Abstract:Gold nanomaterials have become very attractive nanomaterials for biomedical research due to their unique physical and chemical properties, including size dependent optical, magnetic and catalytic properties, surface plasmon resonance (SPR), biological affinity and structural suitability. The performance of biosensing and biodiagnosis can be significantly improved in sensitivity, specificity, speed, contrast, resolution and so on by utilizing multiple optical properties of different gold nanostructures. Lateral flow immunochromatographic assay (LFIA) based on gold nanoparticles (GNPs) has the advantages of simple, fast operation, stable technology, and low cost, making it one of the most widely used in vitro diagnostics (IVDs). However, the traditional colloidal gold (CG)-based LFIA can only achieve qualitative or semi-quantitative detection, and its low detection sensitivity cannot meet the current detection needs. Due to the strong dependence of the optical properties of gold nanomaterials on their shape and surface properties, gold-based nanomaterial modification has brought new possibilities to the IVDs: people have attempted to change the morphology and size of gold nanomaterials themselves or hybrid with other elements for application in LFIA. In this paper, many well-designed plasmonic gold nanostructures for further improving the sensitivity and signal output stability of LFIA have been summarized. In addition, some opportunities and challenges that gold-based LFIA may encounter at present or in the future are also mentioned in this paper. In summary, this paper will demonstrate some feasible strategies for the manufacture of potential gold-based nanobiosensors of post of care testing (POCT) for faster detection and more accurate disease diagnosis.

Abstract:Developing a versatile sensing toolkit with high sensitivity may revolutionize current techniques for analytical applications. This work reports a sensing strategy through the electrical conductivity of gold nanoparticles (AuNPs). By modulating the concentration, aggregation, and surface ligand of AuNPs, a versatile analytical platform for highly sensitive detection based on the electrical conductivity is outlined. The difference in concentration, aggregation, and surface ligand enables a response in the conductivity of AuNPs, offering a great platform for developing AuNPs‐implemented assays using the conductivity as the signal readout. Compared with the plasmonic signal readout of AuNPs, this conductivity‐based sensing method has advantages in high sensitivity, low cost, and simple operation, showing great potential as a powerful technique for versatile applications.

Abstract:<p>The development of nanotechnology promotes the progress of analytical methods, especially gold nanomaterials, which have attracted much interest due to their unique optical properties. As they display different colors depending on their size, shape and state of aggregation, it provides excellent platforms for the development of colorimetric sensors in chemical and biological field. For better understanding the significant role of gold nanomaterials in modern science, it becomes increasing necessary to summarize a review on colorimetric application of gold nanomaterials in sensing. In this review, differently, it proceed from four main strategies, centers on different form of gold nanomaterials, and revolves around its development in the chemical and biological field with a wide range of applications. Finally, this review discusses the main disadvantages of the present and the promising prospects of gold nanomaterials as colorimetric sensors. It will be of significant benefit to to further facilitate the design of novel colorimetric sensors.</p>

Abstract:Gold nanoparticles are the most extensively studied nanomaterials for biomedical application due to their unique properties, such as rapid and simple synthesis, large surface area, strong adsorption ability and facile conjugation to various biomolecules. The remarkable photophysical properties of gold nanoparticles have provided plenty of opportunities for the preparation of gold nanoparticles- based optical biosensors, while the excellent biocompatibility, conductivity, catalytic properties and large surface-to-volume ratio have facilitated the application of gold nanoparticles in the construction of electrochemical biosensors. In this review, we mainly detail the gold nanoparticles-based optical and electrochemical biosensors for biomedical application in the recent two years, which have exhibited greatly enhanced analytical performances in the detection of DNA, proteins and some important small molecules.

Abstract:Molecular biology applications based on gold nanotechnology have revolutionary impacts, especially in diagnosing and treating molecular and cellular levels. The combination of plasmonic resonance, biochemistry, and optoelectronic engineering has increased the detection of molecules and the possibility of atoms. These advantages have brought medical research to the cellular level for application potential. Many research groups are working towards this. The superior analytical properties of gold nanoparticles can not only be used as an effective drug screening instrument for gene sequencing in new drug development but also as an essential tool for detecting physiological functions, such as blood glucose, antigen-antibody analysis, etc. The review introduces the principles of biomedical sensing systems, the principles of nanomaterial analysis applied to biomedicine at home and abroad, and the chemical surface modification of various gold nanoparticles.

Abstract:Abstract: Point of Care Diagnostics (POCD) is quintessential in hospitals and the healthcare sector as the secants uplift the quality of medical care and the life of a patient by facilitating quick identification of the underlying pathological condition. Nanotechnology can provide opportunities and has potential in the development of new-age sensing/diagnostic tools. Owing to extraordinary features (e.g., higher density, effective catalysis, good conduction, biocompatibility, inertness, and greater surface-to-volume ratio), gold nanoparticles (AuNPs) are frequently employed in POCT (Point-of-Care-Testing). Gold nanoparticles-based colorimetric methods are widely used in the rapid, sensitive, and selective detection of analytes/target molecules. AuNPs description is critical for their possible utility in prophylaxis, diagnostics, and treatment of an ailment. AuNPs interact with organic/inorganic target molecules to generate colorimetric shift that enables the accurate, precise, and subtle recognition of biologicals (e.g., microorganisms, cellular components, and proteins) and metal ions. This review focused on the need for AuNPs-based colorimetric application in prophylaxis, diagnostics, and treatment in healthcare and reviewed the future outlook of these AuNPs for biological applications. Different synthesis methods of AuNPs, their morphology, and characterization, including their surface functionalization, will be discussed in detail. AuNPs are very much preferable nanomaterials owing to exclusive optical, electrical, and photothermal features. AuNPs-based colorimetric biosensors are simple and possess great utility, yet these offer a robust technique to enable visual, quantitative analysis.

Abstract:Due to their superiority in the simple design and precise targeting, clustered regularly interspaced short palindromic repeats (CRISPR)-Cas systems have attracted significant interest for biosensing. On the one hand, CRISPR-Cas systems have the capacity to precisely recognize and cleave specific DNA and RNA sequences. On the other hand, CRISPR-Cas systems such as orthologs of Cas9, Cas12, and Cas13 exhibit cis-cleavage or trans-cleavage activities after recognizing the target sequence. Owing to the cleavage activities, CRISPR-Cas systems can be designed for biosensing by degrading tagged nucleic acids to produce detectable signals. To meet the requirements of point-of-care detection and versatile signal readouts, gold nanomaterials with excellent properties such as high extinction coefficients, easy surface functionalization, and biocompatibility are implemented in CRISPR-Cas-based biosensors. In combination with gold nanomaterials such as gold nanoparticles, gold nanorods, and gold nanostars, great efforts are devoted to fabricating CRISPR-Cas-based biosensors for the detection of diverse targets. This review focuses on the current advances in gold nanomaterials-implemented CRISPR-Cas-based biosensors, particularly the working mechanism and the performance of these biosensors. CRISPR-Cas systems, including CRISPR-Cas9, CRISPR-Cas12a, and CRISPR-Cas13a are discussed and highlighted. Meanwhile, prospects and challenges are also discussed in the design of biosensing strategies based on gold nanomaterials and CRISPR-Cas systems.

Abstract:The establishment and rapid progress in sensitive biosensing using immunogold silver enhancement has occurred in the past 30 years. Its high sensitivity and simplicity have made immunogold silver enhancement a revolutionary technique for signal amplification in biosensing. This review focuses on the major applications of immunogold silver enhancement, with special emphasis on quantitative biosensing. In this review, the combinations of immunogold silver enhancement with a series of quantitative techniques, such as colorimetry, electrical and electrochemical methods, gravimetry, chemiluminescence, Raman spectroscopy, and inductively coupled plasma–mass spectrometry (ICP-MS), are discussed in detail. Immunogold silver enhancement has become one of the most useful methods in highly sensitive quantitative bioanalysis. The recent development of ICP-MS detection shows great potential in combination with immunogold silver enhancement.

Abstract:In recent years, nanomaterials of different shape, size, and composition have been prepared and characterized, such as gold and silver nanoparticles, quantum dots, mesoporous silica nanoparticles, carbon nanomaterials, and hybrid nanocomposites. Because of their unique physical and chemical properties, these nanomaterials are increasingly used in point-of-care testing (POCT) to improve analytical performance and simplify detection process. They are used either as carriers for immobilizing biorecognition elements, or as labels for signal generation, transduction and amplification. In this commentary, we highlight recent POCT technologies that employ nanotechnology for the analysis of disease biomarkers, including small-molecule metabolites, enzymes, proteins, nucleic acids, cancer cells, and pathogens. Recent advances in lateral flow tests, printable electrochemical biosensors, and microfluidics-based devices are summarized. Existing challenges and future directions are also discussed.

Using gold-based nanomaterials for fighting pathogenic bacteria: from detection to therapy

Abstract:Owing to the unique quantum size effect and surface effect, gold-based nanomaterials (GNMs) are promising for pathogen detection and broad-spectrum antimicrobial activity. This review summarizes recent research on GNMs as sensors for detecting pathogens and as tools for their elimination. Firstly, the need for pathogen detection is briefly introduced with an overview of the physicochemical properties of gold nanomaterials. And then strategies for the application of GNMs in pathogen detection are discussed. Colorimetric, fluorescence, surface-enhanced Raman scattering (SERS) techniques, dark-field microscopy detection and electrochemical methods can enable efficient, sensitive, and specific pathogen detection. The third section describes the antimicrobial applications of GNMs. They can be used for antimicrobial agent delivery and photothermal conversion and can act synergistically with photosensitizers to achieve the precise killing of pathogens. In addition, GNMs are promising for integrated pathogen detection and treatment; for example, combinations of colorimetric or SERS detection with photothermal sterilization have been demonstrated. Finally, future outlooks for the applications of GNMs in pathogen detection and treatment are summarized.

Abstract:Gold Fluorescence Nanoparticles have revealed the great promise in transforming the drug discovery, photodynamic treatment, and cancer detection, among other biological treatments. The integration of these gold nanoparticles into SERS techniques has unlocked a new scope of possibilities, offering enhanced sensitivity, efficient recognition, and specificity in detecting large/small molecular structures. Through their unique properties, gold fluorescence nanoparticles hold tremendous potential to address the crucial challenges in the biomedical field, that promising the significant advancements in diagnosis and treatment methodologies. Nanotechnology has emerged as a pivotal tool in biomedical research, particularly in developing advanced sensing platforms for disease diagnosis and therapeutic monitoring. Since gold nanoparticles are biocompatible and have special optical characteristics, they are excellent choices for surface‐enhanced Raman scattering (SERS) sensing devices. Integrating fluorescence characteristics further enhances their utility in real‐time imaging and tracking within biological systems. The synergistic combination of SERS and fluorescence enables sensitive and selective detection of biomolecules at trace levels, providing a versatile platform for early cancer diagnosis and drug monitoring. In cancer detection, AuNPs facilitate the specific targeting of cancer biomarkers, allowing for early‐stage diagnosis and personalized treatment strategies. The enhanced sensitivity of SERS, coupled with the tunable fluorescence properties of AuNPs, offers a powerful tool for the identification of cancer cells and their microenvironment. This dual‐mode detection not only improves diagnostic accuracy but also enables the monitoring of treatment response and disease progression. In drug detection, integrating AuNPs with SERS provides a robust platform for identifying and quantifying pharmaceutical compounds. The unique spectral fingerprints obtained through SERS enable the discrimination of drug molecules even in complex biological matrices. Furthermore, the fluorescence property of AuNPs makes it easier to track medication distribution in real‐time, maximizing therapeutic effectiveness and reducing adverse effects. Furthermore, the review explores the role of gold fluorescence nanoparticles in photodynamic therapy (PDT). By using the complementary effects of targeted drug release and light‐induced cytotoxicity, SERS‐guided drug delivery and photodynamic therapy (PDT) can increase the effectiveness of treatment against cancer cells. In conclusion, the utilization of gold fluorescence nanoparticles in conjunction with SERS holds tremendous potential for revolutionizing cancer detection, drug analysis, and photodynamic therapy. The dual‐mode capabilities of these nanomaterials provide a multifaceted approach to address the challenges in early diagnosis, treatment monitoring, and personalized medicine, thereby advancing the landscape of biomedical applications.

Abstract:It is widely acknowledged that gold-based materials are of significant interest in the field of biomedicine. Consequently, considerable efforts have been devoted to identifying gold nanoparticles that exhibit effective performance in tumor diagnosis and treatment. However, the underlying reasons for the enhanced efficacy of these gold-based nanomaterials in cancer therapy and diagnosis remain unclear, primarily due to a lack of in-depth understanding of the mechanisms involved. Therefore, it is essential to summarize progress in the field to facilitate the rational design of more efficient nanodevices. In this review, we present recent achievements drawn from the latest research to demonstrate the broad applications of gold-based materials. We begin by illustrating the mechanisms of gold-derived nanoparticles during therapeutic and diagnostic processes, including photothermal therapy, photodynamic therapy, sonodynamic therapy, photoacoustic tomography, fluorescence imaging, and X-ray computed tomography. We then summarize the advancements of gold-based nanomaterials in cancer diagnosis and treatment, while also analyzing the factors contributing to their enhanced performance. Finally, we highlight key descriptors for evaluating efficacy and strategies for designing high-performance nanomaterials. Abstract:Gold nanoclusters (Au NCs) exhibit broad fluorescent spectra from visible to near-infrared regions and good enzyme-mimicking catalytic activities. Combined with excellent stability and exceptional biocompatibility, the Au NCs have been widely exploited in biomedicine such as biocatalysis and bioimaging. Especially, the long fluorescence lifetime and large Stokes shift attribute Au NCs to good probes for fluorescence sensing and biological detection. In this review, we systematically summarized the molecular structure and fluorescence properties of Au NCs and highlighted the advances in fluorescence sensing and biological detection. The Au NCs display high sensitivity and specificity in detecting iodine ions, metal ions, and reactive oxygen species, as well as certain diseases based on the fluorescence activities of Au NCs. We also proposed several points to improve the practicability and accelerate the clinical translation of the Au NCs.

Abstract:With the rapid development of materials technology, fluorescent gold nanoclusters (AuNCs) are emerging as novel functional materials for diagnostic applications including the detection of biomarkers and bioimaging due to the advantages of their ultra-small size, tunable emissions, size-dependent fluorescence and excellent biocompatibility. In this review, we introduced the synthetic methods, and physical and chemical properties of AuNCs. Subsequently, we described the AuNCs-based design strategies for the detection of biomarkers including small molecules, DNA and proteins. The applications of AuNCs for tumor imaging in vitro and in vivo were also presented. Finally, we discussed the challenges and potential solutions of AuNCs-based nanosensors.

Abstract:Gold nanoparticles (Au NPs) provide a unique platform for biomedical applications. Au NP-based sensors have been widely employed to detect many different chemicals and disease-related biomolecules. Recent research on drug delivery and therapy with Au NPs has also indicated attractive and promising prospects for future applications. In this invited review we will provide an overview of the use of Au NPs for diagnostic sensing and therapy applications.

Abstract:Breast cancer (BC) is one of the most common malignant tumors in women worldwide, and its incidence is increasing every year. Early diagnosis and treatment are critical to improve the curability and prognosis of patients. However, existing detection methods often suffer from insufficient sensitivity and specificity, which limits their clinical application. Fortunately, the rapid development of nanotechnology offers new possibilities for diagnosing BC. For example, the unique physicochemical properties of gold nanomaterials (Au NMs), such as fascinating optical properties and quantum size effect, along with excellent biocompatibility and modifiability, enable them to manifest great potential in the field of biosensing, especially in the detection of BC biomarkers. Through fine surface modification and functionalization, Au NMs can accurately bind to specific antibodies, nucleic acids, and other biomolecules, thus achieving sensitive and precise detection of specific biomarkers. Here, we focus on the research progress of Au NMs as a key biosensing vector in BC biomarker detection. From four major perspectives of early diagnosis, prognostic evaluation, risk prediction, and bioimaging applications, we have thoroughly analyzed the broad application of Au NMs in BC biomarker detection and prospectively addressed its possible future trends. We hope this review will provide more comprehensive ideas for future researchers and promote the further development of this field.Graphical Abstract

Abstract:Ensuring safe high-quality food is an ongoing priority, yet consumers face heightened risk from foodborne pathogens due to extended supply chains and climate change in the food industry. Nanomaterial-based assays are popular and have recently been developed to ensure food safety and high quality. This review discusses strategies for utilizing gold nanoparticles in colorimetric biosensors. The visible-signal biosensor proves to be a potent sensing technique for directly measuring targets related to foodborne pathogens in the field of food analysis. Among visible-signal biosensors, the localized surface plasmon resonance (LSPR) biosensor has garnered increasing attention and experienced rapid development in recent years. This review succinctly introduces the origin of LSPR theory, providing detailed insights into its fundamental principles. Additionally, this review delves into the application of nanotechnology for the implementation of the LSPR biosensor, exploring methods for utilizing gold nanoparticles and elucidating the factors that influence the generation of visible signals. Several emerging technologies aimed at simple and rapid immunoassays for onsite applications have been introduced in the food industry. In the foreseeable future, field-friendly colorimetric biosensors could be adopted in food monitoring systems. The onsite and real-time detection of possible contaminants and biological substances in food and water is essential to ensure human health and safety.
