Abstract:This study investigated the influence of different temperatures (35°C High temperature and average indoor ambient temperature of 25°C) and lactic acid bacterial additives ( Lactiplantibacillus plantarym , Lentilactobacillus buchneri , or a combination of Lactiplantibacillus plantarym and Lentilactobacillus buchneri ) on the chemical composition, fermentation quality, and microbial community of alfalfa silage feed. After a 60-day ensiling period, a significant interaction between temperature and additives was observed, affecting the dry matter (DM), crude protein (CP), acid detergent fiber (ADF), and neutral detergent fiber (NDF) of the silage feed ( p < 0.05). Temperature had a highly significant impact on the pH value of the silage feed ( p < 0.0001). However, the effect of temperature on lactic acid, acetic acid, propionic acid, and butyric acid was not significant ( p > 0.05), while the inoculation of additives had a significant effect on lactic acid, acetic acid, and butyric acid ( p > 0.05). As for the dynamic changes of microbial community after silage, the addition of three kinds of bacteria increased the abundance of lactobacillus . Among all treatment groups, the treatment group using complex bacteria had the best fermentation effect, indicating that the effect of complex lactic acid bacteria was better than that of single bacteria in high temperature fermentation. In summary, this study explained the effects of different temperatures and lactic acid bacterial additives on alfalfa fermentation quality and microbial community, and improved our understanding of the mechanism of alfalfa related silage at high temperatures.

Abstract:The wilting process is required for the preparation of traditional low-moisture silage, which is not only subject to the interference of rainfall during the harvest season in many areas, but also increases labor, economic, and time costs. Therefore, in this experiment, newly harvested alfalfa was supplemented with a complex additive consisting of lactic acid bacteria (LAB), sucrose, and corn flour, and then ensiled directly in order to explore a suitable high-moisture silage processing method. There were four groups: control (CK), 0.01 g/kg LAB plus 2% sugar (S), 0.01 g/kg LAB plus 5% corn flour (C5), and 0.01 g/kg LAB plus 5% corn flour (C10). The results show that all four types of silage had significantly reduced acid detergent fiber, acetic acid content, and ammonia/total nitrogen (p < 0.05). The relative abundance of Lactobacillus spp. was significantly higher (p < 0.05), and that of Weissella, Clostridium sensu stricto 12, and Pantoea genera was significantly lower (p < 0.05) in all four treatments. Group S had significantly increased crude protein content, and groups C5 and C10 had significantly reduced neutral detergent fiber. Overall, group C10 had the highest LA concentration, the highest relative abundance of Lactobacillus spp., and the lowest pH, ammonia/total nitrogen, neutral detergent fiber, and acid detergent fiber. Silage with a complex additive consisting of LAB + 10% corn flour is recommended to achieve good fermentation with high nutritional quality.

Abstract:This study was carried out to determine the effect of fermented natural lactic acid bacteria liquid and water-soluble carbohydrate (WSC) admixture on alfalfa silage fermentation quality, in vitro digestibility and methane production. In the study, analyses (pH, total lactic acid bacteria (LAB), yeast, mold, lactic acid (LA), acetic acid (AA), LA/AA) of naturally fermented lactic acid bacteria liquid (PFJ) prepared with 3% fructose addition to alfalfa plant were conducted. Treatments included alfalfa (control, C), alfalfa + PFJ (PFJ-C), alfalfa + PFJ +1,5% molasses (PFJ-CM), alfalfa + PFJ + 1,5% fructose (PFJ-CF), alfalfa + PFJ + 1,5% sucrose (PFJ-CS). In the study, while the differences between the groups were found statistically insignificant in crude ash (CA) and neutral detergent fiber (NDF) values of the silages, the differences between the groups were found statistically significant in dry matter (DM), crude protein (CP), acid detergent fiber (ADF), in vitro organic matter digestion (IVOMD), metabolizable energy (ME) and methane (CH4) values. Compared to the control group, increases in IVOMD and ME were observed in silages obtained by adding PFJ, molasses, fructose and sucrose. When the fermentation characteristics (pH, ammonia nitrogen (NH3-N), lactic acid (LA), acetic acid (AA), yeast-mold and carbon dioxide (CO2) after aerobic stability) of the silages were analyzed, the differences between the groups were statistically significant. When the CO2 content and post aerobic yeast mold values (PAYMV) of the silages were examined, it was observed that there was a decrease in all experimental groups compared to the control group. LA and AA values of silages increased in all experimental groups compared to the control group.

Abstract:This study examined the effects of different additives on the fermentation quality, nutrient composition, microbial communities, and rumen degradation of ensiled alfalfa. Six treatments were employed in which additives were applied to alfalfa on a fresh weight basis: CK (no additive), FA (0.6% formic acid), CaO (3% calcium oxide and 3% urea), LB (1 × 106 cfu/g Lentilactobacillus buchneri), GLB (2% glucose and 1 × 106 cfu/g L. buchneri), and FLB (2% fucoidan and 1 × 106 cfu/g L. buchneri). After 60 days of ensiling, all treatments altered the bacterial communities, improved the fermentation quality, reduced dry matter (DM) and crude protein (CP) losses, and enhanced the rumen degradation of nutrients. The addition of LB increased the relative abundance of Lactobacillus spp. (p < 0.05), whereas GLB reduced (p < 0.05) the NH3-N:TN ratio and elevated (p < 0.05) the concentrations of Lactobacillus and lactic acid content. The FA treatment reduced (p < 0.05) the pH, as well as the DM and CP degradations, while the CaO treatment increased the degradations of DM, acid detergent fiber, and neutral detergent fiber. We concluded that FA, LB, GLB, and FLB had beneficial effects on alfalfa fermentation, and that CaO increased alfalfa silage rumen degradation.

Effect of Ensiling Density and Storage Temperature on Fermentation Quality, Bacterial Community, and Nitrate Concentration of Sorghum-Sudangrass Silage

Abstract:This study aimed to evaluate the fermentation quality, bacterial community, and nitrate content of sorghum-sudangrass silage with two ensiling densities [550 kg fresh weight (FW)/m 3 (low density, LD) and 650 kg FW/m 3 (high density, HD)] stored at two temperatures [10°C (low temperature, LT) and 25°C (normal temperature, NT)] for 60 days. The fermentation parameters, microbial counts, bacterial community, nutritional composition, and nitrate and nitrite levels were assessed. The pH and ammonia nitrogen (N) in all silages were below 4.0 and 80 g/kg total N, respectively. Compared with LT treatments, NT treatments had lower pH and lactic acid (LA) bacteria and yeasts counts and contained higher LA and LA/acetic acid (LA/AA) ( p < 0.05). The LT-LD contained more ammonia–N than LT-HD ( p < 0.05) and had higher nitrate and lower nitrate degradation than other treatments ( p < 0.05). Lactobacillus was the most dominant genus with all treatments (57.2–66.9%). The LA, LA/AA, and abundances of Pantoea , Pseudomonas , and Enterobacter in the silage negatively correlated with nitrate concentration and positively correlated with nitrate degradation ( p < 0.05). Moreover, pH and ammonia–N were positively correlated with nitrate concentration and negatively correlated with nitrate degradation ( p < 0.05). Overall, all silage had satisfactory fermentation quality, and the silage with HD and NT had better fermentation quality and higher nitrate degradation. The bacterial communities in all silages were dominated by Lactobacillus . The nitrate degradation during the fermentation process might be related to the fermentation quality and the activity of Pantoea , Pseudomonas , and Enterobacter in silage.

Abstract:The aim of this study was to investigate effects of wilting and Lactobacillus plantarum inoculation on the dynamics of the fermentation products, residual non-structural carbohydrates, and bacterial communities in alfalfa silage. Fresh and wilted alfalfa were ensiled with and without L. plantarum for 10, 30, 60, and 90 days. A high-throughput sequencing method for absolute quantification of 16S rRNA was adopted to determine the bacterial community composition at different ensiling periods. For the wilted silage, the bacterial community, pH value, and ammonia nitrogen concentration remained stable in the silage at 30 days. L. plantarum inoculation accelerated lactic acid fermentation and altered the predominant genus in the wilted silage as compared with the non-inoculated group. For the non-wilted group, fast consumption of water-soluble carbohydrates (WSCs) was observed at 10 days in the non-inoculated silage along with rapid growth of undesirable Hafnia . L. plantarum inoculation inhibited growth of Hafnia at 10 days in the non-wilted silage. Clostridia fermentation occurred in the non-wilted silage at 90 days, as indicated by an increased pH, formation of butyric acid (BA), and apparent abundance of genera belonging to Clostridia. L. plantarum inoculation inhibited BA accumulation and growth of Garciella in the non-wilted silage at 90 days as compared with the non-wilted silage without inoculation, but had little effect on the growth of Clostridium sensu stricto . Overall, the high moisture content of the non-wilted alfalfa silage led to rapid consumption of WSCs and growth of harmful microorganisms at the early stage of ensiling, resulting in poor fermentation quality. Wilting and L. plantarum inoculation both improved fermentation quality and inhibited the growth of spoilage microorganisms in alfalfa silage, while L. plantarum inoculation alone failed to achieve optimum fermentation quality of non-wilted alfalfa silage.

Abstract:The aim of this study was to determine the effects of six common commercial lactic acid bacteria (LAB) additives [A1, Lactobacillus plantarum , L. buchneri , and Enterococcus faecalis ; A2, L. plantarum and L. casei ; A3, L. plantarum and L. buchneri ; A4, L. plantarum , L. buchneri , L. casei , and Pediococcus acidilactici ; A5, L. plantarum (producing feruloyl esterase); and A6, L. buchneri , P. acidilactici , β-glucanase, and xylanase] on the bacterial community and fermentation quality of alfalfa silage. Alfalfa was harvested at the squaring stage, wilted in the field for 24 h, and ensiled without any additives (Control) or with A1, A2, A3, A4, A5, or A6. Microbial counts, bacterial community, fermentation parameters, and nutritional composition were determined after ensiling for 90 days. The total abundance of LAB genera on alfalfa pre-ensiling was 0.38% in bacterial community. The abundances of Lactobacillus , Enterococcus , and Pediococcus in the Control silage were 42.18, 40.18, and 8.09% of abundance, respectively. The abundances of Lactobacillus in A1-, A2-, A3-, A4-, and A5-treatments were 89.32, 92.93, 92.87, 81.12, and 80.44%, respectively. The abundances of Pediococcus and Lactobacillus in A6-treatment were 70.14 and 24.86%, respectively. Compared with Control silage, LAB-treated silage had lower pH and less ammonia nitrogen and water-soluble carbohydrates concentrations ( p < 0.05). Further, the A5- and A6-treatments contained lower neutral detergent fiber, acid detergent fiber, and hemicellulose than other treatments ( p < 0.05). Overall, LAB genera were presented as minor taxa in alfalfa pre-ensiling and as dominant taxa in alfalfa silage. Adding LAB additives improved the fermentation quality and altered the bacterial community of alfalfa silage. The main bacterial genera in Control silage were Lactobacillus , Enterococcus , and Pediococcus . Lactobacillus dominated the bacterial communities of A1-, A2-, A3-, A4-, and A5-treatments, while Pediococcus and Lactobacillus were dominant bacterial genera in A6-treatment. Inoculating A5 and A6 degraded the fiber in alfalfa silage. It is necessary to ensile alfalfa with LAB inoculants.

Abstract:Abstract In practical of silage production, variations in the fermentation parameters of silage might be attributable to changes in weather data, especially for ambient temperature. The objective of this study was to investigate how the changed temperature of outside weather condition affects distributions of pH and fermentation products in large round bale silage. The bale silages (1.22 m × 1.25 m ø; density, about 168.9 kg dry matter [DM]/m 3 ) were produced by BR6090 Combi from New Holland Agriculture, wrapped with six layers of stretch film, stored at ambient temperature (2–20°C) and constant temperature (10°C; 15°C; 25°C) for 3, 9, 15, 45 and 90 days. Compared with other silages, silage stored at 25°C had higher ( p < 0.05) lactic acid content, lower ( p < 0.05) pH, butyric acid content and ammonia‐nitrogen (N) ratio of total N. Fluctuant storage temperature limited lactic acid fermentation process with delayed pH decline and continuous butyric production and crude protein (CP) degradation. The differences of pH, lactic acid, propionic acid and ammonia‐N were considerably less evident in the centre of silages stored at different temperatures, and they increased towards the boundaries. The data shows that fluctuant storage temperature had undesirable effect on the fermentation quality of silage, especially for the external part of silo; relatively high and stable storage temperature for initial stage of ensiling process was necessary, but did not promote a positive response to the homogeneous distributions of pH and fermentation products in large round bale silage.

Abstract:The objective of this experiment was to investigate the effects of Lactobacillus plantarum and molasses on the nutrient composition, fermentation quality, bacterial count, aerobic stability, and microflora of alfalfa silage in sandy grasslands. The experimental treatments included control (CK), 10 6 CFU/g Lactobacillus plantarum (L), 5% molasses (M), and 10 6 CFU/g Lactobacillus plantarum + 5% molasses (LM). The nutrient composition, fermentation quality, bacterial count, aerobic stability, and microflora were determined after 14 days and 56 days of ensiling, respectively. The results showed that the addition of L, M, and LM reduced dry matter loss (DM), neutral detergent fiber (NDF), and acid detergent fiber (ADF) content, and increased water-soluble carbohydrates (WSC) and ether extract (EE) content, compared to the CK group. Meanwhile, more lactic acid (LA) and accelerated fermentation were observed, causing the pH value to drop below 4.5 in the L, M, and LM groups after 56 days of ensiling. The addition of L, M, and LM promoted lactic acid bacteria (LAB), and inhibited yeast. The addition of L significantly increased the content of acetic acid (AA). In terms of microflora, the addition of L, M, and LM made Firmicutes become the dominant bacterial phylum earlier, while Lactobacillus, Weissella , and Pediococcus had a higher abundance. According to the result of Pearson's correlation, there is a very significant negative correlation between pH value and Lactobacillus ( P < 0.01) and a very significant positive correlation between pH value and Lactococcus, Enterobacter, Enterococcus , and Leuconostoc ( P < 0.01), which may be inhibited by Lactobacillus under the decreased pH value. The results of the prediction of microbial genes indicated that the addition of M could enhance the carbohydrate metabolism and membrane transport metabolism, which may contribute to LA production by LAB metabolism. In general, L, M and LM all improved the fermentation quality and reduced the loss of nutrients to varying degrees, but considering the fermentation quality, the overall effects of M and LM were better than L. M and LM are recommended to be used as silage additives in the process of alfalfa silage in sandy grasslands to improve the quality.

Abstract:Abstract Background The objective of this study was to evaluate the chemical compositions and microbial communities of salt-tolerant alfalfa silage. Salt-tolerant alfalfa was ensiled with no additive control, and cellulase for 30 and 60 to 90 days. In this study, the dry matter (DM) content of the raw material was 29.9% DM, and the crude protein (CP) content of the alfalfa was 21.9% CP. Results After 30 days of fermentation, the DM content with the cellulase treatment was reduced by 3.6%, and the CP content was reduced by 12.7%. After 60 days of fermentation, compared with alfalfa raw material, the DM content in the control group (CK) was reduced by 1%, the CP content was reduced by 9.5%, and the WSC (water-soluble carbohydrates) content was reduced by 22.6%. With the cellulase, the lactic acid content of the 30- and 60-day silages was 2.66% DM and 3.48% DM. The content of Firmicutes in salinized alfalfa raw material was less than 0.1% of the total bacterial content. Before and after ensiling, the microbes had similar composition at the phylum level, and were composed of Firmicutes , Actinobacteria , Bacteroidetes, and Proteobacteria . The abundance of Pantoea was dominant in fresh alfalfa. In the absence of additives, after 30 days and 60 days of silage, the dominant lactic acid bacteria species became Lactococcus and Enterococcus . Conclusions The results showed that LAB ( Lactobacillus , Lactococcus , Enterococcus, and Pediococcus ) played a major role in the fermentation of saline alfalfa silage. It also can better preserve the nutrients of saline alfalfa silage. The use of cellulase enhances the reproduction of Lactobacillus . The fermentation time would also change the microbial community of silage fermentation.

Impact of Packing Density on the Bacterial Community, Fermentation, and In Vitro Digestibility of Whole-Crop Barley Silage

Abstract:Packing density has a significant influence on the outcome of ensiling forage. In this study, we aim to investigate the effect of packing density on the ensiling properties, microbiome, and in vitro digestibility of barley silages. Barley was ensiled in polyethylene drum silos (30 L) with respective densities of 600, 650, 700, and 750 kg/m3 fresh matter (FM), and stored for 60 days. The bacterial communities, fermentation quality, and in vitro digestibility were analyzed. Fresh barley had a low count of lactic acid bacteria (LAB, 104 cfu/g of FM), and Lactobacillus was nearly undetectable (<1%). Increasing the packing density decreased the pH and the content of ammonia nitrogen (NH3-N), ethanol, neutral detergent fiber (NDF), and acid detergent fiber (ADF) of barley silage (p < 0.05), and increased in vitro digestibility of dry matter, NDF, ADF, and DM recovery (p < 0.05). A higher packing density decreased the abundances of Enterobacter (from 47.4% to 35.4%) and Clostridium (from 13.5% to 3.8%), and increased the abundance of Lactobacillus (from 1.8% to 17.0%). Thus, packing density positively correlated with Lactobacillus (p < 0.05) but negatively correlated with Enterobacter (p < 0.05). The pH and the content of ethanol were positively correlated with Enterobacter (p < 0.05) but negatively correlated with Lactobacillus (p < 0.05). In conclusion, the density of 750 kg/m3 FM resulted in the highest silage quality of the densities tested.

Abstract:Both inoculants treatment and enzyme treatment promote the reproduction of lactic acid bacteria (LAB) to produce enough lactic acid to lower pH in silage. The present study investigated the microbial community and metabolome in cellulase, Lactobacillus casei , and air treated alfalfa silage. Chopped and wilted alfalfa (first cutting, 29% dry matter) was ensiled without (CON) or with L. casei (1 × 10 6 cfu g –1 fresh matter) (LC) or cellulase (20,000IU, 0.5% of fresh matter) (CE) for 56 days, then exposed to air for 3 days (PO). Greater ensiling quality was observed in LC and CE, which had lower pH and higher lactic acid content than CON at 56 days of ensiling and 3 days post-oxygen exposure. Air exposure was associated with decreased lactic acid concentrations and increased yeast and mold counts in all silages. SEM showed that the structure of leaf epicuticular wax crystals were intact in fresh alfalfa, totally decomposed in CON silage, and partly preserved in CE and LC silage. Gas chromatography mass spectrometry revealed that 196 metabolites and 95 differential concentration were present in the 3 days air exposure samples. Most of these metabolites, mainly organic acids, polyols, ketones, aldehydes, are capable of antimicrobial activity. The bacterial communities were obviously different among groups and Lactobacillus developed to a dominant status in all silages. Lactobacillus became dominant in bacterial communities of LC and CE silages from days 7 to 56, and their relative abundances reached 94.17–83.93% at day 56, respectively. For CON silage, until day 56, Lactobacillus dominated the bacterial community with abundance of 75.10%. After 3 days of oxygen exposure, Lactobacillus and Enterococcus were predominant in CON, and Lactobacillus remained dominant in LC and CE silages. The results indicated that, compared to untreated silages, L. casei could be a priority inoculant for alfalfa silage to boost Lactobacillus abundance and improve fermentation quality. Our high-throughput sequencing and gas chromatography mass spectrometry results provide a deep insight into the bacterial community and metabolites in alfalfa silage.

Effect of Homo-Fermentative Lactic Acid Bacteria Inoculants on Fermentation Characteristics and Bacterial and Fungal Communities in Alfalfa Silage

Abstract:We evaluated the effects of a homo-fermentative lactic acid bacteria (homo-LAB) inoculant on the fermentation and microbial communities of alfalfa ensiled at two dry matter (DM) contents of 38 and 46% DM. At both DMs, alfalfa was treated or not with an inoculant containing Pediococcus acidilactici, Enterococcus faecium and Lactobacillus plantarum at a targeted application rate of 165,000 cfu/g of fresh weight and stored for 3, 30 and 60 days. Treatment with the inoculant resulted in a lower drop in pH and, in general, higher lactic acid and lower acetic acid when applied to medium DM silage. For the four most abundant microbial genera, increased abundances of Bacteroides and Lactobacillus (p < 0.05), as well as decreased abundances of Muribaculaceae were observed in high DM and inoculated silages. The abundance of Prevotellaceae-UCG-001 was lower in medium DM control silages than in high DM control silages. Inoculation and DM affected abundances of Vishniacozyma (p < 0.05). Increased abundances of Vishniacozyma, as well as decreased abundances of Leucosporidium were observed in medium DM-inoculated silages. Changes in the relative abundance (RA) of the main populations of bacteria and yeasts did explain the fermentation and nutrition differences among treatments.

Abstract:Ensiling density has significant importance for the quality and preservation of silage. Appropriate ensiling density could improve the nutritional value, extend the storage time of silage and reduce the risk of mold and spoilage. This work aimed to evaluate the effect of ensiling densities on the fermentation quality and aerobic stability of rice straw. The rice straw was obtained after threshing, then chopped and ensiled into a 10 L laboratory silo with three ensiling densities (high density at 700 g/L, medium density at 600 g/L and low density at 500 g/L). Five silos per density were opened after 3, 5, 7, 14, 30 and 60 days of ensiling, and then, the fermentation quality and aerobic stability were analyzed. During ensiling, high density had the highest lactic acid content, and the lowest pH and ammonia nitrogen. There was no difference (p > 0.05) in the propionic acid, butyric acid and ethanol contents among all silage, and the contents of propionic acid and butyric acid were trace amounts. On day 60 of ensiling, the Flieg’s point of high density and medium density were higher than the low density. During aerobic exposure, the continuous lactic acid decrease and pH increase were observed in all silage. The aerobic bacteria and yeasts count in the high density and medium density were lower than that in the low density. The aerobic stability of the high density (26 h) and the medium density (24 h) were higher than that of the low density (13 h). It was suggested that if the ensiling density is higher than 600 g/L, it could effectively improve the fermentation quality and aerobic stability of rice straw.

Abstract:This study aimed to explore the changes in the microbial community on the silage material surface and during the ensiling process of whole-plant maize in different regions. Whole-plant maize silages were sampled in Ziyun, Guanling, and Weinning counties within warm and humid climate areas in southern China. Silages were sampled at 0, 2, 5, 10, 20, and 45 days during ensiling. The nutritional components, fermentation properties, and microbiomes were examined to evaluate the influence of sampling area and fermentation time on the quality of silage. The results showed that the pH values of all silages significantly decreased (<4.2 at ensiling day 2) during fermentation and all silages achieved satisfactory fermentation at 45 days. Butyric acid was not detected during ensiling, and the contents of acetic acid and ammonia nitrogen in the final silages were below 6 g/kg DM and 50 g/kg total nitrogen, respectively. Weissella was the dominant epiphytic bacteria of raw material in Ziyun and Weinning, while Lactobacillus was prevalent in Guanling. Lactobacillus dominated the ensiling process, and its abundance significantly increased with increasing fermentation time in the three groups. Lactobacillus was negatively correlated with pH of all silages ( p < 0.05) and positively correlated with lactic acid, propionic acid and acetic acid ( p < 0.05). Furthermore, the bacterial community was significantly correlated with environmental factors. Altitude had a highly positive correlation with the abundance of Stenotrophomonas , Chryseobacterium , and Massilia ( p < 0.01), while precipitation was negatively correlated with these bacteria. The humidity and average temperature significantly influenced the Lactobacillus and Weissella abundances of fresh whole-plant maize. During the ensiling process, the silages from three regions had similar bacterial dynamic changes, and the Lactobacillus formed and maintained good fermentation characteristics in whole-plant maize silage.

Abstract:OBJECTIVE: To develop the fermentation quality and chemical composition of alfalfa (Medicago sativa Lam.) silage, plants were inoculated with different lactic acid bacteria (LAB) strains at field 24 hours before harvest.METHODS: The treatment groups were as follow: silage without additive as a control and inoculated with each strains of Lactobacillus brevis (LS-55-2-2), Leuconostoc citerum (L-70-6-1), Lactobacillus bifermentans (LS-65-2-1), Lactobacillus plantarum (LS-3-3) and Lactobacillus plantarum (LS-72-2). All the silages were stored at 25 ºC. Parameters such as pH, microorganism and volatile fatty acid contents, crude protein, neutral detergent fiber, acid detergent fiber, net gas, metabolizable energy, organic matter digestibility, dry matter intake and relative feed value were measured to determine fermentation quality, chemical compositions and relative feed value of alfalfa silages.RESULTS: Significant differences were found among the control and treated groups in terms of pH and microorganism contents at all opening times and crude protein, net gas, metabolizable energy and organic matter digestibility of final silage. The pH values ranged from 4.70 to 5.52 for all treatments and control silage had the highest value of overall treatments at T75d silages. Volatile fatty acid of silages was not influenced significantly by inoculations. However, lactic acid content of L. bifermentans (LS-65-2-1) was higher than the other treatments. The highest metabolizable energy and organic matter digestibility were recorded from L. citerum (L-70-6-1) inoculation. In addition, no significant differences were found among treatments in terms of neutral detergent fiber, acid detergent fiber, dry matter intake and relative feed value.CONCLUSION: Among the treated lactic acid bacteria isolates, Lactobacillus bifermentans came into prominence especially in terms of organic acid composition and quality characters of silages.

Abstract:The aim of this study was to gain deeper insights into the dynamics of fermentation parameters and the bacterial community during the ensiling of high-moisture alfalfa. A commercial lactic acid bacteria (YX) inoculant was used as an additive. After 15 and 30 days of ensiling, the control silage (CK) exhibited a high pH and a high concentration of ammoniacal nitrogen (NH3-N); Enterobacter and Hafnia-Obesumbacterium were the dominant genera. At 60 d, the pH value and the concentration of NH3-N in CK silage increased compared with 15 and 30 d, propionic acid and butyric acid (BA) were detected, and Garciella had the highest abundance in the bacterial community. Compared with CK silage, inoculation of YX significantly promoted lactic acid and acetic acid accumulation and reduced pH and BA formation, did not significantly reduce the concentration of NH3-N except at 60 d, and significantly promoted the abundance of Lactobacillus and decreased the abundance of Garciella and Anaerosporobacter, but did not significantly inhibit the growth of Enterobacter and Hafnia-Obesumbacterium. In conclusion, high-moisture alfalfa naturally ensiled is prone to rot. Adding YX can delay the process of silage spoilage by inhibiting the growth of undesirable microorganisms to a certain extent.

Abstract:The objectives of this study were to investigate the adaptation and competition of Lactobacillus plantarum, Pediococcus pentosaceus and Enterococcus faecalis inoculated in alfalfa silage alone or in combination on the fermentation quality, dynamics of bacterial community, and their functional shifts using single‐molecule real‐time (SMRT) sequencing technology. Before ensiling, alfalfa was inoculated with L. plantarum (Lp), P. pentosaceus (Pp), E. faecalis (Ef) or their combinations (LpPp, LpEf, LpPpEf) and sampled at 1, 3, 7, 14 and 60 days. After 60‐days fermentation, the Lp‐, Pp‐ and LpPp‐inoculated silages had lower pH but greater concentrations of lactic acid were observed in Pp, LpEf and LpPpEf‐inoculated silages. The inoculants altered the keystone taxa and the bacterial community dynamics in different manners, where L. plantarum, Weissella cibaria and L. pentosaceus dominated the bacterial communities after 14 days‐fermentation in all treatments. The silages with better fermentation quality had simplified bacterial correlation structures. Moreover, different inoculants dramatically changed the carbohydrate, amino acid, energy, nucleotide and vitamin metabolism of bacterial communities during ensiling. Results of the current study indicate that effect of different inoculants on alfalfa silage fermentation was implemented by modulating the succession of bacterial community, their interactions and metabolic pathways as well during ensiling.

Abstract:Fresh alfalfa is difficult to ensile successfully because of its high moisture content and greater susceptibility to spoilage by Clostridia, Bacilli or Enterobacter. In this study, we evaluated the effects of mixing high-moisture alfalfa with peanut vine in different proportions on the bacterial communities and fermentation characteristics of silage. The high-moisture alfalfa and peanut vine were freshly mixed at ratios of 10:0 (CK), 8:2 (TI), 7:3 (T2), 6:4 (T3) and 5:5 (T4), respectively. For each treatment, silos (25 × 35 cm) were anaerobically fermented in darkness at room temperature and analyzed after 45 days. The results showed that the CK silage was weakly fermented, as indicated by a low lactic acid concentration, a high pH value, and high levels of propionic acid (CP), butyric acid (BA) and ammonia nitrogen (NH3-N). As the proportion of peanut vine in the mixture increased, the pH level decreased, and levels of BA, propionic acid, NH3-N, CP, nonprotein nitrogen and soluble protein also declined (p < 0.05), while the lactic acid concentration increased and levels of neutral detergent fiber (NDF) and water-soluble carbohydrates (WSCs) also rose (p < 0.05). A protein component analysis of silage mixtures using the Cornell Net Carbohydrate and Protein System (CNCPS) showed that the content of the nonprotein nitrogen component (PA) decreased when the proportion of peanut vine increased, whereas the content of rapidly degraded protein (PB1) increased. Mixing with peanut vine also influenced the distribution of the bacterial community. Compared with the CK silage, the relative abundances of Enterococcus, Garciella and Anaerosporobacter in T2, T3 and T4 were significantly lower, while the relative abundance of Lactobacillus was significantly higher. In the T2, T3 and T4 groups, Garciella and Anaerosporobacter were not detected. In summary, in this study, we ensiled high-moisture alfalfa, which was weakly fermented. We found that mixing with peanut vine improved fermentation quality and optimized the structure of the bacterial community. Therefore, to improve the fermentation quality and nutritional value of silage, high-moisture alfalfa should be ensiled with at least 30% peanut vine.

Abstract:The objective of this study was to investigate the effects of cellulolytic lactic acid bacteria on the lignocellulose degradation, sugar profile and lactic acid fermentation of high-moisture alfalfa during anaerobic bioprocessing in low-temperature seasons. Last cut and low dry matter (DM) alfalfa was ensiled with five additives, commercial cellulase (EN), a combination of Lactobacillus plantarum and commercial cellulase (LPEN), a combination of engineered Lactococcus lactis strains (HT2) containing cellulase gene, Enterococcus faecium Y83 (EF83), or Enterococcus faecalis JF85 (EF85), and without additives, as the control, with four replicates each. After anaerobic preservation in a silo from late fall through winter (3 to 20 °C) for 140 days, the ensiled alfalfa was sampled and analysed. Compared with the control, EF85 increased the relative contents of neutral detergent fibre (NDF), acid detergent fibre (ADF), and cellulose and decreased contents of water-soluble carbohydrates (WSCs) and lactic acid, and the conversion of WSCs into lactic acid. Consequently, lactic acid fermentation was decreased in EF85. Compared with the control, supplementation with EN, EF83, HT2, and LPEN decreased contents of NDF, ADF, cellulose, and hemicellulose and increased the contents of WSCs, disaccharide, xylose, and arabinose; interestingly, compared with EN, supplementation with EF83, HT2 and LPEN increased the lactic acid content and the ratio of lactic acid to acetic acid as indicated by strong homo-lactic acid fermentation. Furthermore, HT2 increased the lactic acid concentration to a maximum of 72.2 g/kg DM and had a higher conversion of WSCs into lactic acid than EF83 (36.3%) and LPEN (17.9%), indicating that HT2 had a greater ability to ferment lignocellulose of alfalfa into lactic acid. Therefore, HT2 could enhance the lactic acid production of alfalfa during ensiling in low-temperature seasons.
