Abstract:Cadmium (Cd) is a non-essential heavy metal with high toxicity to plants. Plants have acquired specialized mechanisms to sense, transport, and detoxify Cd. Recent studies have identified many transporters involved in Cd uptake, transport, and detoxification. However, the complex transcriptional regulatory networks involved in Cd response remain to be elucidated. Here, we provide an overview of current knowledge regarding transcriptional regulatory networks and post-translational regulation of the transcription factors involved in Cd response. An increasing number of reports indicate that epigenetic regulation and long non-coding and small RNAs are important in Cd-induced transcriptional responses. Several kinases play important roles in Cd signaling that activate transcriptional cascades. We also discuss the perspectives to reduce grain Cd content and improve crop tolerance to Cd stress, which provides a theoretical reference for food safety and the future research of plant varieties with low Cd accumulation.

Abstract:Cadmium, a high toxicity element, is a potential threat to plant and human health, and a dangerous pollutant in the environment. Uptake and accumulation by crops represent the main entry pathway for potentially health-threatening toxic metals into human and animal food. Crops and other plants take up Cd from the soil or water and may distribute it in their roots and shoots. Soil and/or water are usually contaminated with Cd through natural sources, industrial effluent, and anthropogenic activities. In this review, the sources of Cd contamination, evaluation of the phytotoxic effects on plants, and mode of action of Cd toxicity, were summarized. Plant defensive strategies upon excess Cd are also considered in this review. Cd-induced effects include oxidative stress, disintegration of the photosynthetic apparatus, reduction in gas exchange parameters, nutrient imbalance, and subcellular organelle degradation. In addition, Cd severely impairs biomolecules such as DNA, protein, and lipids. Although plants are sessile in nature, they are equipped with certain mechanisms to cope with unfavorable conditions. These mechanisms include synthesis of metal-chelating proteins, expression of enzymatic and non-enzymatic antioxidants, organic acids, and plant root-mycorrhiza association. The built-in system of plant tolerance to Cd can be further enhanced by the application of exogenous organic and inorganic metal sources. This review will broaden the knowledge about the Cd accumulation in plants and the responses to metal exposure, as well as our understanding of metal tolerance and overcoming this serious issue for sustainable agriculture and human health worldwide.

Abstract:Summary Cadmium (Cd) is one of the most toxic heavy metals faced by plants and, additionally, via the food chain, threatens human health. It is principally dispersed through agro‐ecosystems via anthropogenic activities and geogenic sources. Given its high mobility and persistence, Cd, although not required, can be readily assimilated by plants thereby posing a threat to plant growth and productivity as well as animal and human health. Thus, breeding crop plants in which the edible parts contain low to zero Cd as safe food stuffs and harvesting shoots of high Cd‐containing plants as a route for decontaminating soils are vital strategies to cope with this problem. Recently, multiomics approaches have been employed to considerably enhance our understanding of the mechanisms underlying (i) Cd toxicity, (ii) Cd accumulation, (iii) Cd detoxification and (iv) Cd acquisition tolerance in plants. This information can be deployed in the development of the biotechnological tools for developing plants with modulated Cd tolerance and detoxification to safeguard cellular and genetic integrity as well as to minimize food chain contamination. The aim of this review is to provide a current update about the mechanisms involved in Cd uptake by plants and the recent developments in the area of multiomics approach in terms of Cd stress responses, as well as in the development of Cd tolerant and low Cd accumulating crops.

Abstract:Cadmium accumulation in crops and the possibility of Cd entering the food chain are serious concerns for public health. This review discusses the deleterious effects of Cd in plants and the tolerance and resistance mechanisms that evolved to help mitigate cadmium toxicity. Cadmium reduces seed germination, early seedling growth, and plant biomass. It causes changes in photosynthesis, relative water content, transpiration rate, stomatal conductance, and electrolyte leakage. Cadmium activates the reactive oxygen species that induce chromosomal aberrations, gene mutations, and DNA damage that affect the cell cycle and cell division. In response, plants have applied several adaptive strategies to overcome and reduce the toxic effects of Cd. The primary detoxification mechanisms are exclusion and accumulation of Cd in specific plant parts. Plants also adapt to Cd toxicity with the help of signaling pathways that regulate survival and growth under Cd stress. Other mechanisms such as synthesis of plants hormones, activation of the antioxidant system and the production of phytochelatins and proline are extremely helpful in plant tolerance to Cd. Furthermore, soil microorganisms play a crucial role toward the Cd tolerance in plants by decreasing metal phytoavailability and increasing morphological and physiological parameters of plant.

Abstract:Anthropogenic activities pose a more significant threat to the environment than natural phenomena by contaminating the environment with heavy metals. Cadmium (Cd), a highly poisonous heavy metal, has a protracted biological half-life and threatens food safety. Plant roots absorb Cd due to their high bioavailability through apoplastic and symplastic pathways and translocate it to shoots through the xylem with the help of transporters and then to the edible parts via the phloem. The uptake and accumulation of Cd in plants pose deleterious effects on plant physiological and biochemical processes, which alter the morphology of vegetative and reproductive parts. In vegetative parts, Cd stunts root and shoot growth, photosynthetic activities, stomatal conductance, and overall plant biomass. Plants' male reproductive parts are more prone to Cd toxicity than female reproductive parts, ultimately affecting grain/fruit production and survival. To alleviate/avoid/tolerate Cd toxicity, plants activate several defense mechanisms, including enzymatic and non-enzymatic antioxidants, Cd-tolerant genes up-regulations, and phytohormonal secretion. Additionally, plants tolerate Cd through chelating and sequestering as part of the intracellular defensive mechanism with the help of phytochelatins and metallothionein proteins, which help mitigate the harmful effects of Cd. The knowledge on the impact of Cd on plant vegetative and reproductive parts and the plants' physiological and biochemical responses can help selection of the most effective Cd-mitigating/avoiding/tolerating strategy to manage Cd toxicity in plants.

Abstract:Cadmium (Cd) contamination poses a significant threat to agriculture and human health due to its high soil mobility and toxicity. This review synthesizes current knowledge on Cd uptake, transport, detoxification, and transcriptional regulation in plants, emphasizing the roles of metal transport proteins and transcription factors (TFs). We explore transporter families like NRAMP, HMA, ZIP, ABC, and YSL in facilitating Cd movement within plant tissues, identifying potential targets for reducing Cd accumulation in crops. Additionally, regulatory TF families, including WRKY, MYB, bHLH, and ERF, are highlighted for their roles in modulating gene expression to counteract Cd toxicity. This review consolidates the existing literature on plant–Cd interactions, providing insights into established mechanisms and identifying gaps for future research. Understanding these mechanisms is crucial for developing strategies to enhance plant tolerance, ensure food safety, and promote sustainable agriculture amidst increasing heavy-metal pollution.

Abstract:Medicinal plants, an important source of herbal medicine, are gaining more demand with the growing human needs in recent times. However, these medicinal plants have been recognized as one of the possible sources of heavy metal toxicity in humans as these medicinal plants are exposed to cadmium-rich soil and water because of extensive industrial and agricultural operations. Cadmium (Cd) is an extremely hazardous metal that has a deleterious impact on plant development and productivity. These plants uptake Cd by symplastic, apoplastic, or via specialized transporters such as HMA, MTPs, NRAMP, ZIP, and ZRT-IRT-like proteins. Cd exerts its effect by producing reactive oxygen species (ROS) and interfere with a range of metabolic and physiological pathways. Studies have shown that it has detrimental effects on various plant growth stages like germination, vegetative and reproductive stages by analyzing the anatomical, morphological and biochemical changes (changes in photosynthetic machinery and membrane permeability). Also, plants respond to Cd toxicity by using various enzymatic and non-enzymatic antioxidant systems. Furthermore, the ROS generated due to the heavy metal stress alters the genes that are actively involved in signal transduction. Thus, the biosynthetic pathway of the important secondary metabolite is altered thereby affecting the synthesis of secondary metabolites either by enhancing or suppressing the metabolite production. The present review discusses the abundance of Cd and its incorporation, accumulation and translocation by plants, phytotoxic implications, and morphological, physiological, biochemical and molecular responses of medicinal plants to Cd toxicity. It explains the Cd detoxification mechanisms exhibited by the medicinal plants and further discusses the omics and biotechnological strategies such as genetic engineering and gene editing CRISPR- Cas 9 approach to ameliorate the Cd stress.

Abstract:Cadmium (Cd) is a non-essential trace element for plants, and has strong toxicity at low concentrations. It can suppress the elongation growth of plant cell, inhibit oxidative mitochondrial phosphorylation, induce oxidative stress, inhibit the activities of several antioxidative enzymes, affect photosynthesis by inhibiting ferrous reductase or damaging photosynthesis apparatus, and cause the alteration of chromatin and the change of plasma membrane ATPase activity. In response to Cd stress, the cells of cadmium-resistant plant species can produce a number of proteins such as phytochelatins, metallothioneins and stress proteins to detoxify Cd ions, and efficiently repair Cd damage. The plant cells can also resort to other defense systems to detoxify Cd ions, e.g., the immobilization of Cd by cell wall, exclusion of Cd through the action of plasma membrane, compartmentalization of Cd by vacuolar, and release of plant glands. The phytochelatin synthase (PCS) genes of Arabidopsin, wheat and Schizosaccharomyces pombe had been identified by using different approaches, and the metallothioneins (MT) in plants was also identified recently. By introducing animal MT genes, transgenic plants could increase the resistant ability to Cd toxicity. Subjected to Cd, plant cells often start to synthesize stress proteins such as heat shock proteins, and the plants having been transformed the stress protein genes could enhance their resistant capacity to Cd ions. It was reported that zinc (Zn) ion-transporting proteins could also transport Cd ion. Some minor genes not conferring tolerance on their own could modify the major gene (s), and enhance Cd tolerance. Cd detoxification in wild type plants could be a complex phenomenon, probably under polygenic control to Cd, while acute Cd stress seemed to be a simpler mechanism, apparently involving only one or a few specific major genes.

Abstract:AbstractCadmium (Cd) is one of the non-essential, highly toxic environmental pollutants worldwide causing serious environmental and agricultural problems. Elevated Cd doses are carcinogenic to humans. It is ranked seventh in the list of top 20 toxic metals and classified as a group 1 carcinogen. The median range of Cd dietary intake (66.5–116 µg Cd kg−1 body weight per month) is much higher than maximum limit (25 µg Cd kg−1 body weight per month) reported by FAO/WHO. Toxicity of Cd causes a range of damages to plants from germination to yield; however, the extent of damage is concentration and time-dependent. Reduction in seed germination and plant growth is primarily due to Cd interference with enzymatic and photosynthetic activities and membrane damage. Cadmium exposure at higher rates disturbs the nutritional and water relations of plants and causes oxidative damage. Moreover, Cd-induced structural changes in the photosynthetic apparatus disturb the yield of plants. In this review, adverse effects of Cd on seed germination, stand establishment, plant growth, uptake and assimilation of nutrients, enzymatic activities, ultra-structural and oxidative damages, changes in antioxidant defense system and stress proteins, carbon metabolism, and yield formation are reported. Moreover, Cd dynamics in soil rhizosphere and factors affecting Cd dynamics in soil have also been discussed. Furthermore, remediation strategies (physical, chemical, biological, and amendments) to decontaminate Cd-polluted soils have also been described in this review. Through phytoremediation, Cd can be extracted and stabilized in the soil while through microbes Cd can be sequestrated into their bodies. Increased Cd uptake in hyperaccumulator plants to remediate and convert the toxic form of Cd into nontoxic forms. While in chemical remediation, Cd can be washed out, immobilized and stabilized in the soil through chemical amendments. Bioremediation of polluted sites is considered effective and reliable due to its eco-friendly features. Moreover, Cd uptake and toxicity in rice can be decreased by proper application of essential nutrients such as nitrogen, zinc, iron, and selenium in Cd contaminated soils. The organic amendments may help through an increase in soil pH, adsorption in its functional groups, the formation of complexations, and the conversion of exchangeable to residual forms. Adoption of some agricultural practices are also found to be effective in reducing the Cd uptake and accumulation in plants and harvesting quality food from Cd contaminated soils.

Abstract:Heavy metal (HM) pollution, specifically cadmium (Cd) contamination, is a worldwide concern for its consequences for plant health and ecosystem stability. This review sheds light on the intricate mechanisms underlying Cd toxicity in plants and the various strategies employed by these organisms to mitigate its adverse effects. From molecular responses to physiological adaptations, plants have evolved sophisticated defense mechanisms to counteract Cd stress. We highlighted the role of phytochelatins (PCn) in plant detoxification, which chelate and sequester Cd ions to prevent their accumulation and minimize toxicity. Additionally, we explored the involvement of glutathione (GSH) in mitigating oxidative damage caused by Cd exposure and discussed the regulatory mechanisms governing GSH biosynthesis. We highlighted the role of transporter proteins, such as ATP-binding cassette transporters (ABCs) and heavy metal ATPases (HMAs), in mediating the uptake, sequestration, and detoxification of Cd in plants. Overall, this work offered valuable insights into the physiological, molecular, and biochemical mechanisms underlying plant responses to Cd stress, providing a basis for strategies to alleviate the unfavorable effects of HM pollution on plant health and ecosystem resilience.

Abstract:<p>Cadmium (Cd) is an unessential trace element in plants that is ubiquitous in the environment. Anthropogenic activities such as disposal of urban refuse, smelting, mining, metal manufacturing, and application of synthetic phosphate fertilizers enhance the concentration of Cd in the environment and are carcinogenic to human health. In this manuscript, we reviewed the sources of Cd contamination to the environment, soil factors affecting the Cd uptake, the dynamics of Cd in the soil rhizosphere, uptake mechanisms, translocation, and toxicity of Cd in plants. In crop plants, the toxicity of Cd reduces uptake and translocation of nutrients and water, increases oxidative damage, disrupts plant metabolism, and inhibits plant morphology and physiology. In addition, the defense mechanism in plants against Cd toxicity and potential remediation strategies, including the use of biochar, minerals nutrients, compost, organic manure, growth regulators, and hormones, and application of phytoremediation, bioremediation, and chemical methods are also highlighted in this review. This manuscript may help to determine the ecological importance of Cd stress in interdisciplinary studies and essential remediation strategies to overcome the contamination of Cd in agricultural soils.</p>

Abstract:Heavy metal concentrations exceeding permissible limits threaten human life, plant life, and all other life forms. Different natural and anthropogenic activities emit toxic heavy metals in the soil, air, and water. Plants consume toxic heavy metals from their roots and foliar part inside the plant. Heavy metals may interfere with various aspects of the plants, such as biochemistry, bio-molecules, and physiological processes, which usually translate into morphological and anatomical changes. They use various strategies to deal with the toxic effects of heavy metal contamination. Some of these strategies include restricting heavy metals to the cell wall, vascular sequestration, and synthesis of various biochemical compounds, such as phyto-chelators and organic acids, to bind the free moving heavy metal ions so that the toxic effects are minimized. This review focuses on several aspects of genetics, molecular, and cell signaling levels, which integrate to produce a coordinated response to heavy metal toxicity and interpret the exact strategies behind the tolerance of heavy metals stress. It is suggested that various aspects of some model plant species must be thoroughly studied to comprehend the approaches of heavy metal tolerance to put that knowledge into practical use.

Abstract:Plants take up a wide range of trace metals/metalloids (hereinafter referred to as trace metals) from the soil, some of which are essential but become toxic at high concentrations (e.g. Cu, Zn, Ni, Co), while others are non‐essential and toxic even at relatively low concentrations (e.g. As, Cd, Cr, Pb and Hg). Soil contamination of trace metals is an increasing problem worldwide due to intensifying human activities. Trace metal contamination can cause toxicity and growth inhibition in plants, as well as accumulation in the edible parts to levels that threaten food safety and human health. Understanding the mechanisms of trace metal toxicity and how plants respond to trace metal stresses is important for improving plant growth and food safety in contaminated soils. Accumulation of excess trace metals in plants can cause oxidative stress, genotoxicity, programmed cell death, and disturbance in multiple physiological processes. Plants have evolved various strategies to detoxify trace metals through cell wall binding, complexation, vacuolar sequestration, efflux and translocation. Multiple signal transduction pathways and regulatory responses are involved in plants challenged with trace metal stresses. In this review, we discuss recent progress in understanding the molecular mechanisms involved in trace metal toxicity, detoxification and regulation, as well as strategies to enhance plant resistance to trace metal stresses and reduce toxic metal accumulation in food crops. Abstract:Cadmium (Cd) exerts a toxic influence on numerous crucial growth and development processes in plants, notably affecting seed germination rate, transpiration rate, chlorophyll content, and biomass. While considerable advances in Cd uptake and detoxification of plants have been made, the mechanisms by which plants adapt to and tolerate Cd toxicity remain elusive. This review focuses on the relationship between Cd and plants and the prospects for phytoremediation of Cd pollution. We highlight the following issues: (1) the present state of Cd pollution and its associated hazards, encompassing the sources and distribution of Cd and the risks posed to human health; (2) the mechanisms underlying the uptake and transport of Cd, including the physiological processes associated with the uptake, translocation, and detoxification of Cd, as well as the pertinent gene families implicated in these processes; (3) the detrimental effects of Cd on plants and the mechanisms of detoxification, such as the activation of resistance genes, root chelation, vacuolar compartmentalization, the activation of antioxidant systems and the generation of non-enzymatic antioxidants; (4) the practical application of phytoremediation and the impact of incorporating exogenous substances on the Cd tolerance of plants.

Abstract:Cadmium (Cd) is a heavy metal highly toxic to living organisms. Cd pollution of soils has become a serious problem worldwide, posing a severe threat to crop production and human health. When plants are poisoned by Cd, their growth and development are inhibited, chloroplasts are severely damaged, and respiration and photosynthesis are negatively affected. Therefore, elucidating the molecular mechanisms that underlie Cd tolerance in plants is important. Transcription factors can bind to specific plant cis-acting genes. Transcription factors are frequently reported to be involved in various signaling pathways involved in plant growth and development. Their role in the resistance to environmental stress factors, particularly Cd, should not be underestimated. The roles of several transcription factor families in the regulation of plant resistance to Cd stress have been widely demonstrated. In this review, we summarize the mechanisms of five major transcription factor families–WRKY, ERF, MYB, bHLH, and bZIP–in plant resistance to Cd stress to provide useful information for using molecular techniques to solve Cd pollution problems in the future.

Metalloprotein-Specific or Critical Amino Acid Residues: Perspectives on Plant-Precise Detoxification and Recognition Mechanisms under Cadmium Stress

Abstract:Cadmium (Cd) pollution in cultivated land is caused by irresistible geological factors and human activities; intense diffusion and migration have seriously affected the safety of food crops. Plants have evolved mechanisms to control excessive influx of Cd in the environment, such as directional transport, chelation and detoxification. This is done by some specific metalloproteins, whose key amino acid motifs have been investigated by scientists one by one. The application of powerful cell biology, crystal structure science, and molecular probe targeted labeling technology has identified a series of protein families involved in the influx, transport and detoxification of the heavy metal Cd. This review summarizes them as influx proteins (NRAMP, ZIP), chelating proteins (MT, PDF), vacuolar proteins (CAX, ABCC, MTP), long-distance transport proteins (OPT, HMA) and efflux proteins (PCR, ABCG). We selected representative proteins from each family, and compared their amino acid sequence, motif structure, subcellular location, tissue specific distribution and other characteristics of differences and common points, so as to summarize the key residues of the Cd binding target. Then, we explain its special mechanism of action from the molecular structure. In conclusion, this review is expected to provide a reference for the exploration of key amino acid targets of Cd, and lay a foundation for the intelligent design and breeding of crops with high/low Cd accumulation.

Abstract:Food safety has emerged as a high-urgency matter for sustainable agricultural production. Toxic metal contamination of soil and water significantly affects agricultural productivity, which is further aggravated by extreme anthropogenic activities and modern agricultural practices, leaving food safety and human health at risk. In addition to reducing crop production, increased metals/metalloids toxicity also disturbs plants' demand and supply equilibrium. Counterbalancing toxic metals/metalloids toxicity demands a better understanding of the complex mechanisms at physiological, biochemical, molecular, cellular, and plant level that may result in increased crop productivity. Consequently, plants have established different internal defense mechanisms to cope with the adverse effects of toxic metals/metalloids. Nevertheless, these internal defense mechanisms are not adequate to overwhelm the metals/metalloids toxicity. Plants produce several secondary messengers to trigger cell signaling, activating the numerous transcriptional responses correlated with plant defense. Therefore, the recent advances in omics approaches such as genomics, transcriptomics, proteomics, metabolomics, ionomics, miRNAomics, and phenomics have enabled the characterization of molecular regulators associated with toxic metal tolerance, which can be deployed for developing toxic metal tolerant plants. This review highlights various response strategies adopted by plants to tolerate toxic metals/metalloids toxicity, including physiological, biochemical, and molecular responses. A seven-(omics)-based design is summarized with scientific clues to reveal the stress-responsive genes, proteins, metabolites, miRNAs, trace elements, stress-inducible phenotypes, and metabolic pathways that could potentially help plants to cope up with metals/metalloids toxicity in the face of fluctuating environmental conditions. Finally, some bottlenecks and future directions have also been highlighted, which could enable sustainable agricultural production.

Abstract:Among all non-essential heavy metals, cadmium (Cd) is perhaps the metal that has attracted the most attention in soil science and plant nutrition due to its potential toxicity to humans, and also its relative mobility in the soil–plant system. In plant system, Cd influences entire metabolism. It interferes with physiological activities of plants such as photosynthesis, gaseous exchange, nutrients absorption, and causes reduction in plant growth, dry matter accumulation and yield. It also results alteration in antioxidant production, cell cycle and division that leads to chromosomal aberrations and modification in gene expression. In order to cope with such situation plants have to develop several strategies. Among them phytohormones played an important role, but in presence of high level of Cd in the environment, plants are not able to carry out their normal functions. In such condition, exogenous application of phytohormones improves the plant metabolism. These phytohormones (plant growth regulators) act as active members of signal transduction cascade that involve in metal detoxification. Thus, this review describes mechanism behind application of phytohormones for metal remediation particularly Cd including its impact on morphological, biochemical and molecular characterization of the plants.

Abstract:With the increasing concern on heavy metal contamination in agriculture and other environmental settings, unraveling the mechanisms of cadmium (Cd) tolerance and response in plants has become highly important. Ongoing plant Cd research over the years has focused on strategic and relevant aspects, including molecular, biochemical, and physiological processes. From this perspective, phosphoproteomics appears to be an innovative and powerful approach to investigating plant responses to Cd stress. Here, we summarize progress in plant Cd research across different plant species regarding large-scale phosphoproteomic investigations. Some studies revealed major proteins participating in detoxification, stress signaling, and metabolism, along with their regulation through phosphorylation, which modulates the plant's defense against Cd. However, many pathways remain unexplored. Expanding these studies will help our ability to alleviate Cd stress and provide further information concerning involved mechanisms. Our purpose is to inspire researchers to further explore the use of phosphoproteomics in unraveling such complex mechanisms of Cd tolerance and response across various plant species, with the ultimate aim of enhancing strategies for mitigating Cd stress in agriculture and polluted environments.
