Abstract:Pea pods are by-products of the pea processing industries which are often disposed improperly but are rich reserves of nutrients. In this work, pea pod powder (PPP) was prepared and analysed for its nutritional, physical, functional and structural characteristics for food applications. Results showed that PPP contained 6.3% moisture, 5.2% ash, 3.5% crude fat, 13.3% crude protein, and 35.3% dietary fiber. Further, PPP exhibited 0.47 g/ml bulk density, 0.50 g/ml aerated bulk density, 0.62 g/ml tapped bulk density and had fair flowability as determined by Hausner's ratio and Carr's index. PPP also showed good functional characteristics with 3.24 g/g water absorption index, 7.9% water solubility index, 1.25 g/g oil absorption capacity, and 4.65% swelling power. Based on its excellent qualities, PPP was used to prepare cookies which were analyzed for its structural and spectral characteristics. The X-ray diffraction pattern of PPP and cookies revealed that the crystalline region in the latter remained intact. The FTIR spectra showed the presence of different functional groups in PPP and cookies. The study showed that PPP could be utilized as a beneficial ingredient in dietetic products such as baked goods due to its good water-holding capacity, oil-holding capacity and dietary fiber content.

Abstract:Grass pea (Lathyrus sativus L.) is a nutritious legume crop well-adapted to fragile agro-ecosystems that can survive under challenging climatic conditions. The cultivation of grass pea faces stigma primarily due to the presence of β-N-Oxalyl-L-α, β-diaminopropionic acid (β-ODAP), which is associated with a risk of inducing neurolathyrism upon prolonged consumption of its grains as a staple diet. The grass pea improvement program of the International Center for Agricultural Research in the Dry Areas (ICARDA) aims to reduce β-ODAP content to a safe level along with improving yield potential and nutritional quality of grass pea. In this study, 183 germplasm accessions representing 13 different Lathyrus species and 11 L. sativus breeding lines were evaluated for β-ODAP content based on Rao protocol and mineral concentration using ICP-OES. Significant variability was observed among the accessions for the studied traits. The results showed low β-ODAP content and high mineral concentration in 25 accessions of crop wild relatives, which included L. cicera, L. ochrus, and L. cassius, with one accession IG65277 of L. cassius, in addition to two lines, IG117034 and ACC1335, of L. sativus having very low β-ODAP content. Furthermore, some accessions of L. pseudocicera, L. aphaca, L. cicera, L. marmoratus, L. gorgoni, and L. tingitanus also showed low β-ODAP content. The results showed significant positive correlations among different trait combinations, viz., K and P (r = 0.193 ***), K and Fe (r = 0.177 ***), Mn and Fe (r = 0.210 ***), Mn and Se (r = 0.137 ***), β-ODAP and Mg (r = 0.158 **), and β-ODAP and Ca (r = 0.140 **). L. cicera, L. ochrus, and L. cassius were identified as a great source for improving the mineral concentration and reducing β-ODAP content in the cultivated grass pea.

Abstract:ODAP (N-oxalyl-L-2,3-diaminopropionic acid) is present in the seeds of grass pea. In this study, variation of total ODAP accumulation in leaves throughout the crop growth starting from 40 days after sowing to maturity, and the distribution pattern of ODAP in different plant parts including the seeds at the mature stage was analyzed. Five grass pea accessions were evaluated for two subsequent growing seasons in one location of ICARDA, Aleppo (Syria). The results found that the rate of accumulation of total ODAP varied during plant development. Increased rates of synthesis were noticed in young leaves of grass pea. The highest total ODAP content in leaves was noted in the early growth stage (40–50 days after sowing). Mean total ODAP content in leaves ranged from 0.17 to 0.96 percent during 2010–2011 and from 0.19 to 1.28 percent during 2011–2012. During maturity, the total ODAP content was lowest in the seeds than in leaves, stems, pod cover, seed coat, and cotyledons. The ranges of total ODAP content were 0.13 (seed)–0.34 (stem), 0.20 (seed)–1.01 (leaf), 0.22 (seed)–0.62 (leaf), 0.21 (seed)–0.66 (leaf), and 0.21 (seed)–0.78 (leaf) percent in B387, B222, B390, Bio-520, and B587 accessions, respectively, during maturity. The results indicated that the rate of accumulation and synthesis of total ODAP varied during the plant lifespan. The lowest total ODAP content of leaves was observed after 130 days of sowing. The lower total ODAP content after the early vegetative stage of grass pea plants makes them suitable as a feed.

The Comparative Effect of Lactic Acid Fermentation and Germination on the Levels of Neurotoxin, Anti-Nutrients, and Nutritional Attributes of Sweet Blue Pea (Lathyrus sativus L.)

Abstract:Grass pea (Lathyrus sativus L.), an indigenous legume of the subcontinental region, is a promising source of protein and other nutrients of health significance. Contrarily, a high amount of β-N-oxalyl-l-α,β-diaminopropionic acid (β-ODAP) and other anti-nutrients limits its wider acceptability as healthier substitute to protein of animal and plant origin. This study was aimed at investigating the effect of different processing techniques, viz. soaking, boiling, germination, and fermentation, to improve the nutrient-delivering potential of grass pea lentil and to mitigate its anti-nutrient and toxicant burden. The results presented the significant (p < 0.05) effect of germination on increasing the protein and fiber content of L. sativus from 22.6 to 30.7% and 15.1 to 19.4%, respectively. Likewise, germination reduced the total carbohydrate content of the grass pea from 59.1 to 46%. The highest rate of reduction in phytic acid (91%) and β-ODAP (37%) were observed in germinated grass pea powder, whereas fermentation anticipated an 89% reduction in tannin content. The lactic acid fermentation of grass pea increased the concentration of calcium, iron, and zinc from 4020 to 5100 mg/100 g, 3.97 to 4.35 mg/100 g, and 3.52 to 4.97 mg/100 g, respectively. The results suggest that fermentation and germination significantly (p < 0.05) improve the concentration of essential amino acids including threonine, leucine, histidine, tryptophan, and lysine in L. sativus powder. This study proposes lactic acid fermentation and germination as safer techniques to improve the nutrient-delivering potential of L. sativus and suggests processed powders of the legume as a cost-effective alternative to existing plant proteins.

Abstract:Abstract Background Grass pea ( Lathyrus sativus ) is an underutilised crop with high tolerance to drought and flooding stress and potential for maintaining food and nutritional security in the face of climate change. The presence of the neurotoxin β-L-oxalyl-2,3-diaminopropionic acid (β-L-ODAP) in tissues of the plant has limited its adoption as a staple crop. To assist in the detection of material with very low neurotoxin toxin levels, we have developed two novel methods to assay ODAP. The first, a version of a widely used spectrophotometric assay, modified for increased throughput, permits rapid screening of large populations of germplasm for low toxin lines and the second is a novel, mass spectrometric procedure to detect very small quantities of ODAP for research purposes and characterisation of new varieties. Results A plate assay, based on an established spectrophotometric method enabling high-throughput ODAP measurements, is described. In addition, we describe a novel liquid chromatography mass spectrometry (LCMS)-based method for β-L-ODAP-quantification. This method utilises an internal standard (di- 13 C-labelled β-L-ODAP) allowing accurate quantification of β-L-ODAP in grass pea tissue samples. The synthesis of this standard is also described. The two methods are compared; the spectrophotometric assay lacked sensitivity and detected ODAP-like absorbance in chickpea and pea whereas the LCMS method did not detect any β-L-ODAP in these species. The LCMS method was also used to quantify β-L-ODAP accurately in different tissues of grass pea. Conclusions The plate-based spectrophotometric assay allows quantification of total ODAP in large numbers of samples, but its low sensitivity and inability to differentiate α- and β-L-ODAP limit its usefulness for accurate quantification in low-ODAP samples. Coupled to the use of a stable isotope internal standard with LCMS that allows accurate quantification of β-L-ODAP in grass pea samples with high sensitivity, these methods permit the identification and characterisation of grass pea lines with a very low ODAP content. The LCMS method is offered as a new ‘gold standard’ for β-L-ODAP quantification, especially for the validation of existing and novel low- and/or zero-β-L-ODAP genotypes.

Abstract:Grass pea (Lathyrus sativus L.) is an important legume commonly grown in arid and semi-arid regions. This protein-rich legume performs well even under harsh environmental conditions and is considered to be a strategic famine food in developing countries. Unfortunately, its potential usage is greatly limited as a result of the presence of antinutritional factors, including the neuroexcitatory amino acid β-N-oxalyl-l-α,β-diaminopropionic acid (β-ODAP) and protease inhibitors. β-ODAP is responsible for a neurodegenerative syndrome that results in the paralysis of lower limbs, while protease inhibitors affect protein digestibility, resulting in reduced growth. Concerted research efforts have led to development of grass pea cultivars with reduced β-ODAP content. In contrast, very little information is available on the protease inhibitors of L. sativus. In this study, we have conducted biochemical characterization of 51 L. sativus accessions originating from different geographical regions. Sodium dodecyl sulfate polyacrylamide gel electrophoresis analyses of seed globulins and prolamins revealed striking similarity in their protein profile, although geographic-specific variations in profiles was also evident. Measurement of Bowman-Birk chymotrypsin inhibitor (BBi) and Kunitz trypsin inhibitor (KTi) activities in accessions revealed striking differences among them. Amino acid sequence alignment of grass pea BBi and KTi revealed significant homology to protease inhibitors from several legumes. Real-time polymerase chain reaction analysis demonstrated high-level expression of BBi and KTi in dry seeds and weak expression in other organs. Our study demonstrates substantial variation in BBi and KTi among grass pea accessions that could be exploited in breeding programs for the development of grass pea lines that are devoid of these antinutritional factors.

Abstract:Grass pea (Lathyrus sativus L.) cultivation is limited because of the presence in seeds and tissues of the nonprotein amino acid β-N-oxalyl-L-α,β-diaminopropionic acid (β-ODAP), a neurotoxin that can cause lathyrism in humans. Seven grass pea genotypes differing in seed β-ODAP concentration were grown in pots at three levels of water availability to follow changes in the concentration and amount of β-ODAP in leaves and pods and seeds. The concentration and amount of β-ODAP decreased in leaves in early reproductive development and in pods as they matured, while water stress increased β-ODAP concentration in leaves and pods at these stages. The net amount of β-ODAP in leaves and pods at early podding was positively associated with seed β-ODAP concentration at maturity. We conclude that variation among genotypes in seed β-ODAP concentration results from variation in net accumulation of β-ODAP in leaves and pods during vegetative and early reproductive development.
Accumulation Pattern of Toxin Β-Odap During Lifespan and Effect of Nutrient Elements on Β-Odap Content in Lathyrus Sativus Seedlings

Abstract:In order to assess the influence of nutrient elements on the accumulation of β-N-oxalyl-L-α, β-diaminopropionic acid (β-ODAP; the probable cause of lathyrism) in Lathyrus sativus L. (grass pea), it was first examined under field conditions during the lifespan of a grass pea plant using high-performance liquid chromatography (HPLC). β-ODAP mainly accumulated in young seedlings, in developing and mature seeds and in young leaves, especially in young seedlings. In contrast, all mature leaves, roots, and stems showed a low level of β-ODAP. The β-ODAP accumulation pattern in seedlings grown in a nutrient-deficient solution was highest (3·57 mg/g) in shoots at 7 d growing in the nitrogen-deficient solution and higher compared to the control (2·31 mg/g) in zinc-, calcium-, phosphorus- and molybdenum-deficient shoots (P<0·05). The contents of β-ODAP in seedlings growing in other element-deficient solutions were similar to controls. When the content of β-ODAP in grass pea seedlings fertilized with different forms of organic nitrogen was assayed the results indicated that amino acids such as glutamine and serine, as well as nucleotide nitrogen, all significantly enhanced the accumulation of β-ODAP in young seedlings relative to controls (P<0·05). Taken together, these data suggest that β-ODAP accumulation in grass pea might be related to the level of total free nitrogenous compounds and that nitrogen and phosphate may be the crucial nutrient factors influencing β-ODAP content under field conditions. Thus, the application of appropriate nitrogen and phosphorus fertilizers to the soil could decrease the content of β-ODAP in the seeds and leaves of grass pea.

Abstract:Protein is one of the most abundant substances in plants and plays a major role in human health hence standardization of its analytical quantification method is essential. Various methods for protein quantification exist, such as Kjeldahl, Bradford, Lowry, bicinchoninic acid assay (BCA), Biuret, and total amino acid content methods. These methods are widely applied; however, the development of the rapid and efficient method is the need of the time hence the objective of this research was to analyze and comparing compare the modification of the Kjeldahl method for the determination of protein content in oilseed crops. The study was performed to improve the sample preparation method (processing and digestion) for protein quantification. Generally, the method initially requires homogenization of grains to a fine flour, which involves time and increases the risk of sample cross-contamination and partial loss of oil from the sample during grinding. Moreover at times, it becomes challenging to homogenize oil seeds to fine flour due to high oil content. However, in the present research, the whole grain was digested in place of grounded flour to accomplish quick protein quantification and compared it with the flour matrix of different oil seeds. To further reduce the digestion time and avoid frothing, we have used the modified digestion mixture. The developed method was statistically validated using analysis of variance (ANOVA), Pearson correlation reliability test, paired T -test, and different types of plot analysis. The validation of the sample preparation method in protein quantification demonstrated non-significant differences that the protein content from whole grain of all the five oilseed crops shows 100% non-significant results compared with the flour matrix in both the digestion mixtures. The developed novel method could be used to prepare the sample for protein analysis and reduces the overall analysis time while ensuring the accuracy of the results.

Abstract:Grass pea has the potential to become a miracle crop if the stigma attached to it as a toxic plant is ignored. In light of the following, we conducted transcriptome analyses on the high and low ODAP-containing cultivars i.e., Nirmal and Bidhan respectively in both normal and salt stress conditions. In this study, genes that work upstream and downstream to β-ODAP have been found. Among these genes, AAO3 and ACL5 were related to ABA and polyamine biosynthesis, showing the relevance of ABA and polyamines in boosting the β-ODAP content in Nirmal. Elevated β-ODAP levels in salt stress-treated Bidhan may have evolved tolerance by positively regulating the expression of genes involved in phenylpropanoid and jasmonic acid biosynthesis. Although the concentration of β-ODAP in Bidhan increased under salt stress, it was lower than in stress-treated Nirmal. Despite this, the expression of stress-related genes that work downstream to β-ODAP was found higher in stress-treated Bidhan. This could be because stress-treated Nirmal has lower GSH, proline, and higher H 2 O 2 , resulting in the development of severe oxidative stress. Overall, our research not only identified new genes linked with β-ODAP, but also revealed the molecular mechanism by which a low β-ODAP-containing cultivar developed tolerance against salinity stress.

Abstract:Fermentation of pure teff (Eragrostis teff), pure grass-pea (Lathyrus sativus), and their mixtures, 9:1 and 8:2 (teff/grass-pea) has been done at two temperatures (room temperature and 35 degrees C) in duplicate using the strains of Lactobacillus plantarum, for bacterial fermentation, and Aspergillus oryzae and Rhizopus oligosporus in succession for solid-state fungal fermentation as inocula. In addition, the natural or spontaneous and back-slopping methods of bacterial fermentation have been done on the above four substrate groups. The pH and essential amino acid profiles of the different fermentation processes were compared. The back-slopping in teff at a temperature of 35 degrees C gave the sharpest pH drop. All fermentations done at 35 degrees C showed a steeper slope in their pH versus time plot compared to their room temperature counterpart. Fungal fermentation gave an improved amino acid profile for the essential ones in all of the substrate groups, except in pure grass-pea. Fermented teff/grass-pea (8:2) in this fungal fermentation has been found to be quite comparable in essential amino acid profile to an ideal reference protein recommended for children of 2-5 years of age. None of the bacterial fermentations produced a net change in their essential amino acid profile in any of the substrate groups investigated. Solid state fungal fermentation on pure grass-pea using the fungal strains R. oligosporous and A. oryzae in succession has shown that the neurotoxin beta-N-oxalyl-alpha,beta-diaminopropionic acid (beta-ODAP) in grass-pea has been removed by 80% on average for the high-toxin variety and by up to 97% for the low-toxin variety as determined by an improved chromatographic method with bioelectrochemical detection coupled on-line with refractive index detection.

Abstract:Pigeon pea, a protein-rich legume with low protein digestibility (PD) due to its high polyphenol content and other antinutritional factors (ANFs). Consequently, processing methods are crucial to improve PD. We investigated the effects of thermal treatments (cooking, hydrothermal, autoclaving, infrared rays) treatments and germination on modulation of PD, its properties and association with ANFs in two distinct genotypes based on polyphenol content: high (Pusa Arhar 2018-4) and low (ICP-1452). Treatments improved in vitro PD and essential amino acid content, with autoclaving showing significantly higher PD (ICP-1452: 90.4%, Pusa-Arhar 2018-4: 84.32%) ascribed to disruption of tight protein matrices. Significant increase in β-turn, reduction in protein: starch, protein: polyphenol interactions as well as breakdown of storage proteins revealed by the analysis of protein structural properties. This study suggests thermal treatments, particularly autoclaving, can enhance pigeon pea protein's nutritional quality for its utilization as a new ingredient in development of healthy foods.

Abstract:BACKGROUND Peas provide an excellent plant protein resource for human diets, but their proteins are less readily digestible than animal proteins. To identify the relationship between composition and in vitro digestibility of pea protein, eight pea varieties with a wide range of protein content (157.3-272.7 g kg(-1)) were determined for the proportion of albumins and globulins, their compositions using sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and in vitro protein digestibility (IVPD) before and after heat treatment using a multi-enzyme (trypsin, chymotrypsin and peptidase) method. RESULTS The proportion of albumins based on total seed protein content decreased from 229 to 147 g kg(-1) as seed protein content increased from 157.3 to 272.7 g kg(-1), while the proportion of globulins increased from 483 to 590 g kg(-1). The IVPDs of eight raw pea seeds were 79.9-83.5%, with significant varietal variations, and those were improved to 85.9-86.8% by cooking. Albumins, including (pea albumins 2) PA2, trypsin inhibitor, lectin and lipoxygenase, were identified as proteolytic resistant proteins. Globulins were mostly digested by protease treatment after heating. CONCLUSION The quantitative ratio of albumins and globulins, and the quantitative variations of albumin protein components, including lipoxygenase, PA2, lectins and trypsin inhibitors, appear to influence the protein digestibility of both raw and cooked pea seeds.

Abstract:Macrotyloma uniflorum is a salubrious but underutilized legume mainly consumed in semi-arid zones of Africa, Australia and India. Various antinutritional factors- phytates, oxalates and oligosaccharides- has limited its consumption. Current work describes the influence of various thermal processing technologies - autoclaving, microwave, micronization and extrusion- on antinutritional profile, phenolic acid profile and protein digestibility of two selected varieties. Significant (p ≤ 0.05) reduction in antinutritional content was observed for horse gram grains processed using various thermal technologies. Among all processing technologies extrusion caused marked degradation in antinutritional components. The reduction in oxalic acid, trypsin inhibitor, phytates and tannins ranged from 33 to 87 %, 77-82%, 33-60% and 51-66% respectively. Further, the decline in content of various oligosaccharides viz. raffinose, stachyose and verbascose varied from 36 to 61 %, 25-49% and 30-74% respectively for both the varieties. Although extrusion caused significant (p ≤ 0.05) decrease in the essential amino acid index and protein efficiency ratio, simultaneous increased was observed in biological value. The processing had significant (p ≤ 0.05) impact on grain antinutritional content and also retained its substantial functional properties. This establishes the utility of grain and promote the introduction of these new grains and enlarge the market of novel healthy foods.
Nutritional composition, bioactive compounds and antioxidant potentiality of some indigenous vegetables consumed in Bangladesh

Abstract:Food and nutrition security are essential for accomplishing sustainable development goals, and a growing population requires various food sources to address hunger and nutrition. Indigenous vegetables are nutritious, healthy, and adaptable to local conditions, making them a potential food and nutrition security resource. Thus, this study aimed to evaluate the nutritional richness of indigenous vegetables by comparing the nutritional content of commonly grown indigenous vegetables like Chenopodium album (Bathua), Coccinia grandis (Telakucha), Amaranthus viridis (Shaknotey), Moringa oleifera (Shojne), Alternanthera philoxeroides (Malancha), and Xanthium strumarium (Ghagra) to BARI Lalshak-1 ( Amaranthus tricolor ). Shojne violet had the highest lightness (L*), directions (b*), and chroma (c*) of 43.50, 23.69, and 25.02, respectively and telakucha had the highest luminosity (h°) of 114.11. Shaknotey had more moisture (88.97 %) than bathua green (66.62 %) or ghagra (72.55 %). Ghagra had the most chlorophyll a (1.11 mg/g), b (0.65 mg/g), total chlorophyll (2.04 mg/g), ascorbic acid (22.0 mg/100 g), and flavonoids (50.1 mgQE/100 g). Bathua red had significant levels of anthocyanin, carotenoids, and strongest antioxidant activity (IC50 value): 42.9 μg/g, 0.24 mg/g, and 12.4 μg/mL, respectively. Shojne green and telakucha were found rich in phenolic content (136.0 mgGAE/100 g) and β-carotene (2.05 mg/100 g), respectively. Moreover, the studied indigenous vegetables were rich in minerals, potassium, calcium, magnesium, and iron, with a very high K content (49.4–79.4 mg/g). Correlation matrix and PCA showed that ghagra, shojne, and bathua species were nutritionally superior to others. Therefore, the current findings suggest that ghara, shojne and bathua can become a nutrient-rich indigenous vegetable and improve human nutrition.

Abstract:A simple method is proposed for the simultaneous analysis of the neurotoxin β- N -oxalyl-α, β-diaminopropionic acid (β-ODAP) and its isomer α-ODAP, as well as other free amino acids, in Lathyrus sativus extract based on reversed-phase (C 18 column) high-performance liquid chromatography (HPLC) of their para -nitro-benzyloxycarbonyl chloride (PNZ-Cl) derivatives. Samples were derivatized in 0.5 M NaHCO 3 within 2 min at ambient temperature by adding 50 mM PNZ-Cl in acetonitrile. The PNZ-derivatives were quantified by their UV-absorption at 260 nm. Detection limits were approximately 8 ng mL −1 for both α-, β-isomers, 9 ng mL −1 for homoarginine at a signal-to-noise ratio of 3:1. The coefficients of determination were 0.9967–0.9991. The method was validated by the determination of β-, α-ODAP and homoarginine contents in ethanol-water (30:70, v/v) extracts from the seed and foliage of Lathyrus sativus .

Abstract:Aims: Bambara groundnut is considered as underutilize and commercially forgotten legume in Malaysia. Few studies abroad showed interesting potential on its phytochemical and nutritional contents. This study was carried out to investigate and compare the nutritional, antinutritional, free radical scavenging assay, total phenolic content and fatty acids composition of Bambara groundnut (Vigna subterranea) with comparison with two others commercial legumes (Vigna spp.) red bean (V. angularis) and black-eyed pea (V. unguiculata) in Malaysia. Study Design: Each sample was extracted three times (n=3) for the free radical scavenging assay, total phenolic content, fatty acids composition and antinutritional content. All the data were analysed using ANOVA and Tukey Pairwise tests. Place and Duration of Study: Phytochemistry Laboratory, Kompleks MyGene Bank, Malaysian Agricultural Research and Development Institute (MARDI), Serdang, Selangor, Malaysia from April to December 2022. Methodology: Bambara groundnut (BGN), red bean (RB) and black-eyed pea (BEP) were purchased from various local market situated in the northern state of Peninsular Malaysia. Each sample was dried in the oven at 40°C for 3 days. Samples were grounded and its proximate and minerals content were determined. Each sample was extracted for the free radical scavenging assay, total phenolic content, oxalates (total, soluble and insoluble) content using HPLC and fatty acids composition using GCMS. Results: Bambara groundnut (BGN) was found to have higher total carbohydrate content (66.0 ± 0.01 g/100g) compared with RB and BEP (P<0.05) hence provides more energy (406.67 ± 1.25 kcal/100g) (14% higher than red bean and black-eyed peas). Further analysis on its mineral content revealed the legume is rich in potassium (24% higher than red bean and black-eyed peas) suggesting its potential to regulate body fluid and muscle contractions. The bambara groundnut’s seed oil is rich in fatty acids; omega 3 and 6 (palmitic acid, α-linoleic acid and linolenic acid). BGN also have higher phenolic content (87.3 ± 2.05 g/100g) compared to RB and BEP (P<0.05). The free radical scavenging assay showed moderate antioxidant activity with inhibition concentration (IC50) value of 0.57 ± 0.04 mg/ml (standard: ascorbic acid = 0.06 mg/ml). The underutilize legume also had undetectable oxalate content; hence it is safe for human consumption (lethal dose 660 mg/kg body weight). Conclusion: The potential findings on this legume should elevate the importance of this crop to be commercialised locally and for the future crop that will lead to achieve food security particularly in Malaysia.
Impact of experimental thermal processing of artificially contaminated pea products on ochratoxin A and phomopsin A

Abstract:Abstract Fungi of Aspergillus and Penicillium genus can infect peas ( Pisum sativum ), leading to a contamination with the nephrotoxic and carcinogenic ochratoxin A (OTA). Under unfavourable conditions, a fungus primarily found on lupines, Diapothe toxica , may also grow on peas and produce the hepatotoxic phomopsin A (PHOA). To study the effect of processing on OTA and PHOA content, two model products—wheat/rye-mixed bread with pea flour addition and pea pasta—were manufactured at small-business scale from artificially contaminated pea flour. The decrease of OTA and PHOA contents were monitored along the production process as indicators for toxin transformation. Pea bread dough was subjected to proofing for 30–40 min at 32 °C and baked at 250 °C to 230 °C for 40 min. OTA content (LODs < 0.1 μg/kg) showed a reduction in the bread crust (initially 17.0 μg/kg) to 88% and no reduction in the crumb (110%). For PHOA (LODs < 3.6 μg/kg), a decrease to approximately 21% occurred in the bread crust (initially 12.5 μg/kg), whilst for crumb, a less intense decrease to 91% was found. Pea pasta prepared with two toxin levels was extruded at room temperature, dried and cooked for 8 min in boiling water. In pea pasta, OTA was reduced from 29.8 to 13.9 μg/kg by 22% each after cooking, whilst 15% and 10% of the initial toxin amounts were found in the cooking water, respectively. For PHOA, 60% and 78% of initially 14.3 μg/kg and 7.21 μg/kg remained in the cooked pasta. As only the decrease of the initial content was measured and no specific degradation products could be detected, further research is needed to characterise potential transformation products. Heat treatment reduces the initial PHOA content stronger than the OTA content during pasta cooking and bread making. However, significant amounts of both toxins would remain in the final products.

Abstract:Background: Significant amounts of nutrients, including dietary fibers, proteins, minerals, and vitamins are present in legumes, but the presence of anti-nutritional factors (ANFs) like phytic acid, tannins, and enzyme inhibitors impact the consumption of legumes and nutrient availability. In this research, the effect of a physical process (sonication or precooking) and fermentation with Lactobacillus plantarum and Pediococcus acidilactici on the ANFs of some legumes was evaluated. Results: Total phenolic content was significantly (P < 0.05) reduced for modified and fermented substrates compared with non-fermented controls. Trypsin inhibitory activity (TIA) was reduced significantly for all substrates except for unsonicated soybean and lentils fermented with L. plantarum and P. acidilactici. When physical processing was done, there was a decrease in TIA for all the substrate. Phytic acid content decreased for physically modified soybean and lentil but not significantly for green pea. Even though there was a decrease in ANFs, there was no significant change in in vitro protein digestibility for all substrates except for unsonicated L. plantarum fermented soybean flour and precooked L. plantarum fermented lentil. Similarly, there was a change in amino acid content when physically modified and fermented. Conclusion: Both modified and unmodified soybean flour, green pea flour, and lentil flour supported the growth of L. plantarum and P. acidilactici. The fermentation of this physically processed legume and pulse flours influenced the non-nutritive compounds, thereby potentially improving nutritional quality and usage. © 2021 The Authors. A Comparison of Phenolic, Flavonoid, and Amino Acid Compositions and In Vitro Antioxidant and Neuroprotective Activities in Thai Plant Protein Extracts

Abstract:The leaves of mulberry, Azolla spp., sunflower sprouts, cashew nut, and mung bean are considered rich sources of plant protein with high levels of branched-chain amino acids. Furthermore, they contain beneficial phytochemicals such as antioxidants and anti-inflammatory agents. Additionally, there are reports suggesting that an adequate consumption of amino acids can reduce nerve cell damage, delay the onset of memory impairment, and improve sleep quality. In this study, protein isolates were prepared from the leaves of mulberry, Azolla spp., sunflower sprouts, cashew nut, and mung bean. The amino acid profile, dietary fiber content, phenolic content, and flavonoid content were evaluated. Pharmacological properties, such as antioxidant, anticholinesterase, monoamine oxidase, and γ-aminobutyric acid transaminase (GABA-T) activities, were also assessed. This study found that concentrated protein from mung beans has a higher quantity of essential amino acids (52,161 mg/100 g protein) compared to concentrated protein from sunflower sprouts (47,386 mg/100 g protein), Azolla spp. (42,097 mg/100 g protein), cashew nut (26,710 mg/100 g protein), and mulberry leaves (8931 mg/100 g protein). The dietary fiber content ranged from 0.90% to 3.24%, while the phenolic content and flavonoid content ranged from 0.25 to 2.29 mg/g and 0.01 to 2.01 mg/g of sample, respectively. Sunflower sprout protein isolates exhibited the highest levels of dietary fiber (3.24%), phenolic content (2.292 ± 0.082 mg of GAE/g), and flavonoids (2.014 mg quercetin/g of sample). The biological efficacy evaluation found that concentrated protein extract from sunflower sprouts has the highest antioxidant activity; the percentages of inhibition of 1,1-diphenyl-2-picrylhydrazyl radical (DPPH) and 2,2′-azino-bis-(3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) radical were 20.503 ± 0.288% and 18.496 ± 0.105%, respectively. Five plant-based proteins exhibited a potent inhibition of acetylcholinesterase (AChE) enzyme activity, monoamine oxidase (MAO) inhibition, and GABA-T ranging from 3.42% to 24.62%, 6.14% to 20.16%, and 2.03% to 21.99%, respectively. These findings suggest that these plant protein extracts can be used as natural resources for developing food supplements with neuroprotective activity.
