Abstract:Polyhydroxyalkanoates (PHA) appear as good candidates to substitute conventional petroleum-based plastics since they have similar properties but with the advantage of being biodegradable. Wastewater streams with high organic content are feasible substrates for PHA production resulting in an opportunity for waste recovery. One of the main challenges is the optimization of the selection of microorganisms with high PHA storage capacity. This microbial selection is performed in sequencing batch reactors (SBR) operated under an aerobic feast/famine (F/F) regime. In the present study, a settling stage was added at the end of the feast phase of the enrichment cycle of a SBR fed with pre-acidified cooked mussel processing wastewater (containing up to 12 g NaCl/L). Settling and subsequent supernatant discharge favoured the wash-out of non-accumulating microorganisms as well as the removal of substances that enhanced their undesired development (proteins and carbohydrates). Microbial analysis performed by fluorescence in situ hybridization (FISH) technique showed shifts in the microbial community; the presence of genus Paracoccus increased whereas genera Comamonas decreased. Moreover, the process efficiency was improved with the increase of the PHA production yield (YPHA) and the maximum PHA storage capacity (max. PHA) from 0.48 to 0.72 CmmolPHA/CmmolVFA and from 40 to 60 wt%, respectively. The polymer composition also changed, its HB:HV ratio varied from 83:17 to 70:30. Results obtained in the present study showed that settling after the feast phase promoted the removal of carbon sources that did not contribute to PHA production and the washout of non-storing bacteria, which favoured the culture enrichment.

Abstract:It is urgent to expand the market of biodegradable alternatives to oil-derived plastics owing to (i) increasingly limited oil availability/accessibility, and (ii) the dramatic impact of traditional plastics on aquatic life, the food chain, all Earth ecosystems, and ultimately, human health. Polyhydroxyalkanoates (PHAs) are promising biodegradable polymers that can be obtained through microbial fermentation of agro-industrial byproducts, e.g., milk and cheese whey. Here, the PHA-accumulating efficiency of a mixed microbial culture (MMC, derived from activated sludges) grown on dairy byproducts (cheese and scotta whey) was measured. Bioreactor tests featuring temperature and pH control showed that both scotta and pre-treated Toma cheese whey could be used for PHA production by MMC, although scotta cheese whey supported higher PHA yield and productivity. The advantages of open MMCs include their plasticity and versatility to fast changing conditions; furthermore, no growth-medium sterilization is needed prior to fermentation. However, the use of pure cultures of efficient PHA producers may support better metabolic performances. Therefore, PHA-producing strains were isolated from a MMC, leading to the satisfactory identification of two bacterial strains, Citrobacter freundii and Leuconostoc spp., whose ability to accumulate PHAs in synthetic media was confirmed. A more detailed investigation by mass spectrometry revealed that the strain was L. mesenteroides. Although the validation of L. mesenteroides potential to produce PHA through fermentation of agro-industrial byproducts requires further investigations, this is the first study reporting PHA production with the Leuconostoc genus.

Abstract:An odd number of gapless Dirac fermions is guaranteed to exist at a surface of a strong topological insulator. We show that in a thin-film geometry and under external bias, electron-hole pairs that reside in these surface states can condense to form a novel exotic quantum state which we propose to call "topological exciton condensate" (TEC). This TEC is similar in general terms to the exciton condensate recently argued to exist in a biased graphene bilayer, but with different topological properties. It exhibits a host of unusual properties including a stable zero mode and a fractional charge +/-e/2 carried by a singly quantized vortex in the TEC order parameter.
Enhancement of PHA Production by a Mixed Microbial Culture Using VFA Obtained from the Fermentation of Wastewater from Yeast Industry

Abstract:Wastewater from the yeast production industry (WWY) is potentially harmful to surface water due to its high nitrogen and organic matter content; it can be used to produce compounds of higher commercial value, such as polyhydroxyalkanoates (PHA). PHA are polyester-type biopolymers synthesized by bacteria as energy reservoirs that can potentially substitute petrochemical-derived plastics. In this exploratory work, effluent from WWY was used to produce PHA, using a three-step setup of mixed microbial cultures involving one anaerobic and two aerobic reactors. First, volatile fatty acids (VFA; 2.5 g/L) were produced on an anaerobic batch reactor (reactor A) fed with WWY, using a heat pretreated sludge inoculum to eliminate methanogenic activity. Concurrently, PHA-producing bacteria were enriched using synthetic VFA in a sequencing batch reactor (SBR, reactor C) operated for 78 days. Finally, a polyhydroxybutyrate (PHB)-producing reactor (reactor B) was assembled using the inoculum enriched with PHA-producing bacteria and the raw and distilled effluent from the anaerobic reactor as a substrate. A maximum accumulation of 17% of PHB based on cell dry weight was achieved with a yield of 1.2 g PHB/L when feeding with the distilled effluent. Roche 454 16S rRNA gene amplicon pyrosequencing of the PHA-producing reactor showed that the microbial community was dominated by the PHA-producing bacterial species Paracoccus alcalophilus (32%) and Azoarcus sp. (44%). Our results show promising PHB accumulation rates that outperform previously reported results obtained with real substrates and mixed cultures, demonstrating a sustainable approach for the production of PHA less prone to contamination than a pure culture.

Abstract:Production of polyhydroxyalkanoates (PHAs) by open mixed microbial cultures (MMCs) has been attracting increasing interest as an alternative technology to PHA production by pure cultures, due to the potential for lower costs associated with the use of open systems (eliminating the requirement for sterile conditions) and the utilisation of cheap feedstock (industrial and agricultural wastes). Such technology relies on the efficient selection of an MMC enriched in PHA-accumulating organisms. Fermented cheese whey, a protein-rich complex feedstock, has been used previously to produce PHA using the feast and famine regime for selection of PHA accumulating cultures. While this selection strategy was found efficient when operated at relatively low organic loading rate (OLR, 2g-CODL-1d-1), great instability and low selection efficiency of PHA accumulating organisms were observed when higher OLR (ca. 6g-CODL-1d-1) was applied. High organic loading is desirable as a means to enhance PHA productivity. In the present study, a new selection strategy was tested with the aim of improving selection for high OLR. It was based on uncoupling carbon and nitrogen supply and was implemented and compared with the conventional feast and famine strategy. For this, two selection reactors were fed with fermented cheese whey applying an OLR of ca. 8.5g-CODL-1 (with 3.8g-CODL-1 resulting from organic acids and ethanol), and operated in parallel under similar conditions, except for the timing of nitrogen supplementation. Whereas in the conventional strategy nitrogen and carbon substrates were added simultaneously at the beginning of the cycle, in the uncoupled substrates strategy, nitrogen addition was delayed to the end of the feast phase (i.e. after exogenous carbon was exhausted). The two different strategies selected different PHA-storing microbial communities, dominated by Corynebacterium and a Xantomonadaceae, respectively with the conventional and the new approaches. The new strategy originated a more efficient PHA-production process than the conventional one (global PHA productivity of 6.09g-PHAL-1d-1 and storage yield of 0.96 versus 2.55g-PHAL-1d-1 and 0.86, respectively). Dissociation between the feast to famine length ratio (F/F) and storage efficiency was shown to be possible with the new strategy, allowing selection of an efficient PHA-storing culture with complex feedstock under high organic loading rates.

Abstract:Polyhydroxyalkanoates (PHAs) production with phototrophic mixed cultures (PMCs) has been recently proposed. These cultures can be selected under the permanent presence of carbon and the PHA production can be enhanced in subsequent accumulation steps. To optimize the PHA production in accumulator reactors, this work evaluated the impact of 1) initial acetate concentration, 2) light intensity, 3) removal of residual nitrogen on the culture performance. Results indicate that low acetate concentration (<30CmM) and specific light intensities around 20W/gX are optimal operating conditions that lead to high polyhydroxybutyrate (PHB) storage yields (0.83+-0.07 Cmol-PHB/Cmol-Acet) and specific PHB production rates of 2.21+-0.07 Cmol-PHB/Cmol X d. This rate is three times higher than previously registered in non-optimized accumulation tests and enabled a PHA content increase from 15 to 30% in less than 4h. Also, it was shown for the first time, the capability of a PMC to use a real waste, fermented cheese whey, to produce PHA with a hydroxyvalerate (HV) content of 12%. These results confirm that fermented wastes can be used as substrates for PHA production with PMCs and that the energy levels in sunlight that lead to specific light intensities from 10 to 20W/gX are sufficient to drive phototrophic PHA production processes.

Abstract:Background High-cost production of bioplastics polyhydroxyalkanoates (PHA) is a major concern for their large scale application. In order to produce PHA economically, new technology must be developed to reduce costs on energy consumption, fresh water and substrate usages. It is also important to conduct the PHA production process in a continuous way rather than in a batch process. Results A halophile Halomonas campaniensis strain LS21 was isolated to allow the development of a sea water based open and continuous process for PHA production utilizing mixed substrates consisting of mostly cellulose, starch, lipids and proteins. To study the feasibilities of open and long-term cultivation as well as genetic manipulation of this strain, polyhydroxybutyrate (PHB), the first member of the diverse PHA family, was taken as an example for the application of H. campaniensis LS21 in a robust and long lasting fermentation process. Wild type and recombinant H. campaniensis LS21 containing a PHB synthesis genes phbCAB were allowed respectively to grow in artificial seawater containing mixed substrates similar to kitchen wastes, including soluble and insoluble cellulose, proteins, fats, fatty acids and starch for 65 days without interruption. In the presence of 27 g/L NaCl under a pH around 10 at 37°C, the recombinant produced approximately 70% PHB and the wild type 26% during the 65 days fermentation process without infection. H. campaniensis LS21 secreted extracellular amylase, lipase, protease and cellulase simultaneously during the whole process to allow consumption of the mixed substrates. The recombinant was also found to stably maintain the phbCAB plasmid over the entire 65 days process. Conclusions The seawater based open and continuous process based on halophilic Halomonas campaniensis LS21 allowed the applications of kitchen wastes like mixed substrates as nutrients for production of bioplastic PHB. This study demonstrates the advantages of this technology in terms of energy saving (non-sterilization), seawater based (not fresh water needed), long-lasting and continuous open processing (against batch process), and low cost substrates (non-food mixed substrates). Combined with its ease of genetic manipulation, Halomonas campaniensis LS21 could be developed into a platform for low cost production of chemicals, materials and biofuels.

Abstract:Polyhydroxyalkanoate (PHA) production has been the focus of considerable research to increase productivities and reduce production costs. In this study, a fermented confectionary industry wastewater was used as feedstock for mixed microbial culture PHA production. The feedstock was dominated by lactate and ethanol (60-90 % of all soluble fermentation products). The culture selection reactor was inoculated with municipal activated sludge and was operated at an organic loading rate (OLR) of 100 Cmmol·L-1·d-1, achieving a robust PHA-accumulating enrichment, which produced up to 52.6 ± 0.4 wt% of PHA in accumulation assays. An OLR increase in the culture selection stage to 150 Cmmol·L-1·d-1 led to a PHA content of 59.1 ± 0.6, a yield of 0.93 ± 0.01 Cmol-PHA·Cmol-S-1 and a productivity of 0.93 ± 0.01 g-PHA L-1·h-1. A correlation analysis of the impact of ethanol concentrations from 3.19 to 20.3 Cmmol·L-1 in the reactor showed that ethanol inhibited PHA production rate and yield and the consumption of other carbon sources available. Microbial community analysis revealed the increase of Amaricoccus genus during the bioreactor operation time, a known PHA accumulator. The produced polymer was poly(3-hydroxybutyrate) with an average molecular weight of 4.3 × 105 Da and a polydispersity index of 1.88.

Abstract:In the last years, the production of plastics at large-scale has been an issue of great concern due to their environmental and health impacts. Biobased and biodegradable plastics, such as polyhydroxyalkanoates (PHA), have emerged as a possible more ecological and sustainable alternative to those conventional plastics. PHA can be produced by mixed microbial cultures (MMC) from waste-based feedstocks. This work aimed to maximize the PHA production from fermented brewer's spent grain by selecting an efficient PHA-accumulating MMC, applying different organic loading rates (OLR, 45 or 60 Cmmol FP /(L.d)) and sludge retention times (SRT, 4 or 2 days). Additionally, the biomass reactivation capacity after storage was evaluated. The global PHA productivity depends on the operating conditions, achieving its maximum (3.55 ± 0.8 g PHA /(L.d)) at the highest OLR and SRT tested. After storage, the global PHA productivity was similar to the one achieved before the starvation period. Depending on the operating conditions, the microbiome has its own bacterial composition signature containing various PHA-producers genera (e.g., Acidovorax , Amaricoccus , Brevundimonas , Leucobacter , Leadbetterella and Thauera ) that persisted over time, albeit at different relative abundancies. A comprehensive overview of PHA production with data on system performance and microbiome is presented, which thereby can further contribute to the upscale of such valorisation processes.

Abstract:The use of residual streams as feedstock for the production of polyhydroxyalkanoates (PHAs) is growing steadily, as it allows the valorization of waste and nutrients otherwise disposed of and the potential production of a biodegradable bioplastic. To date, the environmental and economic costs associated with this process limit its scale-up, which is why it is important to identify possible solutions and optimize the costliest steps. With this in mind, a laboratory-scale sequenced batch reactor (SBR, 5 L) was constructed to allow the selection of a mixed microbial culture able to convert volatile fatty acids (VFAs) into PHA. The reactor is fed with synthetic water containing VFAs, ammonium, phosphate, and micronutrients, typical compounds of fermented streams of certain wastes, such as cheese whey, food waste, or wastewater sludge. The biomass selected and produced by this first reactor is sent to an accumulation reactor, which is fed with a solution rich in VFAs, allowing the accumulation of PHAs. The role of aeration and its impacts on the main process parameters were analyzed. Three scenarios corresponding to different aeration rates were analyzed: 0.08, 0.16, and 0.32 vvm. The SBR was operated at an organic load rate of 600 mgCOD L−1d−1, under a dynamic feeding regime (feast–famine) and a short hydraulic retention time (HRT; 1 day). The results obtained showed that a value of 0.32 enabled better selection and better settling of the sludge. Furthermore, a potential correlation between aeration rate and VFA and NH4+ consumption rates was identified. The resulting biomass was able to accumulate up to 0.15 ± 0.02 g PHAgVSS−1.

Abstract:The present study has focused on the mainstream integration of polyhydroxyalkanoate (PHA) production with industrial wastewater treatment by exploiting three different technologies all operating in sequencing batch reactors (SBR): conventional activated sludge (AS-SBR), membrane bioreactor (AS-MBR) and aerobic granular sludge (AGS). A full aerobic feast/famine strategy was adopted to obtain enrichment of biomass with PHA-storing bacteria. All the systems were operated at different organic loading (OLR) rate equal to 1-2-3 kgCOD/m<sup>3</sup>∙d in three respective experimental periods. The AS-MBR showed the better and stable carbon removal performance, whereas the effluent quality of the AS-SBR and AGS deteriorated at high OLR. Biomass enrichment with PHA-storing bacteria was successfully obtained in all the systems. The AS-MBR improved the PHA productivity with increasing OLR (max 35% w/w), whereas the AS-SBR reduced the PHA content (max 20% w/w) above an OLR threshold of 2 kgCOD/m<sup>3</sup>∙d. In contrast, in the AGS the increase of OLR resulted in a significant decrease in PHA productivity (max 14% w/w) and a concomitant increase of extracellular polymers (EPS) production (max 75% w/w). Results demonstrated that organic carbon was mainly driven towards the intracellular storage pathway in the AS-SBR (max yield 51%) and MBR (max yield 61%), whereas additional stressors in AGS (e.g., hydraulic selection pressure, shear forces) induced bacteria to channel the COD into extracellular storage compounds (max yield 50%) necessary to maintain the granule's structure. The results of the present study indicated that full-aerobic feast/famine strategy was more suitable for flocculent sludge-based technologies, although biofilm-like systems could open new scenarios for other biopolymers recovery (e.g., EPS). Moreover, the AS-MBR resulted the most suitable technology for the integration of PHA production in a mainstream industrial wastewater treatment plant, considering the greater process stability and the potential reclamation of the treated wastewater.

Abstract:Olive mill wastewater (OMW), a highly polluting waste from the olive oil industry, was utilized as sole carbon source for the production of polyhydroxyalkanoate (PHA) by extremely halophilic Haloferax Mediterranei (H. mediterranei) in a one stage cultivation step. H. mediterranei showed remarkable cell growth and tolerated the inhibitory effect of polyphenols present in medium containing 25% of OMW. H. mediterranei cultivation conditions were optimized in medium containing 15% OMW by investigating several parameters that affect the production of PHA. The highest polymer yield (0.2g/L) and PHA content (43% PHA/cell dry mass) were achieved at 37°C, 170rpm and 22% salt concentration. Analysis of the produced PHA revealed the production of copolyester poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBHV) containing 6.5mol% 3-hydroxyvalerate (3HV). The production of PHBHV was observed without the need for fermentation step or adding external carbon source. The PHBHV displayed reduced melting points at 140.1°C and 154.4°C when compared to homopolymer polyhydroxybutyrate.
Improvement of the Polyhydroxyalkanoates Recovery from Mixed Microbial Cultures Using Sodium Hypochlorite Pre-Treatment Coupled with Solvent Extraction

Abstract:The use of mixed microbial cultures (MMC) and organic wastes and wastewaters as feed sources is considered an appealing approach to reduce the current polyhydroxyalkanoates (PHAs) production costs. However, this method entails an additional hurdle to the PHAs downstream processing (recovery and purification). In the current work, the effect of a sodium hypochlorite (NaClO) pre-treatment coupled with dimethyl carbonate (DMC) or chloroform (CF) as extraction solvents on the PHAs recovery efficiency (RE) from MMC was evaluated. MMC were harvested from a sequencing batch reactor (SBR) fed with a synthetic prefermented olive mill wastewaster. Two different carbon-sources (acetic acid and acetic/propionic acids) were employed during the batch accumulation of polyhydroxybutyrate (PHB) and poly(3-hydroxybutyrate-co-3-hydroxyvalerate) (PHBV) from MMC. Obtained PHAs were characterized by 1H and 13C nuclear magnetic resonance, gel-permeation chromatography, differential scanning calorimetry, and thermal gravimetric analysis. The results showed that when a NaClO pre-treatment is not added, the use of DMC allows to obtain higher RE of both biopolymers (PHB and PHBV), in comparison with CF. In contrast, the use of CF as extraction solvent required a pre-treatment step to improve the PHB and PHBV recovery. In all cases, RE values were higher for PHBV than for PHB.

Abstract:Industries all over the world generate huge amounts of wastewater, which needs to be treated. The integration of polyhydroxyalkanoates (PHA) production into wastewater treatment is a promising strategy for resource recovery that improves the sustainability of industrial wastewater treatment plants. This study aims to investigate PHA production from several industrial wastewaters using a microbial mixed culture enriched in bacteria of the genus Thauera. The wastewaters were classified based on the volatile fatty acid (VFA) concentration and the presence of ethanol: (W1) wastewater with VFA <12 mM (anaerobically treated dairy, yeast and paper mill wastewaters), (W2) wastewater with VFA >12 mM (acidified sugar, paper mill and brewery wastewaters), and (W3) wastewater with VFA >12 mM and ethanol (acidified starch, distillery and dairy wastewaters). No PHA accumulation was obtained with wastewaters in the category W1 because the VFA concentration was low and was used for biomass growth and maintenance. The maximum PHA accumulation of 46.5% cdw was achieved from acidified paper mill wastewater. For other wastewaters in the categories W2 and W3, PHA accumulation was in the range of 37–45.6% cdw. In the W2 category, PHA accumulation inhibition due to VFA concentration was observed in the case of acidified sugar wastewater, while for wastewaters in the W3 category inhibition was provoked by the wastewater matrix itself. Nevertheless, PHA accumulation inhibition was overcome by wastewater dilution. The presence of ethanol in wastewaters within the category W3 (>27 mM) decreased PHA accumulation circa 20%. The thermal characteristics of the copolymer PHB:PHV obtained in the present study are similar to those of commercial PHAs. Additionally, the produced copolymer was used successfully as a carbon source for nitrate removal (0.72 mg NO3−/mg PHA). This study shows that resource recovery in the form of biopolymers is an option for several industrial wastewaters and that the copolymers obtained are biodegradable.

Abstract:The chlorophyll ethanol-extracted silkworm excrement was hardly biologically reused or fermented by most microorganisms. However, partial extremely environmental halophiles were reported to be able to utilize a variety of inexpensive carbon sources to accumulate polyhydroxyalkanoates. In this study, by using the nile red staining and gas chromatography assays, two endogenous haloarchaea strains: Haloarcula hispanica A85 and Natrinema altunense A112 of silkworm excrement were shown to accumulate poly(3-hydroxybutyrate) up to 0.23 g/L and 0.08 g/L, respectively, when using the silkworm excrement as the sole carbon source. The PHA production of two haloarchaea showed no significant decreases in the silkworm excrement medium without being sterilized compared to that of the sterilized medium. Meanwhile, the CFU experiments revealed that there were more than 60% target PHAs producing haloarchaea cells at the time of the highest PHAs production, and the addition of 0.5% glucose into the open fermentation medium can largely increase both the ratio of target haloarchaea cells (to nearly 100%) and the production of PHAs. In conclusion, our study demonstrated the feasibility of using endogenous haloarchaea to utilize waste silkworm excrement, effectively. The introduce of halophiles could provide a potential way for open fermentation to further lower the cost of the production of PHAs.

Abstract:Annually, the food industry generates large amounts of waste and by-products, causing serious problems in their management and final disposal. In particular, by-products are mainly recovered as livestock feed. A most appealing strategy to valorize them has herein been investigated, through polyhydroxyalkanoate (PHA) production. In this view, a stream rich in volatile fatty acids deriving from the acidogenic fermentation of reground pasta (RP), a farinaceous food-industry by-product, was used as a carbon source for PHA production with a phototrophic purple bacteria (PPB) consortium. PPB are very versatile organisms that present a unique metabolism allowing them to adapt to a variety of environmental conditions. The PPB-PHA enrichment phase was performed in a lab-scale semi-continuous photo-bioreactor under a permanent carbon feast regime, with organic loading rate (OLR) increments from 14 to 19 mmolC/Ld. The results showed that the fermented RP solution composition (with 23.4% of HV precursors on a COD basis) was suitable for the PHBHV copolymer production, with the PPB consortium being capable of reaching a very high content in the hydroxyvalerate (HV) monomer, with a maximum of 60% (gHV/gPHA). Regarding the PHA accumulation stage where the light intensity was increased up to 20.2 W/L, a further increase in the culture PHA content by 76% after 12 h was obtained. Overall, these results open the possibility of valorizing food-industry by-products through the development of a biocatalytic process for PHA production with PPB, thus making the overall approach more sustainable from a green perspective.

Abstract:The feasibility of exopolysaccharides (EPS) production from cheese whey using Chlorella vulgaris was investigated as an example of circular bioeconomy application. The effects of dairy waste utilization in EPS biosynthesis and rheological properties were evaluated, comparing with both control conditions and commercial xanthan gum (CXG). A twofold increase in yield, up to 0.32 g L-1, was observed when Chlorella vulgaris was used for EPS production from whey rather than conventional fertilizers. Additionally, the EPS produced using cheese whey exhibited superior pseudoplasticity in the 0.4-1.0 (w/v) range compared to the control. The EPS from the whey wastewater contained functional groups similar to those of CXG (82.7 %). Moreover, the solutions containing 1 % biopolymer showed rheological profiles similar to those of the 0.4 % CXG. The molecular weight averages predominantly fell within the range of 284 to 324 kDa, as deduced using diffusion NMR, an innovative and rapid determination method for estimating EPS size. The potential applications of EPS notably extend beyond the dairy industry, reaching diverse market sectors, and thereby enhancing the competitiveness of microalgal biorefineries while contributing to the achievement of Sustainable Development Goals.

Abstract:Abstract The synthesis of polyhydroxyalkanoates (PHA) using mixed microbial cultures (MMC) has been studied in three stages: a sequencing batch reactor, a PHA accumulation reactor, and a pyruvate effect. Tainan pig farm, Chiayi pig farm, and Feng Chia University’s sludge were the three seed sludges tested. Two distinct substrates are used, such as effluent from sucrose acidified fermenter and molasses waste acidified fermenter. Both substrates showed significant COD and VFA, but the effluent from molasses waste acidified fermenter had more significant acetic acid and butyric acid levels, with 4,087 ± 128 and 8,151 ± 152 mL/L, respectively. The propionic acid content in effluent from sucrose acidified fermenter is more significant (4,789 ± 36 mg/L). The highest PHA production yield in the three seed sludges was 26.88%, 22.88%, and 21.90%. Tainan pig farm sludge had the highest PHA accumulation of 26.88%. This study validated MMC’s ability to produce PHA in 8 h using VFA efficiency. The present work employed the addition of pyruvate to enhance the MMC. The 1 g addition of pyruvate in a sequencing batch reactor (SBR) can increase the productivity of PHAs yielded 53.58 g PHAs/g VSS (%), respectively. Therefore, MMC boosted with pyruvate used in high organic wastewater to produce PHA could potentially further commercial activities.Graphical abstract

Abstract:Polyhydroxyalkanoate (PHA) is a kind of cellular biopolymers, and a renewable alternative to conventional plastics. To promote low-cost PHA production, this study developed a simple and scalable process to recover this polymer from a phenol-fed mixed culture. Cultures containing >50 % PHA were first acidified then treated with NaClO, leading to digestion of cellular materials by HClO. NaClO dosage, HCl/NaClO ratio, and treatment time were tested for their effects on product Purity and Recovery rate. Compared with NaClO alone which obtained 55-74 % Purity and 57-73 % Recovery, acidification improved the extraction efficiency to 62-83 % Purity and 75-97 % Recovery of cellular PHA. Statistical analysis was performed on dosage, ratio and treatment time. The chemical cost of obtaining 1 kg pure PHA ranged $7-10, but can be greatly reduced by applying a denser culture. However, HClO reduced the molecular weight of the PHA products, which needs to be addressed in future
