Abstract:The present study was undertaken to investigate the impact of heat stress on nutrient digestibility and tibia and reproductive traits, and changes in laying performance, egg qualities, fecal volatile fatty acids, and plasma parameters in laying hens. One-hundred twenty 52-week-old laying hens were raised in three temperature-controlled facilities with constant humidity (50% RH), either normal temperature (LT; 22°C) or heat stress considered being moderate (MT; 27°C) or severe (HT; 32°C) for 42 days. Feed intakes were consistently low (p < 0.01) in HT hens compared with those in LT or MT over the period of 42 days. Egg production kept markedly (p < 0.05) or numerically (p > 0.05) low in hens exposed to HT vs. LT or MT. Egg mass and egg weight were consistently low (p < 0.01) in hens exposed to HT compared with those raised under LT or MT. On the other hand, feed conversion ratio and frequency of dirty and cracked eggs were not significantly affected (p > 0.05) during the experimental period. HT-exposed hens consistently had lowered (p < 0.05) eggshell thickness and breaking strength, eggshell weight, and plasma Ca, P, and Mg levels compared with LT- or MT-treated hens. HT hens had lower (p < 0.01) relative oviduct weight and less number of large yellow follicles compared with those raised under LT or MT conditions at 42 days. Tibia traits measured at 42 days were not affected by any of heat treatments. Fecal volatile fatty acids tended to be higher in HT-exposed laying hens throughout the experiment. It was noted that digestibilities of neutral detergent fiber and dry matter were lowest (p < 0.05) in hens exposed to HT vs. LT or MT environments. Our study suggests that heat stress could lower laying performance, egg quality, and physiological parameters that are coupled with alterations in gut metabolites and mineral/lipid metabolism. The findings emerged from this study will help us design the nutritional and environmental strategies to mitigate the negative effect of heat stress on laying hens.

Abstract:
The effects of simulated transportation conditions on the core body and extremity temperature, blood physiology, and behavior of white-strain layer pullets

Abstract:Transportation of poultry is stressful. The transportation of broilers has been well studied, while the transportation of layer pullets from rearing to laying facilities has not been thoroughly evaluated. This experiment aimed to establish the effects of temperature (T)/RH combinations and duration (D) of transport, via a 5 × 2 factorial arrangement of simulated transport conditions using 5 T/RH combinations (21°C with 30% RH [21/30], 21°C with 80% RH [21/80], 30°C with 30% RH [30/30], 30°C with 80% RH [30/80], and -15°C with uncontrolled RH [-15]), and 2 exposure D (4 or 8 h). Pullets (18-19 wk; n = 240) were obtained from 3 commercial farms (N = 3 farms). Pretreatment, birds were orally administered a miniature data logger to record core body temperature (CBT), an initial blood sample was taken (5 birds/replicate), and initial foot T was recorded. Behavior during exposure was video recorded. Following exposure, a final blood sample was taken (analyzed for heterophil to lymphocyte ratio, partial pressure of CO2, total CO2, bicarbonate, and glucose), birds were slaughtered, and data loggers were retrieved. Data were analyzed as a randomized complete block design via Proc Mixed (SAS 9.4) and significance was declared at P ≤ 0.05. There were no interactions observed for the T/RH and D combinations throughout the study. The CBT and foot T were lowest in pullets exposed to -15 compared with all other treatments. Foot T was also highest in pullets exposed to 30/80 compared with -15, 21/30, and 21/80. There was no impact of T/RH on pullet blood physiology. Activity and thermoregulatory behaviors were impacted by the T/RH combinations. Pullets exposed to 30/30 and 30/80 spent the most time panting. Pullets exposed to 30/80 also spent the least amount of time motionless. Duration had minor impacts on pullet CBT, blood physiology, and behavior. These data indicate that as a response to thermal stress, layer pullets were successful at implementing mechanisms to maintain homeostasis.
The Effect of Supplemental L-Threonine on Laying Performance, Serum Free Amino Acids, and Immune Function of Laying Hens under High-Temperature and High-Humidity Environmental Climates

Abstract:Babcock Brown layers (n = 960), 40 wk of age, were allocated to 1 of 5 dietary treatments groups, each of which included 6 replicates of 32 hens. Each group received the same basal diet formulated with corn, peanut meal, and crystalline amino acids for 8 wk. L-Threonine was added to the basal diet at 0 (control), 0.1, 0.2, 0.3, and 0.4%, to achieve 0.47 (NRC, 1994), 0.57, 0.67, 0.77, and 0.87% threonine, respectively. Although supplementing the diet with L-threonine did not affect ADFI, FCR, egg weight, or egg quality (P > 0.05), the egg production response to supplemental L-threonine was quadratic, and it was maximized at 0.2% supplemental L-threonine. No differences were observed for uric acid, lactate dehydrogenase, alkaline phosphatase, Ca, and P concentrations among the treatments. Serum total protein concentration increased quadratically to supplemental threonine, and the response was maximized at 0.2 and 0.3% supplemental L-threonine. Serum free threonine increased quadratically as supplemental threonine increased, and the response was maximized between 0.2 and 0.3% supplemental L-threonine. The addition of L-threonine at 0.3% of the diet resulted in linearly increasing levels of IgG and total Ig (P < 0.05) as compared with those of the control group. In conclusion, current NRC (1994) recommendations for dietary threonine are insufficient for modern commercial laying hens strains reared under subtropical summer climates. We suggest that 0.2% threonine resulted in optimal egg production, whereas 0.3% L-threonine may have had a positive effect on the humoral immune response of laying hens under conditions of high temperature and humidity.

Abstract:There is no information regarding the influence of heat stress (HS) on host metabolic profile. In this study, we investigated the effects of different environmental temperatures on oxidative status, hormone levels, HS indicators, and plasma metabolites in broilers. A total of 1,680 yellow-feather broilers (28 D old) were randomly allotted to 4 groups with 6 replicates. The broilers (29-57 D old) were maintained in thermostatic rooms (20 degrees C, 25 degrees C, 28 degrees C, and 30 degrees C) for 28 consecutive days. The results showed that the plasma cortisol and adrenocorticotropic hormone levels and creatine kinase and lactate dehydrogenase activities gradually increased when the temperature increased from 20 degrees C to 30 degrees C. However, the insulin-like growth factor-I level decreased gradually. Furthermore, heat shock protein 70 expression significantly increased in the liver and breast muscle (P < 0.01). As the temperature increased, the total anti-oxidant capacity in the plasma and liver gradually decreased, whereas the malondialdehyde level increased. The activity of plasma glutathione peroxidase and total superoxide dismutase in the liver showed a similar increasing trend (P < 0.01). In addition, 15 metabolites were identified at higher (P < 0.05) levels, whereas 2 metabolites were identified at lower (P < 0.05) levels in the 30 degrees C treatment group than those in the 25 degrees C treatment group. Most of these potentially diagnostic biomarkers are involved in carbohydrate, amino acid, lipid, or gut microbiome-derived metabolism, indicating that HS affected the metabolic pathways in broilers. Six candidate metabolites (tartronic acid, l-bethreine, tartaric acid, allose, glut aric acid, and neohesperidin) were selected as biomarkers, as they showed high sensitivity, specificity, and accuracy in diagnosing broilers under HS (P < 0.01). In conclusion, in the final stage of growth, we identified 6 plasma differential metabolites as potential biomarkers of HS-induced metabolic disorders in yellow-feathered broilers. This work offers new insights into the metabolic alterations of broilers exposed to HS and provides a new perspective for further study.
Betaine and feed restriction as potential mitigation strategies against heat stress in two strains of laying hens

Abstract:Climate change is increasingly manifesting in temperate regions. Laying hens are highly sensitive to heat stress and mitigation strategies should be implemented to reduce the negative effects. The goal of this experiment was to evaluate the effects of betaine in drinking water (0.55 g/L) and 4 h feed restriction during peak heat stress on laying performance, egg quality, blood gas parameters, body temperature (Tb), and oxidative stress in 2 different breeds of laying hens. Therefore, 448 ISA Brown hens (25 wk) and 448 Lohmann LSL classic laying hens (26 wk) were housed in 128 cages (7 hens/cage). Thermoneutral (TN) data was collected for 21 d before cyclic heat stress (HS) (21d; 32 ± 2°C; 6 h daily). During HS, hens were divided into 4 treatments: 1) feed restriction (FR), 2) betaine supplementation in drinking water (BET), 3) feed restriction and betaine supplementation in drinking water (FR-BET), or 4) control (CON). The effects were evaluated after 1 wk of HS (acute heat stress; AHS) and 3 wk of HS (chronic heat stress; CHS). Laying rate and egg mass (EM) diminished during CHS but decreased more in white than brown hens (2.78% and 1.94%; -1.57% and -0.81%, respectively; P = 0.004) and remained unaltered by BET or FR. During AHS, average daily feed intake (ADFI) increased compared to TN, but the increase was higher in white than brown hens (6.36% and 2.62%, respectively; P = 0.001). Egg shell quality deteriorated during AHS and CHS, but was most affected in white hens, FR or BET did not impact this. Blood pCO2, HCO3- and base excess significantly decreased during AHS and CHS, but pH and iCa were unaltered. Blood glucose increased in white hens during AHS compared to TN (P < 0.001), while plasma malondialdehyde increased in brown hens (P < 0.001). Results indicated that laying hens experienced HS, but breed differences were observed and white hens were generally most affected. FR affected feed conversion ratio negatively during CHS. However, FR and BET could not improve laying performance, egg quality, Tb, or blood parameters during HS.

Abstract:A total of 245 hens and 35 cocks (32 weeks age) were assigned to seven treatment groups (five replicates with seven hens and one cock) to investigate the effect of dietary electrolyte balance (DEB) and arginine to lysine ratio (Arg/Lys) on birds' physiological and biochemical traits under cyclic heat stress (CHS) condition. Birds were housed in an environmentally controlled facility having four sectors. The first group (positive control, PC) was kept under thermoneutral conditions and fed diet with DEB of 180 mEq and Arg/Lys of 1.25, whereas the other six treatments were kept in the second sector under CHS and fed diet with DEB and Arg/Lys equal to: 180 mEq and 1.25 (negative control, NC); 250 mEq and 1.25; 320 mEq and 1.25; 180 mEq and 1.37; 250 mEq and 1.37; 320 mEq and 1.37, respectively. Hens on NC group had significantly decreased red blood cells (RBCs), white blood cells (WBCs) and its fractions. The groups fed different DEB and Arg/Lys in diet significantly enhanced the blood parameters and plasma lipid profile compared NC group. Hens under CHS fed on 250 and 320 DEB with 1.37 Arg/Lys recorded the lowest concentration of low-density lipoprotein (LDL) compared with the other groups. Triiodothyronine (T<sub>3</sub>) activity was not differed among groups, while T4 activity in layer exposed to CHS (NC group) recorded the highest activity compared to PC. From findings, it can be concluded that laying hens fed a diet having DEB 250 mEq with 1.37 Arg/Lys could be successfully applied to counteract the adverse effect of CHS and to improve blood hematological and biochemical traits, antioxidants, and immunity response.

Abstract:Five hundred seventy-six Hy-Line Brown hens, 40 wk old, were randomly allotted to 4 groups, each of which included 4 replicates of 36 hens. The groups were given a basal corn-soybean meal diet supplemented with 0 (control group), 15, 30, and 60 mg/kg of lead. No differences were observed for laying rate and FCR among the treatments, whereas adding 15, 30, and 60 mg/kg of lead decreased ADFI. Lead concentrations in albumen, yolk, liver, kidney, oviduct, and ovary of the treatment groups were higher than those of the control group. The addition of lead at 60 mg/kg resulted in increased levels of serum urea nitrogen, urea acid, and glutamic oxalacetic transaminase activity. Levels of serum progesterone, follicle-stimulating hormone, and luteinizing hormone in the control group were significantly higher than those of the treatment groups. In conclusion, we indicate that lead concentrations in tissues and eggs have a positive correlation with dietary lead levels, and lead may affect the reproductive performance of hens by reducing the levels of endocrine hormones.
Effect of Mercury Chloride on Oxidative Stress and Nuclear Factor Erythroid 2-Related Factor 2 Signalling Molecule in Liver and Kidney of Laying Hens.

Abstract:This study investigated the effects of mercury chloride (HgCl2) on the deposition of mercury (Hg), histopathology and oxidative stress in liver and kidney of laying hens. The gene expressions of antioxidant enzymes and nuclear factor erythroid 2-related factor 2 (Nrf2)-Kelch-like ECH-associated protein 1 (Keap1) were further studied to uncover the molecular mechanism. A total of 960 40-week-old Hyline brown laying hens were randomly allocated to five treatments with eight pens per treatment and 24hens per pen. The hens were fed with five experimental diets containing graded levels of Hg at 0.270, 1.250, 3.315, 9.405 and 27.230mg/kg respectively. Results revealed that both deposition of Hg and score of injury in liver and kidney were significantly increased as dietary Hg dosage up to 27.230mg/kg diet. Deposition of Hg was positively related to score of injury in liver and kidney of laying hens. Besides, the activities of superoxidative dismutase (SOD), catalase (CAT), glutathione reductase (GR) and glutathione peroxidase (GSH-Px), and glutathione (GSH) content all significantly decreased (p<0.05), while malondialdehyde (MDA) content significantly increased (p<0.05) after Hg exposure in liver and kidney of laying hens. In addition, positive relationships occurred between antioxidant enzyme activities and antioxidant enzyme gene expressions except between SOD activity and manganese superoxide dismutase (MnSOD) gene expression in liver. Meanwhile, Nrf2 gene expression was positively related to antioxidant gene expressions and negatively connected with Keap1 gene expression. Negative relationships occurred between Nrf2 and Keap1 protein levels in liver and kidney. In conclusion, Hg could dose-dependently damage liver and kidney and induced hepatic and renal oxidative stress by means of suppressing Nrf2-Keap1 signalling molecule in laying hens.

Abstract:This study was conducted to elucidate the molecular mechanisms underlying heat stress (HS)-induced abnormal egg-laying in laying hens. Hy-Line brown laying hens were exposed to HS at 32 °C or maintained at 22 °C (control) for 14 days. In addition, granulosa cells (GCs) from preovulatory follicles were subjected to normal (37 °C) or high (41 °C or 43 °C) temperatures in vitro. Proliferation, apoptosis, and steroidogenesis were investigated, and the expression of estrogen and progesterone synthesis-related genes was detected. The results confirmed that laying hens reared under HS had impaired laying performance. HS inhibited proliferation, increased apoptosis, and altered the GC ultrastructure. HS also elevated progesterone secretion by increasing the expression of steroidogenic acute regulatory protein (StAR), cytochrome P450 family 11 subfamily A member 1 (CYP11A1), and 3b-hydroxysteroid dehydrogenase (3β-HSD). In addition, HS inhibited estrogen synthesis in GCs by decreasing the expression of the follicle-stimulating hormone receptor (FSHR) and cytochrome P450 family 19 subfamily A member 1 (CYP19A1). The upregulation of heat shock 70 kDa protein (HSP70) under HS was also observed. Collectively, laying hens exposed to high temperatures experienced damage to follicular GCs and steroidogenesis dysfunction, which reduced their laying performance. This study provides a molecular mechanism for the abnormal laying performance of hens subjected to HS.

Abstract:This study was designed to investigate the effects of γ-aminobutyric acid(GABA) on laying performance and egg quality of laying hens under high temperature in summer,and to study the mechanism of action from antioxidation,immunity and endocrine.Four hundred and eighty 320-day-old Roman hens were randomly divided into five groups with three replicates per group and thirty-two hens per replicate.One group received the basal diet as a control,while the others received the basal diet supplemented with 25,50,75,100 mg/kg GABA as treatments,respectively.The experiment lasted for 60 days.The results showed as follows: 1) laying performance of hens in all treatments was improved(P0.05),while feed intake was not significantly affected(P0.05),and 50 mg/kg was the optimal dosage which significantly increased laying rate and average egg weight(P0.05),and significantly decreased feed/egg ratio(P0.05).2) 50 mg/kg GABA significantly increased eggshell strength(P0.05),improved albumen height and Haugh unit(P0.05),and decreased cholesterol content of egg yolk(P0.05).3) GABA at 50 mg/kg significantly increased serum GSH-Px and SOD activities(P0.05),and decreased serum MDA content(P0.05).It also significantly increased FSH,E2,T3,FT3,IgG,IgA and complement C3 contents in serum(P0.05).In conclusion,supplementing γ-aminobutyric acid in a diet can effectively improve the laying performance,egg quality and heat stress resistance capability of hens by enhancing the antioxidant capacity,immune function and increasing the secretion of gonadal and thyroid hormones.The optimal dosage of GABA is 50 mg/kg.
Evaluating the Impact of Excess Dietary Tryptophan on Laying Performance and Immune Function of Laying Hens Reared under Hot and Humid Summer Conditions

Abstract:1. Tryptophan (Trp), besides its role as an essential amino acid in protein synthesis, may also have other important effects on laying hens under summer conditions. 2. Babcock Brown layers (n = 768), 40 weeks of age, were allocated to 4 treatment groups, each of which included 6 replicates of 32 hens. Each group received the same basal diet, formulated with maize and soybean meal, for 8 weeks. Hens were fed on the basal diet with 0·0, 0·2, 0·4, and 0·8 g/kg L-Trp to achieve dietary concentrations of 1·7, 1·9 g/kg, 2·1 g/kg or 2·5 g/kg of Trp, respectively. 3. Supplementing L-Trp had no affect on laying performance. Adding 0·2 or 0·4 g/kg L-Trp improved egg shell strength compared with those fed on the control diet. Serum albumin concentration increased at 0·4 g/kg compared with those receiving 0·0 or 0·8 g/kg Trp. The addition of Trp at 0·4 g/kg increased serum IgM concentration quadratically. Serum superoxide dismutase (T-SOD) increased linearly and quadratically at 0·4 g/kg. 4. In conclusion, we suggest that 0·2 to 0·4 g/kg Trp may have beneficial effects on laying hens under conditions of high temperature and humidity.
Effect of Dietary Γ-Aminobutyric Acid on Laying Performance, Egg Quality, Immune Activity and Endocrine Hormone in Heat-Stressed Roman Hens.

Abstract:This study was conducted to evaluate the effect of γ-aminobutyric acid (GABA) on laying performance, egg quality, digestive enzyme activity, hormone level and immune activities in Roman hens under heat stress. Roman hens (320 days old) were fed with 0, 25, 50, 75 and 100 mg/kg GABA, respectively during a 60-day experiment. Compared with control, supplementation of 50 mg/kg GABA improved the laying performance and egg quality by significantly increasing egg production, average egg weight and shell strength (P < 0.05), while decreasing the feed-egg ratio and cholesterol level. Anti-oxidation activity was improved by significantly increasing the activity of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px), but decreasing malondialdehyde level in serum (P < 0.05), while significantly increasing the glucose and total protein (TP) level, follicle stimulating hormone (FSH), luteinizing hormone (LH), estradiol (E(2) ), insulin, triiodothyronine (T(3) ) and free triiodothyronine (FT(3) ) levels, and IgG, IgA and complement (C3)activity in serum (P < 0.05). The results indicated that oral GABA improved laying performance and physical condition mainly by modulating hormone secretion, enhancing anti-oxidation and immune activity, and maintaining electrolyte balance. Fifty mg/kg was the optimum level for laying hens under heat stress in the present study.

Abstract:Historical data, epidemiological surveys, laboratory and animal experiments, clinical studies and observations in ordinary dental practice all illustrate that starchy fibrous foods have a low cariogenic potential. The recommended substitution of sucrose in Western diets by starchy foods would lead to a reduction of caries provided that these other carbohydrates replace the between meal consumption of sugar-rich products. The low cariogenic potential of a diet containing carbohydrates with a fibrous character is probably not solely due to its low sucrose content. Such a diet promotes mastication and saliva secretion which tend to reduce the caries risk. Furthermore, unrefined foods may contain caries-protective substances like phytates, trace elements and antibacterial factors. Lectins, which are found in many plants could be of importance by influencing the attachment of micro-organisms to the teeth in a favourable manner. The tradition to end a meal with fruit may thus have other oral effects than hitherto considered. In addition, a diet containing carbohydrates with a fibrous character and a low amount of sucrose probably has a low pathogenic potential for the periodontal tissues. However, no epidemiological data are available to support this assumption.
The effects of simulated transport conditions on the muscle tissue characteristics of white-strain layer pullets

Abstract:The objective of this study was to evaluate the effects of temperature (T)/relative humidity (RH) combinations and exposure duration (D) on the muscle tissue characteristics of layer pullets during simulated transport. While layer pullets are not processed for meat, muscle physiology can be used as an indicator to assess welfare. Pullets (n = 240) were randomly assigned to 1 of 5 T/RH combinations (-15°C uncontrolled RH [-15], 21°C 30%RH [21/30], 21°C 80%RH [21/80], 30°C 30%RH [30/30], and 30°C 80%RH [30/80]) and 2 D (4 or 8 h) in a 5 x 2 factorial arrangement (3 replications). Birds were weighed before exposure, crated (density 45.5 kg/m2) and exposed to the conditions above. After exposure, birds were weighed (live shrink calculated) and slaughtered using a small-scale facility. Postslaughter, carcasses were eviscerated, and an initial pH was obtained from the right breast and thigh. Final breast and thigh pH and color values (lightness [L∗], yellowness [b∗], and redness [a∗]) were obtained 30 h postslaughter. Left breast muscles were frozen and analyzed for thaw and cook loss 4 wk postslaughter. Data were analyzed as a randomized complete block design via ANOVA (Proc Mixed; SAS 9.4), with farm of origin as block. Differences were considered significant when P ≤ 0.05. Live shrink (kg) was higher for pullets exposed to 30/30 and 30/80 compared with those exposed to 21/80 (P = 0.04) and for pullets exposed for 8 h compared with 4 h (P < 0.01). Breast muscle thaw loss (%) was higher in pullets exposed for 4 h compared with 8 h (P = 0.01). Breast and thigh muscle a∗ were higher for pullets exposed to 30/30 compared with 21/30 (P = 0.02). Thigh muscle b∗ was lower for pullets exposed to -15 compared with 21/80 (P = 0.05). Breast b∗ was higher for pullets exposed for 8 h compared with 4 h (P = 0.04). The results from this study demonstrates that increasing exposure D had minor effects on pullet muscle characteristics. In addition, layer pullets coped well with thermal stressors associated with simulated transport.

Abstract:One hundred and fifty Hy-line layer hens, 40 weeks of age were used to study the impact of hot climate on egg production, eggshell quality, intestinal calcium transport and calcium binding protein concentration of the laying hen. Birds were randomly allocated to two experimental groups equally, the 1st group, that served as a control, was reared at room temperature under thermoneutral condition which ambient temperature was 25±1ºC, while the second group was subjected to 40±1ºC for 5 days (4 hrours daily). Egg production, eggshell quality, serum concentrations of calcium, phosphorous, thyroid, aldosterone and estrogen hormones as well as acid phosphatase and alkaline phosphatase activities were estimated. Calcium binding protein concentration and the transport of radioactive calcium ( 45Ca) across the duodenal were also measured. The results indicated that egg weight, hen-day egg production and eggshell quality were inversely related to high temperature peroids. Serum calcium, calcium binding protein, phosphrous and alkaline phosphatase values were significantly lowered in heat stressed birds. The highest values for serum acid phosphatase and duodenal calcium transport were recorded for heat-treated hens at the 5th days of heat stress period. Thyroid and estrogen hormones unlike aldosterone hormone concentrations declined under hot climates. In conclusion, the present study recommends avoiding exposure to heat stress conditions because of its negative impact on the level of blood calcium and intestinal calbindin-D28k and the consequent decrease in the egg production rate and the deterioration of the eggshell quality

Abstract:Abstract The adrenal gland responds to heat stress by epinephrine and glucocorticoid release to alleviate the adverse effects. This study investigated the effect of acute heat stress on the protein profile and histone modification in the adrenal gland of layer-type country chickens. A total of 192 roosters were subject to acute heat stress and thereafter classified into a resistant or susceptible group according to body temperature change. The iTRAQ analysis identified 80 differentially expressed proteins, in which the resistant group had a higher level of somatostatin and hydroxy-δ-5-steroid dehydrogenase but a lower parathymosin expression in accordance with the change of serum glucocorticoid levels. Histone modification analysis identified 115 histone markers. The susceptible group had a higher level of tri-methylation of histone H3 lysine 27 (H3K27me3) and showed a positive crosstalk with K36me and K37me in the H3 tails. The differential changes of body temperature projected in physiological regulation at the hypothalamus–pituitary–adrenal axis suggest the genetic heterogeneity in basic metabolic rate and efficiency for heat dissipation to acclimate to thermal stress and maintain body temperature homeostasis. The alteration of adrenal H3K27me3 level was associated with the endocrine function of adrenal gland and may contribute to the thermotolerance of chickens.

Abstract:High ambient temperature is one of the most important environmental factors negatively impacting poultry production and health. Genetics is an important contributor in mitigating the stress response to heat. Two genetically distinct highly inbred lines of similar body size (Leghorn and Fayoumi) were characterized for phenotypic differences in response to heat. At 14 days of age, birds were exposed to 38°C with 50% humidity for 4 hours, then 35°C until the conclusion of the experiment. Non-treated individuals were kept at 29.4°C for the first week and then 25°C throughout the experiment. Birds in the heat-stress group were inoculated at day (d) 21 with Newcastle disease virus (NDV) La Sota strain to investigate the effects of heat stress and NDV infection. Thirteen blood parameters were measured using the iSTAT blood analyzer at three stages: 4 h, 6 d, and 9 d post heat-stress treatment, representing acute heat (AH) exposure, chronic heat (CH1) exposure, and chronic heat exposure after virus infection (CH2), respectively. Most blood parameters were significantly changed with heat stress in Leghorns at AH and in Fayoumis at CH1 and CH2. The Leghorn line had significant acute responses with disrupted acid-base balance and metabolic disorders. The heat-resilient Fayoumis maintained a relatively well-balanced acid-base balance. The current study provides the comprehensive profile of biomarker signatures in blood associated with heat tolerance and suggests that PO2, TCO2, HCO3, and base excess can be served as potential biomarkers that can be used to genetically improve heat tolerance in poultry.

Abstract:This study was done to evaluate the effects of heat stress (HS) on production performance, redox status, small intestinal barrier-related parameters, cecal microbiota, and metabolome of indigenous broilers. A total of forty female indigenous broilers (56-day-old) were randomly and equally divided into normal treatment group (NT group, 21.3 ± 1.2°C, 24 h/day) and HS group (32.5 ± 1.4°C, 8 h/day) with five replicates of each for 4 weeks feeding trial. The results showed that the body weight gain (BWG) of broilers in HS group was lower than those in NT group during 3–4 weeks and 1–4 weeks ( p < 0.05). The HS exposure increased the abdominal fat rate ( p < 0.05) but decreased the thigh muscle rate ( p < 0.01). Besides, broilers in HS group had higher drip loss of breast muscle than NT group ( p < 0.01). Broilers exposed to HS had lower total antioxidant capacity (T-AOC) in serum and jejunum, activities of total superoxide dismutase (T-SOD) in the jejunum, glutathione peroxidase (GSH-Px) in the thigh muscle, duodenum, and jejunum; and catalase (CAT) in breast muscle, duodenum, and jejunum ( p < 0.05). Whereas the malondialdehyde (MDA) contents in breast muscle, duodenum, and jejunum was elevated by HS exposure ( p < 0.05). Moreover, the relative mRNA expression of Occludin and ZO-1 in the duodenum, Occludin, Claudin-1, Claudin-4, ZO-1, Mucin-2 in the jejunum, and the Claudin-4 and Mucin-2 in the ileum was down-regulated by HS exposure ( p < 0.05). The 16S rRNA sequencing results showed that the HS group increased the relative abundance of Anaerovorax in the cecum at the genus level ( p < 0.05). Cecal metabolomics analysis indicated 19 differential metabolites between the two groups ( p < 0.10, VIP >1). The Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis revealed that the differential metabolites mainly enriched in 10 signaling pathways such as the Citrate cycle (TCA cycle) ( p < 0.01). In summary, chronic HS exposure caused a decline of production performance, reduced antioxidant capacity, disrupted intestinal barrier function, and negatively affected cecal microbiota and metabolome in indigenous broilers.

Abstract:To investigate the effect of ammonia (NH3) and humidity on the immune response of broilers, broilers were exposed to 30 or 70 mg/kg atmospheric NH3 for 21 days. Additionally, birds were exposed to 35, 60, and 85% relative humidity (RH). The relative weights of lymphoid organs, serum total protein, serum globulin, serum albumin, serum lysozyme, proliferation index of peripheral blood lymphocytes, and splenic cytokine gene expression were determined. Exposure to 70 mg/kg NH3 decreased the relative weight of the spleen during the experimental period, serum lysozyme concentration in the first and second weeks, and serum globulin concentration in the third week. The proliferation of peripheral blood lymphocytes was reduced. High levels of NH3 caused increase in IL-1β gene expression in the experimental period and IL-4 gene expression in the first week. Birds exposed to 85% RH had lower thymus and bursa of Fabricius weights in the third week and serum lysozyme concentration in the first week; IL-1β and IL-4 expressions were higher in the second and third weeks and first and second weeks, respectively, than in birds exposed to 60% RH. IL-4 expression was lower during the first week, and IL-1β expression was higher during the second week with 35% RH than with 60% RH. In conclusion, high NH3 level in the poultry house suppressed the immune response of broiler chickens. Neither high nor low RH benefited the immune response of broilers. Furthermore, there was an interactive effect between NH3 and RH on the immune response of broilers.
