Abstract:This study assessed trace metal pollution in the Essaouira protected coastal area (Atlantic coast of Morocco) using the mussel Mytilus galloprovincialis as a bio-indicator and evaluated potential human health risks from mussel consumption. Samples were collected seasonally at 8:00 am from April 2017 to February 2018, and metal concentrations in mussel tissues were analyzed using ICP-AES. The metals were found in descending order: Fe > Zn > Al > Cu > Pb > Cr > Cd > Ag, with significant spatial and seasonal variations. Cluster analysis indicated different sources for the metals, with Fe, Zn, and Al linked to continental inputs and Cd, Cr, Pb, Cu, and Ag to anthropogenic sources. The pollution gradient, assessed using the Individual Multimetal Bioaccumulation Index (IMBI), showed that the area is less polluted than other Moroccan coasts. The Human health risk assessment revealed no significant risks, confirming the safety of mussel consumption from this region.

Abstract:This study reports, for the first time, the baseline levels for fifteen trace metals in M. galloprovincialis tissue from around the Mediterranean, Marmara and Black Seas. The environmental quality in the surrounding seawater was assessed i.e., a mussel farm was investigated by using CF and DC indices, and the water quality was qualified as good for the aquacultural activities. A strong Cu-regulation in the transplanted mussels was observed and it ranged between 3.20 and 3.60 μg/g d.w. The highest bioavailability and bioconcentration of the particulate Fe fraction could present a health risk to consumers with a low risk level (1 < THQ < 9.9). Cr is considered the limiting metal for mussel consumption (< 2 kg/day). The metal contamination gradient was assessed using TEPI and TESVI indices that identified seven reference stations on the large scale and revealed that Cd is the most investigated metal in the literature databases, and found that Pb was the most bioavailable contaminant in the areas examined.

Identification of proteomic signatures of exposure to marine pollutants in mussels (Mytilus edulis)

Abstract:Bivalves and especially mussels are very good indicators of marine and estuarine pollution, and so they have been widely used in biomonitoring programs all around the world. However, traditional single parameter biomarkers face the problem of high sensitivity to biotic and abiotic factors. In our study, digestive gland peroxisome-enriched fractions of Mytilus edulis (L., 1758) were analyzed by DIGE and MS. We identified several proteomic signatures associated with the exposure to several marine pollutants (diallyl phthalate, PBDE-47, and bisphenol-A). Animals collected from North Atlantic Sea were exposed to the contaminants independently under controlled laboratory conditions. One hundred and eleven spots showed a significant increase or decrease in protein abundance in the two-dimensional electrophoresis maps from the groups exposed to pollutants. We obtained a unique protein expression signature of exposure to each of those chemical compounds. Moreover a set of proteins composed a proteomic signature in common to the three independent exposures. It is remarkable that the principal component analysis of these spots showed a discernible separation between groups, and so did the hierarchical clustering into four classes. The 14 proteins identified by MS participate in alpha- and beta-oxidation pathways, xenobiotic and amino acid metabolism, cell signaling, oxyradical metabolism, peroxisomal assembly, respiration, and the cytoskeleton. Our results suggest that proteomic signatures could become a valuable tool to monitor the presence of pollutants in field experiments where a mixture of pollutants is often present. Further studies on the identified proteins could provide crucial information to understand possible mechanisms of toxicity of single xenobiotics or mixtures of them in marine ecosystems.
Heavy metals in sediments, mussels and oysters from Trinidad and Venezuela

Abstract:The Gulf of Paria is bordered by both Trinidad and Venezuela, from which various metallic pollutants and other contaminants can originate. The Gulf is still a significant source of fish, crabs and shellfish for human consumption to both countries, where concerns over the quality of this marine environment have been long expressed but never properly addressed. In addition, the circulatory current patterns in the Gulf ensure that contaminants originating from either country are likely to affect both countries eventually. Heavy metals were determined in oysters (Crassostrea rhizophorae and C. virginica), green mussels (Perna viridis) and sediments from the Gulf of Paria. Samples were obtained at four sites in Trinidad and three sites in Venezuela in the Gulf of Paria, in addition to comparative samples collected from three sites on the north coast of Venezuela. Edible tissues of twelve shellfish from each location were blended and aliquots digested with concentrated nitric acid, for extraction of cadmium, chromium, copper, lead, nickel and zinc. The solutions were analysed by flame atomic absorption spectroscopy. Mercury was extracted with a mixture of nitric, hydrochloric and sulphuric acids and determined by cold vapour atomic absorption. Sediments were oven-dried at 60'C, before being similarly extracted. Results showed that mercury in sediments at all sites in Trinidad and Venezuela exceeded NOAA and Canadian sediment quality guidelines, while cadmium, copper, nickel, lead and zinc also exceeded these guidelines at several sites. Heavy metal levels in oysters and green mussels varied widely with location. However, oysters from the Gulf of Paria contained significantly higher mean levels of cadmium, copper, nickel and zinc than those from the north coast of Venezuela, but this difference was not apparent in mussels. Cadmium, mercury and zinc in sediments were significantly correlated with those of mussels, but not of oysters, in which copper and zinc at several sites in the Gulf of Paria exceeded local maximum permissible levels (Cu = 20 microg g(-1) wet wt; Zn = 50 microg g(-1) wet wt) for human consumption. These findings indicate that while mussels may be better biological indicators of heavy metal pollution in sediments than oysters, the latter may provide copper and zinc contamination. Further research is needed to determine the most appropriate biological indicators of heavy metal and other pollutants in the local marine environment and to develop protocols for their use.

Abstract:Sediments are periodically dredged from the major ports in the Gulf of Gabes (GG) during maintenance operations. These sediments are dumped near the coats of Sfax and Gabes cities. In the present study, 6 trace metals (Cd, Cu, Cr, Ni, Pb, and Zn) concentrations were assessed in both sediments and column water taken from the different basins of Sfax port. This study is the first to focus on the microbial and microalgae contaminations of Sfax port sediments. The spatial distributions of trace metals in dredged sediments from the different basins of Sfax port show that the maximum concentrations of Cd (13.75 μg/g), Cu (892.5 μg/g), and Zn (1447 μg/g) exceeded the Geode standard thresholds. The same elements, also detected in the water column, exceeded the toxicity thresholds for phytoplankton, shellfish, and algae (Cd 0.095 μg/l, Cu 4.52 μg/l, and Zn 37 μg/l). The presence of coliforms, indicators of sewage pollution, as pathogenic germs ( Pseudomonas ), was shown through microbiological examinations of the sediments and water column. The microalgae enumeration showed the abundance of dinoflagellate cysts (stressed form) which attests to the presence of severe conditions in Sfax port. Different toxic species were identified as Prorocentrum lima and Alexandrium minutum. The same trace metal sources and abundance in Sfax port and GG sediments suggest the hypothesis of their transfer from their disposal sites in coastal areas to deeper depths in the GG and until Boughrara lagoon (BL). As trace metals, microalgae species were also transferred by hydrodynamic currents inside the GG where they found suitable conditions to their proliferation causing the seawater coloration phenomenon, eutrophication, and degradation of the aquatic system.

Biomarkers in mussels from a copper site gradient (Visnes, Norway): an integrated biochemical, histochemical and histological study

Abstract:In the present work, mussels (Mytilus edulis) were transplanted into a copper (Cu) gradient in Visnes (Norway) for a period of 3 weeks during November 2003. Sites 1 and 2 showed similar low levels of Cu, site 3 had intermediate Cu levels and site 4 was the most polluted with Cu as confirmed by AAS of digestive gland tissue. Values of lysosomal membrane labilization period were significantly lower at sites 3 and 4 compared to sites 1 and 2. The volume density and size of lysosomes was significantly decreased at site 4. The volume density of neutral lipids was also significantly lower at site 4 compared with the rest of sites. The volume density of lipofuscins showed significantly higher values at sites 2 and 3 compared to the reference site 1. Similar results were obtained regarding bioavailable metal levels measured by autometallography. All together, results are indicative of exocytosis of metal-containing lysosomes and lipofuscins to the digestive tubule lumen in mussels from site 4. In fact, autometallographic metal deposits were detected in digestive tubule lumen, brown cells and stomach in site 4 mussels. In agreement, there was a loss of digestive cells in mussels from site 4 (atrophy of the digestive epithelium) and cell type replacement (diminished volume density of digestive cells and increased volume density of basophilic cells). In conclusion, selected biomarkers indicated that mussels transplanted to sites closest to the Cu mine showed significant differences in metal accumulation pattern and in organization of the digestive gland tissue. Finally, female mussels closest to the Cu mine showed advanced gametogenesis with higher gonad index and vitellogenin-like protein levels than mussels at sites 1 and 2.

Abstract:Information regarding chemical pollutant levels in farmed fish and shellfish, along with the risks associated with their consumption is still scarce. This study was designed to assess levels of exposure to 21 trace elements in fish (Dicentrarchus labrax), mussels (Mytilus galloprovincialis) and oysters (Crassostrea gigas) collected from aquaculture marine ecosystems of the northwestern Mediterranean Sea. Metal concentrations showed great variability in the three species; the highest values of the nonessential elements As and Cd were found in oysters while the highest levels of Al, Pb and V were found in mussels. The essential elements Cu, Mn and Zn were highest in oysters, but Fe, Cr, Ni, Se, Co and Mo levels were highest in mussels. Fish had the lowest concentrations for all trace elements, which were at least one order of magnitude lower than in bivalves. The rare earth elements cerium and lanthanum were found at higher levels in mussels than in oysters, but undetectable in fish. The maximum values set by European regulations for Hg, Cd and Pb were never exceeded in the examined samples. However, comparing the estimated human daily intakes (EHDIs) with the suggested tolerable copper and zinc intakes suggested a potential risk for frequent consumers of oysters. Similarly, people who consume high quantities of mussels could be exposed to concentrations of Al that exceed the proposed TWI (tolerable weekly intake).

Abstract:The mussel M. galloprovincialis is one of the most popular species in world's mariculture and environment pollution bioindicators. Although this mollusk was in a focus of numerous publications, the detoxifying and accumulating role of some of its body parts was insufficiently studied. The goals of the present work were as follows: (a) to study the distribution of potentially toxic elements (Cr, Co, Ni, Cu, Zn, As, Cd, Hg, Pb) in soft tissues, byssus, and shell liquor of this bivalve as a function of sampling location in the Black Sea near the southwestern coast of Crimea and (b) to assess human health risks from consuming soft tissues of mussels cultivated on a mollusk farm. Multivariate analysis showed significant differences in the overall distribution of the elements among the body parts and sampling sites under consideration. The trace element contents in soft tissues of M. galloprovincialis decreased in the following order: Zn > Cu > As > Ni > Pb > Cd > Cr > Co > Hg. The noncarcinogenic hazard index from the cultivated mussel consumption was found to be well below one and the carcinogenic risk index was found within the tolerable limits, which indicate the safety of consuming these mussels for humans. Byssus of M. galloprovincialis was characterized as a perfect indicator of marine environment pollution with Ni, Cu, Pb, Сo, and Cr. For the first time, the concentrations of trace elements were determined in the shell liquor and the function of byssus and shell liquor as the systems of trace element excretion from soft tissues was demonstrated.

Abstract:The bioaccumulation potential of contaminants in marine environments was investigated in biofilms and compared with caged mussels for a wide range of both organic and metallic contaminants across a large geographic area. Marine biofilms were sampled after three months of sub-surface immersion at 49 locations along the 1800 km of the French Mediterranean coast. Ten chemical elements (i.e. As, Cd, Co, Cr, Cu, Hg, Mn, Ni, Pb, and Zn) and 57 organic compounds (i.e., 18 polycyclic aromatic hydrocarbons (PAHs), 8 dioxin-like and 6 non-dioxin-like polychlorinated biphenyls (PCBs) and 25 organochlorine pesticides (OCPs)) were quantified in triplicates, revealing different multi-contaminated profiles depending on sites. Most of contaminants exhibited higher concentrations in biofilms than in mussels. Moreover, a remarkable significant and positive correlation between the concentrations in both biological matrices was observed for PAHs and PCBs, and more contaminant-dependent for OCPs and metals. These results highlighted the potential of biofilms as relevant bioindicators of the marine chemical contamination.

Abstract:BouIsmail (BIB) and Algiers (AB) are the most important bays in Algeria, where busy shipping activities and various industry complexes introduce different pollutants including heavy metals to the aquatic environment. The main goal of this study was to assess the contamination levels of heavy metals (Cd, Cr, Cu, Fe, Hg, Mn, Pb, Zn) in surface sediments and red mullet (Mullus barbatus) of the BIB and AB and to examine the possibility of the use of their enrichment factors (EFs) to track down the sources of metals (natural processes or human activity). The geoaccumulation index (Igeo) was calculated as a criterion to indicate the contamination level for each heavy metal. Moreover, geographical information systems based on spatial analysis methods (inverse distance weighting (IDW)) and statistical approaches (the principal component (PCA)) were performed to assess the spatial influences of multiple anthropogenic sources in different sampled areas. The results of both EF and Igeo revealed that the study area is exposed to various anthropogenic activities. The pollution load index (PLI) values of sediment samples in the different sites of Algiers and BIB ranged from 0.2 to 3.4 illustrating highly contaminated sediments. Significant bioaccumulation of Cd, Cu, Hg, Pb, and Zn (bioaccumulation factor >100%) were observed in muscle and liver of red mullet, suggesting potential health risks through consuming this fish species.

Abstract:Zinc (Zn) isotope compositions in soft mussel tissues help identify internal biological processes and track coastal Zn sources in coastal environments, thus aiding in managing marine metal pollution. This study investigated the seasonal and multi-decadal Zn isotope compositions of blue mussels (genus Mytilus ) from two French coastal sites with contrasting Zn environmental contamination. Concurrently, we characterized the isotope ratios of sediments and plankton samples at each site to understand the associations between organisms and abiotic compartments. Our primary objective was to determine whether these isotope compositions trace long-term anthropogenic emission patterns or if they reflect short-term biological processes. The multi-decadal isotope profiles of mussels in the Loire Estuary and Toulon Bay showed no isotope variations, implying the enduring stability of the relative contributions of natural and anthropogenic Zn sources over time. At seasonal scales, Zn isotope ratios were also constant; hence, isotope effects related to spawning and body growth were not discernible. The multi-compartmental analysis between the sites revealed that Toulon Bay exhibits a remarkably lower Zn isotope ratio across all studied matrices, suggesting the upward transfer of anthropogenic Zn in the food web. In contrast, the Zn isotope variability observed for sediments and organisms from the Loire Estuary fell within the natural baseline of this element. In both sites, adsorptive geogenic material carrying significant amounts of Zn masks the biological isotope signature of plankton, making it difficult to determine whether the Zn isotope ratio in mussels solely reflects the planktonic diet or if it is further modified by biological homeostasis. In summary, Zn isotope ratios in mussels offer promising avenues for delineating source-specific isotope signatures, contingent upon a comprehensive understanding of the isotope fractionation processes associated with the trophic transfer of this element through the plankton.

Abstract:Toxic effects of trace metals on digestive gland and gill cytoarchitecture were studied in the mussel Mytilus platensis after metal detection in the environment. Mussels were collected from the Port of San Antonio Este and from a reference site called Isla Mejillón, both in Rio Negro province (Argentina). Histological techniques, morphometrical analysis and lipofuscin detection with Nile blue method were performed. Mussels from the seaport presented dilated hemolymphatic vessels with numerous hemocytes in the gill filaments and fused filaments in the frontal zone. Morphometric analysis of the gill filaments showed an increase in the epithelial thickness of the lateral zone that bears cilia, and we infer that water filtering might be affected. The sublateral zone was also thickened, probably interfering with gas exchange. A reduction in the abfrontal zone epithelial thickness was also noticed, so water pumping, lubrication and cleaning of the gills might be impaired. Digestive glands of seaport mussels accumulated lipofuscin granules within the digestive cells, accounting for lipid and protein peroxidation. The observed histological alterations in gill and digestive gland in mussels from North Patagonia seaport are relevant findings to be considered as biomarkers of effect after exposure to metals.

Abstract:The main objective of this study was to assess the impact of environmental pollution on the gastropod Stramonita haemastoma by examining various physiological and biochemical parameters. Trace metal elements (TME) were measured in the snail's foot. Over a one-year period from 2013 to 2014, the study investigated the variations in reduced glutathione , malondialdehyde, metallothionein, glutathione S-transferase, acetylcholinesterase and catalase activities in the foot of the gastropod at three different sites. The results showed spatiotemporal fluctuations in biomarkers, physiological parameters and TME. This integrative approach, being the first of its kind in the region using S. haemastoma as a model organism, represents a means of environmental assessment accessible to all countries allowing the monitoring and conservation of coastal marine environments on a national and international scale and which could be used in marine biomonitoring programs to indicate the impact of pollution and assess the quality of the Algerian coastal marine environment.

Abstract:A comparative assessment of the contents of Fe, Zn, Cu, Cd, Mn, Pb, and Ni in the soft tissues of bivalve mollusks Mytilus trossulus , Mytilus galloprovincialis, and their hybrid forms from Minonosok Bay of Peter the Great Bay (Sea of Japan) has been made. No significant differences in the accumulation of Fe and Cd in the tissues of all studied groups of mollusks were found. It has been shown that the hybrid forms of these species had more intensive growth, and against this background they accumulated significantly lower concentrations of such metals as Cu, Mn, Pb, and Ni in their soft tissues compared to the parental species. A significant negative correlation has been found between the accumulation of these metals in the tissues of mollusks and the length and weight of their shells and whole soft tissues M. trossulus , which had a significantly lower weight of valves and soft tissues, differed from M. galloprovincialis in the accumulation of significantly higher concentrations of Zn, for which no relationship has been found between its content and all the studied size and weight characteristics. The conducted testing did not reveal any significant dependence of the accumulation of all studied metals, with the exception of Pb, in the tissues of mollusks on their condition index. The results we obtained indicate the need to take the above features of the trace metal composition of the soft tissues of the studied species into account when using them in monitoring environmental pollution with heavy metals.

Abstract:The concentrations of six metals (Ag, Cd, Cu, Hg, Pb, and Zn) were investigated and compared in three tissues (arms, digestive gland, and mantle) of three cephalopod species from the Tunisian waters: the common octopus (Octopus vulgaris), the common cuttlefish (Sepia officinalis), and the European squid (Loligo vulgaris). Whatever the species or the sites, the digestive gland displayed the highest concentrations of Ag, Cd, Cu, Pb, and Zn, highlighting its major role in their bioaccumulation and detoxification. This is also true for Hg but only for the digestive gland of O. vulgaris. Muscle from the arms and the mantle contained thus relatively low trace metal concentrations except for Hg in L. vulgaris and S. officinalis. Geographic comparison of metal concentrations in Tunisian cephalopods from three locations indicates that higher concentrations of Ag, Pb, and Hg were observed in cephalopods from northern and eastern coasts, whereas the highest Cd levels were detected in the southeastern, reflecting different conditions of exposure. Comparing the trace element concentrations between species, Ag, Cd, Cu, Hg, and Zn concentrations were the highest in the digestive gland of octopuses. This may be related to the differences in ecological features and swimming behavior among different cephalopod species. Effects of length and sex on metal levels were also considered, indicating a limited influence of sex on metal concentration.

Abstract:This study aims to evaluate the liver, gill, and muscle tissue heavy metal accumulation of the thin lip mullet ( Chelon ramada ) from Ain Ziana Lagoon, Benghazi. The heavy metals included zinc, copper, lead, and manganese. Between September 2021 and August 2022, specimens were collected on a monthly basis. They were identified using DNA barcoding, namely the CO1 gene, which is associated with the sequence that is BankIt in the NCBI gene bank (accession number OR533708). The research tracked the lagoon's physicochemical characteristics, finding that heavy metal concentrations spiked in the summer and that water temperature, pH, and salinity varied throughout the year. Except for copper in the winter, lead, manganese, and copper levels were all above acceptable limits, according to a comparison with EPA (2005) regulations. On the other hand, zinc levels were under these limitations. The amounts of all metals in the sediment regularly exceeded the limits set by the EPA. Zinc and manganese levels in fish tissues were within the approved ranges according to WHO and FAO standards but lead and manganese levels were far higher than allowed, indicating possible pollution from sewage or industrial sources. There were noticeable seasonal patterns in bioaccumulation, especially during the warmer months, but manganese did not exhibit a distinct pattern. Also noted that the thin lip mullet's mineral absorption and release capacities varied with the different seasons and tissues. Lastly, the Metal Pollution Index (MPI) showed that there was a larger possibility for bioaccumulation in the liver, gill, and muscle during the study period.

Abstract:A multibiomarker approach helps assess environmental health as it provides a complete tool to understand the effects of environmental stressors on ecosystems and human health. We applied this approach in the central Atlantic Ocean of Morocco, an area subjected to the impact of many types of pollutants, threatening the durability of its resources. In this study, four biomarkers acetylcholinesterase (AChE), glutathione-s-transferase (GST), metallothioneins (MTs), and catalase (CAT) were measured in the digestive gland of the mussel Mytilus galloprovincialis collected from four sites: Imsouane (S1), Cap Ghir (S2), Imi Ouaddar (S3), and Douira (S4). These sites were chosen due to the diversity of impacts ranging from industrial to agricultural and touristic. We also assembled all the enzymatic responses (AChE, GST, CAT, and MTs), using the integrated biomarker response (IBR), to estimate the degree of impact of pollutants at the prospected sites to reveal all the complex interactions between biomarkers and to classify sites via the integrated approach. Results show a seasonal change in biomarker responses with variability between sites. We also recorded the highest levels of AChE inhibition and GST induction in S1, higher levels of catalase activity in S4, and a significant impact on metallothionein concentration in S1 and S3. This project highlights the interest in using a multibiomarker approach to ensure accurate interpretation of biomarker variation to protect the Moroccan coast and its resources.

Abstract:The bioaccumulation of trace metals Cd, Cr, Cu, Fe, and Zn in soft tissues of the barnacle Pollicipes pollicipes was investigated seasonally along the Atlantic coast of northwestern Morocco. Average concentrations (μg g −1 dry weight) exhibited a decreasing order: Fe (548.15 ± 132.43) > Zn (430.80 ± 181.68) > Cd (17.46 ± 9.99) > Cu (7.72 ± 1.26) > Cr (3.12 ± 0.80), with the highest levels during wet seasons. The "Metal Pollution Index" and "Individual Multimetal Bioaccumulation Index" revealed a substantial barnacle contamination in industrialized areas. Additionally, Cd and Zn concentrations surpassed permissible guideline limits. While the "Target Hazard Quotient" and "Hazard Index" unveiled no significant health risks associated with barnacle consumption for humans, Cd posed potential risks, particularly for children consuming barnacles from polluted locations. Regarding the "Maximum Safe Consumption", Cd demonstrated potential harm across all sex and age groups. These findings contribute valuable data on the safety of barnacle consumption, marking the initial assessment of such risks in Morocco. The study offers evidence of metal pollution occurrence and proposes the barnacle species as a reliable biomonitor of trace metal bioavailabilities in marine coastal areas. To our knowledge, this investigation is the first comprehensive report of metal contamination biomonitoring using barnacles from Moroccan Atlantic waters.

Abstract:Size of organisms is critical in controlling metal bioavailability and bioaccumulation, while mechanisms of size-related metal bioaccumulation are not fully understood. To investigate the influences of different sources of particle-associated Cu on body size-related Cu bioavailability and bioaccumulation, zebra mussels (Dreissena polymorpha) of different sizes were exposed to stable Cu isotope (Cu-65) spiked algae (Chlorella vulgaris) or sediments in the laboratory and the Cu tissue concentration-size relationships were compared with that in unexposed mussels. Copper tissue concentrations decreased with mussel size (tissue or shell dry weight) in both unexposed and algal-exposed mussels with similar decreasing patterns, but were independent of size in sediment-exposed mussels. Furthermore, the relative contribution of Cu uptake from algae (65-91%) to Cu bioaccumulation is always higher than that from sediments (9-35%), possibly due to the higher bioavailability of algal-Cu. Therefore, the size-related ingestion of algae could be more important in influencing the size-related variations in Cu bioaccumulation. However, the relative contribution of sediment-Cu to Cu bioaccumulation increased with body size and thus sediment ingestion may also affect the size-related Cu variations in larger mussels (tissue weight >7.5 mg).
