Abstract:Plant and soil elemental ratios of carbon (C), nitrogen (N) and phosphorus (P) play a central role in shaping the composition and structure of microbial communities. However, the relationships between plant and soil elemental C:N:P ratios and microbial diversity are still poorly understood. Here, we evaluated the effects of C:N:P ratios in plant-soil systems on microbial diversity in a chronosequence of restored grasslands (1, 5, 10, 15, 25, and 30 years since restoration) on the Loess Plateau. We found that C and N concentrations, C:N and C:P ratios in leaf, root, soil and microbial biomass, bacterial and fungal diversity (Shannon-Wiener index) gradually increased with year since grassland restoration. Microbial C:N:P ratios ranged from 17.8:4.5:1 to 24.3:6.6:1, and C:P ratio increased from 17.8:1 at the 1-year site to 24.3:1 at the 30-year site, indicating the increasing P limitation for soil microorganisms during grassland development. Soil microbial diversity increased with root, soil, and microbial C and N concentrations, and decreased with P concentration (p < 0.05). Structural equation modeling indicated that soil and microbial C:N and N:P ratios had the greatest influences on soil bacterial and fungal diversity, and elemental C:N:P ratios had a greater effect on soil fungal than bacterial diversity. Our findings emphasize the importance of elemental C:N:P ratios on soil microbial diversity, which is critical for formulating policies for sustainable biodiversity conservation in terrestrial ecosystems.

Abstract:Vegetation restoration is assumed to enhance carbon (C) sequestration in terrestrial ecosystems, where plant producers and microbial decomposers play key roles in soil C cycling. However, it is not clear how the nutrient limitation patterns of plants and soil microbes might change during vegetation restoration. We investigated the nutrient limitations of the plant and microbial communities along a natural vegetation restoration chronosequence (1, 8, 16, 31, and 50 years) following farmland abandonment in Qinling Mountains, China, and assessed their relationships with soil factors. The result showed that following natural vegetation restoration, the nitrogen (N) limitation of plant and microbial communities was alleviated significantly, and thereafter, it began to shift to phosphorus (P) limitation at a later stage. Plants showed P limitation 50 years after restoration, while microbial P limitation appeared 31 years later. The changes in plant nutrient limitation were consistent with those in microbial nutrient limitation, but soil microbes were limited by P earlier than plants. Random forest model and partial least squares path modeling revealed that soil nutrient stoichiometry, especially soil C:N ratio, explained more variations in plant and microbial nutrient limitation. Our study demonstrates that the imbalanced soil C:N ratio may determine the soil microbial metabolic limitation and further mediate the variation in plant nutrient limitation during natural vegetation restoration, which provides important insights into the link between metabolic limitation for microbes and nutrient limitation for plants during vegetation restoration to improve our understanding of soil C turnover in temperate forest ecosystems.

Abstract:<ol class=""><li>Grazing by ungulate herbivores can greatly alter nitrogen ([N]) and phosphorus ([P]) concentrations in plants and soils. It is not clear, however, how grazing might affect N:P co‐limitation in grasslands depending on soil N and P availability.</li><li>Here we selected 173 peer‐reviewed studies, which measured 12 key variables associated with changes in [N], [P] and N:P ratios (i.e. N:P stoichiometry) in soils and plants in the presence or absence of herbivore grazing. Subsequently, we addressed the magnitude and direction of grazing effects on these variables using a meta‐analysis approach.</li><li>Grazing increased leaf [N] and [P] but decreased total and available soil [N] and [P]. Grazing also increased leaf N:P ratios while decreasing root and total soil N:P ratios.</li><li>The response ratio (RR) of leaf N:P was negatively correlated with RR of plant‐available soil [P] and positively correlated with RR of available soil N:P ratio (rather than with RR of total soil N:P).</li><li>Intensive grazing (e.g., heavy grazing or long‐term grazing) had in general more positive effects on plant N:P stoichiometry and negative effects on soil N:P stoichiometry than light grazing. Responses of plant‐soil N:P stoichiometry to grazing greatly varied depending on plant functional group identity, plant organizational level (i.e. species and community) and grassland type.</li><li>Synthesis and applications. Our study suggests that understanding changes in available soil N:P stoichiometry (rather than total soil N:P) in response to grazing is crucial to predict nutrient co‐limitation in grassland biomes. Our findings show that P is more important for plant growth than generally thought due to greater reduction of plant‐available soil [P] under grazing. A better mechanistic understanding of the relationships between plant and available soil N:P stoichiometry under grazing will greatly help improve the sustainability of natural and semi‐natural grassland ecosystems. </li></ol>

Ecological Stoichiometric Characteristics of Plant–Soil–Microorganism of Grassland Ecosystems under Different Restoration Modes in the Karst Desertification Area

Abstract:C, N and P are the key biogenic elements of terrestrial ecosystems, and their biogeochemical processes regulate nutrient cycling and play a key role in restoring degraded ecosystems. In this paper, the grassland ecosystem under artificial restoration measures (Dactylis glomerata (DG), Lolium perenne (LP), Lolium perenne + Trifolium repens (LT)), and the natural restoration measures (NG) in a typical karst plateau desertification control area of southwest China, were taken as the research object. The C, N, and P concentrations and the ecological stoichiometry of the plant–soil–microorganism system in grasslands under different restoration measures in the karst desertification area were explored. We established the following findings: (1) Compared with NG, the C, N and P concentrations of plants and soil in DG, LP and LT were higher, and LT was the highest. (2) The microbial biomass carbon (MBC), microbial biomass nitrogen (MBN) and microbial biomass phosphorus (MBP) concentrations in LT was also the highest. (3) The C:N ratio of plant and soil indicated that the N mineralization and nutrient release rate of DG, LP and LT were stronger than that of NG, and the plant growth of NG was most seriously limited by nitrogen. The N:P ratio in plant and soil indicated that the grassland was limited by P in the four treatments. (4) The result of correlation analysis showed that the cycling process of C, N and P in the plant–soil–microorganism system was coupled, and that the elements closely affected each other. In general, the effect of artificial restoration on a degraded ecosystem was relatively better than the natural restoration for increasing plant, soil and microbial nutrient concentrations, especially in the mixed-seed ecosystem of leguminous and gramineous forage. However, in the future, grassland management, appropriate N fertilizer or N-fixing plants and P fertilizer should be considered to improve the limitation of plant N and P, so as to realize the sustainable development of grasslands and the restoration of degraded ecosystems in the karst desertification control area.

Plant community productivity is associated with multiple ecological stoichiometry in restoration grasslands

Abstract:Ecological stoichiometry links the growth of living organisms at the organ level through the strict elemental ratio (carbon [C], nitrogen [N] as well as phosphorus [P]). And this is conducive to maintaining plant productivity in terrestrial ecosystems. Yet, the identification about the significance of multiple ecological stoichiometry (including leaves, roots, soil and microbial biomass) in shaping plant productivity remains unknown. Here, we evaluated the ecological stoichiometry contributions to plant productivity in restoration grasslands on the Loess Plateau (1-, 5-, 10-, 15-, 25-, 30-years restoration grasslands). We found that grassland restoration for 30 years enhanced plant community productivity by 67.34%, which showed the synchronous increase trend with C and N contents in leaves, roots, soil as well as microbial biomass. However, leaves, roots, soil and microbial biomass P contents displayed the opposite change trend with no significant difference in these restoration grasslands ( p > 0.05). Microbial biomass C/N/P ratios exhibited thesignificant correlations with leaves, roots, soil C/N/P ratios, as well as plant community productivity ( p < 0.01), indicating that stoichiometry of soil microbial biomass is greatly restricted, with the potential to influence plant community productivity. These findings emphasized the crucial role of microbial biomass stoichiometry as a primary regulator on plant community productivity, providing the important data to improve the parameterization of plant growth models in natural grassland of China.

Electronic Prescribing Usability: Reduction of Mental Workload and Prescribing Errors Among Community Physicians.

Abstract:BACKGROUND Medical errors are common in hospitals, and research is always needed to find ways of reducing these. This study attempts to address three gaps in this field. First, the factors leading to the reduction of mental workload and its relationship with the reduction of prescribing errors by improving electronic prescribing (e-prescribing) usability have not been empirically examined before. Second, the past research in the field of e-prescribing usability lacks robust theoretical models. Third, there are no existing studies to examine the direct influences of user interface consistency and error prevention with the reduction of mental workload and prescribing errors. MATERIALS AND METHODS A quantitative survey method was used to collect data from 188 community physicians. The partial least squares path modeling technique was applied to analyze the data. RESULTS Prescribing errors were reduced by improving the information quality, user interface consistency, system ease of use, and mental workload reduction. Mental workload is reduced by ease of use, error prevention, and consistency. No significant relationships between prescribing error reduction with error prevention and also between information quality with mental workload reduction were found. CONCLUSIONS The designers of e-prescribing should improve the error prevention and consistency of the system and make it easy to use if they wish for the system to reduce users' mental workload. They should also improve the system information quality, ease of use, and consistency if they claim that their system reduces physicians' prescribing errors. The system should also reduce users' mental workload to meet this objective.
N and P fertilization enhanced carbon decomposition function by shifting microbes towards an r-selected community in meadow grassland soils

Abstract:Applications of mineral nitrogen (N) and phosphorus (P) fertilizer to grasslands become more frequently in recent decade, aiming to improve the grass production. Nonetheless, the ecological impacts of N and P fertilizer on ecosystem components, especially the belowground soil microbes, are not well acknowledged in grasslands ecosystem. We investigated the responses of plant traits, soil properties, bacterial community composition and potential functions to 5-year N and P fertilization in a mid-high latitude meadow grassland in Inner Mongolia, China. Bacterial species richness and diversity significantly (P < 0.05) decreased under combined N and P fertilization treatment. The overall bacterial community composition was significantly (PERMANOVA, P < 0.05) changed by N and P fertilization. In specific, N and P fertilization increased the relative abundance of the nominated r-strategists (copiotrophs), such as Actiobacteria, Candidatus Saccharibacteria, and Firmicutes; whereas reduced the relative abundance of the putative K-strategists (oligotrophs), including Acidobacteria, Gammaproteobacteria, and Deltaproteobacteria. As a consequence, the ratio of r-strategists to K strategists (r/K ratio) increased under nutrients addition. The shifts towards an r-selected community were further verified by the increased averaged rRNA operon copy number (rrn) of bacterial communigy. Soil N cycling process was inhibited by N fertilization, whereas soil C metabolizing function was accelerated by N and P fertilization. It implies that application of N and P fertilization will increase the risk of C loss in this region. The present study deepens our knowledge on the impacts of N and P fertilization on soil biotic and abiotic parameters in an organic-carbon-rich meadow grassland ecosystem. It provides a reference for designing a sustainable grassland management that can balance the grass yield and environment protection.

Stoichiometric ratios support plant adaption to grazing moderated by soil nutrients and root enzymes

Abstract:Background: Vegetation succession is one of the major driving processes of grassland degradation. Stoichiometry significantly contributes to vegetation dynamics. However, a knowledge gap exists in how soil nutrients and root enzymes influence the stoichiometric ratio to affect vegetation dynamics. Methods: To address these questions, we selected a dominant species (Leymus chinensis (Trin.) Tzvel.) and a degraded-dominant species (Artemisia frigida Willd.) under different management regimes (enclosure and grazing) on the Inner Mongolia steppe. We measured (i) plant nutrient concentrations, (ii) root enzymes and (iii) soil nutrients to investigate how the selected plant species responded to grazing. Results: The results show that: (i) N and P concentrations and the C:N:P ratio in different organs are significantly affected by grazing, and there is variation in the plant species' response. Grazing significantly increased N and P in the leaves and stems of L. chinensis and the stems and roots of A. frigida. (ii) Grazing significantly increased the activities of glutamine synthase but decreased the activities of acid phosphatase in L. chinensis. The nitrate reductase and acid phosphatase activities significantly increased in A. frigida under grazing conditions. (iii) Grazing decreased the total nitrogen, total phosphorus, and available nitrogen, but increased the available phosphorus in the soil. Conclusion: We conclude that A. frigida is better adapted to grazing than L. chinensis, possibly because of its relatively increased stem and root growth, which enhance population expansion following grazing. Conversely, L. chinensis showed increased leaf and stem growth, but suffered nutrient and biomass loss as a result of excessive foraging by livestock, which severely affected its ability to colonize. Root enzymes coupled with soil nutrients can regulate plant nutrients and stoichiometric ratios as an adaptive response to grazing. Thus, we demonstrated that stoichiometric ratios allow species to better withstand grazing disturbances. This study provides a new understanding of the mechanisms involved in grazing-resistance within a plant-soil system.
Decoupling of nitrogen and phosphorus in dominant grass species in response to long-term nitrogen addition in an Alpine Grassland in Central Asia

Abstract:Increased nitrogen (N) availability induced by fertilizer use, rapid urbanization, and livestock cultivation has important effects on the biogeochemical cycles of plant N and phosphorus (P). Knowledge of the long-term N enrichment effects on the biogeochemical cycling of N and P via plant ecological stoichiometry and nutrient resorption remains limited. Nutrient resorption plays an important role in the plant nutrient economy and nutrient cycling. A three-year field experiment was performed to test the effects of N addition on leaf nutrient resorption of two dominant grass species (Leymus tiansecalinus and Festuca ovina) from an 11-year grassland experiment involving four N levels (0, 30, 90, and 150 kg N ha−1 year−1) in an alpine grassland of Tianshan Mountains in northwestern China. Nitrogen addition significantly increased aboveground biomass (AGB) and soil N availability. The N concentrations and N:P ratio in mature and senesced leaves consistently increased with increasing N across all three years. The P concentrations in mature and senesced leaves notably decreased with increasing N. The N addition resulted in decreased N resorption efficiency (NRE) and increased P resorption efficiency (PRE). The divergent responses of plant N and P resorption and N:P ratios resulted in the decoupling of the plant internal nutrient cycles for both grass species, attributed to increased soil N availability and nonsignificant effects on soil available P caused by N addition. In addition, the N:P resorption ratios were negatively correlated with increasing N levels, suggesting different sensitivities of plant N and P to N addition. The aboveground production of both grass species was positively correlated with PRE and negatively correlated with NRE. Under the background of the currently high and steadily increasing atmospheric N deposition, the imbalance of plant nutrient cycling will likely alter plant community compositions and subsequent litter decomposition, ultimately affecting ecosystem stability and function.

Abstract:Vegetation restoration is a critical strategy for addressing ecosystem degradation globally. However, understanding the specific impacts of land-use changes, particularly the conversion of farmland to forestland and grassland, on soil nutrients and microbial biomass in the Loess Plateau remains limited and requires further evaluation. Therefore, this study was conducted to explore how these conversions affect soil organic carbon (SOC), total nitrogen (TN), total phosphorus (TP), and microbial biomass components under various land-use patterns. We studied the SOC, TN, TP, soil microbial biomass carbon (MBC), microbial biomass nitrogen (MBN), microbial biomass phosphorus (MBP) content and their ratios under six land-use patterns (Farmland (FL), Abandoned cropland (ACL), Natural grassland (NG), Alfalfa grassland (Medicago sativa L. (MS)), Spruce forestland (Picea asperata Mast. (PA)) and Cypress forestland (Platycladus orientalis (L.) Franco (PO))). The conversion of FL to grassland and forestland significantly increased C:N and C:P by 9.82%~64.12%, 10.57%~126.05%, and 51.44%~113.40%, 22.10%~116.09%, respectively. The conversion of FL to ACL reduced the C:N and C:P by 5.34%~13.57% and 1.51%~7.55%, respectively. The conversion of FL to NG can increase soil N:P. The conversion of FL to grassland and forestland increased soil MBC, MBN, and MBP by −31.54%~84.48%, −48.39%~1533.93%, −46.55%~173.85%, and −34.96%~17.13%, 68.72%~432.14%, −38.39%~318.46%, respectively. However, the MBC, MBN, and MBP contents in the soil converted from FL to ACL varied from −28.21%~11.95%, 11.17%~531.25%, and −82.64%~70.77%, respectively. Soil SOC, TN, TP, available potassium (AK), pH, and soil bulk density (BD) are the main factors causing microbial biomass differences. These results indicate that converting farmland into forestland and grassland can improve soil nutrient structure and increase soil microbial biomass and carbon accumulation. The results of this study provide theoretical support for the scientific management of regional land.

Abstract:Nitrogen (N) retention is a critical ecosystem function for maintaining soil fertility and mitigating pollution caused by anthropogenic N input. However, it has not yet been elucidated how responses of plant and soil regulate ecosystem N retention. Here, we combined a 14-year N addition experiment in a temperate steppe with a global meta-analysis in grasslands, to assess changes in carbon (C) pool size and stoichiometric C:N ratio of plant and soil components and evaluate the contribution of each component to grassland N retention under increasing N levels. We found that N addition increased N storage in the plant pool by stimulating biomass production and reducing tissue C:N at the community level. However, the non-random loss of forbs and legumes associated with a low C:N ratio partially offset the decline in community-level C:N ratio, thereby diminishing the positive net effect of N enrichment on plant N storage. The observed increase in soil N storage was predominantly determined by the decrease in C:N ratio of topsoil, while no changes were detected in the subsoil. On 14-year time scale, the upper limitation of N retention capacity in our study site was 167.02 g N/m2 . Global meta-analysis further indicated that a decade's N addition significantly increased the N storage in shoot, root and topsoil through enhancing the C pool and decreasing the C:N ratio, while did not affect those of subsoil. However, the positive correlation between the response of subsoil N storage and treatment duration further indicates that, though the accumulation of added N lagged behind that of topsoil, subsoil could play an important role in N retention on a longer time scale. Our study demonstrated that the enhanced plant productivity and altered physiological metabolism indicated by the decreased C:N ratio jointly determined grassland ecosystem N retention. The capacity of the grassland ecosystem to retain exogenous N input is limited, especially for a large amount of N input that occurs in a short period. However, in the context of chronically rising N deposition, the long-term N retention capacity of grasslands should largely depend on the response of subsoil, especially after topsoil N is saturated.

Abstract:The coupling cycles of soil carbon (C), nitrogen (N), and phosphorus (P) have a significant impact on biogeochemical processes and ecosystem services. For centuries, large areas of floodplain wetlands in China have been extensively reclaimed for agricultural purposes due to population growth. However, little is known about the evolution of soil C:N:P stoichiometry along a reclamation chronosequence, particularly across different land uses. In this study, we investigated the variations in soil C:N:P ratios with land use and time gradients along a reclamation chronosequence comprising c. 0, 60, 100, 280, 2000, and 3000 years. Land reclamation induced nutrient decoupling, as it facilitated C and N accumulation from biological processes but restricted P supply controlled by geochemical processes. Soil C and N sequestration reached a stable state after 2000 years, while P declined steadily from 60 years. Soil C/P and N/P increased significantly and were controlled by organic carbon (OC) and total nitrogen (TN), respectively, indicating that an increase in C and N could also promote P uptake. Soil C/N declined in the first 60 years and stabilized at a threshold of 10:1. Different land use patterns following reclamation resulted in distinct soil nutrient structures. Paddies retained more OC and TN but exhibited lower adsorption of total phosphorus (TP) compared to adjacent dryland, leading to significant differences in C/P and N/P between land uses. Based on the redundancy analysis and random forest model, soil OC and TN were mainly affected by the abundance of bacteria metabolizing cellulose, while metal oxides, including Fe2O3 and CaO, could best predict TP. Soil C/P and N/P were mainly driven by soil texture and rose significantly with the increasing proportion of clay particles. Our study suggests that as reclamation proceeds, more anthropogenic management is required to regulate potential nutrient imbalances in order to prevent adverse effects on crop growth, soil quality, and ecosystem health. Additionally, any fertilization strategy should be developed based on dryland C and N deficiencies, and lack of P in paddies.

Abstract:Revegetation influences microbial biomass stoichiometry by altering the substrate conditions, yet the differences in microbial stoichiometry homeostasis and the underlying drivers under different revegetation approaches remain unexplored. Here, we selected sites across three age classes of Robinia pseudoacacia plantation (RP) and abandoned land (AL), and quantified the microbial stoichiometric characteristics during farmland‐initiated restoration. Plant community composition, leaf and soil nutrients, and microbial community composition and diversity were also measured. We found that revegetation of former farmland under both restoration types resulted in non‐isometric changes in soil carbon (C), nitrogen (N), and phosphorus (P) contents, that is, decoupling of soil C and N from P. However, AL and RP succession exhibited homeostatic and plastic microbial biomass stoichiometry, respectively, in response to altered substrate stoichiometry. These differences were associated with adjustments in the above‐ and belowground biomes. Specifically, the synergistic increase of Compositae and Actinobacteria in the late AL succession allowed the ecosystem to reduce P demand and maintain microbial stoichiometric homeostasis. In contrast, higher leaf C and N input during RP succession may have resulted excessive microbial storage of elements, which in turn leads to stoichiometric convergence between microbial biomass and soil resources. In addition, RP succession caused changes in microbial community structure, mainly the continuous increase of Proteobacteria (copiotrophs, r‐strategists), which also potentially increased the requirement for resources to maintain homeostasis and ensure the rapid growth. These findings demonstrate that AL has a comparatively greater efficacy in maintaining microbial stoichiometric homeostasis during long‐term revegetation. Our study also highlights the importance of appropriately managing existing RP plantations to alleviate the pressure of P deficiency and sustainably maintain this fragile ecosystem.

Restoration of ecosystem carbon and nitrogen storage and microbial biomass after grazing exclusion in semi-arid grasslands of Inner Mongolia

Abstract:The semi-arid grasslands in Inner Mongolia have been degraded as a result of long-term grazing and poor grassland management. A series of ecological restoration strategies have been implemented to enhance the ecosystem services of China’s grasslands. To understand the response of ecosystem C and N to grazing exclusion within a nationwide conservation project–Returning Grazing Lands to Grasslands (RGLG), the C and N storages in different parts of the plant–soil system as well as soil microbial biomass carbon (MBC) and nitrogen (MBN) contents were determined and compared between three grazing exclusion sites (6 years) and the paired adjacent grazing sites in the Hulunbuir grassland of Inner Mongolia. Our results showed that the exclusion of grazing for 6 years resulted in significant increases of ecosystem biomass and soil C and N contents, leading to a substantial restoration of ecosystem C and N storage. The average C and N accumulation rates in the whole ecosystem were 173.6gCm−2year−1 and 19.8gNm−2year−1, respectively. The accumulation of C and N in soil contributed to more than 87.6% and 98.2% of the increases in ecosystem C and N, respectively, indicating that the restoration of ecosystem C and N were mainly due to the increase of C and N in soil after exclusion of grazing. The higher MBC and MBN in exclosure sites than grazing sites might be partly because of the enhanced organic matter inputs from plant litter and root exudates. The significant correlations between soil organic matter and microbial biomass suggested that soil microbial biomass could be a sensitive indicator of the soil C and N dynamics. These results indicated that exclusion of grazing pressure within the RGLG project is an effective restoration approach to control grassland degradation in this region. However, more comprehensive multidisciplinary studies are needed to evaluate the effectiveness of the RGLG project and to improve the management strategies for grassland restoration.

Effects of plant functional group loss on soil biota and net ecosystem exchange: a plant removal experiment in the Mongolian grassland

Abstract:The rapid loss of global biodiversity can greatly affect the functioning of above‐ground components of ecosystems. However, how such biodiversity losses affect below‐ground communities and linkages to soil carbon (C) sequestration is unclear. Here, we describe how losses in plant functional groups (PFGs) affect soil microbial and nematode communities and net ecosystem exchange (NEE) in a 4‐year removal experiment conducted on the Mongolian plateau, the world's largest remaining natural grassland. Our results demonstrated that the biomasses or abundances of most components of the two below‐ground communities (microbes and nematodes) were negatively affected by PFG loss and were positively related to above‐ground plant biomass. The removal of dominant PFGs (perennial bunchgrasses and perennial rhizomatous grasses) reduced the biomass or abundance of below‐ground community components while removal of less dominant PFGs (perennial forbs and annuals/biennials) did not change or increased the biomass or abundance of below‐ground community components. The biomass‐based ratio of fungal to bacterial microbes and the number‐based ratio of fungal‐feeding to bacterial‐feeding nematodes decreased with increasing PFG losses. Variation partitioning analyses showed that the identity of PFGs together with above‐ground plant biomass explained most of the total variation in soil microbes and that the identity of PFGs and above‐ground plant biomass together with nematode food resources explained most of the total variation in soil nematodes. The increase in NEE with PFG loss was mainly explained by decreases in above‐ground plant biomass and the ratio of fungi to bacteria. Synthesis. The shift of below‐ground communities from a fungal‐based to a bacterial‐based energy channel as PFG richness decreases indicates that less diverse grassland ecosystems will have lower nutrient retention and hence be more sensitive to land‐use or climate change. The dominant effects of above‐ground plant biomass and below‐ground communities on NEE indicate that PFG loss resulting from land‐use or climate change has the potential to reduce C sequestration in semi‐arid grassland soils. These findings suggest that predictive models may need to consider the composition of above‐ground and below‐ground communities in order to accurately simulate the dynamics of CO2 fluxes in terrestrial ecosystems. Abstract:Microorganisms regulate the responses of terrestrial ecosystems to anthropogenic nutrient inputs. The escalation of anthropogenic activities has resulted in a rise in the primary terrestrial constraining elements, namely nitrogen (N) and phosphorus (P). Nevertheless, the specific mechanisms governing the influence of soil microbial community structure and ecological processes in ecologically vulnerable and delicate semi-arid loess agroecosystems remain inadequately understood. Therefore, we explored the effects of different N and P additions on soil microbial community structure and its associated ecological processes in the farmland of Chinese Loess Plateau based on a 36-a long-term experiment. Nine fertilization treatments with complete interactions of high, medium, and low N and P gradients were set up. Soil physical and chemical properties, along with the microbial community structure were measured in this study. Additionally, relevant ecological processes such as microbial biomass, respiration, N mineralization, and enzyme activity were quantified. To elucidate the relationships between these variables, we examined correlation-mediated processes using statistical techniques, including redundancy analysis (RDA) and structural equation modeling (SEM). The results showed that the addition of N alone had a detrimental effect on soil microbial biomass, mineralized N accumulation, and β-1,4-glucosidase activity. Conversely, the addition of P exhibited an opposing effect, leading to positive influences on these soil parameters. The interactive addition of N and P significantly changed the microbial community structure, increasing microbial activity (microbial biomass and soil respiration), but decreasing the accumulation of mineralized N. Among them, N 24 P 12 treatment showed the greatest increase in the soil nutrient content and respiration. N 12 P 12 treatment increased the overall enzyme activity and total phospholipid fatty acid (PLFA) content by 70.93%. N and P nutrient contents of the soil dominate the microbial community structure and the corresponding changes in hydrolytic enzymes. Soil microbial biomass, respiration, and overall enzyme activity are driven by mineralized N. Our study provides a theoretical basis for exploring energy conversion processes of soil microbial community and environmental sustainability under long-term N and P additions in semi-arid loess areas.

Abstract:Microorganisms are essential components of underground life systems and drive elemental cycling between plants and soil. Yet, in the ecologically fragile Loess Plateau, scant attention has been paid to the response of microbial communities to organic carbon (C) chemistry of both leaves and soils under different revegetation conditions, as well as subsequent alternation in their C metabolic functions. Here, Fourier transform infrared (FTIR) spectrum, amplicon sequencing of 16S rRNA and ITS, and temporal incubation with Biolog-Eco 96 plates were combined to explore the vegetative heterogeneity of microbial community properties and metabolic functions, as well as their regulatory mechanisms in three typical revegetation types including Robinia pseudoacacia L. (RF), Caragana korshinskii KOM. (SL), and abandoned grassland (AG). We observed higher bacterial-to-fungal ratios (B: F = 270.18) and richer copiotrophic bacteria (Proteobacteria = 33.08%) in RF soil than those in AG soil, suggesting that microbes were dominated by r-strategists in soil under RF treatment, which is mainly related to long-term priming of highly bioavailable leaf C (higher proportion of aromatic and hydrophilic functional groups and lower hydrophobicity). Conversely, microbial taxa in AG soil, which was characterized by higher leaf organic C hydrophobicity (1.39), were dominated by relatively more abundant fungi (lower B: F ratio = 149.49) and oligotrophic bacteria (Actinobacteria = 29.30%). The co-occurrence network analysis showed that microbial interactive associations in RF and AG soil were more complex and connective than in SL soil. Furthermore, Biolog-Eco plate experiments revealed that microorganisms tended to utilize labile C compounds (carbohydrates and amino acids) in RF soil and resistant C compounds (polymers) in AG soil, which were consistent with the substrate adaptation strategies of predominant microbial trophic groups in different revegetation environments. Meanwhile, we observed greater microbial metabolic activity and diversity advantages in RF vegetation. Collectively, we suggest that besides the nutrient variables in the leaf-soil system, the long-term regulation of the microbial community by the C chemistry of the leaf sequentially alters the microbial metabolic profiles in a domino-like manner. RF afforestation is more conducive to restoring soil microbial fertility (including microbial abundance, diversity, interactive association, and metabolic capacity). Our study potentially paves the way for achieving well-managed soil health and accurate prediction of C pool dynamics in areas undergoing ecological restoration of the Loess Plateau.

Response of Temperate Leymus chinensis Meadow Steppe Plant Community Composition, Biomass Allocation, and Species Diversity to Nitrogen and Phosphorus Addition

Abstract:Studies on the impacts of fertilization on plant production and species diversity are crucial for better maintaining the stability of grassland ecosystems and restoring degraded grasslands. Using a controlled fertilization experimental platform in a temperate Leymus chinensis meadow steppe ecosystem, the effects of different levels of nitrogen (N) and phosphorus (P) addition on plant community structure, biomass allocation, diversity, and the correlation relationship were explored. The major results were as follows: (1) The structural composition of the plant community changed after different levels of N and P addition; the dominance ratio and biomass of Poaceae plants increased gradually with increasing N and P addition levels. (2) The addition of N and P increased the height, density and coverage of the plant community, the biomass of the dominant L. chinensis and plant community and the total productivity of grassland, and reduced the root–shoot ratio of grassland biomass. For example, plant community biomass, gramineous plant biomass and grassland total productivity increased by 84.46–204.08%, 162.64–424.20%, and 38.12–46.44%, respectively, after N and P addition. (3) The community richness, diversity, and evenness indices decreased overall and showed binomial regression after N and P addition; the functional group of Poaceae plants was highly significantly negatively correlated with species diversity indices and was highly significantly positively correlated with the aboveground biomass of L. chinensis and community; Leguminosae plants and Ranunculaceae plants were highly significantly positively correlated with Margalef and Patrick richness indices; Ranunculaceae plants were highly significantly and negatively correlated with L. chinensis biomass, community biomass, and Poaceae plants. Moderate fertilization not only improved the plant community structure and productivity but was also beneficial for maintaining the grassland species diversity and stability.

Abstract:The knowledge of ecological stoichiometry and stoichiometric homeostasis could contribute to exploring the balance of chemical elements in ecological recovery. However, it is largely unknown how the carbon (C), nitrogen (N), phosphorus (P), and stoichiometric characteristics in the plant-soil-microbe continuum system respond to the spontaneous secondary succession of degraded alpine grasslands. Therefore, we investigated the spontaneous secondary successional recovery of grasslands disturbed by zokor (Myospalax fontanierii) on the Qinghai-Tibetan Plateau, China, via a strategy of substituting space for time. Based on plant richness, biomass, and coverage, plant importance value was employed to assess the recovery degree of zokor-made mounds (ZMMs, large and bare patch areas constructed by zokors). Multiple statistical methods, including stoichiometric homeostatic model, network, and redundancy analysis, were conducted to decipher the stoichiometric patterns. The results indicated that plant C, C:N, and C:P increased with the recovery of ZMMs, contrary to the decrease of plant N and P. In addition, soil C, N, C:N, C:P, and N:P increased with the recovery degree, and the soil became relatively more N rich by increasing organic N under the revegetation of legumes. Meanwhile, soil microbial biomass C, N, and P increased with the recovery of ZMMs, but microbial biomass C:N:P ratios were highly constrained. Soil accessible inorganic nitrogen played an important role in driving plant and microbial nutrient and stoichiometry. Our results demonstrated that the different responses of C, N, and P contents in plant-soil-microbe lead to shifts in C:N:P stoichiometric ratio. Nevertheless, plants and soil microbes exhibited strong stoichiometric homeostasis. Collectively, our study provides new insight into biogeochemical responses to the successional recovery of degraded alpine grassland on the Qinghai-Tibetan Plateau from a stoichiometric perspective.
