Abstract:Nonheme iron enzymes stand out as one of the most versatile biocatalysts for molecular functionalization. They facilitate a wide array of chemical transformations within biological processes, including hydroxylation, chlorination, epimerization, desaturation, cyclization, and more. Beyond their native biological functions, these enzymes possess substantial potential as powerful biocatalytic platforms for achieving abiological metal-catalyzed reactions, owing to their functional and structural diversity and high evolvability. To this end, our group has recently engineered a series of nonheme iron enzymes to employ non-natural radical-relay mechanisms for abiological radical transformations not previously known in biology. Notably, we have demonstrated that a nonheme iron enzyme, ( S )-2-hydroxypropylphosphonate epoxidase from Streptomyces viridochromogenes ( Sv HppE), can be repurposed into an efficient and selective biocatalyst for radical fluorine transfer reactions. This marks the first known instance of a redox enzymatic process for C( sp 3 )F bond formation. This chapter outlines the detailed experimental protocol for engineering Sv HPPE for fluorination reactions. Furthermore, the provided protocol could serve as a general guideline that might facilitate other engineering endeavors targeting nonheme iron enzymes for novel catalytic functions.

Abstract:Due to the scarcity of C-F bond-forming enzymatic reactions in nature and the contrasting prevalence of organofluorine moieties in bioactive compounds, developing biocatalytic fluorination reactions represents a pre-eminent challenge in enzymology, biocatalysis and synthetic biology. Additionally, catalytic enantioselective C(sp(3))-H fluorination remains a challenging problem facing synthetic chemists. Although many non-haem iron halogenases have been discovered to promote C(sp(3))-H halogenation reactions, efforts to convert these iron halogenases to fluorinases have remained unsuccessful. Here we report the development of an enantioselective C(sp(3))-H fluorination reaction, catalysed by a plant-derived non-haem enzyme 1-aminocyclopropane-1-carboxylic acid oxidase (ACCO), which is repurposed for radical rebound fluorination. Directed evolution afforded a C(sp(3))-H fluorinating enzyme ACCO(CHF) displaying 200-fold higher activity, substantially improved chemoselectivity and excellent enantioselectivity, converting a range of substrates into enantioenriched organofluorine products. Notably, almost all the beneficial mutations were found to be distal to the iron centre, underscoring the importance of substrate tunnel engineering in non-haem iron biocatalysis. Computational studies reveal that the radical rebound step with the Fe(III)-F intermediate has a low activation barrier of 3.4 kcal mol(-1) and is kinetically facile.

Abstract:Now, Yunfang Yang, Marc Garcia-Borràs, Xiongyi Huang and colleagues report an enantioselective C( sp 3 )–F bond forming reaction by repurposing an enzyme for radical fluorine transfer of benzylic N- fluoroamide substrates. Based on their experience with an analogous C( sp 3 )–H azidation reaction, the researchers started their investigation by screening a library of over 40 non-haem iron enzymes. This led to the identification of ( S )-2-hydroxypropylphosphonate epoxidase from Streptomyces viridochromogenes ( Sv HppE) to catalyse N -fluoroamide-directed C–H fluorination of 1NF to yield 1F in 3.8% yield with a total turnover number (TTN) of 5 and 74:26 enantiomeric ratio (e.r.) (pictured). Computational analysis was used to search for key active-site residues that could potentially affect the binding and positioning of 1NF in the enzyme. The identified residues and the iron-coordinating carboxylate residue E141 were then subjected to site-saturation mutagenesis. Screening efforts afforded the triple mutant S v HppE N134W E141D Y102C (named S v HppE-Fluor), which exhibited a TTN of 50, 30% yield and 96.5:3.5 e.r. for the radical fluorine transfer of 1NF . Reducing the cell concentration improved the catalytic turnovers to 150. Moreover, preparative-scale synthesis at higher cell concentration yielded 101 mg of 1NF with 57% yield and 96:4 e.r. Substrate scope investigations demonstrated that S v HppE-Fluor accepts N -fluoroamide substrates with substitutions on the arene moiety and secondary and tertiary benzylic C–H bonds for fluorination. The authors further investigated the reaction mechanism by computational simulations, which support the proposed enzymatic radical fluorine transfer mechanism to be operative in this reaction. In principle, non-asymmetric iron-catalysed radical fluorine transfer with fluoroamides has been previously reported with simple Fe(OTf) 2 . However, adding this mechanistic strategy to the biocatalytic toolbox provides possibilities for exquisite control of the reaction selectivity by tuning the protein environment while allowing its integration into prospective biosynthetic pathways.

Abstract:The trifluoromethyl (–CF3) group represents a highly prevalent functionality in pharmaceuticals. Over the past few decades, significant advances have been made in the development of synthetic methods for trifluoromethylation. In contrast, there are currently no metal-loenzymes known to catalyze the formation of C(sp3)‒CF3 bonds. In this work, we demonstrate that a nonheme iron enzyme, hydroxy-mandelate synthase from Amycolatopsis orientalis (AoHMS), is capable of generating CF3 radicals from hypervalent iodine(III) reagents and directing them for enantioselective alkene trifluoromethyl azidation. A high-throughput screening (HTS) platform based on Staudinger ligation was established, enabling the rapid evaluation of AoHMS variants for this abiological transformation. The final opti-mized variant accepts a range of alkene substrates, producing the trifluoromethyl azidation products in up to 73% yield and 96:4 enanti-omeric ratio (e.r.). The biocatalytic platform can be further extended to alkene pentafluoroethyl azidation and diazidation by altering the iodine(III) reagent. In addition, anion competition experiments provide insights into the radical rebound process for this abiological transformation. This study not only expands the catalytic repertoire of metalloenzymes for radical transformations but also creates a new enzymatic space for organofluorine synthesis.

Abstract:We report the reprogramming of nonheme iron enzymes to catalyze an abiological C(sp3)‒H azidation reaction through iron-catalyzed radical relay. This biocatalytic transformation uses amidyl radicals as hydrogen atom abstractors and Fe(III)‒N3 intermediates as radical trapping agents. We established a high-throughput screening platform based on click chemistry for rapid evolution of the catalytic performance of identified enzymes. The final optimized variants deliver a range of azidation products with up to 10,600 total turnovers and 93% enantiomeric excess. Given the prevalence of radical relay reactions in organic synthesis and the diversity of nonheme iron enzymes, we envision that this discovery will stimulate future development of metalloenzyme catalysts for synthetically useful transformations unexplored by natural evolution.
Radical-relay C(sp3)-H azidation catalyzed by an engineered nonheme iron enzyme

Abstract:Nonheme iron enzymes are versatile biocatalysts for a broad range of unique and powerful transformations, such as hydroxylation, chlorination, and epimerization as well as cyclization/ring-opening of organic molecules. Beyond their native biological functions, these enzymes are robust for engineering due to their structural diversity and high evolvability. Based on enzyme promiscuity and directed evolution as well as inspired by synthetic organic chemistry, nonheme iron enzymes can be repurposed to catalyze reactions previously only accessible with synthetic catalysts. To this end, our group has engineered a series of nonheme iron enzymes to employ non-natural radical-relay mechanisms for new-to-nature radical transformations. In particular, we have demonstrated that a nonheme iron enzyme, (4-hydroxyphenyl)pyruvate dioxygenase from streptomyces avermitilis (SavHppD), can be repurposed to enable abiological radical-relay process to access C(sp3)-H azidation products. This represents the first known instance of enzymatic radical relay azidation reactions. In this chapter, we describe the detailed experimental protocol to convert promiscuous nonheme iron enzymes into efficient and selective biocatalyst for radical relay azidation reactions. One round of directed evolution is described in detail, which includes the generation and handling of site-saturation mutagenesis, protein expression and whole-cell reactions screening in a 96-well plate. These protocol details might be useful to engineer various nonheme iron enzymes for other applications.

Abstract:Monfluoromethyl compounds meet the criteria for designing and developing sustainable organofluorines without the presence of per- or polyfluorinated fragments. Nevertheless, the asymmetric synthesis of chiral monofluoromethyl compounds under mild conditions remains a demanding task in this field, partially owing to the challenge that β-fluorine elimination is often observed as a competitive side-reaction during metal-catalysed cross-coupling reactions. In this study, we disclose a nickel-catalysed enantioselective reductive cross-coupling reaction of acid chlorides with a newly developed monofluoromethyl building block—brominated fluoroethanol benzoate (BFEB). This transformation enables the preparation of a series of chiral α-monofluoromethyl acyloin derivatives in good yields with high enantioselectivities. Experimental and computational mechanistic studies reveal that the key to the realization of this protocol hinges upon the generation and harnessing of unprecedented nucleophilic monofluoromethyl ketyl-type radical. The undesired β-fluoride elimination process was largely inhibited attributed to the utilization of bulky bisoxazoline ligand.

Abstract:Here, we reported for the first time an iron-catalyzed highly enantioselective hydrogenation of minimally functionalized 1,1-disubstituted alkenes to access chiral alkanes with full conversion and excellent ee. A novel chiral 8-oxazoline iminoquinoline ligand and its iron complex have been designed and synthesized. This protocol is operationally simple by using 1 atm of hydrogen gas and shows good functional group tolerance. A primary mechanism has been proposed by the deuterium-labeling experiments.

Abstract:With an iron catalyst playing dual roles as a radical initiator and terminator, we report a selective remote C-H functionalization to access delta-azido sulfonamides through a radical relay process. The reaction ofN-fluorosulfonamide furnishes the corresponding products in excellent yields with high regioselective control. The key to success is the highly efficient iron-mediated redox azido transfer to thein situgenerated carbon radical. The products provide incentives for drug discovery and ligand designs.

Abstract:Due to the unique effect of fluorine atoms, the efficient construction of high-value alkyl fluorides has attracted significant interest in modern drug development. However, enantioselective catalytic strategies for the efficient assembly of highly functionalized chiral C(sp3)-F scaffolds from simple starting materials have been underutilized. Herein, we demonstrate a nickel-catalyzed radical transfer strategy for the efficient, modular, asymmetric hydrogenation and hydroalkylation of alkenyl fluorides with primary, secondary, and tertiary alkyl halides under mild conditions. The transformation provides facile access to various structurally complex secondary and tertiary α-fluoro amide products from readily available starting materials with excellent substrate compatibility and distinct selectivity. Furthermore, the utility of this method is demonstrated by late-stage modifications and product derivatizations. Detailed mechanistic studies and DFT calculations have been conducted, showing that the rate-determining step for asymmetric hydrogenation reaction is NiH-HAT toward alkenyl fluorides and the stereo-determining step is alcohol coordination to Ni-enolates followed by a barrierless protonation. The mechanism for the asymmetric hydroalkylation reaction is also delivered in this investigation.
Enantioselective Synthesis of Alkyl Fluorides via Biocatalytic Reduction

Abstract:Here we report the first biocatalytic asymmetric synthesis of alkyl fluorides via reduction of α-fluoroenones and α-fluoroenoates using ene reductase enzymes. The reduction of a wide range of (Z) or (E)-α-fluoroenones was shown to proceed in high yield and selectivity using ene reductases, with the different alkene geometries leading to opposite enantiomers of the chiral fluoroalkane. The reaction could also be successfully extended to α-fluoroenoates to access enantioenriched α-fluoroesters with only the E-alkene isomers under-going reduction, enabling mixtures of alkene geometries to be employed. The selectivity and substrate scope were rationalized using in silico substrate-enzyme molecular docking studies.

Abstract:Regio‐ and stereo‐divergent construction of carbon‐fluorine bond remains a challenge in fluorine chemistry, especially reactions involving fluorination at the α‐position of a carbonyl group. Herein, we present a detailed study for predicting the origins of regio‐ and stereo‐selectivity for organocatalyst catalyzed fluorination reaction. Possible mechanisms and origins of selectivities of NHC (N‐heterocyclic carbene)‐catalyzed fluorination reaction of epoxy‐enals were investigated by using DFT (density functional theory) method. The computational results show that the C‐F bond formation step determines stereoselectivity and regioselectivity. LP•••π and C‐H•••F interactions were identified as the main contributors for the stereoselectivity control, in which R‐configured product was preferred. It was found that α‐fluorination is more energetically favorable than γ‐fluorination, which is possibly due to higher reactivity of the α‐carbon; deduced by the local reactivity index analysis. This work would provide valuable insights for predicting origin of the selectivity of fluorination reactions and developing of more effective organocatalyst with high selectivity.

Abstract:We developed an amide-based Cinchona alkaloids catalyzed enantioselective fluorination of 4-substituted pyrazolones, the products with a quaternary chiral fluorine-attached carbon centre were afforded in high yields (up to 99%) with excellent enantioselectivities (up to 98% ee). Our method is highlighted by low catalyst loading (0.05-0.5 mol %), short reaction times (30 min) and broad substrate scopes (35 examples). Furthermore, we carried out this asymmetric fluorination under sustainable conditions in 10 cylces, the optically pure product (>99.5% ee) could be easily obtained by simple extraction and recrystallization on a multigram scale. Controlled experiments and DFT studies showed that the multiple hydrogen bonding interactions and the steric hindrance at ortho-position in benzene ring of catalyst were the main reasons for high enantioselectivity and reactivity. With a simple operational protocol and low catalyst loading, this transformation provides a practical method for preparation of chiral fluorine-containing compounds.

Abstract:Organofluorides have found wide applications in materials science, medicinal chemistry, agrochemistry, and organic synthesis. The unique reactivity and property of fluorine-containing compounds render them worth investigating in asymmetric synthesis to access valuable enantioenriched molecules. Although metal catalysis paves the way for the chemoselective and regioselective defluorination of fluorine-containing molecules, the functionalization of elusive C-F bond integrated into catalytic enantioselective transformation remains highly sought after. In this review, the latest achievements in transition-metal-catalyzed enantioselective defluorinative coupling reactions have been comprehensively summarized on the basis of the classification of transition-metal catalysts. Compared with other heteroatoms, special emphasis is placed on the function of the fluorine atom as a detachable "chemical handle" for the enantioselective transformation of easily accessible fluorine-containing chemicals, such as trifluoromethyl alkenes, allylic fluorides, propargyl gem-difluorides, gem-difluoroalkenes, polyfluoroarenes, dienyl fluorides, and trifluoromethyl (hetero)arenes. We aim to inspire chemists to continue their interest in both research topics of C-F bond activation and asymmetric catalysis.

Abstract:Organofluorine compounds find extensive application in the fields of agrochemicals, pharmaceuticals, materials science, and molecular imaging. Introducing fluorine atoms can provide organic compounds with unique physicochemical properties or improve their biological activity. Although significant progress has been made in the chemical synthesis of fluorine‐containing compounds, achieving selective fluorination under mild conditions remains extremely challenging. Introducing biocatalytic approaches in organofluorine chemistry is an important strategy given their high efficiency, selectivity, and environmental friendliness. In this review, we present the discovery of fluorinated natural products and fluorinases, the crystal structure and the directed evolution of fluorinases, with a focus on recent advances in the enzymatic synthesis of fluorine‐containing compounds in recent years. It is hoped that this review will help to promote the field of biocatalytic organofluorine compound synthesis.

Abstract:We herein report a new catalytic method for intermolecular olefin aminofluorination using earth-abundant iron catalysts and nucleophilic fluoride ion. This method tolerates a broad range of unfunctionalized olefins, especially nonstyrenyl olefins that are incompatible with existing olefin aminofluorination methods. This new iron-catalyzed process directly converts readily available olefins to internal vicinal fluoro carbamates with high regioselectivity (N vs F), many of which are difficult to prepare using known methods. Preliminary mechanistic studies demonstrate that it is possible to exert asymmetric induction using chiral iron catalysts and that both an iron-nitrenoid and carbocation species may be reactive intermediates.

Iodine(I)/iodine(III) catalysis has proven to be particularly well-suited to this task, enabling abundant alkene substrates to be readily intercepted by in situ-generated λ 3 -iodanes and processed to high-value (di)fluorinated products. These organocatalysis paradigms often emulate metal-based processes by engaging the π bond and, in the case of styrenes, facilitating fluorinative phenonium-ion rearrangements to generate difluoromethylene units. Here we demonstrate that enynes are competent proxies for styrenes, thereby mitigating the recurrent need for aryl substituents, and enabling highly versatile homopropargylic difluorides to be generated in an operationally simple manner. The scope of the method is disclosed, together with application in target synthesis (>30 examples, up to >90% yield).
