Abstract:Binding site interactions of nicotinic insecticides have been resolved by comparative chemical and structural neurobiology approaches using mollusk acetylcholine-binding proteins which serve as structural surrogates of the extracellular ligand-binding domain of the insect and vertebrate nicotinic acetylcholine receptor (nAChR) subtypes. Neonicotinoids with an electronegative pharmacophore are embraced by a reversed position in the binding pocket compared with a nicotinoid cationic functionality. A single dominant binding orientation causes the high affinity for neonicotinoids at the insect nAChR homologue. However, the inferior potency of neonicotinoids at the vertebrate nAChR model is attributable to multiple binding conformations conflicting in the agonist-binding pocket. These findings in molecular recognition conferring potency and selectivity facilitated illustrative studies on nAChR structure-guided insecticide design, yielding outstandingly potent and selective candidate compounds with novel pharmacophores.

Abstract:Insect nicotinic acetylcholine receptors (nAChRs) are pentameric ligand-gated ion channels mainly expressed in the central nervous system of insects. They are the directed targets of many insecticides, including neonicotinoids, which are the most widely used insecticides in the world. However, the development of resistance in pests and the negative impacts on bee pollinators affect the application of insecticides and have created a demand for alternatives. Thus, it is very important to understand the mode of action of these insecticides, which is not fully understood at the molecular level. In this study, we systematically examined the susceptibility of ten Drosophila melanogaster nAChR subunit mutants to eleven insecticides acting on nAChRs. Our results showed that there are several subtypes of nAChRs with distinct subunit compositions that are responsible for the toxicity of different insecticides. At least three of them are the major molecular targets of seven structurally similar neonicotinoids in vivo. Moreover, spinosyns may act exclusively on the α6 homomeric pentamers but not any other nAChRs. Behavioral assays using thermogenetic tools further confirmed the bioassay results and supported the idea that receptor activation rather than inhibition leads to the insecticidal effects of neonicotinoids. The present findings reveal native nAChR subunit interactions with various insecticides and have important implications for the management of resistance and the development of novel insecticides targeting these important ion channels.

Abstract:Acetylcholinesterase (AChE; EC 3.1.1.7) is a primary target of many insecticides including organophosphates (OP) and carbamates (CB). Because AChE is expressed in all invertebrate and vertebrate animals as a key enzyme of the cholinergic system, the toxicity of anticholinesterase insecticides to mammals and non-target species such as beneficial insects has been a great concern. In addition, the intensive use of OP and CB insecticides has resulted in the development of resistance in many insect pests, which has limited the use of anticholinesterase insecticides. Many aces encoding AChEs have been sequenced from a variety of vertebrates, insects and other invertebrates, and crystal structures of four AChEs have been determined in the past 20 years. Although the primary motifs and the three dimensional (3D) structures of different AChEs are similar, differences among AChEs are obvious. The catalytic properties and inhibition kinetics of AChEs from different groups of insects and mammals may be quite different, and two AChEs from a single insect may also show distinct differences. These differences may provide new opportunities for designing more selective insecticides for pest management.

Abstract:A series of ethers containing pyrrolidine and/or pyridine bioisosteres was synthesized and evaluated as nicotinic ligands. The dimethylaminoethoxypridines 6 and 7 inhibited the specific bind of (-)-[3H]Nicotine with Ki values of 300 nM and 450 nM, respectively. Compounds 8 and 9 were found to have Ki values of 3390 nM and 360 nM. These results suggest that dialkylamino and appropriately substituted benzene rings (NO2, 8; OH, 9) are bioisosteric replacements for pyrrolidine and pyridine, respectively.

Abstract:Neonicotinoid agonists with a nitroimino pharmacophore are used worldwide for crop protection and animal health care. Chemical and structural biology investigations on the nicotinic acetylcholine receptor structure in the neonicotinoid-bound state revealed a unique niche beyond the nitro oxygen tip toward the loop D subsite. The nitroimino pharmacophore can be replaced to suitably fit the newly recognized cavity by acylimino [═NC(O)R] and phenoxycarbonylmino [═NC(O)OPh] variants. The ═NC(O)R analogues, where R is a hydrogen acceptor pyridine, pyrazine, or trifluoromethyl, showed high receptor potency, suggesting that the extended pharmacophore undergoes hydrogen bonding with the loop D Arg basic residue. The ═NC(O)OPh analogues had appreciably higher affinity with an electron-donating substituent on the phenyl ring than with an electron-withdrawing group, predicting that the benzene plane and loop D Trp indole form a face-to-edge aromatic interaction. These studies illustrate strategic ligand design combining the chemorational approach with the three-dimensional receptor structure.

Abstract:We detail here distinctive departures from lead classical cholinesterase re-activators, the pyridinium aldoximes, to achieve rapid CNS penetration and reactivation of AChE in the CNS (brain and spinal cord). Such reactivation is consistent with these non-canonical re-activators enhancing survival parameters in both mice and macaques following exposure to organophosphates. Thus, the ideal cholinesterase re-activator should show minimal toxicity, limited inhibitory activity in the absence of an organophosphate, and rapid CNS penetration, in addition to its nucleophilic potential at the target, the conjugated AChE active center. These are structural properties directed to reactivity profiles at the conjugated AChE active center, reinforced by the pharmacokinetic and tissue disposition properties of the re-activator leads. In the case of nicotinic acetylcholine receptor (nAChR) agonists and antagonists, with the many existing receptor subtypes in mammals, we prioritize subtype selectivity in their design. In contrast to nicotine and its analogues that react with panoply of AChR subtypes, the substituted di-2-picolyl amine pyrimidines possess distinctive ionization characteristics reflecting in selectivity for the orthosteric site at the α7 subtypes of receptor. Here, entry to the CNS should be prioritized for the therapeutic objectives of the nicotinic agent influencing aberrant CNS activity in development or in the sequence of CNS ageing (longevity) in mammals, along with general peripheral activities controlling inflammation.
Toxins for decoding interface selectivity in nicotinic acetylcholine receptors

Abstract:Nicotinic acetylcholine receptors (nAChRs) are pentameric ligand-gated ion channels that play crucial roles in neurotransmission and regulate complex processes in brain functions, including anxiety, learning and memory, food intake, drug addiction, cognition and nociception. To perform these and other functions, a diverse array of nAChR subtypes are generated by homomeric or heteromeric assembly of 17 homologous nAChR subunits. Agonists, acetylcholine and nicotine, bind to the interface formed between two α subunits and between α and non-α subunits to activate the nAChR and allow cation influx. The diversity of subunit interfaces determines the channel properties, the responses to different agonists/antagonists, desensitization and downstream signaling and thus, define specialized properties and functions. Over the last several decades, snake venom neurotoxins have contributed to the purification, localization and characterization of molecular details of various nAChRs. Utkin et al. have described the purification and characterization of αδ-bungarotoxins, a novel class of neurotoxins in a recent paper published in the Biochemical Journal [Biochem. J. (2019) 476, 1285-1302]. These toxins from Bungarus candidus venom preferably bind to α-δ site with two orders of magnitude higher affinity compared with α-γ or α-ε sites. The subtle changes in the structure of αδ-bungarotoxins led to variation in interface selectivity. Such new classes of antagonists will offer us great opportunity to delineate the pharmacophores and design new highly selective antagonists. Thus, their findings provide new impetus to re-evaluate molecular details of pharmacological properties of α-neurotoxins with careful consideration towards subtype-, interface- and species-selectivity.

Abstract:The nicotinic acetylcholine receptor (nAChR) is a ligand gated ion channel protein, composed of three domains: a transmembrane domain (TM-domain), extracellular domain (EC-domain), and intracellular domain (IC-domain). Due to its biological importance, much experimental and theoretical research has been carried out to explore its mechanisms of gating and selectivity, but there are still many unresolved issues, especially on the ion selectivity. Moreover, most of the previous theoretical work has concentrated on the TM-domain or EC-domain of nAChR, which may be insufficient to understand the entire structure–function relation. In this work, we perform molecular dynamics, Brownian dynamics simulations and continuum electrostatic calculations to investigate the role of different nAChR domains in ion conduction and selectivity. The results show that although both the EC and IC domains contain strong negative charges that create large cation concentrations at either end of the pore, this alone is not sufficient to create the observed cation selectivity and may play a greater role in determining the channel conductance. The presence of cations in the wide regions of the pore can screen out the protein charge allowing anions to enter, meaning that local regions of the TM-domain are most likely responsible for discriminating between ions. These new results complement our understanding about the ion conduction and selectivity mechanism of nAChR.
The Cyclic Imine Core Common to the Marine Macrocyclic Toxins Is Sufficient to Dictate Nicotinic Acetylcholine Receptor Antagonism

Abstract:Macrocyclic imine phycotoxins are an emerging class of chemical compounds associated with harmful algal blooms and shellfish toxicity. Earlier binding and electrophysiology experiments on nAChR subtypes and their soluble AChBP surrogates evidenced common trends for substantial antagonism, binding affinities, and receptor-subtype selectivity. Earlier, complementary crystal structures of AChBP complexes showed that common determinants within the binding nest at each subunit interface confer high-affinity toxin binding, while distinctive determinants from the flexible loop C, and either capping the nest or extending toward peripheral subsites, dictate broad versus narrow receptor subtype selectivity. From these data, small spiroimine enantiomers mimicking the functional core motif of phycotoxins were chemically synthesized and characterized. Voltage-clamp analyses involving three nAChR subtypes revealed preserved antagonism for both enantiomers, despite lower subtype specificity and binding affinities associated with faster reversibility compared with their macrocyclic relatives. Binding and structural analyses involving two AChBPs pointed to modest affinities and positional variability of the spiroimines, along with a range of AChBP loop-C conformations denoting a prevalence of antagonistic properties. These data highlight the major contribution of the spiroimine core to binding within the nAChR nest and confirm the need for an extended interaction network as established by the macrocyclic toxins to define high affinities and marked subtype specificity. This study identifies a minimal set of functional pharmacophores and binding determinants as templates for designing new antagonists targeting disease-associated nAChR subtypes.

Abstract:The nicotinic acetylcholine receptor (nAChR) and other pentameric ligand-gated ion channels are native to neuronal membranes with an unusual lipid composition. While it is well-established that these receptors can be significantly modulated by lipids, the underlying mechanisms have been primarily studied in model membranes with few lipid species. Here, we use coarse-grained molecular dynamics simulation to probe specific binding of lipids in a complex quasi-neuronal membrane. We ran a total of 50 <i>μ</i>s of simulations of a single nAChR in a membrane composed of 36 species of lipids. Competition between multiple lipid species produces a complex distribution. We find that overall, cholesterol selects for concave inter-subunit sites and polyunsaturated fatty acids select for convex M4 sites, while monounsaturated and saturated lipids are unenriched in the nAChR boundary. We propose the "density-threshold affinity" as a metric calculated from continuous density distributions, which reduces to a standard affinity in two-state binding. We find that the density-threshold affinity for M4 weakens with chain rigidity, which suggests that flexible chains may help relax packing defects caused by the conical protein shape. For any site, PE headgroups have the strongest affinity of all phospholipid headgroups, but anionic lipids still yield moderately high affinities for the M4 sites as expected. We observe cooperative effects between anionic headgroups and saturated chains at the M4 site in the inner leaflet. We also analyze affinities for individual anionic headgroups. When combined, these insights may reconcile several apparently contradictory experiments on the role of anionic phospholipids in modulating nAChR.

Abstract:The biosynthesis of sex pheromone components in many lepidopteran insects is regulated by the interaction between pheromone biosynthesis-activating neuropeptide (PBAN) and the PBAN receptor (PBANR), a class A G-protein-coupled receptor. To identify functionally important amino acid residues in the silkmoth PBANR, a series of 27 alanine substitutions was generated using a PBANR chimera C-terminally fused with enhanced GFP. The PBANR mutants were expressed in Sf9 insect cells, and their ability to bind and be activated by a core PBAN fragment (C10PBAN(R2K)) was monitored. Among the 27 mutants, 23 localized to the cell surface of transfected Sf9 cells, whereas the other four remained intracellular. Reduced binding relative to wild type was observed with 17 mutants, and decreased Ca(2+) mobilization responses were observed with 12 mutants. Ala substitution of Glu-95, Glu-120, Asn-124, Val-195, Phe-276, Trp-280, Phe-283, Arg-287, Tyr-307, Thr-311, and Phe-319 affected both binding and Ca(2+) mobilization. The most pronounced effects were observed with the E120A mutation. A molecular model of PBANR indicated that the functionally important PBANR residues map to the 2nd, 3rd, 6th, and 7th transmembrane helices, implying that the same general region of class A G-protein-coupled receptors recognizes both peptidic and nonpeptidic ligands. Docking simulations suggest similar ligand-receptor recognition interactions for PBAN-PBANR and the orthologous vertebrate pair, neuromedin U (NMU) and NMU receptor (NMUR). The simulations highlight the importance of two glutamate residues, Glu-95 and Glu-120, in silkmoth PBANR and Glu-117 and Glu-142 in human NMUR1, in the recognition of the most functionally critical region of the ligands, the C-terminal residue and amide.
Identification of the potential function-specific sites in subunits of vertebrate neuronal nicotinic acetylcholine receptors

Abstract:The nicotinic acetylcholine receptors (nAChR) are complexes comprising of subunits evolved from common ancestor. Although the subunits are similar in sequences and structures, their molecular function varies significantly. Therefore, detecting the molecular sites specific to each subunit is important to understand the property of the subunits and the receptors formed by them. As we know, the molecular sites critical to the structure and molecular function of a protein family usually are conserved in evolution, and those specific to each member of the family are often closely related to its structural and functional specificity. In this study, we analyzed the sequence specific sites in nAChR subunits by adopting the evolutionary trace method and the two-state model, and explored the relationship between structure and function in combination with the spatial location of the sites. The results showed that the detected sites in α7 subunit were closely related to ligand binding and conformational changes of the energetic coupling pathway. The conserved sites tended to be distributed in the interior of the spatial structure of protein molecules, and the sites potentially related to new functions were distributed on the surface of the spatial structure. In summary, our results could be helpful to understanding the molecular features related to the function specificity and diversity of the nAChR subunits.

Abstract:The alpha 3 beta 2 and alpha 3 beta 4 nicotinic acetylcholine receptors (nAChRs) are widely expressed in the central and peripheral nervous systems, playing critical roles in various physiological processes and in such pathologies as addiction to nicotine and other drugs of abuse. alpha-Conotoxin LvIA, which we previously isolated from Conus lividus, modestly discriminates alpha 3 beta 2 and alpha 3 beta 4 rat nAChRs exhibiting a similar to 17-fold tighter binding to the former. Here, alanine scanning resulted in two more selective analogues [N9A]LvIA and [D11A]LvIA, the former having a >2000-fold higher selectivity for alpha 3 beta 2. The determined crystal structures of [N9A]LvIA and [D11A]LvIA bound to the acetylcholine-binding protein (AChBP) were followed by homologous modeling of the complexes with the alpha 3 beta 2 and alpha 3 beta 4 nAChRs and by receptor mutagenesis, which revealed Phe106, Ser108, Ser113, and Ser168 residues in the beta 2 subunit as essential for LvIA binding. These results may be useful for the design of novel compounds of therapeutic potential targeting alpha 3 beta 2 nAChRs.
Key role of the TM2-TM3 loop in calcium potentiation of the α9α10 nicotinic acetylcholine receptor

Abstract:The α9α10 nicotinic cholinergic receptor (nAChR) is a ligand-gated pentameric cation-permeable ion channel that mediates synaptic transmission between descending efferent neurons and mechanosensory inner ear hair cells. When expressed in heterologous systems, α9 and α10 subunits can assemble into functional homomeric α9 and heteromeric α9α10 receptors. One of the differential properties between these nAChRs is the modulation of their ACh-evoked responses by extracellular calcium (Ca2+). While α9 nAChRs responses are blocked by Ca2+, ACh-evoked currents through α9α10 nAChRs are potentiated by Ca2+ in the micromolar range and blocked at millimolar concentrations. Using chimeric and mutant subunits, together with electrophysiological recordings under two-electrode voltage-clamp, we show that the TM2-TM3 loop of the rat α10 subunit contains key structural determinants responsible for the potentiation of the α9α10 nAChR by extracellular Ca2+. Moreover, molecular dynamics simulations reveal that the TM2-TM3 loop of α10 does not contribute to the Ca2+ potentiation phenotype through the formation of novel Ca2+ binding sites not present in the α9 receptor. These results suggest that the TM2-TM3 loop of α10 might act as a control element that facilitates the intramolecular rearrangements that follow ACh-evoked α9α10 nAChRs gating in response to local and transient changes of extracellular Ca2+ concentration. This finding might pave the way for the future rational design of drugs that target α9α10 nAChRs as otoprotectants.
Evolutionary Relationship Of Nicotinic Acetylcholine Receptor Subunits In Both Vertebrate And Invertebrate Species

Abstract:A significant number of subunits for nicotinic acetylcholine receptor (nAChRs) have been identifi ed in humans and other species. However, the evolutionary relationship and biological functions for most of these subunits are largely unknown. The purposes of this chapter were (1) to infer molecular evolutionary history and divergence times of nAChRs, (2) to identify possible essential amino acid residues for the complementary component of the acetylcholine binding site, and (3) to predict the combinational roles of functionally unassigned nicotinic receptor subunits. A total of 123 nucleotide sequences from 23 species, retrieved from public databases, were aligned and a set of phylogenetic trees was generated using different algorithms. Our results indicate that homooligomer-forming subunits (alpha 7-alpha 10) diverged before the split event between vertebrata and invertebrata. Following this divergence, other a and non-alpha subunits evolved within each lineage independently, suggesting a convergence in the evolution of nAChR subunits. In the invertebrate lineage, this gene duplication event seems to have occurred not long before splitting between nematoda and insecta. Furthermore, we suggest that asparagine at position 4 (N4) in loop E may be essential for the complementary component of the acetylcholine binding site upon examination of the amino acid residues in multiple sequence alignments, which correspond to complementary loops. Finally, the combinational roles for several unverified nAChR subunits are predicted using the information on the conserved residues of the acetylcholine binding site and our generalized quaternary organizational model.

Abstract:Measurements of nitrous acid (HONO) and nitrogen dioxide (NO2) in Beijing City have been performed by means of a developed differential optical absorption spectroscopy (DOAS) system based on photodiode array (PDA), during the autumn of 2004. HONO and NO2 were simultaneously identified by their characteristic absorption bands in the spectral region between 337 nm and 372 nm with high sensibility and time resolution. The concentrations of HONO exhibit obviously diurnal variation with a nocturnal maximum and a daytime minimum. The highest HONO value up to 11.8 microg/m3 was observed during the night of 2/3 September. Possible sources of the observed HONO were discussed. Good correlation to NO2 indicates that NO2 is a main source component. The measurement also shows direct emission of HONO is an important source in strongly polluted urban area.

Abstract:Abstract The ability of the Torpedo nicotinic acetylcholine receptor (nAChR) to undergo agonist-induced conformational transitions requires the presence of cholesterol and/or anionic lipids. Here we use recently solved structures along with multiscale molecular dynamics simulations to examine lipid binding to the nAChR in bilayers that have defined effects on nAChR function. We examine how phosphatidic acid and cholesterol, lipids that support conformational transitions, individually compete for binding with phosphatidylcholine, a lipid that does not. We also examine how the two lipids work synergistically to stabilize an agonist-responsive nAChR. We identify rapidly exchanging lipid binding sites, including both phospholipid sites with a high affinity for phosphatidic acid and promiscuous cholesterol binding sites in the grooves between adjacent transmembrane α-helices. A high affinity cholesterol site is confirmed in the inner leaflet framed by a key tryptophan residue on the MX α-helix. Our data provide insight into the dynamic nature of lipid-nAChR interactions and set the stage for a detailed understanding of the mechanisms by which lipids facilitate nAChR function at the neuromuscular junction.

Abstract:Nicotinic acetylcholine receptors (nAChRs) are critical ligand-gated ion channels in the human nervous system. They are targets for various neurotoxins produced by algae, plants, and animals. While there have been many structures of nAChRs bound by neurotoxins published, the binding mechanism of toxins to the nAChRs remains uncleared. In this work, we have performed extensive Gaussian accelerated molecular dynamics simulations on several Aplysia californica (AC) nAChRs in complex with α-conotoxins, strychnine, and pinnatoxins, as well as human nAChRs in complex with α-bungarotoxin and α-conotoxin for a total of 60 μs of simulation time to determine the binding and dissociation pathways of the toxins to the nAChRs and the associated effects. We uncovered two common binding and dissociation pathways shared by toxins and nAChRs. In the primary binding pathway, the toxins diffused from the bulk solvent to first bind a region near the extracellular pore before moving downwards along the nAChRs to the nAChR orthosteric pocket. The second binding pathway involved a direct diffusion of the toxins from the bulk solvent into the nAChR orthosteric pocket. The dissociation pathways were the reverse of the observed binding pathways. We also found that the toxins enacted their toxicity upon binding by restricting the necessary movements required by the nAChRs to open their extracellular and intracellular pores for the ions to pass through. Notably, the electrostatically bipolar interactions between nAChR orthosteric pocket and toxins provides a molecular level explanation for the common binding mode shared by diverse toxins and serve as a key determinant for toxicity.
