Abstract:Red-crowned cranes (Grus japonensis) consist of two distinct groups: the continental population and the island population. The island population, localized in Hokkaido, Japan, exhibits very low genetic diversity due to its rapid recovery from the brink of extinction. Our previous research in 2018 highlighted a possible mating between a male from the continental population, with the Gj5 haplotype, and a female from the island population, with the Gj2 haplotype, at Hitominuma Sawmp shore in northern Hokkaido. The present study attempted to unravel the distribution of their offspring by examining the major histocompatibility complex (MHC) of this mixed breeding pair compared with samples collected from cranes in northern and southeastern Hokkaido between 2008 and 2022. The analysis identified 55 MHC types, including 10 known types in a dataset of 89 crane samples, based on amino acid sequences. A total of 58 MHC types were recognized, based on nucleotide sequences, as there were many cases in which the same amino acid sequence had different nucleotide sequences. The five DNA types of MHC in the Hitominuma Swamp male were predominantly identified in eight cranes from northern Hokkaido and one chick from southeastern Hokkaido. In addition, population genetic analysis, based on insertion/deletion (InDel) polymorphisms, indicates distinct population differentiation between the northern and southeastern regions of Hokkaido. These results suggest that genetic contributions from the continental red-crowned crane population have already been integrated into the Hokkaido populations, with a more pronounced influence in northern Hokkaido.

Abstract:The red-crowned crane (Grus japonensis) has been demoted to “vulnerable species” because its populations have apparently stabilized in Japan and Korea. Low variation and genetic drift may cause damage to the nascent recovery of the G. japonensis population. The major histocompatibility complex (MHC) is one of the most polymorphic gene families in the vertebrate genome and can reflect information on the adaptive evolution of endangered species. In this study, variations in MHC I exon 3 of captive G. japonensis in China were assessed and compared with those in cranes from Japan. Forty MHC alleles of 274 base pairs were isolated from 32 individuals from two captive populations in China. There was high variability in the nucleotide and amino acid composition, showing the proportion of polymorphic sites of 18.98 and 32.97%, respectively. Comparative analyses of the Chinese and Japanese populations based on 222 base pair sequences revealed more alleles and higher variation in the Chinese population. The lack of significant geographical differentiation of G. japonensis was supported by the genetic differentiation coefficient (0.04506) between the Chinese and Japanese populations. Positive selection of antigen-binding sites was observed, which contributed to maintaining the diversity of MHC class I genes. Phylogenetic analysis suggested the persistence of trans-species polymorphisms among MHC class I genes in Gruidae species. Our results may contribute to optimizing the management of G. japonensis populations and population recovery of this threatened species.

Abstract:Red-crowned crane Grus japonensis is an endangered species in two separate populations: the mainland population in the Eurasian continent and the island population in eastern Hokkaido, Japan. We found 11 insertion/deletion (InDel) markers in the genome of the red-crowned crane and designed primer sets across these InDels that can be analyzed with conventional agarose gel electrophoresis. Sixty-six samples of whole blood and skeletal muscle obtained from red-crowned cranes, including 12 families in eastern Hokkaido from 1994 to 2021, showed different patterns in gel images of 11 InDel PCR reactions except for two pairs. The combined non-exclusion probability of the 11 markers indicates that individuals can be determined with a probability of 99.9%. In 39 non-relative chicks, the expected heterozygosity (He) was 0.316, suggesting low genetic diversity. This might not be caused by high levels of inbreeding since the average FIS was not significantly different from zero (0.095, p = 0.075). The results suggest that the 11 InDel primer sets can be used for fairly accurate individual identification as well as genetic population analyses in red-crowned cranes in the island population.

Abstract:Red-crowned cranes Grus japonensis, which are an endangered species, have two separate populations, a mainland population in the Eurasian continent and an island population in eastern Hokkaido, Japan. Island cranes showed three haplotypes (Gj1, Gj2 and Gj13), whereas ten haplotypes (Gj3-Gj12) were confirmed in captive cranes and stray cranes. We found Gj5 haplotype in feathers of two cranes as well as four new haplotypes in seven wild crane feathers collected in South Korea. We also found feathers in the nest in Sarobetsu Wetland in northwestern Hokkaido. While the haplotype of female-derived feathers was Gj2, that of male-derived feathers was Gj5. The results suggest that there has been crossbreeding between cranes in the island population and cranes in the mainland population.

Abstract:Red-crowned cranes (Grus japonensis) were classified as an endangered species by the International Union for Conservation of Nature, but recently, their population has decreased dramatically. For the purpose of conserving this endangered species, 18 microsatellite markers were developed, including 12 newly isolated ones from a genomic library and 6 modified from another crane species. The markers were characterized in 26 red-crowned cranes. As a result, these markers displayed 3-13 alleles, and the observed and expected heterozygosities ranged from 0.462 to 1.000 and from 0.483 to 0.884, respectively. The marker suite averaged 6.390 alleles per locus with an average polymorphic information content of 0.631. The combined exclusion probability (PE-1) was 0.9985, and the combined exclusion probability (PE-2) was 0.9999. Three of the 18 microsatellite loci presented significant deviations from Hardy-Weinberg Equilibrium (P < 0.05), likely due to sampling bias and unknown founder relatedness in a semi-free population. Our results show that microsatellite loci can provide a standard protocol for genetic information in red-crowned crane populations upon which strategies for effective conservation and management may be based.

Abstract:The red-crowned crane (Grus japonensis) is an endangered wading bird distributed in northeast Asia. The population of G. japonensis is continuing to decline owing to the loss and degradation of habitats. In order to protect G. japonensis, we developed 33 single nucleotide polymorphism (SNP) markers for this species using a genotyping-by-sequencing approach. The minor allele frequency ranged from 0.033 to 0.483, the observed heterozygosity ranged from 0.067 to 0.833, the expected heterozygosity ranged from 0.066 to 0.508, and the polymorphism information content ranged from 0.062 to 0.375. Among these SNPs, two loci showed deviations from the Hardy–Weinberg equilibrium (P < 0.05). The novel polymorphic SNPs can lay the foundation for future studies on evolution and conservation genetics of G. japonensis.

Abstract:Six microsatellite loci were used to assess the genetic variation levels between populations in Zhalong Nature Reserve and levels of gene flow between wild and non-migratory subpopulations of red-crowned crane.Results show that wild,non-migratory and captive populations of red-crowned crane have high genetic variation levels.HE value is 0.833 00 for wild population,0.800 00 for non-migratory population and 0.810 00 for captive population.FST results manifest about 97.5% genetic diversity within population and 2.5% genetic diversity between subpopulations.Levels of gene flow shows 1.9 individuals every generation by private allele method and 1 individual every generation by FST method,and the values are enough to maitain a higher genetic variation level between wild and non-migratory populations in Zhalong Nature Reserve.

Abstract:SummaryThe historical distribution of the Red-crowned Crane Grus japonensis together changes in its breeding and wintering grounds in China are reviewed. According to historical information the bird mainly bred in Heilongjiang and Jilin Provinces and wintered at coastal regions near the Yangtze estuary. Due to geographical and anthropological changes, the breeding grounds gradually moved to the west and south while the wintering grounds moved northward. Efforts have been made to protect this endangered species but factors still exist which restrict further development of the population.

Abstract:Continental populations of Red-crowned Crane(Grus japonensis) are divided into two distinctive parts by the line of Bureinsky Mountains - Xiaoxing' an Mountains - Zhangguangcai Mountains. In 2004 and 2005 , through measurements and statistical testing to the environment factors of 31 nest-sites in the main breeding areas of the eastern and western populations of Red-crowned Crane, we found that three habitat factors; reed density, reed area, and distance to disturbances, show significant differences between the eastern and western populations of the Red-crowned Cranes (p < 0.05). The results of Hierarchical anlysis based on nest-site habitat factors indicated that Red-crowned Cranes breeding in five nature reserves can be divided into two distinctive parts (the eastern and the western populations). These characteristics were present on both quantitative difference of habitat factors and covering tactics by different migratory populations. We found that both eastern and western cranes selected nesting sites with the same water depth and reed height. Habitat loss influenced the crane populations for their K-life history strategies, which formed different characteristics of the nest-site selection between the east and west populations of the Red-crowned Cranes.

Abstract:The analytical system was constructed for the amplified fragment length polymorphism(AFLP) in Red-crowned Crane(Grus japonensis).The 5 couples of Red-crowned Crane were scanned by AFLP marker using the selective 28 primer combinations.The polymorphism and genetic cluster graph were acquired applying 10 individuals belonging to the 5 couples of Red-crowned Crane.A total of 551 polymorphism sites of the total of 1 114 bands were amplified from them.The percentage of polymorphism is 49.5%,each primer couples made 20-66 polymorphism sites.Primer E4M1 made the most,66 polymorphism sites;and primer E6M1 made the least,20 polymorphism sites.The similarity coefficient between the 10 Redcrowned Cranes was calculated,which were all in 0.71-0.88.The genetic distance and the UPGMA tree of 5 couples of Red-crowned Crane had been acquired.Cluster analysis showed that No.1 had close relationship with No.2;No.3 had also very close relationship with No.4;and the other 3 couple cranes had far genetic relationship.These showed that Red-crowned Crane has the behavioral mechanisms to identify the relationship.The study content about breeding behavioral mechanisms of Red-crowned Crane was more abundant,and puts forward some suggestions for studying deeply and founding reasonable breeding system for Semi-captive Red-crowned Crane population.

Abstract:Understanding a species' mating strategies is essential for elucidating their social structures and comprehending the trade-offs involved in optimizing fitness. Our study focuses on the Black-necked Crane ( Grus nigricollis ), an elusive species native to the Qinghai-Tibetan Plateau, which has remained largely mysterious in terms of its mating strategies and group dynamics. Using 13 microsatellite loci, we have conducted precise sex determination and individual identification, which has been instrumental in constructing detailed breeding pedigrees and calculating intricate kinship coefficients. Our comprehensive DNA analysis, combined with meticulous nest-site mapping, reveals that Black-necked Cranes form non-kinship-based groups and exhibit a strong inclination toward nest-site fidelity, especially among males (Male:100%, Female:71%). Significantly, this research documents, for the first time, a notable tendency for extra-pair copulation in this species (16.6%) and reveals that no pair maintained monogamy throughout the four-year study period. These findings challenge conventional views of crane monogamy and deepen our understanding of avian mating systems, suggesting a strategic adaptation to enhance genetic diversity and prevent inbreeding within the population. This research not only unveils new insights into the complex social structures of these cryptic avian populations but also underscores the urgent need for habitat conservation to ensure the species' continued survival and adaptability.

Abstract:The trophic relations of the red-crowned crane in Zhalong wetland, northeastern China may significantly be altered due to the rapid decrease in accessible food resources. To confirm this hypothesis, we determined the trophic status of this vulnerable species by using stable isotope (carbon (δ13C) and nitrogen (δ15N) methods. The δ13Cand 15Nsignatures in the red-crowned crane species were in the range of −17.8‰to −18.5‰and 6.9‰to 8.1‰, respectively. The majority of the ecosystem of Zhalong wetland has an average food-chain length of 3.9, and the trophic level of the examined species, i.e., red-crowned crane, was approximately 3.1. The trophic relations of red-crowned cranes have changed over the period of 2004 to 2014, as the biological species in the bottom trophic level significantly exhausted in the past years. The conflict of food demand by red-crowned cranes versus the livelihood demand of local wetland residents should be resolved to ensure sustainability of food supply sustainable for rare and extinguishable species.

Abstract:•The law of spatial distribution and population transfer of Red-crowned crane in the past 30 years were studied.•The temporal and spatial evolution of land-use in the past 30 years was summarized.•The spatial regression model was used to reveal the effect of land-use change on the distribution of Red-crowned crane.•“Friendly” land-use patterns conducive to habitat conservation of the red-crowned crane were proposed.
