Abstract:The sterile insect technique may prove useful for the suppression of mosquito vectors of medical importance in regions where arboviruses pose a serious public health threat. In the present study, we examined the effects of sterilizing irradiation doses across different ratios of fertile:irradiated males on the mating competitiveness of Ae. aegypti and Ae. albopictus under laboratory and field-cage conditions. For both species, the percentage of females inseminated and the number of eggs laid over two gonotrophic cycles varied significantly in mating treatments involving 1:1, 1:5, and 1:10 fertile:irradiated males compared to controls of entirely fertile or entirely irradiated males but was not generally affected by the irradiation dose. Egg hatching was negatively affected in females exposed to increasing proportions of irradiated males in both laboratory and field cages. Male competitiveness (Fried’s index) values varied from 0.19 to 0.58 in the laboratory and were between 0.09 and 1.0 in field cages, depending on th species. Competitiveness values were negatively affected by th eirradiation dose in both species under field-cage conditions, whereas in the laboratory, Ae. albopictus was sensitive to the dose but Ae. aegypti was not. In general, male competitiveness was similar across all mating regimes. Most importantly, induced egg sterility was positively correlated with the proportion of irradiated males present in the mating treatments, reaching a maximum of 88% under field-cage conditions for both Ae. aegypti and Ae. albopictus males treated with 50 and 40 Gy irradiation, respectively. These results indicate that sterile males produced at our facility are suitable and competitive enough for field pilot SIT projects and provide guidance to decide the optimal sterile:fertile ratios.

Abstract:The Sterile Insect Technique (SIT) is a method of pest control used in the management of Ceratitis capitata (Medfly). This technique is based on the mating and transfer of ejaculate sperm to the wild female population from released sterile males. This transfer will induce sterility in the female through sperm dominant lethal mutations. Consequently, the factors influencing sperm transfer and sperm storage are issues that must be understood to enable successful SIT implementation. Age, size, nutritional status, copula duration, and irradiation are the most important factors that affect sperm transfer and storage. Inconsistent results from studies investigating the influence of these factors on sperm transfer/storage may be due to, the strain of fly studied and variability in the sexual experience of the flies used. In this review, we highlight the interaction between these factors, issues that still need to be addressed, and propose future directions for the improvement of medfly males’ sexual performance and the successful application of SIT against this pest.

Age-dependent variation in mating success of sterile male Mediterranean fruit flies (Diptera: Tephritidae): implications for sterile insect technique

Abstract:The sterile insect technique (SIT) is widely used in integrated programs against the Mediterranean fruit fly, Ceratitis capitata (Wiedemann) (Diptera: Tephritidae). Unfortunately, the mass-rearing procedures inherent to the SIT often lead to a reduction in the mating ability of the released males. To counter this deficiency, SIT programs rely upon the production and release of large numbers of sterile males to achieve high overflooding (sterile:wild male) ratios. To ensure a high release volume, emergence facilities release adult males at a young age (2 d old in some cases). The primary objective of this study was to describe age-dependent variation in the mating propensity and competitiveness of sterile males of C. capitata. Males that were 2 or 3 d old had lower mating propensity than males that were > or =4 d old, and 3-d-old males had lower mating competitiveness than males that were > or =4 d old. Given these results, we measured the effect of a longer holding period on male mortality in storage boxes. With delayed food placement, males held in storage boxes for 4 d after emergence showed no higher mortality than males held for only 2 d (the standard interval). Using large field enclosures, we compared the levels of egg sterility attained via releases of 2- versus 4-d-old sterile males at two overflooding ratios (5:1 and 100:1). At the lower ratio, the proportion of unhatched eggs observed for trials involving 2-d-old sterile males was not, on average, significantly higher than that observed for matings between wild flies (33 versus 25%, respectively), whereas the level of egg sterility observed for releases of 4 d old sterile males was 62%. At the 100:1 overflooding ratio, the proportion of unhatched eggs associated with the 2-d-old sterile males was 58%, a level not significantly different from that induced by 4-d-old sterile males at the 5:1 ratio and significantly lower than the level (79%) observed for 4-d-old sterile males at 100:1 overflooding ratio. The implications of these results for SIT are discussed.

Abstract:Aedes aegypti is an invasive mosquito species and major vector of human arboviruses. A wide variety of control methods have been employed to combat mosquito populations. One of them is the sterile insect technique (SIT) that has recently attracted considerable research efforts due to its proven record of success and the absence of harmful environmental footprints. The efficiency and cost-effectiveness of SIT is significantly enhanced by male-only releases. For mosquito SIT, male-only releases are ideally needed since females bite, blood-feed and transmit the pathogens. Ae. aegypti genetic sexing strains (GSS) have recently become available and are based on eye colour mutations that were chosen as selectable markers. These genetic sexing strains were developed through classical genetics and it was shown to be subjected to genetic recombination, a phenomenon that is not suppressed in males as is the case in many Diptera. The genetic stability of these GSS was strengthened by the induction and isolation of radiation-induced inversions. In this study, we used the red eye mutation and the inversion Inv35 line of the Ae. aegypti red-eye GSS s and introgressed them in six different genomic backgrounds to develop GSS with the respective local genomic backgrounds. Our goal was to assess whether the recombination frequencies in the strains with and without the inversion are affected by the different genomic backgrounds. In all cases the recombination events were suppressed in all Inv35 GSS strains, thus indicating that the genomic background does not negatively affect the inversion result. Absence of any effect that could be ascribed to genetic differences, enables the introgression of the key elements of the GSS into the local genomic background prior to release to the target areas. Maintaining the local background increases the chances for successful matings between released males and wild females and addresses potential regulatory concerns regarding biosafety and biosecurity.

Abstract:Sterile insect technique (SIT) is a promising, environmentally friendly alternative to the use of pesticides for insect pest control. However, implementing SIT with Aedes aegypti (Linnaeus) mosquitoes presents unique challenges. For example, during transport from the rearing facility to the release site and during the actual release in the field, damage to male mosquitoes should be minimized to preserve their reproductive competitiveness. The short flight range of male Ae. aegypti requires elaborate release strategies such as release via Unmanned Aircraft Systems, more commonly referred to as drones. Two key parameters during transport and release are storage temperature and compaction rate. We performed a set of laboratory experiments to identify the optimal temperatures and compaction rates for storage and transport of male Ae. aegypti. We then conducted shipping experiments to test our laboratory-derived results in a 'real-life' setting. The laboratory results indicate that male Ae. aegypti can survive at a broad range of storage temperatures ranging from 7 to 28°C, but storage time should not exceed 24 h. Male survival was high at all compaction rates we tested with a low at 40 males/cm3. Interestingly, results from our 'real-life' shipping experiment showed that high compaction rates were beneficial to survival. This study advances key understudied aspects of the practicalities of moving lab-reared insects into the field and lies the foundation for further studies on the effect of transport conditions on male reproductive fitness.

Abstract:Mosquito Sterile Insect Technique (SIT) programs can be developed in smaller agencies through synchronization of the colony development to take advantage of the natural male early emergence. This paper examined key aspects of Ae. aegypti colony synchronization work, including egg hatching, larval development, and adult emergence to produce sufficient numbers of adult male mosquitoes within a specific timeframe for irradiation and release. Our data indicated that a relatively low percentage of males are required for colony propagation. Additional results highlighted that fresher Ae. aegypti eggs could yield as high as a 93% hatching success than older eggs when placed under vacuum pressure in yeast infused water for 1.5 hours. Eggs that were one-month old hatched (93%) better than older eggs (0-32%). A higher egg density in the hatching flask was correlated to a lower hatch rate, and higher larval density was related to unsynchronized pupae and delayed adult emergence. By keeping Ae. aegypti larvae at reasonable density, over 95% of adults emerged on the first two days of emergence - indicating a high synchronicity. A standardized colony maintenance protocol therefore renders a synchronized larval development and adult male emergence which are critical in SIT programs.

Abstract:The sterile insect technique (SIT) application, as an alternative tool for conventional mosquito control methods, has recently gained prominence. Nevertheless, some SIT components require further development, such as protocols under large-scale conditions, focusing on packing and shipping mosquitoes, and considering transporting time. Immobilization of Aedes aegypti males was tested at temperatures 4, 7, 10, and 14 °C, and each temperature was assessed for 60, 90, and 120 min. The recovery after 24 h was also studied. Chilled and control-reared males had comparable survival rates for all conditions, although 4 °C for 120 min impacted male survival. The male escape rate was affected after 60 min of exposure at 4 °C; this difference was not significant, with 24 h of recovery. First, we defined the successful immobilization at 4 °C for 60 min, thus enabling the evaluation of two transportation intervals: 6 and 24 h, with the assessment of different compaction densities of 100 and 150 mosquitoes/cm3 at 10 °C to optimize the shipment. Compaction during simulated mosquito shipments reduced survival rates significantly after 6 and 24 h. In the mating propensity and insemination experiments, the sterile males managed to inseminate 40 to 66% for all treatments in laboratory conditions. The male insemination propensity was affected only by the highest compaction condition concerning the control. The analysis of the densities (100 and 150 males/cm3) showed that a higher density combined with an extended shipment period (24 h) negatively impacted the percentage of inseminated females. The results are very helpful in developing and improving the SIT packing and shipment protocols. Further studies are required to evaluate all combined parameters' synergetic effects that can combine irradiation to assess sexual competitiveness when sterile males are released into the field.

Abstract:BackgroundMating is a physiological process of crucial importance underlying the size and maintenance of mosquito populations. In sterile and incompatible insect technologies (SIT and IIT), mating is essential for mass production, persistence, and success of released individuals, and is a central parameter for judging the effectiveness of SIT/IIT programs. Some mosquitoes have an enormous reproductive potential for both themselves and pathogens and mating may contribute to persistence of infection in nature. As Aedes albopictus can transmit flaviviruses both sexually and horizontally, and as infected insects are usually derived from laboratory colonies, we investigated the implications of mating between a long-term laboratory colony of Ae. albopictus and wild populations.MethodsThrough a series of mating experiments, we examined the reproductive outcomes of sexual cross-affinity between laboratory-raised and wild adults of Ae. albopictus.ResultsThe results indicated appreciable mating compatibility between laboratory-reared and wild adults, and equivalent levels of egg production among reciprocal crosses. We also observed comparable larval eclosion in lab females mated with wild males, and increased adult longevity in female offspring from wild females|×|laboratory males crosses.ConclusionsTaken together, these data suggest that Ae. albopictus can preserve its reproductive fitness over a long period of time in the laboratory environment and has valuable attributes for SIT application. These observations together with the ability to successfully inseminate heterospecific females indicate the potential of Ae. albopictus to act as an ecological barrier if non-sterilized males are massively released in areas occupied by Aedes aegypti. The observed substantial reproductive fitness combined with the capability to reproduce both, itself and viruses illustrates the potential of Ae. albopictus to pose a serious threat if infected and released accidentally.

Abstract:ABSTRACTBackgroundThe sterile insect technique (SIT), which is based on irradiation-induced sterility, and incompatible insect technique (IIT), which is based on Wolbachia-induced cytoplasmic incompatibility (a kind of male sterility), have been used as alternative methods to reduce mosquito vector populations. Both methods require the release of males to reduce fertile females and suppress the number of natural populations. Different techniques of sex separation to obtain only males have been investigated previously. Our work involves an application of mechanical larval-pupal glass separators to separate Wolbachia-infected Aedes aegypti males from females at the pupal stage, prior to irradiation, and for use in a pilot field release and to assess the quality of males and females before and after sex separation and sterilization.ResultsThis study was the first to demonstrate the efficiency of mechanical glass separators in separating males for use in an Ae. aegypti suppression trial by a combined SIT/IIT approach. Our results indicated that male and female pupae of Wolbachia-infected Ae. aegypti mosquitoes were significantly different (p < 0.05) in weight, size, and emergence-time, which made it easier for sex separation by this mechanical method. During the pilot field release, the percentage of female contamination was detected to be quite low and significantly different between the first (0.10 ± 0.13) and the second (0.02 ± 0.02) twelve-week period. Both males and females were almost completely sterile after exposure to 70 Gy irradiation dose. We observed that both irradiated Wolbachia-infected males and females survived and lived longer than two weeks, but males could live longer than females (p < 0.05) when they were irradiated at the same irradiation dose. When comparing irradiated mosquitoes with non-irradiated ones, there was no significant difference in longevity and survival-rate between those males, but non-irradiated females lived longer than irradiated ones (p < 0.05).ConclusionMechanical sex separation by using a larval-pupal glass separator was practically applied to obtain only males for further sterilization and open field release in a pilot population suppression trial of Ae. aegypti in Thailand. Female contamination was detected to be quite low, and skilled personnel can reduce the risk for female release. The irradiated Wolbachia-infected females accidentally released were found to be completely sterile, with shorter life span than males.

Improving Male Mating Competitiveness and Survival in the Field for Medfly, Ceratitis Capitata(Diptera: Tephritidae) SIT Programs

Abstract:The success of the sterile insect technique (SIT) depends critically upon mating between released sterilized males and wild females. In Hawaii, improvements in the efficiency of sterile males were attempted on two separate fronts – mating enhancement and survival improvement. In the former, two methods have been investigated – selective breeding and aromatherapy. In the latter, flies which survived in field cages for several days were selected and bred to produce progeny with enhanced survival ability compared to control flies. Regarding mating selection, standard laboratory-reared males that successfully mated with wild females in field cages were allowed to breed. F1 offspring were inbred, then the selection procedure was repeated for four additional cycles. In the aromatherapy procedure, laboratory-reared males were exposed to ginger root oil for several hours 1 day prior to testing in field cages. Compared to controls, the selected flies improved the mating competitiveness of male flies ca. 3-fold, irradiation reduced this increase to ca. 2.5-fold. Exposing the selected, hybrid strain raised the fitness of the lab males to ca. 9-fold that of wild males. In the ongoing survival selection study, we have obtained lines in which the selected males survived ca. 2-fold better than laboratory control males over several days in an outdoor field cage, with food and water provided. The goal is to combine the traits of higher survival and mating ability into a single strain for SIT release.

Abstract:Three indigo-reducing obligately alkaliphilic strains, M3(T), 41A and 41C, were isolated. The isolates grew at pH 9-12, but not at pH 7-8. They were Gram-positive, facultatively anaerobic, straight rod-shaped strains with peritrichous flagella. The isolates grew in 0-14% (w/v) NaCl, with optimum growth at 3-13%. They grew at temperatures between 10 and 45 degrees C, with optimum growth at around 30-37 degrees C. They did not hydrolyse starch or gelatin. DL-lactate was the major end-product from D-glucose. No quinones could be detected. The peptidoglycan type was A4beta, Orn-d-Asp. The major cellular fatty acids were C(16:0), C(16:1)7c and C(18:1)9c. The DNA G+C content was 42.6-43.2 mol%. Phylogenetic analysis based on 16S rRNA gene sequence data indicated that the isolates belong to the genus Alkalibacterium. DNA-DNA hybridization revealed low similarity (less than 16%) of the isolates with respect to the two closest phylogenetically related strains, Alkalibacterium olivapovliticus and Alkalibacterium psychrotolerans. On the basis of phenotypic and chemotaxonomic characteristics, phylogenetic data and DNA-DNA relatedness, the isolates merit classification as a novel species of the genus Alkalibacterium, for which the name Alkalibacterium iburiense is proposed. The type strain is M3(T) (=JCM 12662(T)=NCIMB 14024(T)).
On the impact of re-mating and residual fertility on the Sterile Insect Technique efficacy: Case study with the medfly, Ceratitis capitata

Abstract:The sterile insect technique (SIT) can be an efficient solution for reducing or eliminating certain insect pest populations. It is widely used in agriculture against fruit flies, including the Mediterranean fruit fly (medfly), Ceratitis capitata. The re-mating tendency of medfly females and the fact that the released sterile males may have some residual fertility could be a challenge for the successful implementation of the SIT. Obtaining the right balance between sterility level and sterile male quality (competitiveness, longevity, etc) is the key to a cost-efficient program. Since field experimental approaches can be impacted by many environmental variables, it is difficult to get a clear understanding on how specific parameters, alone or in combination, may affect the SIT efficiency. The use of models not only helps to gather knowledge, but it allows the simulation of a wide range of scenarios and can be easily adapted to local populations and sterile male production. In this study, we consider single- and double-mated females. We first show that SIT can be successful only if the residual fertility is less than a threshold value that depends on the basic offspring number of the targeted pest population, the re-mating rates, and the parameters of double-mated females. Then, we show how the sterile male release rate is affected by the parameters of double-mated females and the male residual fertility. Different scenarios are explored with continuous and periodic sterile male releases, with and without ginger aromatherapy, which is known to enhance sterile male competitiveness, and also taking into account some biological parameters related to females that have been mated twice, either first by a wild (sterile) male and then a sterile (wild) male, or by two wild males only. Parameter values were chosen for peach as host fruit to reflect what could be expected in the Corsican context, where SIT against the medfly is under consideration. Our results suggest that ginger aromatherapy can be a decisive factor determining the success of SIT against medfly. We also emphasize the importance of estimating the duration of the refractory period between matings depending on whether a wild female has mated with a wild or sterile male. Further, we show the importance of parameters, like the (hatched) eggs deposit rate and the death-rate related to all fertile double-mated females. In general, re-mating is considered to be detrimental to SIT programs. However, our results show that, depending on the parameter values of double-mated females, re-mating may also be beneficial for SIT. Our model can be easily adapted to different contexts and species, for a broader understanding of release strategies and management options.

Abstract:Balancing process efficiency and adult sterile male biological quality is one of the challenges in the success of the sterile insect technique (SIT) against insect pest populations. For the SIT against mosquitoes, many stress factors need to be taken into consideration when producing sterile males that require high biological quality to remain competitive once released in the field. Pressures of mass rearing, sex sorting, irradiation treatments, packing, transport and release including handling procedures for each step, add to the overall stress budget of the sterile male post-release. Optimizing the irradiation step to achieve maximum sterility while keeping off-target somatic damage to a minimum can significantly improve male mating competitiveness. It is therefore worth examining various protocols that have been found to be effective in other insect species, such as dose fractionation. A fully sterilizing dose of 70 Gy was administered to Aedes aegypti males as one acute dose or fractionated into either two equal doses of 35 Gy, or one low dose of 10 Gy followed by a second dose of 60 Gy. The two doses were separated by either 1- or 2-day intervals. Longevity, flight ability, and mating competitiveness tests were performed to identify beneficial effects of the various treatments. Positive effects of fractionating dose were seen in terms of male longevity and mating competitiveness. Although applying split doses generally improved male quality parameters, the benefits may not outweigh the added labor in SIT programmes for the management of mosquito vectors.

Abstract:Abstract One environmentally friendly method used to manage Bactrocera dorsalis (Hendel), a key agricultural pest of substantial economic importance, is the sterile insect technique (SIT). Nevertheless, several deficiencies related to this strategy impair the success of the SIT, including the inferior performance of released sterile males compared with wild males, which could be partly solved by the utilization of gut symbionts as probiotic dietary components. In this study, a culture-dependent method was used to isolate and characterize gut-associated bacterial species in adult B. dorsalis genetic sexing strain (GSS) males. In addition, three bacterial isolates from the Enterobacteriaceae family, namely, Enterobacter sp., Morganella morganii, and Moellerella wisconsensis, were used as supplements in larval and adult diets to assess their effects on the life-history traits of irradiated males. Consistent with many previous studies, Enterobacter spp. was shown to be beneficial, with some quality control indices, such as adult size, pupal weight, survival rate under stress and nutritionally rich conditions, and mating competitiveness, being significantly increased, while slight nonsignificant increases in emergence rate and flight ability were observed. Conversely, the M. morganii and M. wisconsensis strains both had negative effects on irradiated male fitness and mating competitiveness. Our results, in combination with those of earlier studies, can contribute to improving the effectiveness of SIT application by enhancing the different aspects of augmentative rearing and biological traits of pests under laboratory rearing conditions.

Abstract:Aedes aegypti is the primary vector of diseases such as dengue, chikungunya, Zika fever, and yellow fever. The sterile insect technique (SIT) has been proposed as a species-specific and environment-friendly tool for the suppression of mosquito vector populations as a major component of integrated vector management strategies. As female mosquitoes are blood-feeders and may transmit pathogenic microorganisms, mosquito SIT depends on the release of sterile males. Genetic sexing strains (GSS) can be used for the efficient and robust separation of males from females. Two Ae. aegypti GSS were recently developed by exploiting eye colour mutations, resulting in the Red-eye GSS (RGSS) and the White-eye GSS (WGSS). In this study, we compared two WGSS, with and without the chromosomal inversion 35 (Inv35), and evaluated their biological quality, including genetic stability. Our results suggest that the WGSS/Inv35 presents a low recombination rate and long-term genetic stability when recombinants are removed from the colony (filtering) and a slow accumulation of recombinants when they are not removed from the colony (non-filtering). The two strains were similar with respect to fecundity, pupal and adult recovery rates, pupation curve, and pupal weight. However, differences were detected in fertility, survival rate of females, and flight ability of males. The WGSS/Inv35 presented lower fertility, higher survival rate of females, and better flight ability of males compared to the WGSS.

Abstract:The sterile insect technique (SIT) can be an efficient solution for reducing or eliminating certain insect pest populations. It is widely used in agriculture against fruit flies, including the Mediterranean fruit fly (medfly), . The re-mating tendency of medfly females and the fact that the released sterile males may have some residual fertility could be a challenge for the successful implementation of the SIT. Obtaining the right balance between sterility level and sterile male quality (competitiveness, longevity, etc) is the key to a cost-efficient program. Since field experimental approaches can be impacted by many environmental variables, it is difficult to get a clear understanding on how specific parameters, alone or in combination, may affect the SIT efficiency. The use of models not only helps to gather knowledge, but it allows the simulation of a wide range of scenarios and can be easily adapted to local populations and sterile male production. In this study, we consider single- and double-mated females. We first show that SIT can be successful only if the residual fertility is less than a threshold value that depends on the basic offspring number of the targeted pest population, the re-mating rates, and the parameters of double-mated females. Then, we show how the sterile males release rate is affected by the parameters of double-mated females and the male residual fertility. Different scenarios are explored with continuous and periodic sterile males releases, with and without ginger aromatherapy, which is known to enhance sterile male competitiveness, and also taking into account some biological parameters related to females that have been mated twice, either first by a wild (sterile) male and then a sterile (wild) male, or by wild males only. Parameter values were chosen for peach as host fruit to reflect what could be expected in the Corsican context, where SIT against the medfly is under consideration. The obtained results confirmed that the use of ginger aromatherapy is an essential step for SIT success against medfly. Our work also highlights the importance of estimating the duration of the refractory period between matings and how it may change when a wild (or sterile) female mates first with a sterile (or wild) male. Further, we show the importance of parameters, like the (hatched) eggs deposit rate and the death-rate related to all fertile double-mated females. In general, re-mating is considered to be detrimental to SIT programs. However, our results show that, depending on the parameter values of double-mated females, re-mating may also be beneficial for SIT. Our model, being generic, can be easily adapted to different contexts and species, for a broader understanding of release strategies and management options.

Abstract:Summary The sterile insect technique (SIT) is increasingly used to control pest insect populations. The success of SIT control programs depends on the ability to release sterile males and on the capacity of sterile males to compete with wild males to inseminate wild females. In this study, we evaluated the mating performance of Schistocerca gregaria (Försk.) males irradiated with 4 Gray. We compared reproductive traits, such as duration of precopulation time, mating duration, quantity of sperm stored by females after copulation, number of females mated successively and postmating competition of irradiated males with non-irradiated males. Irradiated males were able to mate but the resulting number of offspring was dramatically reduced compared to the average number of offspring observed during a regular mating. During a single copulation, irradiated males transferred fewer sperm than regular males but, theoretically, this quantity is enough to fertilize all the eggs produced by a female during its reproductive life. Irradiated males also had the ability to remove sperm from a previous mating with unirraditated males. This new information on the mating strategies helps explain the post-copulation guarding behaviour of S. gregaria.

Abstract:Insect symbionts are major manipulators of host's behavior. Their effect on parameters such as fecundity, male mating competitiveness, and biological quality in general, can have a major influence on the effectiveness of the sterile insect technique (SIT). SIT is currently being developed and applied against human disease vectors, including Ae. albopictus, as an environment-friendly method of population suppression, therefore there is a renewed interest on both the characterization of gut microbiota and their exploitation in artificial rearing. In the present study, bacterial communities of eggs, larvae, and adults (both males and females) of artificially reared Ae. albopictus, were characterized using both culture-dependent and culture-independent approaches. Mosquito-associated bacteria corresponding to thirteen and five bacteria genera were isolated from the larval food and the sugar solution (adult food), respectively. The symbiont community of the females was affected by the provision of a blood meal. Pseudomonas and Enterobacter were either introduced or enhanced with the blood meal, whereas Serratia were relatively stable during the adult stage of females. Maintenance of these taxa in female guts is probably related with blood digestion. Gut-associated microbiota of males and females were different, starting early after emergence and continuing in older stages. Our results indicate that eggs contained bacteria from more than fifteen genera including Bacillus, Chryseobacterium, and Escherichia-Shigella, which were also main components of gut microbiota of female adults before and after blood feeding, indicating potential transmission among generations. Our results provided a thorough study of the egg- and gut-associated bacteria of artificially reared Ae. albopictus, which can be important for further studies using probiotic bacteria to improve the effectiveness of mosquito artificial rearing and SIT applications.

Abstract:Abstract The yellow fever mosquito, Aedes aegypti L., can transmit several pathogens responsible for human diseases. With insecticide resistance development becoming a concern, alternative control strategies are needed for Ae. aegypti. Sterile insect technique (SIT) is an increasingly popular option being explored. However, logistical issues in mass production and sterilization make it difficult to maintain a SIT program. Male mosquitoes are typically irradiated as pupae because this is the earliest developmental point at which females can be separated from males, but asynchrony in pupation and high variability in pupal responses to irradiation based on pupal age make it difficult to sterilize mass quantities of pupae on a regular schedule in a rearing facility. Young adult mosquitoes have wider windows for irradiation sterilization than pupae, which can allow facilities to have fixed schedules for irradiation. We produced a workflow for adult Ae. aegypti irradiation in a mosquito control district with an operational SIT program that currently irradiates pupae. The impacts of chilling, compaction, and radiation dose on survival were all assessed before combining them into a complete adult irradiation protocol. Males chilled up to 16 h prior to compaction and compacted to 100 males/cm3 during radiation resulted in low mortality. Males irradiated as adults had increased longevity and similar sterility compared to males irradiated as pupae. Additionally, males sterilized as adults were more sexually competitive than males sterilized as pupae. Thus, we have shown that irradiating adult males can be a viable option to increase the efficiency of this operational mosquito SIT program.
