Abstract:Background Kanamycin (KAN) residues in animal-derived foods continuously enter the human body, which will pose serious threats to human health such as hearing loss, nephrotoxicity and other complications. Therefore, to sensitively detect KAN residues by a reliable technology is extremely urgent in food quality and safety. Compared with traditional methods being limited by cost and complexity, photoelectrochemical (PEC) biosensors benefit from some merits such as rapid response, excellent sensitivity and good stability. In this study, the construction of a highly efficient PEC platform to realize KAN residues detection is discussed. Results Herein, a novel p-n heterojunction consisting of flower-like BiOI microspheres and graphite carbon nitride (g-C 3 N 4 ) nanoflakes was developed to establish a PEC aptasensor for KAN detection at 0 V. The prepared g-C 3 N 4 /BiOI heterostructure showed not only significantly enhanced PEC activity due to the larger specific surface area but also greatly increased charge separation efficiency owing to the strong internal electric field. Meanwhile, using g-C 3 N 4 /BiOI as a highly efficient photoactive material for binding amine-functionalized aptamers to capture KAN, the photocurrent signals showed a 'turn off' mode to achieve the sensitive detection of KAN. The proposed PEC aptasensor exhibited linear response for KAN from 5 × 10 -9 to 3 × 10 -7 mol L -1 with a low detection limit of 1.31 × 10 -9 mol L -1 , and satisfactory recoveries (97.44 -107.38 %) were obtained in real food samples analysis. Significance This work presented a novel p-n heterojunction-based PEC aptasensor with strong selectivity and stability, rendering it allowed to detect KAN in animal-derived foods including milk, honey and pork. Additionally, the detection range satisfied the MRLs for KAN specified by the national standards, demonstrating the potential application for food analysis. The study provides a new insight into the development of efficient and practical biosensors for antibiotic residues detection.

Abstract:An ultrasensitive photoelectrochemical (PEC) aptasensor was originally designed by using ZnIn2S4/ReS2 as a photoactive material and AgInS2 as a signal amplifier. The signal amplifier AgInS2 was incubated on the terminal of H-DNA (immobilized on the ZnIn2S4/ReS2/FTO surface), leading to an enhanced photocurrent response. Then, due to the introduction of DNA2, the formation of a double-stranded structure caused AgInS2 to keep away from the electrode surface, and the photocurrent was reduced. In the presence of kanamycin, DNA2 was released from the system due to the competition relationship, and a restored photocurrent response was obtained. The combination of ZnIn2S4/ReS2 and AgInS2 accelerated the electron transfer and enhanced the separation efficiency of photogenerated electron-hole pairs, resulting in an improved performance of the PEC aptasensor, which was capable of accurate and sensitive detection of kanamycin in actual samples.

Abstract:A"signal-off " photoelectrochemical aptasensor based on p-n type semiconductor competitive quenching effect and strand displacement reaction was constructed for the determination of kanamycin. Au 2S4-graphene composite was used as n-type photoactive semiconductor material. In the presence of the kanamycin, strand displacement reaction was triggered and the p-type CuInS2 quantum dots labeled aptamer was introduced on the Au surface. The CIS QDs can competitive consume the electron donors (AA) and light energy of the PEC system, thus quenched the anodic photocurrent of Au The photocurrent decreased with the increase of kanamycin concentration. The linear range of kanamycin was 1.0 pM-10 μM, and the detection limit was 1.7 pM. In addition, the method can be used for the determination of kanamycin in milk and honey.

Abstract:Herein, a new dual-model photoelectrochemical (PEC)/electrochemical (EC) sensor for the detection of kanamycin (Kana) was developed based on Z-scheme titanium dioxide (TiO2) disk/methylene blue (MB) sensibilization. Metal-organic frameworks-derived porous TiO2 disk were synthesized, and exhibited excellent anodic photocurrent under visible light excitation. Then, amino-labeled double-stranded DNA (dsDNA) was introduced onto the modified electrode. The photocurrent was enhanced with MB embedded in dsDNA to form a Z-scheme TiO2/MB sensibilization. When the target Kana was present, it specifically bound to the aptamer in the dsDNA, leading to the disruption of dsDNA structure and the release of MB. This release of MB and the increase of target spatial resistance, resulted in a significant weakening of the PEC signal and a decreased oxidation peak current of MB. The PEC sensor successfully detected Kana in the range of 2~1000 pM with a LOD of 0.17 pM. Meanwhile, The EC sensor for Kana detection showed a linear range of 5 pM∼500 pM with a LOD of 1.8 pM. Additionally, the sensor exhibited excellent selectivity, reproducibility, stability, and good recoveries when applied to milk and honey samples. As a result, this method has the potential to be applied in ensuring food safety through the rapid determination of antibiotics in food.

Ratiometric Dual Signal-Enhancing-Based Electrochemical Biosensor for Ultrasensitive Kanamycin Detection

Abstract:Based on the signal amplification elements of planar VS2/AuNPs nanocomposites and CoFe2O4 nanozyme, we herein developed an electrochemical biosensor for sensitive kanamycin (Kana) quantification. A ratiometric sensing platform was presented by incorporating VS2/AuNPs nanocomposites as a support material with excellent conductivity and high specific surface area, as well as hairpin DNA (hDNA) with complementary hybridization of biotinylated Kana-aptamer. In addition, streptavidin-functionalized CoFe2O4 nanozyme with superior peroxidase-like catalytic activity were immobilized onto the aptasensor, hence the peroxidase-like catalytic reaction could yield amplified electrochemical signals. With the presence of Kana, the aptamer-biorecognition resulted in a quantitative decrease of nanozyme accumulation and an increase of methylene blue response. Under optimal conditions, the electrochemical signal ratio of the aptasensor revealed a linear relation along with the logarithmic concentration of Kana from 1 pM to 1 μM, with the limit of detection reaching to 0.5 pM. A multiple signal amplification photoelectrochemical biosensor based on biotin-avidin system for kanamycin sensing in fish and milk via synergism of g-C3N4 and Ru@SiO2

Abstract:Background: The residues of kanamycin can accumulate in the human body for a long time and pose serious health risks, including hearing loss, kidney poisoning, and drug allergic reactions. Therefore, it is crucial to develop a rapid, highly sensitive, and low-cost method for detecting kanamycin residues in foods. However, the current methods have limitations such as low sensitivity, expensive instruments, and multiple steps, which make them impractical for use in resource-limited environments and emergencies. In this study, the creation of a multiple-signal amplification photoelectrochemical biosensor to address these aforementioned issues is discussed. Results: Herein, we proposed a multiple signal amplification photoelectrochemical (PEC) biosensor based on carboxylated g-C3N4 and avidin functionalized Ru@SiO2 for the ultrasensitive detection of kanamycin. The carboxylated g-C3N4 was a highly efficient photoactive substance for amplifying photoelectric signals and a substrate for aptamer immobilization. The DOS and PDOS of g-C3N4 were studied by simulation, and the sensing mechanism of the probe at the molecular level was revealed. Meanwhile, using Ru@SiO2 as a signal amplifying unit, through the cooperative work between Ru@SiO2 and g-C3N4, the photoelectric signal could be double amplified to produce an excellent photocurrent response. Under optimized conditions, the photocurrent response of the PEC biosensor to kanamycin was obtained at concentrations from 0.1 nM to 1000 nM with a lower detection limit of 4.1052 × 10-11 mol L-1. This protocol demonstrates high sensitivity, brilliant specific recognition ability, excellent reproducibility, and acceptable stability. Significance: The first combination of g-C3N4 and avidin-Ru@SiO2 as photocurrent materials greatly enhanced the sensitivity of the PEC biosensors. Moreover, the specificity and sensitivity of the PEC biosensor were further improved through the specific interaction between kanamycin and aptamer. The photoelectric conversion mechanism based on g-C3N4 and two pathways for enhancing the photocurrent by Ru(byp)32+ were proposed. Through simulations of the DOS and PDOS of g-C3N4, the sensing mechanism of the probe at the molecular level was revealed. Under the optimum conditions, the PEC biosensor exhibited a wide linear concentration range and a low detection limit.

Abstract:Using staggered-gap ZnO/BiFeO3 heterojunction as photoactive materials and cDNA-CdS as the sensitizer for sensitive Kanamycin (KAN) detection, we have created a unique signal-off biosensing platform. The ZnO/BiFeO3 heterojunction provides active sites for aptamer loading and enhances photocurrent responsiveness. Rapid interfacial charge transfer and the separation efficiency of photo-generated carriers are enhanced by sensitization of the ternary heterojunction ZnO/BiFeO3/CdS. Signal-amplified quenching occurs when sensitizers are replaced with sterically hindered KAN. Because of the aptamer's greater affinity for KAN, the replacement of CdS causes a decrease in photocurrent response. Additionally, the weakly conductive aptamer-KAN complex causes steric hindrance, which exacerbates the photoelectrochemical signal-damping effect even more. The photoelectrochemical aptasensor exhibits excellent selectivity and stability, detecting KAN within the range of 0.00005825-0.233 μg/mL with a detection limit of 0.0466 ng/mL (S/N = 3). This work demonstrates the potential of perovskite oxides and their heterostructures for advanced photoelectrochemical biosensing applications.
Dual-mode colorimetric and photothermal aptasensor for detection of kanamycin using flocculent platinum nanoparticles

Abstract:Chitosan (CS)-stabilized platinum nanoparticles (CS/PtNPs) were employed to develop a novel aptamer-based dual-mode colorimetric and photothermal biosensor for selective detection of kanamycin (KAN). As a peroxidase-like catalyst, the CS/PtNPs showed outstanding catalytic activity for the oxidation of 3,3′,5,5′-tetramethylbenzidine (TMB) in the presence of hydrogen peroxide (H 2 O 2 ). As a stabilizing agent, CS excelled at fixing the KAN binding aptamer on the surface of the CS/PtNPs, amplifying their catalytic activity and enhancing colloidal dispersion and stability. The oxidized TMB (TMB ox ) functioned as a signal for the colorimetric, photothermal aptasensor because of its observable absorbance of light in the visible and near-infrared (NIR) regions. When light from a NIR laser was absorbed by the TMB ox in the reaction solution, heat was generated in inverse proportion to the KAN concentration. The developed colorimetric and photothermal modes of the aptasensor showed a linear detection range of 0.1–50 and 0.5–50 μM, with a limit of detection (LOD) of 0.04 and 0.41 μM, respectively. Moreover, the aptasensor successfully determined KAN concentrations in spiked milk samples, verifying the reliability and reproducibility in practical applications. The dual-mode aptasensor based on CS/PtNPs for KAN detection, utilizing both color change and heat generation signals through a single probe (TMB ox ), demonstrates rapid response, simplicity in operation, cost-effectiveness, and high sensitivity. In addition, unlike typical immunoassays, this aptamer-based peroxidase-like nanozyme activation and inhibition strategy required no washing process, which was very effective in terms of reducing the time required for an assay and sustaining a high sensitivity.

Abstract:A straightforward label-free colorimetric aptasensor utilizing the aptamer-enhanced peroxidase-like activity of cobalt oxyhydroxide (CoOOH) nanosheets has been established for kanamycin detection. In the kanamycin-free state, aptamers adsorb onto the CoOOH surface through electrostatic forces, enhancing the peroxidase-like activity of CoOOH and thereby resulting in a strong absorption signal and a yellow hue in 3,3′,5,5′-tetramethylbenzidine (TMB) upon termination of the reaction with a stop solution. Conversely, upon the introduction of kanamycin, aptamers and CoOOH nanosheets compete for binding to kanamycin, resulting in a significant decrease in the number of aptamers bound to CoOOH. As a result, the activity of CoOOH diminishes, leading to a corresponding reduction in coloration and absorbance of the solution. Hence, the quantitative determination of kanamycin could be realized by analyzing the absorbance variations. Under optimal conditions, the aptasensor demonstrated high sensitivity and specificity, with a linear detection range from 500 nM to 5 μM and a detection limit as low as 54.6 nM. Moreover, the aptasensor effectively identified kanamycin in river water samples, achieving a recovery rate between 91.7% and 102.1%. This approach offers good practicability and provides a novel platform for kanamycin detection in environmental samples.

Abstract:The Ti3C2Tx/MoS2/MWCNT@rGONR nanocomposite was firstly prepared for building a sensitive electrochemical aptasening platform to simultaneous detect kanamycin (Kana) and chloramphenicol (Cap). Owning to the accordion-like structure, rich surface groups, and high charge mobility, the Ti3C2Tx/MoS2/MWCNT@rGONR provided a spacious covalent immobilization surface and a better electrochemical aptasensing platform. The aptamers of Kana and Cap used in sensor enhancing the selectivity. Furthermore, TiP loading more different metal ion were used as label to form a sandwich-type sensor together with Ti3C2Tx/MoS2/MWCNT@rGONR, which further improving the electrochemical sensitivity and obtaining high distinguishable signal readout. Under the optimized conditions, the sensor has good detection limits of Kana and Cap respectively at 0.135 nmol/L and 0.173 nmol/L in a same linearity concentration of 0.5~2500 nmol/L. Finally, it was successfully applied for detection in milk and fish meat and the results were compared with standard method HPLC, indicating a great potential for food safety monitor.

Abstract:We present a novel experimental method for quantitatively characterizing the temperature-dependent mechanical behavior of individual nanostructures during uniaxial straining. By combining a microelectromechanical tensile testing device with a low thermal mass and digital image correlation providing nm-level displacement resolution, we show successful incorporation of a testing platform in a vacuum cryostat system with an integrated heater and temperature control. Characterization of the local sample temperature and time-dependent response at both low and high temperature demonstrates a testing range of ∼90-475 K and steady-state drift rates less than 0.04 K/min. In situ operation of the tensile testing device employing resistively heated thermal actuators while imaging with an optical microscope enables high-resolution displacement measurements, from which stress-strain behavior of the nanoscale specimens is deduced. We demonstrate the efficacy of our approach in measuring the temperature dependence of tensile strength in nominally defect-free ⟨110⟩ Pd nanowhiskers. We uncover a pronounced sensitivity of the plastic response to testing temperature over a range of ∼300 K, with an ultimate strength in excess of 6 GPa at low temperature. The results are discussed in the context of thermally activated deformation mechanisms and defect nucleation in defect-free metallic nanostructures.
Multiply-amplified strategy for the ultrasensitive detection of kanamycin via aptamer-triggered three-dimensional G-quadruplex/Ni–Fe layered double oxide frame networks

Abstract:In this work, a multiply-amplified peroxidase-like colorimetric strategy was proposed for the high-specific recognition and ultrasensitive detection of kanamycin (Kana). Based on two Kana-aptamer triggered sequential reactions, G-quadruplex (G4) and DNA (hairpins) modified Ni–Fe layered double oxides (LDOs) could be obtained simultaneously. Later, a three-dimensional G4/LDO frame networks, as a novel DNAzyme, with enhanced peroxidase-like catalytic activity was assembled through electrostatic interaction. This DNAzyme catalyzed 3,3′,5,5′-tetramethylbenzidine oxidation for the colorimetric detection of Kana. The enhancement principle was discussed and the charge transfer process during the catalytic reaction was investigated. Under the optimal experiment conditions, the proposed method exhibited high sensitivity, where the linear range is from 10 fM to 10 nM (r2 = 0.992), and the limit of detection is 3 fM (S/N = 3). The practicability of this assay was demonstrated by successfully application of residual Kana detection in genuine milk and urine samples.

Abstract:Flexible sensing devices have drawn tremendous attention in the past decades due to their potential applications in future hand-held, potable consumer, and wearable electronics. Here, we firstly developed an ultrasensitive wireless potentiometric aptasensor based on flexible freestanding graphene paper for kanamycin detection. Flexible graphene paper made from a simple vacuum filtration method was used as a biocompatible platform for effective immobilization of aptamer. A nuclease-assisted amplification strategy was introduced into this potentiometric biosensing system in order to significantly improve the detection sensitivity through a classic catalytic recycling reaction of target induced by the nuclease (DNase I). As expected, an ultra-low detection limit of 30.0 fg/mL for kanamycin was achieved. Furthermore, the developed potentiometric enzymatic aptasensor exhibits high selectivity, favorable flexibility, excellent stability and reproducibility, which holds great promising for its routine sensing application.
Mesoporous DNA-Co@C Nanofibers Knitted Aptasensors Performing Onsite Determination of Trace Kanamycin Residues

Abstract:The abuse of kanamycin (KAN) poses an increasing threat to human health by contaminating agricultural and animal husbandry products, drinking water, and more. Therefore, the sensitive detection of trace KAN residues in real samples is crucial for monitoring agricultural pollution, ensuring food safety, and diagnosing diseases. However, traditional assay techniques for KAN rely on bulky instruments and complicated operations with unsatisfactory detection limits. Herein, we developed a novel label-free aptasensor to achieve ultrasensitive detection of KAN by constructing mesoporous DNA-cobalt@carbon nanofibers (DNA-Co@C-NFs) as the recognizer. Leveraging the extended π-conjugation structure, prominent surface area, and abundant pores, the Co@C-NFs can effectively load aptamer strands via π-π stacking interactions, serving as KAN capturer and reporter. Due to the change in DNA configuration upon binding KAN, this aptasensor presented an ultralow detection limit and ultra-wide linear range, along with favorable precision and selectivity. Using real tap water, milk, and human serum samples, the aptasensor accurately reported trace KAN levels. As a result, this convenient and rapid autosensing technique holds promise for onsite testing of other antibiotic residues in agriculture, food safety, and clinical diagnosis.
A universal optical aptasensor for antibiotics determination based on a new high-efficiency Förster resonance energy transfer pair

Abstract:A novel "turn-on" aptasensor for kanamycin (Kana) detection based on a new Förster resonance energy transfer (FRET) pair is reported. A new organic small molecule was employed as a high-efficiency quencher for fluorophore. Based on specific interactions between ssDNA and the quencher, an ingenious and amplified strategy was designed. In the absence of the target, the fluorescence of the fluorophore labeled at the end of the aptamer was quenched. After the binding of the aptamer to the target, the fluorescence was recovered and amplified. The proposed aptasensor showed high specificity, selectivity, and stability in complicated systems. With the P3-based strategy, the limit of detection for Kana is estimated to be 10 nM, which is much lower than the maximum allowable concentration in milk. The recoveries of spiked Kana in milk were in the range 99.8 ~ 105.3% (n = 3). Fortunately, this novel method can be easily extended to other antibiotics such as tobramycin by simply replacing the aptamer, showing great potential as a universal platform for selective, sensitive, and rapid detection of hazardous analytes in food samples.

Abstract:Simple assay format-based SERS methods for sensitive target substance analysis is of great significance for the development of on-site monitoring biosensors. Herein, taking the typical antibacterial kanamycin (KANA) as a subject, a simple, highly sensitive and specific SERS aptasensor was developed by manipulating DNA hydrogel network to fish plasmonic core-shell nanoparticles. A competitive binding mode of aptamer, ligation-rolling circle amplification (L-RCA), gap-containing nanoparticles (GCNPs) with embedded Raman reporters were integrated into the sensor. In the presence of KANA, the double stranded DNA (dsDNA) structure of the aptamer was disrupted, and the released primers were used to construct two kinds of circularized padlock probes (CPPs) which were partially complementary. DNA hydrogel network was formed through the intertwining and self-assembly of two RCA-generated single stranded DNA (ssDNA) chains, during which GCNPs and magnetic beads (MBs) were entangled and incorporated. Finally, KANA quantification was successfully achieved through the quantification of the DNA hydrogel. Overall, this novel SERS aptasensor realized a simple and ultrasensitive quantification of KANA down to 2.3 fM, plus excellent selectivity, and precision even for real food samples. In view of innovative fusion across L-RCA-based DNA hydrogel and SERS technique, the proposed method has promising potential for application in on-site detection and quantification of trace food contaminants.

Abstract:In this work, a novel 3D-DNA walker signal amplification strategy was designed to construct a fluorescent aptasensor for the detection of kanamycin (KAN). The aptasensor utilizes split aptamers for the synergistic recognition of KAN. The presence of KAN induces the split aptamers recombination to form the Mg 2+ -DNAzyme structure, which is activated by Mg 2+ to drive the 3D-DNA walker process for cascading signal amplification. Employing gold nanoflowers (AuNFs) as walking substrate material increases the local DNA concentration to enhance the walker efficiency. The prepared fluorescent aptasensor achieved efficient and sensitive detection of KAN with satisfactory results in the concentration range of 1 × 10 −8 - 1 × 10 −3 μg/kg and the detection limit of 5.63 fg/kg. Meanwhile, the designed fluorescent aptasensor exhibited favorable specificity, anti-interference, storage stability and reproducibility, and verified the feasibility of its application in milk samples. The present work provides an effective tool for the regulation of KAN contamination in animal-derived foods with promising prospects.

Abstract:The American Heart Association estimates that 81% of people who die of coronary heart disease are 65 years old or older. The leading risk health behaviors include physical inactivity, poor diet, smoking, and binge drinking. Using the 2011–2012 California Health Interview Survey (CHIS), this study looked at how self-management, which includes a plan developed by a medical professional and the confidence to manage one’s disease, may decrease negative risk behaviors in older adults. The presence of a plan and increased self-efficacy decreased engagement in negative dietary behaviors and low physical activity. Implications for strategies that address heart disease and self-management are discussed.

Abstract:Photoelectrochemical (PEC) biosensors have exhibited a promising potential for assays of a large variety of analytes; however, how to realize their low background-based "super on" signal output is still a great challenge. Herein, we report a novel multiple nucleic acid amplification-assisted "on-off-super on" signal transition mechanism for the PEC biosensing of kanamycin antibiotics. The biosensing platform was constructed on a perylene-3,4,9,10-tetracarboxylic dianhydride-based photoelectrode, and its strong photocurrent could be well inhibited by an anchored ferrocene (Fc)-labeled hairpin DNA to produce a low background signal. Two target biorecognition-triggered exonuclease III-catalytic reactions were adopted to produce an annular four-footed DNA walker (AFW) and a methylene blue (MB)-labeled DNA strand. By using their synergistic effect to release Fc quenchers and simultaneously capture MB sensitizers, a "super on" signal output was realized. As a result, a very wide linear range from 10 fg mL −1 to 10 ng mL −1 and an ultra-low detection limit of 7.8 fg mL −1 were obtained. Meanwhile, the aptamer recognition-based homogeneous reaction and AFW-based multiple nucleic acid amplification effectively simplified the assay manipulation and well ensured the repeatability of the method. The satisfactory sample experiment results indicated its good reliability and accuracy for the antibiotic residue analysis application.
