Abstract:Schematic representation of four different antimicrobial biomaterial systems based on physical action: carbon‐based materials, proteins and protein assemblies, switchable biomaterials, and bioactive glass. These materials show significant clinical potential by controlling microbial growth through complex nanoscale mechanisms. Green bacteria represent living microbes, while red bacteria indicate dead ones. Developing effective antimicrobial biomaterials is a relevant and fast‐growing field in advanced healthcare materials. Several well‐known (e.g., traditional antibiotics, silver, copper etc.) and newer (e.g., nanostructured, chemical, biomimetic etc.) approaches have been researched and developed in recent years and valuable knowledge has been gained. However, biomaterials associated infections (BAIs) remain a largely unsolved problem and breakthroughs in this area are sparse. Hence, novel high risk and potential high gain approaches are needed to address the important challenge of BAIs. Antibiotic free antimicrobial biomaterials that are largely based on physical action are promising, since they reduce the risk of antibiotic resistance and tolerance. Here, selected examples are reviewed such antimicrobial biomaterials, namely switchable, protein‐based, carbon‐based and bioactive glass, considering microbiological aspects of BAIs. The review shows that antimicrobial biomaterials mainly based on physical action are powerful tools to control microbial growth at biomaterials interfaces. These biomaterials have major clinical and application potential for future antimicrobial healthcare materials without promoting microbial tolerance. It also shows that the antimicrobial action of these materials is based on different complex processes and mechanisms, often on the nanoscale. The review concludes with an outlook and highlights current important research questions in antimicrobial biomaterials.

Abstract:Certain materials can adsorb microbes but quickly become saturated, after which no further microbial adsorption can occur. Moreover, these adsorbed microbes will reproduce at a high rate and return to the environment. The application of adsorption and antibacterial materials represents a new development in air quality improvement. In this study, composite materials, which were prepared by adding an antibacterial material to an adsorbing material, performed the functions of both types of materials. The device based on the composite materials actively adsorbed and killed airborne microbes. This material will be helpful in the sterilization and improvement of the environment.

Abstract:The escalating rise in antimicrobial resistance (AMR) coupled with a declining arsenal of new antibiotics is imposing serious threats to global public health. A pervasive aspect of many acquired AMR infections is that the pathogenic microorganisms exist as biofilms, which are equipped with superior survival strategies. In addition, persistent and recalcitrant infections are seeded with bacterial persister cells at infection sites. Together, conventional antibiotic therapeutics often fail in the complete treatment of infections associated with bacterial persisters and biofilms. Novel therapeutics have been attempted to tackle AMR, biofilms, and persister-associated complex infections. This review focuses on the progress in designing molecular biomaterials and therapeutics to address acquired and intrinsic AMR, and the fundamental microbiology behind biofilms and persisters. Starting with a brief introduction of AMR basics and approaches to tackling acquired AMR, the emphasis is placed on various biomaterial approaches to combating intrinsic AMR, including (1) semi-synthetic antibiotics; (2) macromolecular or polymeric biomaterials mimicking antimicrobial peptides; (3) adjuvant effects in synergy; (4) nano-therapeutics; (5) nitric oxide-releasing antimicrobials; (6) antimicrobial hydrogels; (7) antimicrobial coatings. Particularly, the structure-activity relationship is elucidated in each category of these biomaterials. Finally, illuminating perspectives are provided for the future design of molecular biomaterials to bypass AMR and cure chronic multi-drug resistant (MDR) infections.

Abstract:The emergence of antibiotic-resistant "superbugs" poses a serious threat to human health. Nanomaterials and cationic polymers have shown unprecedented advantages as effective antimicrobial therapies due to their flexibility and ability to interact with biological macromolecules. They can incorporate a variety of antimicrobial substances, achieving multifunctional effects without easily developing drug resistance. Herein, this article discusses recent advances in cationic polymers and nano-antibacterial materials, including material options, fabrication techniques, structural characteristics, and activity performance, with a focus on their fundamental active elements.

Abstract:The bacterial cell envelope and the biofilm matrix are major biological barriers, limiting bacterial bioavailability and propagating antimicrobial resistance. Novel anti‐infectives, such as e.g. pathoblockers, designed to disarm rather than killing the bacteria, require novel functional materials for improving their delivery. Rational development of such innovative antimicrobial therapies will much benefit from complex in vitro models, preferentially human‐based, as alternative to animal testing and for faster translation into the clinic. With the emerging problem of antimicrobial resistance, the world is facing a slow but dangerous pandemic. While the discovery of novel antibiotics is reaching a nearly exhaustive end, new concepts for anti‐infective drugs are emerging. So‐called pathoblockers aim to de‐weaponize bacteria rather than just killing them. As the target of these molecules is typically located intracellularly, however, hitherto almost unnoticed biological barriers are emerging such as the biofilm matrix, the bacterial cell envelope, efflux pumps, and eventual bacterial metabolism. This leads to a new paradigm that is to maximize bacterial bioavailability. To overcome the bacterial barriers, especially when further optimization of the active molecules is not possible, functional materials are needed to engineer innovative delivery systems. Those may not only enable novel anti‐infective molecules to reach their targets, but will also improve the bacterial bioavailability of existing anti‐infectives. Additionally, there is a need for better infection models that allow studying drug effects on both the bacteria and the host in a relevant manner as needed for rational anti‐infective drug development.

Abstract:Antimicrobial resistance is the leading cause of burden on healthcare sector. There is scientific challenge of developing new functional material as a platform to prevent and treat viral and microbial infection and cosmetic utility. In material chemistry, there is progress in the development of functional material with advent of nanotechnology with aid of synthetic organic chemistry. The properties and application of material can be changed significantly by modification of the surface functional groups (namely, COOH, HO, NH2OH, SO4,), formation of composite with inorganic material and incorporation of active pharmaceutical agents. In antibacterial application functional material of copper, silver, gold, platinum, tin, iron, cobalt, ruthenium, zinc and pharmaceutical antimicrobial agents found utility in the treatment of bacterial and hospital acquired infection with different resistant strains of microorganisms. In antiviral application many functional materials have been shown to possess remarkable antiviral ability like quantum dots, gold and silver nanoparticles, nanoclusters, carbon dots, graphene oxide and silicon materials. The polymers and dendrimers functionalized with USFDA approved antiviral agent also has potential therapeutic outcomes. Despite their difference in antiviral mechanism and inhibition efficacy, these functional material structures have unique features as potential antiviral candidates. In cosmetic applications functional material based on mica, sericite, fullerene, charcoal, peptides, mineral, lipids, glucocorticoid, nanocellulose hybrid material are extensively used. In this review, we have highlighted early promise and prospects of functional material for cosmetics, antibacterial and antiviral applications, advantages and disadvantages, Patent scenario, current challenges for translation into commercial products.

Abstract:Biological efficiency of existing antimicrobial agents is still inadequate to ensure optimal therapeutic index. Developing biocompatible advanced functional materials with antimicrobial properties could be promising for environmentally benign applications. Nanoparticles and other nanoscale materials are of great interest due to their multiple potential applications in material science, medicine, and industry. Nanomaterials possess well renowned antimicrobial activity against several microorganisms; however, it has some non-specific toxicity. Biofunctionalization of nanomaterials is one such topic to address this issue. Rational selection of therapeutically active biomolecules for design of nanoparticles will certainly increase the biological applicability. The present paper describes the current status of different types of biofunctionalized nanoparticles and their antibacterial applications. Key principles such as strategies involved at bio-/nanointerface, the structural activity relationship, and mechanism of action involved in the antibacterial activity of functionalized nanoparticles are briefly discussed. This knowledge is important from the objective of generation of advanced functional nanomaterials with antimicrobial properties.

Abstract:Fungi and bacteria afflict humans with innumerous pathogen-related infections and ailments. Most of the commonly employed microbicidal agents target commensal and pathogenic microorganisms without discrimination. To distinguish and fight the pathogenic species out of the microflora, novel antimicrobials have been developed that selectively target specific bacteria and fungi. The cell wall features and antimicrobial mechanisms that these microorganisms involved in are highlighted in the present review. This is followed by reviewing the design of antimicrobials that selectively combat a specific community of microbes including Gram-positive and Gram-negative bacterial strains as well as fungi. Finally, recent advances in the antimicrobial immunomodulation strategy that enables treating microorganism infections with high specificity are reviewed. These basic tenets will enable the avid reader to design novel approaches and compounds for antibacterial and antifungal applications.

Abstract:Pathogenic microbial biofilms that readily form on implantable medical devices or human tissues have posed a great threat to worldwide healthcare. Hopes are focused on preventive strategies towards biofilms, leaving a thought-provoking question: how to tackle the problem of established biofilms? In this review, we briefly summarize the functionalized biomaterials to combat biofilms and highlight current approaches to eradicate pre-existing biofilms. We believe that all of these strategies, alone or in combination, could represent a blueprint for fighting biofilm-associated infections in the postantibiotic era.

Abstract:Contamination of biomedical products with pathogenic microorganisms (bacteria, fungi, and viruses) is one of the main causes of hospital-acquired infections (HAI), and a major burden to the healthcare system. The development of biomaterials that can hamper the contamination of surfaces is vital to decrease patient-related infections in healthcare settings. In this landscape, this review identifies some of the latest antimicrobial strategies while paying particular attention to emerging antimicrobial biomaterials and nature-inspired antimicrobial surface topographies, which are rapidly finding application in the fabrication of biomedical engineering constructs.

Abstract:Microbial colonization on biomaterials is the main cause of failure of a successful implantation. In fact, local infections can eventually evolve in severe sepsis that might finally end up in a multi-organ failure and death of the patient. Besides, infection has become one of the toughest problems in the medical world, as microorganisms become more resistant to known drugs. Scientific research has been focussing on exploring new strategies to combat this life-threatening problem. In this review, information was collected about currently used polymeric biomaterials in the medical field and the main bacterial infections associated with their implantation. Furthermore, drug-free strategies to overcome this complication are explored, and the existing methodology required for assessment of the antibacterial activity is also described.

Abstract:Nanotechnology has emerged as a cornerstone in contemporary research, marked by the advent of advanced technologies aimed at nanoengineering materials with diverse applications, particularly to address challenges in human health. Among these challenges, antimicrobial resistance (AMR) has risen as a significant and pressing threat to public health, creating obstacles in preventing and treating persistent diseases. Despite efforts in recent decades to combat AMR, global trends indicate an ongoing and concerning increase in AMR. The primary contributors to the escalation of AMR are the misuse and overuse of various antimicrobial agents in healthcare settings. This has led to severe consequences not only in terms of compromised treatment outcomes but also in terms of substantial financial burdens. The economic impact of AMR is reflected in skyrocketing healthcare costs attributed to heightened hospital admissions and increased drug usage. To address this critical issue, it is imperative to implement effective strategies for antimicrobial therapies. This comprehensive review will explore the latest scientific breakthroughs within the metal–organic frameworks and the use of mesoporous metallic oxide derivates as antimicrobial agents. We will explore their biomedical applications in human health, shedding light on promising avenues for combating AMR. Finally, we will conclude the current state of research and offer perspectives on the future development of these nanomaterials in the ongoing battle against AMR.

Abstract:Though the history of mankind, we are strongly fighting with infected diseases that are induced by bacteria, fungi, viruses, and other pathogens. Although a variety of medicines, including antibiotics and chemicals, have been developed since about one hundred years ago, many challenges are still threatening human health, such as drug resistance and emerging novel pathogens. Therefore, the development of antimicrobial materials has bloomed. From silver containers in ancient time to synthesized chemicals in the early 20th century, the antimicrobial materials are being improved to fight microorganisms. The efficacies of antimicrobial materials are further promoted by modern technologies. Recently, the efficacies and functions of antimicrobial materials are continuously enriched along with the advances of materials science and engineering, which strengthens the ability against complicated, severe infections and improves the performances on disease treatments. Multifunctional antimicrobial materials have drawn the greater attention of researchers. The multifunctional antimicrobial materials could not only fight against infections, but also promote the efficacy of medical devices, which will play significant roles in promoting the development of advanced medical devices. This review provides an overview on the development of multifunctional antimicrobial materials. The progress in applications of multifunctional antimicrobial materials is also summarized.

Abstract:<p>Microbial colonization on material surfaces is ubiquitous. Biofilms derived from surface‐colonized microbes pose serious problems to the society from both an economical perspective and a health concern. Incorporation of antimicrobial nanocompounds within or on the surface of materials, or by coatings, to prevent microbial adhesion or kill the microorganisms after their attachment to biofilms, represents an important strategy in an increasingly challenging field. Over the last decade, many studies have been devoted to preparing meta‐based nanomaterials that possess antibacterial, antiviral, and antifungal activities to combat pathogen‐related diseases. Herein, an overview on the state‐of‐the‐art antimicrobial nanosized metal‐based compounds is provided, including metal and metal oxide nanoparticles as well as transition metal nanosheets. The antimicrobial mechanism of these nanostructures and their biomedical applications such as catheters, implants, medical delivery systems, tissue engineering, and dentistry are discussed. Their properties as well as potential caveats such as cytotoxicity, diminishing efficacy, and induction of antimicrobial resistance of materials incorporating these nanostructures are reviewed to provide a backdrop for future research.</p>

Abstract:The use of polymers and nanomaterials has vastly grown for industrial and biomedical sectors during last years. Before any designation or selection of polymers and their nanocomposites, it is vital to recognize the targeted applications which require these platforms to be modified. Surface functionalization to introduce the desired type and quantity of reactive functional groups to target a cell or tissue in human body is a pivotal approach to improve the physicochemical and biological properties of these materials. Herein, advances in the functionalized polymer and nanomaterials surfaces are highlighted along with their applications in biomedical fields, e.g., antimicrobial therapy and drug delivery.

Abstract:Significant health issues have lately been brought on by the dramatic increase in pathogenic microbes that are resistant to antibiotics. To manage or fight infections brought on by pathogens, investigators are looking for substitutes for currently used antimicrobial agents. Numerous approaches are being used to create effective antimicrobial agents, with nanotechnology being one of the most significant. Metal/metal oxide nanocomposite–based antibacterial agents work by evading the bacterial defenses against drug resistance and preventing the growth of biofilms or other critical virulence-related operations. Bacterial cell walls and membranes can be penetrated by nanocomposites, which then act by sabotaging crucial molecular processes. Nanocomposite-based antibacterial agents may exhibit synergy when used in conjunction with the proper antibiotics and aid in halting the growing global emergency of bacterial resistance. Moreover, polymer-derived nanocomposites facilitate the creation of a diverse range of healthcare equipment because of properties like improved biodegradability and biocompatibility. Once nanocomposites are incorporated, packed, or covered into various materials, they have antibacterial applications ranging from medicinal and surgical instruments to antimicrobial artificial fabrics. This review study covers the antibacterial properties of nanocomposites, developments in the understanding of their mechanism of action, and promising applications of nanocomposite-based antibacterial agents in biomedicine.

Abstract:Silver‐nanoparticle‐based composites have attracted wide attention owing to their outstanding properties as pathogen‐killing materials. When combined with various metals, polymers, or carbon forms, they exhibit antimicrobial behavior by attacking microbes with synergistic effects. Thus, its unique characteristics make it a highly suitable and promising material for dealing with diverse bacteria, fungi, and viruses, along with surface functionalization to eradicate or minimize the negative effects of microbes. Antimicrobial resistance (AMR), which develops into superbugs, poses a significant challenge to global health leading to 4.95 million deaths in 2019, necessitating the exploration of alternative strategies. AMR is responsible for a significant economic cost estimated by the World Bank, where AMR could result in additional healthcare costs of US$ 1 trillion by 2050. Thus, overcoming these drawbacks is of great importance. Fortunately, the advent of 21st century nanotechnology provides an ample opportunity to develop diverse nanomaterials along with specific functionalization to treat bacteria, fungi, or viruses more effectively with the combination of innovative technologies. Among these, silver nanoparticles (AgNPs) are considered a great boon in the area of AMR, which has a long history of practice. However, in recent years, Ag‐based composites have been designed by scientists to enhance their antimicrobial effects at minimal concentrations, thereby depending on synergism. Thus, in this review, we provide an update on the recent advances in Ag‐based composites with metals, polymers, and carbon for various antimicrobial applications. In addition, we focused on the mechanisms that assist in tackling microbes and the toxicity of AgNPs. Thus, in this review, we highlight the synergistic effects of AgNP‐based composites in combating several microorganisms.

Abstract:Antimicrobial resistance (AMR) is a growing global crisis with an increasing number of untreatable or exceedingly difficult-to-treat bacterial infections, due to their growing resistance to existing drugs. It is predicted that AMR will be the leading cause of death by 2050. In addition to ongoing efforts on preventive strategies and infection control, there is ongoing research towards the development of novel vaccines, antimicrobial agents, and optimised diagnostic practices to address AMR. However, developing new therapeutic agents and medicines can be a lengthy process. Therefore, there is a parallel ongoing worldwide effort to develop materials for optimised drug delivery to improve efficacy and minimise AMR. Examples of such materials include functionalisation of surfaces so that they can become self-disinfecting or non-fouling, and the development of nanoparticles with promising antimicrobial properties attributed to their ability to damage numerous essential components of pathogens. A relatively new class of materials, metal–organic frameworks (MOFs), is also being investigated for their ability to act as carriers of antimicrobial agents, because of their ultrahigh porosity and modular structures, which can be engineered to control the delivery mechanism of loaded drugs. Biodegradable polymers have also been found to show promising applications as antimicrobial carriers; and, recently, several studies have been reported on delivery of antimicrobial drugs using composites of MOF and biodegradable polymers. This review article reflects on MOFs and polymer–MOF composites, as carriers and delivery agents of antimicrobial drugs, that have been studied recently, and provides an overview of the state of the art in this highly topical area of research.
