Abstract:Daylily (Hemerocallis fulva Linn.) flowers were hot air-dried and freeze-dried after harvest. Antioxidant properties of water and ethanol extracts prepared from these dried flowers were evaluated in terms of total antioxidant activity, reducing capacity, and metal chelating activity. Extracts from daylily flowers exhibited strong antioxidant activity. Ethanol was more efficiency to extract antioxidants than water, and freeze-drying preserved higher activities than air-drying. Rutin, (+)-catechin, and gallic acid were identified in the extracts by HPLC, and were highly related to the antioxidant activities. The antioxidant activity was further evaluated by feeding mice with ethanol extract from freeze-dried daylily flowers for 60 days. The results demonstrated that the extract at dosage of 40-225 mg/100 g significantly increased the activity of SOD (superoxide dismutase) and reduced the lipid peroxidation in both blood and liver of rat.

Abstract:The antioxidant properties of methanol extracts from daylily flowers during maturation were determined with antioxidant assays, including antioxidant activity, DPPH (2,2-diphenyl-1-picrylhydrazyl) radical scavenging, superoxide anion radical scavenging and reducing power. Antioxidant compounds, such as phenolic compounds, ascorbic acid and β-carotene were also analysed. Significant variation in antioxidant properties and involved compounds was observed between different maturity stages of daylily flowers. The highest antioxidant activity was observed at stage III (flower opening) accompanying the highest content of ascorbic acid and phenolic compounds, while no significant difference of β-carotene contents was observed among the four maturity stages. Four individual phenolics, such as (+)-catechin, chlorogenic acid, rutin and quercetin were identified and quantified by HPLC. (+)-Catechin was the main phenolic compound identified in daylily flowers, accounting for about 74.11% of total phenolics. Overall, daylily flowers at opening stage possess the highest functional benefit and thus would be the appropriate harvesting stage in view of the nutritional consideration.
Suppression of Nitrosation by Extracts of Daylily (Hemerocallis fulva) Flowers

Abstract:Fresh daylily flowers were processed into powder by the following procedures of blanching, drying and comminution, and the powder was extracted with different solvents. The suppression effects of daylily flower extracts on nitrosation were evaluated in terms of the nitrite scavenging activity and nitrosamine synthesis inhibiting activity determined by spectrophotometric method. The 70% methanol extract of daytily flower obtained by the water bath extraction for 0.5 h in an ultrasonic filed had high nitrite scavenging activity and nitrosamine synthesis inhibiting activity, reaching 66.9% and 54.7%, respectively. The nitrite scavenging activity and nitrosamine synthesis inhibiting activity of daylily flower varied slightly during fridge storage. Among four cultivars (Mengzihua, Baihua, Xiye and Daogan) of daylily flowers, Daogan had the highest nitrite scavenging activity and nitrosamine synthesis inhibiting activity and Xiye the lowest. Stream blanching treatment could maintain higher nitrosamine synthesis inhibiting activity than boiling water blanching treatment, although daylily flowers treated by them both had lower nitrosamine synthesis inhibiting activity than unblanched ones. A highest nitrosamine synthesis inhibiting activity was found in freeze-dried daylily flowers in comparison with sunlight-dried ones followed by hot air-dried ones. In general daylily flowers possess some nitrosamine synthesis inkihiting activity which is affected by cultivars and blanching, drying and cool storage treatments.
Natural deep eutectic solvent enhanced pulse-ultrasonication assisted extraction as a multi-stability protective and efficient green strategy to extract anthocyanin from blueberry pomace

Abstract:A deep understanding upon the relation between anthocyanin extraction performance and physicochemical properties of natural deep eutectic solvents (NADESs) was obtained, and a high-efficiency NADES (Choline chloride-Oxalic acid (ChOa)) was selected and coupled with pulse-ultrasonication for anthocyanin extraction from blueberry pomace. Response surface methodology was adopted to analyzed the significant factors' influence, interactive effects on extraction yield and optimize the extraction conditions. The theoretical optimal conditions with highest yield of total anthocyanins content (TAC) were 3.2 min of ultrasonic time, 325 W of ultrasonic power, 349.15 K of temperature, 60 mL/g of solvent to solid ratio, under which the experimental TAC value (24.27 +/- 0.05 mg C3GE/g DW) was highly correlated with theoretical result (24.28 mg C3GE/g DW). Multi-stability protective effect of ChOa on anthocyanin over acidified ethanol was revealed by stability assays in high temperature, varying pH and light exposure conditions. FT-IR and DSC analysis further characterized the influence of ChOa over acidic ethanol on typical anthocyanins functional groups and thermal behavior of anthocyanin extracts. UPLC-Triple-TOF/MS analysis identified 10 anthocyanins that mainly consisted of delphinidin, petunidin, and malvidin compounds. Overall, this study provided a green, high-efficiency and multistability protective strategy for the recovery of anthocyanins from plant source by-products.

Abstract:This study was performed to investigate the physicochemical properties of lutein microencapsulation (LM) and to assess its bioavailability (BA). LM was prepared with advanced spray and starch-catching drying technology. Its physicochemical properties (characterization, storage stability and dissolution) and BA were evaluated. The BA study of LM was performed using SD rats following a single oral administration of lutein equivalent to 100 mg, and were compared with commercial reference sample (RS). LM was nearly sphericity, its mean particle size was 214.7 nm. It can be free-flowing. Solubility of lutein was increased and it could be directly dissolved in water to form a homogeneity of solution. The storage stability of lutein was enhanced under accelerated and long-term storage conditions. More than 85% lutein could be released from LM after 15 min during the dissolution test. The relative BA of lutein was 139.1% in comparison to RS. The results would be helpful to conduct the application of LM in food industry.

Abstract:Different cultivars of functional vegetables may vary in an antioxidant capacity. In this study, the antioxidant activity of methanol extracts from five cultivars of daylily flower grown in China was compared by various antioxidant assays, including total antioxidant activity, free radical scavenging, superoxide anion radical scavenging and reducing power, the various antioxidant activities were compared to standard antioxidants such as butylated hydroxyanisole and ascorbic acid. All the extracts showed strong antioxidant activity in all tested methods. Among the five cultivars, Panlonghua and Mengzihua were found to possess the highest antioxidant activity, while Xiye and Baihua were found to possess the lowest antioxidant activity. In addition, a high positive correlation between the antioxidant properties and total phenolic content was observed.

Abstract:Daylily is a valuable plant resource with various health benefits. Its main bioactive components are phenolic compounds. In this work, four extraction methods, ultrasonic-assisted water extraction (UW), ultrasonic-assisted ethanol extraction (UE), enzymatic-assisted water extraction (EW), and enzymatic-assisted ethanol extraction (EE), were applied to extract phenolic compounds from daylily. Among the four extracts, the UE extract exhibited the highest total phenolic content (130.05 mg/100 g DW) and the best antioxidant activity. For the UE extract, the DPPH value was 7.75 mg Trolox/g DW, the FRAP value was 14.54 mg Trolox/g DW, and the ABTS value was 15.37 mg Trolox/g DW. A total of 26 phenolic compounds were identified from the four extracts, and the UE extract exhibited a higher abundance range of phenolic compounds than the other three extracts. After multivariate statistical analysis, six differential compounds were selected and quantified, and the UE extract exhibited the highest contents of all six differential compounds. The results provided theoretical support for the extraction of phenolic compounds from daylily and the application of daylily as a functional food.

Abstract:Hemerocallis fulva is a medical and edible plant. In this study, we optimized the ultrasound-assisted extraction (UAE) process of extracting flavonoids from Hemerocallis fulva leaves by single-factor experiments and response surface methodology (RSM). The optimum extraction conditions generating the maximal total flavonoids content was as follows: 70.6% ethanol concentration; 43.9:1 mL/g solvent to sample ratio; 61.7 °C extraction temperature. Under the optimized extraction conditions, the total flavonoid content (TFC) in eight Hemerocallis fulva varieties were determined, and H. fulva (L.) L. var. kwanso Regel had the highest TFC. The cytotoxicity of the extract was studied using the Cell Counting Kit-8 (CCK-8 assay). When the concentration was less than 1.25 mg/mL, the extract had no significant cytotoxicity to HaCaT cells. The antioxidant activity was measured via chemical antioxidant activity methods in vitro and via cellular antioxidant activity methods. The results indicated that the extract had a strong ABTS and •OH radical scavenging activity. Additionally, the extract had an excellent protective effect against H2O2-induced oxidative damage at a concentration of 1.25 mg/mL, which could effectively reduce the level of ROS to 106.681 ± 9.733% (p < 0.001), compared with the 163.995 ± 6.308% of the H2O2 group. We identified five flavonoids in the extracts using high-performance liquid chromatography (HPLC). Infrared spectroscopy indicated that the extract contained the structure of flavonoids. The results showed that the extract of Hemerocallis fulva leaves had excellent biocompatibility and antioxidant activity, and could be used as a cheap and potential source of antioxidants in the food, cosmetics, and medicine industries.

Abstract:The ultrasonic extraction (UE) technology, possessed the advantages of effective, energy‐saving, and environmental‐friendly, was applied to extract the anthocyanin from Lycium ruthenicum (LR). The extraction parameters of UE were optimized by response surface methodology (RSM) with Box–Behnken design (BBD). Anthocyanin composition in LR fruits grown in China was systematically identified and quantified by HPLC‐ESI‐MS. The result showed that PRG was the major anthocyanin, and delphinidin, petunidin, and malvidin were the major anthocyanidins in LR fruits. There was the same anthocyanin composition of LR and great variation in anthocyanins content of LR from different areas in China. However, there was no significant difference between wild and cultivated LR in the same region. A clear separation of LR according to geographical origins was revealed by hierarchical cluster analysis (HCA) and principal component analysis (PCA), and the discrimination model for the anthocyanin concentrations were developed using these two analysis methods. Furthermore, on‐line HPLC‐DPPH assay and scavenging activity of three kinds of radicals (DPPH·, ·OH, and ) in vitro were well applied to evaluate the antioxidant activity of the LR anthocyanin extract (LRAE). And its results indicated the LRAE could be a credible antioxidant agent for applications in cosmetics, food, and medicine.

Abstract:Lutein has several benefits for human health, playing an important role in the prevention of age-related macular degeneration (AMD), cataracts, amelioration of the first stages of atherosclerosis, and some types of cancer. In this work, the Scenedesmus almeriensis microalga was used as a natural source for the supercritical fluid (SF) extraction of lutein. For this purpose, the optimization of the main parameters affecting the extraction, such as biomass pre-treatment, temperature, pressure, and carbon dioxide (CO2) flow rate, was performed. In the first stage, the effect of mechanical pre-treatment (diatomaceous earth (DE) and biomass mixing in the range 0.25–1 DE/biomass; grinding speed varying between 0 and 600 rpm, and pre-treatment time changing from 2.5 to 10 min), was evaluated on lutein extraction efficiency. In the second stage, the influence of SF-CO2 extraction parameters such as pressure (25–55 MPa), temperature (50 and 65 °C), and CO2 flow rate (7.24 and 14.48 g/min) on lutein recovery and purity was investigated. The results demonstrated that by increasing temperature, pressure, and CO2 flow rate lutein recovery and purity were improved. The maximum lutein recovery (~98%) with purity of ~34% was achieved operating at 65 °C and 55 MPa with a CO2 flow rate of 14.48 g/min. Therefore, optimum conditions could be useful in food industries for lutein supplementation in food products.

Abstract:Response surface methodology was used to investigate the ultrasonic-assisted extraction of anthocyanins from Lycium ruthenicum Murr. A Box-Behnken design of three-level-three-factor was used to optimize extractive factors, including ethanol concentration (A), ultrasonic extraction time (B), and liquid-solid ratio (C) and to achieve high extraction yield of the anthocyanins. The three parameters were optimized at 70% (v/v), 28min, 33: 1 (mL/g), respectively, the best extraction yield of anthocyanins was 7.12 ± 0.093%. The AB-8 resin was further to be chosen for purification the anthocyanins. Anthocyanin composition was analyzed by HPLC-MS2, the possible structures can be speculated as petunidin-3-O-(glucosyl-p-coumaroyl)-rutinoside-5-O-glucoside, pentuidin-3-O-(caffeoyl)-rutinoside-5-O-glucoside, delphinidin-3-O-(p-coumaroyl)-rutinoside-5-O-glucoside, petunidin-3-O-(p-coumaroyl)-rutinoside-5-O-glucoside, malvidin-3-O-(p-coumaroyl)-rutinoside-5-O-glucoside, cyanidin-3-O-rutinoside, and petunidin-3-O-(p-coumaroyl)-rutinoside. The thermal stability indicated the high purity of anthocyanins, and purified anthocyanins combined with whey protein isolate (WPI) can enhance the thermal stability of anthocyanins. The thermal stability of the purified anthocyanins was higher than that of the unpurified anthocyanins; whereas the thermal stability of the purified anthocyanin-WPI synthesis was greater than that of the purified anthocyanins. The thermal stable anthocyanins and anthocyanin-protein synthesis has the potential application in the functional food industry.
Optimization of Lutein Recovery from Tetraselmis suecica by Response Surface Methodology

Abstract:Microalgae have been attracting attention as feedstock for biorefinery because they have various advantages, such as carbon fixation, high growth rate and high energy yield. The bioactive compounds and lutein contained in microalgae are known to be beneficial for human health, especially eye and brain health. In this study, in order to improve the recovery of bioactive extracts including lutein from Tetraselmis suecica with higher efficiency, an effective solvent was selected, and the extraction parameters such as temperature, time and solid loading were optimized by response surface methodology. The most effective solvent for lutein recovery was identified as 100% methanol, and the optimum condition was determined (42.4 °C, 4.0 h and 125 g/L biomass loading) by calculation of the multiple regression model. The maximum content of recovered lutein was found to be 2.79 mg/mL, and the ABTS radical scavenging activity (IC50) and ferric reducing antioxidant power (FRAP) value were about 3.36 mg/mL and 561.9 μmol/L, respectively. Finally, the maximum lutein recovery from T. suecica through statistical optimization was estimated to be 22.3 mg/g biomass, which was 3.1-fold improved compared to the control group.

Sustainable extraction of ligustilide and ferulic acid from Angelicae Sinensis Radix, for antioxidant and anti-inflammatory activities

Abstract:The 2030 Agenda for Sustainable Development envisions a rational use of energy and resources in all technological processes. However, in the extraction methods of compounds from medicinal plants and herbs, there is an urgent to reduce the use of organic solvents and increase the energy efficiency of these methods. Therefore, a sustainable extraction method (enzyme and ultrasonic co-assisted aqueous two-phase extraction, EUA-ATPE) of simultaneous extraction and separation of ferulic acid and ligustilide from Angelicae Sinensis Radix (ASR) was developed by integrating enzyme-assisted extraction (EAE) with ultrasonic-assisted aqueous two-phase extraction (UAE- ATPE). The effects of different enzymes, extraction temperature, pH, ultrasonic time, liquid-to-materials ratio, etc., were optimized by single-factor experiments and central composite design (CCD). Under the optimum conditions, the highest comprehensive evaluation value (CEV) and extraction yield were obtained by EUA-ATPE. Furthermore, recovery (R), partition coefficient (K), and scanning electron microscopy (SEM) analysis revealed that enzyme and ultrasonic treatment improved mass transfer diffusion and increased the degree of cell disruption. Besides, the EUA-ATPE extracts have shown great antioxidant and anti-inflammatory activity in vitro. Finally, compared to different extraction methods, EUA-ATPE achieved higher extraction efficiency and higher energy efficiency due to the synergistic effect between EAE and UAE-ATPE. Therefore, the EUA-ATPE provides a sustainable method for extracting bioactive compounds from medicinal plants and herbs, contributing to Sustainable Development Goals (SDG), including SDG-6, SDG-7, SDG-9, SDG-12, and SDG-15.

Abstract:Thirteen patients with severe partially reversible chronic obstructive lung disease received long-term treatment with a sustained-release preparation of theophylline and a beta 2-agonist (terbutaline) alone and in combination. Combination therapy induced significantly greater bronchodilation than treatment with terbutaline alone in the recommended dose. In 9 out of 13 patients the peak flows were higher during combination therapy than during theophylline treatment alone, but this did not reach statistical significance. Only mild side effects were observed.

Abstract:Lutein is the main component of macular and retinal pigment so that it is essential for eye health. In addition, lutein is also related to various metabolic processes in the human body with strong antioxidant properties. The green extraction of lutein in wet microalgae is critical for the further applications as nutraceuticals and pharmaceuticals. Ionic liquid-based aqueous two-phase system (ATPS) is a green approach for multiple product extraction from microalgae. In this study, the extraction performances of ATPS composed of four different cholinium-based compounds (choline geranate, choline chloride, choline dihydrogen citrate and choline bitartrate) and polyethylene glycol 400 (PPG400) were compared. The choline dihydrogen citrate (Ch DHcit) showed a better extraction ability of lutein than other cholinium-based ILs. After optimizing the addition of cholinium-based ILs and PPG400, the ATPS with the best composition (PPG400: Ch Dhcit: water: algal suspension = 42:26:22:10 wt%) was applied to the extraction of lutein from wet Chlorella sp. Six imidazolium-based ionic liquids (1-ethyl-3-methylimidazole chloride, 1-ethyl-3-methylimidazolium bromide, 1-butyl-3-methylimidazolium bromide, 1-butyl-3-methylimidazole chloride, 1-hexyl-3-methylimidazole chloride and 1-hexyl-3-methylimidazolium bromide) were compared as an adjuvant for the lutein extraction from wet Chlorella sp. The introduction of a small amount of 1-ethyl-3-methylimidazole chloride ([C2mim]Cl) improved the extraction efficiency of lutein increased by 9.98 %. In the multiple-times extraction process, the total extraction efficiency of three times distribution reached 102.61 %. This paper provides a new approach to the extraction of lutein from algal biomass.

Assume that the players are better off locating at point B than not locating at the same point, but they are even better off if they locate at point A. Thus, there are gains to coordinating at any point A or B, and further gains to coordinating at point A instead of point B. For this game, there are multiple equilibria. Ultrasound-assisted extraction of polyphenols from lotus rhizome epidermis by alcohol/salt-based aqueous two-phase system: Optimization, extraction mechanism and antioxidant activities

Abstract:In this study, ultrasonic-assisted (UA) alcohol/salt-based aqueous two-phase system (ATPS) method was constructed to extract lotus rhizome epidermis (LRE) polyphenols. The extraction conditions were optimized as salt concentration 26.75 %, ethanol concentration 25.45 %, ultrasonic power 487 W and liquid-solid ratio 35.33 mL/g by comparing response surface methodology (RSM) and artificial neural network (ANN) models. Then, l-dopa (2.35 ± 0.036 mg/g dw), gallocatechin (1.66 ± 0.0035 mg/g dw) and epigallocatechin (1.37 ± 0.0035 mg/g dw) were determined as major polyphenols in LRE by using UA-ATPS method. Moreover, study showed that ultrasound, van der Waals force, hydrogen bond and salting out could accelerate the mass transfer and extraction of polyphenols in LRE cells. The high-pressure cavity and collapse effect of ultrasound could also accelerate the extraction of polyphenols. In vitro antioxidant experiments showed that LRE polyphenols have good antioxidant ability. In sum, this study developed a green and efficient extraction method to enhance the profitability of LRE in food and medicine industries.

Abstract:Lutein is a xanthophyll carotenoid with remarkable applications in the healthcare sector due to its ocular-protective, anti-oxidative and anti-inflammatory activities. Currently, marigold flower petals are the only commercial source for lutein extraction, since chemical synthesis suffers from very low yields. Microalgae cultivation is a promising way for lutein production, as the lutein content of microbial biomass is higher compared to that of marigold petals. In addition, microalgae are shown to have a higher growth rate, higher photosynthetic efficiency, and carbon mitigation potential when compared to terrestrial plants, such as marigolds. For the realization of microalgae as a sustainable and alternative lutein source, crucial factors such as the microalgal strains, critical cultivation parameters (light intensity, nutritional status, metabolic mode), pretreatment, extraction, and purification of lutein from microalgal biomass need to be studied in great detail. In this context, this review summarized the recent research progress in lutein production from microalgae, including competent microalgal strains, cultivation systems, and subsequent biomass treatment technologies. The choice of a high yielding strain, along with effective photobioreactor design will enable maximal lutein production from microalgae in an economically viable manner. This review presents comprehensive information on microalgal cultivation and pretreatment which will enable researchers to design an optimal microalgae-based lutein production system.
