Abstract:Ionic liquid (IL)-based aqueous biphasic systems (ABSs) provide a sustainable and efficient alternative to conventional liquid-liquid extraction techniques and can be used for the extraction, recovery, and purification of diverse solutes. However, the construction of a high-performance ABS that has both excellent phase separation ability and extraction performance remains challenging. This study concerns the preparation of a family of novel ABSs based on poly(ionic liquid)s (PILs) with customized structure and controllable molecular weight for the extraction of bioactive compounds. Several tailor-made PILs consisting of a hydrophobic backbone, hydrophilic imidazolium pendant groups and strong hydrogen bonding basic counteranions are prepared by reversible addition fragmentation chain-transfer polymerization. The PILs have a perfect balance of hydrophobicity/hydrophilicity and functionality, affording outstanding phase separation, which was better than with either the IL monomer poly(1-butyl-3-vinylimidazolium bromide ([BVIm]Br) or the normal free-radical polymer P[BVIm]Br*. More importantly, PIL-based ABSs exhibited unprecedented high partition coefficients for six bioactive compounds including tryptophan, phenylalanine, and caffeine, as well as high extraction yields. The performance of the PIL-based ABSs could also be tuned by changing the molecular weight and anionic character of the PILs. This work shows that tailor-made PIL-based ABSs are a promising platform for bioactive compound extraction and provides significant clues for the design of new ABSs for various applications.
Design and Synthesis of Thermoresponsive Ionic Liquid Polymer in Acetonitrile As a Reusable Extractant for Separation of Tocopherol Homologues

Abstract:We report the design and development of a series of novel ionic liquid polymers (PILs) which possess thermoresponsive properties in organic solvent. The PILs were synthesized via reversible additionfragmentation chain transfer (RAFT) copolymerization of 1-vinyl-3-butylimidazolium bromide and N-isopropylacrylamide, followed by anion exchange of bromide to amino acid d-alanine. The PILs possessed 0.36-0.76 molar fraction of ionic liquid with number-average molecular weight of 2.70-8.17 kg/mol and polydispersity index ranging between 1.12 and 1.25. The copolymerizations followed first-order reaction kinetics, and they were well-controlled, as indicated by the linear increase of molecular weight with monomer conversion. The PILs were thermoresponsive in acetonitrile with upper critical solution temperatures (UCST) varying from 25.7 to 34.8 degrees C, owing to the introduction of anion of amino acid. The PILs could be completely precipitated out by lowering the solution temperature. The PIL/acetonitrile solutions were used as extract phase for separation of tocopherol homologues in hexane. The distribution coefficient of delta-tocopherol between the extract and raffinate phases (D-delta) and the selectivity coefficient of delta-tocopherol to alpha-tocopherol (S-delta/alpha) reached as high as 7.86 and 13.0, respectively, while D-beta &gamma was 3.63 and S-beta &gamma/alpha was 6.0. The mole ratio of alpha-tocopherol in the raffinate phase increased from 0.08 to 0.27 after one stage extraction. The PILs could be reused for multiple extraction cycles with negligible change in the tocopherol distribution and selectivity coefficients. The thermoresponsivity of the PILs is of great benefit by eliminating the normally required back extraction steps. This work demonstrates the potential of thermoresponsive polymers for use in high performance separation of natural products.

Abstract:Ionic liquids (ILs) have been proven to be an effective technology for enhancing drug transdermal absorption. However, due to the unique structural components of ILs, the design of efficient ILs and elucidation of action mechanisms remain to be explored. In this review, basic design principles of ideal ILs for transdermal drug delivery system (TDDS) are discussed considering melting point, skin permeability, and toxicity, which depend on the molar ratios, types, functional groups of ions and inter-ionic interactions. Secondly, the contributions of ILs to the development of TDDS through different roles are described: as novel skin penetration enhancers for enhancing transdermal absorption of drugs; as novel solvents for improving the solubility of drugs in carriers; as novel active pharmaceutical ingredients (API-ILs) for regulating skin permeability, solubility, release, and pharmacokinetic behaviors of drugs; and as novel polymers for the development of smart medical materials. Moreover, diverse action mechanisms, mainly including the interactions among ILs, drugs, polymers, and skin components, are summarized. Finally, future challenges related to ILs are discussed, including underlying quantitative structure-activity relationships, complex interaction forces between anions, drugs, polymers and skin microenvironment, long-term stability, and <i>in vivo</i> safety issues. In summary, this article will promote the development of TDDS based on ILs.

Abstract:The physicochemical properties of ionic liquids (ILs) in transdermal drug delivery (TDD) directly affect drug permeation efficiency. Understanding the relationship between the molecular structure of ILs and their transdermal enhancement effect is essential for optimizing IL performance in transdermal drug administration. This article provides a comprehensive overview of the design principles, evaluation methods and applications for TDD. Ionic liquids (ILs) are salts with melting points typically <100 °C, composed of specific anions and cations. Recently, IL application has expanded into material engineering and biomedicine. Due to their unique properties, ILs have garnered significant interest in pharmacological research as solubilizers, transdermal absorption enhancers, antibacterial agents, and stabilizers of insoluble pharmaceutical active ingredients. The improvement of skin permeability by ILs is closely associated with their specific physicochemical characteristics, which are identified by their ionic composition. However, the existing literature on transdermal medication administration is insufficient in terms of a comprehensive knowledge base. This review provides a comprehensive assessment of the design principles involved in IL synthesis. Additionally, it discusses the methods utilized to assess skin permeability and provides a focused outline of IL application in transdermal drug administration.

Improving Dermal Delivery of Hydrophilic Macromolecules by Biocompatible Ionic Liquid Based on Choline and Malic Acid.

Abstract:The aim of this work was to develop biocompatible ionic liquids (ILs) for enhancement of dermal delivery of hydrophilic macromolecules. The ILs were prepared from malic acid and choline, which are commonly used as food additives and generally regarded as safe. The choline malate IL (CM-IL) was formed via ionic interactions, which has been validated by Fourier transform infrared spectroscopy and nuclear magnetic resonance. Analysis by differential scanning calorimetry confirmed a melting point of -65.2 degrees C for CM-IL. In skin penetration study, CM-IL was proved to be able to deliver a model hydrophilic macromolecule dextran into deep skin. The amount of dextran delivered to epidermis and dermis by CM-IL is approximately two folds that of by dextran solution. Enhanced in vivo skin penetration by CM-IL was proved as well. The fluorescence of FITC-dextran could be observed permeating pervasively throughout the skin from 4 h to 24 h, while in the control groups it was mainly concentrated in stratum cornea (SC) and hair follicles. In addition, CM-IL did not shown any irritation to mice skin within 7 days of successive treatment and any toxicity to human epidermal cells (HEK-A) within 24 h. In conclusion, CM-IL could be potentially used as enhancers or vehicles for dermal delivery of hydrophilic macromolecules.

Abstract:Ionic liquids (ILs) have great potential to facilitate transdermal and topical drug delivery. Here, we investigated the mechanism of action of amphiphilic ILs 1-methyl-3-octylimidazolium bromide (C 8 MIM) and 3-dodecyl-1-methylimidazolium bromide (C 12 MIM) in skin barrier lipid models in comparison to their complex effects in human skin. C 8 MIM incorporated in a skin lipid model was a better permeation enhancer than C 12 MIM for water and model drugs, theophylline and diclofenac. Solid state 2 H NMR and X-ray diffraction indicated that both ILs prefer the cholesterol-rich regions in skin lipids without significantly perturbing their lamellar arrangement and that C 8 MIM induces the formation of an isotropic lipid phase to a greater extent compared to C 12 MIM. C 12 MIM applied topically to the lipid model or human skin as a pretreatment was more potent than C 8 MIM. When co-applied with the drugs to human skin, aqueous C 12 MIM was more potent than C 8 MIM in enhancing theophylline permeation, but neither IL affected (even decreased) diclofenac permeation. Thus, the IL's ability to permeabilize skin lipid barrier is strongly modulated by its ability to reach the site of action and its interactions with drug and solvent. Such an interplay is far from trivial and requires detailed investigation to realize the full potential of ILs.

Abstract:In the drug delivery system, the transdermal drug delivery system has the advantages of painless administration, avoiding the first-pass metabolism, sustained therapeutic effect, and good patient compliance. It is a promising treatment method. Transdermal drug delivery systems have made important contributions in medical practice, but its potential as an alternative to oral drug delivery and subcutaneous injection has not been fully realized. Ionic liquids (ILs) are widely used in biomedicine, especially in transdermal drug delivery, because of their unique properties, such as good chemical stability and biocompatibility. ILs can enhance transdermal transcellular and decellular transport, bypass the barrier of the stratum corneum, and destroy cell integrity. Hydrophilic ILs act by opening tight junctions in the stratum corneum, mainly by enhancing fluidization in protein and lipid regions to promote paracellular transport. Hydrophobic ILs improve their distribution into the epithelial membrane by providing channels, thereby promoting the transcellular transport of lipid regions. Therefore, this review summarizes the latest methods and progress for transdermal delivery of ILs.

Abstract:Transdermal delivery of drugs, a compelling route of systemic drug delivery, provides painless, reliable, targeted, efficient and cost effective therapeutic regimen for patients. However, its use is limited by skin barrier especially the stratum corneum barrier. Moreover, transdermal delivery of macromolecules remains a challenge. Naturally, varieties of physical methods, chemical enhancers and drug carriers have been used to counteract this limitation. Recently, transdermal peptides discovered as safer, more efficient and more specific enhancers could promote the delivery of macromolecules across the skin. Herein, the underlying transdermal peptides are included. Subsequently, we have discussed typical applications and the possible mechanism of two groups of biologically inspired transdermal peptide enhancers, namely cell penetration peptides and transdermal enhanced peptides.
The role of disclosure patterns and unsupportive social interactions in the well‐being of breast cancer patients

Abstract:The purpose of this study was to examine the nature and potential effects of disclosure patterns and unsupportive social interactions in breast cancer patients. Disclosure, the thoughts and feelings people communicate to others, and unsupportive social interactions, the upsetting or unsupportive responses people receive from others, have been infrequently studied in breast cancer patients. Sixty‐six early stage breast cancer patients diagnosed less than one year completed a written questionnaire. Results indicated that the women reported confiding in family and friends more than in mental health workers. Fears of recurrence and worries about the effects of the illness on family members were the most important concerns, whereas concerns about body image were the least troublesome. Failure to disclose concerns was associated with low social support, high unsupportive social interactions, and low emotional well‐being. Most of the unsupportive responses received from other people were either behaviors of minimizing or distancing. Unsupportive reactions were significantly associated with greater role limitations due to emotional problems and decreased social functioning. Abstract:Palmitoylethanolamide (PEA) exhibits multiple skincare functions such as anti-nociceptive and anti-inflammatory effects. However, its topical application is limited due to its difficulty in bypassing the stratum corneum barrier, relatively low bioavailability, and low stability. Herein, elastic nano-liposomes (ENLs) with excellent deformability and elasticity were utilized as a novel drug delivery system to encapsulate PEA to overcome the abovementioned issues and enhance the biological effects on the skin. ENL was prepared with phosphatidylcholine, cholesterol, and cetyl-PG hydroxyethyl palmitamide with a molar ratio mimicking skin epidermal lipids, and PEA was loaded. The PEA-loaded ENL (PEA-ENL) demonstrated efficient transdermal delivery and enhanced skin retention, with negligible cytotoxicity toward HaCaT cells and no allergic reaction in the human skin patch test. Notably, PEA-ENL treatment increased cell migration and induced significant regulation in the expression of genes associated with anti-nociceptive, anti-inflammatory, and skin barrier repair. The mechanism of the anti-nociceptive and anti-inflammatory effects of PEA was further investigated and explained by molecular docking site analysis. This novel PEA-ENL, with efficient transdermal delivery efficiency and multiple skincare functionalities, is promising for topical application.
Synthesis, characterization, and evaluation of novel cell-penetrating peptides based on TD-34

Abstract:In this study, six N-1, N-2, or N-11 derivatives of TD-34 (a cationic cyclic cell-penetrating peptide [CPP], ACSSKKSKHCG) were designed and synthesized including both linear peptides and cyclic peptides, such as DL-1 (KWSSKKSKHCG), DLCC-1 (cyclopeptide, KWSSKKSKHCG), DL-2 (KWSSKKSKHCG-NH2 ), DLCC-2 (cyclopeptide, KWSSKKSKHCG-NH2 ), DL-3 (RWSSKKSKHCG), and DLCC-3 (cyclopeptide, RWSSKKSKHCG). The cyclic peptides were synthesized by disulfide bound linkages formed by N-2 and N-10 cysteine. In vitro penetration experiment was conducted to investigate the transdermal enhancement ability of these derivatives, using triptolide (TP) as model drug. The results display that at the presence of DLCC-2, the accumulative penetration amount of TP increased 1.71-fold (P < .05) within 12 hours, displaying better transdermal enhancing ability than TD-34. Meanwhile, DL-3 and DLCC-3 slightly decreased the transdermal delivery of TP, and the presence of DL-1 and DLCC-1 shows no obvious effect. In order to clarify the factors on the transdermal ability of peptides, the solubility of TP in phosphate buffer saline (PBS) at the presence of different peptides and the mechanism of transdermal delivery of CPPs was investigated. The result shows that most of these peptides have no significant effect on the solubility of TP except DLCC-3 (the solubility of TP slightly increased). And in order to investigate transdermal absorption route of DLCC-2, polyarginine linked to rhodamine b (Rh b) derivative is used. The result proved that the transdermal route of polyarginine is via hair follicle, which may change the transdermal route of its cargo molecule (TP). Our group previously proved that polyarginine and TD-34 have similar transdermal enhancing mechanism (changing the transdermal route of their cargo molecule); it is reasonably speculated that the transdermal route of DLCC-2 is the same as polyarginine and then changes the transdermal absorption route of TP. Furthermore, such results have laid a solid foundation for further investigation of CPPs and paved a way for both designing and synthesizing of new drug delivery system for therapy molecules.

Enhanced Transdermal Delivery of Insulin by Choline-Based Ionic Liquids

Abstract:Ionic liquids (ILs) show promises as chemical penetration enhancers (CPEs) for transdermal delivery of macromolecular drugs. However, their high viscosity and strong drug-IL affinity may limit drug diffusion and release from the drug-loaded IL (one-step strategy). Herein, a two-step strategy was used by applying choline-based ILs as pretreatment agents followed by insulin solution to improve penetration. Insulin remained stable in the ILs and are released slowly from the IL matrices. In vitro and in vivo studies showed that two-step treatment enhanced insulin penetration compared to one-step treatment, with choline citrate ([Ch][Ci]) and choline geranate ([Ch][Ge]) performing the best. In a diabetic rat model, two-step treatment with [Ch][Ge] reduced blood glucose levels to below 80 % within 8 h, while one-step treatment only maintained for 12 h. Trans-epidermal water loss and molecular dynamics simulations suggested that variations in release rates and skin condition accounted for the differences between the two strategies. Physical characterization confirmed that ILs enhanced transdermal delivery of insulin by permeabilize stratum corneum and opened tight junctions. Preliminary safety assessment indicated mild irritation by [Ch][Ge], whereas [Ch][Ci] showed good biocompatibility. It is concluded that ILs hold potential in enhancing transdermal delivery of insulin.

Abstract:Drug delivery remains a challenge in the field of medicine. Current administration methods limit the application of peptide-protein drugs. The non-injectable administration of polypeptide and protein drugs has frequently been a key research topic in the field of drug delivery. Conotoxins (CTX) are one class of these polypeptides, which shows excellent biological efficacy. However, CTX also has drawbacks because of its hydrophilicity and polarity, which restricts its application to invasive drug delivery such as injections and significantly limits its clinical application. In this study, we developed an efficient and practical transdermal delivery system using an ionic liquid based on matrine and coconut oleic acid. In vitro penetration experiments demonstrated that the developed system could effectively deliver CTX to the dermis of the skin, with the cumulative absorption in the skin increased by 4.87 times. Moreover, the synergistic merits of ionic liquids and CTX contribute to the excellent antioxidant, antibacterial, and moisturizing effects at the cellular level. In addition, high safety was confirmed in animal experiments. In particular, the combined system exhibited outstanding efficacy for the improvement of skin elasticity and inhibiting wrinkle formation in population clinical tests. Therefore, this study expands the application of matrine-based ionic liquids in drug delivery and provides a novel methodology that promotes the efficiency of non-invasive drug delivery.

Abstract:<p>The development of hydrophobic drug and protein delivery carriers remains a challenge. To synthesize L-(-)-carnitine-based ionic liquid (IL), this study applies the density functional theory to investigate the hydrogen bonds and van der Waals force that govern L-(-)-carnitine-based IL formation. An ionic liquid-in-oil microemulsion (IL/O ME) is then developed to facilitate the transdermal delivery of proteins and increase the solubility of drugs. IL/O ME is prepared using isopropyl myristate (IPM), Tween 80/Span 20, and L-(-)-carnitine-based IL. The skin permeation studies conducted using mouse skin show that the insulin permeation percentage of the developed IL/O ME is 3.55 folds higher than that of phosphate-buffered saline and 2.91 folds better than that of a hydrophilic L-(-)-carnitine-based IL. In addition, the solubility of two drug molecules, that is, rosiglitazone and bezafibrate, in IL/O ME is at least 49.28 folds higher than their solubility in water or IPM. Therefore, IL/O ME can significantly improve the solubility of drugs and increase the permeability of proteins (e.g., insulin), thus demonstrating a promising potential as a delivery carrier.</p>

Mechanism investigation of ethosomes transdermal permeation.

Abstract:Ethosomes are widely used to promote transdermal permeation of both lipophilic and hydrophilic drugs, but the mechanism of interaction between the ethosomes and the skin remains unclear. In this work, it was exploded with several technologies and facilities. Firstly, physical techniques such as attenuated total reflectance fourier-transform infrared and laser confocal Raman were used and the results indicated that the phospholipids configuration of stratum corneum changes from steady state to unstable state with the treatment of ethosomes. Differential scanning calorimetry reflected the thermodynamics change in stratum corneum after treatment with ethosomes. The results revealed that the skin of Bama mini-pigs, which is similar to human skin, treated by ethosomes had a relatively low Tm and enthalpy. Scanning electron microscopy and transmission electron microscopy showed that the microstructure and ultrastructure of stratum corneum was not damaged by ethosomes treatment. Furthermore, confocal laser scanning microscopy revealed that lipid labeled ethosomes could penetrate the skin via stratum corneum mainly through intercellular route, while during the process of penetration, phospholipids were retained in the upper epidermis. Cell experiments confirmed that ethosomes were distributed mainly on the cell membrane. Further study showed that only the drug-loaded ethosomes increased the amount of permeated drug. The current study, for the first time, elucidated the mechanistic behavior of ethosomes in transdermal application from molecular configuration, thermodynamic properties, ultrastructure, fluorescent labeling and cellular study. It is anticipated that the approaches and results described in the present study will benefit for better design of drug-loaded ethosomes.
Formulation development of transdermal dosage forms: quantitative structure-activity relationship model for predicting activities of terpenes that enhance drug penetration through human skin.

Abstract:Terpenes and terpenoids have been used as enhancers in transdermal formulations for facilitating penetration of drugs into human skin. Knowledge of the correlation between the human skin penetration effect (HSPE) and the physicochemical properties of these enhancers is important for facilitating the discovery and development of more enhancers. In this work, the HSPE of 49 terpenes and terpenoids were compared by the in vitro permeability coefficients of haloperidol (HP) through excised human skin. A first-order multiple linear regression (MLR) model was constructed to link the permeability coefficient of the drug to the lipophilicity, molecular weight, boiling point, the terpene type and the functional group of each enhancer. The Quantitative Structure-Activity Relationship (QSAR) model was derived from our data generated by using standardized experimental protocols, which include: HP in propylene glycol (PG) of 3 mg/ml as the donor solution containing 5% (w/v) of the respective terpene, the same composition and volume of receptor solution, similar human skin samples, in the same set of automated flow-through diffusion cells. The model provided a simple method to predict the enhancing effects of terpenes for drugs with physicochemical properties similar to HP. Our study suggested that an ideal terpene enhancer should possess at least one or combinations of the following properties: hydrophobic, in liquid form at room temperature, with an ester or aldehyde but not acid functional group, and is neither a triterpene nor tetraterpene. Possible mechanisms revealed by the QSAR model were discussed.

Abstract:In recent years, nanoparticles (NPs) including nanostructured lipid carries (NLC) and solid lipid nanoparticles (SLN) captured an increasing amount of attention in the field of transdermal drug delivery system. However, the mechanisms of penetration enhancement and transdermal transport properties of NPs are not fully understood. Therefore, this work applied different platforms to evaluate the interactions between skin and NPs loading triptolide (TPL, TPL-NLC and TPL-SLN). Besides, NPs labeled with fluorescence probe were tracked after administration to investigate the dynamic penetration process in skin and skin cells. In addition, ELISA assay was applied to verify the in vitro anti-inflammatory effect of TPL-NPs.
