Abstract:Objective: The present study was conducted to investigate the effects of lycopene on growth performance, abdominal fat deposition, serum lipids levels, activities of hepatic lipid metabolism related enzymes and genes expression in broiler chickens.Methods: A total of 256 healthy one-day-old male Arbor Acres broiler chicks were randomly divided into four groups with eight replicates of eight birds each. Birds were fed basal diet supplemented with 0 (control), 100, 200, and 400 mg/kg lycopene, respectively.Results: Dietary 100 mg/kg lycopene increased the body weight at 21 day of age compared to the control group (p<0.05). Compared to the basal diet, broilers fed diet with 100 mg/kg lycopene had decreased abdominal fat weight, and broilers fed diet with 100 and 200 mg/kg lycopene had decreased abdominal fat percentage (p<0.05). Compared to control, diets with 100, 200, and 400 mg/kg lycopene reduced the levels of total triglyceride and total cholesterol in serum, and diets with 100 and 200 mg/kg lycopene reduced the level of serum low density lipoprotein cholesterol (p<0.05). The activity of fatty acid synthase (FAS) in 400 mg/kg lycopene treated broilers and the activity of acetyl-CoA carboxylase (ACC) in 100, 200, and 400 mg/kg lycopene treated broilers were lower than those fed basal diet (p<0.05). Lycopene increased the mRNA abundance of adenosine monophosphate activated protein kinase α (AMPK-α), whereas decreased the mRNA abundance of sterol regulatory elementbinding protein 1, FAS, and ACC compared to the control group (p<0.05).Conclusion: Dietary lycopene supplementation can alleviate abdominal fat deposition and decrease serum lipids levels, possibly through activating the AMPK signaling pathway, thereby regulating lipid metabolism such as lipogenesis. Therefore, lycopene or lycopenerich plant materials might be added to poultry feed to regulate lipid metabolism.

Abstract:The objective of this study was to determine the effect of dietary lycopene supplementation on the growth performance, antioxidant enzyme activity of serum and liver, and gene expressions associated with Kelch-like ech-associated protein-1 ( Keap1 )/Nuclear Factor E2-related factor 2 ( Nrf2 ) pathway in liver of Arbor Acres broilers. A total of 288 1-day-old male broilers were randomly divided into 4 treatments with 6 replicates and 12 chickens for each replicate. The control group was fed with the basal diet, while the treated groups were fed with the basal diet with 10, 20, and 30 mg/kg lycopene in powder. Feed and water were provided ad libitum for 42 days. Compared with the control group, (a) the average daily gain increased ( p = 0.002 vs. p = 0.001) and the feed conversion ratio decreased ( p = 0.017 vs. p = 0.023) in groups treated with lycopene in the grower and whole phases, and the average daily feed intake was quadratically affected ( p = 0.043) by lycopene in the grower phase; (b) the serum superoxide dismutase content was linearly affected ( p = 0.035) by lycopene at 21 days; (c) the serum glutathione peroxidase content, superoxide dismutase content, and total antioxidant capability were higher ( p = 0.014, p = 0.003, and p = 0.016, respectively) in the 30 mg/kg lycopene group at 42 days; (d) the liver glutathione peroxidase and superoxide dismutase contents in groups treated with lycopene were higher ( p ≤ 0.001 vs. p ≤ 0.001) at 21 days; (e) the liver glutathione peroxidase content was higher ( p ≤ 0.001) in the 20 and 30 mg/kg lycopene groups, at 42 days; (f) the mRNA expression levels of Nrf2, superoxide dismutase 2, NAD(P)H quinone dehydrogenase 1 , and heme oxygenase 1 genes were higher (21 days: p = 0.042, p = 0.021, p = 0.035, and p = 0.043, respectively; 42 days: p = 0.038, p = 0.025, p = 0.034, and p = 0.043, respectively) in the 20 and 30 mg/kg lycopene groups at 21 and 42 days. The 30 mg/kg lycopene concentration improved the growth performance, antioxidant enzyme activity in serum and liver, and gene expression in the Keap1-Nrf2 signaling pathway of Arbor Acres broilers.

Effects of Broccoli Stem and Leaf Meal on Broiler Performance, Skin Pigmentation, Antioxidant Function, and Meat Quality

Abstract:Three hundred sixty 1-d-old Ross 308 male broilers were used to study the effects of broccoli stem and leaf meal (BSLM) on growth performance, skin pigmentation, antioxidant function, and meat quality. The chicks were fed 4 diets containing different levels (0, 4.0, 8.0, and 12.0%) of BSLM as partial replacement for corn and soybean meal for a period of 42 d. The results showed that dietary supplementation of BSLM had no effect (P > 0.05) on growth performance. As compared with control, dietary 4%, 8%, and 12% BSLM increased (P < 0.05) b value (yellowness) both in shank and breast skin, increased (P < 0.05) the concentrations of xanthophylls in abdominal fat and breast skin, improved (P < 0.05) total antioxidant capability, lowered malondialdehyde concentration, and decreased drip loss percentage of breast muscle. Dietary 8% and 12% BSLM decreased (P < 0.05) shank L values (lightness), increased (P < 0.05) shank a value (redness), and increased (P < 0.05) the activities of superoxide dismutase and catalase of breast muscle as compared with control. The results indicated that dietary supplementation of BSLM in broiler chickens improved the poultry products quality with the more skin pigmentation and the less drip loss percentage of breast meat. The more skin pigmentation mainly related to the high amount of xanthophylls in BSLM. The decreased meat drip loss fed BSLM may be caused by the antioxidative function of BSLM.
Effects of Probiotics and Cello-Oligosaccharide on Nutrient Digestion, mRNA Expression of Intestinal Mucosa Amino Acid Transporters and Nitrogen Emission of Broilers under Heat Stress

Abstract:This experiment was conducted to study the effects of probiotics( PRO) and cello-oligosaccharide( COS) on nutrient digestion,mRNA expression of intestinal mucosa amino acid transporters and nitrogen emission of broilers under heat stress. Nine hundred 21-day-old Ross 308 broilers w ere randomly allocated to 5groups: thermoneutralzone( HN) group,heat stress( HS) group,probiotics( HS+0.15%PRO) group,cellooligosaccharide( HS+0.15%COS) group and synbiotics( HS +SYN,SYN = 0.15%PRO +0.15%COS) group.Birds w ere either raised in a thermoneutral zone( 22 ℃) in HN group,and the others w ere subjected to cyclic heat stress by exposing to 33 ℃ from 08: 00 to 18: 00 and 22 ℃ from 18: 00 to next day 08: 00. Raising experiment period w as 21 days. The results show ed as follow s: 1) compared w ith birds kept in HN group,average daily gain and average daily feed intake of broilers subjected to heat stress w ere significantly reduced 10.65%( P0.05) and 8.04%( P0.05),respectively. Average daily gain,average daily feed intake and feed to gain ratio of broilers subjected to heat stress w ith diet added probiotics or cello-oligosaccharide individually w ere not significantly different( P0.05). Apparent digestibility of crude protein and crude fiber in HS group w as significantly decreased( P0.05),nitrogen intake and nitrogen retention w ere significantly decreased( P0.05),nitrogen excretion and nitrogen excretion rate w ere significantly increased( P0.05),ammonia nitrogen,uric acid contents in excreta and ammonia concentration in chicken house w ere significantly increased( P0.05),the activity of urease in excreta w as significantly increased( P0.05),and mRNA expression of the basic amino acid transporter( rBAT),y+L amino acid transporter-2( y+LAT2),cationic amino acid transporter-1( CAT1) and cationic amino acid transporter-4( CAT4) in intestinal mucosa was significantly down-regulated( P0.05). 2) Compared with HS group,apparent digestibility of crude protein was significantly increased in HS+PRO and HS+COS groups( P0.05),nitrogen excretion and nitrogen excretion rate in HS+PRO and HS+COS groups w ere significantly reduced( P0.05),w hile nitrogen retention w as significantly increased( P0.05). Compared w ith HS group,excreta ammonia nitrogen content in HS+PRO group w as significantly low-ered( P0.05),and ammonia concentration in chicken house and activity of urease in excreta of HS +COS group w ere significantly reduced( P0.05). 3) As compared w ith HS group,apparent digestibility of crude protein and crude fiber w ere significantly increased( P0.05) in HS+SYN group. Nitrogen excretion and nitrogen excretion rate in HS+SYN group w ere significantly reduced( P0.05),w hile nitrogen retention w as significantly increased( P0.05). Ammonia nitrogen concentration in chicken house,excreta ammonia nitrogen content and urease activity of HS+SYN group w ere significantly reduced( P0.05). and mRNA expression of rBAT in intestinal mucosa in HS+SYN group w as significantly up-regulated( P0.05). The results indicate that heat stress decrease nutrient digestion and increase nitrogen emission of broilers,and dietary addition of probiotics and cello-oligosaccharide are effective in partially ameliorating these changes. The combination of probiotics and cello-oligosaccharide is superior to individually addition.

Abstract:Stable gene expression in mammalian cells is a prerequisite for many in vitro and in vivo experiments. However, either the integration of plasmids into mammalian genomes or the use of retro-/lentiviral systems have intrinsic limitations. The use of transposable elements, e.g. the Sleeping Beauty system (SB), circumvents most of these drawbacks (integration sites, size limitations) and allows the quick generation of stable cell lines. The integration process of SB is catalyzed by a transposase and the handling of this gene transfer system is easy, fast and safe. Here, we report our improvements made to the existing SB vector system and present two new vector types for robust constitutive or inducible expression of any gene of interest. Both types are available in 16 variants with different selection marker (puromycin, hygromycin, blasticidin, neomycin) and fluorescent protein expression (GFP, RFP, BFP) to fit most experimental requirements. With this system it is possible to generate cell lines from stable transfected cells quickly and reliably in a medium-throughput setting (three to five days). Cell lines robustly express any gene-of-interest, either constitutively or tightly regulated by doxycycline. This allows many laboratory experiments to speed up generation of data in a rapid and robust manner.

Abstract:There is no information regarding the influence of heat stress (HS) on host metabolic profile. In this study, we investigated the effects of different environmental temperatures on oxidative status, hormone levels, HS indicators, and plasma metabolites in broilers. A total of 1,680 yellow-feather broilers (28 D old) were randomly allotted to 4 groups with 6 replicates. The broilers (29-57 D old) were maintained in thermostatic rooms (20 degrees C, 25 degrees C, 28 degrees C, and 30 degrees C) for 28 consecutive days. The results showed that the plasma cortisol and adrenocorticotropic hormone levels and creatine kinase and lactate dehydrogenase activities gradually increased when the temperature increased from 20 degrees C to 30 degrees C. However, the insulin-like growth factor-I level decreased gradually. Furthermore, heat shock protein 70 expression significantly increased in the liver and breast muscle (P < 0.01). As the temperature increased, the total anti-oxidant capacity in the plasma and liver gradually decreased, whereas the malondialdehyde level increased. The activity of plasma glutathione peroxidase and total superoxide dismutase in the liver showed a similar increasing trend (P < 0.01). In addition, 15 metabolites were identified at higher (P < 0.05) levels, whereas 2 metabolites were identified at lower (P < 0.05) levels in the 30 degrees C treatment group than those in the 25 degrees C treatment group. Most of these potentially diagnostic biomarkers are involved in carbohydrate, amino acid, lipid, or gut microbiome-derived metabolism, indicating that HS affected the metabolic pathways in broilers. Six candidate metabolites (tartronic acid, l-bethreine, tartaric acid, allose, glut aric acid, and neohesperidin) were selected as biomarkers, as they showed high sensitivity, specificity, and accuracy in diagnosing broilers under HS (P < 0.01). In conclusion, in the final stage of growth, we identified 6 plasma differential metabolites as potential biomarkers of HS-induced metabolic disorders in yellow-feathered broilers. This work offers new insights into the metabolic alterations of broilers exposed to HS and provides a new perspective for further study.
Effects of Compound Organic Acid Calcium on Growth Performance, Hepatic Antioxidation and Intestinal Barrier of Male Broilers under Heat Stress

Abstract:Objective: The aim of this study was to evaluate the effects of compound organic acid calcium (COAC) on growth performance, hepatic antioxidant status and intestinal barrier of male broilers under high ambient temperature (32.7°C).Methods: Nine hundred healthy one-d-old Cobb-500 male broiler chicks were randomly assigned into three groups with six replicates of 50 birds each. A basal diet supplemented with 0% (control), 0.4% and 0.8% COAC, respectively were fed to birds for 6 weeks. All treatments were under high ambient indoor temperature of 32.7°C, and had a constant calcium and available phosphorus ratio.Results: The results showed that, compared with control, the average daily gain of broilers in 0.4% and 0.8% was significantly increased and the ratio of feed to gain in in 0.4% and 0.8% was significantly decreased at 1 to 21, 22 to 42 and 1 to 42 days of age (p<0.05). Compared with control, 0.8% COAC slightly decreased (p = 0.093) the content of malondialdehyde in liver at 42 days of age while 0.4% COAC significantly decreased (p<0.05) the activity of alkaline phosphatase. Furthermore, 0.4% COAC significantly enhanced the intestinal barrier function via increasing jejunal and ileal ocln transcription, promoting jejunal mucin 2 transcription at 42 days of age (p<0.05), and decreasing jejunal toll-like receptor 2 (TLR-2) and ileal TLR-15, inducible nitric oxide synthase compared with control group (p<0.05). Whereas, no significant differences on the transcription of interleukin-1β in jejunum and ileum were observed among three treatments (p>0.05). Overall, heat stress caused by high natural environment temperature may induce the damage to hepatic antioxidation and intestinal barrier.Conclusion: Dietary inclusion of COAC can improve the tolerance of broilers to thermal environment through the modification of antioxidative parameters in liver and the mRNA expression of genes in intestinal barrier, resulting in an optimal inclusion level of 0.4%.
Dietary supplementation with synbiotics improves growth performance, antioxidant status, immune function, and intestinal barrier function in broilers subjected to cyclic heat stress

Abstract:This study investigated the effects of synbiotics supplementation on growth performance, antioxidant status, immune function, and intestinal barrier function in broilers subjected to cyclic heat stress. One hundred and forty-four 22-day-old male broilers were randomly assigned to one of three treatment groups of six replicates each for a 21-day study, with eight birds per replicate. Broilers in the control group were reared at a thermoneutral temperature and received a basal diet. Broilers in the other two heat-stressed groups were fed a basal diet supplemented without (heat-stressed group) and with 1.5 g/kg synbiotic (synbiotic group). One and a half gram of the synbiotic consisted with 3 × 109 colony forming units (CFU) Clostridium butyricum, 1.5 × 109 CFU Bacillus licheniformis, 4.5 × 1010 CFU Bacillus subtilis, 600 mg yeast cell wall, and 150 mg xylooligosaccharide. Compared with the control group, heat stress increased rectal temperatures at 28, 35, and 42 days of age, respectively (P < 0.05). Birds subjected to heat stress had reduced weight gain, feed intake, and feed efficiency during 22 to 42 days (P < 0.05). In contrast, supplementation with the synbiotic decreased rectal temperature at 42 days of age and elevated weight gain of heat stress-challenged broilers (P < 0.05). Heat-stressed broilers exhibited a lower superoxide dismutase (SOD) activity in jejunal mucosa and a higher malondialdehyde accumulation in serum, liver and jejunal mucosa (P < 0.05), and the regressive SOD activity was normalized to control level when supplementing synbiotic (P < 0.05). Heat stress increased interleukin-1β (IL-1β) and interferon-γ (IFN-γ) levels in serum and IL-1β content in jejunal mucosa of broilers (P < 0.05). Synbiotic reduced IL-1β level in serum of broilers subjected to heat stress (P < 0.05). Compared with the control group, elevated serum diamine oxidase activity and reduced jejunal villus height were observed in broilers of the heat-stressed group (P < 0.05), and the values of these two parameters in the synbiotic group were intermediate (P > 0.05). Heat stress upregulated mRNA abundance of IL-1β and IFN-γ and downregulated gene expression levels of occluding and zonula occluden-1 (ZO-1) in jejunal mucosa of broilers (P < 0.05). The alterations in the mRNA expression levels of jejunal IL-1β and ZO-1 were reversed by the synbiotic (P > 0.05). In conclusion, dietary synbiotics could improve growth performance, antioxidant capacity, immune function, and intestinal barrier function in heat-stressed broilers.
Effects of Compound Polysaccharides Derived from Astragalus and Glycyrrhiza on Growth Performance, Meat Quality and Antioxidant Function of Broilers Based on Serum Metabolomics and Cecal Microbiota

Abstract:This study aimed to evaluate the effects of dietary supplementation of compound polysaccharides derived from Astragalus and Glycyrrhiza on growth performance, meat quality, antioxidant function, cecal microbiota and serum metabolomics of broilers. A total of 480 one-day-old male Arbor Acres (AA) broilers were randomly divided into four treatments with six replicates comprising 20 broilers each. Treatments: CON group was the basal diet; ANT group was supplemented with Terramycin calcium; LAG group was supplemented with 150 mg/kg Astragalus polysaccharides and 75 mg/kg Glycyrrhiza polysaccharides; HAG group was supplemented with 300 mg/kg Astragalus polysaccharides and 150 mg/kg Glycyrrhiza polysaccharides. The results showed that LAG and HAG supplementation increased growth performance, antioxidant function and meat quality compared with the CON group and ANT group and, especially, the effect of LAG treatment was better than HAG. Analysis of cecal microbiota showed that LAG and HAG supplementation altered cecal microbial diversity and composition in broilers. Serum metabolomics analysis showed that a total of 193 differential metabolites were identified in CON and LAG groups, which were mainly enriched in linoleic acid metabolism and glutathione metabolism pathways. Moreover, there was a close correlation between serum metabolites, cecal microbiota and phenotypic indicators. Conclusion: Dietary supplementation of 150 mg/kg Astragalus polysaccharides and 75 mg/kg Glycyrrhiza polysaccharides could improve the growth performance, antioxidant function and meat quality of broilers by changing the serum metabolites and cecal microbiota composition.

Effects of dietary L-citrulline supplementation on nitric oxide synthesis, immune responses and mitochondrial energetics of broilers during heat stress

Abstract:L-Citrulline is a non-protein amino acid that promotes arginine recycling and muscle protein synthesis. This study investigated whether L-citrulline can exert these functions in heat-stressed chickens. Arbor acre broilers were fed either basal diets (Control) or basal diets supplemented with 1% L-citrulline (L-Cit). At 28 d old, broilers were subjected to two environmental temperatures, 35 °C for 8 h/d (HS) or 24 °C for 24 h/d (TNZ) for 2 weeks. The experiment was designed as a 2 by 2 factorial arrangement. Results showed that HS increased the core body temperature (CBT) and rectal temperature of broilers compared to the TNZ condition. The CBT and mean CBT decreased by ∼0.5oC in TNZ + L-Cit broilers compared to the TNZ + Control group (P < 0.05). L-Cit supplementation at TNZ significantly (P < 0.05) lowered the plasma malondialdehyde content but this was increased during HS. Exposure to HS significantly (P < 0.05) elevated plasma malondialdehyde compared to TNZ condition. Plasma immunoglobulin A, G, and M were increased (P < 0.05) by TNZ + L-Cit compared to the TNZ + Control group. Plasma nitric oxide and endothelial nitric oxide synthase (NOS) activity was increased during HS, whereas the total and inducible NOS enzymes were decreased. In addition, L-Cit supplementation increased both the inducible and endothelial NOS isoforms (P < 0.05). The mRNA expression of peroxisome proliferator-activated receptor γ coactivator 1-α, avian uncoupling protein, cytochrome C oxidase subunit 3, and ATP synthase F1 subunit beta (ATP5β) in the breast muscle were downregulated (P < 0.05) during HS. However, L-Cit supplementation upregulated the mitochondrial transcription factor A and during HS, L-Cit increased ATP5β expression similar to TNZ housed broilers. Therefore, this study demonstrates that dietary L-Cit can lower the body temperature, decrease lipid peroxidation and promote the immune status of broilers under thermoneutrality. Also, L-Cit would act to promote muscle ATP generation during heat stress in broilers.
Lycopene: a natural antioxidant for prevention of heat-induced oxidative stress in poultry

Abstract:Heat stress is one of the most important physiological factors challenging poultry production throughout the world especially in tropical and sub-tropical countries. Oxidative stress induced by heat stress not only compromises productivity and performance but also results in morbidity and mortality losses leading to the economic burden for poultry producers. It reduces the shelf life of poultry product in addition to poor meat and egg quality. Recently, a trend towards using phytochemicals derived from natural sources with potential antioxidant activities has increased. Lycopene is a predominant carotenoid pigment which is universally found in fruits and vegetables, with tomatoes and their products being key sources. Several in vivo and in vitro studies have suggested that lycopene is a powerful antioxidant compared to other carotenoids. Supplementation of tomato (Solanum lycopersicum) derived lycopene revealed numerous health promoting activities in poultry birds, such as anti-oxidant, anti-inflammatory, immunomodulation, improved performance and better meat as well as egg quality. Lycopene maintains oxidative balance in birds through various ways including serving as a free radical scavenger, inhibiting signalling pathways and activating host antioxidant enzymes, such as superoxide dismutase (SOD), glutathione peroxidase (GSH-Px) and catalase (CAT). The aims of this current review are to summarise mechanisms of action through which lycopene quench reactive species and maintain oxidative balance, highlight the potential role of lycopene as a natural antioxidant agent for poultry industry and examine the benefits to poultry producers when using this natural antioxidant on commercial basis in poultry diets to alleviate the adverse effects of heat stress.

Abstract:This experiment aimed to investigate the effect of Lonicerae flos and Turmeric extracts (LTE) added to diets on growth performance and intestinal health of broilers. A total of 720 healthy 21-day-old yellow-feathered broilers were randomly divided into 3 treatment groups, with 6 replicates and 40 broilers per replicate. These 3 dietary treatments included a basal diet + 0 g/t LTE (CON), a basal diet + 300 g/t LTE (LTE300), and a basal diet + 500 g/t LTE (LTE500). The results showed that dietary supplementation of LTE linearly increased (P < 0.05) average daily gain (d 21-38) and average daily feed intake (d 21-60). At d 60, LTE300 had the highest serum total antioxidant capacity and total superoxide dismutase (P < 0.05), and LTE500 had the lowest malondialdehyde level (P < 0.05) among the three groups. Moreover, compared to CON, LTE300 significantly (P < 0.05) reduced endotoxin (d 38 and d 60) and diamine oxidase activity (d 38); LTE500 significantly (P < 0.05) reduced endotoxin (d 38 and d 60) and diamine oxidase levels (d 60) in the serum. LTE groups significantly (P < 0.05) increased ileal the ratio of villus height to crypt depth and serum immunoglobulin G. Furthermore, dietary supplementation of LTE also improved the intestinal epithelial barrier by the up-regulated mRNA expression of Claudin-1, Occludin and zonula occludens-1, and decreased the mRNA expression of interleukin-2, interleukin-8, tumor necrosis factor-α, nuclear factor κB, myeloid differentiation factor 88 and toll-like receptor 4. Compared to CON, 16S rRNA sequencing analysis showed that LTE300 had a better effect on the microbial diversity and composition in the ileum, and Bacillus and Lactobacillus_agilis were significantly enriched in LTE300. PICRUSt results showed that LTE300 was significantly (P < 0.05) enriched in four pathway pathways at KEGG level 2. In conclusion, dietary supplementation with LTE improved growth performance and intestinal health by enhancing antioxidant capacity, intestinal barrier and immune function, and regulating intestinal flora of yellow-feathered broilers.
Effect of chronic heat stress on the carbonylation of glycolytic enzymes in breast muscle and its correlation with the growth performance of broilers

Abstract:Chronic heat stress has detrimental effects on the growth performance of broilers, and the potential mechanism is under exploration. In this study, the protein carbonyl modification was introduced to glycolytic enzymes to evaluate its relationship with the growth performance of heat-stressed (HS) broilers. A total of 144 male 28-day-old broilers were assigned to 3 treatments: the normal control group (NC, raised at 22°C with free access to feed and water), the HS group (raised at 32°C with free access to feed and water), and the pair-fed group (PF, raised at 22°C with an amount of feed equal to that consumed by the HS group on a previous day). Results showed that heat stress decreased the average daily growth, increased the feed-to-gain ratio (F/G), decreased breast muscle rate, and increased abdominal fat rate compared with the NC and PF groups (P < 0.05). Higher cloacal temperature and serum creatine kinase activity were found in the HS group than those of the NC and PF groups (P < 0.05). Heat stress increased the contents of carbonyl, advanced glycation end-products, malonaldehyde, and the activities of catalase, glutathione peroxidase, and total antioxidant capacity compared with the NC and PF groups (P < 0.05). Heat stress increased the contents of glucose and lactate, declined the glycogen content, and lowered the relative protein expressions of pyruvate kinase muscle type, lactate dehydrogenase A type (LDHA), and citrate synthase compared to those of the NC group (P < 0.05). In contrast to the NC and PF groups, heat stress intensified the carbonylation levels of phosphoglucomutase 1, triosephosphate isomerase 1, β-enolase, and LDHA, which were positively correlated with the F/G (P < 0.05). These findings demonstrate that heat stress depresses growth performance on account of oxidative stress and glycolysis disorders. It further increases the carbonylation of glycolytic enzymes, which potentially correlates with the F/G by disturbing the mode of energy supply of broilers.

Abstract:This study was done to evaluate the effects of heat stress (HS) on production performance, redox status, small intestinal barrier-related parameters, cecal microbiota, and metabolome of indigenous broilers. A total of forty female indigenous broilers (56-day-old) were randomly and equally divided into normal treatment group (NT group, 21.3 ± 1.2°C, 24 h/day) and HS group (32.5 ± 1.4°C, 8 h/day) with five replicates of each for 4 weeks feeding trial. The results showed that the body weight gain (BWG) of broilers in HS group was lower than those in NT group during 3–4 weeks and 1–4 weeks ( p < 0.05). The HS exposure increased the abdominal fat rate ( p < 0.05) but decreased the thigh muscle rate ( p < 0.01). Besides, broilers in HS group had higher drip loss of breast muscle than NT group ( p < 0.01). Broilers exposed to HS had lower total antioxidant capacity (T-AOC) in serum and jejunum, activities of total superoxide dismutase (T-SOD) in the jejunum, glutathione peroxidase (GSH-Px) in the thigh muscle, duodenum, and jejunum; and catalase (CAT) in breast muscle, duodenum, and jejunum ( p < 0.05). Whereas the malondialdehyde (MDA) contents in breast muscle, duodenum, and jejunum was elevated by HS exposure ( p < 0.05). Moreover, the relative mRNA expression of Occludin and ZO-1 in the duodenum, Occludin, Claudin-1, Claudin-4, ZO-1, Mucin-2 in the jejunum, and the Claudin-4 and Mucin-2 in the ileum was down-regulated by HS exposure ( p < 0.05). The 16S rRNA sequencing results showed that the HS group increased the relative abundance of Anaerovorax in the cecum at the genus level ( p < 0.05). Cecal metabolomics analysis indicated 19 differential metabolites between the two groups ( p < 0.10, VIP >1). The Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis revealed that the differential metabolites mainly enriched in 10 signaling pathways such as the Citrate cycle (TCA cycle) ( p < 0.01). In summary, chronic HS exposure caused a decline of production performance, reduced antioxidant capacity, disrupted intestinal barrier function, and negatively affected cecal microbiota and metabolome in indigenous broilers.

Abstract:The current study aimed to investigate the effect of increasing supplementation of dietary glycine (Gly) on growth performance, meat quality, liver characteristics, and intestinal health in broiler chickens raised under heat stress (HS) conditions. A total of one thousand six hundred 25-d-old broiler chickens were randomly allotted to 1 of 5 dietary treatments with 8 replicates. Each replicate comprised 20 male and 20 female birds. A negative control (NC) diet was prepared to meet or exceed energy and nutrient requirement estimates, whereas a positive control (PC) diet was formulated to contain increasing concentrations of AMEn by 50 kcal/kg as well as those of digestible amino acids, total Ca, and available P by 10% compared with the respective concentrations in the NC diet. Three additional diets were prepared by supplementing the NC diet with 0.4, 0.8, or 1.6% Gly. All chickens were raised under cyclic HS conditions at 29°C ± 0.89°C for 10 h/d and 23°C ± 1.45°C for the remaining time over an 18-d feeding trial. Results indicated that broiler chickens fed the NC diet had a greater (P < 0.05) FCR than those fed the PC diet under HS conditions. Increasing supplementation of up to 1.6% Gly in diets decreased (linear, P < 0.001) FCR in broiler chickens. Increasing supplementation of dietary Gly tended to increase (linear, P = 0.070) water holding capacity in the breast meat. Increasing supplementation of dietary Gly decreased (linear, P < 0.05) serum aspartate aminotransferase concentrations and tended to decrease blood heterophil:lymphocyte (linear, P = 0.083) and liver malondialdehyde concentrations (quadratic, P = 0.084). A tendency for increased villus height (linear, P = 0.086) and a significant increase in villus height:crypt depth ratio and goblet cell numbers (linear, P < 0.05) were identified following increasing Gly supplementation. In conclusion, increasing supplementation of dietary Gly improved feed efficiency, meat quality, liver health, and intestinal morphology possibly by mitigating oxidative stress and stress response in broiler chickens raised under HS conditions.
Hepatic lipid metabolomics in response to heat stress in local broiler chickens breed (Huaixiang chickens)

Abstract:High-temperature environment-induced heat stress (HS) is a hazard environmental element for animals, leading to dramatic changes in physiological and metabolic function. However, the metabolomic-level mechanisms underlying lipid metabolism in liver of slow-growing broilers are still obscure. The present study investigated the effects of HS on hepatic lipidomics in Chinese indigenous slow-growing broilers (Huaixiang chickens). The study includes two treatments, each treatment had 5 replicates with 4 broilers per cage, where a total of 40 eight-week-old female Huaixiang chickens (average initial body weight of 840.75 ± 20.79 g) were randomly divided into normal temperature (NT) and HS groups for 4 weeks, and the broilers of NT and HS groups were exposed to 21.3 ± 1.2℃ and 32.5 ± 1.4℃ respectively. The relative humidity of the two groups was maintained at 55%-70%. The liquid chromatography-mass spectrometry (LC-MS)-based metabolomics were conducted to evaluate the changes in hepatic lipidomics of broilers. The results showed that there were 12 differential metabolites between the two treatments. Compared with the NT group, HS group reduced the levers of hepatic phosphatidylcholine (PC) (16:0/16:0), PC (16:0/18:2), triglyceride (TG) (16:0/16:1/18:1), TG (18:0/18:1/20:4) (VIP > 1 and p < 0.05), while increased PC (18:1/20:3), PC (18:0/18:1), PC (18:1/18:1), PC (18:0/22:5), dimethyl-phosphatidyl ethanolamine (dMePE) (14:0/18:3), dMePE (18:0/18:1) and dMePE (16:0/20:3) levels (Variable Importance in the Projection; VIP > 1 and p < 0.05). In addition, according to the analysis of metabolic pathway, the pathways of linoleic acid, alpha-linolenic acid, glycerolipid and glycerophospholipid metabolism were involved in the effects of HS on hepatic lipid metabolism of broilers (p < 0.05). In conclusion, HS altered the hepatic lipid metabolism mainly through linoleic acid, alpha-linolenic acid, glycerolipid and glycerophospholipid metabolism pathway in indigenous broilers. These findings provided novel insights into the role of HS on hepatic lipidomics in Chinese indigenous broiler chickens.

Abstract:Heat stress is the most common environmental stressor in poultry production, negatively affecting growth performance, meat quality, and welfare. Therefore, the aim of this study was to compare the nutritional effects of dietary supplementation with selenomethionine, Bacillus subtilis (BS), and a combination of selenomethionine and BS on broilers challenged with heat stress. A total of 300 21-day-old male broilers (Ross 308) were randomly assigned to 5 groups with 6 replicates of 10 broilers per each: control group (CON, broilers raised at 22 ± 2 °C), heat stress exposure group (HS, broilers raised at 32 ± 2 °C for 8 h/d), HSS group (HS group supplemented with 0.3 mg/kg selenomethionine), HSB group (HS group supplemented with 1 × 109 cfu/kg BS), and HSBS group (HS group supplemented with 0.3 mg/kg selenomethionine and × 109 cfu/kg BS). The experiment lasted for 21 d. The results indicated that, compared to the CON group, heat stress reduces (P < 0.05) broiler growth performance and damages the meat quality in breast and thigh muscles. Dietary supplementation with selenomethionine and BS did not improve the growth performance of broilers under heat stress. However, compared to the HS group, the HSS, HSB, and HSBS groups showed significantly increased (P < 0.05) pH45 min, redness (a*) and yellowness (b*), muscle fiber density, intramuscular fat, triglyceride content, and expression levels of Myf5, CAPN 2, FM, SLC27A1, A-FABP, H-FABP, APOB-100, and ACC in breast and thigh muscles. Meanwhile, these groups showed reduced (P < 0.05) lightness (L*), drip loss, shear force, muscle fiber cross-sectional area, and FM gene expression level. The HSBS group showed greater improvement in the physicochemical quality of muscle and volatile substances compared to the HSS and HSB groups. In conclusion, selenomethionine and BS improved meat quality and flavor in broilers under heat stress by modulating muscle fiber composition and characteristics, as well as increasing intramuscular fat deposition.

Abstract:Heat stress adversely affects both the productivity and well-being of chickens. Probiotics offer beneficial impacts on the health and growth performance of broilers. The current study investigates the influence of administering of Bacillus (including B. subtilis , B. licheniformis , B. coagulans , and B. indicus ) and Lactobacillus (consisting of L. acidophilus , L. plantarum , L. buchneri , and L. rhamnosus ) probiotics via drinking water, either singular or combined, on various aspects including growth performance, oxidative stress markers, carcass characteristics, fecal microbial composition, intestinal structure, and intestinal pH in broilers exposed to chronic heat stress. A total of 150 one-day-old broiler chicks were divided into 5 groups: (1) NC, negative control; (2) HS, birds exposed to chronic heat stress; (3) HSpBacil, exposed to chronic heat stress and received Bacillus probiotic; (4) HSpLAB, subjected to chronic heat stress and provided with Lactobacillus probiotic; (5) HSpMix, subjected to chronic heat stress and administered a combined probiotic from Bacillus and Lactobacillus . The HS group exhibited significantly reduced levels of growth performance, carcass traits, and notably affected oxidative stress indices, as well as intestinal pH and histomorphology in the birds. Additionally, the administered probiotics led to increased weight of lymphoid organs, enhanced body weight gain, and improved intestinal histomorphology. Furthermore, the probiotics decreased malondialdehyde and increased total antioxidant capacity in broilers. In conclusion, Bacillus and Lactobacillus probiotics, as single or multi-species, particularly Lactobacillus and combined probiotic, demonstrated potential in alleviating the adverse effects of heat stress in broiler chickens. They could serve as beneficial feed additives and growth enhancers.
