Abstract:The aqueous two-phase extraction process (APTE) for polysaccharides from the fruits of Ilex cornuta (Lindl. et Paxt.) (PFIC) was studied, and its monosaccharide composition and antioxidant activities were analyzed. After screening and selection of isopropanol/ammonium sulfate as the aqueous two-phase system, various parameters of crude extract amount, ultrasonic time, extraction temperature and extraction time on the extraction process was investigated in detail and optimized by response surface methodology (RSM). The obtained PFIC was characterized by FT-IR and its monosaccharide composition was analyzed by GC-MS. Furthermore, the antioxidant activities of PFIC were evaluated by measuring the scavenging rates of DPPH radical, hydroxyl radical and superoxide anion radical. Under the optimal conditions in which crude extract amount was 2.23 g, ultrasonic time was 17.5 min, extraction temperature was 69 â„ƒ and the extraction time was 1.55 h, the yield of polysaccharides reached 50.87% which was close to the predicted value of 50.78%. FT-IR analysis confirmed that the extract was polysaccharide, and GC-MS analysis indicated that the monosaccharide components of the polysaccharides were rhamnose, arabinose, xylose, mannose, glucose and galactose. Antioxidant results showed that PFIC showed strong scavenging ability to DPPH radical, fine scavenging ability to hydroxyl radical and certain scavenging ability to superoxide anion radical. The ultrasonic-assisted APTE process for PFIC optimized by RSM is reasonable and reliable, and the obtained PFIC has the potential to be utilized as natural antioxidants.

Abstract:A series of novel histone deacetylase (HDAC) inhibitors were designed, synthesized and evaluated based on the strategies of a hybrid of the classic pharmacophore of HDAC inhibitors with the thiazolidinone scaffold. Some of the compounds 12i showed potent HDAC1 inhibition with nM IC50 values, more importantly, compound displayed much better anti-metastatic effects than vorinostat (SAHA) against migration of the A549 cell line. Further mechanism exploration implied that compound 12i may inhibit tumor metastasis via modulating the epithelial-mesenchymal transition (EMT) and upregulating the acetylation of α-tubulin.

Abstract:In the present work, the ultrasonic-assisted extraction (UAE) of total flavonoids (TF) from Pteris cretica L. was optimized by response surface methodology (RSM) on the basis of a single-factor experiment. The optimized UAE parameters were as follows: Ethanol concentration 56.74%, extraction time 45.94 min, extraction temperature 74.27 °C, and liquid/solid ratio 33.69 mL/g. Under the optimized conditions, the total flavonoids yield (TFY) was 4.71 ± 0.04%, which was higher than that obtained by heat reflux extraction (HRE). The extracts were further analyzed by HPLC, and five major flavonoids, including rutin, quercitrin, luteolin, apigenin, and luteolin-7-O-glucoside, were identified and quantified. Furthermore, the results of the antioxidant test showed that the TF extract obtained under optimized UAE conditions exhibited good 2,2-diphenyl-1-picrylhydrazyl radical (DPPH•) and 2,2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) radical (ABTS+•), nitric oxide radical (NO•) scavenging activities, and ferrous ion (Fe2+) chelating capacity, with IC50 values of 74.49, 82.92, 89.12, and 713.41 μg/mL, respectively. Results indicated that the UAE technique developed in this work was an efficient, rapid, and simple approach for the extraction of flavonoids with antioxidant activity from P. cretica.

Abstract:Polyphenols are indispensable as the main antioxidant substances of Phyllanthus emblica. In this study, ultrasonic-assisted extraction of emblic leafflower fruit polyphenol was performed with ethanol, and a response surface method was used to optimize the extraction results and determine parameters for the best conditions. These parameters included a drying temperature of 80 °C emblic leafflower fruit pulp, 60% alcohol concentration, solid‒liquid ratio, ultrasonic extraction time of 24 min and ultrasound temperature of 45 °C. Under the experimental conditions, the yield of polyphenols obtained from P. emballiae was 10.40%, which was almost the same as the ideal prediction value (10.39%). The established regression model shows a good fitting degree.

Abstract:A series of natural deep eutectic solvents (NADESs) were prepared with choline chloride, betaine, and a variety of natural organic acids in order to find new environmentally-friendly green solvents to replace the traditional solvents. The NADESs were employed to extract flavonoids from Potentilla fruticosa L. (PFL) with the help of ultrasound. The eutectic solvent diluted with an appropriate amount of water improved the extraction ability of flavonoids due to the decrease of solution viscosity. The microstructure of the raw sample and the samples subjected to ultrasonic bath in different solutions were observed using scanning electron microscope (SEM) to determine the role of the NADESs in the extraction process. The DPPH method and glucose consumption method were used to study the antioxidant and hypoglycemic ability of flavonoid compounds in PFL. Single factor method and response surface methodology (RSM) were designed to analyze the effects of three extraction parameters, including solvent/solid ratio, ultrasonic power, and extraction time, on the extraction yield, antioxidant capacity, and hypoglycemic capacity, and the corresponding second-order polynomial prediction models were established. The optimal extraction conditions for the maximum extraction yield, antioxidant capacity, and hypoglycemic capacity were predicted by RSM, and the reliability of RSM simulation results was verified by a one-off experiment.

Abstract:To optimize the ultrasonic‐assisted biphasic aqueous extraction conditions for polyphenolic compounds from Vaccinium dunalianum Wight leaves and investigate their antioxidant and tyrosinase inhibition activities, single‐factor experiments were conducted to investigate the effects of ethanol volume fraction (%), ammonium sulfate mass fraction (%), solid‐liquid ratio (g/mL), ultrasonic temperature (°C), and ultrasonic time (min) on polyphenolic content during extraction. Based on these experiments, three key factors influencing extraction were selected for response surface methodology (RSM) optimization. The results indicated that under conditions of 26% ethanol, 20% ammonium sulfate, a solid‐liquid ratio of 1:30, and extraction for 35 minutes at 50°C, the polyphenol content reached 61.62 mg/g. The relative contents of 6'‐O‐caffeoylarbutin, β‐arbutin, and chlorogenic acid were 34.45%, 4.56%, and 31.06%, respectively. The DPPH· and ABTS+· scavenging rates were above 95% and 96%, respectively, and the ferric reducing ability exhibited a significant dose‐effect relationship. The inhibition rates of monophenolase and diphenolase activities of tyrosinase were 43.84% and 35.73%, respectively. The optimized process for ultrasonic‐assisted biphasic aqueous extraction of polyphenols from Vaccinium dunalianum Wight leaves demonstrated significant antioxidant and tyrosinase inhibition activities.

Abstract:The objective of this study was to develop an ultrasonic-assisted procedure for the extraction of total phenolics from Citrus aurantium L. blossoms (CAB) and evaluate the free radical scavenging activity and anti-HMG-CoA reductase activity of the total phenolics. In this work, a Box– Behnken design based on single-factor experiments was used to explore the optimum extraction process. Under the optimum conditions (extraction solvent 70.31% ethanol, extraction temperature 61.94 °C, extraction time 51.73 min, and liquid-to-solid ratio 35.63 mL/g), the extraction yield of total phenolics was 95.84 mg gallic acid equivalents (GAE)/g dry matter (DM), which was highly consistent with the theoretical value (96.12 mg GAE/g DM). The higher contents of total phenolics and five main phenolic compounds obtained from the optimized ultrasonic-assisted extraction (UAE) proved its efficiency when compared with conventional heat reflux extraction (HRE). The total phenolic extract showed excellent free radical scavenging properties against 1,1-diphenyl-2-picrylhydrazyl radical (DPPH·), 2,2’-azino-bis-(3-ethylbenzthiazoline-6-sulphonic acid) radical (ABTS+·), hydroxyl radical (·OH) and superoxide anion radical (·O2−), with IC50 values of 197.007, 83.878, 218.643, and 158.885 μg/mL, respectively; the extracts also showed good inhibition of β-hydroxy-β-methylglutaryl-CoA reductase (HMG-CoA reductase) activity, with an IC50 value of 117.165 μg/mL. Total phenolics from CAB could be a potential source of natural free radical scavenger and HMG-CoA reductase inhibitor.

Abstract:The study aimed to improve the extraction rate of Platycodon grandiflorum roots polysaccharides (PGPs) using ultrasound-assisted extraction (UAE). A comparative analysis was undertaken to evaluate polysaccharides content, molecular weight distribution, monosaccharide composition, preliminary structure, antioxidant, and hypoglycemic activity of UAE in comparison with heating water extraction (HWE). The optimum extraction conditions included a liquid-to-material ratio of 20 mL/g, ultrasonic power of 150 W, extraction temperature of 70 °C, and extraction time of 20 min, resulting in a significantly greater polysaccharides (12.011 ± 0.91 %) compared to HWE (7.62 ± 0.18 %). Through Sephacryl S-100 column elution, two homogenous fraction (PGP-U extracted with UAE and PGP-H extracted with HAE) were obtained. The molecular weight of PGP-U and PGP-H was 3.14 kDa and 3.44 kDa, respectively, mainly composed of different proportions of fourteen monosaccharides. Fourier transform infrared spectroscopy (FT-IR) and Nuclear Magnetic Resonance (NMR) spectra experiment results showed that the two polysaccharides were pyranose ring with α- and β-glycoside bond. PGP-U and PGP-H exhibited specific antioxidant activities, encompassing total reducing force, scavenging of DPPH radicals, ABTs radicals and hydroxyl radicals in vitro, along with mitigation of H 2 O 2 -induced damage in HepG2 cells. Moreover, PGP-U exerted significantly stronger inhibitory activities against α-amylase and α-glucosidase and could significantly enhances the glucose uptake capacity and intracellular glycogen content of insulin-resistant HepG2 (IR-HepG2) cells.

Abstract:Polygonatum sibiricum Polysaccharide (PsP) with antioxidant function is the main active component of Polygonatum sibiricum (P.sibiricum). The currently poor extraction yield and extraction methods of PsP cannot meet the application of that in food industrial production. In this research, an ultrasound-assisted extraction-deep eutectic solvents (UAE-DESs) method, which has never been used in the PsP industry, was first used to extract PsP. The extraction conditions were optimized by the response surface method (RSM). Both the extraction yield and antioxidant function were simultaneously considered during the optimization process. The indicators of PsP's level and antioxidant activity in vitro were used to present the extraction yield of the UAE-DESs method, the purity, and the antioxidant effect of PsP. Under the optimal conditions, which included a liquid–solid ratio of 26:1 (mL:g), extraction temperature of 80 °C, ultrasonic time of 51 min, and ultrasonic power of 82 W, the PsP extraction yield could reach (43.61 ± 0.09)%, which was obviously higher than single DESs (33.81%) and UAE (5.83%), respectively, and the PsP appeared favorably antioxidant function. This research proposed an efficient extraction method for PsP, filled the basic research gap, and further improved the development of PsP as a dietary supplement with antioxidant function in the food industry.

Abstract:Ultrasound-assisted extraction (UAE) or ultrasonic extraction technology has been used for the isolation of bioactive compounds from Argyreia speciosa (Linn. f.) Sweet plant (family: Convolvulaceae). Argyreia speciosa is an Indian traditional medicinal plant having strong evidence of various properties widely used in Ayurveda. Dried flowers were used for extraction and the response surface methodology (RSM) was used to optimize the conditions of extraction for the yield of total phenolics and antioxidants from A. speciosa flowers (ASF) in respect of three variables: temperature, power, and time. Damage and structural changes of the bioactive compounds were studied by scanning electron microscopy (SEM) and Fourier-transform infrared (FTIR) spectroscopy techniques. The optimum UAE conditions obtained by RSM for the yield of total phenolic content from ASF were 42.41°C temperature, 43.62 % power, and 26.51 min process time and conditions for the maximum antioxidant activity by DPPH radical scavenging were 41.36°C temperature, 57.02 % power, and 26.63 min. These findings showed that the UAE under optimum conditions increased the percentage yield of bioactive compounds. SEM results showed that there was no relevant damage to the cells, and FTIR spectroscopy verified that there were no changes in chemical structure. It has been concluded that UAE method is an effective technique for enhancing the yields of major phenolic compounds from A. speciosa flowers.

Abstract:Ultrasound-assisted extraction for bioactive compound retrieval is a viable alternative to traditional extraction methods. Employing ultrasound-assisted extraction, this study aimed to enhance the scavenging capacity of 2,2-diphenyl-1-picrylhydrazyl (DPPH), 2,2′-azino-bis(3-ethylbenzothia-zoline-6-sulfonic acid) (ABTS), total polyphenol content (TPC), and flavonoid content in Ainsliaea acerifolia (A. acerifolia) through response surface methodology (RSM). Initially, the impact of extraction temperature, time, and ethanol concentration on DPPH scavenging capacity, ABTS, TPC, and flavonoid content was assessed. Optimal conditions for maximizing antioxidant activity and TPC were determined as 78% (v/v) ethanol, 60 °C extraction temperature, and 91 min of extraction time. High-performance liquid chromatography (HPLC) analysis of the optimized extract revealed dicaffeoylquinic acid as the primary polyphenol in A. acerifolia extracts, comprising the majority of phenolic compounds (102.06 mg/g DW). This model enabled the optimization of conditions for phenolic compound extraction with antioxidant properties from A. acerifolia, highlighting its potential as a source of antioxidant compounds for industrial, pharmaceutical, and food applications.

Polysaccharides from Trichosanthes Fructus via Ultrasound-Assisted Enzymatic Extraction Using Response Surface Methodology

Abstract:An efficient procedure for ultrasound-assisted enzymatic extraction of crude polysaccharides from Trichosanthes Fructus (crude TFP) using response surface methodology (RSM) was developed. The Box–Behnken design was applied to optimize the effects of pH ( X 1 ), enzyme amount ( X 2 ), extraction temperature ( X 3 ), and liquid-to-solid ratio ( X 4 ) on the extraction. The statistical analysis indicated that the independent variables ( X 4 , X 2 , and X 3 ), the quadratic coefficients ( X 1 2 , X 2 2 , X 3 2 , and X 4 2 ), and the interaction coefficient ( X 1 X 3 ) had significant impact on the yield of crude TFP. The optimal conditions were determined as follows: pH 4.5, enzyme amount 5000 u/g, extraction temperature 45°C, and liquid-to-solid ratio 30 ml/g. The experimental yield of crude TFP was 6.58%, which was very close to the predicted yield of 6.71%. TFPI was then purified and characterized with Sephadex G-100 column, UV-Vis, GPC, and FT-IR. The average molecular weight of TFPI was calculated to be 1.49 × 10 5 Da. TFPI exhibited strong reducing power and possessed not only remarkable scavenging activities against ABTS •+ and DPPH radicals, but also high antitumor activities in C4-2, DU145, and PC3 cells. The results suggest that Trichosanthes Fructus and TFPI could be a novel potent natural medicine with antioxidant and antitumor activities.

Abstract:We have compared, in 60 adult patients, the cardiovascular effects of glycopyrronium 5 micrograms kg-1 and 10 micrograms kg-1 given either simultaneously or 1 min before edrophonium 1 mg kg-1. Significant differences between the four groups were detected (P less than 0.001). Both groups receiving 10 micrograms kg-1 showed increases in heart rate of up to 30 beat min-1 (95% confidence limits 28-32 beat min-1). Use of glycopyrronium 5 micrograms kg-1 provided greater cardiovascular stability and, given 1 min before the edrophonium, was sufficient to minimize early, edrophonium-induced bradycardias. This low dose of glycopyrronium provided good control of oropharyngeal secretions.

Abstract:In this study, ultrasonic-assisted (UA) alcohol/salt-based aqueous two-phase system (ATPS) method was constructed to extract lotus rhizome epidermis (LRE) polyphenols. The extraction conditions were optimized as salt concentration 26.75 %, ethanol concentration 25.45 %, ultrasonic power 487 W and liquid-solid ratio 35.33 mL/g by comparing response surface methodology (RSM) and artificial neural network (ANN) models. Then, l-dopa (2.35 ± 0.036 mg/g dw), gallocatechin (1.66 ± 0.0035 mg/g dw) and epigallocatechin (1.37 ± 0.0035 mg/g dw) were determined as major polyphenols in LRE by using UA-ATPS method. Moreover, study showed that ultrasound, van der Waals force, hydrogen bond and salting out could accelerate the mass transfer and extraction of polyphenols in LRE cells. The high-pressure cavity and collapse effect of ultrasound could also accelerate the extraction of polyphenols. In vitro antioxidant experiments showed that LRE polyphenols have good antioxidant ability. In sum, this study developed a green and efficient extraction method to enhance the profitability of LRE in food and medicine industries.

Abstract:Plant-derived flavonoids have been attracting increasing research interest because of their multiple health promoting effects, where numerous investigations were carried out on the optimization of extraction and bioactivities. This study aims to optimize the extraction process of compound flavonoids (CFs) from Chinese herbal medicines and detect their antioxidant activity in vitro. CFs were extracted from the raw materials named "medicine food homology," composed of hawthorn, lotus leaf, tartary buckwheat, cassia seed, Lycium barbarum, and Poria cocos in a mass ratio of 4 : 2 : 2 : 1.5 : 1 : 1. L9 (34) orthogonal design, level effect and engineering average estimation, and quantification theory were utilized to improve the extraction method of CFs, and the predictive model for CFs yield was constructed. The 2,2ʹ-diphenyl-1-picrylhydrazyl (DPPH), 2,2ʹ-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), hydroxyl radical scavenging rate, and reducing power of CFs were measured. The highest CFs yield was obtained under the following extraction condition: liquid-solid ratio of 35 : 1 mL/g, extraction temperature of 75°C, extraction duration of 75 min, and extraction mode enzyme-assisted extraction. The forecasted yield was 37.62%. The result was accurate and the established prediction equation was reliable (R = 0.95). The antioxidant activity of CFs was significantly positively correlated with the concentration from 0.05 to 0.4 mg/mL. The DPPH, ABTS, hydroxyl radical scavenging abilities, and the reducing power of CFs were 81.82 ± 1.75%, 49.35 ± 0.09%, 89.78 ± 0.66%, and 0.232 ± 0.001 at the concentration of 0.4 mg/mL, respectively. CFs could be exploited as natural antioxidants in pharmaceuticals and functional foods.

Abstract:With the proposal of the 2030 Agenda for Sustainable Development, the Chinese medicine extraction technology has been innovatively improved to prioritize low energy consumption, sustainability, and minimized organic solvent utilization. Forsythia suspensa (FS) possesses favorable pharmacological properties and is extensively utilized in traditional Chinese medicine. However, due to the limitations of the composition and extraction methods, its potential has not been fully developed. Thus, a combination of ultrasound-assisted extraction (UAE), enzyme-assisted extraction (EAE), and β-cyclodextrin extraction (β-CDE) was employed to isolate and purify rutin, phillyrin, and forsythoside A from FS. The results demonstrated that the efficiency of extracting enzymatic and ultrasound assisted β-cyclodextrin extraction (EUA-β-CDE) was highly influenced by the temperature and duration of hydrolysis, as well as the duration of the extraction process. According to the results of the single-factor experiment, Box-Behnken design (BBD) in Response surface method (RSM) was used to optimize the experimental parameters to achieve the maximum comprehensive evaluation value (CEV) value. The EUA-β-CDE compared with other extraction methods, has good extraction effect and low energy consumption by high performance liquid chromatography (HPLC), scanning electron microscopy (SEM), calculation of power consumption and CO2 emission The EUA-β-CDE compared with other extraction methods, has good extraction effect and low energy consumption by HPLC, SEM, calculation of power consumption and CO2 emission. Then, the structural characteristics of EUA-β-CDE of FS extract had significant interaction with β-CD by Fourier infrared spectroscopy (FT-IR) and differential scanning calorimetry (DSC). In addition, EUA-β-CDE extract has good antioxidant and anti-inflammatory activities. The establishment of EUA-β-CDE of FS provides a new idea for the development and application of other sustainable extraction methods of traditional Chinese medicine.

Abstract:Using ultrasound (US) in proanthocyanidin (PA) extraction has become one of the important emerging technologies. It could be the next generation for studying the US mechnophore impact on the bioactive compound’s functionality. The objective of this study was to demonstrate the potential of US treatment on PAs extracted from kiwifruit (Actinidia chinensis) leaves, and to provide a comprehensive chemical composition and bioactivity relationship of the purified kiwifruit leaves PAs (PKLPs). Several methods like single-factor experiments and response surface methodology (RSM) for the four affected factors on US extraction efficiency were constructed. HPLC-QTOF-MS/MS, cytotoxicity analysis, and antioxidant activity were also demonstrated. In the results, the modeling of PA affected factors showed that 40% US-amplitude, 30 mL/g dry weight (DW) solvent to solid ration (S/S), and 70 °C for 15 min were the optimum conditions for the extraction of PAs. Furthermore, PKLPs exhibited significant radical scavenging and cellular antioxidant activity (p < 0.05). In conclusion, this study’s novelty comes from the broad prospects of using US in PKLP green extraction that could play an important role in maximizing this phytochemical functionality in drug discovery and food science fields.

Abstract:For thousands of years, plant has been widely applied in the medical area and is an important part of human diet. A high content of nutrients could be found in all kinds of plants, and the most outstanding group of nutrients that attracts scientists' attention is the high level of phenolic compounds. Due to the relationship between high phenolic compound content and high antioxidant capacity, plant extracts are expected to become a potential treatment for oxidation stress diseases including diabetes and cancer. However, according to the instability of phenolic compounds to light and oxygen, there are certain difficulties in the extraction of such compounds. But after many years of development, the extraction technology of phenolic compounds has been quite stable, and the only problem is how to obtain high-quality extracts with high efficiency. To further enhance the value of plant extracts, concentration and separation methods are often applied, and when detailed analysis is required, characterization methods including HPLC and LC/GC–MS will be applied to evaluate the number and type of phenolic compounds. A series of antioxidant assays are widely performed in numerous studies to test the antioxidant capacity of the plant extracts, which is also an important basis for evaluating value of extracts. This paper intends to provide a view of a variety of methods used in plants' phenolic compound extraction, separation, and characterization. Furthermore, this review presents the advantages and disadvantages of techniques involved in phenolic compound research and provides selected representative bibliographic examples.

Optimization and Spectrum-Effect Analysis of Ultrasonically Extracted Antioxidant Flavonoids from Persicae Ramulus

Abstract:The objectives of this study were to optimize the ultrasonic-assisted flavonoid extraction process from PR and to establish fingerprints in order to analyze the spectrum-effect relationship of antioxidant activity. The ultrasonic-assisted flavonoid extraction process from PR was optimized using RSM, and the fingerprints of twenty-eight batches of flavonoids from PR were established using UHPLC. Meanwhile, the in vitro antioxidant activity of PR was evaluated in DPPH and ABTS free radical-scavenging experiments. Then, the peaks of the effective antioxidant components were screened using the spectrum-effect relationships. The results show that the optimal extraction yield of flavonoids from PR was 3.24 ± 0.01 mg/g when using 53% ethanol, a 1:26 (g/mL) solid-liquid ratio, and 60 min of ultrasonic extraction. Additionally, the clearance of two antioxidant indices by the flavonoids extracted from PR had different degrees of correlation and showed concentration dependence. Simultaneously, the similarity of the UHPLC fingerprints of twenty-eight batches of PR samples ranged from 0.801 to 0.949, and four characteristic peaks, namely peaks 4, 12, 21, and 24, were screened as the peaks of the components responsible for the antioxidant effect of PR using a GRA, a Pearson correlation analysis, and a PLS-DA. In this study, characteristic peaks of the antioxidant effects of PR were screened in an investigation of the spectrum-effect relationship to provide a scientific basis for the study of pharmacodynamic substances and the elucidation of the mechanism of action of the antioxidant effect of PR.
