Abstract:Fluorescence resonance energy transfer (FRET) nanoprobes based on aggregated induced emission (AIE) molecules are promising for the monitoring of biomolecules in living cells, but this nanoprobe for reactive oxygen species (ROS) has rarely been reported. Here, a novel AIE-based FRET nanoparticles (NPs) is developed for ratiometric detection and imaging of hypochlorite in live cells. Different to common FRET-based nanosensors, the NPs is prepared via facile co-assembly strategy in water using amphiphilic tetraphenylethene (TPE1, energy donor) and thienyl-diketopyrrolopyrmle (NDPP, energy acceptor). This hybrid NPs are uniform spherical, with size of approximately 120 nm. NDPP maintains bright fluorescence inside the hybrid NPs and acts as the reactive unit for hypochlorite in this FRET nanosystem. As a ratiometric fluorescent nanoprobe, the hybrid NPs shows high selectivity to hypochlorite with a detection limit calculated to be 105 nM, thanks to its stable particle size in hypochlorite sensing. Furthermore, the successful ratiometric fluorescent imaging of ClO- in HeLa cells demonstrates that our NPs can effectively pass through the cell membrane and recognize the ClO- in living cells.

Abstract:As a powerful tool for medical diagnosis and bioanalysis, conventional optical spectrometers are generally expensive, bulky and always require an accompanying data processing device. In this work, we developed a novel smartphone-based CD-spectrometer (SCDS) for high sensitive and ultra-portable colorimetric analysis, with the advantage of cost-effective and simplicity. The distance between the light source and slit, the structure of SCDS and the parameters of camera in the smartphone were all optimized to ensure the best analytical performance. Besides, the SCDS employed HSV color model and utilized the overall intensity calculated by summing V-value of adjacent position for the absorbance measurement. In this way the errors caused by the low resolution of CD-grating can effectively be eliminated to promote the sensitivity of the SCDS. The performance of the SCDS was first validated for colorimetric detection of BSA with a detection limit of 0.0073 mg/mL, which is superior compared to that of the microtiter plate reader (MTPR). Moreover, by combining with 3,3',5,5'-tetramethylbenzidine-manganese dioxide (TMB-MnO2) nanosheets reaction, a high sensitive and specific system for ascorbic acid detection was established. The SCDS gives a detection range from 0.6250 mu M to 40 mu M with a detection limit of 0.4946 mu M for AA detection. Compared to other studies, the SCDS features wide detection range and very low detection limit with low cost instrument. Therefore, the SCDS will be an ideal and promising colorimetric system for point-of-care (POC) application in food security, disease diagnosis and environmental monitoring. (C) 2019 Elsevier B.V. Abstract:The development of oxidase mimic with favorable catalytic activity is of great significance for the rapid and convenient detection of ascorbic acid (AA). Herein, in this work, a synergistic regulation strategy of heterogeneous interface and hollow structure is proposed to effectively solve the problem of limited active sites obtained with Co3O4. The charge rearrangement at heterointerface could improve the catalytic activity of oxidase mimic. Moreover, the hollow nanocubes structure could increase specific surface area and provide rich active sites. As a result, the target material possesses enhanced oxidase-like catalytic activity and desirable stability compared to bare Co3O4 and MnO2, which also catalyze the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) to form a blue product (ox-TMB) even without hydrogen peroxide. Ox-TMB is reduced to TMB owing to the strong reducibility of AA, which represents a decrease in absorbance at 652 nm and a colorless solution. Moreover, an efficient biosensing method for the detection of AA is developed. The relative absorbance (∆A) of the sensing platform is linearly related to the concentration of AA in the range of 0–200 μM, and the detection limit is 0.11 μM. This work provides a novel method for the rapid, cost-effective and sensitive detection of AA, and possesses a wide range of potential application value.

Abstract:Highly sensitive and selective detection for hypochlorite anion (OCl-) is of extreme significance due to its important role and function in biological systems. Herein, a diaminomaleonitrile derivative (DMD) has been developed as a novel ratiometric and colorimetric fluorescent probe for OCl- detection. Consistent with the UV-vis spectral change, the probe DMD exhibited promptly a color change from canary yellow to gules upon the addition of OCl-, which could be easily observed by naked-eyes. In addition, the detection limit was found to be 0.5 nM, which is much lower than that of many reported OCl- probes. More importantly, owing to its excellent sensing properties and negligible cytotoxicity, probe DMD can be conveniently applied to image intracellular OCl- in living cells, indicating it a promising tool in biological applications. Abstract:A highly selective thiophene-thioimidazole hydrazine-based ratiometric chemodosimeter (TPBN) was designed and synthesized to detect hypochlorite (ClO–). The probe showed yellow fluorescence and exhibited ultra sensitivity towards hypochlorite (detection limit 8.74 nM) through the oxidative intramolecular cyclization process to give a blue fluorescent triazole product (TPBN-P). Additionally, the as-designed sensor displayed a fast response (80s) to hypochlorite with excellent selectivity over other competing analytes. DFT calculations, ESI-MS, and 1H NMR titration experiments supported the detection mechanism. The probe was a valuable and practical ratiometric sensor for test strips, commercial disinfectants, and water samples. The probe was successfully used in the bio-imaging of hypochlorite in human breast cancer cells due to its noteworthy photophysical characteristics and good cell permeability.

Abstract:A flexible chemosensor has been developed based on colorimetric and fluorescent dual modes using tetraphenylethylene-centered tetraaniline (TPE4A) for rapid and sensitive detection of hypochlorite anion. The fluorescent probe TPE4A exhibits a unique aggregation-induced emission (AIE) character which is proved by a blue shift of the fluorescent peak from 544 to 474 nm with the water equivalents increasing. With the addition of hypochlorite in solution, the absorbance of the probe changes and the responding fluorescence color can be observed to change from light green to purple. The detection limit of hypochlorite is 1.80 × 10−4 M in solution, and the visual detection limit is 1.27 μg/cm2 with the naked eye for the flexible paper-based chemosensor. The proposed flexible chemosensors show a good selectivity and sensitivity which has great potential for effective detection of hypochlorite anions without any spectroscopic instrumentation.

Abstract:By using Fe 7 S 8 nanosheets (NSs) as catalyst, O -phenylenediamine (OPD) as chromogenic substrate, H 2 O 2 as oxidant, and ascorbic acid (AA) as regulator, a dual ratio colorimetric and fluorescence sensing system for determining AA was constructed. The inherent peroxidase like activity of Fe 7 S 8 NSs enables it to catalyze the oxidation of OPD in the existence of H 2 O 2 , resulting in the formation of yellow-colored 2, 3-diaminophenazine (DAP) with a characteristic absorption peak at 450 nm and a fluorescence emission peak at 560 nm. However, the presence of AA could suppress the oxidation of OPD and induce the generation of quinoxaline derivative (QXD) 3-(dihydroxyethyl) furo [3,4-b] quinoxaline-1-one with absorption and fluorescence emission peaks located at 345 and 435 nm, respectively. Thus, employing the absorption of QXD at 345 nm and emission at 435 nm as the reporting signals, and the absorption of DAP at 450 nm and emission at 560 nm as the reference signals, a ratio colorimetric ( A 345 / A 450 ) and ratio fluorescence ( F 435 / F 560 ) dual signal detection method for AA was developed. Under the optimized conditions, the value of A 345 / A 450 was found to be linear with AA concentration in the ranges of 1.0–40 μM and 40–100 μM, and the value of F 435 / F 560 was strongly correlated with AA concentration in the ranges of 1.0–30 μM and 30–100 μM. When applied for the assay of AA in human serum samples, an acceptable recovery ranging from 93.1 to 106.6 % was obtained, confirming the potential of the sensing system in practical applications.

Abstract:Ratiometric fluorescent sensors, which can provide a built-in correction for environmental effects, have attracted significant attention for analytical sensing and optical imaging with the potential to provide a precise and quantitative analysis. Herein, we report a strategy based on dye-sensitized upconversion for the design of dual-excitation upconverion ratiometric probes possessing same emission peaks under a large separation in the excitation spectra (980 nm and 808 nm). Specifically, effective enhancement of upconversion luminescence could be attributed to Cy787 dyes present on the surface of nanoparticles, and it subsequently decreased upon the addition of ClO- under an 808 nm irradiation, whereas the signal under 980 nm excitation remained essentially constant, thus allowing for quantitative ratiometric monitoring of ClO-. The rationally designed dye-sensitized upconverion nanosystem exhibits excellent sensitivity for ClO- with a quantification limit of 3.6 nM in aqueous solutions. We have also demonstrated that the designed nanoprobe is a promising material for semi-quantitative detection of ClO- in an arthritis mouse model.
Colorimetric ascorbic acid sensing from a synergetic catalytic strategy based on 5,10,15,20-tetra(4-pyridyl)-21H,23H-porphyrin functionalized CuS nanohexahedrons with the enhanced peroxidase-like activity

Abstract:Artificial peroxidase mimics are indispensable for establishing the colorimetric sensing platform to detect rapidly H2O2 and related biomolecules. In this work, 5,10,15,20-tetra(4-pyridyl)-21H,23H-porphyrin (TPyP) functionalized CuS nanohexahedrons were prepared and was found to possess an excellent peroxidase-like activity. TPyP-CuS effectively catalyzed the oxidation of colorless 3,3´,5,5´-tetramethylbenzidine (TMB) by H2O2 into a blue oxTMB, distinguished visually only within 1 min. Fluorescent data reveal that the higher catalytic activity of TPyP-CuS is from lots of OH generated from H2O2. Based on the peroxidase-like activity of TPyP-CuS, a visual sensing platform was constructed to rapidly detect H2O2 in a wide linear range of 1.0–8.0 mM with a relatively lower limit of detection (LOD) of 121.8 μM as well as ascorbic acid (AA) in a linear range from 1 to 30 μM with a low detection limit of 0.419 μM.

Abstract:A free radical scavenging system based on FeO@Au/MOF-ABTS˙ has formed the basis of a novel method for the highly sensitive and specific spectrophotometric determination of ascorbic acid (AA). The FeO@Au/MOF nanozyme with magnetic separation properties was effectively prepared and evaluated using an environmentally friendly technique. Nanomaterials have the advantages of superparamagnetism, biocompatibility, chemical stability, and enhanced synergistic peroxidase-like activity, which can be utilized in catalysis to oxidise the peroxidase substrate 2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) (ABTS) into a green-colored product in the presence of HO. AA as an antioxidant has scavenging effects on ABTS radicals and can reduce green ABTS˙ to uncolored ABTS, contributing to a substantial reduction in green color. Based on such a premise, a highly selective and sensitive chromogenic sensing method depending on the peroxidase-like activity of the nanocomposites was developed in order to achieve the efficient detection of AA in real samples. Under optimum conditions, the proposed technique had a detection range of 0.001-0.1 mmol L, a limit of detection of 0.098 μmol L, and a detection time of only 30 seconds. The newly proposed colorimetric analysis method devoid of enzymes has broad application potential in the areas of quality control and quality and safety detection.

Abstract:Hypochlorite (ClO − ) mediates various physiological processes and diseases as a reactive oxygen species. The visualization of the concentration changes of endogenous hypochlorite in cells and in vivo faces great challenges due to the single-signal (spectrum) variation. In this study, a multifunctional ratiometric fluorescent probe IBA was designed and developed for analyzing hypochlorite and Cu 2+ independently through a synergetic FRET-PET mechanism. The sensor displayed excellent capacities in anti-interference capacity and sensitivity during the analysis processes of hypochlorite and Cu 2+ . In particular, IBA showed different fluorescence signal for hypochlorite and Cu 2+ meeting the need of independent analysis. And taking advantage of excellent optical properties of the probe in detecting hypochlorite, it successfully monitored the fluctuation of exogenous and endogenous hypochlorite in A549 cells. Moreover, IBA could visualize the changes of hypochlorite in living zebrafish with the fluorescent change between red and green. Overall, the designed novel probe IBA can serve as a potential tool to analyze hypochlorite and Cu 2+ , and the exploration of the variations of hypochlorite through living imaging shows potential to study the relevant molecular mechanisms and metabolism routes during multifarious physiological activities.

Abstract:Hypochlorous acid (HClO) as well as its ionic form (ClO−), representative of reactive oxygen species (ROS), are essential players in all sorts of biological processes. The abnormal level of each can lead to the onset of various diseases. Besides, Sodium hypochlorite, a commonly-used bleaching agent in our daily lives, could also result in breathing and skin problems when overexposed. Therefore, developing a molecular chemosensor for sensing HClO is of biological and environmental importance. Though many such chemosensors have been reported, new HClO chemosensors with different sensing performances may still come in handy in certain situations. In this work, we have developed a new coumarin-based chemosensor, CM-hbt, for realizing both ratiometric and colorimetric imaging detection of HClO in live cells. Notably, we further explored its application in sensing HClO in plant mung beans as well as fabricated an easy-to-use paper strip apparatus for facilitating its quick detection, which is seldomly seen in other HClO chemosensors. All the analysis results confirmed the high sensitivity and selectivity of this novel chemosensor. DFT calculations were used to decipher the underlying sensing mechanism of CM-hbt. Overall, this work presents a novel chemosensor, CM-hbt, as a colorimetric and ratiometric chemosensor for realizing imaging detection of HClO in a variety of different model systems, which highlights its broad spectrum of application potentials.

Abstract:A novel dual-responsive colorimetric and fluorescent chemosensor L based on diketopyrrolopyrrole derivative for Fe3+ detection was designed and synthesized. In presence of Fe3+, sensor L displayed strong colorimetric response as amaranth to rose pink and significant fluorescence enhancement and chromogenic change, which served as a naked-eye indicator by an obvious color change from purple to red. The binding constant for L-Fe3+ complex was found as 2.4×104 with the lower detection limit of 14.3nM. The sensing mechanism was investigated in detail by fluorescence measurements, IR and 1H NMR spectra. Sensor L for Fe3+ detection also exhibited high anti-interference performance, good reversibility, wide pH response range and instantaneous response time. Furthermore, the sensor L has been used to quantify Fe3+ ions in practical water samples with good recovery.

Abstract:In this study, using diethylenetriaminepentaacetic acid (DPTA), diethylenetriamine (DETA) and Fe (NO3)3.9H2O, Fe, N co-doped carbon dots (Fe, N-CDs) were successfully synthesized by a simple one-pot hydrothermal method. Fe, N-CDs possessed fluorescence and peroxidase-like properties. When H2O2 was present, o-phenylenediamine (OPD) can be oxidized by Fe, N-CDs to oxidized phenylenediamine (oxOPD), and oxOPD further quenched the fluorescence of Fe, N-CDs because of inner filter effect. There were two peaks at 450 nm and 560 nm for the system of Fe, N-CDs and oxOPD. Results showed that the concentration of H2O2 can not only affect the fluorescence intensity but also the color of the oxOPD. Considering that the xanthine oxidase can oxidize xanthine to produce H2O2, thus, a ratiometric fluorescent and colorimetric dual-signal sensor for H2O2 and xanthine was developed. This new method was demonstrated to have high sensitivity and excellent selectivity for determination of H2O2 and xanthine. The detection limits (LOD) for H2O2 and xanthine were 0.047 μM and 0.02 μM with a ratiometric fluorescence method, and were 0.05 μM and 0.023 μM with a colorimetric method, respectively. This dual-signal sensor was used successfully for xanthine detection in human serum and urine.

Abstract:A new water-soluble colorimetric and ratiometric fluorescent probe for detecting hypochlorite ion (ClO − ) based on a phenothiazzine group was designed and synthesized. As ratiometric fluorescent probe, LD - Lyso showed rapid, accuracy, and selective fluorescence sensing effect for ClO − in PBS buffer solution with a large Stokes shift (195 nm), it displayed a significant blue-shift phenomenon and a 25-fold enhancement of the fluorescence intensity ratio (I 550 /I 635 ) toward ClO − , fluorescence notably changing from red to green, the detection limit as low as 8.74 μM; As colorimetric probe, LD - Lyso also showed a significant shift in absorption band (from 450 to 395 nm) upon treatment with ClO − along with the visible color changed from faint yellow to colorless, the detection limit as low as 0.01 μM. In addition, the detecting mechanism of LD - Lyso towards ClO − via the intramolecular charge transfer (ICT) mechanism was proved by ESI-MS and DFT calculation, and the application as testing strips for detecting ClO − was realized.

Abstract:Zinc-substituted myoglobin (ZnMb) is a naturally occurring photosensitizer that generates singlet oxygen with a high quantum yield. Using a combination of photophysical and fluorescence imaging techniques, we demonstrate the interaction of ZnMb with Gram-positive Staphylococcus aureus and Gram-negative Escherichia coli. An efficient antibacterial action against S. aureus was observed, with a reduction up to 99.9999% in the number of colony-forming units, whereas no sizable effect was detected against E. coli. Because ZnMb is known to form during the maturation of additive-free not-cooked cured ham, the use of this protein as a built-in photodynamic agent may constitute a viable method for the decontamination of these food products from Gram-positive bacteria.

Abstract:Hypochlorite (ClO − ) is a well-known redox regulator among reactive oxygen species and plays a crucial role in bacterial disinfection and body immunity. Therefore, there is a high demand for an analytical technique to monitor ClO − under physiological conditions. Herein, a novel fluorescence probe, RES-DPPT , was developed for detecting ClO − . This probe employs diphenylphosphinothioyl unit as the recognition group and resorufin as the fluorescent platform. RES-DPPT has high sensitivity and specificity, rapid response, and wide pH tolerance for monitoring ClO − . Abstract:Hypochlorite (ClO- ), as a kind of essential reactive oxygen species, plays a crucial role in vitro and in vivo. Here, a ratiometric fluorescent probe (TPAM) was designed and constructed for sensing ClO- based on substituted triphenylamine and malononitrile, which exhibited obvious colour transfer from orange to colourless under daylight accompanied by noticeable fluorescence change from red to green in response to ClO- . TPAM could effectively monitor ClO- with the merits of fast response, excellent selectivity, high sensitivity and a low detection limit of 0.1014 μM. 1 H NMR, mass spectra and theoretical calculations proved that ClO- caused the oxidation of the carbon-carbon double bond in TPAM, resulting in compound 1 and marked changes in colour and fluorescence. In addition, TPAM was utilized for imaging ClO- in living cells successfully with good photostability and biocompatibility.

Abstract:The development of reliable method for detection of hypochlorite (ClO-) in vivo is of great importance to understand the precise roles of ClO- in a variety of pathological and physiological processes. Herein, three novel camphor-based fluorescence probes (2a-2c) for the ratiometric detection of ClO- were designed and synthesized, which exhibited both a distinct red shift of the fluorescence emission and a marked enhancement of the fluorescence intensity ratio in response to ClO-. The probes 2a-2c could selectively and sensitively detect ClO- with rapid response time, wide pH application range and low detection limit. The sensing mechanism was carefully confirmed by the ESI-MS, H-1 NMR and DFT studies. In addition, the cell imaging experiments demonstrated that the probes 2a-2c could be successfully applied to imaging ClO- in living cells.
