is common in nature, yet the mechanisms by which environmental inputs are detected and alter developmental programs are not well understood. Even less explored are the mechanisms underlying transgenerational plasticity. Here we address the problem of how asexual aphids produce winged or wingless offspring in response to external cues. We find that gene expression patterns suggest the importance of the hormone ecdysone in this polyphenism. We show that aphids injected with ecdysone or its analog produce fewer winged offspring, and aphids with inhibited ecdysone signaling produce more winged offspring. Our results provide insight into the hormonal control of alternative aphid morphs, which have long been a textbook example of polyphenism, but whose control mechanisms have long proven elusive.
A novel gene REPTOR2 activates the autophagic degradation of wing disc in pea aphid

Abstract:Wing dimorphism in insects is an evolutionarily adaptive trait to maximize insect fitness under various environments, by which the population could be balanced between dispersing and reproduction. Most studies concern the regulatory mechanisms underlying the stimulation of wing morph in aphids, but relatively little research addresses the molecular basis of wing loss. Here, we found that, while developing normally in winged-destined pea aphids, the wing disc in wingless-destined aphids degenerated 30-hr postbirth and that this degeneration was due to autophagy rather than apoptosis. Activation of autophagy in first instar nymphs reduced the proportion of winged aphids, and suppression of autophagy increased the proportion. REPTOR2, associated with TOR signaling pathway, was identified by RNA-seq as a differentially expressed gene between the two morphs with higher expression in the thorax of wingless-destined aphids. Further genetic analysis indicated that REPTOR2 could be a novel gene derived from a gene duplication event that occurred exclusively in pea aphids on autosome A1 but translocated to the sex chromosome. Knockdown of REPTOR2 reduced autophagy in the wing disc and increased the proportion of winged aphids. In agreement with REPTOR's canonical negative regulatory role of TOR on autophagy, winged-destined aphids had higher TOR expression in the wing disc. Suppression of TOR activated autophagy of the wing disc and decreased the proportion of winged aphids, and vice versa. Co-suppression of TOR and REPTOR2 showed that dsREPTOR2 could mask the positive effect of dsTOR on autophagy, suggesting that REPTOR2 acted as a key regulator downstream of TOR in the signaling pathway. These results revealed that the TOR signaling pathway suppressed autophagic degradation of the wing disc in pea aphids by negatively regulating the expression of REPTOR2.
De novo assembled transcriptome of horned gall aphid, Schlechtendalia chinensis Bell, suggest changes in functional gene expression during host alternation

Abstract:The horned gall aphid, Schlechtendalia chinensis Bell, is the most economically valuable galling aphid. It accounts for approximately 75% of Chinese gallnut production, which is important for medical and chemical purposes. The horned gall aphid feeds on the Chinese sumac tree, Rhus chinensis, during the summer, induces galls, and overwinters on the moss Plagiomnium spp. However, the sequence information of S. chinensis is limited, and the functional gene expression changes during host alternation remain obscure. In this study, the transcriptome of the horned gall aphid was sequenced using high-throughput sequencing technology. Approximately 49 million reads were obtained and were assembled into 99 366 transcripts and 47 400 unigenes. Annotation was performed using a BLAST search against primary databases such as NCBI nr and Swiss-Prot, as well as enrichment analysis by GO, KOG and KEGG. Differentially expressed genes between spring and autumnmigrant aphids were compared, and the expression levels of many genes related to detoxification, digestion and other functional genes were found to have changed, reflecting the adaptation ability of horned gall aphids in different seasons and hosts. Functional genes in wax biosynthesis were upregulated in spring migrant aphids. This study analyzed the expression profile difference between spring and autumn migrant aphids to approach the real functional genes change during host alternation. The transcriptome provides a basis to understanding the functional genes during gall aphid host alternation and the related physiological and molecular characteristics.
Effect of Binodoxys Communis Parasitism on Flight Behavior of the Soybean Aphid, Aphis Glycines

Abstract:Many aphid species possess wingless (apterous) and winged (alate) stages, both of which can harbor parasitoids at various developmental stages. Winged aphids bearing parasitoid eggs or young larvae can potentially still engage in long-distance flights and thereby facilitate parasitoid dispersal, which has important implications for biological control of aphids by parasitoids. In this study, we determined the effect of parasitism by Binodoxys communis (Hymenoptera: Braconidae) on wing development and flight of the summer alate stage of the soybean aphid, Aphis glycines (Hemiptera: Aphididae), and quantified the effect of aphid flight duration on subsequent B. communis development. Parasitism by B. communis was allowed at alatoid 3rd (L3) and 4th instar (L4) nymphal stages of A. glycines and subsequent aphid flight was measured using a computer-monitored flight mill. Only 25% of aphids parasitized as L3 alatoid nymphs produced normal winged adults, compared to 100% of L4 alatoids. While flight performance of aphids that had been parasitized as 4th-instar alatoid nymphs 24 or 48h prior to testing was similar to that of unparasitized alates of identical age, it declined sharply for alates that had been parasitized as 4th-instar alatoid nymphs 72 and 96h prior to testing. Parasitoid larval and pupal development times, percent mummification and adult emergence rates did not differ between treatments in which aphids had flown over varying durations. Our results have implications for natural biological control of A. glycines in Asia and classical biological control of the soybean aphid in North America.

Abstract:Aphids present a fascinating example of phenotypic plasticity, in which a single genotype can produce dramatically different winged and wingless phenotypes that are specialized for dispersal versus reproduction, respectively. Recent work has examined many aspects of this plasticity, including its evolution, molecular control mechanisms, and genetic variation underlying the trait. In particular, exciting discoveries have been made about the signaling pathways that are responsible for controlling the production of winged versus wingless morphs, including ecdysone, dopamine, and insulin signaling, and about how specific genes such as REPTOR2 and vestigial are regulated to control winglessness. Future work will likely focus on the role of epigenetic mechanisms, as well as developing transgenic tools for more thoroughly dissecting the role of candidate plasticity-related genes.
Differences in Oxidative Stress Markers and Antioxidant Enzyme Activities in Black Bean Aphid Morphs (Aphis fabae Scop.) Fed on the Primary Host Viburnum opulus L

Abstract:Changes in the level of oxidative stress markers-superoxide anion radical (O2-), hydrogen peroxide (H2O2) and malondialdehyde (MDA) and the activity of antioxidant enzymes-superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX) in the black bean aphid occurring on the primary host (viburnum plants) were studied. Among the aphid morphs, the lowest contents of O2-, H2O2 and MDA were noted for winged adults (alatae), which were also characterized by the highest activity of antioxidant enzymes. These metabolic features indicate the adaptation of winged morphs to the colonization of new host plants. During spring migration, an increase in the content of oxidative stress markers and antioxidant enzyme activities in wingless females (fundatrigeniae) was observed. The significance of the biochemical adaptation of the black bean aphid to its winter host is discussed.
Expression patterns and functional analysis of the short neuropeptide F and NPF receptor genes in Rhopalosiphum padi

Abstract:The short neuropeptide F (sNPF) and NPF receptor (NPFR) genes play important roles in many physiological processes. However, information on the survival-related functions of sNPF and NPFR under different stress conditions is lacking in aphids. In this study, we cloned sNPF and NPFR, and investigated the expression levels of these genes in different developmental stages, wing morphs, and stress conditions of the bird cherry-oat aphid (Rhopalosiphum padi L.), an important agricultural pest. The sNPF and NPFR transcript levels varied among developmental stages, and their expression levels in alate females were significantly higher than those in apterous females. In addition, starvation resulted in significantly increased sNPF expression, which then recovered after refeeding. Heat stress and insecticides significantly affected transcription of both genes. sNPF and NPFR knockdown in R. padi using RNA interference revealed optimal interference efficiency at 48 h post-injection. sNPF knockdown significantly decreased adult longevity, 15-d fecundity, and food intake. Additionally, mortality under starvation, insecticides, and heat stress conditions was significantly higher after injection with double-stranded sNPF in R. padi. NPFR knockdown significantly affected food intake and starvation resistance in R. padi. These results strongly indicate that sNPF plays vital roles in food intake, longevity, and reproduction in R. padi, and it can significantly affect the pest's response to stress conditions.
Gene Expression Profiling Indicated Diverse Functions and Characteristics of Core Genes in Pea Aphid

Abstract:The pea aphid is a global insect pest, and variable phenotypes can be produced by pea aphids in the same genotype in response to changes in external environmental factors. However, detailed dynamic gene regulation networks and the core markers involved in different biological processes of pea aphids have not yet been reported. In this study, we obtained the published genomic and transcriptomic data, and performed transcriptome profiling of five pea aphid morphs (winged asexual female, wingless asexual female, wingless sexual female, winged male and wingless male) from each of three pea aphid genotypes, i.e., the transcriptomes from a total of 15 types of pea aphids were analyzed and the type-specific expression of genes in five different morphs was identified. The expression profiling was verified by quantitative real-time PCR (qPCR) analysis. Moreover, we determined the expression features and co-expression networks of highly variable genes. We also used the ARACNe method to obtain 263 core genes related to different biological pathways. Additionally, eight of the identified genes were aligned with transcription factor families, indicating that they act as transcription factors and regulate downstream genes. Furthermore, we found reliable markers using random forest methodology to distinguish different morphs of pea aphids. Our study provides a systematic and comprehensive approach for analyzing the core genes that may play important roles in a multitude of biological processes from the insect transcriptomes.

Abstract:Insect populations feeding on different plant species are under selection pressure to adapt to these differences. A study integrating elements of the ecology, behavior and gene expression of aphids on different host plants has not yet been well-explored. The present study explores the relationship between host fitness and survival, feeding behavior and salivary gland gene expression of a pea ( Pisum sativum ) host race of Acyrthosiphon pisum feeding on a common host Vicia faba and on three genetically-related hosts ( Vicia villosa , Medicago truncatula and Medicago sativa ). Life table data indicated that aphids on non-favored hosts exhibited small size, low reproduction rate, slow population increase and individual development and long lifespan. Electrical penetration graph results showed that the aphids spent significantly less time in passive ingestion of phloem sap on all non-preferred host plants before acclimation. After a period of acclimation on M. truncatula and V. villosa , pea host race individuals showed improved feeding behavior. No individuals of the pea host race completed its life history on M. sativa . Interestingly, the number of host-specific differentially-expressed salivary gland genes was negatively correlated with the fitness of aphids on this host plant. This study provided important cues in host plant specialization in aphids.
