Abstract:Xylitol is a five-carbon sugar alcohol with potential for use as a sweetener. Industrially, xylitol is currently produced by chemical hydrogenation of D-xylose using Raney nickel catalysts and this requires expensive separation and purification steps as well as high pressure and temperature that lead to environmental pollution. Highly efficient biotechnological production of xylitol using microorganisms is gaining more attention and has been proposed as an alternative process. Although the biotechnological method has not yet surpassed the advantages of chemical reduction in terms of yield and cost, various strategies offer promise for the biotechnological production of xylitol. In this review, the focus is on the most recent developments of the main metabolic engineering strategies for improving the production of xylitol.

Abstract:Xylitol as a five carbon sugar alcohol and low caloric content,has been widely used in medicine,food and chemical fields.Compared with chemical synthesis method,biological synthesis method has become a research focus because of its mild operation conditions,low energy consumption,and little environmental pollution.However,there also exist the leading obstacles to the commercial production of xylitol by bioconversion,such as the different xylose metabolic pathways existed in natural microorganisms,nonspecific nature of xylose reductase(XR),and requiring nicotinamide cofactors(NADH and NADPH) in biosynthesis of xylitol.Based on reviewing and analyzing the current understanding on biocatalytic routes to xylitol production,novel genetic engineering strategies through improving metabolic pathways of microorganisms which can improve the biological catalytic conversion of xylitol yield were emphasised.

Abstract:Xylitol is the first rare sugar that has global market because of its excellent properties. Considering its superiority to chemosynthesis, biosynthesis of xylitol became hot issue in recent studies. The production of xylitol from glucose experienced a development from three-step process to two-step process, or even only one-step process. The microbial and enzymatic process involving key enzymes, molecular cloning and expression and transgenic bacteria construction is introduced in this paper. This study may provide novel thought to explore new resource for better control of biological reaction conditions and obtainment of higher xylitol yield.

Abstract:Cofactor supply is a rate-limiting step in the bioconversion of xylose to xylitol. Strain WZ04 was first constructed by a novel simultaneous deletion-insertion strategy, replacing ptsG, xylAB and ptsF in wild-type Escherichia coli W3110 with three mutated xylose reductase genes (xr) from Neurospora crassa. Then, the pfkA, pfkB, pgi and/or sthA genes were deleted and replaced by xr to investigate the influence of carbon flux toward the pentose phosphate pathway and/or transhydrogenase activity on NADPH generation. The deletion of pfkA/pfkB significantly improved NADPH supply, but minimally influenced cell growth. The effects of insertion position and copy number of xr were examined by a quantitative real-time PCR and a shake-flask fermentation experiment. In a fed-batch fermentation experiment with a 15-L bioreactor, strain WZ51 produced 131.6 g L-1 xylitol from hemicellulosic hydrolysate (xylitol productivity: 2.09 g L-1 h(-1)). This study provided a potential approach for industrial-scale production of xylitol from hemicellulosic hydrolysate.

Abstract:A microbial and enzymatic process for xylitol production that initiates from D-glucose occurs by virtue of a three-step process in which D-arabitol is the first intermediate product and D-xylulose is the second. To date, the steps of the formation of D-arabitol and the conversion of xylitol from D-xylulose are key points of departure for promoting the overall process for xylitol production from D-glucose. This review provides comprehensive insights regarding potential methods of xylitol production from D-glucose, which might be an alternative to the classical process that utilizes D-xylose as substrate.

Abstract:Cofactor availability is often a rate-limiting factor in the bioconversion of xylose to xylitol. The overexpression of pentose phosphate pathway genes and the deletion of Embden-Meyerhof-Parnas pathway genes can modulate the glucose metabolic flux and increase the intracellular NADPH supply, enabling Escherichia coli cells to produce xylitol from corncob hydrolysates. The effects of zwf and/or gnd overexpression and pfkA, pfkB, and/or pgi deletion on the intracellular redox environment and xylitol production were examined. The NADPH-enhanced strain 2bpgi produced 162 g/L xylitol from corncob hydrolysates after a 76 h fed-batch fermentation in a 15 L bioreactor, which was 13.3% greater than the 143 g/L xylitol produced by the IS5-d control strain. Additionally, the xylitol productivity and xylitol yield per glucose for 2bpgi were 2.13 g/L/h and 2.50 g/g, respectively. Thus, the genetic modifications in 2bpgi significantly enhanced NADPH regeneration, making 2bpgi a potentially useful strain for the industrial-scale production of xylitol from detoxified corncob hydrolysates.

Abstract:Xylitol is a white crystalline, amorphous sugar alcohol and low-calorie sweetener. Xylitol prevents demineralization of teeth and bones, otitis media infection, respiratory tract infections, inflammation and cancer progression. NADPH generated in xylitol metabolism aid in the treatment of glucose-6-phosphate deficiency-associated hemolytic anemia. Moreover, it has a negligible effect on blood glucose and plasma insulin levels due to its unique metabolism. Its diverse applications in pharmaceuticals, cosmetics, food and polymer industries fueled its market growth and made it one of the top 12 bio-products. Recently, xylitol has also been used as a drug carrier due to its high permeability and non-toxic nature. However, it become a challenge to fulfil the rapidly increasing market demand of xylitol. Xylitol is present in fruit and vegetables, but at very low concentrations, which is not adequate to satisfy the consumer demand. With the passage of time, other methods including chemical catalysis, microbial and enzymatic biotransformation, have also been developed for its large-scale production. Nevertheless, large scale production still suffers from high cost of production. In this review, we summarize some alternative approaches and recent advancements that significantly improve the yield and lower the cost of production.

Abstract:Erythritol, as a new type of natural sweetener, has been widely used in food, medical, cosmetics, pharmaceutical and other fields due to its unique physical and chemical properties and physiological functions. In recent years, with the continuous development of strategies such as synthetic biology, metabolic engineering, omics-based systems biology and high-throughput screening technology, people's understanding of the erythritol biosynthesis pathway has gradually deepened, and microbial cell factories with independent modification capabilities have been successfully constructed. In this review, the cheap feedstocks for erythritol synthesis are introduced in detail, the environmental factors affecting the synthesis of erythritol and its regulatory mechanism are described, and the tools and strategies of metabolic engineering involved in erythritol synthesis are summarized. In addition, the study of erythritol derivatives is helpful in expanding its application field. Finally, the challenges that hinder the effective production of erythritol are discussed, which lay a foundation for the green, efficient and sustainable production of erythritol in the future and breaking through the bottleneck of production.

Abstract:Xylitol production from lignocellulose-hydrolyzed glucose and xylose is a sustainable and environment-friendly process. In this study, Kluyveromyces marxianus YZB194, constructed by overexpressing ScGAL2N376F from Saccharomyces cerevisiae and NcXYL1 from Neurospora crassa, and deleting GPD1, KU70, PGI1, and XYL2, was used for xylitol production by co-utilizing glucose and xylose. By deleting PGI1, the glucose repression effect on xylose in K. marxianus was released, besides, glucose metabolism in YZB194 was refactored through the pentose phosphate pathway, a pathway yielding the largest supply of NADPH in yeast. The metabolic pathway of xylitol was blocked by deleting of XYL2. By simultaneously co-utilizing 70 g/L glucose and 140 g/L xylose, 139.96 g/L xylitol with a productivity of 0.83 g/L/h was obtained by YZB194 at 37 °C under aerobic conditions. Through fed-batch fermentation in a fermenter at 42 °C, 203.57 g/L xylitol with a productivity of 2.99 g/L/h and yield on xylose of 0.99 g/g were obtained using K. marxianus YZB194. A platform for xylitol production by co-utilizing glucose and xylose via rewriting the glucose metabolism pathway was implemented.

Abstract:Xylitol classified as an A-level food additive by the FAO and WHO, is being increasingly used in the food and pharmaceutical products with a predicted market worth of US$ 1 billion. Xylitol is commercially produced by catalytic hydrogenation of pure xylose which is a cost intensive method. Biotechnological methods using microbes offer a more sustainable approach but face challenges in purification and recovery due to the complex composition of fermentation broth. Crystallization emerges as a viable method for purifying xylitol in both chemical and biological processes, offering advantages such as high purity, uniform crystal sizes, and low operating costs. The present review aims to provide a comprehensive compilation of physico-chemical factors influencing xylitol crystallization, including nucleation, crystal growth rates, and crystal morphology, alongside techno-economic considerations. By addressing research gaps on xylitol crystallization, this review enhances the understanding of downstream processing of bioprocessed xylitol, to meet its increasing demand.

Abstract:In this study, four engineered Saccharomyces cerevisiae carrying xylanase, β-xylosidase and xylose reductase genes by different transcriptional regulations were constructed to directly convert xylan to xylitol. According to the results, the high-copy number plasmid required a rigid selection for promoter characteristics, on the contrast, the selection of promoters could be more flexible for low-copy number plasmid. For cell growth and xylitol production, glucose and galactose were found more efficient than other sugars. The semi-aerobic condition and feeding of co-substrates were taken to improve the yield of xylitol. It was found that the strain containing high-copy number plasmid had the highest xylitol yield, but it was sensitive to the change of fermentation. However, the strain carrying low-copy number plasmid was more adaptable to different processes. By optimization of the transcriptional regulation and fermentation processes, the xylitol concentration could be increased of 1.7 folds and the yield was 0.71 g xylitol/g xylan.

Abstract:Xylitol, a naturally occurring five-carbon sugar alcohol derived from d-xylose, is currently in high demand by industries. Trichoderma reesei, a prolific industrial cellulase and hemicellulase producing fungus, is able to selectively use d-xylose from hemicelluloses for xylitol production. The xylitol production by T. reesei can be enhanced by genetic engineering of blocking further xylitol metabolism in the d-xylose pathway. We have used two different T. reesei strains which are impaired in the further metabolism of xylitol including a single mutant in which the xylitol dehydrogenase gene was deleted (∆xdh1) and a double mutant where additionally l-arabinitol-4-dehydrogenase, an enzyme which can partially compensate for xylitol dehydrogenase function, was deleted (∆lad1∆xdh1). Barely straw was first pretreated using NaOH and Organosolv pretreatment methods. The highest xylitol production of 6.1 and 13.22 g/L was obtained using medium supplemented with 2 % Organosolv-pretreated barley straw and 2 % d-xylose by the ∆xdh1 and ∆lad1∆xdh1 strains, respectively.

Abstract:Lignocellulose is the most abundant renewable resource on earth. Constructing microbial cell factories for synthesizing value-added chemicals with lignocellulose is the key to realize green biomanufacturing. Xylose is the second most fermentable sugar in lignocellulose after glucose. Building microbial cell factories that can efficiently metabolize xylose is of great significance to achieve full utilization of lignocellulose. However, the lower metabolism efficiency of xylose than that of glucose in most microorganisms limits the application of xylose. In recent years, the deepening understanding of microbial metabolic mechanisms and the continuous advancement of synthetic biology have greatly improved the efficiency of microbial metabolism of xylose and expanded the spectrum of xylose-derived products. This article introduces several xylose metabolic pathways that exist in the nature and the derived products, summarizes the strategies for constructing recombinant strains that can co-utilize xylose and glucose, and reviews the research progress in the application of lignocellulose hydrolysates in the synthesis of target products. Finally, this article discusses the current technical bottlenecks and prospects the future development directions in this field.

Abstract:Lignocellulosic biomass, encompassing cellulose, lignin and hemicellulose in plant secondary cell walls (SCWs), is the most abundant source of renewable materials on earth. Currently, fast-growing woody dicots such as Eucalyptus and Populus trees are major lignocellulosic (wood fiber) feedstocks for bioproducts such as pulp, paper, cellulose, textiles, bioplastics and other biomaterials. Processing wood for these products entails separating the biomass into its three main components as efficiently as possible without compromising yield. Glucuronoxylan (xylan), the main hemicellulose present in the SCWs of hardwood trees carries chemical modifications that are associated with SCW composition and ultrastructure, and affect the recalcitrance of woody biomass to industrial processing. In this review we highlight the importance of xylan properties for industrial wood fiber processing and how gaining a greater understanding of xylan biosynthesis, specifically xylan modification, could yield novel biotechnology approaches to reduce recalcitrance or introduce novel processing traits. Altering xylan modification patterns has recently become a focus of plant SCW studies due to early findings that altered modification patterns can yield beneficial biomass processing traits. Additionally, it has been noted that plants with altered xylan composition display metabolic differences linked to changes in precursor usage. We explore the possibility of using systems biology and systems genetics approaches to gain insight into the coordination of SCW formation with other interdependent biological processes. Acetyl-CoA, s-adenosylmethionine and nucleotide sugars are precursors needed for xylan modification, however, the pathways which produce metabolic pools during different stages of fiber cell wall formation still have to be identified and their co-regulation during SCW formation elucidated. The crucial dependence on precursor metabolism provides an opportunity to alter xylan modification patterns through metabolic engineering of one or more of these interdependent pathways. The complexity of xylan biosynthesis and modification is currently a stumbling point, but it may provide new avenues for woody biomass engineering that are not possible for other biopolymers.

Abstract:Xylitol is one of the value-added polyalcohols with diverse applications in the food, nutraceuticals, cosmetic, and pharmaceutical industries. Commercial xylitol production is extensively carried out via chemical technology through catalytic xylose dehydrogenation under high temperatures and pressure. The biotechnological synthesis of xylitol from lignocellulosic biomass is a promising and sustainable substitute for chemical xylitol synthesis, which requires strong reaction conditions and removal of the undesirable coproducts produced during the process. Numerous reviews target culture conditions and metabolic pathways in recombinant and wild xylitol-producing organisms for enhanced xylitol accretion. However, fewer studies focus on the engineering and bioprocess aspects essential for large-scale xylitol production. This review emphasizes recent advancements in xylitol bioproduction using immobilization methods. Cell immobilization improves biocatalyst reusability, stability, and tolerance against inhibitors, decreases process costs, and denotes a fundamental prerequisite for industrial application. The main immobilization techniques, mode of operations, and bioreactors employed for xylitol bioproduction using immobilization have been comprehensively summarized in this review.

Abstract:Sorbitol is a polyol widely used in the food industry,pharmaceutical industry and other chemical related industries,not only as a sweetener but also as a humectant,texturizer,and softener.With the increasing demand of sorbitol,the biological production of sorbitol attracted a lot of attention in recent years.This article provides an overview on the biological prudction of sorbitol by the bacterium Zymomonas mobilis,the Z.moblilis is able to produce sorbitol and gluconic acid from fructose and glucose respectively,and this process is finished in a one-step reaction via a glucose-fructose oxidoreductase.Different methods which improve the productivity and economic efficiency for sorbitol production are discussed,besides,the properties,applications,market and possible industrial biological production are also discussed.

Abstract:A metabolically engineered Escherichia coli has been constructed for the production of xylitol, one of the top 12 platform chemicals from agricultural sources identified by the US Department of Energy. An optimal plasmid was constructed to express xylose reductase from Neurospora crassa with almost no inclusion bodies at relatively high temperature. The phosphoenolpyruvate-dependent glucose phosphotransferase system (ptsG) was disrupted to eliminate catabolite repression and allow simultaneous uptake of glucose and xylose. The native pathway for D-xylose catabolism in E. coli W3110 was blocked by deleting the xylose isomerase (xylA) and xylulose kinase (xylB) genes. The putative pathway for xylitol phosphorylation was also blocked by disrupting the phosphoenolpyruvate-dependent fructose phosphotransferase system (ptsF). The xylitol producing recombinant E. coli allowed production of 172.4gL−1 xylitol after 110h of fed-batch cultivation with an average productivity of 1.57gL−1h−1. The molar yield of xylitol to glucose reached approximately 2.2 (mol xylitol mol−1 glucose). Furthermore, the recombinant strain also produced about 150gL−1 xylitol from hemicellulosic sugars in modified M9 minimal medium and the overall productivity was 1.40gL−1h−1, representing the highest xylitol concentration and productivity reported to date from hemicellulosic sugars using bacteria. Thus, this engineered E. coli is a candidate for the development of efficient industrial-scale production of xylitol from hemicellulosic hydrolysate.

Abstract:BackgroundThe process of industrial xylitol production is a massive source of organic pollutants, such as waste xylose mother liquor (WXML), a viscous reddish-brown liquid. Currently, WXML is difficult to reuse due to its miscellaneous low-cost sugars, high content of inhibitors and complex composition. WXML, as an organic pollutant of hemicellulosic hydrolysates, accumulates and has become an issue of industrial concern in China. Previous studies have focused only on the catalysis of xylose in the hydrolysates into xylitol using one strain, without considering the removal of other miscellaneous sugars, thus creating an obstacle to subsequent large-scale purification. In the present study, we aimed to develop a simple one-pot biotransformation to produce high-purity xylitol from WXML to improve its economic value.ResultsIn the present study, we developed a procedure to produce xylitol from WXML, which combines detoxification, biotransformation and removal of by-product sugars (purification) in one bioreactor using two complementary strains, Candida tropicalis X828 and Bacillus subtilis Bs12. At the first stage of micro-aerobic biotransformation, the yeast cells were allowed to grow and metabolized glucose and the inhibitors furfural and hydroxymethyl furfural (HMF), and converted xylose into xylitol. At the second stage of aerobic biotransformation, B. subtilis Bs12 was activated and depleted the by-product sugars. The one-pot process was successfully scaled up from shake flasks to 5, 150 L and 30 m3 bioreactors. Approximately 95 g/L of pure xylitol could be obtained from the medium containing 400 g/L of WXML at a yield of 0.75 g/g xylose consumed, and the by-product sugars glucose, l-arabinose and galactose were depleted simultaneously.ConclusionsOur results demonstrate that the one-pot procedure is a viable option for the industrial application of WXML to produce value-added chemicals. The integration of complementary strains in the biotransformation of hemicellulosic hydrolysates is efficient under optimized conditions. Moreover, our study of one-pot biotransformation also provides useful information on the combination of biotechnological processes for the biotransformation of other compounds.

Abstract:Microbial production of chemicals from renewable biomass has emerged as a crucial route for sustainable bio-manufacturing. Lignocellulose with a renewable property and wide sources is supposed to be a promising feedstock for the second-generation biorefinery. The efficient co-utilization of mixed sugars from lignocellulosic hydrolysates represents one of the key challenges in reducing the production cost. However, most microorganisms prefer glucose over xylose due to carbon catabolite repression, which constrains the efficiency of lignocellulosic conversion. Therefore, developing the microbial platforms capable of simultaneously utilizing glucose and xylose is paramount for economically viable industrial-scale production. This article reviews the key strategies and studies of metabolic engineering for promoting efficient co-utilization of glucose and xylose by microorganisms. The representative strategies include relieving glucose repression, enhancing xylose transport, constructing xylose metabolic pathways, and directed evolution.
