Abstract:At present, many researchers are exploring biological tissue inspection using terahertz time-domain spectroscopy (THz-TDS) techniques. In this study, based on a modified hard modeling factor analysis method, terahertz spectral unmixing was applied to investigate the relationships between the absorption spectra in THz-TDS and certain biomarkers of gastric cancer in order to systematically identify gastric cancer. A probability distribution and box plot were used to extract the distinctive peaks that indicate carcinogenesis, and the corresponding weight distributions were used to discriminate the tissue types. The results of this work indicate that terahertz techniques have the potential to detect different levels of cancer, including benign tumors and polyps.

Abstract:Liver cancer usually has a high degree of malignancy and its early symptoms are hidden, therefore, it is of significant research value to develop early-stage detection methods of liver cancer for pathological screening. In this paper, a biometric detection method for living human hepatocytes based on terahertz time-domain spectroscopy was proposed. The difference in terahertz response between normal and cancer cells was analyzed, including five characteristic parameters in the response, namely refractive index, absorption coefficient, dielectric constant, dielectric loss and dielectric loss tangent. Based on class separability and variable correlation, absorption coefficient and dielectric loss were selected to better characterize cellular properties. Maximum information coefficient and principal component analysis were employed for feature extraction, and a cell classification model of support vector machine was constructed. The results showed that the algorithm based on parameter feature fusion can achieve an accuracy of 91.6% for human hepatoma cell lines and one normal cell line. This work provides a promising solution for the qualitative evaluation of living cells in liquid environment.

Abstract:Terahertz technology has been widely used as a nondestructive and effective detection method. Herein, terahertz time-domain spectroscopy was used to detect drug-induced liver injury in mice. Firstly, the boxplots were used to detect abnormal data. Then the maximal information coefficient method was used to search for the features strongly correlated with the degree of injury. After that, the liver injury model was built using the random forests method in machine learning. The results show that this method can effectively identify the degree of liver injury and thus provide an auxiliary diagnostic method for detecting minor liver injury. Abstract:Terahertz wave technique is very prospective in the biomedical application field, where there exists a practical problem that it is hard to register a terahertz time-domain spectroscopy (THz-TDS) detection point and its accurate corresponding position in pathology image. In this paper, complex human gastric tissue is taken as the experimental subject to search for accurate detection approach. This paper uses tissue microarray technology to process paraffin-embedded gastric tissue, and then contrasts the Terahertz absorption coefficient and refractive index spectrum of detection subjects which are divided into registered experimental group and unregistered control group using principal component analysis method. Meanwhile, this paper contrasts the gastric tubular adenocarcinoma THz-TDS discrimination methods using support vector machine and logistic regression respectively. The research result reveals that the detection accuracy brought by tissue microarray technology helps better discriminate tumor samples and normal samples.

Abstract:Human dehydrated normal and cancerous gastric tissues were measured using transmission time-domain terahertz spectroscopy. Based on the obtained terahertz absorption spectra, the contrasts between the two kinds of tissue were investigated and techniques for automatic identification of cancerous tissue were studied. Distinctive differences were demonstrated in both the shape and amplitude of the absorption spectra between normal and tumor tissue. Additionally, some spectral features in the range of 0.2 similar to 0.5 THz and 1 similar to 1.5 THz were revealed for all cancerous gastric tissues. To systematically achieve the identification of gastric cancer, principal component analysis combined with t-test was used to extract valuable information indicating the best distinction between the two types. Two clustering approaches, K-means and support vector machine (SVM), were then performed to classify the processed terahertz data into normal and cancerous groups. SVM presented a satisfactory result with less false classification cases. The results of this study implicate the potential of the terahertz technique to detect gastric cancer. The applied data analysis methodology provides a suggestion for automatic discrimination of terahertz spectra in other applications.

Abstract:At present, the terahertz time-domain spectroscopic information of each component in multi-composition compounds detection is comprehensively combined. The lower resolution level of mixed spectra has posed many difficulties in signal analysis due to the overlapping characteristic information in the mixed ones. In this paper, to compare the performances of Nonnegative Matrix Factorization (NMF), Self-modeling Mixture Analysis (SMMA) and Multivariate Curve Resolution-Alternating Least Squares (MCR-ALS) on complex systems, a binary mixture and a ternary mixture are employed during THz-TDS testing. The position of the absorption peak (PK) and the correlation coefficient are used to evaluate the decomposition effects. The experimental results show that the component spectra resolved by MCR-ALS demonstrate good consistency with the sample components. Further, MCR-ALS presents excellent results in comparison with NMF and SMMA in terms of decomposing precision and computing speed. MCR-ALS thus appears a promising algorithm to resolve the THz multi-way data, and may be useful for many applications, such as medicine quality assurance and unknown component identification in the general area of terahertz science and technology.

Abstract:Terahertz (THz) radiation is the electromagnetic spectrum lying between the microwave and infrared portions, and covers wavelength between 3mm and 30um. Terahertz spectroscopy has been used in qualitative and quantitative analysis since molecular vibrations and rotations of a great large number of materials lie in this region. Many techniques, such as principal component regression (PCR) and partial least square regression (PLSR), have been applied in quantitative analysis of terahertz time domain spectroscopy. However, those "soft-modeling" methods, though effective in many cases, all require a large number of samples and have poor extrapolation capability, and the utilization is also restricted in some cases (samples lack or requirement of extrapolation). A parametric method of spectrum, Indirect Hard Modeling (IHM), was proposed to resolve terahertz spectral matrix and quantify the mixtures. Savitzky-Golay method was used to eliminate the influence of noise. Moreover, the features of terahertz spectra were considered and the artificial baseline was also resolved as a signal spectrum to improve spectrum extraction accuracy. Subsequently, the analysis of IHM was carried out, and the ability of prediction and accuracy of the model in the case of low training samples were discussed. In order to validate the capability of the proposed method, quaternary systems of Rifampicin, Lactose monohydrate, microcrystalline cellulose and magnesium stearate were tested. The regression correlation coefficient (R) and root mean square error (RMSE) were employed to estimate the performance of the model. Finally, the comparison of IHM and PLSR were carried out, and theoretical analysis and experimental results indicated that the number of samples required by IHM modeling could be reduce to two during calibration, at the same time, the accuracy of quantitative and extrapolation capabilities were also improved.

Abstract:Tumor genesis is accompanied by glycosylation of related proteins. Glycoprotein is usually regarded as a tumor marker since glycoproteins are consumed remarkably more by the cancer cells than the normal ones. In this paper, the terahertz time-domain attenuated total reflection (ATR) technique is applied to inspect the glycoprotein solution from a concentration gradient of 0.2 mg/ml to 50 mg/ml. A significant nonlinear relationship between the absorption coefficient and the concentrations has been discovered. The influence of the dynamical hydration shell around glycoprotein molecules on the absorption coefficient is discussed and the phenomenon is explained by the concepts of THz excess and THz defect. In order to identify glycoproteins, features are obtained by composite multiscale entropy (CMSE) method and clustered by the K-means algorithm. The results indicate that features extracted by the CMSE method are better than the Principal Component Analysis (PCA) method in both specificity and sensitivity of recognition. Meanwhile, the absorption coefficient and dielectric loss angle tangent are more suitable for qualitative identification. Research shows that the CMSE method has important directive significance for analyzing glycoprotein terahertz spectroscopy. And it has the potential for glycoprotein related tumor markers identification using terahertz technology in medical applications.

Abstract:Terahertz technology is a non-invasive and label-free inspection method, promising for applications in various fields, especially in biomedical diagnosis. Terahertz radiation is capable for cell physiological and pathological status recognition due to its sensitivity to cell composition and structural information of biomacromolecular. In this paper, the transmission properties of glass, polymethyl methacrylate and polypropylene in THz regime were compared and polypropylene was selected as the petri dish material for cell adhesion due to its high transmission of terahertz wave. The time-domain and frequency-domain spectra, refractive index and absorption coefficient of hepatoma cell line Huh7 and normal cell line MIHA were analyzed. The experimental results showed significant distinction between the terahertz characteristic spectra of hepatoma cells and normal cells. This work can be helpful for applying terahertz technology in cancer detection at the cellular level.
Blind Spectral Unmixing in Terahertz Domain Using Nonnegative Matrix Factorization

Abstract:Recovering component spectra from terahertz measurements of unknown mixtures has been studied in this paper using nonnegative matrix factorization (NMF). NMF mathematically decomposes the spectra data into two nonnegative matrixes which describe the component spectra and the corresponding fractional abundance. Two basic algorithms in the class of this method, NMF and NMF with smoothness constraint (cNMF), were adopted to resolve the terahertz absorption spectra matrix obtained from a ternary mixture with varying compositions of Nitrofurantoin, L-Leucine and D-Tyrosine. The quality of the decomposition results was evaluated. The performance of the two algorithms on extracting component terahertz spectra was compared. The optimal result reached by cNMF in this study implies the capability of the NMF method for blind terahertz spectral unmixing. The attempt made in our work helps to further investigate unknown mixtures by terahertz spectroscopy.

Abstract:In this paper, we propose a discrimination model for abnormal sialic acid content using terahertz time and frequency-domain features of samples in an attempt to explore the significance of human serum sialic acid content in early cancer screening. Specifically, the time-domain spectral features were obtained by applying variational mode decomposition (VMD) for time-domain spectrum preprocessing and composite multi-scale entropy (CMSE) for feature extraction. The frequency-domain spectral features were acquired using principal component analysis (PCA). The model was then constructed on the basis of support vector machine (SVM). To verify the reliability of this discrimination model, we used three different concentrations of albumin solutions to simulate the serum environment in the human body and mixed them with sialic acid solutions of eight different concentrations. The results indicate that our discrimination model achieved an accuracy of 91.01% for the detection of abnormal sialic acid content. This study provides a foundation for the accurate quantitative detection of biomarkers in human serum.

Abstract:Terahertz waves are known for their bio-safety and spectral fingerprinting features, and terahertz spectroscopy technology holds great potential for both qualitative and quantitative identification in the biomedical field. There has been a substantial amount of research utilizing this technology in conjunction with machine learning algorithms for substance identification. However, due to the strong absorption of water for terahertz waves, the single-dimensional features of the sample become indistinct, thereby diminishing the efficiency of the algorithmic recognition. Building upon this, we propose a method that employs terahertz time-domain spectroscopy (THz-TDS) in conjunction with multidimensional feature spectrum identification for the detection of blood sugar and glucose mixtures. Our research indicates that combining THz-TDS with multidimensional feature spectrum and linear discriminant analysis (LDA) algorithms can effectively identify glucose concentrations and detect adulteration. By integrating the multidimensional feature spectrum, the identification success rate increased from 68.9% to 96.0%. This method offers an economical, rapid, and safe alternative to traditional methods and can be applied in blood sugar monitoring, sweetness assessment, and food safety.

Abstract:At present, researchers are exploring biological tissue detection method using terahertz techniques. In this paper, techniques to inspect mouse liver injury by using terahertz spectroscopy were studied. The boxplots were applied to remove abnormal data, and the maximal information coefficient was employed to select crucial features from the absorption coefficient and refractive index spectra. Random Forests and AdaBoost were applied to recognize different levels of liver injury. We found that AdaBoost had better performance on low-level injury classification. This work suggests that terahertz techniques have the potential to detect liver injury at an early stage and evaluate liver treatment strategies.

Abstract:Terahertz time-domain spectroscopy (THz-TDS) has been applied to medical applications as THz waves are sensitive to biomolecules. In this paper, terahertz time-domain attenuated total reflection spectroscopy (THz-ATR) was used to identify human colon cancer cell line. The refractive indices of cancerous and normal cell lines with six different concentrations were analyzed in the frequency domain from 0.1-1.2THz. t-Distributed Stochastic Neighbor Embedding (t-SNE) and Principal Components Analysis (PCA) methods were adopted to extract the key features from the THz spectrum. Experimental results showed that the features extracted by t-SNE had better recognition accuracy owing to enhanced clustering efficacy. This work may be helpful for detecting human colon cancer cells by using terahertz techniques.

Abstract:Sialic acid is one of the key constituents in tumor-specific glycans and an important biomarker for a diversity of cancer types. Its content change in the blood serum can be used for early diagnosis of cancer, grading of cancer, and evaluation of therapeutic effect and prognosis. In this paper, we studied and analyzed the terahertz (THz) response of solid sialic acid samples with five concentrations. Based on the characteristics of the peak value among absorption coefficient spectrum, linear fitting is performed to achieve the quantitative detection of sialic acid.

Abstract:Terahertz spectroscopy has been widely used for investigating the fingerprint spectrum of different substances. For cancerous tissues, the greatest difficulty is the absorption peaks of various substances contained in tissues overlap with each other, which are hard to identify and quantitative analyze. As a result, it is very hard to measure the presence of cancer cell and then to diagnose accurately. In this paper, we select three typical neurotransmitters (γ-aminobutyric acid, L-glutamic acid, dopamine hydrochloride) and two typical metabolites (inositol and creatine) in neurons to measure their terahertz spectra with different mixture ratios. By choosing characteristic absorption peaks, removing baseline and using the least square method, we can identify the components and proportions of each mixture, where the goodness of fit to practical situation is up to 94%. These results provide important evidences for identifying nerve substances and obtaining exact quantitative analysis.

Abstract:The distinctive vibrational features in terahertz (THz) spectroscopy characterize a "fingerprint" of the single-component molecular substance. However, due to componential spectral overlapping and baseline drift, the identification and quantification of multicomponent mixtures are quite challenging for THz spectral analysis. A systematic and feasible strategy has been proposed by combining machine learning with THz spectroscopy for both qualitative and quantitative analysis. After the component number was effectively determined by singular value decomposition (SVD), nonnegative matrix factorization (NMF) and self-modeling mixture analysis (SMMA) were applied to extract componential THz spectra. The difficulties of NMF and SMMA encountered in handling ternary mixtures were solved. The results show component spectra extracted by SMMA are highly consistent with the experimental spectra of pure substances after standardization to correct baseline drift, which greatly facilitates rapid identification of compositions in mixtures. Additionally, compared to back-propagation neural network (BPNN), support vector regression (SVR) predict the contents of each individual component with high robustness and the decision coefficient R 2 greater than 0.949. Fingerprint terahertz spectroscopy enhanced by machine learning provided an effective strategy for mixture analysis in practical applications.

Abstract:The aim of this work is to extract component spectra from unknown mixtures in the terahertz region. To that end, a method, hard modeling factor analysis (HMFA), was applied to resolve terahertz spectral matrices collected from the unknown mixtures. This method does not require any expertise of the user and allows the consideration of nonlinear effects such as peak variations or peak shifts. It describes the spectra using a peak-based nonlinear mathematic model and builds the component spectra automatically by recombination of the resolved peaks through correlation analysis. Meanwhile, modifications on the method were made to take the features of terahertz spectra into account and to deal with the artificial baseline problem that troubles the extraction process of some terahertz spectra. In order to validate the proposed method, simulated wideband terahertz spectra of binary and ternary systems and experimental terahertz absorption spectra of amino acids mixtures were tested. In each test, not only the number of pure components could be correctly predicted but also the identified pure spectra had a good similarity with the true spectra. Moreover, the proposed method associated the molecular motions with the component extraction, making the identification process more physically meaningful and interpretable compared to other methods. The results indicate that the HMFA method with the modifications can be a practical tool for identifying component terahertz spectra in completely unknown mixtures. This work reports the solution to this kind of problem in the terahertz region for the first time, to the best of the authors' knowledge, and represents a significant advance toward exploring physical or chemical mechanisms of unknown complex systems by terahertz spectroscopy.
Terahertz Spectroscopy for Quantifying Solid Mixtures

Abstract:A quantitative detection method based on the terahertz absorption spectra was developed to analyze solid mixture. Specifically mixtures of anhydrous theophylline and lactose monohydrate were analyzed experimentally. Samples with known compositions were used to calibrate the system which was then used to measure the theophylline and lactose concentrations with average errors of 5.39% and 6.91%, respectively. With lactose as an unknown component, the absorption peak was fitted with the Lorentzian lineshape and a polynomial. The theophylline concentrations calculated using the peak intensity had an average error of 6.64%. These results demonstrate that the system can be used to quantitatively analyze mixture components based on the characteristic information in terahertz spectra for quality control applications.

Abstract:In this paper, we use the terahertz time-domain spectroscopy system to measure the terahertz absorption spectra of three common food additives, namely, benzoic acid, sorbic acid, and xylitol, along with their mixtures. In addition, we select three machine learning algorithms to analyze the binary and ternary mixtures of food additives, namely, the partial least squares regression (PLS), the least squares support vector machine (LS-SVM), and the backpropagation neural network (BPNN). We find that in the quantitative analysis of multivariate mixtures, the nonlinear models LS-SVM and BPNN are more advantageous than the linear model PLS. As the mixture composition increased, the advantages of using a nonlinear model for analysis become more obvious. Among the two nonlinear models, we find that LS-SVM has a fixed modeling step compared with that of BPNN and does not require complicated parameter discussion and optimization, which can efficiently realize the quantitative analysis of multivariate mixtures. Moreover, by observing the spectral characteristics of the analyse, it is found that in addition to the discussion of the applicability of the algorithm, the spectral characteristics of the analyte also affect the accuracy of quantitative detection to a certain extent.
