Abstract:Expansins (EXPs) are unique plant cell wall proteins with the ability to induce cell wall expansion and play potential roles in xylem development. In the present study, a total of 25 BpEXP genes were identified in Betulaplatyphylla. Results of bioinformatics analysis described that BpEXP gene family was highly conserved in the process of evolution. All these genes were clustered into four groups, EXPA, EXPB, EXPLA and EXPLB, according to phylogenetic analysis and BpEXP1 was highly homologous to PttEXP1 and PttEXP2. The results of RT-qPCR showed that BpEXP1 was expressed higher in stems and preferentially expressed in the first internodes, followed by apical buds and the third internodes, promoter expression analysis with GUS assay demonstrated that it was expressed in developing xylem, suggesting that BpEXP1 might be involved in the development of the primary stems of birch. Overexpression of BpEXP1 can promote cortex cell expansion and then enlarge the cortex cell area and layer, however inhibit the secondary cell wall deposition and result in the thinner cell wall and larger lumens of xylem fiber in transgenic plants. In addition, yeast one hybrid (Y1H) assays showed that the BpMYB4 was identified as the upstream transcription factor of BpEXP1 via ABRE elements to participate in regulating xylem development. This study will provide information for investigating the regulation mechanism of BpEXP genes and gene resources for birch genetics improvement.
RNA-Seq Analysis of Differential Gene Expression in Betula Luminifera Xylem During the Early Stages of Tension Wood Formation.

Abstract:BACKGROUND:Betula luminifera H. Winkler, which is widely distributed in southern China, is an economically important broadleaf tree species. However, little genomic information of B. luminifera is available, and little is known about the molecular mechanisms of wood formation in this species. Meanwhile, few efforts have focused on investigating the early transcriptional changes during tension wood formation in woody plants.RESULTS:A reference transcriptome dataset was first generated containing 45,700 Unigenes, and 35,135 (76.9%) Unigenes were annotated by a BLAST similarity search against four public databases. Then, based on an anatomical investigation, the global gene expression changes during the early stages of tension wood formation were analyzed. Gene expression profiling showed that a total of 13,273 Unigenes were differentially regulated during the early stages of tension wood formation. Most genes involved in cellulose and lignin biosynthesis were highlighted to reveal their biological importance in tension wood formation. In addition, the transcription levels of many genes involved in the auxin response pathway were significantly changed during the early stages of tension wood formation. Furthermore, 18 TFs co-expressed with key enzymes of cellulose synthesis were identified.CONCLUSIONS:Our results revealed the transcriptional changes associated with TW formation and identified potential key genes in the regulation of this process. These results will help to dissect the molecular mechanism of wood formation and provide key candidate genes for marker-assisted selection in B. luminifera.

Abstract:P>Root hair growth requires intensive cell-wall modification. This study demonstrates that root hair-specific expansin As, a sub-clade of the cell wall-loosening expansin proteins, are required for root hair elongation in rice (Oryza sativa L.). We identified a gene encoding EXPA17 (OsEXPA17) from a rice mutant with short root hairs. Promoter::reporter transgenic lines exhibited exclusive OsEXPA17 expression in root hair cells. The OsEXPA17 mutant protein (OsexpA17) contained a point mutation, causing a change in the amino acid sequence (Gly104 -> Arg). This amino acid alteration is predicted to disrupt a highly conserved disulfide bond in the mutant. Suppression of OsEXPA17 by RNA interference further confirmed requirement for the gene in root hair elongation. Complementation of the OsEXPA17 mutant with other root hair EXPAs (OsEXPA30 and Arabidopsis EXPA7) can restore root hair elongation, indicating functional conservation of these root hair EXPAs in monocots and dicots. These results demonstrate that members of the root hair EXPA sub-clade play a crucial role in root hair cell elongation in Graminaceae.
Sequence characterization and expression analysis of NAC genes from Betula platyphylla

The NAC family has many members; however, the functions of the majority of them are unknown, especially in woody plants. In this study, to characterize the NAC members and explore NAC genes’ association with xylem development, 21 full-length NAC genes were identified in Betula platyphylla, a deciduous woody plant. Most of the 21 NAC proteins contain a complete NAC DNA-binding domain and a variable transcriptional regulation domain. Motif analysis showed that proteins within the same group mostly have similar motifs; however, there were no obvious similarities in gene structures among the identified BpNACs, based on exon/intron analysis. Cis-elements analysis indicated that promoters of the 21 BpNAC genes were enriched in ABRE and AC-box elements, suggesting that BpNAC genes might be related to ABA signal transduction pathways and xylem development. Quantitative real-time PCR analysis indicated that the 21 BpNACs were differentially expressed in the stem of B. platyphylla. In different stages of the growth season, BpNACs showed expression differences in the developing xylem. These genes also responded to artificial bending in tension wood and opposite wood, to varying degrees, and exhibited distinct expression patterns at different treatment times. The results indicated that these BpNAC genes might participate in tension wood development. Our study provided the basis for further functional investigations of BpNAC genes in birch.

Abstract:The growth of plant cells is inseparable from relaxation and expansion of cell walls. Expansins are a class of cell wall binding proteins, which play important roles in the relaxation of cell walls. Although there are many members in expansin gene family, the functions of most expansin genes in plant growth and development are still poorly understood. In this study, the functions of two expansin genes, AtEXPA4 and AtEXPB5 were characterized in Arabidopsis thaliana. AtEXPA4 and AtEXPB5 displayed consistent expression patterns in mature pollen grains and pollen tubes, but AtEXPA4 also showed a high expression level in primary roots. Two single mutants, atexpa4 and atexpb5, showed normal reproductive development, whereas atexpa4 atexpb5 double mutant was defective in pollen tube growth. Moreover, AtEXPA4 overexpression enhanced primary root elongation, on the contrary, knocking out AtEXPA4 made the growth of primary root slower. Our results indicated that AtEXPA4 and AtEXPB5 were redundantly involved in pollen tube growth and AtEXPA4 was required for primary root elongation.
Genome-wide identification of expansin genes in Brachypodium distachyon and functional characterization of BdEXPA27

Abstract:Plant expansin belongs to a group of cell wall proteins and functions in plant growth and development. However, limited data are available on the contributions of expansins in Brachypodium distachyon. In the present study, a total of 38 expansins were identified in B. distachyon genome. Phylogenetic analysis divided the expansins into four groups, namely EXPA, EXPB, EXLA, and EXLB. Chromosomal distribution showed that they were unevenly distributed on 4 chromosomes. A total of six tandem duplication pairs and four segmental duplication pairs were detected, which contributed to the expansion of the B. distachyon expansin gene family. Expansins in the same group shared similar gene structure and motif composition. Three types of cis-elements, development-related, hormone-related, and abiotic stresses-related elements were found in the B. distachyon expansin gene promoters. Expression profiles indicated that most of B. distachyon expansin genes participate in plant development and abiotic stress responses. Overexpression of BdEXPA27 increased seed width and length, root length, root hair number and length in Arabidopsis and showed higher germination rate in transgenic lines. This study establishes a foundation for further investigation of B. distachyon expansin genes and provides novel insights into their biological functions.
The BpMYB4 Transcription Factor From Betula platyphylla Contributes Toward Abiotic Stress Resistance and Secondary Cell Wall Biosynthesis

Abstract:The MYB (v-myb avian myeloblastosis viral oncogene homolog) family is one of the largest transcription factor families in plants, and is widely involved in the regulation of plant metabolism. In this study, we show that a MYB4 transcription factor, BpMYB4, identified from birch ( Betula platyphylla Suk.) and homologous to EgMYB1 from Eucalyptus robusta Smith and ZmMYB31 from Zea mays L. is involved in secondary cell wall synthesis. The expression level of BpMYB4 was higher in flowers relative to other tissues, and was induced by artificial bending and gravitational stimuli in developing xylem tissues. The expression of this gene was not enriched in the developing xylem during the active season, and showed higher transcript levels in xylem tissues around sprouting and near the dormant period. BpMYB4 also was induced express by abiotic stress. Functional analysis indicated that expression of BpMYB4 in transgenic Arabidopsis ( Arabidopsis thaliana ) plants could promote the growth of stems, and result in increased number of inflorescence stems and shoots. Anatomical observation of stem sections showed lower lignin deposition, and a chemical contents test also demonstrated increased cellulose and decreased lignin content in the transgenic plants. In addition, treatment with 100 mM NaCl and 200 mM mannitol resulted in the germination rate of the over-expressed lines being higher than that of the wild-type seeds. The proline content in transgenic plants was higher than that in WT, but MDA content was lower than that in WT. Further investigation in birch using transient transformation techniques indicated that overexpression of BpMYB4 could scavenge hydrogen peroxide and O 2 .– and reduce cell damage, compared with the wild-type plants. Therefore, we believe that BpMYB4 promotes stem development and cellulose biosynthesis as an inhibitor of lignin biosynthesis, and has a function in abiotic stress resistance.

Abstract:Expansins are primary agents inducing cell wall extension, and are therefore obvious targets in biotechnological applications aimed at the modification of cell size in plants. In trees, increased fibre length is a goal of both breeding and genetic engineering programmes. We used an alpha-expansin PttEXPA1 that is highly abundant in the wood-forming tissues of hybrid aspen (Populus tremula L. x P. tremuloides Michx.) to evaluate its role in fibre elongation and wood cell development. PttEXPA1 belongs to Subfamily A of alpha-expansins that have conserved motifs at the N- and C-termini of the mature protein. When PttEXPA1 was over-expressed in aspen, an extract of the cell wall-bound proteins of the transgenic plants exhibited an increased expansin activity on cellulose-xyloglucan composites in vitro, indicating that PttEXPA1 is an active expansin. The transgenic lines exhibited increased stem internode elongation and leaf expansion, and larger cell sizes in the leaf epidermis, indicating that PttEXPA1 protein is capable of increasing the growth of these organs by enhancing cell wall expansion in planta. Wood cell development was also modified in the transgenic lines, but the effects were different for vessel elements and fibres, the two main cell types of aspen wood. PttEXPA1 stimulated fibre, but not vessel element, diameter growth, and marginally increased vessel element length, but did not affect fibre length. The observed differences in responsiveness to expansin of these cell types are discussed in the light of differences in their growth strategies and cell wall composition.

Abstract:Expansin plays a crucial role in plant growth and stress resistance as a cell wall relaxation protein. The expansin family consists of four subfamilies: EXPA, EXPB, EXLA, and EXLB. However, a few reports have been previously published investigating EXLA genes. The research here aimed to characterize the PtEXLA1 gene from a popular species ( P. alba × P. glandulosa CV.84K) and evaluate its role through genetic transformation to understand its contribution to plant growth and stress resistance. The results showed that the PtEXLA1 gene was 780 bp in length, encoded 259 amino acids, and had typical characteristics of EXLA. The PtEXLA1 transgenic tobacco plants had a larger corolla in comparison to wild-type plants, and exhibited higher resistance to drought, high temperature, and salt stress based on the evaluation of chlorophyll content, relative conductivity, and malondialdehyde content. PtEXLA1 can be an efficient gene resource for stress resistance breeding of plants.

Abstract:Cold, salt and drought are the main environmental stresses affecting plant growth and crop yield. ERF1, a member of the well-studied ERF gene family, has been proven to play an important role in abiotic stress in plants. In this study, an ERF1, BpERF1.1, was overexpressed in birch (Betula Platyphylla Suk.) and 11 transgenic lines were obtained. Effects of cold, salt and drought treatments were checked on the BpERF1.1 overexpressing lines and the corresponding WT line. The transgenic lines showed improved tolerance against these abiotic stresses than the WT line. Further RNA-seq analysis identified 689 differentially expressed genes (DEGs) in the transgenic birch compared with WT, including 228 up-regulated genes and 461 down-regulated genes. The result of gene ontology enrichment analysis showed that among these DEGs, 273 genes were involved in various plant biological process, and 83% of them were involved in cellular process, metabolic process, biological regulation and response to stimulus (11%). All these results demonstrate that BpERF1.1 gene improves the tolerance and resistance of birch against cold, salt and drought stress, probably by interconnecting with other genes involved in plant response to abiotic stresses.

Abstract:Expansins are a kind of structural proteins of the plant cell wall, and they enlarge cells by loosening the cell walls. Therefore, expansins are involved in many growth and development processes. The complete genomic sequences of Brassica rapa, Brassica oleracea and Brassica nigra provide effective platforms for researchers to study expansin genes, and can be compared with analogues in Arabidopsis thaliana. This study identified and characterized expansin families in B. rapa, B. oleracea, and B. nigra. Through the comparative analysis of phylogeny, gene structure, and physicochemical properties, the expansin families were divided into four subfamilies, and then their expansion patterns and evolution details were explored accordingly. Results showed that after the three species underwent independent evolution following their separation from A. thaliana, the expansin families in the three species had increased similarities but fewer divergences. By searching divergences of promoters and coding sequences, significant positive correlations were revealed among orthologs in A. thaliana and the three basic species. Subsequently, differential expressions indicated extensive functional divergences in the expansin families of the three species, especially in reproductive development. Hence, these results support the molecular evolution of basic Brassica species, potential functions of these genes, and genetic improvement of related crops.
BpWOX11 promotes adventitious root formation in Betula pendula

Abstract:Abstract Adventitious root formation is a key step in vegetative propagation via cuttings. It is crucial for establishing birch plantations and preserve birch varieties. Although previous studies have highlighted role of WOX11 in controlling adventitious root formation, no such study has been conducted in birch. Understanding the mechanism of adventitious root formation is essential for improvement of rooting or survival rate using stem cuttings in birch. In this study, we cloned BpWOX11 and produced BpWOX11 overexpression (OE) transgenic lines using the Agrobacterium -mediated plant transformation. OE lines exhibited early initiated adventitious root formation, leading to increase the rooting rate of stem cuttings plants. RNA sequencing analysis revealed that OE lines induced the gene expression related to expansin and cell division pathway, as well as defense and stress response genes. These may be important factors for the BpWOX11 gene to promote adventitious root formation in birch cuttings. The results of this study will help to further understand the molecular mechanisms controlling the formation of adventitious roots in birch.

Genome-wide Identification, Expression and Salt Stress Tolerance Analysis of the GRAS Transcription Factor Family in Betula Platyphylla

Abstract:The GRAS gene family is a plant-specific family of transcription factors and play a vital role in many plant growth processes and abiotic stress responses. Nevertheless, the functions of the GRAS gene family in woody plants, especially in Betula platyphylla (birch), are hardly known. In this study, we performed a genome-wide analysis of 40 BpGRAS genes (BpGRASs) and identified typical GRAS domains of most BpGRASs. The BpGRASs were unevenly distributed on 14 chromosomes of birch and the phylogenetic analysis of six species facilitated the clustering of 265 GRAS proteins into 17 subfamilies. We observed that closely related GRAS homologs had similar conserved motifs according to motif analysis. Besides, an analysis of the expression patterns of 26 BpGRASs showed that most BpGRASs were highly expressed in the leaves and responded to salt stress. Six BpGRASs were selected for cis-acting element analysis because of their significant upregulation under salt treatment, indicating that many elements were involved in the response to abiotic stress. This result further confirmed that these BpGRASs might participate in response to abiotic stress. Transiently transfected birch plants with transiently overexpressed 6 BpGRASs and RNAi-silenced 6 BpGRASs were generated for gain- and loss-of-function analysis, respectively. In addition, overexpression of BpGRAS34 showed phenotype resistant to salt stress, decreased the cell death and enhanced the reactive oxygen species (ROS) scavenging capabilities and proline content under salt treatment, consistent with the results in transiently transformed birch plants. This study is a systematic analysis of the GRAS gene family in birch plants, and the results provide insight into the molecular mechanism of the GRAS gene family responding to abiotic stress in birch plants.
PagEXPA1 combines with PagCDKB2;1 to regulate plant growth and the elongation of fibers in Populus alba × Populus glandulosa

Abstract:Expansins are important plant cell wall proteins. They can loosen and soften the cell walls and lead to wall extension and cell expansion. To investigate their role in wood formation and fiber elongation, the PagEXPA1 that highly expressed in cell differentiation and expansion tissues was cloned from 84K poplar ( Populus alba × P. glandulosa ). The subcellular localization showed that PagEXPA1 located in the cell wall and it was highly expressed in primary stems and young leaves. Compared with non-transgenic 84K poplar, overexpression of PagEXPA1 can promote plant-growth, lignification, and fiber cell elongation, while PagEXPA1 Cas9-editing mutant lines exhibited the opposite phenotype. Transcriptome analysis revealed that DEGs were mainly enriched in some important processes, which are associated with cell wall formation and cellulose synthesis. The protein interaction prediction and expression analysis showed that PagCDKB2:1 and PagEXPA1 might have an interaction relationship. The luciferase complementary assay and bimolecular fluorescence complementary assay validated that PagEXPA1 can combined with PagCDKB2;1. So they promoted the expansion of xylem vascular tissues and the development of poplar though participating in the regulation of cell division and differentiation by programming the cell-cycle. It provides good foundation for molecular breeding of fast-growing and high-quality poplar varieties.

Abstract:In the present study, “Dangshansuli” (wild type, WT) and its russet mutant “Xiusu” (mutant type, MT) were used to characterize the PbEXPs in “Dangshansuli” as well as the role of IAA in mediating the expression of PbEXPss. Forty-one expansins were identified in WT and divided into EXPA, EXPB, EXLA and EXLB families. In the same EXP family, they shared similar exon–intron structure and phylogenetic characters. Eight of 41 expansin genes, screened from the transcriptome of WT and its russet mutant “Xiusu”, exhibited differential expression patterns in the epidermis of both WT and MT with the development of pear fruit. In addition, the correlation coefficients between IAA content and expression of expansins of PbEXPB3b and PbEXP4a in MT were 0.763 and 0.762, respectively. After treatments with IAA, NPA and AVG, the expression levels of PbEXPB3b and PbEXP4a changed significantly 15, 30, or 45 days after full bloom (DAFB). The longitudinal and vertical diameters of the epidermis of WT and MT at different stages were measured. We found that high IAA levels enhanced the expression of PbEXPB3b and PbEXP4a in epidermis of MT at the russet turning stage of 75–100 DAFB, which promoted cell expansion and microcrack occurrence and led to russet on the pear fruits.

In this study, the functions of two expansin genes, AtEXPA4 and AtEXPB5 were characterized in Arabidopsis thaliana. AtEXPA4 and AtEXPB5 displayed consistent expression patterns in mature pollen grains and pollen tubes, but AtEXPA4 also showed a high expression level in primary roots. Two single mutants, atexpa4 and atexpb5, showed normal reproductive development, whereas atexpa4atexpb5 double mutant was defective in pollen tube growth. Moreover, AtEXPA4 overexpression enhanced primary root elongation, on the contrary, knocking out AtEXPA4 made the growth of primary root slower. Our results indicated that AtEXPA4 and AtEXPB5 were redundantly involved in pollen tube growth and AtEXPA4 was required for primary root elongation.

Characterization and Functional Analysis of the Poplar Pectate Lyase-Like Gene PtPL1-18 Reveal Its Role in the Development of Vascular Tissues

Abstract:Pectin is a major component of plant cell walls, and the structure of pectin impacts on the properties of wood. Although we know that pectate lyase (PL, EC 4.2.2.2) has a major influence on the structure of pectin, our knowledge of Pectate lyase-like genes (PLL) in tree species remains limited. To better understand the characteristics of PLL genes in trees and to identify novel PLL genes that are potentially involved in the development of wood, we performed comprehensive analyses of gene structures, phylogenetic relationships, chromosomal locations, gene duplication events, conserved protein motifs, and gene expression patterns of 30 PLLs in Populus trichocarpa (PtPL1s). We performed an in silico gene expression profiling and quantitative real-time PCR analysis and found that most of the PtPL1 genes from subgroups Ia and Ib were highly expressed in xylem. PtPL1-18 from subgroup Ia was preferentially expressed in developing primary xylem and in xylem cells that were developing secondary walls. Overexpression of PtPL1-18 in poplar reduced plant growth and xylem development. Reduced secondary cell wall thickening and irregular xylem cells were observed in the transgenic trees, probably due to their lower pectin content. Although pectin is not a major component of plant secondary cell walls, our results are consistent with the PtPL1 genes performing important functions during wood formation.

Abstract:The WRKY family of transcription factors (TFs) is one of the most diverse families in plants, playing crucial roles in various plant growth and stress response processes. Asian white birch (Betula platyphylla) is a globally distributed tree species that holds ecological, medical, and economic significance. However, the regulatory mechanisms of WRKY TFs in birch remain poorly understood. Herein, we cloned and characterized the BpWRKY49 gene from birch. Through bioinformatics analyses, we revealed the potential involvement of BpWRKY49 in both biotic and abiotic stress responses. In addition, BpWRKY49 was found to be localized in the nucleus and exhibited transcriptional activity in yeast. Transactivation assays further confirmed that BpWRKY49 exhibited transcriptional activity at its C-terminal end. Notably, our binding specificity assays demonstrated the specific interaction of BpWRKY49 with the W-box cis element in vitro. Furthermore, tissue-specific expression analysis demonstrated that BpWRKY49 exhibited the highest expression level in the roots. Real-time quantitative PCR (RT-qPCR) analysis of birch plants subjected to salt and drought treatments revealed that BpWRKY49 displayed significant 30-fold and 10-fold upregulations under salt and drought stress conditions, respectively. DAP-seq analysis of BpWRKY49 identified a total of 21,832 peaks, with 3477 occurring in the promoter region of genes. Gene ontology (GO) enrichment analysis highlighted prominent terms related to defense against biotic stress, followed by terms associated with abiotic stress and development. Y1H assays of three genes provided evidence for the binding ability of BpWRKY49 to the promoters of BpPUB21, BpBTL15, and BpHIP47 in vitro. Collectively, our findings strongly suggest that BpWRKY49 possesses diverse functions and may activate multiple genes to contribute to various biological processes, including salt stress tolerance, in birch.
