Abstract:This study describes an attempt to develop and validate a measure of patient satisfaction with a district nursing service in a south-west London inner city borough. The patients were 126 house-bound elderly people living alone and dependent upon district nursing services. The patient satisfaction measure was derived from an earlier North American questionnaire assessing satisfaction with hospital nursing services. The results indicated that the measure chosen reflected multiple dimensions of patient satisfaction that were differentially sensitive to factors such as the degree of personalization of care and the impact of disruptions to the service. Scores on the questionnaire were also related in a systematic way with the number and type of spontaneous comments made about the service. Overall the results suggest that self-report questionnaire measures of satisfaction can be devised that are sensitive to variations in the style and level of community nursing offered to older house-bound people.

Abstract:In this study, two novel fluorescence quenching biomimetic enzyme-linked immunosorbent assays were constructed for the sensitive detection of enrofloxacin (ENRO) in animal-derived foods. In this strategy, the fluorescence of quantum dots (QDs) was efficiently quenched by the enzymatic formation of horseradish peroxidase (HRP) and Prussian blue nanoparticles (PBNPs) in biomimetic enzyme-linked immunosorbent (BELISA). Under the optimal conditions, the detection limit (IC 15 ) of the method using HRP was 1.67 ng/L, and the sensitivity (IC 50 ) was 0.57 μg L −1 , while IC 15 of the method using PBNPs reached to 1.02 ng/L, and IC 50 was 0.04 μg L −1 . The recoveries of these proposed methods ranged from 83.37 to 118.08 %, with the CV values of 0.32–17.01 %. The results from the proposed methods showed high agreement with results yielded by using HPLC. In a conclusion, this conversion of the absorption signal into fluorescent signals led to an increase in the sensitivity of ENRO detection in animal-derived foods. And it is worth noting that compared to the proposed fluorescence quenching BELISA using HRP as catalyst, the method using PBNPs as catalyst showed a higher sensitivity with a 14-fold lower IC 50 , demonstrating the good application prospect of nano-enzyme in biomimetic immunoassay.

Abstract:In recent years, due to exceptional properties like broad interlayered spacing and low working potential, MXene-derived titanate nanoribbons have been established as promising electrode materials. Herein, the electrocatalytic activity of MXene-derived potassium titanate nanoribbon was employed to develop a voltammetric sensor for the detection of enrofloxacin. The sensor's significance is to provide a sustainable solution to quantify the presence of enrofloxacin regarding food safety and environmental monitoring. Moreover, to achieve the United Nations' Sustainable Development Goals by preventing antimicrobial resistance to accomplish the One Health approach. Potassium titanate nanoribbons were synthesized using 2D Ti3C2 MXene as an active precursor material, while X-ray diffraction spectroscopy, field emission scanning electron microscopy, high-resolution transmission electron microscopy, selected area electron diffraction pattern, elemental mapping, and energy-dispersive X-ray spectroscopy were used to characterize the crystallinity, surface and layered morphology of synthesized nanoribbons. The Brunauer-Emmett-Teller (BET) technique was applied to calculate the specific surface area of the synthesized materials. The materials underwent electrochemical characterization using cyclic voltammetry (CV), differential pulse voltammetry (DPV), and electrochemical impedance spectroscopy (EIS). Later on, the nanoribbons were fabricated on the surface of a glassy carbon electrode, and the electro-oxidative behaviour of enrofloxacin was studied by CV, DPV, square wave voltammetry (SWV) in 0.1 M phosphate buffer (optimized pH 8). The developed sensor depicts a significantly lower limit of quantification of 0.007 μM (≈2.5 μg/L), and an upper limit of quantification of 18 μM (≈6.5 mg/L) along with a limit of detection (LOD) of 0.00279, 0.00803, 0.00881 μM obtained from CV, DPV, and SWV respectively. Furthermore, the developed electrodes show a reliable selectivity to be examined in real complex matrices, i.e. marine water, river water, agricultural soil, organic fertilizer, milk, honey, and poultry egg.
A colorimetric-aptamer-based assay for the determination of enrofloxacin through triggering the aggregation of gold nanoparticles

Abstract:Although enrofloxacin (ENR) is a widely used broad-spectrum antibiotic in veterinary medicine, its residues in animals can pose a risk to human health. Thus, we developed a new method for detecting ENR based on aptamers and AuNPs. In the absence of ENR, the aptamers attached to the surface of the AuNPs via electrostatic interactions to protect the AuNPs from NaCl, and the solution remained red. Conversely, the aptamer bonded with ENR, leading the aptamer to detach from the AuNP surface, and the color of the solution changed from red to blue. Based on this principle, ENR can be qualitatively detected by the naked eye and quantitatively detected by measuring the absorbance ratio at 650 nm and 530 nm. The experimental results showed a good linear relationship within the ENR concentration range of 0-400 nM, with a limit of detection (LOD) of 1.72 nM, which is satisfactory for detection in food safety. Additionally, this method has also been successfully applied to the detection of ENR in tap water, river water, milk, serum and urine, with good recovery rates and RSD values of less than 7%, indicating its great potential for ENR detection in environmental water samples. More importantly, the combination of this method with a smartphone platform provided great convenience for on-site and visual detection of ENR, offering promising applicability prospects.

Abstract:The controllable modulation of the response mode is highly attractive for the construction of photoelectrochemical (PEC) sensors with improved sensitivity and anti-interference ability in complex real samples matrix. Here, we present a charming proof-of-concept ratiometric PEC aptasensor of enrofloxacin (ENR) analysis via the controllable signal transduction. Different with the traditional sensing mechanism, this ratiometric PEC aptasensor integrates the anodic PEC signal induced by PtCuCo nanozyme-catalyzed precipitation reaction and the polarity-switching cathodic PEC response mediated by Cu 2 O nanocubes on S-scheme 2 S 4 heterostructure. Taking advantages of the photocurrent-polarity-switching signal response model and the superior performance of the photoactive substrate material, the proposed ratiometric PEC aptasensor displays a good detection linear range for ENR analysis from 0.01 pg mL −1 to 10 ng mL −1 , with a detection limit of 3.3 fg mL −1 . This study provides a general platform for detecting interested trace analytes in real samples and expands the diversity of sensing strategy design.

Abstract:Enrofloxacin (ENR) is a broad-spectrum fungicide that has been largely applied in pharmacy and animal-specific medicine. In this paper, a simple, novel and highly sensitive molecularly imprinted electrochemiluminescence (MIP-ECL) sensor based on mercaptopropionic acid-functionalized copper nanoclusters (MPA-Cu NCs) was developed to selectively detect enrofloxacin (ENR). MPA-Cu NCs prepared by a one-step method were used to modify the glassy carbon electrode. A molecularly imprinted polymer film containing the cavity was constructed after electropolymerization and elution. Under optimized conditions, the MIP-ECL sensor could detect ENR in the range of 0.1 nM–1 μM (R2 = 0.9863) with a low limit of detection of 27 pM, and the recovery rates of ENR in biological and lake water samples were 88.20–105.0%. The MIP-ECL sensor provided path to improve the stability issues of Cu NCs, which might open promising avenues to develop new ECL systems for biological analysis and environmental water monitoring.

Abstract:A high conductivity composite based on covalent organic frameworks/gold nanoparticles (TAPB-PDA-COFs/AuNPs, TAPB: 3,5-tris(4-aminophenyl)benzene, PDA: p-phthalaldehyde) was prepared by a simple in-situ synthesized method and a novel electrochemical sensor based on TAPB-PDA-COFs/AuNPs was constructed for detection of Enrofloxacin (ENR). A variety of different characterization techniques including ultraviolet‐visible spectrophotometer (UV–vis), fourier transform infrared spectroscopy (FTIR), transmission electron microscopy (TEM) and X-ray photoelectron spectroscopy (XPS) were used to characterize the TAPB-PDA-COFs/AuNPs. ENR was detected by square wave stripping voltammetry (SWV) according to the relationship between the ENR concentration and the oxidation peak current. The result showed that TAPB-PDA-COFs/AuNPs was synthesized successfully. The electrochemical sensor showed two linear ranges in the range of 0.05–10 μmol L−1 and 10–120 μmol L−1 with the limit of detection of 0.041 μmol L−1 (S/N = 3). The good recoveries (96.7–102.2%) and low RSDs (0.9–6.4%) indicated the possibility of using this sensor for actual sample detection. Therefore, TAPB-PDA-COFs/AuNPs-based electrochemical sensor showed good performance in detecting ENR, and would be a potential candidate for the development of fluoroquinolones determination.

Abstract:Recently, up-conversion luminescent (UCL) materials have caught extensive sight on account of their excellent biocompatibility and weak automatic fluorescence background, but the low optical signal makes researchers shy away. Organic dye-sensitized UCL materials can improve the low optical signal drawback of UCL and rejuvenate it with adjustable optical properties and unique antenna effects. In this work, an efficient, simple and selective electrochemiluminescence (ECL) sensing platform was developed for determination of enrofloxacin (ENR). 3,4,9,10-perylene tetracarboxylic acid (PTCA) was successfully used as an "antenna" to improve the ECL performance of the UCL nanoparticles (PEI-NaYF4: Yb, Er) due to its appropriate excitation spectrum position and superior electron transfer rate. The specific recognition function of the aptamer enabled the sensor to eliminate the interference from conspecific impurity. In the presence of ENR, the specific combination of ENR with aptamer made the aptamer fall from surface of the electrode, thus we could see a considerable enhancement of signal. Under the most favourable conditions, the aptasensor based on antenna effect displayed a wide detection range (1.0 × 10-14∼1.0 × 10-6 M), low limit of detection (LOD = 3.0 × 10-15 M) and receivable recoveries (96.0%-102.4%) during water samples analysis. At this point, antenna effect provides a powerful strategy to expand the application of UCL in the field of ECL biosensing.

Abstract:In this project, we developed an electrochemical aptamer sensor based on the coordinated catalysis toward thionine (Thi) of nickel metal-organic framework (Ni-MOF) and gold nanoparticles (AuNPs) to sensitively detect the enrofloxacin (ENR), which belonged to a third-generation quinolone antibiotic for treat diseases caused by bacteria. In order to improve the analytical performance of the sensor, Ni-MOF and gold AuNPs, as the substrate material, could increase the load of the aptamer (Apt) and synergistically catalyze thionine (Thi). Therefore, ultrathin sheets of gold triangular nanosheets (AuTNSs) were prepared as signal labeling materials, which were loaded with a large amount of Thi and complementary deoxyribonucleic acid (cDNA). With the help with Ni-MOF and AuNPs catalysis toward Thi, under optimal conditions, the aptasensor exhibited a massive detection range of 1×10−2 pg·mL−1 to 1×103 pg·mL−1 (S/N = 3) and a detection limit of 5.6×10−3 pg·mL−1 for ENR. Meanwhile, the proposed sensor has a high sensitivity, satisfactory repeatability and long-time stability, and could be well applied to the detection of food samples.

Abstract:In this study, an electrochemical aptasensor was established for the ultrasensitive detection of enrofloxacin (ENR) based on a CoNi-based metal-organic framework (MOF) and gold nanoparticles (AuNPs). Co-MOF, Ni-MOF, and CoNi-MOF were optimized by characterization techniques like scanning electron microscope (SEM), Fourier-transform infrared spectrophotometer (FT-IR), and X-ray diffraction (XRD). The optimal CoNi-MOF has waxberry-like structure, mixed metal valences of, and π-conjugated structure, which show biocompatibility, high hydrophilicity, and good electrochemical stability. In order to enhance the electrochemical performance of the sensor, electrodeposition of AuNPs was used to improve conductivity and increase the load of sulfhydryl group-modified aptamer (Apt) by Au-S bond. The electrochemical behavior was studied by cyclic voltammetry (CV) and electrochemical impedance spectrum (EIS). The Apt/AuNPs/CoNi-MOF/GCE exhibits an extensive detection range of 1×10−3 pg·mL−1 to 1×105 pg·mL−1 (S/N = 3) and a detection limit of 3.3×10−4 pg·mL−1 for ENR under optimal conditions. The arranged sensor demonstrates a high sensitivity, satisfactory reproducibility, and long-time stability. Furthermore, the obtained sensor reveals a high practical prospect for the sensitive detection of ENR in meat samples.

Abstract:The emergence and development of low-cost and high-efficiency nanozymes are promising to replace natural enzymes promoting the application of chemiluminescence immunoassays. Herein, a rapid and highly sensitive flow injection chemiluminescence immunoassay based on cobalt hydroxide (Co(OH)2) nanozyme was established to detect enrofloxacin (ENR) residues in food. In this system, Co(OH)2 nanosheets act as nanozymes to catalyze and amplify the chemiluminescence signal of the luminol-PIP-H2O2 system, as well as a carrier for immobilizing antibodies to form stable immunoprobes. In addition, carboxyl resin beads with good stability and biocompatibility were used as the base of the immunosensor to carry more coating antigens, based on the principle of competitive immunity and to achieve the rapid detection of ENR. Under optimal conditions, the linear working range is 0.0001 ~ 1000 ng/mL, and the limit of detection (LOD) is 0.041 pg/mL (S/N = 3). The method has been successfully applied to the analysis of aquatic products and poultry food.

Abstract:Enrofloxacin, a fluoroquinolone widely used in animal husbandry, presents environmental and human health hazards due to its stability and incomplete hydrolysis leading to residue accumulation. To address this concern, a highly sensitive aptamer biosensor utilizing a localized surface plasmon resonance (LSPR) sensing chip and microfluidic technology was developed for rapid enrofloxacin residue detection. AuNPs were prepared by the seed method and the AuNPs-Apt complexes were immobilized on the chip by the sulfhydryl groups modified on the end of the aptamer. The properties and morphologies of the sensing chip and AuNPs-Apt complexes were characterized by Fourier transform infrared spectroscopy (FTIR), UV-Vis spectrophotometer, and scanning electron microscope (SEM), respectively. The sensing chip was able to detect enrofloxacin in the range of 0.01–100 ng/mL with good linearity, and the relationship between the response of the sensing chip and the concentration was Δλ (nm) = 1.288log ConENR (ng/mL) + 5.245 (R2 = 0.99), with the limit of detection being 0.001 ng/mL. The anti-interference, repeatability, and selectivity of this sensing chip were studied in detail. Compared with other sensors, this novel aptamer biosensor based on AuNPs-Apt complexes is expected to achieve simple, stable, and economical application in the field of enrofloxacin detection.

Abstract:Exploring new functions of nanomaterials can help facilitate the development of biosensors for the detection of antibiotics. Herein, a new detection modality based on monovalent antigen-induced aggregation (MAA) of immunomagnetic beads (IMBs) was proposed for rapid and label-free detection of enrofloxacin (ENR), which endowed IMBs with the abilities of both sample separation and signal generation. In the presence of ENR, the initially well-dispersed IMBs were aggregated and the degree of aggregation was in a concentration-dependent manner. After exploring the mechanism underlying IMB aggregation and investigating the key parameters affecting it, a label-free biosensing platform was developed for rapid and sensitive detection of ENR. Based on the significant differences in the magnetic separation speed and size between the aggregated and well-dispersed IMBs, two methods were proposed for quantitatively determining ENR, i.e., measuring the turbidity of the IMB supernatant after magnetic separation for a given time and visualizing and calculating the grayscale value of the aggregated IMBs trapped on the surface of a nitrocellulose membrane. A three-dimensional (3D)-printed syringe was designed and fabricated for automatic filtration of IMBs. This immunosensor allowed for sensitive detection of ENR in less than 15 min without any labels. It exhibited a satisfactory limit of detection of 0.79 ng mL-1 and showed the feasibility for ENR detection of spiked chicken meat with recovery rates ranging from 74.8 to 98.3%. The MAA immunosensor can act as a promising tool to detect trace levels of ENR and has the potential to be applied to complex food samples.

Abstract:A novel electrochemical sensor for the detection of enrofloxacin (ENR) in aqueous solutions has been developed using a carbon paste electrode modified with a mixture of metal-organic frameworks (MOFs) of CuBTC and FeBTC. These MOFs were successfully synthesized via a solvothermal method and characterized using various techniques, including X-ray diffraction, Fourier-transform infrared spectroscopy, Brunauer–Emmett–Teller analysis, and X-ray photoelectron spectroscopy. The MOF mixture exhibited a particle size ranging from 40 to 100 nm, a high surface area of 1147 m 2 /g, a pore volume of 0.544 cm 3 /g, and a capillary diameter of 1.50 nm. Additionally, energy-dispersive X-ray mapping demonstrated the uniform distribution of the two MOFs within the electrode composition. The synergistic effect of the electrocatalytic properties of CuBTC and the high conductivity of FeBTC significantly enhanced the electrochemical response of ENR, increasing the signal by more than ten times compared to the unmodified electrode. Under optimal analytical conditions, the sensor exhibited three dynamic ranges for ENR detection, that is, 0.005 to 0.100 μM, 0.1 to 1.0 μM, and 1 to 13 μM, with coefficients of determination of 0.9990, 0.9954, and 0.9992, respectively, depending on the accumulation duration. The sensor achieved a low detection limit of 3 nM and demonstrated good reproducibility, with a relative standard deviation of 3.83%. Furthermore, the sensor demonstrated effective performance in analysing tap and lake water samples, with recovery rates ranging from 90.2% to 121.3%. Beilstein J. Herein, a PEC aptasensor was designed and constructed by g-C3N4/Bi24O31Cl10 heterojunction as photoelectric active material for realizing sensitive detecting enrofloxacin (ENR) antibiotic. The formation of heterojunction can facilitate separation and transfer of photogenerated carriers and reduce the recombination rate, leading to the enhancement of photocurrent response compared to that of individual semiconductor. The ENR aptamer was introduced as the specific recognition element to realize excellent selectivity of the PEC aptasensor. Because ENR-aptamer complex can block the transfer of charge, the ENR-PEC aptasensor presented the decreased photocurrent response with the increasing of ENR concentration without external voltage. The developed ENR-PEC aptasensor presented excellent sensitivity and selectivity for detecting ENR with a detection range from 0.5 to 100 fM and a low detection limit of 0.167 fM (S/N = 3). The ENR-PEC aptasensor with good reproducibility may be potential for the determination of the actual samples.

Abstract:A lateral flow immunochromatographic assay (LFIA) based on surface-enhanced Raman scattering (SERS) for sensitive and specific detection of antibiotic enrofloxacin (ENR) in food samples was developed. 1,4-benzenedithiol (BDT) was selected as the Raman reporter, and the BDT mediated-gap AuNR@Au nanoparticles (NPs) were synthesized, characterized and used as the substrate in SERS-LFIA due to the existence of the anisotropic gold nanorods (AuNRs) and the nano-gap with the high SERS enhancement. AuNRs were prepared, then covered by monolayer BDT. Under reduction condition and in presence of HAuCl4, the reduced gold was deposited and grown on AuNRs to form AuNRBDT@Au NPs. As the two thiol groups on para-positions in BDT were respectively linked to AuNR (core) and Au (shell), the gap size inside the NPs was uniform. The immunoprobe (e.g. AuNRBDT@Au-Ab) was obtained by immobilizing Ab against ENR on the surface of AuNRBDT@Au NPs. The performance of SERS-LFIA was similar to that in colloidal gold based-LFIA, and the entire assay time was within 15 min. After LFIA procedures, the specific SERS intensity of BDT at 1560 cm-1 on the test line was measured for the quantitative detection of ENR. The IC50 and limit of detection (LOD) of the LFIA for ENR were 59 pg mL-1 and 0.12 pg mL-1 (e.g. 71 pg g-1 and 0.14 pg g-1 in real sample), respectively. There was no cross-reactivity (CR) of the LFIA with other five antibiotics. The recoveries of ENR from spiked food samples were in range of 89.2%-102.4% with the relative standard deviation (RSD) of 1.70%-6.38%. It was proven that the proposed method was able to simply and rapidly detect ENR in food samples with high sensitivity, specificity, accuracy and precision. The platform can be also an alternative platform for the detection of other target analytes using corresponding Abs.

Congenital disorders in the cattle population of the Czech Republic

Abstract:The aim of the paper was to analyse congenital disorders in the Czech cattle population in 1986-2001. The offspring of 474 sires - 215 Czech Simmental, 236 Holstein, and 23 beef - were diagnosed with congenital disorders which were unevenly distributed because only 18 occurred in the progeny of 10 and more sires, in contrast to 88 occurring in the progeny of 1 sire only. Umbilical hernia was the most frequently noted disorder, and 136 sires fathered progeny with limb anomalies. The most frequent gestational accident was schistosomus reflexus, the results suggesting a familial burden. Three sires fathering offspring with the afflicted spinal column and limbs were heterozygous for Complex Vertebral Malformation (CVM) though they had not been reported as such. Foetal defects and stillbirth were quite frequent, and the calves affected were fathered by 56 sires. In rare disorders with a low incidence, an accurate genetic analysis or even simple discrimination between inherited and acquired defects is problematic. It would be our recommendation that those sires with a higher incidence of defects among their offspring should be disqualified from fathering stock bulls, or culled.

Abstract:Magnetic biosensor takes advantage of rapid and facile magnetic separation/collection of targets, however, generally relies on additional signal labels to generate signal in a tedious and high-cost way. Here, we proposed a chemical and electrochemical conversion (C-ECC) method to develop a label-free electrochemical magnetic biosensor to detect antibiotics enrofloxacin (ENR). The C-ECC method integrates the chemical decomposition of magnetic beads (MBs) to release ironic ions and the simultaneous electrochemical deposition of Prussian blue (PB) analogs through the reaction of ironic ions and co-existing K4Fe(CN)6. Unlike conventional method that relies on the physical magnetic property of MBs, the C-ECC method fully exploited the chemical/electrochemical properties of MBs to produce electrochemically active PB to generate signal, thus endowing MBs with dual roles in both sample treatment and signal generation. The incorporation of chemical and electrochemical conversion produced more PB with higher electroactivity when compared with sole chemical or electrochemical conversion. Moreover, an interesting electrochemical refreshment (ER) was designed to remove insulative species on the electrode surface to improve electroactivity of electrode and benefit amperometric detection significantly. Under optimized conditions, the C-ECC-based biosensor presented limit of detection (LOD) of 4.17 pg mL−1 for ENR, which is lower than most analogs, as well as satisfactory specificity. The biosensor also performed well in fish and chicken meat samples, with LODs lower than maximum residue limits of national standards. The C-ECC method may create a new way to design magnetic sensors and contribute to rapid, facile and sensitive detection in agriculture/food, clinic diagnosis and environmental monitoring.

Abstract:We used mutant Fas-deficient (lpr) or Bim-deficient mice to investigate the role of the death receptor and Bcl-2-regulated apoptotic pathways in terminating a physiological T cell response to herpes simplex virus infection. In WT and lpr mice CD8+ antigen-specific T cells were deleted after viral clearance. In contrast, the immune response was not terminated in Bim-deficient mice despite viral clearance, and CD8+ antigen-specific T cells accumulated in the spleen. Thus, Bim is dispensable for viral clearance but is necessary for the death of activated T cells when immune responses are terminated. These findings have implications for the therapeutic manipulation of immune responses to infections and immunization.
