Abstract:To determine the impact of oral physiology on the volatility of typical wine aroma compounds, mixtures of a synthetic wine with oral components (centrifuged human saliva (HS), artificial saliva with mucin (AS), and buccal epithelial cells (BC)) were prepared. Each wine type was independently spiked with four relevant wine odorants (guaiacol, β-phenyl ethanol, ethyl hexanoate, and β-ionone). Additionally, the impact of four types of phenolic compounds (gallic acid, catechin, grape seed extract, and a red wine extract) on aroma volatility in the HS, AS, and BC wines was also assessed. Static headspace was measured at equilibrium by solid phase microextraction–GC/MS analysis. Results showed a significant impact of oral components on the volatility of the four tested odorants. Independently of the type of aroma compound, aroma volatility was in general, higher in wines with BC. Moreover, while guaiacol and ethyl hexanoate volatility was significantly lower in wines with HS compared to wines with AS, β-ionone showed the opposite behavior, which might be related to metabolism and retention of mucin, respectively. Phenolic compounds also showed a different effect on aroma volatility depending on the type of compound and wine. Gallic acid had little effect on polar compounds but it enhanced the volatility of the most hydrophobic ones (ethyl hexanoate and β-ionone). In general, flavonoid type polyphenols significantly reduced the volatility of both polar (guaiacol and β-phenyl ethanol) and hydrophobic compounds (β-ionone in HS and BC wines), but through different mechanisms (e.g., π–π interactions and hydrophobic binding for polar and apolar odorants respectively). On the contrary, flavonoids enhanced the volatility of ethyl hexanoate, which might be due to the inhibition exerted on some salivary enzymes (e.g., carboxyl esterase) involved in the metabolism of this odorant molecule.

Abstract:The impact of commercial proanthocyanidic tannins on fruity pool of esters, representing the fruitiness of Bordeaux red wines, was assessed in model solutions. It was shown that the presence of tannins in the matrix significantly attenuated perception of fruity notes (p = 0.013). Physicochemical analysis demonstrated that the presence of proanthocyanidic tannins in dilute alcohol solution resulted in a decrease in ester partition coefficients and thus in a decrease in ester contents in the headspace (p < 0.05). This fact highlighted the changes that may occur in wines at a pre-sensory level, prior to sensory evaluation. Finally, a new sensory tool was developed, consisting in an ISO glass containing two compartments separated by a glass wall, providing a way to compare perceived odors according to whether or not the components of the odor mixtures were actually mixed in solution (p < 0.001). This new tool was used to demonstrate the impact and the only implication of pre-sensory level in the consequences of physical mixture between proanthocyanidic tannins and esters on their odor perception.
Effect of condensed tannins addition on the astringency of red wines

Abstract:Astringency has been defined as a group of sensations involving dryness, tightening, and shrinking of the oral surface. It has been accepted that astringency is due to the tannin-induced interaction and/or precipitation of the salivary proline-rich proteins (PRPs) in the oral cavity, as a result of the ingestion of food products rich in tannins, for example, red wine. The sensory evaluation of astringency is difficult, and the existence of fast and reliable methods to its study in vitro is scarce. So, in this work, the astringency of red wine supplemented with oligomeric procyanidins (condensed tannins), and the salivary proteins (SP) involved in its development were evaluated by high-performance liquid chromatography analysis of human saliva after its interaction with red wine and by sensorial evaluation. The results show that for low concentration of tannins, the decrease of acidic PRPs and statherin is correlated with astringency intensity, with these families having a high relative complexation and precipitation toward condensed tannins comparatively to the other SP. However, for higher concentrations of tannins, the relative astringency between wines seems to correlate's to the glycosylated PRPs changes. This work shows for the first time that the several families of SP could be involved in different stages of the astringency development.

Impact of non-volatile compounds on volatiles perception: How proanthocyanidic tannins affect red wine fruity aroma?

Abstract:Previous research on the fruity character of red wines highlighted that the perception of a “fruity pool” of esters was modified by the presence of fruity or non-fruity compounds with synergistic and/or masking effects. Moreover, some works described that non-volatile compounds may have an important impact on wine perception, but also on odorant compounds volatility. The goal of this work was to assess the olfactory consequences of a mixture between esters and proanthocyanidic tannins, through sensory and physico-chemical approaches. Sensory analyses including triangle tests, detection thresholds, and sensory profiles, were conducted in order to evaluate the sensory impact of pure tannins on red wines “fruity pool” perception. Then, the impact of these non-volatile molecules on esters volatility, and thus taster stimulation, was evaluated thanks to the determination of partition coefficients. Triangular test revealed a significant difference on global perception between the fruity pool containing esters and the same fruity pool in mixture with proanthocyanidic tannins. Sensory profiles evaluation showed fruity notes perception were significantly lower for the fruity pool supplemented with tannins compared to the fruity pool alone in dilute alcohol solution. These results confirmed the sensory importance of some non-volatile compounds on odour perception. Finally, esters partition coefficient evaluation revealed a decrease of the volatility of esters when tannins were present in the matrix, thus corroborating sensory evaluation results. when they are added in the matrix, thus reducing orthonasal taster stimulation and consequently reducing red wine fruity notes perception. Such a study should be extended to other wine tannins and anthocyanins, including their concentration ranges, to assess the impact of the phenolic matrix on red wines aroma perception. work was to provide a global overview on the impact of proanthocyanidic tannins on the aroma of a 13-ester mixture representative of Bordeaux red wines fruity aroma. The sensory consequences of the mixture were studied as well as the presence of tannins on ester volatility, which was measured by calculating their partition coefficients, thus allowing us to evaluate the involvement of pre-sensory changes. 1, adding FAR into TS led to significant changes in the perception of various descriptors, compared to FAR in DAS. The presence of tannins at 5 g/L in the matrix led to an attenuation of the perception of overall aroma (p < 0.05), red-berry- and fresh-fruit (p < 0.01). These results were in agreement with those observed in the triangle tests, with a significant difference between the perception of FAR in DAS and in TS.

Abstract:Wine polyphenols (PPhs) and volatile organic compounds (VOCs) are responsible for two of the main sensory characteristics in defining the complexity and quality of red wines: astringency and aroma. Wine VOCs’ volatility and solubility are strongly influenced by the matrix composition, including the interactions with PPhs. To date, these interactions have not been deeply studied, although the topic is of great interest in oenology. This article reviews the available knowledge on the main physicochemical and sensory effects of polyphenols on the release and perception of wine aromas in orthonasal and retronasal conditions. It describes the molecular insights and the phenomena that can modify VOCs behavior, according to the different chemical classes. It introduces the possible impact of saliva on aroma release and perception through the modulation of polyphenols–aroma compounds interactions. Limitations and possible gaps to overcome are presented together with updated approaches used to investigate those interactions and their effects, as well as future perspectives on the subject.

Abstract:Wine is one of the most consumed beverages around the world. It is composed of alcohols, sugars, acids, minerals, proteins and other compounds, such as organic acids and volatile and phenolic compounds (also called polyphenols). Polyphenols have been shown to be highly related to both (i) wine quality (color, flavor, and taste) and (ii) health-promoting properties (antioxidant and cardioprotective among others). Polyphenols can be grouped into two big families: (i) Flavonoids, including anthocyanidins, flavonols, flavanols, hydrolysable and condensed tannins, flavanones, flavones and chalcones; and (ii) Non-flavonoids, including hydroxycinnamic acids, hydroxybenzoic acids, stilbenes, tyrosol and hydroxytyrosol. Each group affects in some way the different properties of wine to a greater or a lesser extent. For that reason, the phenolic composition can be managed to obtain singular wines with specific, desirable characteristics. The current review presents a summary of the ways in which the phenolic composition of wine can be modulated, including (a) invariable factors such as variety, field management or climatic conditions; (b) pre-fermentative strategies such as maceration, thermovinification and pulsed electric field; (c) fermentative strategies such as the use of different yeasts and bacteria; and (d) post-fermentative strategies such as maceration, fining agents and aging. Finally, the different extraction methods and analytical techniques used for polyphenol detection and quantification have been also reviewed.

Abstract:Polysaccharides have an important role in the technological and sensory characteristics of wines. The aim of this work was to study the effects of the addition of four polysaccharide extracts obtained from grape products and byproducts to red wines during storage for 2 months on their phenolic composition and color. The four extracts rich in polysaccharides were obtained from grape must, white grape pomace, red grape marc, and red wine, and they were compared with a commercial inactivated yeast. These products were studied in three wines selected for their highest astringency and acidity characteristics. The highest differences were found in the red wines with high initial phenolic concentrations, which reduced their values. The addition of polysaccharide extracts from grape pomace or marc, must, or yeast can mainly be interesting in wines with high phenolic content since they may be useful to modulate the astringency of red wines. This is the first work that studies the effect of polysaccharide extracts obtained from grape byproducts in red wines, showing great possibilities of these products.
Impact of Perceptive Interactions on Red Wine Fruity Aroma

Abstract:A preparative HPLC method was applied to aromatic red wine extracts. Twenty-five fractions with various flavors were thus obtained, and several aromatic reconstitutions were produced by mixing some of these fractions. Discriminative tests revealed that the omission of some fractions from the mixture of fruity fractions or the addition of others affected the overall expression of fruity aroma. Sensory profile analyses identified significant differences among aromatic reconstitutions in terms of intensity of black-berry, as well as fresh-, and jammy-fruit descriptors. A fraction with a very low fruity note (fraction 17) had an additive effect on the fresh fruity aroma, while fractions with caramel and lactic notes (fractions 3-5) had a masking effect on this aroma and an additive effect on the jammy-fruit aroma. Further analysis revealed that ethyl 2-hydroxy-4-methylpentanoate was eluted in fraction 17, while diacetyl, acetoin, acetic acid, and γ-butyrolactone were eluted in fractions 3-5. Omissions tests established that ethyl 2-hydroxy-4-methylpentanoate was responsible for enhancing black-berry and fresh-fruit aroma and that a combination of diacetyl, acetoin, acetic acid, and γ-butyrolactone, at levels between 2 and 40% of their perception thresholds, had the same hypoadditive effect on the overall and fresh fruity aroma as fractions 3-5.

Abstract:Astringency and bitterness in red wines are generally considered to be elicited by phenolic compounds and especially by tannins. This study explored the effects of pH, ethanol level and tartaric acid concentration in modifying astringency and bitterness of oligomeric tannins in model wine solutions. Bitterness and overall astringency intensities were successively rated by 16 judges. As ethanol level and pH values increased, the astringency perception was lowered. While pH affected only astringency, ethanol contributed also to the perceived bitterness of tannin oligomers, especially at typical wine ethanol levels (11–15%). The variation of tartaric acid concentration at constant pH did not have any effect on either of these sensory properties.

Abstract:Extracts containing anthocyanins (ACN), and skin tannins (SKIN) and seed tannins (SEED) were prepared from Vitis vinifera cv. Cabernet Franc grapes grown in the Loire Valley, and characterised. Phenolic fractions from Cabernet Franc wines made from three Loire Valley locations were also isolated and characterised. Bitterness and astringency of ACN, SEED and SKIN as well as the wine extracts were evaluated by time intensity procedures in citric acid solution and in a base white wine. SEED and SKIN were equally astringent when tasted at the same concentration in spite of differences in tannin composition. The lower molecular weight (MW) of SEED was equal in astringency to larger MW SKIN which had a lower percentage of galloylation. The SEED fraction was slightly more bitter than the SKIN fraction in the citric acid solution, although no difference could be detected between samples in base white wine. Astringency of ACN alone was much lower than either SKIN or SEED. Addition of ACN to either tannin fraction produced very small sensory effects in citric acid. In wine, addition of ACN to either SEED or SKIN increased astringency significantly over either fraction alone, but had no effect on bitterness. The wine fractions differed only in astringency, which was correlated with tannin units as determined by thiolysis.
Effects of Plant-Derived Polyphenols on the Antioxidant Activity and Aroma of Sulfur-Dioxide-Free Red Wine

Abstract:Significant efforts have been made in recent years to produce healthier wines, with the primary goal of reducing the use of sulfur dioxide (SO2), which poses health risks. This study aimed to assess the effectiveness of three plant-derived polyphenols (dihydromyricetin, resveratrol, and catechins) as alternatives to SO2 in wine. After a three-month aging process, the wines were evaluated using analytical techniques such as high-performance liquid chromatography, colorimetry, gas chromatography–olfactometry–mass spectrometry, as well as electronic nose and electronic tongue analyses, with the purpose to assess parameters including antioxidant activity, color, contents of volatile aroma compounds, and sensory characteristics. The results demonstrated various degrees of improvement in the antioxidant activity, aromatic intensity, and sensory characteristics of wines using polyphenols. Notably, dihydromyricetin (200 mg/L) exhibited the strongest antioxidant activity, with increases of 18.84%, 23.28%, and 20.87% in 2,2-diphenyl-1-picrylhydrazyl, 2,2'azino-bis(3-ethylbenzothiazoline-6-sulfonic acid), and ferric-ion-reducing antioxidant power assays, respectively. Resveratrol (200 mg/L) made the most significant contribution to volatile aroma compounds, with an 8.89% increase in the total content of alcohol esters. In E-nose analysis, catechins (200 mg/L) showed the highest response to aromatic compounds and the lowest response to volatile sulfur compounds, while also exhibiting the best sensory characteristics. Therefore, the three plant-derived polyphenols investigated here exhibited the potential to enhance wine quality as alternatives to SO2. However, it is important to consider the specific impact of different polyphenols on wine; hence, suitable antioxidants should be selected in wine production according to specific requirements.

Towards an understanding of bitterness in white wines: Contribution of 27 compounds assessed by LC-HRMS and sensory analysis

Abstract:Whereas bitterness perception can modify the taste balance of white wines, its molecular origin remains largely unclear. This work aimed at determining the influence of a selection of the most cited bitter compounds on the bitterness of commercial dry white wines. Forty-two wines were sensorially characterized by a trained panel and divided into two statistically different groups depending on their bitterness. Twenty-seven bitter compounds were selected and five quantitation methods were developed and validated. The methods were used to measure the levels of all the 27 compounds in dry wine, 25 of them in sweet wine and 22 of them in grape juice. The detected concentrations were generally below the taste detection thresholds. No significant positive correlation between the bitterness intensity of the tasted samples and the concentration of the assayed bitter compounds was observed, suggesting the existence of other markers of bitterness in white wines.

Abstract:The effect of vinification techniques on phenolic compounds and antioxidant activity of wine lees are poorly understood. The present study investigated the antioxidant activity of white and red wine lees generated at early fermentation and during aging. In this study, the total phenol content (TPC), total tannin content (TTC), mean degree of polymerization (mDP), and antioxidant activities of five white and eight red wine lees samples from different vinification backgrounds were determined. The results showed that vinification techniques had a significant (p < 0.05) impact on total phenol and tannin content of the samples. White wine lees had high mDP content compared with red ones. Catechin (50-62%) and epicatechin contents were the predominant terminal units of polymeric proanthocyanidin extracted from examined samples. Epigallocatechin was the predominant extension unit of white wine lees, whereas epicatechin was the predominant compound in red wine marc. The ORAC (oxygen radical absorbance capacity) assay was strongly correlated with the DPPH (α, α-diphenyl-β-picrylhydrazyl) assay, and the results showed the strong antioxidant activities associated with red wine lees (PN > 35 mg Trolox/g FDM) (PN: Pinot noir lees; FDM: Freeze-dried Material). This study indicates that tannin is one of the major phenolic compounds available in wine lees that can be useful in human and animal health applications.

Abstract:Due to the increase of the use of yeast derivatives (YDs) in winemaking to improve the technological and sensory properties in wines, in this work we evaluated the effect of the post-fermentation application of different yeast derivative products on the physical and chemical properties and astringency of red wines during two consecutive harvests. A commercial and two experimental new yeast derivatives were applied at a medium‒high dosage (30 g/hL). The addition of different yeast derivatives in red wine increased the concentration of different polysaccharide fractions and, therefore, the total polysaccharide content, producing a decrease in the duration of the wine astringency perception over time. The use of yeast derivatives could produce an adsorption/clarification and/or protective effect on the phenolic compounds. However, it did not produce an important modification of the colour parameters. An intensification or a lower decrease of the most volatile compound groups was produced, but it depended on the YDs and yeast strain used in fermentation and post-fermentation processes.

Abstract:Tannin addition as an enological practice has been widely used in the winemaking process because of their ability of improving the aroma and sensory characteristics and stabilizing of color of red wine. In this study, hydrolysable, condensed tannins and their mixtures in different ratios were added into two Merlot wines to investigate their effect on the wine overall quality. The contents of 15 phenolic compounds were detected by HPLC-DAD, CIELAB color parameters were measured using a chromatic aberration meter, sensory evaluation was accomplished using the assessment standards established by the American Wine Association, and antioxidant activities were analyzed using DPPH and ABTS radical tests. The results indicated that adding tannins affected phenolic composition, contents and color of wine. The specific effects varied by tannins. Furthermore, tannin addition, especially the mixed tannins, improved the sensory qualities and antioxidant activities greatly. The mixed tannins added with a ratio of 1 : 1 between hydrolyzable and condensed tannins exhibited a better effect on both sensory qualities and antioxidant activities, and it could be recommended as an ideal tannin addition for wine quality improvement.

Abstract:Red wines and model solutions containing either grape seed phenols or (+)-catechin were assessed for perceived astringency by a panel of trained assessors using time−intensity (T−I) methodology. The effect of acid on the perception of astringency was evaluated by using three different sensory protocols. Maximum intensity (I max), total duration (T tot), and time to reach maximum intensity (T max) of astringency were significantly increased when the acid solution was tasted after the astringent model solutions as well as when a mixture of acid and astringent compounds was assessed. However, when the panelists rinsed their mouths with the acid solution before tasting the model solutions, astringency parameters remained unaffected. For wine samples, I max of astringency decreased significantly when the acid was tasted after the control wine, whereas it remained unaffected when it was tasted before. Abstract:The wine industry has increased its interest in finding new fining agents, and vegetable proteins have received increasing attention. Quinoa and kiwicha are native pseudocereals to the Andean highlands with high protein content and are considered gluten-free products. This work aimed to determine the effect of fining with quinoa and kiwicha protein extracts (QP and KP respectively) at different doses (30 and 50 g/hL) on the aroma composition, color, and phenolics of a Monastrell wine compared to pea proteins and gelatin. Fining treatments produced no significant reduction of TPI. Except for QP at 30 g/hL and the lowest dose of pea proteins, the fining treatments decreased the content of total anthocyanins. The doses of 50 g/hL of QP and KP were particularly efficient, producing the largest decrease in total tannins. Analyses by size exclusion chromatography showed that treatments with QP and KP were capable of selectively removing high molecular weight phenolics. All fining agents slightly decreased the color intensity. All fining treatments showed a significant decline in total ester concentration (26-45%). Instead, total alcohols remained constant upon the fining treatments. Five terpenes were identified in all wines, and their content was not affected by the fining treatments.
Characterization of sensory perceptions elicited by white wine spiked with different aroma, taste and mouth-feel active molecules

Abstract:The present work was aimed at understanding the sensory responses induced by dry white wine modified with increasing concentrations of different sensory active molecules. The tasting panel was composed by 34 trained subjects characterized according to gender, smoking habits, 6-n-propylthiouracil (PROP) taster status, and sensitivity to tartaric acid, tannic acid and sucrose. Additional taste/mouthfeel responsiveness was evaluated in a basal white wine added of tartaric acid, tannic acid and sucrose. The addition of a fruity odorant mixture to the base white wine enabled the assessment of orthonasal aroma and taste intensities (sweetness, sourness, saltiness). The diversity of taste/mouthfeel responsiveness in water or wine enabled to group individuals as high or low sensitives. Likewise, the tasting panel showed two groups responding differently to aroma and flavor. Both high and low aroma sensitivity individuals showed equal (p>0.05) and congruent response to in-mouth sweet flavor perception. Moreover, the high smell sensitive group was less sensitive to sourness and saltiness than the low smell sensitives for the spiked wines across all flavor concentrations. Differences in PROP taster status and sensitivity to other tastants in water solutions were not correlated with the taste/mouthfeel perceptions in wine. The individuals most sensitive to sucrose in wine taste showed higher response to the fruity aroma. In conclusion, taste, mouthfeel and flavor perceptions showed a high variability among individuals evidencing the advantage of grouping tasters with different chemosensory sensitivities to understand cross-modal sensory interactions.

Abstract:Two stereoisomeric phenolic compounds, (+)‐catechin and (‐)‐epicatechin, were rated for perceived intensity of oral astringency and bitterness by trained judges using the scalar method. Mouth drying and mouth roughening were also assessed, since they are often associated with astringency. Amounts of 375, 750 and 1500 mg/L of each compound were tasted in red wine, and in a model system, similar in composition to a dry table wine. Preliminary tests showed that these concentrations were above the threshold level but within the range found in wine. A control sample (model solution or wine without the addition of the above phenolic compounds) was also evaluated. The results showed that the two compounds were both bitter and astringent. The high (‐)‐epicatechin concentration was significantly more bitter and astringent than the equal concentration of (+)‐catechin in the model solution. Mouth drying and roughening ratings showed a similar increasing pattern with the ratings of astringency particularly at the higher concentrations. However, these attributes were rated differently from astringency suggesting that although they contribute to astringency, they are not subsumed by it.
