Abstract:Changes in the starch digestibility and the properties of the starches isolated from waxy, low-, and high-amylose rice kernels subjected to heat-moisture treatment (HMT) at 100 degrees C for 16 h at a moisture content of 25% were investigated. The results showed that amylose content is important in determining pasting properties for both HMTrice starches and in-kernel HMTrice starches. Amylose leaching, swelling power, and solubility index decreased in hydrothermal treatment of both isolated rice starches and rice kernels. The gelatinization temperatures of both hydrothermal-treated samples were affected by different treatments to different extents, and the changes were more pronounced for starches isolated from the treated rice kernels. The relative crystallinity increased in the starch from treated waxy and low-amylose rice kernels but decreased in the starch from treated high-amylose rice kernels. Starch digestibility study suggested that thermal treatment of rice kernels was more effective in producing low-glycemic-index starch-based foods than the same treatment of isolated starch. The study showed that HMT on rice kernels had significant impacts of physicochemical properties and starch digestibility of waxy, low-, and high-amylose rice starches to various extents.
Physicochemical and Crystalline Properties of Heat-Moisture-Treated Rice Starch: Combined Effects of Moisture and Duration of Heating

Abstract:BACKGROUND Currently there is much interest in the application of physical modification techniques such as heat-moisture treatment (HMT). The effects of HMT on normal and waxy rice starches, subject to different levels of moisture content and duration of heating, were investigated. RESULTS Water solubility index (determined at 90 °C) decreased after HMT for normal and waxy rice starches, while swelling power (determined at 90 °C) showed inconsistent results (decrease for normal type, increase for waxy type) after HMT. Values in pasting parameters of normal and waxy rice starch increased initially, but the extent of increase slowed down with moisture content and length of treatment increasing. HMT decreased gelatinization temperatures with 4 h and 8 h treatment, but when length of treatment was prolonged to 16 h gelatinization temperature increased. Degree of crystallinity decreased for all treatments, and decreased much more at higher levels of moisture content. CONCLUSION Variations in levels of moisture content and duration of heating had significant effects on physicochemical and crystalline properties to different extents.

Abstract:In this study, quinoa starch was subjected to microwave and heat moisture treatment (MHT) with various moisture content (15 %, 25 %, 35 %) and microwave power (4.8, 9.6, 14.4 W/g), and its structure and digestibility were investigated. SEM and particle size analysis indicated that MHT caused the agglomeration of starch granules and increased the particle size. Moreover, MHT increased the short-range order structure and relative crystallinity, except for MHT with moisture content (35 %). DSC results demonstrated that the gelatinization temperature and gelatinization enthalpy had a slight improvement after MHT. Moreover, MHT increased the amylose content to some extent. It was worth noting that the digestibility of quinoa starch significantly decreased. After MHT, a part of rapidly digestible starch (RDS) was converted into slowly digestible starch (SDS) or resistant starch (RS). Particularly, when moisture content was 25 %, the starch had a highest SDS + RS content. Thus, this study provided a potential approach using MHT to modulate the digestibility of starch.

Physicochemical Properties and in Vitro Digestibility of Proso Millet Starch Modified by Heat-Moisture Treatment and Annealing Processing.

Abstract:Heat-moisture treatment (HMT) and annealing (ANN) were applied to modify the proso millet starch, and then the physicochemical properties as well as the in vitro digestion of the modified starch were investigated sys-tematically. Results indicated that HMT and ANN did not change the typical A-type crystallinity. However, both processes cause cracks and dents on the surface of the granule. The gelatinization temperature increased while peak viscosity value, relative crystallinity and gelatinization enthalpy of proso millet starch decreased signifi-cantly after HTM and ANN. Meanwhile, a remarkable increase of the slowly digestible starch(SDS) and resistant starch(RS) content was noticed after HTM and ANN modification (the highest content of SDS and RS after HTM and ANN were 9.52 +/- 0.82 %, 12.03 +/- 1.36 % and 12.15 +/- 0.89 %, 8.75 +/- 1.63 %, respectively). Those results indicated that the ANN and HMT processes could modify the physicochemical properties and in vitro digestion of proso millet starch efficiently and showed potential application to produce healthy starch food with lower digestion.
Appropriate Microwave Improved the Texture Properties of Quinoa Due to Starch Gelatinization from the Destructed Cyptomere Structure

Abstract:Texture characteristics of quinoa under microwave (MW) irradiation were studied from the perspective of starch gelatinization. MW reduced the light transmittance and increased the hardness and stickiness of quinoa. Microstructure showed that MW dispersed the vesicular structure of starch aggregates into separate starch particles, resulting in the full hydration of starch and water molecules to form denser network structures. The value of peak viscosity and setback decreased in RVA after MW treatment, but the gelatinization temperature remained stable. DSC further proved that moderate MW treatment could reduce the gelatinization enthalpy of starch and made quinoa accessible to gelatinize. However, XRD showed that the crystal structure of starch was preserved, but the crystallinity increased. Finally, low field NMR showed that moderate MW stimulated the full hydration of starch to form denser network structures, while excessive MW treatment made starch partially gelatinize and form rigid structure, resulting in negative texture properties.

Abstract:Effects of microwave and traditional water bath treatment at different temperatures (70, 80, 90 degrees C) on in vitro digestion rate and antioxidant activity of digestion products of quinoa protein were investigated. The results indicated microwave treatment at 70 degrees C produced the highest quinoa protein digestion rate and the strongest antioxidant activities of its digestion products (P < 0.05), which was further verified by the results of free amino, sulfhydryl group, gel electrophoresis, amino acid profiles and the molecular weight distribution of the digestion products. However, limited exposure of active groups induced by water bath treatment might decrease the susceptibility of digestive enzymes and subsequently lower the digestibility and antioxidant activities of quinoa protein. The results suggested that a moderate microwave treatment could be used as a potential way to enhance the in vitro digestion rate of quinoa protein, as well as increase the antioxidant activities of its digestion products.

Abstract:To investigate the effect of heat-moisture treatment (HMT) on the physiochemical properties and in vitro di-gestibility of corn starch, the pasting behavior, viscoelasticity, thermal properties, long/short range structure, morphology and in vitro digestion of corn starch treated with different HMT conditions (HMT-20, 25, 30, 35 and 40 %) were characterized. Results indicated that after HMT, the pasting and disintegration behaviors of corn starch were affected and correlated with the moisture content. The dynamic viscoelasticity of corn starch was changed, and when glassy conditions were reached, the elastic properties decreased with increasing moisture while the viscous properties increased, especially for the HMT-40 %. The thermal stability of starch was improved by HMT, although the enthalpy of pasting (& UDelta;H) was reduced. Additionally, the HMT processing also promoted the conversion of RDS to SDS and/or RS (SDS and RS increased to 39.80 % and 31.68 % for HMT-40 %, respectively), which might attribute to the rearrangement of free starch molecules. The present work provides a potential approach to make functional starches with healthy properties.
Impacts of Hydrothermal Treatments on the Morphology, Structural Characteristics, and In Vitro Digestibility of Water Caltrop Starch

Abstract:The influence of hydrothermal treatments on the structural properties and digestibility of water caltrop starch was investigated. Scanning electron microscopy (SEM) showed some small dents on the surface of starch granules for samples treated with heat moisture treatment (HMT), but not for samples treated with annealing (ANN) which generally showed smoother surfaces. The gelatinization temperature of starch was generally increased by hydrothermal treatments, accompanied by a trend of decreasing breakdown viscosity. These results implied the improvement of thermal and shearing stability, particularly for HMT in comparison to ANN. After being cooked, the native and ANN-modified water caltrop starch granules were essentially burst or destroyed. On the other hand, the margin of starch granules modified by HMT and dual hydrothermal treatments remained clear with some channels inside the starch granules. X-ray diffraction revealed that the crystalline pattern of water caltrop starch changed from the CA-type to the A-type and the relative crystallinity reduced with increasing moisture levels of HMT. Results of ANN-modified water caltrop starch were mostly similar to those of the native one. Moreover, water caltrop starch modified with HMT20 and dual modification contained a pronouncedly higher resistant starch content. These results suggested that HMT, ANN, and dual modification effectively modified the functional properties of water caltrop starch.

Abstract:The effects of adenosine and the adenosine receptor agonist (-)-N(6)-phenyl-isopropyladenosine (PIA) in the presence of isoprenaline on isometric force of contraction and calcium dependent slow action potentials were studied in papillary muscles from guinea pigs pretreated with pertussis toxin and control guinea pigs. Hearts from guinea pigs treated in the same way with pertussis toxin or solvent alone underwent histological examination. For comparison, hearts from isoprenaline treated guinea pigs were also studied. Pertussis toxin specifically inactivates guanine nucleotide binding proteins (N proteins) involved in transmembrane signal transduction in many receptor systems (for example, adenosine receptors, m-cholinoceptors, and and alpha 2 adrenoceptors). In papillary muscles from control guinea pigs adenosine and PIA in the presence of isoprenaline produced a negative inotropic effect and inhibited the maximal rate of depolarisation of slow calcium dependent action potentials in potassium depolarised papillary muscles. After pretreatment with pertussis toxin the inhibitory effects both on force of contraction and on the maximal rate of depolarisation of adenosine and PIA were abolished. Treatment with pertussis toxin produced disseminated myocardial necrosis and a disseminated cellular calcium overload evidenced by glyoxal-2-bis-hydroxyanil (GBHA) staining. Similar lesions (for example, myocardial necrosis and cellular calcium overload) were also observed after treatment with isoprenaline. In controls neither myocardial necrosis nor cellular calcium overload was found. It is concluded that pertussis toxin sensitive N proteins are involved in the inhibitory effects of adenosine and PIA on force of contraction and on slow calcium inward current during beta adrenergic stimulation.(ABSTRACT TRUNCATED AT 250 WORDS)
Removal of proteins and lipids affects structure, in vitro digestion and physicochemical properties of rice flour modified by heat-moisture treatment

Abstract:Background: The objective of this experiment was to investigate the role of endogenous proteins and lipids in the structural and physicochemical properties of starch in heat-moisture treatment (HMT) rice flour and to reveal their effect on starch digestibility under heat. Results: The findings indicate that, in the absence of endogenous proteins and lipids acting as a physical barrier, especially proteins, the interaction between rice flour and endogenous proteins and lipids diminished. This reduction led to fewer starch-protein inclusion complexes and starch-lipid complexes, altering the granule aggregation structure of rice flour. It resulted in a decrease in particle size, an increase in agglomeration between starch granules, and more surface cracking on rice granules. Under HMT conditions with a moisture content of 30%, slight gelatinization of the starch granules occurred, contributing to an increased starch hydrolysis rate. In addition, the elevated thermal energy effect of HMT enhanced interactions between starch molecular chains. These resulted in a decrease in crystallinity, short-range ordering, and the content of double-helix structure within starch granules. These structural transformations led to higher pasting temperatures, improved hot and cold paste stability, and a decrease in peak viscosity, breakdown, setback, and enthalpy of pasting of the starch granules. Conclusion: The combined analysis of microstructure, physicochemical properties, and in vitro digestion characteristics has enabled us to further enhance our understanding of the interaction mechanisms between endogenous proteins, lipids, and starches during HMT. © 2024 Society of Chemical Industry.
Changes of Components and Organizational Structure Induced by Different Milling Degrees on the Physicochemical Properties and Cooking Characteristics of Quinoa

Abstract:The pasting characteristics induced by different milling degrees on the cooking quality of quinoa were investigated. The results showed that significant variations were observed in the basic composition of quinoa with different milling degrees, which indicated the existence of effects due to the degree of milling. The combination of starch and water promoted the pasting of quinoa, and with increasing milling, quinoa textural properties tended to produce softer and more viscous. LF NMR results indicated that more water was present in the dense network formed by the starch gelatinization. Rheological properties further demonstrated that milling degree enhanced the interactions between starch molecules and correspondingly increased the energy storage modulus and loss modulus. The microstructures (SEM and CLSM) confirmed that the physical property changes in quinoa were attributed to a dense network structure caused by starch pasting, which was similar to the results of RVA. The higher the amylose/amylopectin ratio (between 0.113 and 0.167), the more pronounced the characteristic peaks. The different degrees of milling of quinoa changed the quinoa crystallinity. Consequently, the degree of milling improved the cooking characteristics and enhanced the physicochemical properties. These results help the quinoa processing industry to develop healthy quinoa products with moderate milling.

Abstract:This study sought approaches to improve the textural properties of quinoa protein (QP) from the perspective of processing methods. The effect of microwave (MW) heating on the aggregation changes and gel properties of QP was investigated. The QP was treated with water bath and MW at the same heating rate. Ultraviolet absorption spectrum, intrinsic fluorescence spectrum and circular dichroism showed that MW accelerated the denaturation and unfolding of protein structure, thus more hydrophobic residues were exposed and the surface hydrophobicity was enhanced. The significant depression of free sulfhydryl groups and solubility indicated that MW promoted the production of protein aggregates. Larger protein aggregates observed in turbidity, particle size and atomic force microscopy images. MW treatment improved the gel strength and maintained the water holding capacity of QP gel. This study provided a theoretical reference for the application of MW in gluten-free foods.
Synergistic Improvement of Quinoa Protein Heat-Induced Gel Properties Treated by High-Intensity Ultrasound Combined with Transglutaminase

Abstract:BACKGROUNDQuinoa protein is enriched with a wide range of amino acids, including all nine essential amino acids necessary for the human body, and in appropriate proportions. However, as the main ingredient of gluten-free food, it is difficult for quinoa to form a certain network structure for lack of gluten protein. The aim of this work was to enhance the gel properties of quinoa protein. Therefore, the texture characteristics of quinoa protein treated with different ultrasound intensities coupled with transglutaminase (TGase) were investigated. RESULTSThe gel strength of quinoa protein gel increased markedly by 94.12% with 600 W ultrasonic treatment, and the water holding capacity increased from 56.6% to 68.33%. The gel solubility was reduced and free amino content increased the apparent viscosity and the consistency index. Changes in the free sulfhydryl group and hydrophobicity indicated that ultrasound stretched protein molecules and exposed active sites. The enhanced intrinsic fluorescence intensity at 600 W demonstrated that ultrasonic treatment affected the conformation of quinoa protein. New bands emerged in sodium dodecylsulfate-polyacrylamide gel electrophoresis indicating that high-molecular-weight polymers were generated through TGase-mediated isopeptide bonds. Furthermore, scanning electron microscopy showed that the gel network structure of TGase-catalyzed quinoa protein was more uniform and denser, thereby improving the gel quality of quinoa protein. CONCLUSIONThe results suggested that high-intensity ultrasound combined with TGase would be an effective way to develop higher-quality quinoa protein gel. & COPY; 2023 Society of Chemical Industry.
A Study on the Structural and Digestive Properties of Rice Starch–Hydrocolloid Complexes Treated with Heat–Moisture Treatment

Abstract:Rice starch-hydrophilic colloid complexes (SHCs) were prepared by incorporating xanthan gum and locust bean gum into natural rice starch. Subsequently, they underwent hygrothermal treatment (H-SHC) to investigate their structural and digestive properties with varying colloid types and added amounts of H-SHC. The results demonstrated that heat–moisture treatment (HMT) led to an increase in resistant starch (RS) content in rice starch. This effect was more pronounced after the addition of hydrophilic colloid, causing RS content to surge from 8.42 ± 0.39% to 38.36 ± 3.69%. Notably, the addition of locust bean gum had a more significant impact on enhancing RS content, and the RS content increased with the addition of hydrophilic colloids. Enzyme digestion curves indicated that H-SHC displayed a lower equilibrium concentration (C∞), hydrolysis index (HI), and gluconeogenesis index (eGI). Simultaneously, HMT reduced the solubility and swelling power of starch. However, the addition of hydrophilic colloid led to an increase in the solubility and swelling power of the samples. Scanning electron microscopy revealed that hydrophilic colloid encapsulated the starch granules, affording them protection. X-ray diffraction (XRD) showed that HMT resulted in the decreased crystallinity of the starch granules, a trend mitigated by the addition of hydrophilic colloid. Infrared (IR) results demonstrated no formation of new covalent bonds but indicated increased short-range ordering in H-SHC. Rapid viscosity analysis and differential scanning calorimetry indicated that HMT substantially decreased peak viscosity and starch breakdown, while it significantly delayed the onset, peak, and conclusion temperatures. This effect was further amplified by the addition of colloids. Rheological results indicated that H-SHC displayed lower values for G′, G′′, and static rheological parameters compared to natural starch. In summary, this study offers valuable insights into the development of healthy, low-GI functional foods.

Abstract:With the gradual stripping of cellulose and protein in the milling process, the proportion of starch gradually increased to 91.35 %. In particular, milling reduced the content of saponins from 5.06% to 0.23%. Milling increased the swelling power of quinoa flour but decreased the gelatinization enthalpy of quinoa starch. Accordingly, the microstructure of quinoa with higher milling degree after cooking was more uniform and denser. Fourier transform infrared (FTIR) analysis showed that starch-protein interaction was beneficial to protect the ordered structure of quinoa starch. The digestibility of protein and starch was interrelated, and the digestibility rate increased with the raising milling degree. By fitting the starch digestograms and combining the digestion kinetic model, three digestion patterns of quinoa starch during milling were determined. The results of the confocal laser scanning microscope showed that the physical barrier composed of protein matrix coated starch and starch-protein complex affected starch digestion.
Structural changes and components' interactions alter the digestion property of in-kernel starch from thermally processed Tibetan Qingke

Abstract:Thermal processing has been applied to the property modification of the pure Tibetan Qingke (TQ) starch, but few studies attempt to elucidate the changes in physicochemical properties of in-kernel starches under whole TQ kernels thermal processing. In this study, the molecular, structural, and digestive characteristics of SUTQ, SHTQ, SMTQ, and SBTQ (starches isolated from unprocessed, heat fluidized, microwave irradiated, and baked TQ, respectively) were measured to elucidate the effects of different thermal treatments on the in-kernel starch. After TQ thermal treatments, the amylose/amylopectin ratio increased to 0.71, 0.64, and 0.68 for SHTQ, SMTQ, and SBTQ, respectively, while the molecular weight of in-kernel starches decreased by 39.29 %-82.14 %. Furthermore, the damage degree of starch surface morphology ranked as: SHTQ > SBTQ > SMTQ. The entanglement action between protein network resulting from protein polymerization and gelatinized starch aggregations exhibited most obvious in SHTQ. Moreover, the increased fluorescence intensity, newly formed dark color rings, and disrupted Maltese cross were observed in SHTQ, SMTQ, and SBTQ, and their variation trends were consistent with the damage degree of starch surface morphology. SHTQ, SMTQ, and SBTQ showed the decreased relative crystallinity (1.79 %, 15.98 %, and 14.64 %), short-range order (0.64, 0.81, and 0.80), and double helix structures (0.48, 0.61, and 0.58) and increased degree of gelatinization (100 %, 38 %, and 45.51 %), which partially transformed slowly digested starch into rapidly digested starch. In particularly, the formation of more ordered structures (V-type crystalline and interaction between protein and starch) induced by heat fluidization treatment provided the greatest thermal stability and highest resistant starch content (42.1 %) for SHTQ. Heat fluidization was a promising thermal processing for the development of TQ or TQ starch-based functional foods.

Abstract:This study investigated the effect of removing proteins, lipids and starch on the structure, physicochemical properties and digestion properties of rice flour (with 30% moisture) treated with heat moisture treatment (HMT). According to the results, HMT caused the adhesion and agglomeration of the rice flour, promoted the binding between starch, protein and lipid molecular chains and led to the formation of complexes (especially starch-lipid complexes), which hindered the removal of non-starch components. Compared to the untreated rice flour, the HMT treated lipid-removal rice flour had small changes in their crystallinity, gelatinization temperature and viscosity property. After removing protein, the crystallinity, peak viscosity, final viscosity, breakdown and starch digestibility were sharply increased. In particular, the peak viscosity increased from 811 cP to 1746 cP and the enthalpy change increased from 5.33 J/g to 10.18 J/g. These findings are helpful in understanding the contribution of removing endogenous proteins and lipids to the physicochemical changes of HMT treated rice flour during its heating process and thus can be helpful in controlling the quality of rice flour through HMT.
Effects of adding proteins from different sources during heat-moisture treatment on corn starch structure, physicochemical and in vitro digestibility

Abstract:Proteins impact starch digestion, but the specific mechanism under heat-moisture treatment remains unclear. This study examined how proteins from various sources-white kidney bean, soybean, casein, whey-altered corn starch's structure, physicochemical properties, and digestibility during heat-moisture treatment (HMT). HMT and protein addition could significantly reduce starch's digestibility. The kidney bean protein-starch complex under HMT had the highest resistant starch at 19.74 %. Most proteins effectively inhibit α-amylase, with kidney bean being the most significantly (IC50 = 1.712 ± 0.085 mg/mL). HMT makes starch obtain a more rigid structure, limits its swelling ability, and reduces paste viscosity and amylose leaching. At the same time, proteins also improve starch's short-range order, acting as a physical barrier to digestion. Rheological and low-field NMR analyses revealed that protein enhanced the complexes' shear stability and water-binding capacity. These findings enrich the understanding of how proteins from different sources affect starch digestion under HMT, aiding the creation of nutritious, hypoglycemic foods.
Effect of the phytate and hydrogen peroxide chemical modifications on the physicochemical and functional properties of wheat starch.

Abstract:To investigate the characteristics of chemically modified wheat starch, such as oxidized (OWS), cross-linked (CLWS) and dual-modified wheat starches, cross-linked oxidized (COWS) and oxidized cross-linked (OCWS) wheat starches were obtained by 12% hydrogen peroxide (H2O2) and 2% sodium phytate treatments, respectively. After modifications of wheat starch by cross-linking and oxidization, it was determined that native wheat starch was effectively modified with respect to the structure and physicochemical characteristics, as detected by Fourier transform infrared spectroscopy (FT-IR), differential scanning calorimetry (DSC), Rapid Visco Analyzer (RVA), scanning electron microscopy (SEM) and X-ray diffraction (XRD). DSC analysis demonstrated that peak temperature (Tp) 64.41°C of the COWS was the highest. The RVA viscosity indexes of CLWS were noticeably increased compared with the other four starches, of which three oxidized starch samples showed lower RVA profiles because of oxidation. The XRD result indicated that cross-linked phosphates from sodium phytate were primarily located in the amorphous regions of starch granules. After modifications by different treatments, the starch samples exhibited different morphological characteristics, including A-type and B-type wheat starch granules, which showed much closer contact with each other by the cross-linking reaction under SEM observations. The functional characteristics, including solubility, swelling power, light transmittance and freeze-thaw stability (FTS), of all five treated and untreated wheat starches demonstrated that COWS had the highest solubility at 0.57 and that CLWS showed the best swelling power at 12.63 (g/g). The paste clarity of COWS was improved to 47.72% higher than that of native starch (7.54%), and the water loss of COWS (21.62%) was the lowest, which is beneficial to the production of quick-frozen food. Our results showed that dual-modified starch by using sodium phytate and hydrogen peroxide had significantly altered structural and functional properties. The present study provides fundamental information of dual-modified wheat starch for its potential industrial application.

Abstract:The objective of this work was to study the effects of heat-moisture treatment (HMT) of freshly harvested mature high-amylose maize (HAM) kernels on its starch structure, properties, and digestibility. Freshly harvested HAM kernels were sealed in Pyrex glass bottles and treated at 80 °C, 100 °C, or 120 °C. HMT of HAM kernels had no impact on its starch X-ray diffraction pattern but increased the relative crystallinity. This result together with the increased starch gelatinization temperatures and enthalpy change indicated starch molecules reorganization forming long-chain double-helical crystalline structure during HMT of HAM kernels. The aggregation of starch granules were observed after HMT, indicating interaction of starch granules and other components. This interaction and the high-temperature crystalline structure led to reductions in the starch digestibility, swelling power, solubility, and pasting viscosity of the HAM flours. Some starch granules remained intact and showed strong birefringence after the HAM flours were precooked at 100 °C for 20 min and followed by enzymatic hydrolysis, and the amount of undigested starch granules increased with increasing HMT temperatures. This result further supported that HMT of HAM kernels with high moisture level could increase the starch thermal stability and enzymatic resistance.
