Abstract:Transition metal coordination complexes have provided cancer treatment with new insights to overcome the limitations of current chemotherapeutic agents. Utilization of bifunctional tetrazole–carboxylate ligands with Zn( II ) obtained two self-assembled complexes [Zn(HL 1 )(bipy) 3/2 (H 2 O)]·CH 3 OH·4(H 2 O) ( 1 ) (H 3 L 1 = 1,3,5-tri(2-carboxymethyltetrazol-5-yl) benzene) and [Zn(L 2 ) 2 (H 2 O) 2 ] 2 ·2H 2 O ( 2 ) (HL 2 = (5-pyridin-3-yl-tetrazol-2-yl)-acetic acid). The X-ray diffraction results showed that the two complexes displayed a two-dimensional (2D) layer structure and a one-dimensional (1D) layer structure. Nanocoprecipitation with DSPE-PEG-2000 resulted in the formation of complex nanoparticles (NPS) with excellent water dispersion. In vitro CCK-8 assay indicated the two NPs exert high cytotoxicity and sensitivity and a low half-maximum inhibitory concentration (IC 50 ) towards HeLa than HepG2 cells. In addition, the cytotoxicity was also confirmed by live/dead co-stained experiments. The presented experimental results showed the 1 and 2 NPs were capable of inhibiting cell proliferation in vitro and may help design coordination complex-based anticancer candidates for cancer cells.

Abstract:For the antitumor advantages, Zn(II) complexes have attracted more and more attention. Herein, three novel tetranuclear Zn(II) complexes 1-3 based on dihydrazone pyrimidine derivatives H2L1-H2L3 were synthesized and characterized using IR, 1H NMR, single crystal X-ray diffraction analysis, XRD, TG and elemental analysis. Single crystal X-ray diffraction analysis revealed that 1-3 all displayed [2x2] grid-like topology. The stability in solution, lipophilicity, confocal imaging and antitumor activities were investigated. Complexes 1-3 displayed high structural stability, membrane permeability and different lipophilicity. They can target mitochondria due to the cation charge. MTT assay indicated that they all exhibited stronger antiproliferative activity than the corresponding derivatives H2L1-H2L3 and the well-known cisplatin against all the selected tumor cells (BGC-823, BEL-7402, MCF-7 and A549), with IC50 values ranging from 2.83 μM to 7.97 μM. AO/EB double staining, flow cytometry and ROS detection suggested that complexes 1 and 2 could induce BGC-823 apoptosis in a dose-dependent manner. UV-Vis spectra, CD spectra, viscosity analysis and molecular docking revealed that complexes 1 and 2 interact with DNA mainly via partial intercalation and groove binding. Tetranuclear [2x2] grid-like Zn(II) complexes have the potential to be promising antitumor agents in the future.

Abstract:Non-platinum-metal complexes show great potential as anticancer agents. Herein, a series of dithiocarbazate non-Pt-metal complexes, including [FeIII(L)2]·Cl·2H2O 1, [CoIII(L)2]·NO3·2.5H2O 2, [NiII(L)2] 3, and [ZnII(L)2] 4, have been designed and evaluated for their efficacy as antineoplastic agents. Among them, complex 2 exhibited higher anticancer efficacy than complexes 1, 3, 4, and cisplatin against several cancer cell lines. Hemolysis assays revealed that complex 2 showed comparable hemolysis with cisplatin. In vivo anticancer evaluations showed that complex 2 could retard tumor xenograft growth effectively with low systemic toxicity. Further studies revealed that complex 2 suppressed cancer cells by triggering multiple mechanisms involving the simultaneous inhibition of mitochondria and glycolytic bioenergetics. Overall, our study provides new insights into the anticancer mechanism of Co complexes, which can be used as a good strategy to overcome the flexibility of cancer cells to chemotherapy adaptation.

Abstract:Many strategies have been used in treatment of cancer, but currently the chemotherapy is still the main force, unfortunately, general agents exhibit inevitable toxicity and inferior selectivity. Therefore, less toxic and high selectivity anticancer reagents have been extensively sought and developed. Here we report a series of Copper (II) complexes CuP1~CuP5 which have good biological activities. Due to the influence of the molecular size, CuP1 displayed the strongest bound with ct-DNA. All of the complexes show positive anticancer activities toward A549, H1975, HepG2 and T47D cancer cells. Additionally, flow cytometry experiments indicated that complexe CuP2 arrest cell cycles at S and G2 phase and mitochondrial membrane potential tests were also performed to explore the mechanism of action. Our findings disclosed that this type of complex represents a potent platform for development of metal anticancer drugs.

Abstract:Primary murine CD4+ and CD8+ T helper (Th) cells provide help for various immune responses by secreting lymphokines which activate effector cells. The purpose of the present study was to investigate the co-stimulatory signals that, together with T cell receptor (TCR) cross-linking, induce phenotypically distinct primary Th cells to secrete IL-2 and proliferate. We isolated highly purified populations of primary CD4+ or CD8+ T cells and stimulated them in vitro with platebound anti-CD3 mAb. TCR cross-linking by anti-CD3 mAb induced both IL-2 receptor expression and responsiveness to exogenous IL-2, but was not sufficient to induce either IL-2 secretion or T cell proliferation. Rather, for both CD4+ and CD8+ primary Th cells, IL-2 secretion and proliferation required both TCR cross-linking and antigen presenting cell (APC)-derived co-stimulatory signals. Based on G-10 adherence and sensitivity to gamma-irradiation, the APC populations able to induce primary CD4+ Th cells and primary CD8+ Th cells to secrete IL-2 were indistinguishable. In addition, we found that either IL-1 or IL-6 could replace the requirement for APC-derived co-stimulatory signals for IL-2 secretion and proliferation by both primary CD4+ Th cells and primary CD8+ Th cells. Thus, the present study has examined and compared the co-stimulatory requirements of rigorously purified subsets of IL-2-secreting primary CD4+ and primary CD8+ T cells.(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract:A series of solid-state coordination compounds of Mn(II), Fe(II), Ni(II), Cu(II) and Zn(II) with 5-((1-methyl-pyrrol-2-yl)methyl)-4-(2-chlorophenyl)-1,2,4-triazoline-3-thione as a ligand were synthesized and studied in terms of their chemical composition (elemental analysis, ICP), spectroscopic properties (FTIR) and magnetic measurements. Single crystal X-ray diffraction analysis (SC-XRD), HRMS and NMR studies were carried out to confirm the structure of the organic ligand. Its biological potential was evaluated in silico , using molecular docking studies and ADME analysis. The anticancer activity of all five metal complexes and the organic ligand were tested in vitro against A549 and HT-29 cancer cells. We found that the metal complexes in general exhibited better anticancer activity in comparison with the free ligand, with the Cu(II) complex exhibiting the highest activity against both tested cell lines.

Abstract:To inhibit the growth and metastasis of triple‐negative breast cancer (TNBC), two Fe(III) thiosemicarbazone complexes (Fe1 and Fe2) were designed and synthesized. The structures of the Fe(III) complexes were characterized by single crystal X‐ray diffraction. The antiproliferative activity of Fe1 and Fe2 against four cancer lines (MDA‐MB‐231, T98G, HepG2, 143B) and human renal proximal tubular epithelial cell line (HK‐2) was evaluated by MTT assay. Among all cells, Fe2 showed significant cytotoxicity to TNBC cells (MDA‐MB‐231), with an IC50 value of 12.38 μM. Furthermore, Fe2 showed less toxicity to HK‐2 cells. The two Fe(III) complexes can produce excess of reactive oxygen species, decrease of mitochondrial membrane potential, and induce DNA damage, then lead to apoptosis of MDA‐MB‐231 cells. In addition, Fe1 and Fe2 can also inhibit migration and invasion of MDA‐MB‐231 cells. This study provides guidance for the development of metal complexes that inhibit the growth and metastasis of TNBC.

Abstract:Copper complexes are considered as potential candidates for anticancer therapy and medical applications. In this paper, three new Cu(II) complexes, [Cu(IPY)2](ClO4)2·H2O (CuI1), [Cu(IPY)(L-Phe)H2O]ClO4·0.5H2O (CuI2) and [Cu(IPY)(L-Val)H2O]ClO4 (CuI3) (where IPY = 2-(1H-imidazol-2-yl)pyridine, L-Phe = L-phenylalanine, L-Val = L-valine), with good amphipathic properties were synthesized and characterized. The single crystal X-ray diffraction results revealed that CuI1 was four-coordinated, while CuI2 and CuI3 both adopted a five-coordinated tetragonal pyramidal configuration. Multi-spectral methods, viscosity experiment and molecular docking technique showed that three complexes interacted with DNA through insertion. The results of the gel electrophoresis experiments indicated that DNA was oxidatively cleaved by all the complexes in a concentration-dependent manner. Moreover, singlet oxygen (1O2), hydrogen peroxide (H2O2) and superoxide anion radicals (·O2−) were associated with oxidative cleavage of DNA. All the complexes also had good binding affinity with human serum albumin (HSA). The MB degradation assay revealed that all complexes could react with H2O2 to form ·OH through Fenton-like processes. The complexes displayed good antiproliferative activity against the tested human cancer cells in vitro, including cervical carcinoma cells (HeLa), liver cancer cells (HepG2 and BEL-7402) and gastric adenocarcinoma cells (SGC-7901), but showed lower toxicity to normal liver cells (LO2). The anticancer mechanism research revealed that CuI1, CuI2 and CuI3 arrested the cell cycle at S phase, elevated intracellular reactive oxygen species (ROS) levels and induced loss of mitochondrial membrane potential (MMP). The results indicated that these Cu(II) complexes could induce DNA damage and ROS-mediated mitochondrial dysfunction, leading to cancer cell apoptosis. Our work provided a theoretical basis for the design of new low-toxic and highly efficient anticancer Cu(II) complexes by incorporating biological metabolites and aromatic heterocycle ligands.

Abstract:The 3d transition metal (Mn(II), Fe(II), Co(II), Ni(II), Cu(II) and Zn(II)) complexes, supported by anions of sterically demanding β-diketones, 1,3-dimesitylpropane-1,3-dione (HLMes) and 1,3-bis(3,5-bis(trifluoromethyl)phenyl)-3-hydroxyprop-2-en-1-one (HLCF3), were synthesized and evaluated for their antitumor activity. To assess the biological effects of substituents on phenyl moieties, we also synthesized and investigated the analogous metal(II) complexes of the anion of the less bulky 1,3-diphenylpropane-1,3-dione (HLPh) ligand. The compounds [Cu(LCF3)2], [Cu(LMes)2] and ([Zn(LMes)2]) were characterized by X-ray crystallography. The [Cu(LCF3)2] crystallizes with an apical molecule of solvent (THF) and features a rare square pyramidal geometry at the Cu(II) center. The copper(II) and zinc(II) complexes of diketonate ligands, derived from the deprotonated 1,3-dimesitylpropane-1,3-dione (HLMes), adopt a square planar or a tetrahedral geometry at the metal, respectively. We evaluated the antitumor properties of the newly synthesized (Mn(II), Fe(II), Co(II), Ni(II), Cu(II) and Zn(II)) complexes against a series of human tumor cell lines derived from different solid tumors. Except for iron derivatives, cellular studies revealed noteworthy antitumor properties, even towards cancer cells endowed with poor sensitivity to the reference drug cisplatin.

Abstract:In these studies, we designed and investigated the potential anticancer activity of five iron(II) cyclopentadienyl complexes bearing different phosphine and phosphite ligands. All complexes were characterized with spectroscopic analysis viz. NMR, FT–IR, ESI–MS, UV–Vis, fluorescence, XRD (for four complexes) and elemental analyses. For biological studies, we used three types of cells—normal peripheral blood mononuclear (PBM) cells, leukemic HL-60 cells and non-small-cell lung cancer A549 cells. We evaluated cell viability and DNA damage after cell incubation with these complexes. We observed that all iron(II) complexes were more cytotoxic for HL-60 cells than for A549 cells. The complex CpFe(CO)(P(OPh) 3 )(η 1 - N -maleimidato) 3b was the most cytotoxic with IC 50 = 9.09 μM in HL-60 cells, IC 50 = 19.16 μM in A549 and IC 50 = 5.80 μM in PBM cells. The complex CpFe(CO)(P(Fu) 3 )(η 1 - N -maleimidato) 2b was cytotoxic only for both cancer cell lines, with IC 50 = 10.03 μM in HL-60 cells and IC 50 = 73.54 μM in A549 cells. We also found the genotoxic potential of the complex 2b in both types of cancer cells. However, the complex CpFe(CO) 2 (η 1 - N -maleimidato) 1 which we studied previously, was much more genotoxic than complex 2b , especially for A549 cells. The plasmid relaxation assay showed that iron(II) complexes do not induce strand breaks in fully paired ds-DNA. The DNA titration experiment showed no intercalation of complex 2b into DNA. Molecular docking revealed however that complexes CpFe(CO)(PPh 3 ) (η 1 - N -maleimidato) 2a , 2b , 3b and CpFe(CO)(P(O i Pr) 3 )(η 1 - N -maleimidato) 3c have the greatest potential to bind to mismatched DNA. Our studies demonstrated that the iron(II) complex 1 and 2b are the most interesting compounds in terms of selective cytotoxic action against cancer cells. However, the cellular mechanism of their anticancer activity requires further research.

A Tetranuclear copper(II) Complex with a Pyridine‐2,6‐dicarboxamide Ligand: Structural and Magnetic Properties and In vitro Antiproliferative Activity Against Human Cancer Cells

Abstract:A tetranuclear Cu(II) complex of a pentadentate ligand, containing pyridine2,6‐diamide motif, is reported. The X‐ray structure of the complex reveals that it has an open book type architecture. The magnetic property of the complex is explored by VT susceptibility measurements and DFT calculations. The complex shows strong antiproliferative activity against HCT116 cancer cells. Possible mechanism of anticancer activity is also discussed. A pentadentate diamide ligand, N,N′‐di(pyridine‐2‐yl)pyridine‐2,6‐dicarboxamide (L1H2) and its tetranuclear Cu(II) complex [Cu4(L1)2(μ3‐OH)2(H2O)3(ClO4)](ClO4) ⋅ H2O are reported. X‐ray crystal structure of the complex reveals that it has an open‐book type architecture. Two central copper atoms are connected to each other by two hydroxo bridges. Each of these two hydroxo bridges also connects the two central copper atoms to one of the two terminal copper atoms. Variable temperature susceptibility measurements show that χMT decreases with the temperature, indicating a strong antiferromagnetic behavior. With the help of DFT calculations the magnetic data was analyzed using the spin Hamiltonian H=−2 J1S3 ⋅ S4 −2 J2(S3 ⋅ S1+ S3 ⋅ S2+ S4 ⋅ S1+ S4 ⋅ S2)−2 J3S1 ⋅ S2 (where subscripts 1 and 2 refer to the terminal copper atoms and 3 and 4 refer to the central copper atoms). The best fit was obtained with 2 J1=−322 cm−1, 2 J2=+34.8 cm−1, 2 J3=−8.8 cm−1 and g=2.18, indicating a δ‐type interaction between terminal copper atoms. The complex proves to be an avid binder of ct‐DNA, with apparent binding constant (Kapp) value estimated as 1.536×107 M−1. In vitro experiments indicate that the Cu(II) complex reduced the proliferation of HCT116 cells among other cell lines. This reduction of cell proliferation may be attributed to the enhancement of intracellular ROS, along with modulation and disruption of cell cycle associated proteins. SKC conceptualized the project, arranged funding and involved in overall coordination among collaborators, analyzing the data, and writing the paper. AM was involved in doing the experimental work on synthesis of the compounds, carrying out analytical and spectroscopic measurements, collecting X‐ray crystallographic data and DNA binding experiments. MS, SM and KDS were involved in study of anticancer activities of the complex. SS helped in refining X‐ray structure of the complex. MC did the magnetic measurements and analysis whereas AF did the DFT calculations.

Abstract:Developing new drugs for the treatment of cancer is of great significance. Coordination compounds based on tetrazole-carboxylates are one of the most potential candidates for the treatment of cancer. Two metallic compounds coordinated and controlled by bifunctional tetrazolate-carboxylate H3L1 and L2 ligands [H3L1= N,N-di(2'- carboxymethyl terazole-5-yl) amine and L2 = 2-(5-pyridin-2-yl-tetrazol-2-yl)malonic acid diethyl ester] have been synthesized and characterized by single crystal X-ray diffraction analysis. The [Sm(L1)(H2O)3]·4H2O (1) displayed a 2-D network while [Sm(L2)3(OH)3] (2) with a mononuclear cluster structure. The effect of the main ligands in the two compounds on ablation of cancer cells were explored. Nanoparticles (NPs) of the two compounds can be prepared by PEG-2000 (Polyethylene glycol-2000) co-precipitation method. In vitro MTT assay on HeLa cells showed that 2 NPs with a lower IC50 (half maximal inhibitory concentration) of only 1.3 μM is lower than that of 1 NPs (1.6 μM), indicating that the cytotoxicity of compound 2 NPs is superior to that of compound 1 NPs. The results showed that [Sm(L1)(H2O)3]·4H2O (1) and [Sm(L2)3(OH)3] (2) NPs were capable of inhibiting cell proliferation in vitro and may be a potential candidate against cancer.

Abstract:A series of bi- or mononuclear hexacoordinate iron(III) complexes, [Fe(L)][Fe(bpb)(CN)2]·CH3OH·0.5H2O (1), [Fe(L)][Co(bpb)(CN)2]·CH3OH (2) [(Fe (L))2(4,4'-bipy)](BPh4)2 (3), [Fe(L)(py)](BPh4) (4) and [Fe(L)(dmap)](BPh4) (5) (bpb = 1,2-bis(pyridine-2-carboxamido)benzenate, L = N,N'-bis(2-hydroxybenzyliden)-1,7-diamino-4-azaheptane, dmap = 4-dimethylaminopyridine), have been prepared with the pentadentate Schiff base iron(III) compound as assemble precursor and characterized by element analysis, IR and X-ray diffraction. Single crystal structural determination revealed the neutral cyanide-bridged binuclear entity for complexes 1 and 2 and the cationic di- or mononuclear structure for complexes 3-5 with the positive charge(s) balanced by BPh4- ion(s). The experimental study and theoretical simulation of the magnetic property discovered the ferromagnetic coupling between the Fe(III) ions bridged by cyanide group in complex 1 and the always high spin state of the Fe(III) ion coordinated to the Schiff base ligand in both complexes 1 and 2. The temperature dependent magnetic susceptibility investigation over complexes 3-5 showed the occurrence of the thermo-induced gradual complete spin crossover (SCO) property at about 115, 170 and 200 K, respectively.
Synthesis, cytotoxicity and DNA binding of novel Ni(II), Co(II) and Zn(II) complexes bearing pyrimidinyl hydrazone ligand

Abstract:To develop efficient new antitumor agents, Ni(II), Co(II) and Zn(II) complexes 1 – 3 derived from 2-((2-pyrimidin-2-yl)hydrazono)methyl)quinoline (HL) were prepared and structurally characterized using IR, elemental analysis, UV-Vis, fluorescence spectrometry and single crystal X-ray diffraction analysis. The in vitro cytotoxicity of complexes 1 – 3 against MCF-7, BGC-823, A549, BEL-7402 tumor cell lines and human normal cell line HL-7702 was evaluated using MTT assay. Complexes 1 - 3 display higher cytotoxic activity than cisplatin as well as HL against BGC-823 tumor cell line, with IC 50 values ranging from 5.57 μM to 11.20 μM, and less toxicity toward the normal cell line HL-7702. In particular, complex 3 exhibits the most prominent antiproliferative activity. Complex 3 was found to induce apoptosis of BGC-823 cells greatly in a dose-dependent manner, which was suggested by Annexin V-FITC/PI and AO/EB dual-staining assays. The DNA binding studies, which were performed using UV-Vis, fluorescence and CD spectral analyzes, revealed that complex 3 interacts with CT-DNA mainly via a partial intercalative binding mode. The molecular docking of the interaction of complex 3 with DNA is in accordance with the experimental data.

Abstract:Four novel mononuclear complexes, [Cd(L)(2)center dot 2H(2)O] (1), [Ni(L)(2)center dot 2H(2)O] (2) [Cu(L)(2)center dot H2O] (3), and [Zn(L)(2)center dot 2H(2)O] (4) (CCDC number: 1444630-1444633 for complex 1 - 4) (HL=4-(2,3-dichlorophenyl)piperazine -1-carboxylic acid) were synthesized, and have been characterized by IR spectroscopy, elemental analysis, and X-ray crystallography. Molecular docking study preliminarily revealed that complex 1 had potential telomerase inhibitory activity. In accordance with the result of calculation, in vitro tests of the inhibitory activities of complexes 1 against telomerase showed complex 1 (IC50 = 8.17 +/- 0.91 mu M) had better inhibitory activities, while complexes 2, 3 and 4 showed no inhibitory activities. Antiproliferative activity in human cancer cell line HepG2 was further determined by MTT assays. The IC50 value (6.5 +/- 0.2 mu M) for the complex 1 having good inhibitory activity against HepG2 was at the same micromolar concentrations with cis-platinum (2.2 +/- 1.2 mu M). While the IC50 value for the metal-free ligand, complex 2, 3 and 4 was more than 100 mu M. These results indicated that telomerase was potentially an anticancer drug target and showed that complex 1 was a potent inhibitor of human telomerase as well as an antiproliferative compound. Synthesis, Characterization and Anticancer Activities of Transition Metal Complexes with a Nicotinohydrazone Ligand.

Abstract:The reaction of divalent transition metal salts and (E)-N'-(1-(pyridin-2-yl)ethylidene)nicotinohydrazide (penh) led to the formation of [Mn(penh)2] (complex 1), [Co(penh)2] (complex 2), [Cu(penh)2] (complex 3) and [Cd (penh)2] (complex 4) complexes. The four complexes were characterized using elemental analyses, infrared spectra and single-crystal X-ray diffraction analyses. Subsequently, the complexes were used for in vitro cell level experiments to determine potential antitumor effects. The results demonstrated that the complexes exhibited a similar structure; however, they were crystallized with distinct space groups. In comparison with the uncomplexed penh ligand, all four complexes were able to markedly decrease the proliferation rate of various types of tumor cell, including the human lung cancer cell line A549, human gastric cancer cell line BGC823 and human esophageal cancer cell line Eca109, in a concentration-dependent manner. Furthermore, the complexes promoted tumor cell apoptosis, as demonstrated in the apoptosis assay, and this was confirmed using electrophoresis.

Abstract:We synthesized an organic bimetallic complex (compound 2) with a cross configuration, which was prepared by a two-step reaction. The ferrocene Schiff base was obtained by the reaction of Ferrocenecarboxaldehyde and DL-alaninol, which formed the ferrocene-palladium(Ⅱ) complex through coordination bond. The crystal structure showed that 2 has a trans structure opposite to cisplatin and behaves as a racemate. In this study, compound 2 was found to suppress the viability of different tumor cell lines excellently with potency up to 20-fold better than cisplatin as well as possessing lower toxicity in normal cells. The mechanism of antiproliferation activities was through inducing caspase 3-dependent apoptosis in vitro. Molecular docking was conducted to predict the cellular target of compound 2 was XIAP, which suggested that 2 may bind to the BIR3 domain of XIAP protein by forming hydrogen bonds with TRY-323a.

Abstract:Two bis(tridentate) Schiff base ligands H2L(x) were used to construct three 2×2 grid-type tetranuclear Fe(II) complexes 1-3 to obtain polynuclear spin-crossover materials. Magnetic susceptibility measurements show that the spin states of the complexes are related to the substituents of H2L(x), and that spin transition occurs only in complexes 1 and 2, which are derived from a bulky ligand, whereas complex 3 is diamagnetic. The transition temperatures of complexes 1 and 2 are close to room temperature and are dependent on counteranions. The spin transition of complex 1 can be reversibly tuned by the dehydration and hydration process.
