Abstract:This study has identified the active cerebral areas of normal Chinese that are associated with Chinese semantic processing using functional brain imaging. According to the traditional cognitive theory, semantic processing is not particularly associated with or affected by input modality. The functional brain imaging experiments were conducted to identify the common active areas of two modalities when subjects perform Chinese semantic tasks through reading and listening respectively. The result has shown that the common active areas include left inferior frontal gyrus (BA 44/45), left posterior inferior temporal gyrus (BA37); the joint area of inferior parietal lobules (BA40) and superior temporal gyrus, the ventral occipital areas and cerebella of both hemispheres. It gives important clue to further discerning the roles of different cerebral areas in Chinese semantic processing.
The Nature and Neural Correlates of Semantic Association versus Conceptual Similarity

Abstract:The ability to represent concepts and the relationships between them is critical to human cognition. How does the brain code relationships between items that share basic conceptual properties (e.g., dog and wolf) while simultaneously representing associative links between dissimilar items that co-occur in particular contexts (e.g., dog and bone)? To clarify the neural bases of these semantic components in neurologically intact participants, both types of semantic relationship were investigated in an fMRI study optimized for anterior temporal lobe (ATL) coverage. The clear principal finding was that the same core semantic network (ATL, superior temporal sulcus, ventral prefrontal cortex) was equivalently engaged when participants made semantic judgments on the basis of association or conceptual similarity. Direct comparisons revealed small, weaker differences for conceptual similarity > associative decisions (e.g., inferior prefrontal cortex) and associative > conceptual similarity (e.g., ventral parietal cortex) which appear to reflect graded differences in task difficulty. Indeed, once reaction time was entered as a covariate into the analysis, no associative versus category differences remained. The paper concludes with a discussion of how categorical/feature-based and associative relationships might be represented within a single, unified semantic system.

Distinct Roles for the Anterior Temporal Lobe and Angular Gyrus in the Spatiotemporal Cortical Semantic Network

Abstract:Abstract Semantic knowledge is supported by numerous brain regions, but the spatiotemporal configuration of the network that links these areas remains an open question. The hub-and-spokes model posits that a central semantic hub coordinates this network. In this study, we explored distinct aspects that define a semantic hub, as reflected in the spatiotemporal modulation of neural activity and connectivity by semantic variables, from the earliest stages of semantic processing. We used source-reconstructed electro/magnetoencephalography, and investigated the concreteness contrast across three tasks. In a whole-cortex analysis, the left anterior temporal lobe (ATL) was the only area that showed modulation of evoked brain activity from 100 ms post-stimulus. Furthermore, using Dynamic Causal Modeling of the evoked responses, we investigated effective connectivity amongst the candidate semantic hub regions, that is, left ATL, supramarginal/angular gyrus (SMG/AG), middle temporal gyrus, and inferior frontal gyrus. We found that models with a single semantic hub showed the highest Bayesian evidence, and the hub region was found to change from ATL (within 250 ms) to SMG/AG (within 450 ms) over time. Our results support a single semantic hub view, with ATL showing sustained modulation of neural activity by semantics, and both ATL and AG underlying connectivity depending on the stage of semantic processing.

Characterizing Differences in Functional Connectivity Between Posterior Cortical Atrophy and Semantic Dementia by Seed-Based Approach.

Abstract:Background:Posterior cortical atrophy (PCA) and semantic dementia (SD) are focal syndromes involving different cerebral regions. This study aimed to demonstrate the existence of abnormal functional connectivity (FC) with an affected network in PCA and SD.Methods:A total of 10 patients with PCA, 12 patients with SD, and 11 controls were recruited to undergo a detailed clinical history interview and physical examination, neuropsychological assessments, and PET/MRI scan. Seed-based FC analyses were conducted to construct FC in language network, visual network, and salience network. The two-sample t-test was performed to reveal distinct FC patterns in PCA and SD, and we further related the FC difference to cognition. Meanwhile, the uptake value of fluorodeoxyglucose in regions with FC alteration was also extracted for comparison.Results:We found a global cognitive impairment in patients with PCA and SD. The results of FC analyses showed that patients with PCA present decreased FC in left precentral gyrus to left V1 and increased FC in right inferior frontal gyrus to right V1 in the visual network, right medial frontal gyrus and left fusiform to left anterior temporal lobe and post-superior temporal gyrus in the language network, and left superior temporal gyrus to left anterior insula in the salience network, which were related to cognitive function. Patients with SD had decreased FC from right superior frontal gyrus, right middle frontal gyrus and right superior frontal gyrus to left anterior temporal lobe, or post-superior temporal gyrus in the language network, as well as left superior frontal gyrus to right anterior insula in the salience network, positively relating to cognitive function, but increased FC in the right superior temporal gyrus to left anterior temporal lobe in the language network, and right insula and left anterior cingulum to right anterior insula in the salience network, negatively relating to cognitive function. Most of the regions with FC change in patients with PCA and SD had abnormal metabolism simultaneously.Conclusion:Abnormal connectivity spread over the cortex involving language and salience networks was common in patients with PCA and SD, whereas FC change involving the visual network was unique to patients with PCA. The FC changes were matched for cognitive deficits.

Abstract:Abstract Studies in semantics traditionally focus on knowledge of objects. By contrast, less is known about how objects relate to each other. In an fMRI study, we tested the hypothesis that the neural processing of categorical spatial relations between objects is distinct from the processing of the identity of objects. Attending to the categorical spatial relations compared with attending to the identity of objects resulted in greater activity in superior and inferior parietal cortices (especially on the left) and posterior middle frontal cortices bilaterally. In an accompanying lesion study, we tested the hypothesis that comparable areas would be necessary to represent categorical spatial relations and that the hemispheres differ in their biases to process categorical or coordinate spatial relations. Voxel-based lesion symptom mapping results were consistent with the fMRI observations. Damage to a network comprising left inferior frontal, supramarginal, and angular gyri resulted in behavioral impairment on categorical spatial judgments. Homologous right brain damage also produced such deficits, albeit less severely. The reverse pattern was observed for coordinate spatial processing. Right brain damage to the middle temporal gyrus produced more severe deficits than left hemisphere damage. Additional analyses suggested that some areas process both kinds of spatial relations conjointly and others distinctly. The left angular and inferior frontal gyrus processes coordinate spatial information over and above the categorical processing. The anterior superior temporal gyrus appears to process categorical spatial information uniquely. No areas within the right hemisphere processed categorical spatial information uniquely. Taken together, these findings suggest that the functional neuroanatomy of categorical and coordinate processing is more nuanced than implied by a simple hemispheric dichotomy.

Functional integration and segregation during semantic cognition: Evidence across age groups

Abstract:Semantic cognition is underpinned by ventral anterior temporal lobe (vATL) which encodes knowledge representations and inferior frontal gyrus (IFG), which controls activation of knowledge based on the needs of the current context. This core semantic network has been validated in substantial empirical findings in the past. However, it remains unclear how these core semantic areas dynamically communicate with each other, and with other neural networks, to achieve successful semantic processing. Here, we investigated this question by testing functional connectivity in the core semantic network during semantic tasks and whether these connections were affected by cognitive ageing. Compared to a non-semantic task, semantic tasks increased the connectivity between left and right IFGs, indicating a bilateral semantic control system. Strengthened connectivity was also found between left IFG and left vATL, and this effect was stronger in the young group. At a whole-brain scale, IFG and vATL increased their coupling with multiple-demand regions during semantic tasks, even though these areas were deactivated relative to non-semantic tasks. This suggests that the domain-general executive network contributes to semantic processing. In contrast, IFG and vATL decreased their interaction with default mode network (DMN) areas during semantic tasks, even though these areas were positively activated by the task. This suggests that DMN areas do not contribute to all semantic tasks: their activation may sometimes reflect automatic retrieval of task-irrelevant memories and associations. Taken together, our study characterizes a dynamic connectivity mechanism supporting semantic cognition within and beyond core semantic regions.
Macroscale brain states support the control of semantic cognition

Abstract:Understanding how the human brain adapts to varying cognitive demands is crucial in neuroscience. Here, we examined how networks involved in controlled semantic retrieval reconfigure themselves to generate neurocognitive states appropriate to different task contexts. We parametrically varied the demands of two semantic tasks - global association and feature matching judgments - and contrasted these effects of cognitive control with those of non-semantic tasks. We then characterized these effects on the cortical surface and within a whole-brain state space, anchored by the top three dimensions of intrinsic connectivity. Our results revealed that demanding semantic association tasks elicited more activation in the anterior regions of the prefrontal and temporal cortex. In contrast, difficult semantic feature matching tasks produced more posterior activation, aligning closely with regions engaged during multiple demanding non-semantic tasks. In both semantic feature matching and non-semantic contexts, the difficulty effects were situated towards the controlled end of a dimension capturing functional separation between cognitive control and default mode regions. Conversely, in semantic association tasks, the difficulty effects elicited similar responses across both cognitive control and default mode networks. Furthermore, controlled association and non-semantic control were located towards the heteromodal end of a heteromodal- unimodal dimension, while semantic feature matching involved a brain state that was more visual and unimodal. These findings demonstrate that a variety of brain states underpin controlled cognition. Specifically, cognitive control regions with heteromodal semantic knowledge system to identify contextually relevant conceptual overlaps (e.g., associating ’DOG’ with ’BEACH’), and from heteromodal memory regions for modality-specific conceptual overlaps (e.g., connecting ’DALMATIAN’ with ’BLACK AND WHITE’).

Abstract:Exploring the functional mechanism of the human brain during semantics categorization and subsequently leverage current semantics-oriented multimedia analysis by functional brain imaging have been receiving great attention in recent years. In the field, most of existing studies utilized strictly controlled laboratory paradigms as experimental settings in brain imaging data acquisition. They also face the critical problem of modeling functional brain response from acquired brain imaging data. In this paper, we present a brain decoding study based on sparse multinomial logistic regression (SMLR) algorithm to explore the brain regions and functional interactions during semantics categorization. The setups of our study are two folds. First, we use naturalistic video streams as stimuli in functional magnetic resonance imaging (fMRI) to simulate the complex environment for semantics perception that the human brain has to process in real life. Second, we model brain responses to semantics categorization as functional interactions among large-scale brain networks. Our experimental results show that semantics categorization can be accurately predicted by both intra-subject and inter-subject brain decoding models. The brain responses identified by the decoding model reveal that a wide range of brain regions and functional interactions are recruited during semantics categorization. Especially, the working memory system exhibits significant contributions. Other substantially involved brain systems include emotion, attention, vision and language systems.
Going beyond Inferior Prefrontal Involvement in Semantic Control: Evidence for the Additional Contribution of Dorsal Angular Gyrus and Posterior Middle Temporal Cortex

Abstract:Abstract Semantic cognition requires a combination of semantic representations and executive control processes to direct activation in a task- and time-appropriate fashion [Jefferies, E., & Lambon Ralph, M. A. Semantic impairment in stroke aphasia versus semantic dementia: A case-series comparison. Brain, 129, 2132–2147, 2006]. We undertook a formal meta-analysis to investigate which regions within the large-scale semantic network are specifically associated with the executive component of semantic cognition. Previous studies have described in detail the role of left ventral pFC in semantic regulation. We examined 53 studies that contrasted semantic tasks with high > low executive requirements to determine whether cortical regions beyond the left pFC show the same response profile to executive semantic demands. Our findings revealed that right pFC, posterior middle temporal gyrus (pMTG) and dorsal angular gyrus (bordering intraparietal sulcus) were also consistently recruited by executively demanding semantic tasks, demonstrating patterns of activation that were highly similar to the left ventral pFC. These regions overlap with the lesions in aphasic patients who exhibit multimodal semantic impairment because of impaired regulatory control (semantic aphasia)—providing important convergence between functional neuroimaging and neuropsychological studies of semantic cognition. Activation in dorsal angular gyrus and left ventral pFC was consistent across all types of executive semantic manipulation, regardless of whether the task was receptive or expressive, whereas pMTG activation was only observed for manipulation of control demands within receptive tasks. Second, we contrasted executively demanding tasks tapping semantics and phonology. Our findings revealed substantial overlap between the two sets of contrasts within left ventral pFC, suggesting this region underpins domain-general control mechanisms. In contrast, we observed relative specialization for semantic control within pMTG as well as the most ventral aspects of left pFC (BA 47), consistent with our proposal of a distributed network underpinning semantic control.

Abstract:Abstract Studies of patients with brain damage suggest that specific brain regions may be differentially involved in representing/processing certain categories of conceptual knowledge. With regard to the dissociation that has received the most attention—between the domains of living things and artifacts—a debate continues as to whether these category-specific effects reflect neural implementation of categories directly or some more basic properties of brain organization. The present positron emission tomography (PET) study addressed this issue by probing explicitly for differential activation associated with written names of objects from the domains of living things or artifacts during similarity judgments about different attributes of these objects. Subjects viewed triads of written object names and selected one of two response words as more similar to a target word according to a specified perceptual attribute (typical color of the objects) or an associative attribute (typical location of the objects). The control task required a similarity judgment about the number of syllables in the target and response words. All tasks were performed under two different stimulus conditions: names of living things and names of artifacts. Judgments for both domains and both attribute types activated an extensive, distributed, left-hemisphere semantic system, but showed some differential activation-particularly as a function of attribute type. The left temporooccipito-parietal junction showed enhanced activity for judgments about object location, whereas the left anteromedial temporal cortex and caudate nucleus were differentially activated by color judgments. Smaller differences were seen for living and nonliving domains, the positive findings being largely consistent with previous studies using objects; in particular, words denoting artifacts produced enhanced activation in the left posterior middle temporal gyrus. These results suggest that, within a distributed conceptual system activated by words, the more prominent neural distinction relates to type of attribute.

Abstract:Contemporary theories assume that semantic cognition emerges from a neural architecture in which different component processes are combined to produce aspects of conceptual thought and behaviour. In addition to the state-level, momentary variation in brain connectivity, individuals may also differ in their propensity to generate particular configurations of such components, and these trait-level differences may relate to individual differences in semantic cognition. We tested this view by exploring how variation in intrinsic brain functional connectivity between semantic nodes in fMRI was related to performance on a battery of semantic tasks in 154 healthy participants. Through simultaneous decomposition of brain functional connectivity and semantic task performance, we identified distinct components of semantic cognition at rest. In a subsequent validation step, these data-driven components demonstrated explanatory power for neural responses in an fMRI-based semantic localiser task and variation in self-generated thoughts during the resting-state scan. Our findings showed that good performance on harder semantic tasks was associated with relative segregation at rest between frontal brain regions implicated in controlled semantic retrieval and the default mode network. Poor performance on easier tasks was linked to greater coupling between the same frontal regions and the anterior temporal lobe; a pattern associated with deliberate, verbal thematic thoughts at rest. We also identified components that related to qualities of semantic cognition: relatively good performance on pictorial semantic tasks was associated with greater separation of angular gyrus from frontal control sites and greater integration with posterior cingulate and anterior temporal cortex. In contrast, good speech production was linked to the separation of angular gyrus, posterior cingulate and temporal lobe regions. Together these data show that quantitative and qualitative variation in semantic cognition across individuals emerges from variations in the interaction of nodes within distinct functional brain networks.
Pre-frontal cortex guides dimension-reducing transformations in the occipito-ventral pathway for categorization behaviors

Abstract:To interpret our surroundings, the brain uses a visual categorization process. Current theories and models suggest that this process comprises a hierarchy of different computations that transforms complex, high-dimensional inputs into lower-dimensional representations (i.e. manifolds) in support of multiple categorization behaviors. Here, we tested this hypothesis by analyzing these transformations reflected in dynamic MEG source activity while individual participants actively categorized the same stimuli according to different tasks: face expression, face gender, pedestrian gender, vehicle type. Results reveal three transformation stages guided by pre-frontal cortex. At Stage 1 (high-dimensional, 50-120ms), occipital sources represent both task-relevant and task-irrelevant stimulus features; task-relevant features advance into higher ventral/dorsal regions whereas task-irrelevant features halt at the occipital-temporal junction. At Stage 2 (121-150ms), stimulus feature representations reduce to lower-dimensional manifolds, which then transform into the task-relevant features underlying categorization behavior over Stage 3 (161-350ms).

Disentangling Semantic Composition and Semantic Association in the Left Temporal Lobe

Abstract:Although composing two words into a complex representation (e.g., "coffee cake") is conceptually different from forming associations between a pair of words (e.g., "coffee, cake"), the brain regions supporting semantic composition have also been implicated for associative encoding. Here, we adopted a two-word magnetoencephalography (MEG) paradigm which varies compositionality ("French/Korean cheese" vs "France/Korea cheese") and strength of association ("France/French cheese" vs "Korea/Korean cheese") between the two words. We collected MEG data while 42 English speakers (24 females) viewed the two words successively in the scanner, and we applied both univariate regression analyses and multivariate pattern classification to the source estimates of the two words. We show that the left anterior temporal lobe (LATL) and left middle temporal lobe (LMTL) are distinctively modulated by semantic composition and semantic association. Specifically, the LATL is mostly sensitive to high-association compositional phrases, while the LMTL responds more to low-association compositional phrases. Pattern-based directed connectivity analyses further revealed a continuous information flow from the anterior to the middle temporal region, suggesting that the integration of adjective and noun properties originated earlier in the LATL is consistently delivered to the LMTL when the complex meaning is newly encountered. Taken together, our findings shed light into a functional dissociation within the left temporal lobe for compositional and distributional semantic processing. SIGNIFICANCE STATEMENT Prior studies on semantic composition and associative encoding have been conducted independently within the subfields of language and memory, and they typically adopt similar two-word experimental paradigms. However, no direct comparison has been made on the neural substrates of the two processes. The current study relates the two streams of literature, and appeals to audiences in both subfields within cognitive neuroscience. Disentangling the neural computations for semantic composition and association also offers insight into modeling compositional and distributional semantics, which has been the subject of much discussion in natural language processing and cognitive science.

Abstract:Abstract Most contemporary theories of semantic memory assume that concepts are formed from the distillation of information arising in distinct sensory and verbal modalities. The neural basis of this distillation or convergence of information was the focus of this study. Specifically, we explored two commonly posed hypotheses: (a) that the human middle temporal gyrus (MTG) provides a crucial semantic interface given the fact that it interposes auditory and visual processing streams and (b) that the anterior temporal region—especially its ventral surface (vATL)—provides a critical region for the multimodal integration of information. By utilizing distortion-corrected fMRI and an established semantic association assessment (commonly used in neuropsychological investigations), we compared the activation patterns observed for both the verbal and nonverbal versions of the same task. The results are consistent with the two hypotheses simultaneously: Both MTG and vATL are activated in common for word and picture semantic processing. Additional planned, ROI analyses show that this result follows from two principal axes of convergence in the temporal lobe: both lateral (toward MTG) and longitudinal (toward the anterior temporal lobe).

Abstract:Recognising an object involves rapid visual processing and activation of semantic knowledge about the object, but how visual processing activates and interacts with semantic representations remains unclear. Cognitive neuroscience research has shown that while visual processing involves posterior regions along the ventral stream, object meaning involves more anterior regions, especially perirhinal cortex. Here we investigate visuo-semantic processing by combining a deep neural network model of vision with an attractor network model of semantics, such that visual information maps onto object meanings represented as activation patterns across features. In the combined model, concept activation is driven by visual input and co-occurrence of semantic features, consistent with neurocognitive accounts. We tested the model’s ability to explain fMRI data where participants named objects. Visual layers explained activation patterns in early visual cortex, whereas pattern-information in perirhinal cortex was best explained by later stages of the attractor network, when detailed semantic representations are activated. Posterior ventral temporal cortex was best explained by intermediate stages corresponding to initial semantic processing, when visual information has the greatest influence on the emerging semantic representation. These results provide proof of principle of how a mechanistic model of combined visuo-semantic processing can account for pattern-information in the ventral stream.

Abstract:Multiple sensory-motor and non-sensory-motor dimensions have been proposed for semantic representation, but it remains unclear how the semantic system is organized along them in the human brain. Using naturalistic fMRI data and large-scale semantic ratings, we investigated the overlaps and dissociations between the neural correlates of six semantic dimensions: vision, motor, socialness, emotion, space, and time. Our findings revealed a more complex semantic atlas than what is predicted by current neurobiological models of semantic representation. Brain regions that are selectively sensitive to specific semantic dimensions were found both within and outside the brain networks assumed to represent multimodal general and/or abstract semantics. Overlaps between the neural correlates of different semantic dimensions were mainly found inside the default mode network, concentrated in the left anterior superior temporal gyrus and angular gyrus, which have been proposed as two connector hubs that bridge the multimodal experiential semantic system and the language-supported semantic system.

Abstract:The representation of object categories is a classical question in cognitive neuroscience and compelling evidence has identified specific brain regions showing preferential activation to categories of evolutionary significance. However, the potential contributions to category processing by tuning the connectivity patterns are largely unknown. Adopting a continuous multicategory paradigm, we obtained whole-brain functional connectivity (FC) patterns of each of four categories (faces, scenes, animals and tools) in healthy human adults and applied multivariate connectivity pattern classification analyses. We found that the whole-brain FC patterns made high-accuracy predictions of which category was being viewed. The decoding was successful even after the contributions of regions showing classical category-selective activations were excluded. We further identified the discriminative network for each category, which span way beyond the classical category-selective regions. Together, these results reveal novel mechanisms about how categorical information is represented in large-scale FC patterns, with general implications for the interactive nature of distributed brain areas underlying high-level cognition. Abstract:Abstract How concepts are coded in the brain is a core issue in cognitive neuroscience. Studies have focused on how individual concepts are processed, but the way in which conceptual representation changes to suit the context is unclear. We parametrically manipulated the association strength between words, presented in pairs one word at a time using a slow event-related fMRI design. We combined representational similarity analysis and computational linguistics to probe the neurocomputational content of these trials. Individual word meaning was maintained in supramarginal gyrus (associated with verbal short-term memory) when items were judged to be unrelated, but not when a linking context was retrieved. Context-dependent meaning was instead represented in left lateral prefrontal gyrus (associated with controlled retrieval), angular gyrus, and ventral temporal lobe (regions associated with integrative aspects of memory). Analyses of informational connectivity, examining the similarity of activation patterns across trials between sites, showed that control network regions had more similar multivariate responses across trials when association strength was weak, reflecting a common controlled retrieval state when the task required more unusual associations. These findings indicate that semantic control and representational sites amplify contextually relevant meanings in trials judged to be related.

Abstract:Neurocognitive models of semantic memory have proposed that the ventral anterior temporal lobes (vATLs) encode a graded and multidimensional semantic space—yet neuroimaging studies seeking brain regions that encode semantic structure rarely identify these areas. In simulations we show that this discrepancy may arise from a crucial mismatch between theory and analysis approach. Utilizing an analysis recently formulated to investigate graded multidimensional representations, (RSL), we decoded semantic structure from ECoG data collected from the vATL cortical surface while participants named line drawings of common items. The results reveal a graded, multidimensional semantic space encoded in neural activity across the vATL, which evolves over time and simultaneously expresses both broad and finer-grained semantic structure amongst animate and inanimate concepts. The work resolves the apparent discrepancy within the semantic cognition literature and, more importantly, suggests a new approach to discovering representational structure in neural data more generally.
