Abstract:Thienoisoindigo-based semiconducting polymer with a strong near-infrared absorbance is synthesized and its water-dispersed nanoparticles (TSPNs) are investigated as a contrast agent for photoacoustic (PA) imaging in the second near-infrared (NIR-II) window (1000-1350 nm). The TSPNs generate a strong PA signal in the NIR-II optical window, where background signals from endogenous contrast agents, including blood and lipid, are at the local minima. By embedding a TSPN-containing tube in chicken-breast tissue, an imaging depth of more than 5 cm at 1064 nm excitation is achieved with a contrast-agent concentration as low as 40 mu g mL(-1). The TSPNs under the skin or in the tumor are clearly visualized at 1100 and 1300 nm, with negligible interference from the tissue background. TSPN as a PA contrast in the NIR-II window opens new opportunities for biomedical imaging of deep tissues with improved contrast.

Abstract:Photoacoustic tomography (PAT) is an emerging molecular imaging modality. Here, we demonstrate use of semiconductor copper sulfide nanoparticles (CuS NPs) for PAT with an Nd:YAG laser at a wavelength of 1064 nm. CuS NPs allowed visualization of mouse brain after intracranial injection, rat lymph nodes 12 mm below the skin after interstitial injection, and CuS NP-containing agarose gel embedded in chicken breast muscle at a depth of ~5 cm. This imaging approach has great potential for molecular imaging of breast cancer.
Iron-Content-Dependent, Quasi-Static Dielectric Resonances and Oxidative Transitions in Bornite and Chalcopyrite Copper Iron Sulfide Nanocrystals

Abstract:Manipulation of material composition can often induce unprecedented optoelectronic changes in semiconductor nanocrystals (NCs). Here, we demonstrate that bornite-phase copper iron sulfide (Cu5FeS4) NCs exhibit tunable optical characteristics from visible to near-infrared (NIR) that are strongly dependent on their iron content. These stoichiometry-dependent optical modulations confirm that the origin of the spectral response in these NCs is due to the occurrence of two fundamentally different types of resonant excitation: (i) a quasi-static dielectric resonance (DR) and (ii) a localized surface plasmon resonance (LSPR). Electronic band structure calculations by density functional theory (DFT) show that the presence of an intermediate band (IB) of states formed by empty Fe d-orbitals gives rise to the occurrence of a DR in the visible-frequency regime, analogous to the observed optical response in all-dielectric IB chalcopyrite (CuFeS2) NCs. Moreover, as the Fe content of the NCs is decreased via aerobic oxidation, the intensity of the DR diminishes, accompanied by a concomitant rise in the intensity of a hole-induced LSPR response in the NIR, reminiscent of self-doped copper sulfide NCs. Notably, the spectral position of the DR does not change as its intensity is reduced, clearly differentiating this mode of resonant excitation from the well-known LSPR response, which shifts substantially in frequency as the free-carrier concentration is modified. We further demonstrate an analogous DR-to-LSPR evolution in CuFeS2 NCs subjected to the influence of oxidizing agents and added ions. The decreased Fe content significantly reduces the contributions of Fe 3d orbitals to the IB in these ternary Cu–Fe–S NCs, resulting in a reduced DR intensity, and as the composition approaches a more Cu–S-like system, the vacancy-induced LSPR intensity becomes more pronounced. Abstract:Functional Materials Letters, Ahead of Print. Copper-based nanomaterials have garnered significant attention for their potential applications in photothermal (PT) conversion and photoacoustic (PA) imaging due to their unique optical properties. However, current strategies often involve complex synthesis processes and the incorporation of additional materials, which limit their practical applicability. Here, we employed a straightforward nanocrystallization method to synthesize copper phosphate nanoparticles (Cu3(PO4)2 NPs), converting free copper ions into solid-phase nano-sized particles. This approach not only simplifies the synthesis process but also enhances the optical and thermal properties of the resulting Cu3(PO4)2 NPs. The Cu3(PO4)2 NPs achieved a temperature increase of over 20∘C and a PT conversion efficiency of 10.5% under 808 nm laser irradiation. Furthermore, Cu3(PO4)2 NPs exhibited strong, concentration-dependent PA signals, which enabled effective deep tissue imaging in vivo. The enhanced properties of Cu3(PO4)2 NPs can be attributed to the nanocrystallization of the particles, which alters the local environment of copper ions and enhances d−d transitions through specific ligand field effects. These effects increase light absorption, particularly in the NIR region, by facilitating better overlap of electronic states and enabling more efficient non-radiative relaxation processes. This highlights the significance of nano-structuring in developing advanced nanomaterials with tailored functionalities for biomedical applications.

Abstract:Photoacoustic (PA) imaging has received more and more attention on disease diagnosis and fundamental scientific research. It is still challenging to amplify their imaging ability and reduce the toxicity of inorganic materials and exogenous contrast agents. Semiconducting polymer nanoparticles (SPNs), as a new type of contrast agent, have the advantages of low toxicity, flexible structure adjustment, good photostability, and excellent photothermal conversion efficiency. SPNs containing benzo(1,2- c ;4,5- c ′)bis(1,2,5)thiadiazole (BBT) units, as the most classic second near-infrared window (NIR-II, 1,000–1700 nm) PA contrast agents, can achieve light absorption in the NIR-II region, thereby effectively reducing light loss in biological tissues and improving imaging resolution. This mini review summarizes the recent advances in the design strategy of BBT and its derivative-based semiconducting polymer nanoparticles for second near-infrared photoacoustic imaging. The evolution process of BBT blocks provides a unique perspective for the design of high-performance NIR-II PA contrast agents.

Unlocking NIR-II Photoluminescence in 2D Copper Tetrasilicate Nanosheets Through Flame Spray Pyrolysis

Abstract:Expanding fluorescence bioimaging into the second near-infrared spectrum (NIR-II, 1000-1700 nm) unlocks advanced possibilities for diagnostics and therapeutics, offering superior tissue penetration and resolution. Two-dimensional copper tetrasilicate (CTS) pigments (MCuSi4O10, M = Ca, Sr, Ba) are known for their brightness and stability, yet synthetic challenges have curbed their integration into bioimaging. Here, we introduce flame-spray-pyrolysis (FSP) as a versatile and scalable synthesis approach to produce ultra-bright, metastable CTS nanosheets (NS) by annealing multi-element metal oxide nanoparticles into 2D crystals. Group-II ion incorporation shifts emission into the NIR-II range, with Ba0.33Sr0.33Ca0.33CuSi4O10 peaking at 1007 nm, while minor Mg-doping induces a hypsochromic shift and extends fluorescence lifetimes. The engineered CTS achieve quantum yields up to 34%, supporting NS high-frame-rate imaging (>200 fps). These unique properties enable CTS-NS to serve as powerful contrast agents for super-resolution NIR bioimaging, demonstrated in vivo through transcranial microcirculation mapping and macrophage tracking in mice using diffuse optical localization imaging (DOLI). This pioneering synthesis strategy unlocks wavelength-tunable NS for advanced NIR-II bioimaging applications.

Abstract:Photoacoustic (PA) imaging using contrast agents with strong near-infrared-II (NIR-II, 1000–1700 nm) absorption enables deep penetration into biological tissue. Besides, biocompatibility and biodegradability are essential for clinical translation. Herein, we developed biocompatible and biodegradable germanium nanoparticles (GeNPs) with high photothermal stability as well as strong and broad absorption for NIR-II PA imaging. We first demonstrate the excellent biocompatibility of the GeNPs through experiments, including the zebrafish embryo survival rates, nude mouse body weight curves, and histological images of the major organs. Then, comprehensive PA imaging demonstrations are presented to showcase the versatile imaging capabilities and excellent biodegradability, including in vitro PA imaging which can bypass blood absorption, in vivo dual-wavelength PA imaging which can clearly distinguish the injected GeNPs from the background blood vessels, in vivo and ex vivo PA imaging with deep penetration, in vivo time-lapse PA imaging of a mouse ear for observing biodegradation, ex vivo time-lapse PA imaging of the major organs of a mouse model for observing the biodistribution after intravenous injection, and notably in vivo dual-modality fluorescence and PA imaging of osteosarcoma tumors. The in vivo biodegradation of GeNPs is observed not only in the normal tissue but also in the tumor, making the GeNPs a promising candidate for clinical NIR-II PA imaging applications.

Abstract:Objective: Photoacoustic (PA) tomography (PAT) has attracted extensive interest because of its optical absorption contrast and ultrasonic detection. This study aims to develop a biocompatible and biodegradable PA contrast agent particularly promising for clinical applications in human body. Methods: In this study, we presented a PA contrast agent: 1, 2-distearoyl-sn-glycero-3-phosphoethanolamine- N-[methoxy (polyethylene glycol)] (DSPE-PEG)-coated superparamagnetic iron oxide (SPIO) nanoparticles (NPs) loaded with indocyanine green (ICG). We used ICG and SPIO NPs because both drugs are approved by the U.S. Food and Drug Administration. Given the strong absorption of near-infrared laser pulses, SPIO@DSPE-PEG/ICG NPs with a uniform diameter of ~28 nm could significantly enhance PA signals. Results: We demonstrated the contrast enhancement of these NPs in phantom and animal experiments, in which the in vivo circulation time of SPIO@DSPE-PEG/ICG NPs was considerably longer than that of free ICG. These novel NPs also displayed a high efficiency of tumor targeting. Conclusions: SPIO@DSPE-PEG/ICG NPs are promising PAT contrast agents for clinical applications.
Near-infrared-II-activatable sulfur-deficient plasmonic Bi2S3-x-Au heterostructures for photoacoustic imaging-guided ultrasound enhanced high performance phototherapy

Abstract:Bismuth sulfide is widely used as an n-type semiconductor material in photocatalytic reactions. However, bismuth sulfide has poor absorption in the near-infrared region and low charge separation efficiency, limiting its application in phototherapy and sonodynamic therapy (SDT). In this study, we successfully synthesized an "all-in-one" phototheranostic nanoplatform, namely Bi2S3-x-Au@HA, based on a single second near-infrared (NIR-II) light-responsive Schottky-type Bi2S3-x-Au heterostructure for photoacoustic (PA) imaging-guided SDT-enhanced photodynamic therapy (PDT)/photothermal therapy (PTT). Bi2S3-x-Au@HA exhibits excellent NIR-II plasmonic and photothermal properties, rendering it with NIR-II PA imaging capabilities for accurate diagnosis. Additionally, the high-density sulfur vacancies constructed on the Bi2S3 surface cause it to possess a reduced band gap (1.21 eV) that can act as an electron trap. Using the density functional theory, we confirmed that the light and ultrasound-induced electrons are more likely to aggregate on the Au nanoparticle surface through interfacial self-assembly, which promotes electron-hole separation and enhances photocatalytic activity with increased reactive oxygen species (ROS) generation. With a further modification of hyaluronic acid (HA), Bi2S3-x-Au@HA can selectively target cancer cells through HA and CD44 protein interactions. Both in vitro and in vivo experiments demonstrated that Bi2S3-x-Au@HA effectively suppressed tumor growth through SDT-enhanced PTT/PDT under a single NIR-II laser and ultrasound irradiation with negligible toxicity. Our findings provide a framework for fabricating Schottky-type heterostructures as single NIR-II light-responsive nanotheranostic agents for PA imaging-guided cancer phototherapy.

Abstract:The emerging technique of photoacoustic imaging, especially in the near infra-red (NIR) window, permits high resolution, deep-penetration, clinically reliable sensing. However, few contrast agents are available that can specifically respond to intricate biological environments, and which are biodegradable and biocompatible. Herein, we introduce a new class of pH-sensitive organic photoacoustic contrast agent that operates in the second NIR window (NIR-II, 960–1,700 nm), which is derived from the self-assembled charge-transfer nanocomplex (CTN) by 3,3’,5,5’-tetramethylbenzidine (TMB) and its dication structure (TMB++). The unique NIR-II-responsive CTN can specifically respond to pH change in the physiological range and allows noninvasive and sensitive visualization of the tumor acidic microenvironment (e.g. at pH 5) in mice with higher signal-to-noise ratio. The CTN is biodegradable under physiological conditions (e.g. pH 7.4), which alleviates the biosafety concern of nanoparticle accumulation in vivo. These results clearly show the potential of the TMB/TMB++-based CTN as a promising pH-activated and biodegradable molecular probe for specific tumor photoacoustic imaging in the NIR-II region.
Rapid Degradation and High Renal Clearance of Cu3BiS3 Nanodots for Efficient Cancer Diagnosis and Photothermal Therapy in Vivo

Abstract:A key challenge for the use of inorganic nanomedicines in clinical applications is their long-term accumulation in internal organs, which raises the common concern of the risk of adverse effects and inflammatory responses. It is thus necessary to rationally design inorganic nanomaterials with proper accumulation and clearance mechanism in vivo. Herein, we prepared ultrasmall Cu3BiS3 nanodots (NDs) as a single-phased ternary bimetal sulfide for photothermal cancer therapy guided by multispectral optoacoustic tomography (MSOT) and X-ray computed tomography (CT) due to bismuth's excellent X-ray attenuation coefficient. We then monitored and investigated their absorption, distribution, metabolism, and excretion. We also used CT imaging to demonstrate that Cu3BiS3 NDs can be quickly removed through renal clearance, which may be related to their small size, rapid chemical transformation, and degradation in an acidic lysosomal environment as characterized by synchrotron radiation-based X-ray absorption near-edge structure spectroscopy. These results reveal that Cu3BiS3 NDs act as a simple but powerful "theranostic" nanoplatform for MSOT/CT imaging-guided tumor ablation with excellent metabolism and rapid clearance that will improve safety for clinical applications in the future.
A computational framework for distinguishing direct versus indirect interactions in human functional protein-protein interaction networks.

Abstract:Recognition of indirect interactions is instrumental to in silico reconstruction of signaling pathways and sheds light on the exploration of unknown physical paths between two indirectly interacting genes. However, very limited computational methods have explicitly exploited the indirect interactions with experimental evidence thus far. In this work, we attempt to distinguish direct versus indirect interactions in human functional protein-protein interaction (PPI) networks via a predictive l2-regularized logistic regression model built on the experimental data. The l2-regularized logistic regression method is adopted to counteract the potential homolog noise and reduce the computational complexity on large training data. Computational results show that the proposed model demonstrates promising performance even though the training data are highly skewed. From the 304 799 PPIs that are curated in several databases, the proposed method detects 23 131 indirect interactions, most of which have been verified by the breadth-first graph search algorithm to find dozens of physical paths between the interacting partners. Pathway enrichment analysis shows that most of the physical paths can be mapped onto more than one human signaling pathway, indicating that there do exist a series of biochemical signals between the two indirectly interacting genes. The interactome-scale computational results promise to provide useful cues to the following applications: (1) exploration of unknown physical PPIs or physical paths between two indirectly interacting genes; (2) amending or extending the existing signaling pathways; (3) recognition of the physical PPIs for druggable target discovery.
A Photoacoustic Contrast Nanoagent with a Distinct Spectral Signature for Ovarian Cancer Management

Abstract:Photoacoustic imaging (PAI) has tremendous potential for improving ovarian cancer detection. However, the lack of effective exogenous contrast agents that can improve PAI diagnosis accuracy significantly limits this application. This study presents a novel contrast nanoagent with a specific spectral signature that can be easily distinguished from endogenous chromophores in cancer tissue, allowing for high-contrast tumor visualization. Constructed as a 40 nm biocompatible polymeric nanoparticle loaded with two naphthalocyanine dyes, this agent is capable of efficient ovarian tumor accumulation after intravenous injection. The developed nanoagent displays a spectral signature with two well-separated photoacoustic peaks of comparable PA intensities in the near-infrared (NIR) region at 770 and 860 nm, which remain unaffected in cancer tissue following systemic delivery. In vivo experiments in mice with subcutaneous and intraperitoneal ovarian cancer xenografts validate that this specific spectral signature allows for accurate spectral unmixing of the nanoagent signal from endogenous contrast in cancer tissue, allowing for sensitive noninvasive cancer diagnosis. In addition, this nanoagent can selectively eradicate ovarian cancer tissue with a single dose of photothermal therapy by elevating the intratumoral temperature to ≈49 °C upon exposure to NIR light within the 700-900 nm range.
Biogenic Copper Selenide Nanoparticles for Near-Infrared Photothermal Therapy Application.

Abstract:Near-infrared (NIR) photothermal therapy (PTT) is attractiveforcancer treatment but is currently restricted by limited availabilityand insufficient NIR-II photoactivity of photothermal agents, forwhich artificial nanomaterials are usually used. Here, we report thefirst use of biogenic nanomaterials for PTT application. A fine-controlledextracellular biosynthesis of copper selenide nanoparticles (bio-Cu2-x Se) by Shewanellaoneidensis MR-1 was realized. The resulting bio-Cu2-x Se, with fine sizes (similar to 35.5nm) and high product purity, exhibited 76.9% photothermal conversionefficiency under 1064 nm laser irradiation, outperforming almost allthe existing counterparts. The protein capping also imparted goodbiocompatibility to bio-Cu(2-x )Seto favor a safe PTT application. The in vivo PTT with injected bio-Cu2-x Se in mice (without extraction norfurther modification) showed 87% tumor ablation without impairingthe normal organisms. Our work not only opens a green route to synthesizethe NIR-II photothermal nanomaterial but may also lay a basis forthe development of bacteria-nanomaterial hybrid therapy technologies.
Biocompatible PEGylated Bismuth Nanocrystals: "All-in-one" Theranostic Agent with Triple-Modal Imaging and Efficient in Vivo Photothermal Ablation of Tumors.

Abstract:Biocompatible single-component theranostic agents integrating multimodal imaging and therapeutic functions (namely, "all-in one" agents) are highly desired for clinical cancer treatments. Herein, PEGylated pure metallic bismuth nanocrystals (Bi-PEG NCs) have been developed to be a competent theranostic agent for in vivo high-performance multimodal bio-imaging and photothermal ablation of tumors. The resultant Bi-PEG NCs show excellent physiological stability, biocompatibility, prolonged blood circulation half-life and preferential tumor accumulation. Thanking to the strong. near-infrared (NIR) absorbance as well as the high photothermal conversion efficiency and conversion stability, highly effective in vivo photothermal ablation on tumors has been realized upon NIR irradiation, without noticeable toxicity. Impressively, the Bi-PEG NCs show ultrahigh X-ray computed topography (CT) enhancement efficiency (similar to 60.3 HU mL mg(-1)), overwhelming all CT contrast agents reported so far. Combining the strong CT contrast ability and photoacoustic/photothermal effect, high-contrast CT, photoacoustic (PA) and infrared thermal (IRT) triple-modal imaging have been demonstrated both in vitro and in vivo. This work highlights the potentials of such NCs as a powerful "all-in-one" theranostic nanoplatform for bioimaging and antitumor therapy, and may have provided a rather promising candidate for clinically-applied antitumor treatments based on single-component agents. Abstract:This study introduces CuS‐loaded polydopamine‐coated CaCO3 microparticles (CaCO3@CuS/PDA MPs) combined with localization optoacoustic tomography (LOT) for high‐resolution imaging of brain microvasculature and precise mapping of blood flow velocity in the NIR‐II window, overcoming the resolution limits imposed by acoustic diffraction. The new contrast agent enhances optoacoustic imaging capabilities and facilitates studies into vascular‐related pathologies. Optoacoustic imaging offers label‐free multi‐parametric characterization of cerebrovascular morphology and hemodynamics at depths and spatiotemporal resolution unattainable with optical microscopy. Effective imaging depth can greatly be enhanced by employing photons in the second near‐infrared (NIR‐II) window. However, diminished absorption by hemoglobin along with a lack of suitable contrast agents hinder an efficient application of the technique in this spectral range. Herein, copper sulfide (CuS) micro‐ and nano‐formulations for multi‐scale optoacoustic imaging in the NIR‐II window are introduced. Dynamic contrast enhancement induced by intravenously administered CuS nanoparticles facilitated visualization of blood perfusion in murine cerebrovascular networks. The individual calcium carbonate microparticles carrying CuS are further shown to generate sufficient responses to enable super‐resolution microvascular imaging and blood flow velocity mapping with localization optoacoustic tomography.

Abstract:Aberrant copper contents implicate numerous diseases including Alzheimer's disease and Wilson's disease. Conventional copper detection technologies are difficult to offer non-invasive and accurate deep tissue detection of copper. Here, we report a photoacoustic (PA) nanoprobe (NRh-IR-NMs) for ratiometric PA imaging of Cu2+. The nanoprobe consists of a selective Cu2+-responsive probe (NRh) as the indicator and a nonresponsive dye (IR) as the internal reference. In the presence of Cu2+, a selective Cu2+-induced structure change of NRh would take place, resulting in the increase of PA signal intensity increment at 716 nm (ΔPA716). However, the ΔPA834 which attributes to IR shows negligible change. Therefore, the ratiometric PA signal (ΔPA716/ΔPA834) could be used as an indicator for Cu2+ detection. This ratiometric PA detection method offers a noninvasive technology with high selectivity and tissue penetration depth, which is a promising tool for deep-tissue detection of Cu2+ in living organisms.
Optimization of Bi3+ in Upconversion Nanoparticles Induced Simultaneous Enhancement of Near-Infrared Optical and X-ray Computed Tomography Imaging Capability.

Abstract:Bioimaging probes have been extensive studied for many years, while it is still a challenge to further improve the image quality for precise diagnosis in clinical medicine. Here, monodisperse NaGdF4:Yb3+,Tm3+,x% Bi3+ (abbreviated as GYT-x% Bi3+, x = 0, 5, 10, 15, 20, 25, 30) upconversion nanoparticles (UCNPs) have been prepared through the solvothermal method. The near-infrared upconversion emission intensity of GYT-25% Bi3+ has been enhanced remarkably than that of NaGdF4:Yb3+,Tm3+ (GYT) with a factor of ∼60. Especially, the near-infrared upconversion emission band centered at 802 nm is 150 times stronger than the blue emission band of GYT-25% Bi3+ UCNPs. Such high ratio of NIR/blue UCL intensity could reduce the damage to tissues in the bioimaging process. The possibility of using GYT-25% Bi3+ UCNPs with strong near-infrared upconversion emission for optical imaging in vitro and in vivo was performed. Encouragingly, the UCL imaging penetration depth can be achieved as deep as 20 mm. Importantly, GYT-25% Bi3+ UCNPs exhibit a much higher X-ray computed tomography (CT) contrast efficiency than GYT and iodine-based contrast agent under the same clinical conditions, due to the high X-ray attenuation coefficient of bismuth. Hence, simultaneous remarkable enhancement of NIR emission and X-ray CT signal in upconversion nanoparticles could be achieved by optimizing the doping concentration of Bi3+ ions. Additionally, Gd3+ ions in the UCNPs endow GYT-25% Bi3+ UCNPs with T1-weighted magnetic resonance (MR) imaging capability.
Nd3+-sensitized Multilayered Rare-Earth Nanocrystals with Enhanced NIR-IIb Luminescence for High Resolution Optical Imaging

Abstract:Luminescence imaging in the second near-infrared (NIR-II) window, particularly within the NIR-IIb (1500-1700 nm) sub-window, has been utilized for both fundamental research and clinical applications due to its exceptional spatiotemporal resolution. Nevertheless, efficient contrast agents designed for NIR-IIb imaging are still scarce. Herein, a multilayered rare-earth nanocrystal (NaYF4:Yb,Er,Ce@NaYF4:Yb,Nd@NaYF4) with strong NIR-IIb luminescence was designed and synthesized. The doping of Ce3+ ions in the core effectively suppresses the upconversion and boosts the downconversion luminescence by populating the Er 4I13/2 level. Additionally, by separating the sensitizer (Nd3+) and the activator (Er3+) in different layers, we are able to maximize the light absorption around 800 nm and reduce the cross relaxations between rare-earth elements, which leads to a strong downconversion emission around 1530 nm under 808 nm excitation. After passivation with NaYF4 shell and phase transfer with highly PEGylated mesoporous silica layer, the nanocomposites with excellent colloidal stability and bright NIR-IIb emission were obtained. With long blood circulation time in vivo, the fabricated nanocomposites enable high resolution and contrast luminescence imaging of the blood vessels and tumors in living mice. Moreover, the rare-earth nanocrystals with superior optical properties also hold great promise for various applications in optical sensing and biomedical imaging.
