Abstract:Antimicrobial resistance (AMR) has arisen due to antibiotic overuse and misuse. Antibiotic resistance renders standard treatments less effective, making it difficult to control some infections, thereby increasing morbidity and mortality. Medicinal plants are attracting increased interest as antibiotics lose efficacy. This study evaluates the antibacterial activity of solvent extracts prepared using Terminalia bellirica and Terminalia chebula fruit against six bacterial pathogens using disc diffusion and broth microdilution assays. The aqueous and methanol extracts of T. bellirica and T. chebula showed substantial zones of inhibition (ZOIs) against Staphylococcus aureus and methicillin-resistant S. aureus (MRSA). The activity against those bacteria was strong, with minimum inhibitory concentrations (MIC) ranging from 94 μg/mL to 392 μg/mL. Additionally, the T. bellirica methanolic extract showed noteworthy antibacterial activity against Escherichia coli and an extended spectrum β-lactamase (ESBL) E. coli strain (MIC values of 755 μg/mL for both). The aqueous T. bellirica and T. chebula extracts also inhibited Klebsiella pneumoniae growth (MIC values of 784 μg/mL and 556 μg/mL, respectively). The corresponding methanolic extracts also inhibited ESBL K. pneumoniae growth (MIC values of 755 μg/mL and 1509 μg/mL, respectively). Eighteen additive interactions were observed when extracts were combined with reference antibiotics. Strong antagonism occurred when any of the extracts were mixed with polymyxin B. Liquid chromatography-mass spectroscopy (LC-MS) analysis of the extracts revealed several interesting flavonoids and tannins, including 6-galloylglucose, 1,2,6-trigalloyl-β-D-glucopyranose, 6-O-[(2E)-3-phenyl-2-propenoyl]-1-O-(3,4,5-trihydroxybenzoyl)-β-D-glucopyranose, propyl gallate, methyl gallate, sanguiin H4, hamamelitannin, pyrogallol, gallic acid, ellagic acid, chebulic acid, and chebuloside II. All extracts were nontoxic in brine shrimp assays. This lack of toxicity, combined with their antibacterial activities, suggests that these plant species may be promising sources of antibacterial compound(s) that warrant further study.

Abstract:Development of cheap and reliable diagnostics is pivotal in the control of infectious diseases. Currently, one of the best strategies to tackle infectious disease is the use of natural products from plant origin, due to the perception that long term use of western medicine induces severe complications and also to stop the prevalence of antibacterial infection. This study aims at substantiating the traditional use of selected medicinal plants with antibacterial claim for possible lead for the development of more potent drugs in the treatment of bacterial infections. The preliminary phytochemical content, antioxidant and antibacterial potential of the selected medicinal plants were evaluated using standard procedures. Thirtheen plants were obtained through questionnaire administration: Garcinia kola, Costus afer, Vitellaria paradoxa, Pycnanthus angolensis, Cola acuminata Acanthospermum hipida, Aloe vera, Euadenia trifoliate, Microglossa abzelii, Moringa oleifera, Bryophyllum pinnatum, Baphia nitida and Afromonium melegueta. Thin layer chromatography (TLC) profiling of the various selected plants were carried out using two solvent system consisting of n-Hexane and ethyl acetate in varying ratio (8:2 and 7:3). Eight (8) plants, which showed good chromatographical profiling with the used solvent, were selected for further experiment (Garcinia kola, Vitellaria paradoxa, Acanthospermum hispidum, Aloe vera, Microglossa abzelii, Baphia nitida, Moringa oleifera and Bryophyllum pinnatum). These plants were subjected to qualitative phytochemistry and five plants (Moringa oleifera, Vitellaria paradoxa, Baphia nitida, Bryophyllum pinnatum and Garcinia kola) selected for quantitative phyto-screening for alkaloids, flavonoids, saponins and tannins. The antibacterial screening was carried out using Agar well diffusion and minimum inhibitory concentration (MIC) against the following isolate Staphylococcus aureus, Escherichia coli, Salmonella typhi and Bacillus subtilis. The antioxidant potential of the selected five plants were evaluated using in vitro (DPPH and FRAP) and the best plant was used for in vivo (SOD and MDA) analysis. The phytochemistry of the 13 plants revealed the presence or absence of Alkaloids, saponins, tannins, flavonoids, terpenoids, cardiac glycosides, carbohydrates and protein in the selected plants, and the best five were selected for quantitative evaluation showed they all have good quantity of alkaloids, flavonoids, saponins and tannins. The plants exhibited mild to strong antibacterial activities, having an MIC value ranging from 2.5 to >20 μg/mL when compared to ciprofloxacin (control) that is 5μg/mL. The plants showed good free radical scavenging activities and efficient ferric reducing power in a concentration dependent manner and a good enzyme activity of the superoxide dismutase (SOD) at a concentration of 100 μ/mL with a reduction in malondialdehyde (MDA) production as a marker of lipid peroxidation. The various plants have potentials for development of drugs for the treatment of infectious diseases; and a potential for further evaluation of their antibacterial claim.
The antibacterial activity of extracts of nine plant species with good activity against Escherichia coli against five other bacteria and cytotoxicity of extracts

Abstract:BackgroundThe development of antibiotic resistant bacteria stems from a number of factors, including inappropriate use of antibiotics in human and animal health and their prolonged use as growth promoters at sub-clinical doses in poultry and livestock production. We were interested in investigating plants that could be useful in protecting humans or animals against diarrhoea. We decided to work on extracts of nine plant species with good activity against Escherichia coli based on earlier work in the Phytomedicine Programme. Leaves of nine medicinal plant species with high antibacterial activity against Escherichia coli were extracted with acetone and their minimal inhibitory concentration (MIC) values determined using a microplate serial dilution technique against Gram-positive (Staphylococcus aureus, Enterococcus faecalis and Bacillus cereus) and Gram-negative (Escherichia coli, Salmonella Typhimurium and Pseudomonas aeruginosa) bacteria. Bioautography was used to determine the number of bioactive compounds in each extract. In vitro safety of the extracts was determined using the 3-(4,5-dimethylthiazolyl-2)-2,5-diphenyltetrazolium bromide reduction assay on Vero cells.ResultsThe extracts were active against all the pathogens with average MICs ranging from 0.02 to 0.52 mg/ml. As expected E. coli was relatively sensitive, while E. faecalis and S. Typhimurium were more resistant to the extracts (average MICs of 0.28 mg/ml and 0.22 mg/ml respectively). Cremaspora triflora and Maesa lanceolata leaf extracts had higher activity than the other extracts against Gram-positive and Gram-negative pathogens with mean MICs of 0.07 mg/ml and 0.09 mg/ml respectively. Extracts of Maesa lanceolata and Hypericum roeperianum had the highest total antibacterial activity (TAA) at 1417 and 963 ml/g respectively. All extracts with the exception of that of Maesa lanceolata, Elaeodendron croceum and Calpurnia aurea had relatively low cytotoxicity with LC50 > 20 μg/ml. Cremaspora triflora had the best selectivity index (SI) against S. aureus and E. coli of 2.87 and 1.15 respectively. Hypericum roeperianum had a SI of 1.10 against B. cereus. Bioautography revealed 1–6 visible antimicrobial compounds that were generally non-polar.ConclusionsThere was a weak positive, but statistically non-significant correlation between the potency of the extracts and their cytotoxicity (R = 0.45, ρ > 0.05). The activity of the extracts on the test bacteria was in some cases not correlated with cytotoxicity, as shown by selectivity indices >1. This means that cellular toxicity was probably not due to compounds with antibacterial activity. Some of the extracts had a good potential for therapeutic use against the bacterial pathogens or for application in treating diarhoea. It does not appear that activity against E. coli is a good predictor of activity against Gram-negative rather than Gram-positive bacteria. Further investigation is in progress on C. triflora and H. roeperianum, both of which had promising activities and potential safety based on cytotoxicity.
Effect of certain bioactive plant extracts on clinical isolates of beta-lactamase producing methicillin resistant Staphylococcus aureus

Abstract:Ethanolic extracts and some fractions from 10 Indian medicinal plants, known for antibacterial activity, were investigated for their ability to inhibit clinical isolates of beta-lactamase producing methicillin-resistant Staphylococcus aureus (MRSA) and methicillin-sensitive S. aureus (MSSA). Synergistic interaction of plant extracts with certain antibiotics was also evaluated. The MRSA test strains were found to be multi-drug resistant and also exhibited high level of resistance to common beta-lactam antibiotics. These strains produced beta-lactamases, which hydrolyze one or other beta-lactam antibiotics, tested. The extract of the plants from Camellia sinensis (leaves), Delonix regia (flowers), Holarrhena antidysenterica (bark), Lawsonia inermis (leaves), Punica granatum (rind), Terminalia chebula (fruits) and Terminalia belerica (fruits) showed a broad-spectrum of antibacterial activity with an inhibition zone size of 11 mm to 27 mm, against all the test bacteria. The extracts from the leaves of Ocimum sanctum showed better activity against the three MRSA strains. On the other hand, extracts from Allium sativum (bulb) and Citrus sinensis (rind) exhibited little or no activity, against MRSA strains. The antibacterial potency of crude extracts was determined in terms of minimum inhibitory concentration (MIC) by the tube dilution method. MIC values, of the plant extracts, ranged from 1.3 to 8.2 mg/ml, against the test bacteria. Further, the extracts from Punica granatum and Delonix regia were fractionated in benzene, acetone and methanol. Antibacterial activity was observed in acetone as well as in the methanol fractions. In vitro synergistic interaction of crude extracts from Camellia sinensis, Lawsonia inermis, Punica granatum, Terminalia chebula and Terminalia belerica was detected with tetracycline. Moreover, the extract from Camellia sinensis also showed synergism with ampicillin.TLC of the above extracts revealed the presence of major phytocompounds, like alkaloids, glycosides, flavonoids, phenols and saponins. TLC-bioautography indicated phenols and flavonoids as major active compounds.

Abstract:INTRODUCTION Recently, there have been growing attention on the modification and optimisation of new extraction and quantification methods, caused by the lack of environmentally friendly methodologies for the extraction of phytochemicals from complex matrices. In the case of pharmaceutical compounds, not only the extraction procedure but also the analysis method should be efficient, precise, fast and easy. OBJECTIVES The essential pharmaceutical characteristics and trace concentration of withanolides led us to modify and optimise the previously reported extraction and quantification procedure for withaferin A (WA) as a candidate for withanolides. MATERIAL AND METHODS The WA from the air-dried aerial part of Withania somnifera Dunal. was extracted using a microwave-assisted extraction (MAE) technique. Four variables affecting the extraction procedure were optimised using the central composite design approach. The method of high-performance thin-layer chromatography assay was validated and applied for the quantification of each experiment. RESULTS The optimum values of factors were: extraction time (150 s), extraction temperature (68°C) and 17 mL of methanol : water in the ratio 25 : 75 as extracting solvent. The solvent system consisted of ethyl acetate : toluene : formic acid : 2-propanol (7.0 : 2.0 : 0.5 : 0.5, v/v/v/v), and densitometric scanning at 220 nm was applied for the analysis. The dynamic linear range, LOD, LOQ and recovery with the inter-day, and intra-day RSDs of the developed method indicated the validity of the method. CONCLUSION A pressurised MAE method for extracting WA from the plant's aerial part was optimised using factorial-based design. The net effect of time, temperature, solvent volume and its ratio suggests that the yield of WA increases until each factor reaches its optimum value, and decreases with further increase in temperature or solvent ratio.

Abstract:Compared with the use of a single herb alone, herb combinations can significantly increase their effectiveness in treating various diseases. The objective of this research was to determine the appropriate proportions of Garcinia mangostana (GM), Curcuma comosa (CC), and Acanthus ebracteatus (AE) to enhance their antioxidant and anti-inflammatory properties. The study employed computer modeling (in silico) and laboratory testing (in vitro). The optimal levels of phenolic and flavonoid compounds were achieved with a ratio of 15 parts GM, 5 parts CC, and 10 parts AE. In contrast, the 10 : 15 : 5 ratio resulted in the highest antioxidant activity, reducing DPPH radicals by 80.33% and ABTS radicals by 92.20%. The ratio had a synergistic effect and was within the safe range, ranging from 82.97 to 147.04 µg/mL. The ratio of 5 : 10 : 15 had the greatest anti-inflammatory effects, with an exceptional activity level of 98.86%. It effectively suppressed 23 genes or proteins, including the prominent NFE2L2 gene, and had a synergistic effect. This combination effectively inhibits inflammation (IC50: 12.05 µg/mL) and is safe for macrophages at concentrations up to 50 µg/mL. These findings emphasize the potential of these herbal mixtures for antioxidant and anti-inflammatory applications.
Biochemical and molecular docking-based strategies of Acalypha indica and Boerhavia diffusa extract by targeting bacterial strains and cancer proteins

Abstract:Antibiotic-resistant microbes have emerged around the world, presenting a risk to health. Plant-derived drugs have become a potential source for the production of antibiotic-resistant drugs and cancer therapies. In this study, we investigated the antibacterial, cytotoxic and antioxidant properties of Acalypha indica and Boerhavia diffusa, and conducted in silico molecular docking experiments against EGFR and VEGFR-2 proteins. The metabolic extract of A. indica inhibited Streptococcus iniae and Staphylococcus sciuri with inhibition zones of 21.66 ± 0.57 mm and 20.33 ± 0.57 mm, respectively. The B. diffusa leaf extract produced inhibition zones of 20.3333 ± 0.5773 mm and 20.33 ± 0.57 mm against Streptococcus iniae and Edwardsiella anguillarum, respectively. A. indica and B. diffusa extracts had toxicities of 162.01 μg/ml and 175.6 μg/ml, respectively. Moreover, B. diffusa (IC50 =154.42 µg/ml) leaf extract exhibited moderately higher antioxidant activity compared with the A. indica (IC50 = 218.97 µg/ml) leaf extract. Multiple interactions were observed at Leu694, Met769 and Leu820 sites for EGFR and at Asp1046 and Cys1045 sites for VEGFR during the molecular docking study. CID-235030, CID-70825 and CID-156619353 had binding energies of -7.6 kJ/mol, -7.5 kJ/mol and -7.6 kJ/mol, respectively, with EGFR protein. VEGFR-2 protein had docking energies of -7.5 kJ/mol, -7.6 kJ/mol and -7.3 kJ/mol, respectively, for CID-6420353, CID-156619353 and CID-70825 compounds. The MD simulation trajectories revealed the hit compound; CID-235030 and EGFR complex, CID-6420353 and VEGFR-2 exhibit stable profile in the root mean square deviation (RMSD), radius of gyration (Rg), solvent accessible surface area (SASA), hydrogen bond and root mean square fluctuation (RMSF) and the binding free energy by MM-PBSA method. This study indicates that methanol extracts of A. indica and B. diffusa may play a crucial role in developing antibiotic-resistant and cancer drugs.Communicated by Ramaswamy H. Sarma.
In vitro total phenolics, total flavonoids, antioxidant and antibacterial activities of selected medicinal plants using different solvent systems

Abstract:Recently, an interest has surged in utilizing indigenous medicinal plants to treat infectious illnesses and extract bioactive substances, highlighting the need to analyze medicinal plants for phytochemicals and bioactivities. The present study was aimed to evaluate the impact of different solvent systems (aqueous, ethanol, and methanol) used for extraction on total phenolics, total flavonoids, antioxidant, and antibacterial activity of three medicinal plants of Azad Kashmir ( Achillea millefolium , Bergenia ciliata, and Aloe vera ). High phenolic content was found in methanol extracts of B. ciliata (27.48 ± 0.58 mg GAE/g dry weight), A. vera (25.61 ± 0.33 mg GAE/g dry weight), and A. millefolium (24.25 ± 0.67 mg GAE/g dry weight). High flavonoid content was obtained in the ethanol extract of A. millefolium (27.13 ± 0.64 mg QE/g dry weight), methanol extract of B. ciliata (17.44 ± 0.44 ± 0.44 mg QE/g dry weight), and the methanol extract of A. vera (14.68 ± 0.67 mg QE/g dry weight). Strong 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity (DPPH) was obtained with a methanol extract of B. ciliata (IC 50 = 60.27 ± 0.20 μg/mL). With a zone of inhibition and a minimum inhibitory concentration ranging from 10.00 ± 0.66 to 24.67 ± 1.21 mm and 78 to 625 μg/mL, respectively, all of the studied plants demonstrated notable antibacterial activity against Staphylococcus aureus and Escherichia coli . A. vera showed greater antibacterial activity as compared to other plants under study while methanolic extract showed greater antibacterial activity than ethanolic and aqueous extract. The findings of this research support the use of these medicinal plants to treat a variety of diseases.

Comprehensive in vitro evaluation of Indigofera hochstetteri Baker extract: Effect of chemicals in antimicrobial, anticancer, anti-inflammatory, and anti-diabetic activities

Abstract:The study aimed to analyze the pharmacological properties of medicinal plant Indigofera hochstetteri Baker extracts. Preliminary phytochemical analysis revealed a diverse range of secondary metabolites present in it. TLC analysis detected numerous phytochemicals with varying Rf values, aiding in different solvent systems. GC-MS analysis revealed the presence of 29 bioactive compounds with diverse pharmacological activities, including anti-inflammatory, antioxidant, analgesic and antimicrobial properties. Antimicrobial effect of I. hochstetteri Baker methanolic extract showed significant inhibitory effects against E. coli, E. aerogenes, S. flexneri, P. aeruginosa, S. aureus, E. faecalis, B. cereus, and fungal strain C. albicans. The methanol extract also showed significant antifungal activity by inhibiting the growth of Sclerotium rolfsii in food poisoning method. MTT assays revealed significant cytotoxic activity of methanolic extract against human leukemia HL-60 cancer cells with IC50 of 116.01 μg/mL. In apoptotic study, I. hochstetteri Baker methanolic extract showed 28.84% viable cells, 30.2% early apoptosis, 35.54% late apoptosis, and 5.86% necrosis comparatively similar with standard used. The extract showed significant anti-inflammatory effect on HRBC stabilization, and protein denaturation of BSA and egg albumin denaturation with IC50 of 193.62 μg/mL, 113.94 μg/mL respectively. In anti-diabetic assays like α-amylase, α-glucosidase, and Glucose uptake assay, I. hochstetteri extract showed good anti-diabetic effect with IC50 of 60.64 μg/mL, 169.34 μg/mL, and 205.63 μg/mL respectively. In conclusion I. hochstetteri Baker have promising bioactive metabolites with significant biological activities, it can be good substitute for the chemical drugs after successful clinical studies.
Molecular docking of bioactive compounds extracted and purified from selected medicinal plant species against covid-19 proteins and in vitro evaluation

Abstract:Bioactive compounds are secondary metabolites of plants. They offer diverse pharmacological properties. Peganum harmala is reported to have pharmaceutical effects like insecticidal, antitumor, curing malaria, anti-spasmodic, vasorelaxant, antihistaminic effect. Rosa brunonii has medicinal importance in its flower and fruits effective against different diseases and juice of leaf is reported to be applied externally to cure wounds and cuts. Dryopteris ramosa aqueous leaf extract is used to treat stomach ulcers and stomachaches. Each of these three medicinal plants have been indicated to have anticancer, antiviral, antioxidant, cytotoxic and antifungal effects but efficacy of their bioactive compounds remained unexplored. Study was aimed to explore In-vitro and In-silico anticancer, antiviral, antioxidant, cytotoxic and antifungal effects of bioactive compounds of above three medicinal plants. DPPH and ABTS assay were applied for assessment of antioxidant properties of compounds. Antibacterial properties of compounds were checked by agar well diffusion method. Brine shrimp lethality assay was performed to check cytotoxic effect of compounds. Molecular docking was conducted to investigate the binding efficacy between isolated compounds and targeted proteins. The compound isomangiferrin and tiliroside presented strong antioxidant potential 78.32% (± 0.213) and 77.77% (± 0.211) respectively in DPPH assay while harmaline showed 80.71% (± 0.072) at 200 μg/mL in ABTS assay. The compound harmine, harmaline and PH-HM 17 exhibited highest zone of inhibition 22 mm, 23 mm, 22 mm respectively against Xanthomonas while Irriflophenone-3-C-β- D-glucopyranoside showed maximum zone of inhibition 34 mm against E. coli. The compound isomangiferrin and vasicine contained strong antibacterial activity 32 mm and 22 mm respectively against S. aureus. The compound mangiferrin, astragalin, tiliroside, quercitin-3-O-rhamnoside showed maximum inhibitory zone 32 mm, 26 mm, 24 mm and 22 mm respectively against Klebsiella pneumoniae. Highest cytotoxic effect was observed by compound tiliroside i.e. 95% with LD 50 value 73.59 μg/mL. The compound tiliroside showed the best binding mode of interaction to all targeted proteins presenting maximum hydrophobic interactions and hydrogen bonds. The binding affinity of tiliroside was − 17.9, − 14.9, − 14.6, − 13.8, − 12.8 against different proteins 6VAR, 5C5S, IEA3, 2XV7 and 6LUS respectively. Bioactive compounds are significant natural antioxidants, which could help to prevent the progression of various diseases caused by free radicals. Based on molecular docking we have concluded that phytochemicals can have better anticancer and antiviral potential.

Abstract:In the present study, evaluation of antimicrobial, antioxidant, phytochemical constituents and toxicological properties of six coastal medicinal plants (CMP's) such as Ipomea biloba, Cantharanthus roseus, Cymbopogon citratus, Vitex negundo, Thespesia populnea and Pandanus tectorius was done. The maximum antimicrobial activity was recorded by methanolic extracts of V. negundo and T. populnea against bacterial and fungal pathogens. Similarly, methanolic extracts of V. negundo and T. populnea evidenced highest antioxidant properties. The extract of T. populnea showed the maximum cytotoxicity against Artemia salina with the LC50 value of 478.11μg/ml. The hemolytic property of CMP's extracts was V. negundo (8.91%), T. populnea (21%) and C. citratus (64%) also the hemolytic index did not show any hemolysis of human erythrocytes. Furthermore, the TLC separation of V. negundo and T. populnea extracts exhibited the presence of Flavonoids (Rf-0.74) and Terpenoids (Rf-0.64). The present findings propose the promising clinical applications of selected CMP's.

Abstract:Tamarindus indica is one of the tropical medicinal plants that has been attributed curative potential of numerous diseases by many rural dwellers. This study was designed to evaluate the antioxidant, antibacterial activities and also to determine the various chemical constituents responsible for its pharmacological activities. The methanol extract of Tamarindus indica fruit pulp was analyzed by Gas Chromatography/Mass Spectrometer to determine the volatile compounds present. The antioxidant activities were performed using DPPH and FRAP method and the antibacterial activity was tested against some common pathogens by macro broth dilution method. The GCMS analysis shows the presence of 37 compounds, out of which 14 had their peak area percentages ≥ 1% and only two compounds had no reported pharmacological activities. Most of the bioactive compounds including 5-Hydroxymethylfurfural (31.06%)-3-O-Methyl-d-glucose (16.31%), 1,6-anhydro-β-D-Glucopyranose (9.95%), 5-methyl-Furancarboxaldehyde (3.2%), Triethylenediamine (1.17%), 1-(2-furanyl)-1-Propcanone (2.18%), Methyl 2-furoate (3.14%), Levoglucosenone (3.21%), methyl ester-Hepta-2,4-dienoic acid, (8.85%), 2,3-dihydro-3,5-dihydrox-4H-Pyran-4-one (3.4%), O-α-D-glucopyranosyl-(1.fwdarw.3)-β-D-fructofuranosyl-α-D-Glucopyranoside (2.18%), n-Hexadecanoic acid (1.38%), 2-Heptanol, acetate (1.29%), 5-[(5-methyl-2-fur-2-Furancarboxaldehyde (1.08%), 3-Methyl-2-furoic acid (1.05%) and cis-Vaccenic acid (2.85%)have been reported with different activities such as antibacterial, antifungal, antitubercular, anticancer, antioxidant and other prophylactic activities. The extract demonstrated inhibitory potential against all tested pathogen. However, Plesiomonas shigellosis ATCC 15903 and Bacillus pumillus ATCC 14884 are more sensitive with the MIC of 0.22 and 0.44 mg/ml respectively. The antioxidant activity was relatively low due to the low phenolic content of the extract. This shows that there is a strong correlation between antioxidant activities and phenolic content. GC–MS analysis revealed the presence of bioactive phytoconstituents with various biological activities and this justifies the rationale behind its usage as a curative therapy by many local dwellers.

Abstract:Introduction and Aim Amid the rapid advancements in contemporary medicine, the resurgence of phytomedicine as a therapeutic avenue has garnered substantial attention. Nearly 30% of FDA-approved pharmaceuticals trace their origins to botanical sources. Phytomedicine has been shown to hold promising applications to attenuate bacterial virulence and source new of bioactive compounds to battle multidrug resistant pathogens. In this context, the current investigation delves into the antimicrobial potential of four indigenous plant species. Methodology This study is primed to unveil the antibacterial and antifungal potential of n-hexane and methanolic extracts of Bismarckia nobilis , Choysia ternata , Chamaedora cataractarum , and Beaucarnea recurvate against strains of S. aureus and C. neoformans . These plant extracts' Minimum Inhibitory Concentration (MIC) was discerned via the agar well diffusion assay, microbroth dilution assay, and MTT reduction assay. Results Notably, the n-hexane extracts of B. nobilis and C. ternata exhibited robust activity against S. aureus strains, with 100 mg/mL concentrations yielding inhibition zones measuring 12.1–13.1 mm and 13.1–15.1 mm, respectively. Correspondingly, the methanolic extracts (100 mg/mL) of B. nobilis , C. ternata , C. cataractarum , and B. recurvata presented notable antifungal activity against Cryptococcus neoformans , as evidenced by zones of inhibition measuring 14.25 mm, 13.25 mm, 16.25 mm, and 17.35 mm, respectively. Microbroth dilution assays revealed that the MIC of CT and BN plants against S. aureus ranged from 0.78 to 3.125 mg/mL and 1.56–12.5 mg/mL, respectively, with a consequential MIC index of 0.1248 for BN and CT plants against S. aureus . The n-hexane extract of B. nobilis and C. ternata showed antibacterial activity against pathogenic S. aureus. Similarly, the methanolic extracts of B. nobilis, C. ternata, C. cataractarum, and B. recurvata exhibited potent antifungal activity against C. neoformans. Conclusion This study postulates indigenous plant-derived extracts as potent and multifaceted antifungal and antibacterial resources for antimicrobial development.

Abstract:BackgroundThe extracts of the ten selected Sri Lankan medicinal plants have been traditionally used in the treatment of inflammatory mediated diseases. The extracts were investigated for anti-inflammatory and anti-oxidant potential in vitro to identify bio-active extracts for further chemical characterization.MethodsIn vitro anti-inflammatory activities of total ethanol extracts were investigated measuring the inhibitory activities of four pro-inflammatory enzymes, arachidonate-5- lipoxygenase (A5-LOX), hyaluronidase (HYL), xanthine oxidase (XO) and inducible nitric oxide (iNO) synthase. Cytotoxicity of extracts were determined by MTT assay. Oxidative burst inhibition (OBI) on human whole blood (WB) and isolated polymorphoneutrophils (PMNs) was carried out for a selected bio-active extract. Anti- oxidant activities of the extracts were determined by 2,2-diphenyl-1-picrylhydrazyl (DPPH) free radical scavenging, ferric reducing antioxidant power (FRAP), ferrous ion chelation (FIC) and oxygen radical absorbance capacity (ORAC) assays. Total polyphenol and total Flavonoid contents of the extracts were also determined. The most active plant extract was analysed using Gas chromatography-Mass spectrometry (GC-MS) and High Performance Liquid Chromatography (HPLC).ResultsThe ethanol bark extract of Flacourtia indica showed the highest A5-LOX (IC50: 22.75 ± 1.94 g/mL), XO (70.46 ± 0.18%; 250 μg/mL) and iNOs inhibitory activities on LPS- activated raw 264.7 macrophage cells (38.07 ± 0.93%; 500 μg/mL) with promising OBI both on WB (IC50: 47.64 2.32 μg/mL) and PMNs (IC50: 5.02 0.38 μg/mL). The highest HYL inhibitory activity was showed by the leaf extracts of Barathranthus nodiflorus (42.31 ± 2.00%; 500 μg/mL) and Diospyros ebenum (41.60 ± 1.18%; 500 μg/mL). The bark and leaf extracts of Callophyllum innophyllum (IC50: 6.99 ± 0.02 μg/mL) and Symplocus cochinchinesis (IC50: 9.85 ± 0.28 μg/mL) showed promising DPPH free radical scavenging activities. The GC-MS analysis of ethanol bark extract of F. indica showed the presence of two major bio-active compounds linoleic acid ethyl ester and hexadecanoic acid, ethyl ester (> 2% peak area). The HPLC analysis showed the presence polyphenolic compounds.ConclusionThe ethanol bark extract of F. indica can be identified as a potential candidate for the development of anti-inflammatory agents, which deserves further investigations. The bio-active plant extracts may be effectively used in the applications of cosmetic and health care industry.

Abstract:Herbs are making a comeback, and the herbal renaissance is taking place all over the world. Medicinal plants continue to play an essential role in developing and emerging countries. Considering the chemical and therapeutic elements obtained in natural form from plants and plant extracts, they play an essential role in modern medicine. The present study aims to evaluate the phytochemicals, antimicrobial and anti-inflammatory activity of the aqueous extract of . The phytochemical examination revealed the presence of alkaloids, steroids, coumarins, and glycosides in the root's aqueous extract. The antimicrobial activity against the Gram-negative organisms (that cause urinary and secondary infections) was performed. The aqueous extract showed maximum activity against sp with a zone of inhibition of 28 mm and an almost similar zone size was measured against antibiotics Ciprofloxacin. The anti-inflammatory tests were also performed and the extract exhibited an appreciable presence of anti-inflammatory metabolites. The test extracts inhibited protein (albumin) denaturation in a concentration-dependent manner, according to the current data. Secondary metabolites comprise a vast number of functional moieties that interact to create a broad range of biological functions. Many modern drugs are based on plants and plant-based compounds, and they are currently being used to treat various ailments.

This study focuses on the antimicrobial activity and phytochemical characterization of C. oppositifolia extracts, aiming to identify novel plant-based therapeutic agents. Methods: C. oppositifolia specimens-leaves and inflorescence. Specimens were cleaned, sterilized, dried, and ground into a fine powder. Extracts were obtained using methanol and petroleum ether via a Soxhlet apparatus, followed by fractionation with chloroform, n-butanol, and ethyl acetate. Volatile oil was extracted through hydro distillation using a Clevenger apparatus. Phytochemical analysis was conducted to identify bioactive compounds. Biophysical techniques, including UV-visible spectrophotometry, TLC, HPLC, GC-MS, FTIR, and NMR, were employed for characterization. Antimicrobial activity was tested against S. aureus ATCC25922 and E. coli ATCC25922 using agar well and disc diffusion methods, and synergistic effects were assessed with erythromycin and amoxicillin. Results: Methanol extract exhibited bacteriostatic activity with inhibition zones of 13.0 ± 0.2 mm for both S. aureus and E. coli . Petroleum ether, chloroform, n-butanol, and ethyl acetate fractions showed varying inhibition zones. Erythromycin demonstrated bactericidal activity, which was enhanced synergistically when combined with methanol extract and volatile oil, increasing inhibition zones against S. aureus . Phytochemical analysis identified phenols, flavonoids, tannins, coumarins, alkaloids, terpenoids, saponins, and glycosides. FTIR analysis revealed functional groups such as amines, aldehydes, nitriles, alkenes, and sulfones. GC-MS identified 24 compounds, with α-pinene, caryophyllene, and carene as major components. NMR spectra indicated no complex formation between oils and antibiotics, suggesting the compounds act as synergists. Conclusion: The C. oppositifolia extracts possess significant antimicrobial activity and synergistic potential, particularly against S. aureus . The presence of various bioactive compounds suggests a promising role in developing new plant-based therapeutics. THE EFFECT OF LAI BASED MODIS VEGETATION INDEX IN DIFFERENT SCALE TO ESTIMATE GROSS PRIMARY PRODUCTIVITY

Abstract:The Moderate Resolution Imaging Spectroradiometer (MODIS) is the primary NASA Earth Observing System instrument monitoring the seasonality of global terrestrial vegetation. MODIS products, such as the MODIS Vegetation Index (VI), Normalized Difference Vegetation Index (NDVI) and Enhanced Vegetation Index (EVI), are commonly used in ecological modeling. The difference spatial resolutions (250, 500, 1000 and 5600 m) may cause the error in the output. In this study, the 3-PGS (Physiological Principles Predicting Growth using Satellite data) model was used to examined the errors of gross primary productivity (GPP) based difference spatial resolutions. First, in situ Leaf Area Index (LAI) was compared with NDVI and EVI from Hemispherical Spectroradiometer (HSSR) to find the relationship. Then, these relationships were applied to the model for estimating GPP as the final output. From the results, GPP based on MODIS NDVI at spatial resolution 250 m was performed the less error around 9.12%, whereas GPP based MODIS on EVI at spatial resolution 1000 m was performed the less error around 5.76%. GPP based on MODIS NDVI and EVI at spatial resolution 5600 m were showed the biggest error with 18.47% and 13.02%, respectively. However, the global spatial resolution can be accepted for applying to the model, based on one point, with the error around 20%.
Phytochemical Analysis and Demonstration of Antioxidant, Antibacterial, and Antibiofilm Activities of Ethnomedicinal Plants of North East India

Abstract:Ethnomedicinal plants are a rich reservoir of active compounds with potent pharmacological properties. Therefore, plants could serve as a source for the discovery of active antimicrobial and antioxidant agents and are focused because of their low toxicity, economic viability, easy availability, etc. In this regard, phytochemical analyses, viz. β-carotene, total sugar, reducing sugar, vitamin C, total carotenoids, protein, total phenolic content (TPC), and total flavonoid content (TFC) of 20 ethnomedicinal plants of North East India (NEI) were evaluated in this study. The antibacterial activity against human pathogens and antioxidant potential of plant extracts was also demonstrated. The minimum inhibitory concentration (MIC80), minimum bactericidal concentration (MBC), and total antibacterial activity (TAA) of the active extracts were evaluated against Pseudomonas aeruginosa and Chromobacterium violaceum. The active extracts were also examined for antibiofilm as well as anti-pyocyanin activities against P. aeruginosa and anti-QS activity against C. violaceum at sub-MICs. The study demonstrated variable concentration of phytochemicals of the extracts, viz. β-carotene (0.29-8.91 mg g-1), total sugar (2.92-30.6 mM), reducing sugar (0.44-14.5 mM), vitamin C (8.41-31.3 mg g-1), total carotenoids (14.9-267.0 mg g-1), protein (5.65-283 mg g-1), TPC (5.32-31.0 mg GAE/g DW), and TFC (1.74-68.2 mg QE/g DW). The plant extracts also exhibited potent antioxidant and antibacterial activities against both Gram-positive and Gram-negative bacteria. Some of the extracts also demonstrated significant biofilm inhibition and eradication, anti-pyocyanin, and anti-QS activities at sub-MICs. The selected ethnomedicinal plants are rich in phytochemicals and demonstrated potent antioxidant, antibacterial, and antibiofilm activities, thus could serve as the important source of novel antioxidant and antimicrobial agents.
In vitro antibacterial and antioxidant activity of flavonoids from the roots of Tephrosia vogelii: a combined experimental and computational study

Abstract:Tephrosia vogelii is a traditional medicinal plant used to treat hypertension, diarrhea and urinary disorders. Silica gel chromatographic separation of CH2Cl2/MeOH (1:1) roots extract of T. vogelii afforded seven compounds namely; β-sitosterol (1a), stigmasterol (1b), 6a, 12a-dehydro-deguelin (2), tephrosin (3), maackiain (4), obovatin (5) and 6-oxo, 6a, 12a-dehydro-deguelin (6). GC-MS analysis of essential oils from the root of T. vogelii displayed a total of 17 compounds of which cis-nerolidol (41.7 %) and cadinol (19.7 %) were the major constituents. CH2Cl2/MeOH (1:1) extract, MeOH extract, maackiain (4) and obovatin (5) showed moderate inhibitory activity against Pseudomonas aeruginosa with MIC value of 0.5, 0.66, 0.83 and 0.83 mg/mL, respectively, compared to ciprofloxacin (MIC of 0.078 μg/mL). 6a, 12a-dihydro-deguelin (2), and 6-oxo, 6a, 12a-dehydro-deguelin (6) displayed significant activity against S. epidermis with MIC values of 0.66 mg/mL. Tephrosin (3) and maackiain (4) also showed moderate antibacterial activity against Staphylococcus aureus and Proteus mirabilis with MIC values of 0.83 and 0.5 mg/mL, respectively, compared to ciprofloxacin (0.312 μg/mL). The radical scavenging activity results indicated that tephrosin (3), obovatin (5) and 6-oxo, 6a, 12a-dehydro-deguelin (6) showed potent DPPH scavenging activity with IC50 values of 10.97, 10.43 and 10.73 μg/mL, respectively, compared to ascorbic acid (IC50 of 5.83 μg/mL). The docking prediction results revealed that 6a, 12a-dehydro-deguelin (2) displayed the best binding energy of -8.1 kcal/mol towards pyruvate kinase of S. aureus (PDB ID: 3T07) and -7.9 kcal/mol towards P. mirabilis urease (PDB ID: 1E9Y) and DNA gyrase B of Escherichia coli (PDB: 4F86) receptors compared to ciprofloxacin (-7.2 to -8.0 kcal/mol). Maackiain (4) and obovatin (5) displayed the minimum binding energy of -7.9 and -8.2 kcal/mol towards the LasR protein of P. aeruginosa (PDB: ID 2UV) and S. epidermidis FtsZ (PDB: ID 4M8I), respectively. The SwissADME drug-likeness and Pro Tox II toxicity prediction results indicated that compounds (2-6) obeyed Lipinski's rule of five with 0 violations and none of them were found to be hepatotoxic, mutagenic, and cytotoxic, respectively. The in vitro assessment results supported by the in silico analysis revealed that crude extracts and isolated compounds showed promising antibacterial and antioxidant activity, which proves the therapeutic potential of the roots of T. vogelii.
