Abstract:Objective: To study at the single-cell level whether a sperm's motility and morphology parameters reflect its DNA integrity, and to establish a set of quantitative criteria for selecting single sperm with high DNA integrity. Design: Prospective study. Setting: In vitro fertilization center and university laboratories. Patient(s): Male patients undergoing infertility treatments. Intervention(s): None. Main Outcome Measure(s): The motility and morphology parameters of each sperm were measured with the use of computer vision algorithms. The sperm was then aspirated and transferred for DNA fragmentation measurement by single-cell gel electrophoresis (comet assay). Result(s): We adapted the World Health Organization criteria, which were originally defined for semen analysis, and established a set of quantitative criteria for single-sperm selection in intracytoplasmic sperm injection. Sperm satisfying the criteria had significantly lower DNA fragmentation levels than the sample population. Both normal motility and normal morphology were required for a sperm to have low DNA fragmentation. The quantitative criteria were integrated into a software program for sperm selection. In blind tests in which our software and three embryologists selected sperm from the same patient samples, our software outperformed the embryologists and selected sperm with the highest DNA integrity. Conclusion(s): At the single-cell level, a sperm's motility and morphology parameters reflect its DNA integrity. The developed technique and criteria hold the potential to mitigate the risk factor of sperm DNA fragmentation in intracytoplasmic sperm injection. (Fertil Steril (R) 2021;116:1308-18. Abstract:The increasing infertility rate has become a worrying global challenge in recent years. According to the report of the World Health Organization, the male factor is responsible for over half of infertility cases, which includes the lack of desirable characteristics in sperm motility, morphology, and DNA integrity. In recent years, it has been shown that clinical methods including density gradient centrifugation cause damage to sperm DNA and besides being invasive, they are costly and time-consuming. In contrast, microfluidics has been used as a promising and non-invasive approach to manipulate biological cells. Here, by using the microvortices created by the oscillation of the bubbles caused by the bulk acoustic waves, we were able to trap sperms with less motility. In contrast, the highly motile sperms overcame the force of the microvortices and were guided to the outlet pool by following the channel boundaries. As a result, over 50% and 44% improvement in sperm progressive motility and viability, respectively, as well as 40% improvement in DNA integrity, were observed in the analysis of sperms retrieved from the output pool. In addition to being fast and non-invasive, the proposed device benefits from an easy method for sperms retrieval and does not require any preprocessing of the raw sperm sample.

Abstract:Microfluidics represent a quality sperm selection technique. Human couples fail to conceive and this is so in a significant population of animals worldwide. Defects in male counterpart lead to failure of conception so are outcomes of assisted reproduction affected by quality of sperm. Microfluidics, deals with minute volumes (μL) of liquids run in small-scale microchannel networks in the form of laminar flow streamlines. Microfluidic sperm selection designs have been developed in chip formats, mimicking in vivo situations. Here sperms are selected and analyzed based on motility and sperm behavioral properties. Compared to conventional sperm selection methods, this selection method enables to produce high-quality motile sperm cells possessing non-damaged or least damaged DNA, achieve greater success of insemination in bovines, and achieve enhanced pregnancy rates and live births in assisted reproduction-in vitro fertilization (IVF) and intracytoplasmic sperm injection (ICSI). Besides, the concentration of sperm available to oocyte can be controlled by regulating the flow rate in microfluidic chips. The challenges in this technology are commercialization of chips, development of fully functional species-specific microfluidic tools, limited number of studies available in literature, and need of thorough understanding in reproductive physiology of domestic animals. In conclusion, incorporation of microfluidic system in assisted reproduction for sperm selection may promise a great success in IVF and ICSI outcomes. Future prospectives are to make this technology more superior and need to modify chip designs which is cost effective and species specific and ready for commercialization. Comprehensive studies in animal species are needed to be carried out for wider application of microfluidic sperm selection in in vitro procedures.
High-Throughput Sperm Sorting Microfluidic Device for Livestock’s Sperm Motility Enhancement

Abstract:Sperm sorting by microfluidic chips has flourished lately. Most traditional chips are used to collect upstream-swimming top-tier sperms, which numbers are 10 5 ~10 6 sperm cells and 1~10 μL. However, the spec of traditional chips could only reach mankind in-vitro fertilization (IVF). As a result, a device is proposed that conducts a milliliter-level high-throughput sorting process and has a passive sorting property with a stable flow field. Furthermore, the chip is manufactured using a low-cost technique. The results demonstrate that sperm motility and progressive motility (PR) have increased by more than 20% and 15%, respectively, and that the number of sperms is 5*10 7 ~10 8 cells. Additionally, the chip is discussed in several experimental environments. We believe that the motility sorting device is applicable for livestock.
Integration of Sperm Motility and Chemotaxis Screening with a Microchannel-Based Device

Abstract:Objective Sperm screening is an essential step in IVF procedures. The swim-up method, an assay on sperm motility, is used clinically to select the ideal sperm for subsequent manipulation. However, additional parameters, including acrosome reaction capability, chemotaxis, and thermotaxis are also important indicators of mammalian sperm health. To monitor both sperm motility and chemotaxis simultaneously during sperm screening, we designed and constructed a microdevice comprising a straight channel connected with a bi-branch channel that mimics the mammalian female reproductive tract. Methods The width and length of the straight channel were optimized to select the motile sperm. Cumulus cells were selectively cultured in the bi-branch channel to generate a chemoattractant-forming chemical gradient. Sperm chemotaxis was represented by the ratio of the sperm swimming towards different branches. Results The percentage of motile sperm was improved from ( 58. 5 ± 3. 8 ) % to ( 82. 6±2.9)% by a straight channel 7 mm in length and 1 mm in width. About 10% of sperm were found chemotactically responsive in our experiment, which is consistent with previous studies. Conclusion The combined evaluation of both sperm motility and chemotaxis was achieved for the first time, and the motile and chemotactically responsive sperm can be easily enriched on a lab-on-a-chip device to improve IVF outcome. Abstract:Since male abnormality has now been reported to account at least thirty percent of couples with fertility problem. Therefore, distinguishing and classification of sperm has become more and more important, especially when Assisted Reproductive Technology (ART) like in vitro fertilization (IVF) is involved. However, traditional sperm sorting methods all requires manual selection, which is tedious and often cause sperm damage during the screen process. Although, new design from our recent study, seemed to point for a possibility to make these kind of MISS device capable of separating sperms into various mobility sub-class. In addition, the Hoechst 33258 and propidium iodide double staining results, also confirmed that the most motile and health sperm is indeed able to leave the laminar stream and being sorted out at outlet-h, with least dead sperm or debris. Meanwhile, we observed the dead sperm had been dominate from clear red stains and live sperm are not as active as is corresponded with more diffused blue stains. In order to confirm the real sorting situation, we counted the amount of sperm in collected sample by flow cytometry and tried to establish the statistic model. Though the flow cytometry, we could detect the sperm fluorescent distribution and count the size of sperm population. Nevertheless, t-test with 95% confidence level also shown significant difference in live/dead sperm ratio. Therefore, from the combined result among flow cytometry, fluorescent stain, and the statistic model test, it is confirmed that live sperms were able to cross the laminar stream created by the device and reach the sorting chamber with corresponding range from non-motile sperms to debris, as well as from living to dead ones. It is concluded that the proposed method is cost effective and is possible to serve as a protocol for rapid sperm quantitative assessment.

Abstract:Sperm preparation is a necessary step in all methods of assisted reproductive technology. Here, we demonstrate the utility of a microfluidic spiral channel device for preparation of sperm for intrauterine insemination (IUI) directly from semen samples. After optimization of operating conditions, we use data from 130 washed semen samples to show that the microfluidic chip can retain 89% of all sperm cells and 90% of motile sperm cells, while removing 40% of the fluid volume and 82% of white blood cells. Then, we show that, when combined with a centrifugal wash, the microfluidic chip can be used in a protocol for sperm preparation. This protocol is capable of selecting 40–70% of motile sperm in 15 min. Compared to the clinical gold standard, density gradient centrifugation, this offers a higher sperm recovery, a gentler approach, and reduced reagent costs.

Abstract:In this paper, we propose a microfluidic device capable of generating a retarding flow field for the sorting and separation of human motile sperm in a high-throughput manner. The proposed sorting/separation process begins with a rapid flow field in a straight-flow zone to carry sperm into a sorting zone to maintain the sperm's mobility. The sorting zone consists of a diffuser-type sperm sorter to differentiate sperm with different motilities based on the flowing upstream nature of human sperm in a retarding flow field. The dead sperm will then be separated from the live ones by passing through a dumbbell flow field to the outlet for disposal. The proposed flowing upstream sperm sorter (FUSS) is designed to imitate the selection mechanism found in the female body when sperm swim into the uterus. The experimental results demonstrate the utility of this device with regard to throughput (approximately 200 000 sperm per minute and a maximum of 200 million cells per mL), efficiency (90% of selected sperm are mobile), and the ability to select sperm with high motility (∼20% of sperm with a velocity exceeding 120 μm s-1). The proposed device is suitable for intrauterine insemination as well as in vitro fertilization thanks to the highly efficient sorting process not interfering with the natural function and energy resource of human sperm.

Abstract:The same sperm selection techniques in assisted reproduction clinics have remained largely unchanged despite their weaknesses. Recently, microfluidic devices have emerged as a novel methodology that facilitates the sperm selection process with promising results. A prospective case-control study was conducted in two phases: 100 samples were used to compare the microfluidic device with Density Gradient, and another 100 samples were used to compare the device with the Swim-up. In the initial phase, a significant enhancement in progressive motility, total progressive motile sperm count, vitality, morphology, and sperm DNA fragmentation were obtained for the microfluidic group compared to Density Gradient. Nevertheless, no statistically significant differences were observed in sperm concentration and chromatin structure stability. In the subsequent phase, the microfluidic group exhibited significant increases in sperm concentration, total progressive motile sperm count, and vitality compared to Swim-up. However, non-significant differences were seen for progressive motility, morphology, DNA structure stability, and DNA fragmentation. Similar trends were observed when results were stratified into quartiles. In conclusion, in a comparison of microfluidics with standard techniques, an improvement in sperm quality parameters was observed for the microfluidic group. However, this improvement was not significant for all parameters.

Abstract:This paper proposes a selection and extraction strategy to acquire high quality sperms from a biomimetic microfluidic device capable of generating a dual gradient flow field for motile sperm sorting. The sorter consists of a straight flow zone, a rapidly expansion sector for sperm sorting by gradient up-stream flow, and a dumbbell channel for dead sperms separation at the outlet. A collateral runner is set at the front area of the expansion sector, but mostly kept in dry state without affecting the sorting process. At the end of sorting, a pressure was applied from both the inlet and outlet to flush the high quality sperms into the collateral runner for collection. Controllable numbers, ranging from 1 to 15000, of sperms can be collected with a motility (>60μm) ratio higher than 60%, suitable for in-vitro fertilization (IVF).
Sorting of Sperms with Reverse Progressive Characteristic May Provide Another Option for Acquiring Spermatozoa with Significant Improvement in Fertility Related Quality for Patients with Oligospermia

Abstract:Previous study had reported a sub-group from progressive motile sperm sub-population with characteristics often referred to as upstream swimming or reversed progressiveness [1]. However, only until recently this sub-group can be separated with minimum damage [2]. Therefore, through the use of a novel microfluidic biochip design, it is the purpose of current study to exam whether or not spermatozoa thus acquired could show improvements not only in sperm motility but also in DNA integrity for normal as well as oligospermia patents. Soring of sperms with reverse progressiveness characteristic was performed by the construction of a flow velocity gradient between 20-180 μm/sec within the flow channel of a diffusor type microfluidic system [3]. Thereafter, targeted sperms would include sperms that were able to withstand or swim against the forward flow stream, but not slower ones, ones with forward progressive characteristic, or debris. In this study, 17 males subjects were recruited from couples undergoing ART cycles in Taipei Medical University Hospital, pre-approval by Joint IRB of Taipei Medical University (TMU-JIRB: 201209014). Samples were grouped according WHO and ASRM guidelines [4]: normal (concentration ≥ 15 x 106 cells/ml, motility ≥ 40 %), sub-fertile (concentration < 15 x 106 cells/ml) and severe oligospermia (concentration < 5 x 106 cells/ml). Two semen processes would be used for comparative purpose, centrifugation and chip-sorting. The adopted key quality measures included sperm motility (via Computer-assisted sperm analysis, C.A.S.A.), sperm apoptosis (via TUNEL assay) and DNA fragmentation (via Halo sperm assay). In normal and sub-fertile, although centrifugation and flow chip process would both promote sperm motility (in progressiveness, and linearity). However, no statistical differences were observed in all experiment measures. Meanwhile, in oligospermia group, chip sorted sample showed significantly (P<0.01) improvement in sperm apoptosis rate (from 86% down to 69% in TUNEL assay), as well as in DNA fragmentation rate (from 92% down to 74% in Halo sperm assay). Sorting of sperms with reverse progressive characteristic may have significant clinical efficacy in ART treatment, especially for patients with severe oligospermia.
Selection of functional sperm by using hyaluronic acid modified magnetic microbeads and an electromagnetic manipulation system

Abstract:In recent years, the incidence of infertility has increased year by year. Assisted reproductive technology (ART) is one of the effective strategies to treat infertility. In the process of ART, commonly used methods for sperm separation have shortcomings and there is still room for improvement. In this study, a functional sperm selection strategy was established based on hyaluronic acid (HA) modified magnetic microbeads (MBs) and a supporting two-dimensional electromagnetic manipulation device system. Due to the modification of HA on the surface of MBs, the HA-MBs have the ability to target and bind to specific receptors on the sperm membrane to form a sperm-MB complex. A disulfide linker was introduced to connect HA and MBs. After modifying HA and connecting it with the disulfide linker, the sperm of sperm-MBs can be released under the combined effect of hyaluronidase and reduced GSH with the disulfide bond broken and HA degraded. A two-dimensional electromagnetic manipulation system was introduced to generate a magnetic field and control the directional movement of the sperm-MB complex under the guidance of an inverted microscope. The free MBs can also be removed after the sperm is released. Furthermore, the mouse sperm selected through this strategy can achieve normal insemination via ICSI and the obtained blastocysts have normal morphology and developmental milestones. This strategy has potential to be developed into an automated screening solution for the screening of functional sperm for assisted reproductive technology.
The Design and Optimization of a Microfluidic Device for Sperm Motility Screening

Abstract:Objectiye To optimize the depth of the microchannel and the time point for sperm collection,and improve the efficiency of sperm screening on a microfluidic device. Methods Microchannels with four different depths of 25, 50, 100 and 200 μm were tested. Mice sperm were added to the inlet of the microchannel. The relative quantity and motility of sperm in the outlet were recorded at different collection times, i.e. ,5, 15, 30 and 60 min. Statistical method one-way ANOVA and appropriate post-hoc testing were applied to analyze differences between different groups, and further to select the best-fit depth of the microchannel and the time point for collection. Results In microchannels with depths of 25, 50, 100 and 200 μm, the sperm motilities measured in each outlet were (85.4 ± 2.3)%, (85.8 ± 5.8)%,( 87. 2 ± 2. 8 ) %, (76. 5 ± 2. 8 ) % respectively with statistical significance ( F = 5.8, P ＜ 0. 05 ). No obvious differences were found among 25-100 μm channels, however the motility dramatically decreased in the 200 μm group. The relative sperm quantities were (5.2 ±2.0)%, (7.2 ±2.5)%,(12.3 ±2.0)%,(7. 7 ± 1.1 ) % respectively with statistical significance ( F = 6. 9, P ＜ 0. 05), which increased with channel depth from 25 to 100 μm,while it decreased in the 200 μm channel Taking 2 indexes into account, 100 μmwas the most fit channel depth for sperm motility screening. The sperm motility in the outlet gradually decreased with time. At the time points of 5, 15, 30 and 60 min after adding sperm, the sperm motilities were (99. 6 ±0. 7)%, (87.2 ±2. 8)%, (79. 3 ±2. 2)% and (62. 6 ±8.0)% respectively with statistical significance ( F = 37. 3, P ＜ 0. 01 ). Yet the relative quantities of sperm in the outlet increased almost three times in this process. At the time points mentioned above, the relative quantities of sperm were (5.8±1.1)%, (10.6 ± 0.9)%, (12.1 ± 1.7)%, (17.9 ± 3.4)% respectively with statistical significance ( F = 17.8, P ＜ 0. 01 ). Thus 15-30 min was the ideal screening time. Conclusion An effective microdevice for sperm screening with optimized depth and collection time period is developed,which may contribute significantly for the screening of healthy sperm on microfluidic chips.

Abstract:Background and Aims: While density gradient centrifugation (DGC) is a conventional method in sperm preparation, this method can induce reactive oxygen species (ROS) which cause sperm dysfunction, sperm DNA damage, a recent advancement in sperm processing using microfluidic system is believed to reduce damage and to select the good quality sperm. The aim of this study was to evaluate the effectiveness of sperm preparation between DGC method and microfluidic system. Method: 30 semen samples were separated into 2 equal small samples, each sample was processed by DGC methods (control) and microfluidic (intervention) respectively. Pre- and post processing parameters was evaluated according to WHO-2010 and the DNA fragmentation index (DFI) was determined by flow cytometry technique. Results: The result showed that DFI values from the microfluidic preparation (0.67 ± 0.53%) was significantly lower than that of the DCG group (1.18 ± 13.73%). In particular, for semen samples with high DFI (>15%), the microfluidic system showed a noticeable reduction in DFI compared to the results from the DGC method. The progressive motile sperm recovery rate was higher in the intervention group (39.75 ± 19.20 vs 32.14 ± 18.14). Our data also showed that the vitality and mobility of sperm in the microfluidic group was higher than those of the DGC group (97.9 ± 2.21 vs 96.56 ± 4.11%, 96.36 ± 2.60 vs 95.06 ± 2.35%, respectively). However, there was no difference in sperm progressive motility and sperm morphology between two groups. Conclusions: There were significant improvement in sperm DFI and progressive motility in the microfluidic system. With this result, microfluidic system is a potential approach to reduce DNA fragmentation for ICSI procedures.
Lab-on-chip (LoC) application for quality sperm selection: An undelivered promise?

Abstract:Quality sperm selection is essential to ensure the effectiveness of assisted reproductive techniques (ART). However, the methods employed for sperm selection in ART often yield suboptimal outcomes, contributing to lower success rates. In recent years, microfluidic devices have emerged as a promising avenue for investigating the natural swimming behavior of spermatozoa and developing innovative approaches for quality sperm selection. Despite their potential, the commercial translation of microfluidic-based technologies has remained limited. This comprehensive review aims to critically evaluate the inherent potential of lab-on-chip technology in unraveling sophisticated mechanisms encompassing rheotaxis, thermotaxis, and chemotaxis. By reviewing the current state-of-the-art associated with microfluidic engineering and the swimming of spermatozoa, the goal is to shed light on the multifaceted factors that have impeded the broader commercialization of these cutting-edge technologies and recommend a commercial that can surmount the prevailing constraints. Furthermore, this scholarly exploration seeks to enlighten and actively engage reproductive clinicians in the profound potential and implications of microfluidic methodologies within the context of human infertility.
Microfluidic sperm selection yields higher sperm quality compared to conventional method in ICSI program: A pilot study

Abstract:An appropriate preparation technique, should be capable of isolating highquality spermatozoa for intracytoplasmic sperm injection (ICSI). The aim was to assess sperm quality parameters, DNA integrity, embryo development, and clinical outcomes using a practical and accessible Microfluidic Sperm Sorting (MSS) technique. A total of 95 ICSI cases performed using sperm samples were prepared with our MSS (group 1) or by Direct Swim Up (DSU; control) method (group 2). Both sperm quality parameters and sperm DNA fragmentation (SDF) were compared between the groups. DNA fragmentation was assessed using Sperm Chromatin Dispersion (SCD) test and fine morphology was assessed using Motile Sperm Organelle Morphology Examination (MSOME). Embryo development and clinical outcomes were compared between the groups. In the MSS group, progressive motility and the fraction of Class I sperm morphology sperm were significantly higher compared to DSU group (P < 0.01 and P < 0.001, respectively). Moreover, the rates of DNA fragmentation and immotile spermatozoa were significantly lower in MSS when compared to DSU group (P < 0.001). Also, higher rates of high-quality embryo formation (P < 0.001), implantation (P = 0.04) and pregnancy (P = 0.05) were achieved in the MSS compared to DSU groups. The MSS technique proved to be a noninvasive, disposable, easy to use, and inexpensive method for separation of high-quality spermatozoa. Both laboratory parameters and clinical outcomes were improved with application of MSS for neat sperm collection in ICSI. Abstract:About 18% of reproductive-age adults worldwide are affected by infertility. In vitro fertilization (IVF) and intracytoplasmic sperm injection (ICSI) are widely used assisted reproductive technologies (ARTs) aimed at improving clinical outcomes. Efficient and noninvasive selection and isolation of highly motile sperm with intact DNA are essential for the success of IVF and ICSI and can potentially impact the therapeutic efficacy and the health of the offspring. Compared to traditional methods, microfluidic technology offers significant advantages such as low sample consumption, high efficiency, minimal damage, high integration, similar microenvironment, and high automation, providing a new platform for ARTs. Here, we review the current situation of microfluidic technology in the field of sperm motility screening and evaluation and IVF research. First, we focus on the working principle, structural design, and screening results of sperm selection microfluidic platforms. We then highlight how the multiple steps of the IVF process can be facilitated and integrated into a microfluidic chip, including oocyte capture, sperm collection and isolation, sperm sorting, fertilization, and embryo culture. Ultimately, we summarize how microfluidics can complement and optimize current sperm sorting and IVF protocols, and challenges and possible solutions are discussed.

[Analyze on volatile compounds of Antrodia camphorata using HS-SPME-GC-MS].

Abstract:OBJECTIVE To analyze the volatile compounds of Antrodia camphorata in solid-state and submerged cultures. METHODS A headspace solid-phase microextraction (HS-SPME) coupled with gas chromatography-mass spectrometry(GC-MS) were used to evaluate the profile of the volatile compounds. RESULTS 49 volatile compounds were identified in A. camphorata mycelia in submerged culture, while 43 volatile compounds were identified in mycelia in solid-state culture. 1-octen-3-ol, 3-octanone, 1-octen-3-ylacetate, acetic acid octyl ester and ethanol were the main volatile compounds in A. camphorata mycelia in submerged culture, while 1-octen-3-ol, 3-octanone, 3-methyl-butyraldenhyde, gamma-podecalactone and methyl 2-furozte were the most potent key volatile compounds in mycelia in solid-state culture. CONCLUSION The volatile compounds in the mycelia of A. camphorata in solid-state and submerged cultures are similar but their relative contents are different.

Abstract:Abstract STUDY QUESTION Is a microfluidic sperm sorter (MSS) able to select higher quality sperm compared to conventional methods? SUMMARY ANSWER The MSS selects sperm with improved parameters, lower DNA fragmentation, and higher fertilizing potential. WHAT IS KNOWN ALREADY To date, the few studies that have compared microfluidics sperm selection with conventional methods have used heterogeneous study population and have lacked molecular investigations. STUDY DESIGN, SIZE, DURATION The efficiency of a newly designed MSS in isolating high-quality sperm was compared to the density-gradient centrifugation (DGC) and swim-up (SU) methods, using 100 semen samples in two groups, during 2023–2024. PARTICIPANTS/MATERIALS, SETTING, METHODS Semen specimens from 50 normozoospermic and 50 non-normozoospermic men were sorted using MSS, DGC, and SU methods to compare parameters related to the quality and fertilizing potential of sperm. The fertilizing potential of sperm was determined by measurement of phospholipase C zeta (PLCζ) and post-acrosomal sheath WW domain-binding protein (PAWP) expression using flow cytometry, and the chromatin dispersion test was used to assess sperm DNA damage. MAIN RESULTS AND THE ROLE OF CHANCE In both normozoospermic and non-normozoospermic groups, the MSS-selected sperm with the highest progressive motility, PLCζ positive expression and PLCζ and PAWP fluorescence intensity the lowest non-progressive motility, and minimal DNA fragmentation, compared to sperm selected by DGC and SU methods (P < 0.05). LIMITATION, REASONS FOR CAUTION The major limitations of our study were the low yield of sperm in the MSS chips and intentional exclusion of severe male factor infertility to yield a sufficient sperm count for molecular experiments; thus testing with severe oligozoospermic semen and samples with low count and motility is still required. In addition, due to ethical considerations, at present, it was impossible to use the sperm achieved from MSS in the clinic to assess the fertilization rate and further outcomes. WIDER IMPLICATIONS OF THE FINDINGS Our research presents new evidence that microfluidic sperm sorting may result in the selection of high-quality sperm from raw semen. This novel technology might be a key to improving clinical outcomes of assisted reproduction in infertile patients.
