Abstract:A deep understanding upon the relation between anthocyanin extraction performance and physicochemical properties of natural deep eutectic solvents (NADESs) was obtained, and a high-efficiency NADES (Choline chloride-Oxalic acid (ChOa)) was selected and coupled with pulse-ultrasonication for anthocyanin extraction from blueberry pomace. Response surface methodology was adopted to analyzed the significant factors' influence, interactive effects on extraction yield and optimize the extraction conditions. The theoretical optimal conditions with highest yield of total anthocyanins content (TAC) were 3.2 min of ultrasonic time, 325 W of ultrasonic power, 349.15 K of temperature, 60 mL/g of solvent to solid ratio, under which the experimental TAC value (24.27 +/- 0.05 mg C3GE/g DW) was highly correlated with theoretical result (24.28 mg C3GE/g DW). Multi-stability protective effect of ChOa on anthocyanin over acidified ethanol was revealed by stability assays in high temperature, varying pH and light exposure conditions. FT-IR and DSC analysis further characterized the influence of ChOa over acidic ethanol on typical anthocyanins functional groups and thermal behavior of anthocyanin extracts. UPLC-Triple-TOF/MS analysis identified 10 anthocyanins that mainly consisted of delphinidin, petunidin, and malvidin compounds. Overall, this study provided a green, high-efficiency and multistability protective strategy for the recovery of anthocyanins from plant source by-products.

Abstract:An excellent high-efficiency natural deep eutectic solvent (NADES, ChCl-MA) was screened out and integrated with pulse-ultrasonication technique for extracting phenolic compounds from Carya cathayensis Sarg. peels (CCSPs). Single factor experiment combined with response surface methodology (RSM) using Box-Behnken design (BBD) were employed to investigate significant factors and optimize their influence on extraction of phenolic compounds. Significant synergistic effect triggered by ChCl-MA based pulse-ultrasonication over other methods used alone were proved by comparative study concerning a variety of bioactive components and antioxidant activities. The second-order kinetic model was developed and validated (R2 > 0.99) to describe the extraction process and its mechanism; and second-order kinetic extraction rate constant (k), saturation concentration (Cs), and initial extraction rate (h) were calculated. FT-IR, DSC and SEM results further demonstrated synergistic effect and influence during extraction. Overall, this study provided a green and high-efficiency alternative for the recovery of various phenolics compounds from plant source by-products.

In this study, phytochemicals in Carya cathayensis Sarg. peels extracted with Choline chloride-Malic acid (ChCl-MA) were identified using UPLC-Triple-TOF/MS. Effect of NADES on phenolic composition, antioxidant properties and inhibition of α-glucosidase and α-amylase were evaluated. Furthermore, extraction mechanism caused by different solvents were investigated by quantum chemical calculation combined with molecular dynamic simulation. A total of 29 phytochemicals were identified, and catechin, procyanidin B1, 2,3-dihydroxybenzoic acid, pinocembrin, procyanidin B3, myricetrin were the most abundant compounds. The extract using ChCl-MA exhibited the highest phenolic compounds content, antioxidant capacity, and α-glucosidase and α-amylase inhibition activities. Larger solvent accessible surface area, more hydrogen bonds between ChCl-MA and extract, longer lifetime of the hydrogen bonds, and lower intermolecular interaction energy account for higher extraction efficiency of ChCl-MA.

Abstract:This study proposed natural deep eutectic solvent (NADES) based pulse-ultrasonication-assisted extraction (PUAE) for green recovery of crocins from gardenia fruits (Gardenia jasminoides Ellis). Choline chloride-1,2-propylene glycol (ChCl-PG, 1:2.5, mol/mol) with 35 % (v/v) water content was tailor-made to obtain the highest extraction efficiency with high surface tension, polarity, conductivity, pH, and low viscosity, density for affinity-based extraction of crocins. The optimized P-UAE conditions (extraction time 120 s, ultrasonic power 400 W, solvent-to-solid ratio 40 mL g(-1), extraction temperature 35 celcius) led to the maximum yield of crocins (36.97 mg g(-1) DW). The second-order kinetic model was fitted to the extraction process and the synergistic effect was verified by SEM and FTIR. The crocetin was identified as the main compound by UPLC-Triple-TOF/MS. The antiglycative effect and protective effect on DNA indicated the bioactive activities of crocins. Therefore, the ChCl-PG based P-UAE could serve as a green strategy to effectively extract crocins from gardenia fruits.
Ultrasonic Impact on Viscosity and Extraction Efficiency of Polyethylene Glycol: A Greener Approach for Anthocyanins Recovery from Purple Sweet Potato

Abstract:Purple sweet potatoes are known for its vibrant purple color due to high level of anthocyanins. A polyethylene glycol based ultrasonic-assisted green extraction (PEG-UAE) of anthocyanins from purple sweet potato was proposed. Different types of PEG were tested for anthocyanin extraction along with PEG concentration, liquid-tosolid ratio, ultrasonic temperature and time were investigated for its impact on viscosity and extraction efficiency. The optimum extraction condition, 42 mL/g of ratio, 83% of PEG 200 concentration, 64 degrees C of ultrasonic temperature and 80 min of sonication time, resulted in better extraction of anthocyanins (83.78 mg CE/100 g DW) and phenolics (994.88 mg GAE/100 g DW). Using UPLC-Triple-TOF/MS, ten anthocyanin and six non-anthocyanin compounds were identified and characterized, with the highest peak area for cyanidin-3-caffeoyl-p-hydroxybenzoyl sophoroside-5-glucoside (25.9%). Moreover, the anthocyanins and phenolics extraction yield along with antioxidant activity were negatively correlated with PEG viscosity, on which ultrasonication has profound effects.

Abstract:This study presents a novel approach for converting cottonseed hulls (CSHs) into valuable proanthocyanidins (PAs) through deep eutectic solvent (DES)-based ultrasound-assisted extraction (UAE-DES). Response surface methodology (RSM) was applied to optimize and model this process, resulting in maximum yields of 78.58 mg/g. The ideal PA extraction conditions were determined to be a liquid-to-material ratio of 36.25 mL/g, a water content of 33.21%, and an extraction period of 7.4 min. Molecular dynamic simulations (MDS) were performed to study the interactions between the solvent and target chemicals. Increased van der Waals forces and stronger interactions between DES and the target chemical catechin (CA) compared to those observed with methanol or water were observed. Furthermore, the optimized extract exhibited a higher PA content than can be obtained with conventional extraction methods and demonstrated antioxidant activity in vitro. The cottonseed hulls residues (CSRs) remaining after the extraction process can be used to produce activated carbon (AC CSR ), which has some capacity to adsorb methylene blue (MB) contaminants. This study offers a reference for the fruitful transformation of waste biomass into high-value products.

Abstract:The sustainable extraction of saponins was investigated using natural deep eutectic solvents (NADESs) combined with ultrasound-assisted extraction. A novel NADES (butyric acid-urea) that was responsive to ionic strength was designed and used as the extractant. Ultrasound treatment and a catalyst ferric chloride with plant cell wall breaking function were applied to improve the extraction efficiency.Since the solubility of the NADES varied significantly with ionic strength, 95% of NADES was readily separated from the water phase after the addition of sodium chloride, while saponins remained in the water phase for easy collection. The reuse capacity of NADES, the eco-friendliness of the extraction method, and the antioxidant activity of the extract were further evaluated.NADES was continuously recovered and used to extract Polygonatum sibiricum powder: the yield of saponins did not decrease after five cycles of recovery and re-extraction. The penalty point on the "Eco-scale" suggested that the extraction method was "green" (i.e. eco-friendly).Compared with ethanol extracts, the NADES extracts showed a higher saponin concentration and antioxidant activity.The study can contribute to the sustainable and green extraction of hydrophilic active substances in the food and pharmaceutical industries.

Abstract:In this study, we employed ultrasound-assisted extraction in conjunction with natural deep eutectic solvents (NADES) to investigate an innovative and sustainable approach for the selective extraction of Panax notoginseng (PN) saponins. Fifteen NADES were prepared, of which choline chloride-urea (ChCl-Ur) was selected as the optimal solvent because of the higher solubility of PN saponins. Response surface methodology using Box-Behnken design was adopted to optimize the extraction process. According to the results, NADES with a water content of 45 % (v/v) and a ChCl/Ur ratio of 1:2 (mol/mol) exhibited superior efficacy as a solvent for extracting saponins from PN. The optimal extraction conditions include a liquid–solid ratio of 40 mL/g, an extraction temperature of 47 °C, and an extraction duration of 36 min. The extraction efficiency of NADES for saponins was superior to that of water and comparable to that of 70 % (v/v) methanol. Additionally, we assessed the anti-inflammatory and anticoagulant properties of the resulting extraction solution. The TNF- α and IL-1 β inhibiting test showed that the NADES extract of saponins exhibited a stronger inhibitory effect on the mRNA expression of inflammatory cytokines TNF- α and IL-1 β when compared to an aqueous extract. And a dose-dependent relationship was observed. In the anticoagulation activity assay, the NADES extract of saponins at a concentration of 25 mg/mL significantly prolonged the activated partial thromboplastin time ( p < 0.05). Additionally, concentrations of 12.5 and 25 mg/mL exhibited significant prolongation of the prothrombin time ( p < 0.01). These findings suggest that NADES can be used as a solvent to extract PN saponins in an efficient way, and the specific concentrations of the extract possess potent bioactivities.

Abstract:Polygonatum sibiricum Polysaccharide (PsP) with antioxidant function is the main active component of Polygonatum sibiricum (P.sibiricum). The currently poor extraction yield and extraction methods of PsP cannot meet the application of that in food industrial production. In this research, an ultrasound-assisted extraction-deep eutectic solvents (UAE-DESs) method, which has never been used in the PsP industry, was first used to extract PsP. The extraction conditions were optimized by the response surface method (RSM). Both the extraction yield and antioxidant function were simultaneously considered during the optimization process. The indicators of PsP's level and antioxidant activity in vitro were used to present the extraction yield of the UAE-DESs method, the purity, and the antioxidant effect of PsP. Under the optimal conditions, which included a liquid–solid ratio of 26:1 (mL:g), extraction temperature of 80 °C, ultrasonic time of 51 min, and ultrasonic power of 82 W, the PsP extraction yield could reach (43.61 ± 0.09)%, which was obviously higher than single DESs (33.81%) and UAE (5.83%), respectively, and the PsP appeared favorably antioxidant function. This research proposed an efficient extraction method for PsP, filled the basic research gap, and further improved the development of PsP as a dietary supplement with antioxidant function in the food industry.

Abstract:Agricultural industries produce substantial quantities of phenolic-rich by-products, which have gained much attention due to their antioxidant properties. Ultrasonic technology was applied for extraction of antioxidants from the walnut green husk using ethanol as a food grade solvent. Response surface methodology (RSM) was used to optimize experimental conditions. The responses were total phenolic content (TPC), ferric reducing antioxidant power (FRAP), scavenging activity of 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical and yield. TPC varied from 6.28 to 7.23mg GA g−1 dry sample. FRAP and DPPH values varied from 0.33 to 0.46 mmol Fe2+ g−1 of dry sample and 33.98% to 56.31% inhibition, respectively. Extraction yields ranged from 33.04% to 38.72%. The optimal conditions were 60% ethanolwater mixture as solvent, temperature of 60 °C and extraction time of 30 min. Comparison of ultrasonic-assisted extraction (UAE) and conventional extraction was shown that TPC, FRAP, DPPH and yield obtained by UAE during 30 min were significantly higher than by conventional extraction during 16 hours. The extract can be used as substitute of synthetic antioxidants for food products, color and oxidative stabilization.

Abstract:A series of natural deep eutectic solvents (NADESs) were prepared with choline chloride, betaine, and a variety of natural organic acids in order to find new environmentally-friendly green solvents to replace the traditional solvents. The NADESs were employed to extract flavonoids from Potentilla fruticosa L. (PFL) with the help of ultrasound. The eutectic solvent diluted with an appropriate amount of water improved the extraction ability of flavonoids due to the decrease of solution viscosity. The microstructure of the raw sample and the samples subjected to ultrasonic bath in different solutions were observed using scanning electron microscope (SEM) to determine the role of the NADESs in the extraction process. The DPPH method and glucose consumption method were used to study the antioxidant and hypoglycemic ability of flavonoid compounds in PFL. Single factor method and response surface methodology (RSM) were designed to analyze the effects of three extraction parameters, including solvent/solid ratio, ultrasonic power, and extraction time, on the extraction yield, antioxidant capacity, and hypoglycemic capacity, and the corresponding second-order polynomial prediction models were established. The optimal extraction conditions for the maximum extraction yield, antioxidant capacity, and hypoglycemic capacity were predicted by RSM, and the reliability of RSM simulation results was verified by a one-off experiment.

Abstract:Using ultrasound (US) in proanthocyanidin (PA) extraction has become one of the important emerging technologies. It could be the next generation for studying the US mechnophore impact on the bioactive compound’s functionality. The objective of this study was to demonstrate the potential of US treatment on PAs extracted from kiwifruit (Actinidia chinensis) leaves, and to provide a comprehensive chemical composition and bioactivity relationship of the purified kiwifruit leaves PAs (PKLPs). Several methods like single-factor experiments and response surface methodology (RSM) for the four affected factors on US extraction efficiency were constructed. HPLC-QTOF-MS/MS, cytotoxicity analysis, and antioxidant activity were also demonstrated. In the results, the modeling of PA affected factors showed that 40% US-amplitude, 30 mL/g dry weight (DW) solvent to solid ration (S/S), and 70 °C for 15 min were the optimum conditions for the extraction of PAs. Furthermore, PKLPs exhibited significant radical scavenging and cellular antioxidant activity (p < 0.05). In conclusion, this study’s novelty comes from the broad prospects of using US in PKLP green extraction that could play an important role in maximizing this phytochemical functionality in drug discovery and food science fields.

Abstract:A time-saving, efficient, and environmentally friendly ultrasonic-microwave-assisted natural deep eutectic solvent (UMAE-NADES) extraction method was developed for the extraction of anthocyanins from Aronia melanocarpa . Eight different natural eutectic solvents were screened initially, and choline chloride-glycerol was selected as the extraction solvent. The extraction conditions were optimized using the response surface methodology, and the extraction rate of anthocyanins was higher than those achieved using the traditional ethanol method, natural deep eutectic solvent extraction method, and ultrasonic-microwave-assisted ethanol method. Six anthocyanins, including cyanidin-3-O-galactoside, cyanidin-3-O-glucoside, cyanidin-3-O-arabinoside, cyanidin-3-O-xyloside, cyanidin-3,5-O-dihexoside, and the dimer of cyanidin-hexoside were identified and extracted at a purity of 448.873 mg/g using high performance liquid chromatography-mass spectrometry (HPLC-MS). The compounds extracted using UMAE-NADES had higher antioxidant capacities than those extracted by the other three methods. The UMAE-NADES demonstrated significant efficiency toward the extraction of bioactive substances and has potential utility in the food and pharmaceutical industries.

Abstract:In this study, ultrasonic-assisted natural deep eutectic solvents method (UAE-NADESs) was applied to greenly and efficiently extract five polyphenols (chlorogenic acid, (+)-catechin, protocatechuic acid, caffeic acid and gallic acid) from Chaenomeles speciosa (CSP), and their contents were simultaneously determined by HPLC. On the basis of 9 kinds of customized NADESs, the best compositions were selected. Choline chloride-urea-malonic acid (ChCl-Ur-MA) was screened as the optimal compositions of NADES. The effects of six single factors on polyphenols contents were investigated. Afterwards, the combination of response surface methodology (RSM) and genetic algorithm (GA) was adopted to optimize process parameters, and results of optimal parameters were as follows: molar ratio of 1:3:3, water content of 31%, liquid-solid ratio of 11: 1 mL/g, vortex time of 30 s, extraction time of 40 min and extraction temperature of 60°C. The total yield of five components was 3.92 ± 0.03 mg/g, which was higher than traditional organic solvents. Among them, the extraction yield of (+)-catechin was improved significantly. Meanwhile, the hydrogen bond of ChCl-Ur-MA was verified by Fourier Transform Infrared Spectroscopy (FT-IR). The Sigma profiles (σ-profiles) of (+)-catechin, different NADESs, methanol and water were calculated and analyzed by COSMO model. The mechanism of discrepancie in extraction yield of (+)-catechin between NADESs and traditional organic solvents were discussed. In addition, the antioxidant activities of the extracts were determined. Compared with 50% methanol extract, the optimal NADES extract has better DPPH, hydroxyl and ABTS radicals scavenging abilities. In a word, the ternary NADES used in this study is a new and green extraction solvent that can replace organic solvents to extract five polyphenols. Natural deep eutectic solvent-ultrasound assisted extraction: A green approach for ellagic acid extraction from Geum japonicum

Abstract:In China and other Asian nations, Geum japonicum (GJ) is used as functional vegetables or as a type of folk medicine. Ellagic acid (EA) is one of the main active ingredients in GJ and has been utilized in food, cosmetics, and medicinal goods worldwide. Natural deep eutectic solvents (NADESs) have gradually replaced organic solvents for efficient extraction of plant-derived active compounds due to its environmental protection, low toxicity, low solubility, reusability, etc. In this work, several NADESs were tailored for the ultrasound assisted extraction (UAE) of EA from GJ, among which choline chloride-oxalic acid (ChCl:Oa) was the most effective. In optimal conditions, ChCl:Oa extract produced higher EA yields than common organic solvents including methanol, ethanol and acetone. In vitro antioxidant experiments showed that ChCl:Oa extract had stronger DPPH radical scavenging ability than other solvent extracts. Mechanically, Fourier transform infrared (FT-IR) results indicated that ChCl:Oa could form a hydrogen bonding with EA, which enhanced the stability of EA. Meanwhile, ChCl:Oa-UAE treatment could destroy the tissue structure of GJ, thereby improving EA yield. In conclusion, these results imply that the ChCl:Oa-UAE method might be an environmentally friendly approach for extracting EA from GJ.

Abstract:With the development of the walnut industry, the high-value utilization of walnut green husk (WGH), an agricultural byproduct arising from walnut cultivation, has emerged as a pressing concern. WGH is rich in natural bioactive compounds and possesses extremely high medicinal value. Therefore, extracting bioactive compounds from WGH for application is a potential solution. This study aimed to investigate the influence mechanism of electron beam irradiation (EBI) and X-ray irradiation (XRI) pretreatment on the extraction of bioactive compounds from WGH, focusing on structural, physicochemical, and functional properties. Scanning electron microscopy, Fourier transform infrared spectroscopy, and X-ray diffraction analysis showed that irradiation induced notable disruptions in the surface morphology and cell wall architecture of WGH. The alteration in cell wall structure facilitated the penetration of extracting solvents, thereby aiding in the extraction of active compounds from walnut green husk. EBI and XRI pretreatment at 30 kGy significantly enhanced the extraction yield (up by 17.46%–38.81%) of polyphenols, triterpenoids, flavonoids, and quinones from WGH and augmented the antioxidant (up by 19.02%–41.06%), hypoglycemic (up by 25.68%–28.64%), and anti-inflammatory (up by 28.36%–35.63%) activities of WGH extracts. Moreover, XRI exhibited a more pronounced effect than EBI. However, elevated doses caused the decomposition of bioactive compounds. Correlation analysis revealed a relationship between the structural characteristics, bioactive compound contents, and functional properties of irradiated WGH. In conclusion, this research shows the considerable potential of EBI and XRI in the industrial application of WGH.

Novel Green Three-Constituent Natural Deep Eutectic Solvent Enhances Biomass Extraction from Acanthopanax senticosus and the Extraction Mechanism

Abstract:In this work, a novel three-constituent natural deep eutectic aqueous solution (3c-NADES) composed of betaine, triethanolamine, and MgCl2·6H2O was synthesized and used for extracting three main kinds of active compounds from Acanthopanax senticosus stems with a microwave-ultrasound-assisted technique. The target analytes were determined by an ultraperformance liquid chromatography-tandem triple-quadrupole mass spectrometry (UPLC-MS/MS) approach. Single-factor experiments and Box–Behnken design were used to obtain the optimized extraction conditions: a microwave power of 600 W, an extraction temperature of 59 °C, an ultrasound power of 500 W, an extraction time of 42 min, and a solid–liquid ratio of 0.07 g/g. The yields of isofraxidin, eleutheroside E, eleutheroside B, protocatechuic acid, chlorogenic acid, and ursolic acid reached 18.76, 49.38, 174.82, 2.98, 457.39, and 16.62 μg/g, respectively, which increased from 1.04- to 3.19-fold from those of conventional extraction methods. In addition, the reusability of 3c-NADES was stable after six runs. Furthermore, the results with molecular dynamics combined with density functional theory analyses revealed that this kind of 3c-NADES is compatible with the target compounds, and the changes of the hydrogen-bond-related parameters were analyzed. Abstract:The extraction efficiency, recovery, quantification, and stability of isoflavones (daidzein, genistein and puerarin) from the wastes /by-products of kudzu roots and soy molasses were evaluated by using natural deep eutectic solvents (NADESs) coupled with ultrasound-assisted extraction. The NADES extraction was compared with the conventional Soxhlet extraction technique, and the extracted isoflavones were quantified using HPLC-UV/Vis. Results showed extractability of puerarin to be significantly increased (p < 0.0001) in proportion to the increased water concentration of NADES (9.7 ± 0.2, 12.6 ± 0.2, and 14.8 ± 0.3 mg/g in NADESs containing 10%, 20%, and 30% water, respectively). Despite having less isoflavone content, kudzu roots and soy molasses extracts obtained using NADES exhibited higher antioxidant activity when compared with methanolic extracts. Further, NADES extracts also markedly reduced the degradation degree of isoflavones. The results of this investigation support the use of green extraction techniques to obtain value-added components like isoflavones from wastes/by-products like that of kudzu roots and soy molasses, which are envisaged to find future potential applications in the food and pharmaceutical industries.
