Abstract:OBJECTIVE To screen the suitable resin for separation and purification of total flavones from Radix tetrastigmae,and to establish the optimum parameters of separation and purification process.METHODS The adsorption capacities and desorption ratios of ten selected macroporous resins were evaluated with UV method.Suitable resins were chosen to study the performance of static-dynamic adsorption and desorption,and to optimize the process parameters.RESULTS HPD826 resin showed advantageous adsorption and desorption properties with thermodynamic parameter according with the Freundlich equation.The optimal conditions were determined as follows: for adsorption,the flow speed was 2 BV·h-1;for desorption,wash the adsorbate-laden column first with 2 BV water,then with 3 BV 70% ethanol-water.Under this condition,the content of total flavones increased by 2.44-fold.CONCLUSION It is feasible to separate and purify Radix tetrastigmae total flavones with HPD826 resin.
In Vitro and Vivo Antioxidant Activities of Daylily Flowers and the Involvement of Phenolic Compounds

Abstract:Daylily (Hemerocallis fulva Linn.) flowers were hot air-dried and freeze-dried after harvest. Antioxidant properties of water and ethanol extracts prepared from these dried flowers were evaluated in terms of total antioxidant activity, reducing capacity, and metal chelating activity. Extracts from daylily flowers exhibited strong antioxidant activity. Ethanol was more efficiency to extract antioxidants than water, and freeze-drying preserved higher activities than air-drying. Rutin, (+)-catechin, and gallic acid were identified in the extracts by HPLC, and were highly related to the antioxidant activities. The antioxidant activity was further evaluated by feeding mice with ethanol extract from freeze-dried daylily flowers for 60 days. The results demonstrated that the extract at dosage of 40-225 mg/100 g significantly increased the activity of SOD (superoxide dismutase) and reduced the lipid peroxidation in both blood and liver of rat.

Abstract:The relationship between market structure and advertising has been extensively studied, but has generated sharply opposing theoretical predictions, as well as inconclusive empirical findings, likely because of severe endogeneity concerns. We exploit the 2008 merger of Miller and Coors in the U.S. brewing industry to examine how changes in local concentration affect firms’ advertising behavior. Well-established regional preferences over beer brands, and the sharp increase in concentration from the merger, make this an excellent setting to analyze this question. We find a significant positive effect of local market concentration on advertising expenditures: a 100-point increase in the Herfindahl–Hirschmann Index measure of concentration increases advertising per capita by about 5%. Abstract:Euphorbia hirta has anti-inflammatory effects in traditional medicine, but its anti-inflammatory mechanism has not been explored at the cellular and molecular levels. To unravel these mechanisms, the main active components in the 65 and 95% ethanol extracts of Euphorbia hirta were first identified by UPLC-Q-TOF/MS. Subsequently, potential anti-inflammatory targets and signaling pathways were predicted using network pharmacology and experimentally validated using RT-PCR and flow cytometry in a lipopolysaccharide (LPS)-induced inflammation model of RAW264.7 cells. The results revealed flavonoids as the key active components. Network pharmacology uncovered 71 potential anti-inflammation targets, with a protein-protein interaction (PPI) network highlighting 8 cores targets, including IL-6, TNF, NFκB and Nrf2 et al. Furthermore, Euphorbia hirta exerts anti-inflammation effects through modulation of Nrf2 and NF-κB signaling pathways. Specifically, the 65% ethanol extract of Euphorbia hirta (EE65) and quercitrin (HPG) exerted anti-inflammatory activity by inhibiting the expression of inflammatory genes associated with the NF-κB signaling pathway, whereas baicalein (HCS) suppressed cellular inflammation by promoting Nrf2-mediated antioxidant gene expression and enhancing apoptosis of inflammatory cells. The results of the study suggest that Euphorbia hirta has potential for the development of anti-inflammatory drugs.

Abstract:The effects of purification methods and its hypolipidemic function on the total flavonoids of Aurea Helianthus flower were investigated. Liquid-liquid extraction of ethanol extract from Aurea Helianthus flower was carried out by using different polar solvents. The extract with the highest total flavonoid content was selected, and the optimal conditions for purification of total flavonoids were determined by purification with macroporous resin. The human digestive environment was simulated in vitro, and the binding ability of different flavonoid samples to three kinds of cholate was compared. The results showed that the purity of total flavonoids in ethanol extract was 27.8%, the purity of total flavonoids in ethyl acetate extract was 46.4%, and the purity was increased by 18.6%. Subsequent purification with AB-8 macroporous resin; loading of total flavonoids at a concentration of 5.5 mg/mL, flow rate of 1.5 mL/min, 110 mL; use of 75% ethanol, 80 mL as eluent at a flow rate of 1.5 mL The elution at /min resulted in a total flavonoid purity of 83.5 % and an increase of 37.1%, and a good purification effect was obtained. The binding rate of total flavonoids purified by AB-8 macroporous resin to sodium taurocholate, sodium glycocholate and sodium cholate was 88.2%, 73.2% and 75.8 %, respectively. The binding ability was the strongest, and the others were ethyl acetate. Extract, ethanol extract. The purity of total flavonoids showed a good correlation with the binding capacity of cholate, and the correlation coefficient was between 0.963 and 0.988. The total flavonoids of Aurea Helianthus flower have good bile acid binding ability and can be used as the focus of natural hypolipidemic substances.

Abstract:The health benefits and promising medical treatment potential of total flavonoids from Eupatorium lindleyanum DC. (TFELDC) have been recognized. The process parameters of extracting total flavonoids from Eupatorium lindleyanum DC. by ultrasonic–microwave synergistic extraction (UMSE) were optimized, and they were purified by AB‐8 macroporous resin in the current study. In addition, the antioxidant and enzyme inhibitory activities of the purified TFELDC (PTFELDC) were evaluated. The results showed that the optimal parameters of UMSE were as follows: ethanol volume fraction 71.5%, L/S ratio 12.2 ml/g, microwave power 318 W, and extraction time 143 s. After TFELDC were purified by AB‐8 macroporous resin, the total flavonoid contents of PTFELDC increased from 208.18 ± 1.60 to 511.19 ± 3.21 mg RE/g FDS. Compared with TFELDC, the content of total flavonoids in PTFELDC was increased by 2.46 times. The antioxidant activities of PTFELDC were assessed using DPPH radical, superoxide anion radical, reducing power, and ferric reducing antioxidant power assays, and the IC50 values were found to be 37.13, 19.62, 81.22, and 24.72 μg/ml, respectively. The enzyme inhibitory activities of PTFELDC were measured using lipase, α‐amylase, α‐glucosidase, and acetylcholinesterase assays with the IC50 values 1.38, 2.08, 1.63, and 0.58 mg/ml, respectively. By comparing with their positive controls, it was found that PTFELDC had good antioxidant activities, and lipase, α‐amylase, and α‐glucosidase inhibitory activities, However, the acetylcholinesterase inhibitory activity was relatively weaker. These results suggested that PTFELDC have a promising potential as natural antioxidant, antilipidemic, and hypoglycemic drugs used in functional foods or pharmaceuticals.

Optimization of Extraction of Compound Flavonoids from Chinese Herbal Medicines Based on Quantification Theory and Evaluation of Their Antioxidant Activity

Abstract:Plant-derived flavonoids have been attracting increasing research interest because of their multiple health promoting effects, where numerous investigations were carried out on the optimization of extraction and bioactivities. This study aims to optimize the extraction process of compound flavonoids (CFs) from Chinese herbal medicines and detect their antioxidant activity in vitro. CFs were extracted from the raw materials named "medicine food homology," composed of hawthorn, lotus leaf, tartary buckwheat, cassia seed, Lycium barbarum, and Poria cocos in a mass ratio of 4 : 2 : 2 : 1.5 : 1 : 1. L9 (34) orthogonal design, level effect and engineering average estimation, and quantification theory were utilized to improve the extraction method of CFs, and the predictive model for CFs yield was constructed. The 2,2ʹ-diphenyl-1-picrylhydrazyl (DPPH), 2,2ʹ-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), hydroxyl radical scavenging rate, and reducing power of CFs were measured. The highest CFs yield was obtained under the following extraction condition: liquid-solid ratio of 35 : 1 mL/g, extraction temperature of 75°C, extraction duration of 75 min, and extraction mode enzyme-assisted extraction. The forecasted yield was 37.62%. The result was accurate and the established prediction equation was reliable (R = 0.95). The antioxidant activity of CFs was significantly positively correlated with the concentration from 0.05 to 0.4 mg/mL. The DPPH, ABTS, hydroxyl radical scavenging abilities, and the reducing power of CFs were 81.82 ± 1.75%, 49.35 ± 0.09%, 89.78 ± 0.66%, and 0.232 ± 0.001 at the concentration of 0.4 mg/mL, respectively. CFs could be exploited as natural antioxidants in pharmaceuticals and functional foods.

Abstract:The aim of this study was to investigate the antioxidant and free-radical scavenging activity of extract and fractions from various parts of Elsholtzia ciliata. The inflorescences, leaves, stems and roots of E. ciliata were extracted separately and two phenolic component enrichment methods: ethyl acetate-water liquid-liquid extraction and macroporous resin adsorption-desorption, were adopted in this study. The antioxidant activities of water extracts and fractions of E. ciliata were examined using different assay model systems in vitro. The fraction root E (purified by HPD300 macroporous resin) exhibited the highest total phenolics content (497.2 ± 24.9 mg GAE/g), accompanied with the highest antioxidant activity against various antioxidant systems in vitro compared to other fractions. On the basis of the results obtained, E. ciliata extracts can be used potentially as a ready accessible and valuable bioactive source of natural antioxidants.
Purification and Identification of Flavonoid Molecules from Rosa setate x Rosa rugosa Waste Extracts and Evaluation of Antioxidant, Antiproliferative and Antimicrobial Activities

Abstract:Rosa setate x Rosa rugosa is widely used in the essential oil industry and generates large amounts of waste annually. The purpose of this research is the recycling of bioactive flavonoids from rose waste biomass to develop high-value products. Resin screening and adsorption/desorption dynamic analysis showed that HP20 resin was suitable to purify the flavonoids from R. setate x R. rugosa waste extracts. Under the optimal enrichment process, the product had a 10.7-fold higher purity of flavonoids with a satisfactory recovery of 82.02%. In total, 14 flavonoids were identified in the sample after purification by UHPLC-QTOF-MS. Moreover, the DPPH and ABTS assays revealed that the flavonoids-purified extracts exhibited higher antioxidant activities than the crude extracts. Meanwhile, the purified extracts presented stronger antiproliferative activity against HepG2, Caco-2, MCF-7 and A549 cell lines. The bacteriostatic effects of the purified extracts against four bacteria (Staphylococcus aureus (S. aureus), Escherichia coli (E. coli), Staphylococcus epidermidis (S. epidermidis), Pseudomonas aeruginosa (P. aeruginosa)) and yeast (Candida albicans (C. albicans)) were stronger compared with the crude extracts. It was concluded that flavonoids-enriched extracts from R. setate x R. rugosa waste had the potential to be applied in functional food and pharmaceutical industries.

Abstract:Macroporous resin was used to enrich flavonoids in the ethyl acetate extract of Artemisia Selengensis Turcz. Based on a single factor experiment, the enrichment process was optimized using the response surface method. The optimal parameters of the enrichment process were a sample concentration of 0.3 mg/mL, a loading rate of 1 mL/min, an elution flow rate of 2 mL/min, and a total flavonoid content of 155.38 ± 0.97 mg/g. The flavonoids enriched by AB‐8 macroporous resin demonstrated significant scavenging activities against DPPH, ABTS+, and hydroxyl free radicals, and also exhibited certain inhibitory effects on α‐amylase and α‐glucosidase. Among them, the scavenging ability of the flavonoids enriched by AB‐8 macroporous resin on hydroxyl free radical (IC50 = 30.31 ± 1.92 μg/mL) was the closest to Vc, and the inhibitory effect on α‐glucosidase (IC50 = 16.19 ± 1.35 μg/mL) was the best. These findings confirmed the potential of Artemisia Selengensis Turcz. is a natural antioxidant and hypoglycemic drug.

Adsorption of Myricetrin, Puerarin, Naringin, Rutin, and Neohesperidin Dihydrochalcone Flavonoids on Macroporous Resins

Abstract:The adsorption properties (equilibrium, kinetics, and column breakthrough) of five model flavonoids (myricetrin, puerarin, naringin, rutin and neohesperidin dihydrochalcone) on selected macroporous resins were investigated in order to identify a suitable resin adsorbent for effective separation and purification of flavonoids from the extracts of herbal plants. It was observed that the resins with a low polarity and a high specific surface area have high adsorption capacities for all five flavonoids. Both the Langmuir and Freundlich isotherm equations correlate well the adsorption equilibrium data of the five flavonoids on four selected resins, and adsorption enthalpy, entropy, and free energy of the five flavonoids on HPD300 resin were calculated from the adsorption isotherms by the Freundlich equation constants. The pseudo-second-order adsorption rate equation fits the kinetic data on four selected resins better than the pseudo-first-order adsorption rate equation, and the initial adsorption rates were calculated and discussed. The HPD300 resin was selected as the most promising adsorbent for a preliminary separation and purification of flavonoids because of its excellent adsorption/desorption properties including high adsorption rates for all five flavonoids. The adsorption breakthrough experiment with a synthetic flavonoid mixture solution on the HPD300 resin further confirmed that the HPD300 resin can separate the five flavonoids effectively, especially for purifying neohesperidin dihydrochalcone from the flavonoid mixtures.

Abstract:This study was focused on extraction, radical scavenging activities, and chemical composition identification of total flavonoids in sunflower (Helianthus annuus L.) receptacles (TFSR). We investigated the optimal extract parameters of TFSR using response surface methodology. The highest yield of TFSR was 1.04% with the ethanol concentration 58%, the material-to-liquid ratio 1:20 (v/w), the extraction time 2.6 h, and the extraction temperature 67 °C. The results of radical scavenging activities showed that ethyl acetate fraction (EAF) was the strongest by using 2-diphenyl-1-picrylhydrazyl (DPPH), 2, 2’-azino-bis (3-ethylbenzo thiazoline-6-sulfonic acid) (ABTS) and iron ion reducing analysis. The EAF had the highest flavonoids contents. Four fractions A, B, C and D were enrichment from EAF by polyamide resin. Fraction B had the highest flavonoids content. Thirteen chemical components of flavonoids in fraction B were first identified by Ultimate 3000 Nano LC System coupled to a Q Exactive HF benchtop Orbitrap mass spectrometer (UHPLC-HRMS/MS). Among of the thirteen chemical components, isoquercetin and daidzein were identified accurately by comparing with standard samples. Radical scavenging analysis showed that isoquercetin and EAF had strong activities. Therefore, sunflower receptacles can be used as a source of natural flavonoids. TFSR as a natural radical scavenger has potential applications in pharmaceutical industry.

Abstract:The flavonoids from Perilla leaves were extracted using flash extraction assisted by ultrasonic extraction with ethanol. Subsequently, macroporous resin was employed for the isolation and purification of these flavonoids, followed by an investigation into their antioxidant activity. The process conditions for the extraction of flavonoids from Perilla leaves were designed and optimized using a one-way experiment combined with a response surface methodology. The optimal extraction conditions were determined as follows: the liquid–solid ratio was 20:1, ethanol volume fraction of 60%, ultrasound temperature of 60 °C, ultrasound time of 10 min and flash evaporation time of 60 s. The optimal extraction rate of flavonoids is 9.8 mg/g. In terms of separation and purification, a high-performance macroporous resin (HPD450 resin) with high purification efficiency was selected through static analysis and adsorption experiments. The optimal enrichment conditions were as follows: loading concentration of 0.06 mg/mL, optimal loading concentration of 20 mL, elution concentration of 70% and 76 mL, providing a reference for the further development and utilization of Perilla leaf flavonoids.

Abstract:The effect of tablet thickness on the hydration kinetics of types I and II theophylline anhydrate tablets at 95% relative humidity and 35 degrees C was studied with various kinetic equations. Samples of 1- and 2-cm-diameter tablets (1 g) were compressed at 1000 kg/cm2. Types I and II tablets expanded by 11-17% in volume during hydration to the monohydrate. The thickness expansion of all tablets exceeded the diameter expansion. After the hygroscopicity test, the final expansion ratio of type I tablets was more than that of type II tablets. The hydration kinetics analysis suggested that the hydration of theophylline anhydrate tablets proceeds as follows: Hydration of the 1-cm-diameter type I tablet followed the two-dimensional phase boundary equation, and that of the 1-cm-diameter type II tablet followed the three-dimensional phase boundary equation, but those of the 2-cm-diameter types I and II tablets followed the equation for two-dimensional growth of nuclei. The hydration kinetics of tablets does not depend on the nature of bulk powder but on geometrical factors (tablet thickness and porosity).
Extraction and purification of antioxidative flavonoids from Chionanthus retusa leaf

Abstract:In this work, flavonoids from the leaves of Chionanthus retusa were extracted using alcohol, and the extraction yield was optimized by single-factor and orthogonal experiments. Then, the extracted solution with flavonoids was purified via macroporous resin by elution with different concentrations of ethanol. The antioxidative activity of total flavonoid in purified extracted solution was evaluated by detecting its ability to scavenge DPPH free radicals. The results demonstrated that ethanol with a concentration of 60%, ultrasonic power of 140 W, liquid–solid ratio of 25:1 ml g −1 , and water-bath temperature of 80°C were the optimal conditions for the extraction of total flavonoids from C. retusa leaf, achieving a yield of 121.28 mg g −1 . After purification by macroporous resin using different concentrations of ethanol, the highest content of total flavonoids (88.51%) in the extracted solution can be obtained with the 50% ethanol eluant. The results of scavenging DPPH free radicals suggest that the purified flavonoids in the 50% ethanol eluant had the best antioxidant capacity over the flavonoids in other ethanol eluants. In addition, it is confirmed the antioxidant capacity of the extractives was associated with the content of total flavonoids and kinds of flavonoids. These results may provide a feasible pathway to make full use of total flavonoids from C. retusa leaf.

Abstract:In the present work, the ultrasonic-assisted extraction (UAE) of total flavonoids (TF) from Pteris cretica L. was optimized by response surface methodology (RSM) on the basis of a single-factor experiment. The optimized UAE parameters were as follows: Ethanol concentration 56.74%, extraction time 45.94 min, extraction temperature 74.27 °C, and liquid/solid ratio 33.69 mL/g. Under the optimized conditions, the total flavonoids yield (TFY) was 4.71 ± 0.04%, which was higher than that obtained by heat reflux extraction (HRE). The extracts were further analyzed by HPLC, and five major flavonoids, including rutin, quercitrin, luteolin, apigenin, and luteolin-7-O-glucoside, were identified and quantified. Furthermore, the results of the antioxidant test showed that the TF extract obtained under optimized UAE conditions exhibited good 2,2-diphenyl-1-picrylhydrazyl radical (DPPH•) and 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical (ABTS+•), nitric oxide radical (NO•) scavenging activities, and ferrous ion (Fe2+) chelating capacity, with IC50 values of 74.49, 82.92, 89.12, and 713.41 μg/mL, respectively. Results indicated that the UAE technique developed in this work was an efficient, rapid, and simple approach for the extraction of flavonoids with antioxidant activity from P. cretica.

Abstract:Hemerocallis fulva is a medical and edible plant. In this study, we optimized the ultrasound-assisted extraction (UAE) process of extracting flavonoids from Hemerocallis fulva leaves by single-factor experiments and response surface methodology (RSM). The optimum extraction conditions generating the maximal total flavonoids content was as follows: 70.6% ethanol concentration; 43.9:1 mL/g solvent to sample ratio; 61.7 °C extraction temperature. Under the optimized extraction conditions, the total flavonoid content (TFC) in eight Hemerocallis fulva varieties were determined, and H. fulva (L.) L. var. kwanso Regel had the highest TFC. The cytotoxicity of the extract was studied using the Cell Counting Kit-8 (CCK-8 assay). When the concentration was less than 1.25 mg/mL, the extract had no significant cytotoxicity to HaCaT cells. The antioxidant activity was measured via chemical antioxidant activity methods in vitro and via cellular antioxidant activity methods. The results indicated that the extract had a strong ABTS and •OH radical scavenging activity. Additionally, the extract had an excellent protective effect against H2O2-induced oxidative damage at a concentration of 1.25 mg/mL, which could effectively reduce the level of ROS to 106.681 ± 9.733% (p < 0.001), compared with the 163.995 ± 6.308% of the H2O2 group. We identified five flavonoids in the extracts using high-performance liquid chromatography (HPLC). Infrared spectroscopy indicated that the extract contained the structure of flavonoids. The results showed that the extract of Hemerocallis fulva leaves had excellent biocompatibility and antioxidant activity, and could be used as a cheap and potential source of antioxidants in the food, cosmetics, and medicine industries.

Abstract:It is known that Flavonoid is abundant in Eucommia ulmoides pollen. Flavonoid has various biological functions. Based on this, the study aimed to further exploit the flavonoid from Eucommia ulmoides pollen, optimizing the microwave extraction process of flavonoid from Eucommia ulmoides pollen by means of single factor experiment combined with response surface analysis, investigating the scavenging ability of the extract to DPPH free radicals, detecting its antibacterial properties on Escherichia coli and golden yellow staphylococcus and evaluating inhibitory effect of human breast cancer cells (MCF‐7). The results showed that the dry Eucommia ulmoides pollen was extracted under the condition of 74% (v/v) ethanol concentration, solid‐to‐liquid (S/L) ratio of 1: 42 (g: mL), microwave power 500 W and extraction time of 165 s, yielding 3.28 g flavonoid per 100 g pollen, which is closely consistent with predicted value. The IC50 of flavonoid for scavenging DPPH radical was 0.04 mg/mL. The extract has strong inhibitory effect on Escherichia coli and Staphylococcus aureus. At the same time, the extract had a significant inhibitory effect on the growth and proliferation of MCF‐7 cells in a dose‐dependent and time‐dependent manner, it has no obvious toxic side effects on fibroblasts (L929) at low doses.

Optimization of Preparative Separation and Purification of Total Flavonoids from Radix Puerariae by Macroporous Resin Method

Abstract:Aim To screen the optimum macroporous resin and conditions for the isolation and purification of flavonoids from Radix Puerariae. Methods The static and dynamic adsorption/desorption methods were used, and the separation and purification process was evaluated by measuring the concentration of total flavonoid in the fractions with UV spectrophotometer. Results The SP70 macroporous resin was the most effective compared with other macroporous resins. The optimum conditions were screened, which were 0. 5 g· mL- 1 corresponding to crude drug for concentration of extract, pH 5 - 6, and appended 60 times the volume of the resin bed (BV) with the adsorption speed 2 BV· h - 1,and the volume of aq. 70％ (V/V) ethanol as eluant was 5 BV with desorption speed 2 BV·h-1. By this method, the final contents of total flavonoids exceeded 80%. Conclusion The SP70 macroporousresin is the most effective one for large-scale isolation and purification of flavonoids from Radix Pueraria, which meets industrial needs.
