Abstract:With the rapid development of cattle industry, bovine viral diarrhea virus (BVDV) is becoming widespread in China, which causes serious economic losses to the industry. Effective vaccination and viral surveillance are critical for the prevent and control of BVDV infection. In the present study, the immunogenic domain of E2 protein of BVDV-1 was expressed by prokaryotic pET-28a vector. Monoclonal antibodies (mAbs) against E2 protein were prepared and systemically examined by western blot, immunofluorescence assay, blocking ELISA (bELISA) and virus neutralization test (VNT). The mAb 1E2B3, which showed good reactivity and neutralizing activity to BVDV-1 strains, was selected for ELISA establishment. After a series of screening and optimization, a novel bELISA for highly sensitive and specific detection of BVDV-1 antibodies was established, using HRP-labeled 1E2B3 and recombinant E2 protein. ROC analysis of 91 positive and 84 negative reference bovine serum samples yielded the area under the curve (AUC) of 0.9903. A diagnostic specificity of 96.43 % and a sensitivity of 95.6 % were achieved when the cutoff value was set at 24.31 %. There was no cross reaction to the positive sera of classical swine fever virus (CSFV), BVDV-2, border disease virus (BDV), bovine parainfluenza virus type 3 (BPIV3), infectious bovine rhinotracheitis virus (IBRV), foot-and-mouth disease virus (FMDV), Mycoplasma bovis (M.bovis) and Brucella. The total agreement rate of bELISA with VNT was 93.96 % (249/265). In addition, the result of bELISA was positively correlated with neutralizing antibody titer, and the bELISA could well distinguish the serum samples before and after BVDV vaccination. These results indicate that the established bELISA in this study is specific, sensitive, simple and convenient, which provides technical support for the vaccine efficacy evaluation, prevention and control of BVD in the future.
Comparison of indirect ELISA and AGID for Detecting EBLV Antibody in the Serum Samples Dairy Cattle

Abstract:Objective to analyze the consistency of ELISA and AGID for detecting BLV antibody. Method The 3998 Serum samples of cows imported from Uruguay were collected and tested for the antibodies against BLV using ELISA and AGID. Result The results showed that the positive coincidence rate of the two methods was 100%(18/18),the negative coincidence was 99.42%(3975/3998) and the total coincidence rate was 99.87%(3993/3998). Conclusion Both ELISA and AGID could be applied in diagnosis and serological screening of EBL,and the ELISA positive rate was higher than that of AGID method.

Abstract:Porcine epidemic diarrhea virus (PEDV) causes severe infectious diseases in all ages of swine and leads to serious economic losses. Serologic tests are widely accepted and used to detect anti-PEDV antibodies that could indicate PEDV infection or vaccination. In this study, PEDV recombinant S1 protein (rS1) was expressed with the Bac-to-Bac system and purified by nickel-affinity chromatography. An indirect enzyme-linked immunosorbent assay based on rS1 (rS1-ELISA) was then developed and optimized by checkerboard assays with serial dilutions of antigen and serum. Serum samples from 453 domestic pigs and 42 vaccinated pigs were analyzed by the indirect fluorescent antibody (IFA) test and rS1-ELISA. Taking IFA as a gold standard, rS1-ELISA produced a high sensitivity (90.7%) and specificity (94.6%) by a receiver operating characteristic (ROC) curve. In addition, ROC analysis also revealed that rS1-ELISA was consistent with IFA (area under the curve 0.9583 ± 0.0082). This rS1-ELISA was then applied to antibody detection in inactivated PEDV vaccinated pigs. The antibody could be detected 2–4 weeks after the first inoculation. These results indicated that the rS1-ELISA established in this study provides a promising and reliable tool for serologic detection of anti-PEDV IgG antibodies in infected or vaccinated pigs.

Abstract:Bovine viral diarrhea virus (BVDV) is a major cause of economic loss in the cattle industry, worldwide. Infection results in reduced productive performance, growth retardation, reduced milk production, and increased susceptibility to other diseases leading to early culling of animals. There are two primary methods used to control the spread of BVDV: the elimination of persistently infected (PI) animals and vaccination. Currently, modified live or inactivated vaccines are used in BVDV vaccination programs, but there are safety risks or insufficient protection, respectively, with these vaccines. Here we report the development and efficacy of the first targeted subunit vaccine against BVDV. The core of the vaccine is the fusion of the BVDV structural protein, E2, to a single-chain antibody, APCH, together termed, APCH-E2. The APCH antibody targets the E2 antigen to the major histocompatibility type II molecule (MHC-II) present on antigen-presenting cells. Industrial production of the vaccine is carried out using the baculovirus expression vector system (BEVS) using single-use manufacturing technologies. This new subunit vaccine induces strong BVDV-specific neutralizing antibodies in guinea pigs and cattle. Importantly, in cattle with low levels of natural BVDV-specific neutralizing antibodies, the vaccine induced strong neutralizing antibody levels to above the protective threshold, as determined by a competition ELISA. The APCH-E2 vaccine induced a rapid and sustained neutralizing antibody response compared to a conventional vaccine in cattle.

Abstract:Infection with bovine leukemia virus (BLV) leads to enzootic bovine leukosis, the most prevalent neoplastic disease in cattle. Due to the lack of commercially available vaccines, reliable eradication of the disease can be achieved through the testing and elimination of BLV antibody-positive animals. In this study, we developed a novel competitive ELISA (cELISA) to detect antibodies against BLV capsid protein p24. Recombinant p24 protein expressed by Escherichia coli , in combination with the monoclonal antibody 2G11 exhibiting exceptional performance, was used for the establishment of the cELISA. Receiver-operating characteristic curve analysis showed that the sensitivity and specificity of the assay were 98.85 % and 98.13 %, respectively. Furthermore, the established cELISA was specific for detecting BLV-specific antibodies, without cross-reactivity to antisera for six other bovine viruses. Significantly, experimental infection of cattle and sheep with BLV revealed that the cELISA accurately monitors seroconversion. In a performance evaluation, the established cELISA displayed a high agreement with Western blotting and the commercial BLV gp51 cELISA kit in the detection of 242 clinical samples, respectively. In conclusion, the novel p24 cELISA exhibited the potential to be a reliable and efficient diagnostic tool for BLV serological detection with a broad application prospect.

Abstract:BACKGROUND: Bovine viral diarrhea (BVD) is an economically important cattle disease with a worldwide distribution. Detection and elimination of animals persistently infected (PI) with bovine viral diarrhea virus (BVDV) is essential for the control of BVD and eradication of BVDV. There are usually no pathognomonic clinical signs of BVDV infection. Diagnostic investigations therefore rely on laboratory-based detection of the virus, or virus-induced antigens or antibodies.OBJECTIVES: Erns as an immunogenic protein of BVDV, is genetically and antigenically conserved among different isolates and therefore, is a candidate antigen for development of the enzyme linked immunosorbent assay (ELISA) for serological studies or identification of PI animals. The aim of this study was to produce a monoclonal antibody (MAb) against recombinant Erns.MATERIALS AND METHODS: For this purpose, recombinant maltose-binding protein (MBP)-Erns protein was expressed in Escherichia coli and purified using amylose resin chromatography column and used as an antigen in MAb production. Spleen cells of the immunized mice with the recombinant antigen were fused with SP2/0 myeloma cells. Next, culture supernatants of primary clones of fused cells were screened by indirect ELISA. After three rounds of cloning, the reactivity of the MAbs with recombinant and natural antigen was established by Western blotting.RESULTS: Based on our results, MAb against recombinant Erns was produced and reacted successfully with recombinant and natural antigens.CONCLUSIONS: With regards to the role of Erns in the identification of PI animals, it appears that Erns recombinant antigen and the specific monoclonal antibodies produced against it may be suitable for developing BVDV laboratory diagnostic assays.

Abstract:Summary Two-day-old chicks uniformly developed leptospiremia persisting for at least 5 days following intraperitoneal inoculation of leptospires. No clinically detected illness resulted from this infection nor was there constant significant serologically demonstrable antibody response. Employment of chicks for recovery of leptospires form materials contaminated with other microorganisms and hence unsuitable for direct culture is suggested.

Abstract:In order to determine the level of antibody against PEDV in swine serum .The PEDV recombinant N protein was expressed by E.coli and purified through native conditions purification method .Based on purified re-combinant N protein ,an indirect ELISA for detection of anti-PEDV antibodies was developed and its optimal reac-tion conditions were determined:antigen working concentration was 0.5 μg/mL,it was coated at 37 ℃for 2 h and 4 ℃overnight,serum samples dilution was 1∶100,incubated at 37 ℃for 45 min.HRP-labeled rabbit anti-pig IgG was diluted at 1∶10 000 ,incubated at 37 ℃for 60 min,the substrate for showing color was incubated at 37 ℃for 15 min.It was judged to be positive when the OD was greater than the cut off OD 450nm of 0.30,as negative when OD450nm was smaller than 0.27,and as suspicious between 0.27 and 0.30.The established ELISA assay was confirmed to be good in specificity,sensitivity and repeatability by repeatability and cross test .A total of 184 clinical serum samples obtained from pig farms in Zhejiang Province were detected by using the established ELISA and the positive rate was 68.5%.

Abstract:Bovine viral diarrhea virus (BVDV) causes a significant economic impact on the beef and dairy industries worldwide. Fetal infection with a non-cytopathic strain may lead to the birth of persistently infected (PI) offspring, the main event in the epidemiological chain of BVDV infection. This report describes the birth of 10 PI dairy calves from Brazil and the infecting BVDV subgenotype. Serum and blood samples were collected from 10 cows and 10 calves; all 10 calves were previously deemed BVDV positive by the ear notch rapid test. Serum samples were used in the virus neutralization technique to detect anti-BVDV antibody. Blood samples were used to detect BVDV RNA using the reverse transcription polymerase chain reaction (RT-PCR) assay of the 5' UTR and Npro genes. All 10 cows were negative for BVDV RT-PCR, while all 10 calves were RT-PCR positive. Phylogenetic analyses were performed and the strain was classified as BVDV-1d. High titers of BVDV-specific antibodies in the serum of cows indicated recent circulation of BVDV in the dairy herd, whereas calves presented intermediate, low, or no anti-BVDV-1a antibody titers. The monitoring of circulating BVDV subgenotypes and the detection of PI animals is of great importance in disease control, and regular vaccination alone is insufficient to prevent BVDV infection.

Abstract:The objective of this study was to validate an indirect enzyme-linked immunoassay (iELISA) using the recombinant proteins, malate dehydrogenase (MDH) and superoxide dismutase (SOD) [CuZn], as antigens and to evaluate its ability to discriminate antibodies produced by vaccination from those induced by infection in the diagnosis of bovine brucellosis. Sera from six groups were evaluated: G1 - culture-positive animals (52 serum samples) (naturally infected); G2 - non-vaccinated animals (28 serum samples) positive in RBT (Rose Bengal test) and 2ME (2-mercaptoethanol test) selected from brucellosis-positive herds; G3 - animals from a brucellosis-free area (32 serum samples); G4 - S19 vaccinated heifers (114 serum samples); G5 - RB51 vaccinated heifers (60 serum samples); G6 - animals inoculated with inactivated Yersinia enterocolitica O:9 (42 serum samples). Diagnostic sensitivity (DSe) and diagnostic specificity (DSp) were estimated using the frequentist approach and the confidence interval (CI) (95%) calculated by the Clopper-Pearson (exact) method. The DSe for iELISA_MDH in the G1 group was 71.7% (CI 95%: 57.6-83.2%) and for the G2 100.0% (CI 95%: 87.7-100.0%), whereas the DSp was 84.4% in the G3 (CI 95%: 67.2-94.7%). For the iELISA_SOD the DSe was estimated 67.3% for the G1 (CI 95%: 52.9-79.7%) and 71.4% for G2 (CI 95%: 51.3-86.8%), while the DSp for G3 was 87.5% (CI 95%: 71.0-96.5%). iELISA_MDH and iELISA_SOD showed potential to be used in the diagnosis of infected animals, increasing the range of serological tests available for the diagnosis of bovine brucellosis, with the advantage of being S-LPS-free. However, none of the tests could differentiate between infection and vaccination.

Abstract:Swine vesicular disease (SVD) is a contagious viral disease of pigs. The clinical signs of SVD are indistinguishable from other vesicular diseases, such as Senecavirus A infection (SVA) and foot-and-mouth disease (FMD). Rapid and accurate diagnostic tests of SVD are considered essential in countries free of vesicular diseases. Competitive ELISA (cELISA) is the serological test used routinely. However, although cELISA is the standard test for SVD antibody testing, this test produces a small number of false-positive results, which caused problems in international trade. The current project developed a SVD isotype antibody ELISA using recombinant SVD virus-like particles (VLP) and an SVD-specific monoclonal antibody (mAb) to reduce the percentage of false-positives. The diagnostic specificities of SVD-VLP isotype-ELISAs were 98.7% and 99.6% for IgM and IgG. The SVD isotype-ELISAs were SVD specific, without cross-reactivity to other vesicular diseases. A panel of 16 SVD positive reference sera was evaluated using the SVD-VLP isotype-ELISAs. All sera were correctly identified as positive by the two combined SVD-VLP isotype-ELISAs. Comparison of the test results showed a high level of correlation between the SVDV antigen isotype-ELISAs and SVD-VLP isotype-ELISAs. 303 sera from animals lacking clinical signs and history of SVDV exposure were identified positive using SVD cELISA. These samples were examined using SVD-VLP isotype-ELISAs. Of the 303 serum samples, five were positive for IgM, and five of 303 were positive for IgG. Comparable to virus neutralization test results, SVD isotype-ELISAs significantly reduced the false-positive samples. Based on above test results, the combined use of cELISA and isotype-ELISAs can reduce the number of false-positive samples and the use of time-consuming virus neutralization tests, with benefit for international trade in swine and related products.

Validation of a competitive enzyme-linked immunosorbent assay to improve the serological diagnosis of swine vesicular disease.

Abstract:Swine vesicular disease (SVD) is an infectious viral disease of pigs. The clinical symptoms of SVD are indistinguishable from other vesicular diseases. In countries free of vesicular diseases, rapid SVD diagnosis and differentiation from other vesicular diseases are essential. In this report, a competitive enzyme-linked immunosorbent assay (cELISA) was developed and validated to improve the current SVD serological diagnosis. In this cELISA, an anti-SVD monoclonal antibody (mAb) captures the recombinant SVD virus-like particle (SVD-VLP) antigen, and 5B7 mAb is used as a competitor to compete with polyclonal antibodies in SVD-positive sera. The cut-off value of the SVD-VLP based cELISA (SVD-VLP cELISA) is ≥ 65% inhibition (%). The determined diagnostic specificity was 99.2%. SVD-VLP cELISA successfully detected SVD antibodies in the sera of SVD-infected animals and produced a diagnostic sensitivity of 100%. A panel of SVD positive sere including outbreak samples (n = 11) and samples (n = 5) from experimentally inoculated pigs, were correctly identified as positive by the SVD-VLP cELISA. In terms of reducing false positives detected by the currently used cELISA (5B7 cELISA), the performance of SVD-VLP cELISA is comparable to the gold standard virus neutralization test.

Abstract:Infectious bursal disease virus is the causative agent of Gumboro disease, a severe infection that affects young chickens and is associated with lymphoid depletion in the bursa of Fabricius. Traditional containment strategies are based either on inactivated or live-attenuated vaccines. These approaches have several limitations such as residual virulence or low efficacy in the presence of maternally derived antibodies (MDA) but, most importantly, the impossibility to detect the occurrence of natural infections in vaccinated flocks. Therefore, the development of novel vaccination strategies allowing the differentiation of infected from vaccinated animals (DIVA) is a priority. Recently, commercial vectored and experimental subunit vaccines based on VP2 have been proved effective in protecting from clinical disease and posed the basis for the development of novel DIVA strategies. In this study, an engineered version of the VP3 protein of IBDV (His-VP3) was produced in plants, successfully purified from Nicotiana benthamiana leaves, and used to develop an enzyme-linked immunosorbent assay (ELISA) for the detection of anti-VP3 antibodies. The His-VP3 ELISA was validated with a panel of 180 reference sera and demonstrated to have 100% sensitivity (95% CI: 94.7–100.0) and 94.17% specificity (95% CI: 88.4–97.6). To evaluate the application of His-VP3 ELISA as a DIVA test, the novel assay was used to monitor, in combination with a commercial kit, detecting anti-VP2 antibodies, the immune response of chickens previously immunized with an inactivated IBDV vaccine, a recombinant Turkey herpes virus carrying the VP2 of IBDV (HVT-ND-IBD) or with plant-produced VP2 particles. The combined tests correctly identified the immune status of the vaccinated specific pathogen free white-leghorn chickens. Moreover, the His-VP3 ELISA correctly detected MDA against VP3 in commercial broiler chicks and showed that antibody titers fade with time, consistent with the natural decrease of maternally derived immunity. Finally, the novel assay, in combination with a VP2-specific ELISA, demonstrated its potential application as a DIVA test in chickens inoculated with VP2-based vaccines, being able to detect the seroconversion after challenge with a very virulent IBDV strain.

Abstract:The level of neutralizing antibodies in vaccinated animals is directly related to their level of protection against a virus challenge. The virus neutralization test (VNT) is a "gold standard" method for detecting neutralizing antibodies against foot-and-mouth disease virus (FMDV). However, VNT requires high-containment facilities that can handle live viruses and is not suitable for large-scale serological surveillance. In this study, a bovine broadly neutralizing monoclonal antibody (W145) against FMDV serotype A was successfully produced using fluorescence-based single-B-cell antibody technology. Using biotinylated W145 as a detector antibody and another bovine cross-reactive monoclonal antibody, E32, which was produced previously as a capture antibody, a competitive enzyme-linked immunosorbent assay for the detection of neutralizing antibodies (NAC-ELISA) against FMDV serotype A was developed. The specificity and sensitivity of the assay were evaluated to be 99.04% and 100%, respectively. A statistically significant correlation ( = 0.9334, < 0.0001) was observed between the NAC-ELISA titers and the VNT titers, suggesting that the NAC-ELISA could detect neutralizing antibodies against FMDV serotype A and could be used to evaluate protective immunity.
Development and Preliminary Application of Double Antibody Sandwich ELISA for Detection of Swine Vesicular Disease Virus.

Abstract:[Objective] The aim of this study was to develop an indirect sandwich ELISA method for detection of swine vesicular disease virus (SVDV). [Method] High titer serum against SVDV was prepared respectively by inoculated rabbits and guinea pigs with purified virus. To develop an indirect sandwich ELISA, the optimum concentrations of capture antibody, detection antibody, enzyme conjugate and standard antigen were determined using block titration, and positive threshold value was also determined. The specificity, sensitivity and repeatability of the developed ELISA were evaluated using cross-reaction test, comparison test and intra-assay repeated test. In addition, standard samples and clinical samples were detected by this method. [Result] The best working conditions of the developed ELISA are as follows: capture antibody, 1∶400; detection antibody, 1∶200; enzyme conjugate, 1∶8 000; and standard antigen, 1∶4. The positive threshold value was found to be 0.20. For the detection by the developed ELISA, no cross-reaction with foot and mouth disease was observed. The developed ELISA had close sensitivity with ELISA recommended by the World Organisation for Animal Health (OIE) but had sensitivity 2-4 times higher than that of reverse indirect hemagglutination test. In addition, the developed method also had good reproducibility, and the detection results of standard samples were in line with their own background. All the 36 clinical samples were negative in the developed ELISA. [Conclusion] The developed indirect sandwich ELISA can be used for diagnosis of swine vesicular disease.

Abstract:Diagnostic assays that are able to detect foot-and-mouth disease (FMD) virus infection in the vaccinated population are essential tools in the progressive control pathway for the FMD. However, testing of serum samples using a single diagnostic assay may not completely substantiate freedom from the virus infection. Therefore, viral non-structural proteins (NSPs)-based various serological assays have been developed for the detection of FMD infection. Nevertheless, the NSPs-based ELISAs have been developed in the indirect-ELISA format, thereby necessitating the use of species-specific conjugated secondary-antibodies for the detection of anti-NSP antibodies in various FMD-susceptible species. Therefore, this study presents a novel recombinant 2B-NSP-based indirect ELISA, employing HRP-conjugated protein-A/G detection system which can detect anti-NSPs antibodies from multiple FMD-susceptible species in a single ELISA platform. Recombinant 2B (r2B) protein was expressed as His-SUMO tagged protein in the E. Coli cells and purified using NI-NTA affinity column chromatography. Using the r2B protein and HRP-conjugated protein A/G, an indirect ELISA was developed and validated for the detection of anti-2B antibodies in serum samples collected from multiple FMD-susceptible animal species with known FMD status. Further, a resampling based statistical technique has been reported for determination of optimal cut-off value for the diagnostic assay. Through this technique, the optimal cut-off of 44 percentage of positivity value was determined for the assay. At this optimal cut-off value, the developed diagnostic assay provided diagnostic sensitivity, specificity, and accuracy, positive and negative predictive values (PPV and NPV) of 92.35 %, 98.41 %, 95.21 %, 98.58 %, and 91.67 %, respectively. The assay was validated further by analyzing random serum samples collected across multi-locations in India. The assay can be used as a single platform for testing serum samples from different species of FMDV-susceptible animals and will be useful for NSP-based serosurveillance of FMDV.
